In the recent years, tissue engineering researchers have exploited a variety of biomaterials that can potentially mimic extracellular matrix (ECM) for tissue regeneration. Natural cellulose, mainly obtained from bacterial (BC) and plant-based (PC) sources, can serve as a high potential scaffold material for different regenerative purposes. Natural cellulose has drawn the attention of researchers due to its advantage over synthetic cellulose in terms of availability, cost-effectiveness, perfusablility, biocompatibility, negligible toxicity, mild immune response and due to imitating native tissues. In this article, we will review the recent in vivo and in vitro studies aimed to assess the potentials of natural cellulose for the purpose of soft (skin, heart, veins, nerve, among others) and hard (bone and tooth) tissue engineering.

The regeneration of bone tissue is a main purpose of most therapies in dental medicine. For bone regeneration, calcium phosphate (CaP)-based substitute materials based on natural (allo- and xenografts) and synthetic origins (alloplastic materials) are applied for guiding the regeneration processes. The optimal bone substitute has to act as a substrate for bone ingrowth into a defect, while it should be resorbed even in the time frame needed for complete regeneration up to the condition of restitution ad integrum. In this context, the modes of action of CaP-based substitute materials have been frequently investigated and it has been shown that such materials strongly influence regenerative processes such as osteoblast growth or differentiation and also on osteoclastic resorption due to different physicochemical properties of the materials. However, the material characteristics needed for the required ratio between the formation of new bone tissue and material degradation has not been found until now. The addition of different substances such as collagen or growth factors and also of different cell types have already been tested but did not allow for sufficient or prompt application. Moreover, metals or metal ions are differently used as basis or as supplement for different materials in the field of bone regeneration. Moreover, it has already been shown that different metal ions are integral components of bone tissue playing functional roles in the physiological cellular environment as well as in the course of bone healing. The present review focuses on frequently used metals as integral parts of materials designated for bone regeneration with the aim to give an overview of currently existing knowledge about the effects of metals in the field of bone regeneration.

Qinghai spruce (Picea crassifolia) is a ecologically important species in the forest ecosystem of the Qilian Mountains region in China. Natural regeneration of this species is critical to maintaining forest ecosystem function. We here analyzed several biological indicators among naturally re-generating Qinghai spruce across several elevations in the Pailugou watershed. Specifically, seedling density, basal diameter (BD), and plant height were measured, as were soil physico-chemical parameters, at 2700 m, 3000 m, and 3300 m above sea level. Differences in the biological indicators and correlations between the indicators and soil parameters were then assessed at each elevation. The results showed that soil stoichiometry was more sensitive to changes in el-evation than seedling indicators were. Furthermore, seedling density was positively correlated with soil pH, whereas BD was positively correlated with the carbon to nitrogen ratio (C/N), the carbon to phosphorus ratio, and soil organic carbon (SOC) contents. None of the analyzed soil parameters had a significant impact on elevation-specific differences in seedling density. However, soil pH, SOC, and C/N significantly affected variations in seedling diameter at each elevation. Finally, soil pH, SOC, C/N, and the carbon to phosphorus ratio significantly affected variations in seed-ling height at each elevation. This study provides a strong theoretical basis for further under-standing of the mechanisms associated with Qinghai spruce regeneration, ulti-mately contrib-uting to rational protection and management strategies for this important natural resource.

Forest managers are often required to restore forest stands following natural disturbances, a situation that may become more common and more challenging under global change. In parts of Central Europe, particularly in mountain regions dominated by mixed temperate forests, the use of relatively low intensity, uneven-aged silviculture is a common management approach. Because this type of management is based on mimicking less intense disturbances, the restoration of more severe disturbance patches within forested landscapes has received little attention within the context of uneven-aged silviculture in the region. The goal of this paper is to synthesize research on the restoration of forests damaged by disturbances in temperate forests of Slovenia and neighbouring regions of Central Europe, where uneven-aged silviculture is practiced. We place particular emphasis on the most important biotic and abiotic drivers of post-disturbance regeneration, and use this information to inform silvicultural decisions about applying natural or artificial regeneration in disturbed areas. We conclude with guidelines for restoration silviculture in uneven-aged forest landscapes.

Annelids are well-known for their ability to regenerate. The morphological development in terms of regeneration physiology, involves several cellular factors which play crucial roles in the developmental phenomena. The physiological process of regeneration involves critical biomechanical processes which is effectively regulated by the cellular factors. In today’s biological research, the fundamental analysis of regeneration is necessary for the scientific assessment of cellular-biomechanical advancements, which eventually helps to develop adequate insight in the biomedical research in regenerative sciences. In this review we have elucidated the recent research on explaining the developmental physiology of regeneration in annelids.

Background: It has been demonstrated that pressotherapy used post-exercise (Po-E) can influence training performance, recovery, and physiological properties. This study examined the effec-tiveness of pressotherapy on these parameters. Methods: The systematic review and meta-analysis were performed according to PRISMA guidelines. A literature search of MEDLINE, PubMed, EBSCO, Web of Science, SPORTDiscus and ClinicalTrials has been done up to March 2021. Inclusion criteria were: randomized control trials (RCTs) or cross-over studies, mean participant age be-tween 18-65 yrs., ≥ 1 exercise mechanical pressotherapy intervention. The risk of bias was assessed by the Cochrane risk-of-bias tool for RCT (RoB 2.0). Results: 12 studies comprised of 322 partici-pants have been selected. The mean sample size was n = 25. Pressotherapy significantly reduce muscle soreness(Standard Mean Difference;SMD= -0.33; CI = -0.49, -0.18; p < 0.0001; I2 = 7%). Pres-sotherapy did not significantly affect jump height (SMD = -0.04; CI =-0.36, -0.29; p = 0.82). Presso-therapy did not significantly affect creatine kinase level 24-96h after DOMS induction (SMD = 0.41; CI = -0.07, 0.89; p = 0.09; I2 = 63%). Conclusions: Only moderate benefits of using pressotherapy as a recovery intervention have been observed. Results varied between the type of exercise and used protocol. Pressotherapy should only be applied as an additional component of a more compre-hensive recovery strategy. Study PROSPERO registration number- CRD42020189382.

Injured peripheral nerves regenerate their axons in contrast to those in the central nervous system. However, functional recovery after surgical repair is often disappointing. The basis for the poor recovery is the progressive deterioration with time and distance, of the growth capacity of the neurons that lose their contact with targets (chronic axotomy) and the growth support of the chronically denervated Schwann cells (SC) in the distal nerve stumps. This is despite the retained capacity of chronically denervated and atrophic muscle to accept reinnervation. Progressive decline in regeneration associated genes in both axotomized neurons and denervated SCs accounts for the decline in regenerative success in association with silencing of neural activity in sensory neurons due to their disconnection from their sense organs and, in motoneurons due to loss of their synaptic contacts in the spinal cord. Whilst exogenous neurotrophic factors promote nerve regeneration, the profuse axonal outgrowth and difficulties in delivery are avoided by promoting their endogenous expression with brief (1 hour) low frequency (20Hz) electrical stimulation (ES) proximal to the injury site. ES accelerates axon outgrowth and in turn, target reinnervation in both animals and human subjects. Applying ES to intact nerve days prior to nerve injury, conditional ES (CES) increases axonal outgrowth and regeneration rate with the potential for application in nerve transfer surgeries and end-to-side neurorrhaphies. However, the additional surgery for applying CES electrodes may be a hurdle. ES is applicable in all surgeries with excellent outcomes.

Diabetes is a group of diseases characterized by loss of β cell mass and/or -function, resulting in hyperglycemia. Approx. 537 million people worldwide suffer from diabetes – a number which is expected to increase. Diabetes is primarily treated by exogenous insulin, which comes with the challenges of maintaining glycemic control to prevent ketoacidosis and severe complications. The need for a curative treatment has initiated the research in β cell regeneration. Several studies in mice have identified essential genes for β cell fate, which can be manipulated in other cells to induce generation of new β cells. Zebrafish, a regeneration-positive animal model, has shown several different sources of new β cells, including regeneration by self-replication, neogenesis by duct-associated progenitor cells, and transdifferentiation of other endocrine islet cells. The animal models used in this research area are either limited by their low regenerative ability (mice), or their small size and remoteness from humans (zebrafish). There is a need for new animal models of diabetes, in which the molecular pathways of endogenous regeneration can be studied. This study proposes the axolotl salamander (Ambystoma mexicanum) as a model for studying the regeneration of β cells. The axolotl has shown great regenerative capability, as they have proven capable of regenerating amputated limbs, and hearts with myocardial infarction, among other organs. This study aims to establish a diabetic axolotl model, investigate their regenerative ability in the pancreas, and examine the potential systemic effects of the induced disease. In a pilot study, five different protocols using STZ (streptozotocin) were tested, and the most optimal protocol was found. Furthermore, the glucose tolerance test was optimized to characterize the glycemic state of the animals. The effect of the treatment on blood glucose levels was measured to characterize the development and decline of the disease. The histological changes in the pancreas were examined. Moreover, the systemic effects of the STZ treatment were investigated in blood and urine. The study indicated that it is possible to induce diabetes in the axolotl, but variations between the animals should be minimized, or the sample size should be increased to conduct a satisfying experiment, as it was not possible to induce diabetes in all animals. Regeneration was not observed histologically, but a restoration of blood glucose levels was seen over the span of the experiment. Lastly, edema formation was observed in some of the STZ-treated animals, but the cause of edema remains undetermined.

Pericytes, as perivascular cells, are present in all vascularized organs and tissues, and they actively interact with endothelial cells in capillaries and microvessels. Their involvement includes functions like blood pressure regulation, tissue regeneration, and scarring. Studies have confirmed that pericytes play a crucial role in bone tissue regeneration through direct osteodifferentiation processes, paracrine actions, and vascularization. Recent pre-clinical and clinical experiments have shown that combining perivascular cells with osteogenic factors and tissue-engineered scaffolds can be therapeutically effective in restoring bone defects. This approach holds promise for addressing bone-related medical conditions. In this review, we have emphasized the characteristics of pericytes and their involvement in angiogenesis and osteogenesis. Furthermore, we have explored recent advancements in the use of pericytes in preclinical and clinical investigations, indicating their potential as a therapeutic resource in clinical applications.

Objective: There are currently no known treatment methods for ensuring complete recovery from facial nerve damage. Thus, various basic and clinical studies continue to be conducted to achieve this end. As part of this research, autophagy is being investigated for its role in nerve regeneration. Here, we review the existing literature regarding the involvement and signifi-cance of autophagy in facial nerve injury and regeneration. Methods: A comprehensive litera-ture review was conducted to assess the induction and role of autophagy in facial nerve injury and subsequent regeneration. Results: Studies conducted on rats and mice have demonstrated activation of autophagy and expression of related factors in facial and sciatic nerves. Among the most studied of these factors in relation to degeneration and regeneration of facial and sciatic nerves are LC3II/I, PI3K, mTOR, Beclin-1, ATG3, ATG5, ATG7, ATG9, and ATG12. Conclusion: This analysis indicates that autophagy is involved in the process of nerve regeneration follow-ing facial and sciatic nerve damage. Inadequate autophagy induction or failure of autophagy re-sponses can result in regeneration issues after facial or sciatic nerve damage. Animal studies suggest that autophagy plays an important role in facial and sciatic nerve degeneration and re-generation.

The c-Jun N-terminal Kinase (JNK) signalling pathway is a conserved response to a wide range of internal and external cellular stress signals. Besides the stress response, the JNK pathway is involved in a series of vital regulatory mechanisms during development and adulthood that are critical to maintain tissue homeostasis. These mechanisms include the regulation of apoptosis, growth, proliferation, differentiation, migration and invasion. The JNK pathway has such a diverse functionality and cell-tissue specificity, that it has emerged as a key player in regeneration, tumorigenesis and other pathologies such as neurodegenerative diseases. The JNK pathway is highly active in the central nervous system (CNS), and plays a central role for the cells to cope with pathophysiological insults during both development and adulthood. Among the many mechanisms described in the literature, in this review we focus on the JNK pathway functions in pathologies of the CNS. More specifically, we discuss some newly identified examples and mechanisms of JNK-driven tumor progression in glioblastoma, regeneration/repair after an injury in the CNS, neurodegeneration, and neuronal cell death. Recent studies have shown that the JNK pathway regulates matrix metalloproteinases (MMPs) production in response to cytoneme/tumor microtubes formation and Wingless (Wg)/WNT pathway activation in glioblastoma cells. Thus, JNK pathway is essential for glioblastoma progression, infiltration and non-autonomous induction of neurodegeneration. In regeneration, the JNK pathway controls Draper (Drpr) expression in glial cells that mediate engulfment and regeneration of the CNS upon injury.

Composite membranes formed by sulfonated poly(ether ether ketone) (SPEEK) with embedded carbon quantum dots (CQDs) were used for detection and removal of heavy metal ions, such as lead and cadmium, from water. SPEEK is responsible for the capture of heavy metals based on the cation exchange mechanism, while CQDs detect their contamination by exhibiting changes in fluorescence. Water-insoluble “red” carbon quantum dots (rCQDs) were synthesized from p-phenylenediamine so that their photoluminescence was shifted from that of the polymer matrix. CQDs and the composites were characterized by several techniques: FTIR, Raman, UV/VIS, photoluminescence, XPS spectroscopies and AFM microscopy. The heavy metal ion concentration was analyzed by inductively coupled plasma–optical emission spectroscopy (ICP-OES). The concentration ranges were 10.8 - 0.1 mM for Pb2+ and 10.0 - 0.27 mM for Cd2+. The composite achieved 100% removal efficiency for lead and cadmium if the concentration was below a half of the ion exchange capacity of SPEEK. The regeneration of membranes in 1 M NaCl was also studied. SPEEK/rCQDs showed a turn-off effect more pronounced for lead. A second order law was effective to describe the kinetics of the process.

Bone/fracture healing is a complex process with different steps and four basic tissue layers being affected: cortical bone, periosteum, fascial tissue surrounding the fracture, and bone marrow. Stem cells and their derivatives, including embryonic stem cells, induced pluripotent stem cells, mesenchymal stem cells, hematopoietic stem cells, skeletal stem cells, and multipotent stem cells, can function to artificially introduce highly regenerative cells into decrepit biological tissues and augment the healing process at the tissue level. Stem cells are molecularly and functionally indistinguishable from standard human tissues. The widespread appeal of stem cell therapy lies in its potential benefits as a therapeutic technology that, if harnessed, can be applied in clinical settings. This review aims to establish the molecular pathophysiology of bone healing, the current stem cell interventions that disrupt or augment the bone healing process, and finally, consider the future direction/therapeutic options related to stem cells and bone healing.

The contribution aims to reconstruct the trajectory of urban morphology studies focused on the graphical understanding of the transformation phenomena within historical and contemporary fabrics. Highlighting some analogies with urban morphology studies present in the theories and design experiments of authors such as Aldo Rossi, Carlo Oswald W. Ungers, Hans Kollhoff, Saverio Muratori, Gianfranco Caniggia, and Giancarlo De Carlo. These studies mostly matured and de-veloped within a favorable cultural environment, which, albeit with due distinctions, saw them aligned on the same front in the practice of analogical procedures (Zambelli, 2021), expressing and anticipating in architectural facts the contemporary concept of urban acupuncture (M. Casagrande, 2010). An emerging notion used to designate the episodic and local character, with urban and ter-ritorial impact of new interventions that aim to oppose the contemporary practice of grand self-celebratory authorial interventions intentionally in dissonance with the context. These inter-ventions are promoted both by liberal and capitalist culture as well as by socialist-inspired culture (Sudjic, 2012). Lastly, it highlights the multiscale nature present in urban morphology studies as well as, at least in intentions, in urban acupuncture projects. Evident in the correlation between expression and content, form and emerging significance, wherein (Foucault, 1969) each change in form, to-gether with a change in scale, corresponds to a new morphological adaptation and consequently a redefinition of rules and 'urban grammar', usable in urban projects with territorial significance. Today, enriched by new digital monitoring tools that graphically render real-time socio-economic flows and dynamics, confirming those insights and syntheses that morphology and typology had suggested in forms that we could now define as acupuncture urban interventions.

Background The distinctive feature of liver fibrosis is the progressive replacement of healthy hepatic cells by extracellular matrix protein, which is abundant in collagen I and III with impaired matrix remodelling. The activation of myofibroblastic cells enhances a fibrogenic response of complex interactions of hepatic stellate cells, fibroblasts and inflammatory cells to produce excessive deposition of extracellular protein matrix. This process is activated by multiple fibrogenic mediators and cytokines such as TNF-α and IL-1ß with decrease in the anti-fibrogenic factor NF-ҡß. Mesenchymal stem cells (MSCs) represent a promising therapy for liver fibrosis giving more advanced regenerative influence when cultured with extrinsic or intrinsic proliferative factors, cytokines, anti-oxidants, growth factors, and hormones such as Melatonin (MT). However, previous studies showed conflicted findings concerning the therapeutic effects of adipose (AD) and bone marrow (BM)-MSCs, therefore, the present work aimed to accomplish a comparative and comprehensive study investigating the impact of MT pre-treatment on the immunomodulatory, anti-inflammatory and anti-apoptotic effects of AD and BM-MSCs and to critically analyse whether MT pre-treated both AD-MSCs and BM-MSCs reveal equal or different therapeutic and regenerative potentials in CCl4-injured liver of experimental rat models. Materials and methods Six groups of experimental rats were used with ten rats in each group; group I (control group), group II (CCl4-treated group), group III (CCl4 and BM-MSCs treated group), group IV (CCl4 and MT pre-treated BM-MSCs group), group V (CCl4 and AD-MSCs treated group) and group VI (CCl4 and MT pre-treated AD-MSCs group). Liver function tests, gene expression of inflammatory, fibrogenic, apoptotic and proliferative factors were analysed. Histological and immunohistochemical changes were assessed. Results The present study compared the capability of AD and BM-MSCs, with and without the pre-treatment of MT, to reduce hepatic fibrosis. Both types of MSCs improved hepatocyte function by reducing the serum levels of (ALT), aspartate aminotransferase (AST), alkaline phosphatase (AKP), and total bilirubin (TBIL). In addition, the changes in the hepatocellular architecture including the hepatocytes, liver sinusoids, central veins, portal veins, biliary ducts and hepatic arteries showed decrease hepatocyte injury and cholestasis with reduction of inflammation, apoptosis and necrosis of the hepatic cells together with inhibition of liver tissue fibrosis. These results were augmented by the analysis of the expression of the anti-inflammatory cytokines TNFα and IL-1β, the anti-fibrogenic factor NF-ҡß, the apoptotic factor caspase-3 and the proliferative indicators antigen Ki-67 and proliferating cell nuclear antigen (PCNA). These findings were found statistically significant, with the best results in the rats received AD-MSCs pre-treated with MT, denoting better regenerative and therapeutic effects. Conclusion AD-MSCs, pre-treated with MT, are superior to BM-MSCs due to their better efficacy in improving hepatic fibrosis and promoting the therapeutic and regenerative capability of liver tissue. They represent a very significant tool for future stem cell use in tissue regeneration strategy for the treatment of liver diseases.

In Italy, the number of buildings that have fallen into disuse is huge; also the normative and regulative framework promoting public portfolio’s re-use and revitalisation is still unclear and blurred. Nevertheless, in the last ten years, these buildings have become fertile ground for innovation and creative experiences by civic actors. The rise of this new category of civic actors plays an important role, both from the institutional side and the kind of initiatives and practices they process. They act in different manners but according to an in-depth analysis they share similar patterns of behaviour. What has emerged is that, although the different contexts, where they operate, the institutional performances might be successful if only certain kinds of conditions are taken into consideration. Based on some basic features, civic actors and their dynamics with public administrations are analysed to understand conditions that allow revitalisation processes to be successful in unused public buildings.

Such activities have also put some effects on environment in both positive and negative way. In this study only positive impacts of COVID-19 on Environment have been highlighted that played role in regeneration of natural systems i.e., reduced air emissions, cleaner air, beaches &water bodies, reduced noise levels, biodiversity rehabilitation etc.

Space shuttle has been a hall mark of American space program since its inception. Despite its temporary shutdown few years ago, with the recent interest in space exploration which includes revitalizing human outpost in microgravity and transportation required to build it, realize other experiments (e.g. in space telescopes, in space manufacturing) and interplanetary voyages, it has regained attention. Its superior design, manufacturing, materials, performance, durability, and efficiency place it among the best, in fact, the only effort by mankind to build a reusable craft horizontally, launch vertically like a rocket and fly back like a plane. Various requirements emerge during its design (thermal, fluid, acoustics, vibration and structural) and design of its main engine (Aerojet Rocketdyne RS 25) which requires considerable attention, heat transfer being most important. This facilitated and necessitated use of various types of heat exchangers such as single coil, heat pipe, built in internal heat exchanger (IHEX), external heat exchanger (EHEX), condensing heat exchanger (CHEX), Interface heat exchangers (InHEX), regenerative heat exchanger (RHEX) and compact heat exchangers (CoHEX), change, manipulate and optimize their configurations in piping and instrumentation diagrams (PIDs). In this short narrative, an effort has been made to summarize them, and their developments over time with a focus on the application, design, manufacturing, materials, and performance (in service and final operation).

Public participation in the decision-making process in Urban Interventions is the key to the success of the project for improving the quality of life of its citizens. The citizen has the democratic right to express his needs and aspiration; he is the final user who experiences the outcomes of the policy decisions. Non involvement of the citizens in the planning process can bring about the misinterpretation of the intention of political leadership and lead to opposition and protest. The inadequate understanding of citizens of the urban context makes public participation ineffective. In this context, the decision-makers are often faced with the challenges of the level of confidence of the citizens about their ideas and responses being incorporated in the project and the confidence of the citizens in the local urban authority in its ability to carry out the project. However, the decision-makers base their decision on the assumption that the citizens have a general understanding of the urban issues. This research work investigates the basis of this assumption. 1. Do the citizens have confidence that the local urban authority considers their choices and responses in the course of decision making 2. Do the citizens have the confidence that the local urban authority can undertake the Urban Regeneration project 3. Whether in the decision-making process of urban regeneration intervention, citizen's responses are backed by a general understanding of urban issues. The case study taken up is of Hassan city. Five areas of crucial importance have been selected based on the development plan report of the city. The integrated approach aims to find the most appropriate area for proposing the Urban Regeneration project. The framework adopted includes 1. Questionnaire survey: to collect citizens’ responses 2. Analysis of variance (ANNOVA) for analysis of the data collected.

Various techniques have been used to “clean-up” industrial phosphoric acid: precipitation, flotation, activated charcoal or clay treatment. To address membrane processes potential in phosphoric acid clarification process, this study explores the advantage of membrane techniques as new route for phosphoric acid clarification in an eco-efficient way through the use of “regenerated spent membrane”. Regeneration of the spent membranes was performed on of 0.15 m2 active area regeneration. These membrane samples were used to study the phosphoric acid clarification at a laboratory scale. They were immersed in an oxidizer for at most seven days. The samples were characterized systematically before immersion in an oxidant media. In this study, the potential to regenerate spent membranes and application of this media to clarify the 29% P2O5 phosphoric acid was demonstrated. This study shows, by tests that the reverse osmosis (RO) membranes achieve an abatement of 70% and 65% for solids and organic materials, respectively. These positive results will pave the way for implementing these membranes phosphoric acid treatment process. Moreover, besides being economically advantageous, the use of the spent membrane is likely an environmentally friendly route (no waste, no organic solvent and effluent to be regenerated later on).

The aim of this article is to evaluate and compare, retrospectively, the efficacy of two regenerative periodontal procedures in patients suffering from aggressive periodontitis (AgP). Twenty-eight patients were diagnosed with AgP, suffering from multiple intra-bony defects (IBD); that were treated by one of two regenerative modalities of periodontal therapy randomly assigned to each patient: a. guided tissue regeneration (GTR) or b. an application of enamel matrix derivatives (EMD) combined with DBX. The monitoring of the treated sites included recordings of probing depth (PD), clinical attachment level (CAL), and gingival recession. Pre-treatment and follow-up (up to 10 years from the surgery) recordings were analyzed statistically within and between groups. A significant reduction was shown at time on PD and CAL values, however, not between subject groups. CAL values decreased in all sites. At EMD group (44 sites) CAL gain was 1.92 mm (±1.68) from pre-treatment to follow-up (p<0.001) and at the GTR group (12 sites) CAL gain of 2.27 (±1.82) mm. In conclusion, 1-10 years observations have shown that surgical treatment of AgP patients by either GTR or by application of EMD/DBX results in similar successful clinical results.

Heart disease, including myocardial infarction (MI), remains a leading cause of morbidity and mortality worldwide, necessitating the development of more effective regenerative therapies. Direct reprogramming of cardiomyocyte-like cells from resident fibroblasts offers a promising avenue for myocardial regeneration, but its efficiency and consistency in generating functional cardiomyocytes remain limited. Alternatively, reprogramming induced cardiac progenitor cells (iCPCs) could generate essential cardiac lineages, but existing methods often involve complex procedures. These limitations underscore the need for advanced mechanistic insights and refined reprogramming strategies to improve reparative outcomes in the heart. Partial cellular fate transitions, while still a relatively less well-defined area and primarily explored in longevity and neurobiology, hold remarkable promise for cardiac repair. It enables the re-programming or rejuvenation of resident cardiac cells into a stem or progenitor-like state with enhanced cardiogenic potential, generating the reparative lineages necessary for comprehen-sive myocardial recovery while reducing safety risks. As an emerging strategy, partial cellular fate transitions play a pivotal role in reversing myocardial infarction damage and offer sub-stantial potential for therapeutic innovation. This review will summarize current advances in these areas, including recent findings involving two transcription factors that critically regulate stemness and cardiogenesis. It will also explore considerations for further refining these ap-proaches to enhance their therapeutic potential and safety.

Cyclin-dependent kinase 1 (Cdk1) is a master regulator of the G2-M transition between DNA replication and cell division. This study investigates the regulation of cardiomyocyte (CM) proliferation during the early neonatal period and following ischemic injury in adult mice. We analyzed cell cycle dynamics with the assessment of DNA synthesis, and cytokinesis in murine hearts during the first 15 days after birth. A distinct proliferative block was observed at 1 day, followed by a second wave of DNA synthesis at 4 days, leading to CM binucleation (CMBN) by day 5. Genome-wide mRNA profiling revealed differential expression of cell cycle regulatory genes during this period, with a downregulation of factors involved in cell division and mitosis. The loss of Cdk1 impaired CMBN but extended the neonatal CM proliferation window until day 10 post-birth. In adult hearts, cardiac-specific ablation of Cdk1 triggered CM proliferation post-myocardial infarction (MI) in specific zones, driven by the activation of EGFR1 signaling and suppression of the anti-proliferative p38 and p53 signalling. This was accompanied by restoration of fractional shortening, mitochondrial function, and decreased reactive oxygen species. Additionally, cardiac hypertrophy was mitigated, and survival rates post-MI were increased in Cdk1 knockout mice. These findings reveal a novel role of Cdk1 in regulating cell cycle exit and re-entry in differentiated CM and offer insights into potential strategies for cardiac repair.

Extremely low-frequency electromagnetic fields (ELF-EMF) are ubiquitous in industrial-11 ized environments due to the continuous use of electrical devices. Our previous studies demon-12 strated ELF-EMF affects muscle cells by modulating oxidative stress and enhancing myo-genesis. 13 This pilot study investigated these effects on skeletal muscles of sedentary adult mice, assessing 14 physiological responses to ELF-EMF exposure and potential modulation by antioxidant supplemen-15 tation. Male C57BL/6 mice were exposed to ELF-EMF (0.1 or 1.0 mT) 1 hour/day up to 5 weeks, and 16 fed a standard diet without or with N-acetyl-cysteine (NAC). Results showed transient increases in 17 muscle strength (after 2-weeks’ exposure at 1.0 mT), potentially linked to muscle fiber recruitment 18 and activation, revealed by higher PAX7 and MyH expression levels. After ELF-EMF exposure, ox-19 idative status assessment revealed transient increases in expression levels of SOD1 and catalase en-20 zymes, in total antioxidant capacity and in protein carbonyl levels, marker of oxidative damage. 21 These effects were partially reduced by NAC. In conclusion, ELF-EMF exposure affects skeletal 22 muscle physiology and NAC supplementation partially mitigates these effects, highlighting the 23 complex interactions between ELF-EMF and antioxidant pathways in vivo. Further investigations 24 on ELF-EMF as a therapeutic modality for muscle health are necessary.

In the ongoing shift towards electric vehicles (EVs) primarily utilizing lithium-ion battery technology, a significant population of hybrid electric vehicles (HEVs) remains operational, reliant on established NiMH battery systems. Over the last twenty years, these HEVs have generated a substantial quantity of NiMH batteries that are either inoperable, experiencing performance degradation, or approaching the end of their service life. This results in a twofold challenge: a growing volume of environmentally hazardous waste due to the difficulty of NiMH battery reclamation, and escalating maintenance costs for HEV owners necessitated by replacement battery purchases. Patent WO2015092107A1, published in 2015, proposed a "Method for regenerating NiMh batteries." This method claims the ability to restore NiMH batteries to their original functionality. However, a comprehensive review of relevant scientific literature fails to identify any empirical evidence supporting the efficacy of the regeneration technique detailed in the patent. Within this context, this paper presents a detailed analysis and evaluation of the feasibility of the aforementioned patented method. To achieve this objective, the authors propose a prototype constructed utilizing NiMH battery cells sourced from a Toyota Prius. The experimental results obtained from this prototype demonstrate the potential for battery regeneration using the proposed method. These findings offer a promising solution for mitigating the challenges associated with NiMH battery disposal and maintenance within the HEV domain.

The study evaluated the regenerative responses of the lacrimal functional unit (LFU) after Lacrimal Gland (LG) ablation. The LG of Wistar rats were submitted to G1) partial LG ablation, G2) partial ablation and transplantation of an allogeneic LG, or G3) total LG ablation, (n=7-10/group). The eye wipe test, slit-lamp image, tear flow, and histology were evaluated. RT-PCR analyzed inflammatory and proliferation mediators. The findings were compared to naïve controls after 1 and 2 months (M1 and M2). G3 presented increased corneal sensitivity, and the 03 groups have shown corneal neovascularization. Histology revealed changes in LG, and corneal inflammation. In the LG, there was an increase in MMP-9 mRNA of G1 and G2 at M1 and M2 and RUNX-1 at M1 and M2 in G1 and RUNX-3 mRNA at M1 in G1, and in G2 at M2. TNF-α mRNA rose in the corneas of G1 and G2 at M2. There was an increase in the IL-1β mRNA in the Trigeminal Ganglion of G1 at M1. Without changes in tear flow or evidence of LG regeneration, LG ablation or grafting are unreliable models for dry eye or LG repair in rats. The surgical manipulation extended inflammation to the LFU.

Abstract: Nowadays, there is an increasing interest in the development of novel bioresorbable membranes for Guided Bone Regeneration (GBR). Hydroxyapatite of different resources in com-bination with chitosan has been used successfully. In this work, armored catfish (Pterygoplich-thys spp) bone as a source of hydroxyapatite is proposed; it was isolated through bone calcina-tion at a temperature of 1000 °C. The hydroxyapatite obtained was analyzed for its chemical composition by means of Energy Dispersive Spectroscopy (EDS), its functional groups by Fouri-er Transform Infrared Spectroscopy (FT-IR), its crystallographic structures were analyzed by X-ray diffraction (DRX) and Scanning electron microscopy (SEM) was used to observe the mor-phology of the particle. The composed membrane presents a major roughness morphology sur-face than the pristine chitosan membrane. The hydroxyapatite particles added to the polymeric matrix increased the mechanical tensile properties of the membrane. These findings confirm that armored catfish bones are a viable, economic, and environmentally friendly source of hydroxy-apatite, which combined with chitosan, is a suitable alternative to develop biocompatible GBR membranes after it is combined with chitosan.

Herein, activated red mud particles are used as an adsorbent for adsorbing phosphorus. Different concentrations of HCl and deionized water are used for desorption tests, and the optimal desorption mechanism is investigated. The Langmuir isothermal model and pseudo-second-order kinetic linear model produce consistent results: the desorption is dominated by the surface desorption of the monomolecular layer, and chemical desorption limits the rate of surface desorption. The desorption thermodynamics indicates that the desorption of phosphorus by the desorbent is spontaneous and high temperature promotes desorption. The particle diffusion model demonstrates that the removal of phosphorus in the form of precipitation from the surface of an activated hematite particle adsorbent occurs primarily via a chemical reaction, and an analysis of the microscopic morphology indicates that the desorption process is primarily accompanied by the dissolution of the metal element Ca, followed by Al and Fe. The desorbent reacts with the active elements in red mud, and the vibration of the [SiO4]2− group calcite or aragonite group of calcium–iron garnet in the functional group disappears. The intensity of the wave peaks of the PO43− groups decreases considerably. Thus, desorption primarily involves the chemical desorption of the monomolecular layer.

The development of biomaterial as substitution to connective tissue graft for treating gingival recession is growing nowadays. Hydrogels received attention as potential scaffold to be used in gingival tissue engineering since their capability to mimic soft tissue characteristic as they can hold amount of water. Gelatin is a natural polymer that widely used in hydrogel preparation with good biological properties, but it has poor physical stability for the used as scaffold. Chemical crosslinking strategy using EDC/NHS was performed in this study to overcome its limitation. One uncrosslinked gelatin and two crosslinked gelatin hydrogels different concentration of EDC/NHS were prepared in this study. This study investigated the difference of physical characterization from the prepared gelatin hydrogels. Our findings suggest the crosslinking strategy using EDC/NHS increase physical properties of the gelatin hydrogels without interfering the chemical structure of the gelatin. The higher crosslinker concentration will result in better morphology and more resilient to biodegradation. As a result, the crosslinked gelatin hydrogel with gelatin:EDC:NHS mass ratio of 12:1:1 met the physical characteristic for future use as scaffold for gingival tissue regeneration.

Proximal tubular epithelial cells (PTEC) are constantly exposed to potentially toxic metabolites and xenobiotics. The regenerative potential of the kidney enables the replacement of damaged cells either via the differentiation of stem cells or the regain of proliferative properties of the PTEC. Nevertheless, it is known that renal function declines, suggesting that the deteriorated cells are not replaced by fully functional cells. To understand the possible causes of this loss of kidney cell function, it would be beneficial to understand the role of toxins during the regeneration process. Therefore, we investigated the sensitivity and function of human induced pluripotent stem cells (hiPSC), hiPSC differentiating, and hiPSC differentiated into proximal tubular epithelial-like cells (PTELC) to known nephrotoxins. hiPSC were differentiated into PTELC, exhibiting a similar morphology to PTEC, expressing prototypical PTEC markers and performing albumin endocytosis. When treated with two nephrotoxins, hiPSC and differentiating hiPSC were more sensitive to cisplatin than differentiated PTELC, whereas all stages were equally sensitive to cyclosporin A. Both toxins also had an inhibitory effect on albumin uptake. Our results suggest a high sensitivity of differentiating cells towards toxins, which could have an unfavorable effect on regenerative processes. To study this, our model of hiPSC differentiating into PTELC appears suitable.

Normally, people lose between 50 and 100 hairs a day and generate new ones from the stem cells of hair follicles, but in people suffering from baldness, the condition called alopecia or androgenetic alopecia, the stem cells remain inactive and are unable to regenerate new hair. The stages of hair growth have been studied to better understand how hair grows and what might be done to prevent or treat premature hair loss. Anagen is referred to as the active phase of hair growth in which the hair shaft is produced by the hair follicle, whereas during catagen and telogen phases the lower part of hair follicle regresses until its growth stops. Although 9% of hair follicles remain in telogen at any one time, a variety of factors including growth factors and cytokines promote the transition from telogen to anagen and the subsequent stimulation of hair growth. In this work, we discuss a new possible treatment, AMPLEX PLUS that could help significantly slow hair loss and encourage new hair growth.

In regenerated oak forests, weeds are present throughout the year, the ones appearing in early spring representing a major problem. Hence, the aim of this study was to examine herbicides for early spring broadleaved weed control in the regenerated oak forests while the seedlings are in a dormant stage. During 2019 and 2020 two experiments were set up in regenerated pedunculate oak forests with 2-yar and 3-year-old seedlings and two herbicides were applied in two doses: fluroxypyr at dose 360 g a.i. ha-1 and 540 g a.i. ha-1 and clopyralid at dose 100 g a.i. ha-1 and 120 g a.i. ha-1. The results from the two-year investigations showed that fluroxypyr and clopyralid significantly reduced early spring broadleaved weeds in the regenerated pedunculate oak forests but both doses of fluroxypyr provided greater control of presented weeds than the applied doses of clopyralid. The manual weeding reduced broadleaved weeds in experiments, but that method did not have a long-term effect on the reduction of weeds. Applied doses of herbicides fluroxypyr and clopyralid did not cause phytotoxicity symptoms in dormant oak seedlings. All investigated treatments significantly reduced fresh broadleaved weed biomass compared to the control. Generally, it can be concluded that fluroxypyr and clopyralid can be successfully used for control of many early spring broadleaved weeds in the regenerated pedunculate oak forests, but oak seedlings must be in the dormant stage.

Tropical forests provide habitats for diverse flora and fauna. In addition, they are essential in regulating the climate while at the same time millions of people depend on them for their livelihoods. As seen by rising political commitment and worldwide promises to reduce rates of deforestation and boost restoration of degraded forest ecosystems, forests are becoming more widely acknowledged for their role as a nature-based solution to many sustainable development concerns. Understanding tropical forest dynamics and their conservation have so become more important. This study analyzed, the forest stand structure, tree species composition, abundance and diversity and the regeneration status of Londiani Forest. Londiani Forest is divided into three blocks, namely Kedowa, Chebewor and Londiani and from each of these blocks belt transects 25 m wide and 1 km long were established 100 m from the edge of the forest to reduce edge effects. At every 200 m along the transect, 25 m × 25 m quadrats were set up in which, an inventory of all tree species was done. For tree species that could not be identified in the field, the local name was provided by para-taxonomists who participated in the data collection and the species name was later identified with the help of a taxonomist or a manual of woody tree species of Londiani Forest. The diameter at breast height (DBH) was measured at 1.3 m from the ground using a diameter tape. Tree height was measured using a Suunto angular clinometer. A nested quadrat 5 m x 5 m quadrat within the 25 m x 25 m quadrat was used to sample saplings while a 1 m x 1 m quadrat was used for sampling seedlings. Regeneration status was assessed using the number of seedlings and saplings. Stumps of trees cut were counted including identifying tree species from which the tree stump was derived. Data from the quadrats were entered into Microsoft Excel. Total stem density, species density, basal area, species basal area, relative density and species diversity were determined. A total of 1,308 individual trees belonging to 34 different species from 24 families were recorded. Kedowa had the highest (27) species richness followed by Chebewor (19) then Londiani (14). There was a statistically significant difference in species richness among the three forest blocks (p<0.05). There were no statistically significant differences in the mean DBH (F=0.560; p=0.729) and height class distribution (F= 0.821, p=.558) among the three forest blocks. There was a statistically significant difference in stem density (F=12.22; p=0.005) and woody species diversity (F=0.32; p=0.001) within the three forests blocks. There was no statistically significant difference in the basal area distribution within the three forest blocks p>0.005. The similarity index ranged from 0.34 to 0.47. In all the forest blocks, there was evidence of seedlings and saplings indicating that there was regeneration. Results clearly show that while the Londiani Forest shows variation in characteristics, there is clear evidence that it is regenerating.

Physical activity is well-established as an important protective factor against degenerative conditions and a promoter of tissue growth and renewal. The discovery of FNDC5 as the precursor of irisin in 2012 sparked significant interest in its potential as a diagnostic biomarker and a therapeutic agent for various diseases. Clinical studies have examined the correlation between plasma irisin levels and pathological conditions using a range of assays, but the lack of reliable measurements for endogenous irisin has led to uncertainty about its prognostic/diagnostic potential as an exercise surrogate. Animal and tissue-engineering models have shown the protective effects of irisin treatment in reversing functional impairment and potentially permanent damage, but dosage ambiguities remain unresolved. This review provides a comprehensive examination of the clinical and basic studies of irisin in the context of degenerative conditions and explores its potential as a therapeutic approach in the physiological processes involved in tissue repair/regeneration.

Growing cells is the typical mode of operation in many aspects of biotechnology and metabolic engineering. This comes about due to cell growth processes creating a driving force that pull metabolic flux along different metabolic pathways, that indirectly help move substrate to product. But, there is an alternative mode of operation that uses resting (non-growing) cells to achieve similar or even higher productivities. In general, resting cells are provided with carbon substrates for biocatalytic reactions but starved of nitrogen or phosphorus. Such resting cells have been usefully employed in many forms of biocatalysis and biotransformation, with or without cofactor regeneration. However, much remains unknown about the transcriptome and metabolome of resting cells in biotransformation settings. This short writeup provides the backdrop of resting cells in biocatalysis, documents their use in biotransformation with application examples, and identifies research gaps that could be filled with contemporary RNA-seq and mass spectrometry proteomics technology. Overall, utility of resting cells in biocatalysis and the extant knowledge gap in their fundamental physiology are highlighted in this resource.

Nervous systems and brains’ computational abilities are an evolutionary innovation, specializing and speed-optimizing ancient biophysical dynamics. Bioelectric signaling originated in cells’ communication with the outside world and with each other, in order to cooperate toward adaptive construction and repair of multicellular bodies. Here we review the emerging field of developmental bioelectricity, which links the field of basal cognition to state-of-the-art questions in regenerative medicine, synthetic bioengineering, cognitive science, and even machine learning and artificial intelligence. One of the predictions of this view is that regeneration and regulative development are able to restore correct large-scale anatomies from diverse starting states because, like the brain, they exploit bioelectric encoding of distributed goal states - in this case, pattern memories. Based on this idea, we propose a new interpretation of recent stochastic regenerative phenotypes in planaria, by appealing to computational models of memory representation and processing in the brain. Moreover, we discuss novel findings showing that bioelectric changes induced in planaria can be stored in tissue for over a week, thus revealing that somatic bioelectric circuits in vivo can implement a long-term, re-writable memory medium. A consideration of the mechanisms, evolution, and functionality of basal cognition makes novel predictions and provides an integrative perspective on the evolution, physiology, and biomedicine of information processing in vivo.

Background: Recently, the management of musculoskeletal disorders with the patients' own stem cells, isolated from the walls of small blood vessels, which can be found in great numbers in the adipose tissue, has received considerable attention. On the other hand, there are still misconceptions about these adipose-derived regenerative cells (ADRCs) that contain vascular-associated pluripotent stem cells (vaPS cells) in regenerative medicine. Methods: Based on our previous publications on this topic, we have developed a concept to describe the significance of the ADRCs/vaPS cells in the field of orthobiologics as briefly as possible and at the same time as precisely as possible. Results: The ADRCs/vaPS cells belong to the group of orthobiologics that are based on autologous cells. Because the latter can both stimulate a patient’s body's localized self-healing power and provide new cells that can integrate into the host tissue during the healing response when the localized self-healing power is exhausted, this group of orthobiologics appears more advantageous than cell-free orthobiologics and orthobiologics that are based on allogeneic cells. Within the group of orthobiologics that are based on autologous cells, enzymatically isolated, uncultured ADRCs/vaPS cells have several advantages over non-enzymatically isolated cells/microfragmented fat as well as over uncultured bone marrow aspirate concentrate and cultured cells (adipose-derived stem cells, bone marrow-derived mesenchymal stem cells). Conclusions: The use of ADRCs/vaPS cells can be seamlessly integrated into modern orthopedic treatment concepts, which can be understood as the optimization of a process which - albeit less efficiently - also takes place physiologically. Accordingly, this new safe and effective type of treatment is attractive in terms of holistic thinking and personalized medicine.

When profit-driven renewal is difficult to implement, many historic districts are “frozen protected” in China. In the social context of “building cities for the people” in recent years, the public value-oriented renewal of urban historic areas plays an important role in the transformation of cities. This study investigates the strategies of historic district regeneration in China by integrating spatial form, urban governance, and public value. We propose a novel framework to analyze the regeneration of historic districts, using Taiyuan Bell Tower Street as a case study. The framework distinguishes between two dimensions: spatial and economic-social. In the spatial dimension, we focus on the transformation of spatial form to regenerate public value, emphasizing the attribute of "publicness" as central to urban regeneration efforts. This involves a tiered approach of excavating and upgrading historic districts at macro, meso, and micro levels. In the economic-social dimension, we explore urban governance to strengthen public value, involving government, market, and societal actors in a collaborative renewal process. The analysis highlights that a public value-oriented approach to heritage regeneration offers greater sustainability and could serve as a model for other cities seeking similar outcomes.

The widespread use of the herbicide glyphosate has resulted in the global presence of its degra-dation product, aminomethylphosphonic acid (AMPA), in the global environment. AMPA is commonly found in water bodies, including freshwater systems. We investigated the effects of chronic and acute AMPA exposure on the survivorship, regenerative abilities, and locomotion of the brown planarian (Girardia tigrina), a water-dwelling flatworm commonly found in freshwa-ter ecosystems. We exposed planarians to two concentrations of AMPA, consistent with the high and low ranges of concentrations observed in US water systems. We measured endpoints includ-ing survivorship, regenerative abilities of head and tail segments, eyespot regeneration, and swimming speed. Compared to the control group, we found that planarians chronically exposed to AMPA exhibited (1) modestly reduced survivorship while regenerating and (2) slower regen-eration from the tail segment. No significant differences were observed in intact body growth during chronic exposure. Our findings highlight the potential ecological impacts of AMPA con-tamination on planarian populations. Understanding the effects of AMPA exposure on planarians contributes to our understanding of the ecological consequences of glyphosate use in freshwater ecosystems.

Nanotechnology has become an increasingly promising medical field, particularly scar removal. Scar removal is a complex process involving regenerating damaged skin tissue, and nanotechnology presents unique solutions to this issue. One potential application of nanotechnology is using nanofibers as scaffolds to support the growth of new skin tissue. These fibres can also be loaded with drugs or growth factors to promote tissue regeneration and reduce scarring. Another potential application is nanocarriers for drug delivery to specific body areas, which can promote tissue regeneration and reduce scarring.Additionally, nanotechnology has been utilized to create new materials for skin regeneration, such as "nano skin" that mimics the structure of natural skin. Nanoprobes have also been developed for the detection of scar tissue and the monitoring of its progression. These potential nanotechnology applications offer exciting possibilities for the future of scar removal and skin repair. With further research and development, nanotechnology has the potential to revolutionize scar removal and provide more effective solutions for tissue regeneration.

De novo regeneration of Cannabis sativa L. (cannabis) using tissue culture techniques remains unreliable and infrequent. Conventional methods for the regeneration and transformation of cannabis have not achieved the reliability and replicability needed to be integrated into research and breeding programs. Protoplast systems are effective for gene expression studies, transformation and genome editing technologies, and opens the possibility of somatic hybridization to create interspecific hybrids. To date, leaf-derived protoplast have been isolated for transient gene expression studies, but attempts at protoplast-to-plant regeneration have not been reported. The present study aims to test the efficacy of using a callus culture system, as an abundant source for protoplast isolation and lays the groundwork for a protoplast-to-plant regeneration system. Using hypocotyl-derived callus cultures, which are known to have relatively greater regenerative potential, the efficacy of protoplast isolation and initial cell division were assessed. In this study, the effect of 2-aminoindane-2-phosphonic acid (AIP) in callus culture media and the effect of subculture frequency on protoplast yield were assessed. This study found that inclusion of AIP at 1 mM resulted in a 334% increase in protoplast yield compared with AIP-free medium, representing the first known use of AIP in cannabis tissue culture. Inclusion of AIP led to a 28% decrease in total soluble phenolics and 52% decrease in tissue browning compared to the control medium. Lastly, a two-phase culture system for protoplast regeneration was tested. At a concentration of 2.0×10^5 protoplasts per mL, division was observed, providing the first know report of cell division from cannabis protoplasts and setting the stage for future development of a protoplast-to-plant regeneration system.

Exosomes are membrane-bound, biologically active nanovesicles of size 30-200 nm produced by many cell types, such as both mammalian and plant cells. They are wrapped in a phospholipid bilayer and play a significant role in intercellular communications. The ease in their isolation and the ability of plant-derived exosome-like nanovesicles (PDEVs) to efficiently deliver bioactive constituents into mammalian cells have made them popular in contemporary research. PDEVs share many characteristics with mammalian EVs (MEVs), including shape, size, surface charge, and consist of bioactive molecules like lipids, proteins, nucleic acids, and tiny metabolites. However, the chemical composition profile of PDEVs and their biogenesis mechanism differ significantly from those of MEVs. They have been widely explored as potential therapeutic agents and are considered as good alternatives to act as a carrier for drug delivery. The present work elucidates the isolation of exosome-like-nanovesicles (henceforth exosomes) from the culture supernatants of an in vitro cultured callus tissue derived from a bone healing plant known for its osteogenic activity, i.e., Cissus quadrangularis. The physical and biological properties of exosomes were successfully studied using different characterization techniques. To assess their therapeutic potential, we studied the internalisation of calcein-AM labeled exosomes by human derived mesenchymal stromal cells (hMSCs). Additionally, we evaluated the potential of exosomes in the migration of cells in a cell scratch assay with hMSCs and their effect on amelioration of oxidative stress was investigated on preosteoblast MC3T3-E1 cells that were pre-treated with these exosomes. Furthermore, we investigated their proliferation and differentiation to osteoblasts like cells with the help of resazurin assay and alkaline phosphatase assay (ALP). The obtained results provide a primary justification for the use of Cissus quadrangularis-derived exosomes as a nanocarrier for drug molecules for various therapeutic bone applications.

Trace metals are essential elements that play key roles in a number of biochemical processes governing human visual physiology in health and disease. Several trace metals, such as zinc, have been shown to play important roles in the visual phototransduction process. In spite of this, there has been little research conducted on the direct effect of trace metal elements on the visual pho-toreceptor rhodopsin. In the current study, we have determined the effect of several metal ions, such as iron, copper, chromium, manganese, and nickel on the conformational stability of rho-dopsin. To this aim, we analyzed, by means of UV-visible and fluorescence spectroscopic methods, the effects of these trace elements on the thermal stability of dark rhodopsin, the stability of its active Metarhodopsin II conformation, and its chromophore regeneration. Our results show that copper prevented rhodopsin regeneration and slowed down the retinal release process after il-lumination. In turn, Fe3+, but not Fe2+, increased the thermal stability of the dark inactive con-formation of rhodopsin, whereas copper ions markedly decreased it. These findings stress the important role of trace metals in retinal physiology at the photoreceptor level, and may be useful for the development of novel therapeutic strategies to treat retinal disease.

This paper describes the influence of the regeneration treatments on 4-chloro-2-methyl-phenoxyacetic acid (MCPA)-loaded activated carbons (ACs) on adsorption-desorption cycles with two commercial ACs, Merck and Norit 1240 X ( Norit 1240 X was used in a granular and powder form). The ACs were saturated with MCPA and then submitted to four regeneration cycles, by washing with ethanol, NaOH solutions and washed with NaOH solutions followed by a thermal treatment. The ACs regenerated with ethanol showed a better performance concerning the successive adsorption-desorption cycles. On the first cycle, the MCPA desorbed, from washing with ethanol, from all ACs was higher than 99%. After a fourth adsorption-desorption cycle, washing regeneration combined with thermal treatment allowed excellent regeneration results. The amount of MCPA adsorbed on Norit 1240 X AC was higher than the amount adsorbed in the first cycle. MCPA adsorption kinetic data were analyzed by applying pseudo-first-order, pseudo-second-order, and Weber-Morris models. The pseudo-second-order fit better fit to the data and the Weber-Morris representation allows confirming that on Norit 1240 X, in a granular form, the pore diffusion was the limiting factor concerning the MCPA adsorption.

Hydrogels can provide instant relief to pain and facilitate the fast recovery of wounds. Currently the incorporation of medicinal herbs/plants in polymer matrix is being investigated due to their anti-bacterial and wound healing properties. Herein, we investigated the novel combination of chitosan (CS), chondroitin sulfate (CHI) and garlic (Gar) to synthesize hydrogels through freeze gelation process for faster wound healing and resistance to microbial growth at the wound surface. The synthesized hydrogels were characterized via Fourier transform infrared spectroscopy (FTIR), which confirmed the presence of relevant functional groups. The scanning electron microscopy (SEM) images exhibited the porous structure of the hydrogels, which is useful for the sustained release of Gar from the hydrogels. The synthesized hydrogels showed significant inhibition zones against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). Furthermore, cell culture studies confirmed the cyto-compatibility of the synthesized hydrogels. Thus, the novel hydrogels presented in this study can offer antibacterial effect during wound healing and promote tissue regeneration.

There is a growing appreciation in the fields of Cell and Developmental Biology that cells collectively process information in time and space. While many powerful molecular tools exist to observe biophysical dynamics biologists must find ways to quantitatively understand these phenomena at the systems level. Here, we present a guide for application of well-established information theory metrics to biological datasets and explain these metrics using examples from cell, developmental and regenerative biology. We introduce a novel computational tool (CAIM) for simple, rigorous application of these metrics to timeseries datasets. Finally, we use CAIM to study calcium and cytoskeletal Actin information flow patterns between Xenopus laevis embryonic animal cap stem cells. The tools that we present here will enable biologists to apply information theory to develop systems level understanding of a diverse array of experimental systems.

Regeneration is usually regarded as a unique plant or some animal species process. In reality, regeneration is a ubiquitous process in all multicellular organisms. It ranges from response to wounding by healing the wounded tissue to whole body neoforming (remaking of the new body). In a larger context, regeneration is one facet of two reproduction schemes that dominate the evolution of life. Multicellular organisms can propagate their genes asexually or sexually. Here I present the view that the ability to regenerate tissue or whole-body regeneration is also determined by the sexual state of the multicellular organisms (from simple animals like hydra and planaria to plants and complex animals). The above idea is manifested here by showing evidence that many organisms, organs, or tissues show inhibited or diminished regeneration capacity when in reproductive status compared to the same organism organs or tissues in nonreproductive conditions or by exposure to sex hormones.

Cancer cachexia is a multifactorial syndrome that is identified by ongoing muscle atrophy, along with functional impairment, anorexia, weakness, fatigue, anemia, reduced tolerance to antitumor treatments. Thus, reducing the patients’ quality of life. Cachexia alone causes about 22-25% of cancer deaths. This review covers the symptoms, mediators, available treatment, and prospects of 3D bioprinting for cancer cachexia. Studies about cachexia have shown several factors that drive this disease – protein breakdown, inflammatory cytokines activation, and mitochondrial alteration. Even with proper nutrition, physical exercises, anti-inflammatory agents, chemotherapy, and grafting attempts, standard treatment has been unsuccessful for cachexia. But the use of 3D bioprinting shows much promise compared to conventional methods by attempting to fabricate 3D constructs mimicking the native muscle tissues. In this review, some 3D bioprinting techniques with their advantages and drawbacks, along with their achievements and challenges in in-vivo applications have been discussed. Constructs with neural integration or muscle-tendon units aim to repair muscle atrophy. But it is still difficult to properly bio-print these complex muscles. Although progress can be made by developing new bio-inks or 3D printers to fabricate high-resolution constructs. Using secondary data, this review study shows prospects of why 3D bioprinting can be a good alternate approach to fight cachexia.

Zebrafish is an excellent model for observing human genetic disorders. Hearing impairment is the most common genetic disorder including syndromic & non-syndromic hearing loss. Stem cell therapies are considered a new hope in case of hearing impairment. Stem cells are the master cells of the human body and having the capability to differentiate into any other form of cells in more than 200 types of cells (regeneration). This review article describes zebrafish as a biomedical model for stem cell research in hearing impairment, which revolutionized the biomedical arena to compete for the challenges.

Nanomedicine has allowed for emerging advances in imaging, diagnostics and therapeutics. Regenerative Medicine has taken advantage of a number of nanomaterials for reparation of diseased or damaged tissues in the nervous system involved in memory, cognition and movement. Electrical, thermal, mechanical and biocompatibility aspects of carbon-based nanomaterials (nanotubes, graphene, fullerenes and their derivatives) make them suitable candidates to drive nerve tissue repair and stimulation. This review article focuses on recent advances on the use of carbon nanotube (CNT)-based technologies on nerve tissue engineering; outlining how neurons interact with the nanomaterials interface for promoting neuronal differentiation, growth and network reconstruction for their possible use in therapies of neurodegenerative pathologies and spinal cord injuries.

A total of five protoclones were successfully cultured on PDA medium out of regenerated twenty two colonies of Termitomyces protoplast and further studied. Liquid MYG grown mycelial tissue is used for protoplast isolation by enzymatic digestionin a mixture containing Lysing enzyme 2% and Cellulase R10 2% in 0.6 M mannitol. The incubation conditions like temperature, shaking and time were standardized at 24ºC, 60 rpm and 10 hours, respectively for healthy protoplasts liberation. The purified protoplasts showed an average yield of 1.2 × 10⁷ cells/gm tissue with 31.60 ± 9.31% regeneration efficiency on specific medium and 77.12 ± 2.72% viability by FDA test. Four ISSR primers were used in this study resulting a total of 27 reproducible bands with mean value of 6.75. They showed similar banding pattern in all the lines with zero percent polymorphism ranged from 280 bp–2700 bp. The amplified rRNA-ITS gene showed ~600 bp size in gel and found a single restriction site for enzyme HaeIII in all the protoclones and parent with similar fragment size in all.

Previous study demonstrated the reconstituted type I collagen matrix extracted from rabbit tendons enabled to regenerate the TMJ disc in the rabbit. The aim of this study was to investigate changes in the extracellular matrix (ECM) and mechanisms of regeneration in TMJ disc. In 36 New Zealand rabbits that underwent a partial discectomy, discs were replaced with reconstituted collagen templates for 3 months. A histological analysis showed that moderate to severe degeneration appeared in partially discectomized joints without implantation. In contrast, discs that received the reconstituted collagen template regenerated, and returned to normal to protect the joint. Cells in the regenerative tissue expressed ECM, and fibers became regular and compact due to tissue remodeling over time. Reparative cells differentiated into chondroblasts, and showed highly dense pericellular fibers. The morphology and collagen composition of the disc and condyle in the 3-month experimental group were similar to those of normal tissues. In conclusion, the reconstituted collagen template facilitated the regeneration of surgically discectomized discs. Type I and type II collagens play a crucial role in the regeneration of articular discs.

Regeneration of lost tail is of great importance to lizards. Anolis carolinensis, a green lizard, is capable of regenerating its tail efficiently after autotomy. Hence, it is considered as a model organism in regeneration study. A. carolinensis shed its tail in order to distract the predator’s attention and thus makes a way to escape. Restoring of the amputated tail takes several days and the mechanism is currently clearly understood. Although save its life, tail regeneration is associated with the impairment of several vital functions in Anoles. In addition, various differences have been observed between original and regenerated tail in terms of mechanism and structure. To date, very little work has been conducted on tail autotomy and regeneration at molecular and genetic level. The genes responsible for regeneration in anoles are identified recently. These genes are evolutionarily conserved through all tetrapod vertebrates. They are, however, in a state of ‘switched-off’ in other vertebrates including humans. Consequently, a throughout study of these so called ‘switched-off’ genes may provide a way of restoring lost organs in human, and thus could revolutionize the modern medical science.

Multicellularity is one of the major evolutionary transitions, and its rise provided the ingredients for the emergence of a biosphere inhabited by complex organisms. Over the last decades, the potential for bioengineering multicellular systems has been instrumental in interrogating nature and exploring novel paths to regeneration and disease, as well as cognition and behaviour. Here, we provide a list of open problems that encapsulate many of the ongoing and future challenges in the field, and we suggest conceptual approaches that may facilitate progress.

Preconditioned mesenchymal stem cells (MSCs) represent an advanced method to overcome the challenges related to their survival rate, differentiation, and activity maintenance. L-carnitine has demonstrated antioxidant and anti-apoptotic effects on injured cardiac cells. However, limited research has been conducted to explore their combined effect on cardiac tissue injury. Therefore, the present study aimed to conduct a comparative and comprehensive study investigating the role of L-carnitine-pre-treatment on the immunomodulation of inflammation, apoptosis, and differentiation of MSCs and their impact on cardiac toxicity induced by Doxorubicin (DOX). Rats were divided into group I (control), group II (DOX), group III (DOX+MSCs), group IV (DOX+L-carnitine), group V (DOX+L-carnitine pre-treated MSCs). CK-MB, troponin I, MDA, and catalase levels were improved in the treated groups (III, IV, and V). The degeneration and necrosis of the cardiomyocytes were reduced in the treated groups. They regained their normal architecture in the L-carnitine pre-treated MSCs group. These findings were augmented by analyzing the immunomodulators; NF-ҡβ, TNFα, and IL-1β, the proliferative indicators Ki-67, and hsp90, the cardiac differentiation marker TH and the apoptotic regulators; caspase 3 and Bcl2. These results demonstrate the optimal regenerative and therapeutic effects of L-carnitine pre-treated MSCs representing an efficient tool for future stem cell therapy.

Objectives: This study aims to explore the use of murine models in peripheral nerve transection research, evaluating and synthesizing key methods for analyzing nerve regeneration to guide future research and clinical interventions. Methodology: A systematic review was conducted in February 2024, adhering to Cochrane and PRISMA 2020 guidelines, using Medline, Scielo, and Lilacs databases. The focus was on experimental studies of nerve regeneration in animal models post-transection. Only experimental clinical trials reporting nerve regeneration outcomes were included; studies lacking a comparator group or functional evaluation methods were excluded. Results: Out of 273 studies initially identified from MEDLINE, 19 were selected for detailed analysis. The average study included 32.5 subjects, with about 10.16 subjects per intervention subgroup. The predominant model was the sciatic nerve injury with a 10mm gap. The most common intervention involved unprocessed adipose-derived stem cells, utilized in 14 articles. Conclusions: The review underscores the significant potential of current methodologies in peripheral nerve regeneration, particularly highlighting the use of murine models and thorough evaluation techniques. These studies significantly contribute to our understanding of nerve regeneration processes and inform directions for future research.

This study aimed to evaluate the effectiveness of decellularized nerves for promoting nerve regeneration by transplanting them from the sciatic nerve of a rabbit to that of a rat. The Sciatic nerves were harvested from New Zealand White Rabbits (n=4). A 1-cm segment of each nerve was decellularized by Sondell’s protocol and preserved at –4℃ in Roswell Park Memorial Institute (RPMI) 1640 medium for one week. Subsequently, a defect of a similar size was created in the sciatic nerves of rats. The decellularized nerves treated with detergent were then transplanted into these defects in the rats. The study included the three following groups: Group 1 (n=4) was the decellularized xenogeneic nerve group; Group 2 (n=7) was the negative control group in which the defect remained un-repaired; and Group 3 (n=5) was composed of the positive control group where the nerve was immediately transplanted from one rat to another. The sciatic functional index (SFI) was considered to evaluate the motor function by comparing it with the normal side at 1, 2, 3, and 4 weeks, and 1, 2, and 3 months postoperatively. Furthermore, electrophysiological and immunohistochemical assessments were conducted at 4 months. Immunohistochemically, at 16 weeks postoperative, the experimental group did not show statistically significant differences compared to controls in terms of schwann cell comparison, yet exhibited favorable outcomes when compared to the positive control group. Additionally, analysis of motor function test confirmed recovery of latency and nerve conduction velocity to levels similar to those of the normal side. While the nerve allograft demonstrated superior nerve regeneration in motor function test at 3 months, markedly low values were observed in all groups. When reconstructing peripheral nerves using detergent-based decellularized xenogenic nerves, analysis of immuno-histochemistry and motor nerve conduction study reveals a promising degree of nerve regeneration. However, when compared with fresh allograft group at 3 months after operation, the sciatic functional index was lower. Significantly lower values observed across all groups compared to previous assessments indicate the need for further research into potentially influential alternative factors.Histologically, at 16 weeks postoperative, the experimentally treated group did not show statistically significant differences compared to the controls in terms of a Schwann cell comparison, but exhibited favorable outcomes when compared to the positive control group. Additionally, an analysis of the motor function testing confirmed recovery of the latency and nerve conduction velocity to levels similar to those of the normal side. Several types of guided bone regeneration materials are available with various properties and clinical indications. Among them, dentin-derived matrix from extracted teeth is an excellent alternative to traditional graft materials, such as allografts and xenografts. Different clinical preparation protocols exist: block form versus powder form, demineralized versus non-demineralized form. Out of these methods, demineralization protocols for dentin-derived matrix from extracted teeth have been noted for a wide range of successful clinical outcomes. The purpose of this animal study was to find out the most effective method of demineralizing dentin-derived matrix to result in the highest regenerative ability in the bone graft material in rabbit calvaria. The study conducted a comparative histomorphometric analysis of four groups in distinguished matrix conditions: a control group without graft placement, a non-demineralized dentin-derived matrix graft placement group, a 15-minute demineralized dentin-derived matrix graft placement group, and a 30-minute demineralized dentin-derived matrix graft placement group. Regenerative processes were evaluated three times at 2, 4, and 8 weeks post-graft placement. These groups of rabbits were evaluated randomly in a blinded setting to prevent bias. Among the four groups compared, the group with a 15-minute demineralized dentin-derived matrix graft showed the most efficient bone regeneration in the histomorphometric analysis. Based on this animal study, it was apparent that a 15-minute demineralization of dentin-derived matrix is the preferred preparation protocol to maximize the regenerative potential in the matrix.

Synthetic bone made of calcium phosphate is widely used for the clinical treatment of bone defects caused by trauma and bone tumors. Although synthetic bone is easy to use, its effects are only partial depending on the size and location of the bone defect, and there are many treatment options other than the use of synthetic bone. Various biodegradable polymers that are being considered for use as scaffolds for regenerative medicine are also being considered for use as synthetic bones; however, commercial synthetic bone products with performance comparable to that of calcium phosphate bioceramics have not yet been clinically applied. This review introduces the practice of bone regeneration therapy with synthetic bone and recent research on bone regeneration therapy with biodegradable polymers.

Abstract : Aims: Regeneration of myocardial tissue and improvement of myocardial activity after ischemia is one of the major fields of interest in cardiovascular research. We have developed a novel experimental rat model, and used it to examine the effect of mesenchymal stem cell transplantation (MSCT) on myocardial ischemia evaluated by single photon emission computed tomography (SPECT/CT) and immunohistochemistry. Methods and results: Forty adult female Wistar rats underwent open thoracotomy with (n=30) or without (n=10) surgical ligation of the left anterior descending coronary artery (LAD), to create myocardial ischemia, which was confirmed with ECG. Myocardial viability was evaluated via SPECT/CT at 7 days before as well as at 7 and 14 days post-surgery. At day 0, 15 animals received homologous stem cells injected at the ischemic myocardium area. SPECT/CT evaluation confirmed the reduction of functional myocardial cells of the left ventricle one week post-infarction. Regeneration of the ischemic myocardium fifteen days post infarction was recorded, only in animals submitted to stem cell transplantation. Findings were also confirmed by histology and immunohistochemical analysis, with significant higher expression of GATA4 and Nkx2,5. Conclusion: The positive effect of mesenchymal stem cells transplantation in the ischemic myocardium was recorded. The application of SPECT- CT qualitative and quantitative evaluation of myocardial function may lead to a new approach in experimentation with an anticipated significant impact in the ongoing cardiovascular laboratory research. In clinical perspective, MSCT may be beneficial when accompanied with myocardial revascularization procedures.

Preclinical evidence suggests that voltage gradients can act as a kind of top-down master regulator during embryogenesis and orchestrate downstream molecular-genetic pathways during organ regeneration or repair. Moreover, electrical stimulation shifts response to injury towards regeneration instead of healing or scarring. Cancer and embryogenesis not only share common phenotypical features, but also commonly upregulated molecular pathways. Ion channel activity is directly or indirectly linked to the pathogenesis of cancer hallmarks, while experimental and clinical studies suggest that their modulation may exert antitumor effects. A large recent clinical trial served as a proof-of-principle for the benefit of preoperative use of topical sodium channel blockade as a potential anticancer strategy against early human breast cancers. Apart from the obvious importance in perioperative medicine, it calls for the planning of further carefully designed clinical trials to expand on the concept of ion channels as tumor drivers.

Exosomes have shown promising potential as a therapeutic approach for wound healing. Nevertheless, the translation from experimental studies to commercially available treatments is still lacking. To assess the current state of research in this field, a systematic review was performed to examine the methodological heterogeneity among studies conducted over the past five years. Additionally, the review analyzed the suitability of animal models used and their relevance to human medicine. A PubMed search was performed for english-language, full-text available papers published from 2018 to June 2023, focusing on exosomes derived from mammalian sources and their application in wound healing, particularly those involving in vivo assays. Out of 531 results, 148 papers were selected for analysis. The findings revealed significant variability in terms of cell sources and types, biomaterials, and administration routes under investigation, indicating the need for further research in this field. Additionally, a comparative examination encompassing diverse cellular origins, types, administration pathways, or biomaterials is imperative. Furthermore, the predominance of rodent-based animal models raises concerns, as there have been limited advancements towards more complex in vivo models and scale-up assays. These constraints underscore the substantial efforts that remain necessary before attaining commercially viable and extensively applicable therapeutic approaches using exosomes.

The odontogenic cysts can lead to a bone destruction that can hamper the dental implant place-ment in the ideal 3D position. Different biomaterials and techniques that allow bone regeneration are described in the literature, each with its advantages and disadvantages. This clinical case with 18 months of follow-up shows the enucleation of an odontogenic cyst fol-lowing the guide bone regeneration with fence technique. After 6 months 2, dental implants were placed at the region of tooth 11 and 21, being rehabilitated nine weeks after. The fence technique allowed the recovery of the bone morphology and the placement of the dental implants in the 3D ideal position.

Insufficient oral ridge tissue presents a challenge in the treatment of dental implants. One method to enhance alveolar crest dimensions is guided bone regeneration (GBR); however, existing membranes have certain limitations. To address this issue, we aimed to compare the effectiveness of a resorbable bilayer membrane composed of poly(l-lactic acid) and poly(-caprolactone) （PLACL）with that of a collagen membrane（COL） in a rat GBR model. The rat calvaria was used as an experimental model by placing a plastic cylinder. Forty male Fisher rats underwent surgery, and micro-computed tomography and histomorphometric analyses were performed to assess bone regeneration. The results showed that bone regeneration was similar across all the groups. However, after 24 weeks, the PLACL membrane demonstrated significant resilience, occasional partial degradation, and intermittent air bubble formation. This extended preservation of the barrier effect has great potential to facilitate optimal bone regeneration. In conclusion, this study shows that the PLACL membrane is a promising alternative to GBR. By providing a durable barrier and supporting bone regeneration over an extended period, this resorbable bilayer membrane may address the limitations of the current membranes. Further studies and clinical trials are warranted to validate the efficacy and safety of this drug in humans.

Composite biomaterials that combine osteoconductive and osteoinductive properties are a promising approach for bone tissue engineering (BTE) since it allows osteogenesis stimulation while mimicking the extracellular matrix (ECM) morphology. In this context, the aim of the present research work was to produce polyvinylpyrrolidone (PVP) nanofibers containing mesoporous bioactive glass (MBG) 80S15 nanoparticles. These composite materials were produced by electrospinning technique. The design of experiments (DOE) was used to estimate the optimal electrospinning parameters to reduce average fiber diameter. The polymeric matrices were thermally crosslinked under different conditions, and the fiber’s morphology was studied using scanning electron microscopy (SEM). Evaluation of the mechanical properties of nanofibrous mats revealed a dependence on thermal crosslinking parameters and on the presence of MBG 80S15 particles inside the polymeric fibers. Degradation tests indicated that the presence of MBG led to a faster degradation of nanofibrous mats. On the other hand, MBG presence also leads to higher swelling capacity. The assessment of in vitro bioactivity in simulated body fluid (SBF) was performed in MBG pellets and PVP/MBG (1:1) composites to assess whether the bioactive properties of MBG 80S15 were kept when it was incorporated into PVP nanofibers. FTIR and XRD analysis along with SEM–EDS results indicated that a hydroxy-carbonate apatite (HCA) layer was formed on the surface of MBG pellets and nanofibrous webs after soaking in SBF over different time periods. In general, the materials revealed no cytotoxic effects on Saos-2 cell line. The overall results for the produced materials show the potential of the composites to be used in BTE.

Bacterial nanocellulose (BNC) surface has a negative charge that allows the adsorption of calcium ions to initiate the nucleation of different calcium phosphate phases. The aim of this study was to investigate different methods of mineralization on three-dimensional microporous bacterial nanocellulose, to mimetize the composition, structure, and biomechanical properties of natural bone. To generate 3D microporous biomaterial, the porogen particles were incorporated during the BNC fermentation with the strain Komagataeibacter medellinensis. Calcium phosphates (CPs) were deposited on BNC scaffolds by five alternating immersing cycles with calcium and phosphate solutions. Scanning electron microscopy micrograph showed that the scaffolds have different pore sizes, between 70 and 350 µm, but the porous interconnectivity was affected by the biomineralization method and time. The crystals on the BNC surface are shown to be rod-shaped with a calcium phosphate ratio similar to that of immature bone, increasing from 1.13 to 1.6 with cycle numbers. The main mineral phases obtained by X-ray diffraction were Octacalcium Dihydrogen Hexakis (phosphate (V)) Pentahydrate (OCP). In vitro cell studies showed good cellular adhesion and higher cell viability to 95% in all the scaffolds. Osteogenic differentiation of human bone marrow mesenchymal stem cells on the scaffolds was evaluated by bone expression markers like alkaline phosphatase, osteocalcin, and osteopontin. In conclusion, it is possible to prepare 3D BNC scaffolds with controlled microporosity, which could allow osteoblast adhesion, proliferation, and differentiation.

The evaluation of the possibility of obtaining dihaploids by the method of anther culture in vitro to accelerate selection for resistance to prolonged flooding of seeded rice with water was carried out. The object of research is rice hybrids F2 of the Rice Breeding and Seed Production Laboratory of the “Donskoy” ASC, obtained as a result of interbreeding the best economically valuable varieties with samples bearing genes of resistance to prolonged flooding with water. Basic nutrient medium with an optimal composition of nutrition elements and growth hormones stimulating callo- and morphogenesis were used. For the induction of callus formation, 12604 anthers were planted, according to 26 hybrid combinations represented by 68 plants, as a result, 716 neoplasms were obtained, including 586 non–morphogenic callus, 130 regenerating plants. Cultivation of anthers revealed large genotypic differences in the samples. According to the responsiveness to neoplasms, 60% of the panicles showed a positive result, the rest did not give callus. The most responsive to the formation of callus were hybrid combinations: 5009/2 – 84 pcs., 5010/2 – 94 pcs., 4565/3 – 85 pcs., 4641/2 – 69 pcs. They also showed the ability to morphogenesis. Androgenic plants were obtained by 14 hybrid combinations, their share was 1.03% of the total number of inoculated anthers. 30 green regenerative lines from four rice hybrids were obtained, differing in visual morphological assessment: 5009/2 - 5 pcs., 5010/2 – 5 pcs., 4565/3 – 2 pcs., 4641/2 – 18 pcs. The isolated lines are characterized by good responsiveness in the culture of anthers in vitro, carry genes for resistance to prolonged flooding, and can be used in rice breeding programs using DG technologies.

This work presents the results of an experimental socio-economic study conducted in two shanty towns of Messina as part of a systemic urban regeneration and fight against poverty program called Capacity. The study has shown that the development of a positive attitude towards the future and the confidence in others are associated with the development of the riskiest option, which is the one that can give the highest pay-off. The paper also illustrates the expected and unexpected outcomes of projects for individuals and the community, as well as the economic benefits for the public administration and the society of a strategy that reduces the reliance on social welfare measures as well as the local control exercised by organized crime.

Cities consume over 75% of natural resources, produce over 50% of global waste and emit 60 - 80% of greenhouse gases. The scenario that by 2050 two thirds of the world population will live in cities, highlights how cities are still responsible of the growing consumption characterized by linear economy processes, with the production of various types of waste. In this unsustainable framework, the Circular Economy offers the opportunity to shape the urban system by means of rethinking the possibility to produce and use goods and services exploring new ways to ensure long-term prosperity. The Circular City paradigm contains in fact all the principles of the Circular Economy: recovery, recycling and sharing. In particular, Circular City also introduces actions related to the development of renewable energy communities, use of green materials, CO2 absorption approaches and Proximity Cities. The aim of this work is to develop a methodology to build a composite index (Circular City Index) capable of measuring the degree of implementation of urban policies that enable the territory to initiate an ecological transition of public assets. The city of Cagliari (Sardinia, Italy) represents the case study to apply circular urban policies in public properties, for civil and military use.

Despite a significant advance in the pathophysiological understanding of peripheral nerve damage, the successful treatment of large nerve defects remains an unmet medical need. In this article, axon growth guidance for peripheral nerve regeneration is systematically reviewed and discussed mainly from the engineering perspective. In addition, the common approach to surgery, bioengineering approaches to emerging technologies (i.e. optogenetic stimulation and magnetic stimulation) for functional recovery are discussed, with pros and cons. Alternatively, clear future perspectives of axon guidance and nerve regeneration are addressed.

Peripheral nerve injuries pose a major clinical concern world-wide, and functional recovery after segmental peripheral nerve injury is often unsatisfactory, even in case of autografting. Although it is well established that angiogenesis plays a pivotal role during nerve regeneration, the influence of lymphangiogenesis is strongly underinvestigated. In this study, we analyzed the presence of lymphatic vasculature in healthy and regenerated murine peripheral nerves, revealing that nerve autografts contained increased numbers of lymphatic vessels after segmental damage. This led us to elucidate the interaction between lymphatic endothelial cells (LECs) and Schwann cells (SCs) in vitro. We show that SC and LEC secretomes do not influence the respective other cell types’ migration and proliferation in 2D scratch assay experiments. Furthermore, we successfully created lymphatic microvascular structures in SC-embedded 3D fibrin hydrogels in the presence of supporting cells, whereas SCs seemed to exert anti-lymphangiogenic effects when cultured with LECs alone. Here, we describe for the first time increased lymphangiogenesis after peripheral nerve injury and repair. Furthermore, our findings indicate a potential lymph-repellent property of SCs, thereby providing a possible explanation for the lack of lymphatic vessels in the healthy endoneurium. Our results highlight the importance to elucidate the molecular mechanisms of SC-LEC interaction.

Abstract: Urban regeneration decision-making is a complex process, as it involves a wide range of decision-makers, public-private partnerships in finance and implementation, including the inevitable considerable amount of risk on a long-term basis. There are a multitude of stakeholders, the citizens being the key stakeholders. It is necessary to involve the citizens in the planning process. Such involvement allows the communities to express their needs and aspirations, which is useful in the policymaking, delivery of planning programs, and in the monitoring process. In such a context, Factor analysis was the statistical technique used (1) Carry out factor analysis based on the principal component analysis method using the software XLSTAT 2021.4.1.1205 - (2) Construct a factor model of Urban Regeneration. (3) Interpret and label the factor dimensions. The results of the analysis indicated that the first two principal components accounted for 60.04% of the total variance of the original dataset. All variables seemed to be, positively correlated to each other and contributed similarly to principal components PC1 &PC2. The observations were well clustered; except for very few outliers. The limitation of the work was that the perceptions of the citizens were limited to the variables derived by the researcher.

Hearing dysfunctions can be classified by type, degree, configuration, time of onset, aetiology, and finally, consequences on speech development. They can be divided into conductive, mixed, central types and sensorineural. Conductive hearing loss (CHL) results from interference with the mechanical transmission of sound through the external and middle ear; it can be congenital, as a consequence of anatomic abnormalities, but it can commonly be acquired following middle ear inflammatory pathologies. Sensorineural hearing loss (SNHL) results from failure to transduce vibrations to neural impulses in the cochlean and is a consequence of an irreversible damage to the differentiated cells which make up the organ of hearing and the acoustic paths at various levels. Mixed hearing loss involves a combination of these two types in the same ear. Studies in neuroscience field have shown that the prevention of cell degeneration is only possible if all the factors taken at the different stages of stem cells’ multiplication and differentiation are administered together. We have demonstrated this in a recent study on the ability of SCDSFs to prevent neurodegeneration in hippocampal cells of the CA1 zone in mice. This study confirms previous findings demonstrating that early developmental zebrafish embryo extracts could act as a modulator of senescence in human mesenchymal stem cells (hMSC) isolated from many adult tissues. These findings have open a promising way for the approaches promoting the rejuvenation and regeneration of different tissues, by-passing stem cell transplantation. In the present clinical trial we have used SCDSFs to study the possible reversion of neurosensory hearing loss, until now considered an irreversible condition.

Dental pulp tissue engineering (TE) quests to regenerate dentin/pulp complex by combining a suitable supporting matrix, stem cells, and biochemical stimuli. Such procedures foresee a matrix that can be easily introduced into the root canal system (RCS) and tightly adhere to dentin walls to assure the dentin surface's proper colonization with progenitor cells capable of restoring the dentin/pulp complex. Herein was investigated an injectable self-setting hyaluronic acid-based (HA) hydrogel system, formed by aldehyde-modified (a-HA) with hydrazide-modified (ADH), enriched with platelet lysate (PL), for endodontic regeneration. The hydrogels' working (wT) and setting (sT) times, the adhesion to the dentine walls, the hydrogel's microstructure, and the delivery of human Dental Pulp Cells (DPCs) were studied in vitro. Hydrogels incorporating PL showed a suitable wT and sT and a porous microstructure. The tensile tests showed that the breaking point occurs after 4.13 mm deformation. While in the indentation test after 1.3 mm deformation. Both breaking points occur in the hydrogel extension. The HA/PL hydrogels exhibited supportive properties and promoted cell migration toward dentin surfaces in vitro. Overall, these results support using PL-laden HA injectable hydrogels (HA/PL) as a biomaterial for DPCs encapsulation, thereby displaying great clinical potential towards endodontic regenerative therapies.

In 1998, the Lisbon Universal Exhibition – Expo'98 – led to an urban regeneration process on Lisbon’s waterfront. Following other cities, this event was a pretext to replace a depressed area and to re-connect it with the river, through the creation of a set of new spaces for common use along the water. For them, it was promoted a public art program, which can be considered quite innovative in the Portuguese context, and that resulted in a monumentalisation of Lisbon’s eastern riverfront, later extended to other areas. Behind this framework, this article aims to debate the relations between public art and the dynamics of urban regeneration at the end of the 20th century. For that, it will analyse: 1) the Expo'98’s public art program, comparing its initial assumptions with the final results; 2) the impact of this program, through the identification of public art’s placements before (1974-1998) and after (1999-2009) the event. As a result, it is possible to find that the placement of public art reveals the spaces that were "conquered" to the port system, and a dialectic between functional/economic and leisure/symbolic values. It is concluded that public art had a significant role in the urban processes of the late 20th century, which is quite evident in a discourse that considers it as a qualifying factor of urban space and a mean of economic and social development.

Kandelia obovata (Ko) and Aegiceras corniculatum (Ac) are common and dominant plant species in mangrove wetlands in South China, and distribute in the similar tidal zones along the coastline. The present study aimed to determine the allelopathic effects of leaf litter leachates (LLLs) from Ko and their purified condensed tannins (PCTs) on the germination and growth of Ac by mangrove microcosms. Replicate pots containing five different levels of LLLs and PCTs were separately prepared and propagules of Ac were placed in each treatment. Both LLLs and PCTs significantly inhibited the germination and growth of Ac, especially in high levels. The final germination rates of roots, stems, and the number of fine roots declined continuously while other growth indicators, including the lengths of fine roots, nutritive roots, the biomasses of roots, stems, leaves, increased firstly and then decreased with increasing levels. These results indicated that LLLs from the leaf litter of Ko, in particular, their PCTs exerted an inhibition effect on propagule germination and seedling growth of Ac, and the inhibitory effects were concentration dependent. This study suggested that condensed tannins from leaf litter, acting as allelochemicals, could regulate the natural regeneration of a mangrove forest.

Cannabis has long been used for its medicinal and psychoactive properties. With the relatively new adoption of formal medicinal cannabis regulations worldwide, the study of cannabinoids, both endogenous and exogenous, has similarly flourished in more recent decades. In particular, research investigating the role of cannabinoids in regeneration and neurodevelopment has yielded promising results in vertebrate models. However, regeneration-competent vertebrates are few, whereas a myriad of invertebrate species have been established as superb models for regeneration. As such, this review aims to provide a comprehensive summary of the endocannabinoid system, with a focus on current advances in the area of endocannabinoid system contributions to invertebrate neurodevelopment and regeneration.

Our recent study detected the expression of a tissue Renin-Angiotensin System (tRAS) in human intervertebral discs (IVDs). The present study sought to investigate the impact of angiotensin II receptor type 1 (AGTR1) antagonist losartan on human nucleus pulposus (NP) cell inflammation and degeneration induced by tumor necrosis factor-α (TNF-α). Human NP cells (4 donors, Pfirrmann grade 2-3, 30-37 years old, male) were isolated and expanded. TNF-α (10 ng/mL) was used to induce inflammation and degeneration. We examined the impact of losartan supplementation and measured gene expression of tRAS, anabolic, catabolic, and inflammatory markers in NP cells after 24 h and 72 h of exposure, respectively. T0070907, a PPAR-gamma antagonist, was applied to examine the regulatory pathway of losartan. Losartan (1 mM) significantly impaired TNF-α induced increase of pro-inflammatory (nitric oxide and TNF-α), catabolic (matrix metalloproteinases), and tRAS (AGTR1a and Angiotensin Converting Enzyme) markers. Further, losartan maintained the NP cell phenotype by upregulating aggrecan and downregulating collagen type I expression. In summary, losartan showed anti-inflammatory, anti-catabolic, and positive phenotype modulating effects on human NP cells. These results indicate that tRAS signaling plays an important role in IVD degeneration, and tRAS modulation with losartan could represent a novel therapeutic approach.

Cranio-maxillofacial structure is a region of particular interest in the field of regenerative medicine due to both its anatomical complexity and the numerous abnormalities affecting this area. However, this anatomical complexity is what makes possible the coexistence of different microbial ecosystems in the oral cavity and the maxillofacial region, contributing to the increased risk of bacterial infections. In this regard, different materials have been used for their application in this field. These materials can be obtained from natural and renewable feedstocks or by synthetic routes with desired mechanical properties, biocompatibility and antimicrobial activity. Hence, in this review, we have focused on bio-based polymers, which by their own nature, by chemical modifications of their structure, or by their combination with other elements, provide a useful antibacterial activity as well as the suitable conditions for cranio-maxillofacial tissue regeneration. This approach has not been reviewed previously, and we have specifically arranged the content of this article according to the resulting material and its corresponding application, reviewing guided bone regeneration membranes; bone cements; and devices and scaffolds for both soft and hard maxillofacial tissue regeneration, including hybrid scaffolds, dental implants, hydrogels and composites.

The natural vegetation study was conducted in Kafta-sheraro national park (KSNP) North, Ethiopia to explore floristic composition, structure and regeneration of woody species in the home of African elephant. In the park, the above information is not well documented which is necessary for conservation. Data were collected From August to December 2018. The vegetation data were collected from 161 quadrats of size 20m×20m, 5mx5m for shrub ̸ tree, sapling and seedling respectively. Individual trees and shrubs DBH >=2.5cm and height >=2m were measured using Tape meter and Clinometer respectively. DBH, frequency, density, basal area, and IVI were used for vegetation structure. A total of 70 woody species 46 (65.7%) trees, 18 (25.7%) shrubs and 6 (8.6%) tree ̸ shrub) were identified. The total basal area and density of 79.3 m2 ha-1, and 466 ±12.8 (S.E.) individuals ha-1 were calculated for 64 woody species. Fabaceae was the most dominant family occupied 16 species (23.0%) followed by Combretaceae 8 species (11.4%). Acacia mellifera and Combretum hartmannianum were the most dominant and frequent species. Abnormal patterns of selected woody species were dominantly identified. Regenerating status all the woody plant species was categorized as “Fair” (18.75%), “Poor” (7.81 %) and “None” (73.44%). However, there is good initiation for conservation of the park; still the vegetation of the park was threatened by firewood collection, charcoal production, fire, intensive farming, mining and over grazing. Therefore, the study area as the habitat for the population of the African elephant; the KSNP should be recommended the highest conservation priority and studied the soil seed bank of species having poor regeneration condition.

Liver cirrhosis leads to hepatic dysfunction and life-threatening conditions. Though clinical efficacy of autologous bone marrow-drived mesenchymal stem cells (BM-MSC) transplantation in alcoholic cirrhosis (AC) was demonstrated, the relevant mechanism has not been elucidated. We aimed to identify predictive factors and gene/pathways for responders after autologous BM-MSC transplantation. Fifty-five patients with biopsy-proven AC underwent autologous BM-MSC transplantation. The characteristics of responders who showed improvement in fibrosis score (≥ 1) after transplantation were compared with those of non-responders. BM-MSCs were analyzed with cDNA microarrays to identify genes and pathways that were differentially expressed in responder after transplantation. Thirty-three patients (66%) were responders. In the multivariate analysis, initial high Laennec score (p=0.007, odds ratio 3.73) and performance of BM-MSC transplantation (p=0.033, odds ratio 5.75) were predictive factors for responder. Three genes (olfactory receptor 2L8, microRNA4520-2, and chloride intracellular channel protein 3) were upregulated in responders and 11 metabolic pathways (inositol phosphate, ATP-binding cassette transporters, protein kinase signaling, extracellular matrix-receptor interaction, endocytosis, phagosome, hematopoietic cell lineage, adipocytokine, peroxisome proliferator-activated receptor, fat digestion/absorption, and insulin resistance) were upregulated in non-responders (p<0.05). BM-MSC transplantation is warranted treatment for AC patients with high Laennec score. Cell-based therapy utilizing response-relating genes or pathway can be treatment candidate.

Reactive oxygen species (ROS), which were originally classified as exclusively deleterious compounds, have gained increasing interest in the recent years given their action as bona fide signalling molecules. The main target of ROS action is the reversible oxidation of cysteines, leading to the formation of disulfide bonds, which modulate protein conformation and activity. ROS endowed with signalling properties are mainly produced by NADPH oxidases at the plasma membrane, but their action also involves a complex machinery of multiple redox-sensitive protein families that differs in their subcellular localization and their activity. Given that the levels and distribution of ROS are highly dynamic in part due to their limited stability, the development of various fluorescent ROS sensors, some of which are quantitative (ratiometric), represents a clear breakthrough in the field and have been adapted to both ex vivo and in vivo applications. The physiological implication of ROS signalling will be presented mainly in the frame of morphogenetics processes, embryogenesis, regeneration, and stem cell differentiation. Gain and loss of function as well as pharmacological strategies have demonstrated the wide but specific requirement of ROS signalling at multiple stages of these processes and its intricate relationship with other well-known signalling pathways.

Background: In spinal cord injury, radical treatment is still a persistent hope for patients and clinicians. Our study aimed to determine the different histological changes in central, cranial and caudal sites of compressed spinal cord as a result of neuroectodermal stem cells (NESCs) transplantation in rats. Material and methods: For extraction of NESCs, future brains were extracted from mice embryos (10-days old) and cultured. Eighty, male rats were divided randomly into control, sham (20 rats each); while 40 rats were subjected to compressed spinal cord injury (CSCI). Seven days after spinal cord injury, rats were subdivided into 2 groups (20 rats each); an untreated and treated with NESCs injected cranial and caudal to the site of the spinal cord injury. Rats were sacrificed 4 weeks after transplantations of NESCs and specimens from the spinal cord at the central, cranial and caudal to site of spinal cord injury were proceeded to be stained with haematoxylin & eosin, osmic acid and Immunohistochemistry of glial fibrillary acidic protein (GFAP). Results: Sections of CSCI revealed areas of hemorrhages, necrosis and cavitation limited by reactive astrocytosis, with upregulation of GFAP expression. Evidence of remyelination and mitigation of histopathological features, reactive astrocytosis in CSCI sections were more pronounced in cranial than in caudal region. Conclusions: NESCs transplantation ameliorated the pathological changes, promoted remyelination.

Na-magadiite exchanged with cetyl-trimethylammonium cations provided organophilic silicate materials that allowed the effective removal of the acidic dye "eosin". The organic cations were intercalated into the interlayer spacing of the layered silicate via an exchange reaction between the organic cations from their bromide salt and the solid Na-magadiite at room temperature. Different techniques were used to characterize the effect of the initial concentration of the surfactant on the structure of the organo-magadiites. The C, H, N analysis indicated that a maximum of organic cations of 1.03 mmol/g was achieved at initial concentrations higher than 0.25 mmol/g and was accompanied by an expansion of the basal spacing of 3.15 nm, with a tilted angle of 64.5° to the silicate layers. The conformation of the organic surfactants was probed using solid-state ¹³C, finding mainly the trans conformation similar to that of the starting cetyl trimethylammonium bromide salt (C16TMABr). Thermal gravimetric analysis was carried out to study the thermal stability of the resulting organo-magadiites. The intercalated surfactants started to decompose at 200 °C, with a mass loss percentage of 8 % to 25 %, depending on the initial loading of the surfactant, and was accompanied by a decrease of the basal spacing from 3.20 nm to 2.51 nm, as deduced from the in situ X-ray diffraction studies. At temperatures below 220 °C, an expansion of the basal spacing from 3.15 to 3.35 nm occurred. These materials were used as a removal agent for the anionic dye "eosin". The maximum amount of the dye removed was related to the organic cation content and to the initial concentration of eosin, with an improvement from 2.5 mg/g to 98 mg/g. This value decreased when the organo-magadiite was preheated at temperatures above 200 °C. The regeneration tests indicated that an 85 % removal efficiency was maintained after 6 cycles of use for the organo-magadiites.

Various tissue resident stem cells are receiving attention from basic scientists and clinicians as they hold certain promise for regenerative medicine. This paper is intended to clarify and facilitate the understanding, development and adoption of regenerative medicine in general and specifically of therapies based on unmodified, autologous adipose-derived regenerative cells (UA-ADRCs). To this end, results of landmark experiments on stem cells and stem cell therapy performed in the labs of the authors are summarized, the most intriguing of which are the following: (i) vascular associated mesenchymal stem cells (MSCs) can be isolated from different organs (adipose tissue, heart, skin, bone marrow and skeletal muscle) and differentiated into ectoderm, mesoderm and endoderm, providing significant support for the hypothesis of the existence of a small, ubiquitously distributed, universal vascular associated stem cell with full pluripotency; (ii) the orientation and differentiation of MSCs are driven by signals of the respective microenvironment; and (iii) these stem cells irrespective of the tissue origin exhibit full pluripotent differentiation potential without any prior genetic modification or the need for culturing. They can be obtained from a small amount of adipose tissue when using the appropriate technology for isolating the cells, and can be harvested from and re-applied to the same patient at the point of care without the need for complicated processing, manipulation, culturing, expensive equipment, or repeat interventions. These findings demonstrate the potential of UA-ADRCs for triggering the development of an entire new generation of medicine for the benefit of patients and of healthcare systems.

Background: Traditional root canal therapy (RCT) has been the primary method for treating necrotic pulp, but it presents limitations such as loss of vitality and postoperative fractures. Recent advancements in regenerative endodontics offer alternative strategies to restore pulp tissue vitality and function. These approaches involve stem cell transplantation, cell homing, and the use of organic scaffolds to promote pulp regeneration. Highlight: This article reviews various regenerative endodontic techniques, including autogenic and allogenic dental pulp transplantation, the use of amniotic membrane scaffolds, platelet-rich fibrin therapy, specialized pro-resolving mediators, nanofibrous and bioceramic-based scaffolds, injectable scaffolds, and dentin matrix proteins. These strategies leverage stem cells, growth factors, and biomaterials to create conducive microenvironments for cell attachment, proliferation, and differentiation, thereby facilitating pulp tissue regeneration. Methods: The literature search was conducted using databases such as PubMed, Scopus, and Google Scholar, employing keywords like "regenerative endodontics," "pulpitis," "pulp regeneration," and "platelet-rich fibrin." Conclusion: The reviewed approaches in regenerative endodontics show promising potential for overcoming the limitations of traditional RCT and promoting pulp tissue regeneration. However, further research is needed to optimize protocols, address challenges, and translate these advancements into routine clinical practice. Ultimately, these innovations have the potential to enhance patient outcomes in endodontic treatment by restoring pulp vitality and function effectively.

Chitosan-based hydrogels are promising in bone tissue regeneration due to their biocompatibility, tunable properties, and ability to mimic the extracellular matrix. With high water content and porous structure, they can encapsulate bioactive agents for controlled release. Chitosan, derived from chitin in crustacean exoskeletons and fungi, possesses mechanical qualities ideal for biomedical applications. This study will evaluate recent advances in chitosan hydrogels for bone regeneration, focusing on their efficacy, mechanisms, and challenges in clinical settings.

Newts, a type of urodele amphibian, offer remarkable insights into regenerative medicine due to their extraordinary tissue regeneration capabilities—a challenging feat in humans. During limb regeneration of adult newts, fascinating cellular and molecular processes are revealed, including scarless healing, de-differentiation of mature cells, and regeneration of limbs and digits. Sonic hedgehog (Shh), crucial for vertebrate limb development, is regulated by the zone of polarizing activity regulatory sequence (ZRS) in the limb bud zone of polarizing activity (ZPA). The metamorphosed (terrestrial) newt can reactivate Shh during regeneration, facilitating proper limb patterning. Cell types capable of regulating the ZRS in metamorphosed newts remain unknown. The identification of such cell types provides invaluable insight to novel regenerative mechanisms. Therefore, in this study, we developed the first newt ZRS reporter. First, we isolated and characterized the newt ZRS enhancer (nZRS), identifying conserved DNA binding sites. Several binding sites with medical relevance were conserved in the newt ZRS. In a functional analysis, we developed a system composed of a transgenic nZRS reporter newt and a new newt anti-Shh antibody, together allowing Shh monitoring during limb regeneration. This system provides a valuable in vivo approach for future genetic studies of patterning during limb regeneration.

Rationale: Post-hepatectomy liver failure (PHLF) remains a significant risk for patients undergoing partial hepatectomy (PHx). Reliable prognostic markers and treatments to enhance liver regeneration are lacking. Plasma nanoparticles, including lipoproteins, exosomes, and extracellular vesicles (EVs), can reflect systemic and tissue-wide proteostasis and stress, potentially aiding liver regeneration. However, their role in PHLF is still unknow. Methods: Our study included 9 patients with hepatocellular carcinoma (HCC) undergoing PHx: 3 patients with PHLF, 3 patients undergoing the Associating Liver Partition and Portal vein Ligation for Staged hepatectomy (ALPPS) procedure and three matched controls without complications after PHx. Patient plasma was collected before (PRE), 1 and 5 days (POD1, POD5) after PHx. EVs were isolated by ultracentrifugation and extracted proteins subjected to quantitative mass spectrometry using a super-SILAC mix prepared from primary and cancer cell-lines. Results: We identified 2625 and quantified 2570 proteins in EVs in PHx patients. Among these, 53 proteins were significantly upregulated and 32 were downregulated in patients with PHLF compared to those without PHLF. Furthermore, 110 proteins were upregulated and 78 were downregulated in PHLF compared to patients undergoing ALPPS. The EV-proteomic signature in PHLF indicates significant disruptions in protein translation, proteostasis, intracellular vesicle biogenesis, as well as alterations in proteins involved in extracellular matrix (ECM) remodeling, metabolic and cell cycle pathways, present already before PHx. Conclusion: Longitudinal proteomic analysis of EV´s circulating in human patient plasma undergoing PHx uncovers proteomic signatures associated with PHLF which reflect dying hepatocytes and endothelial cells and are present before PHx already. Our results support EVs as potential biomarkers and therapeutic targets for liver regeneration and failure.

Background: Fingertip injuries with volar pulp tissue defects are challenging to manage due to the complex anatomy and critical role of the fingertip in sensory and motor functions. Traditional surgical approaches are associated with donor site morbidity, prolonged immobilization, and suboptimal functional outcomes. This study aimed to evaluate the efficacy of a conservative treatment protocol using artificial dermis and semi-occlusive dressing for these injuries. Methods: A single-center, prospective study was conducted on 31 patients with fingertip injuries involving volar pulp defects. The treatment protocol involved wound debridement, application of artificial dermis (Pelnac®), and semi-occlusive dressing (IV3000®). Outcomes were assessed using subjective questionnaires and objective measures, including fingerprint regeneration, sensory function, pain, and cosmetic appearance. The impact of injury characteristics on outcomes and treatment duration was evaluated. Complications were recorded, and patient satisfaction was assessed. Results: The mean treatment duration was 45.29 days (SD=17.53), with no significant differences based on injury characteristics. Complications were minimal, with only one case (3.22%) directly attributable to the treatment. Fingerprint regeneration was considerable (mean score=2.58, SD=0.67), with slightly better scores in bone-exposed injuries. Sensory disturbances were minimal, with no significant differences across injury types. Post-treatment pain was low (mean=0.45, SD=0.67), and cosmetic satisfaction was high (mean=4.09, SD=0.94). Overall patient satisfaction was high (mean=4.41, SD=0.67), regardless of injury severity. Discussion: This study demonstrates the efficacy of a conservative approach using artificial dermis and semi-occlusive dressing for fingertip injuries with volar pulp defects. The treatment protocol achieved favorable outcomes in tissue regeneration, sensory recovery, pain management, and cosmetic appearance while minimizing complications and morbidity associated with traditional surgical interventions. The high patient satisfaction across all injury types highlights the patient-centered nature of this approach. Conclusions: The conservative treatment protocol using artificial dermis and semi-occlusive dressing is a promising strategy for managing fingertip injuries with volar pulp defects. This approach harnesses the body's regenerative potential, minimizes surgical morbidity, and achieves excellent functional and aesthetic outcomes. Further research is needed to optimize treatment protocols and investigate long-term outcomes.

Glycosphingolipids (GSLs), a subtype of glycolipids containing sphingosine, are critical components of vertebrate plasma membranes, playing a pivotal role in cellular signaling and interactions. In human articular cartilage in osteoarthritis (OA), GSL expression is known notably to decrease. This review focuses on the roles of gangliosides, a specific type of GSL, in cartilage degeneration and regeneration, emphasizing their regulatory function in signal transduction. The expression of gangliosides, whether endogenous or augmented exogenously, is regulated at the enzymatic level, targeting specific glycosyltransferases. This regulation has significant implications for the composition of cell surface gangliosides and their impact on signal transduction in chondrocytes and progenitor cells. Different levels of ganglioside expression can influence signaling pathways in various ways, potentially affecting cell properties, including malignancy. Moreover, gene manipulations against gangliosides have been shown to regulate cartilage metabolism and chondrocyte differentiation in vivo and in vitro. This review highlights the potential of targeting gangliosides in the development of therapeutic strategies for osteoarthritis and cartilage injury and addresses promising directions for future research and treatment.

Low efficiency of in vivo transfection of a few fibers revealed a novel tissue network which temporally amplified growth stimulation in the entire regenerating rat soleus muscle. This acupuncture-like effect was demonstrated when the fibers began to grow after complete fiber degradation, synchronous inflammation, myoblast and myotube formation. The first detection of neonatal sarcoplasmic/endoplasmic reticulum ATPase (SERCA1b) has been made in this system. The neonatal, fast and slow SERCA isoforms displayed consequent change with innervation and differentiation, recapitulating events in muscle development. In vivo transfection of myotubes with plasmids expressing dominant negative Ras or a calcineurin inhibitor peptide (Cain/cabin) proved that the expression of the slow myosin heavy chain and the slow muscle type SERCA2a are dif-ferently regulated. In vivo transfection of a few nuclei of myotubes with dnRas or SER-CA1b shRNA stimulated fiber size growth in the whole regenerating muscle but only until the full size had been reached. Growth stimulation by Ras and SERCA1b antisense was abolished in co-transfection with Cain or in case of perimuscular injection with IL4 antibody. This revealed a novel signalling network resembling to scale-free networks which, starting from transfected fiber myonuclei as “hubs”, can amplify growth stimula-tion uniformly in the entire regenerating muscle.

Parkland contributes economic, social, and ecological benefits worldwide. However, parklands degradation is an encounter to biodiversity loss needs generating information on woody species. This study was aimed to investigate woody species composition, diversity, structure, and management of parklands along Agroecology, and farmers’ wealth status. Multistage sampling technique was employed. Highland Bede Kebele, and midland Durgi Kebele were randomly selected. A total of 60 quadrats with size of 100m x 50m were used for inventory. A total of 276 respondents were used. Species richness, Shannon, Simpson, and evenness diversity indices were subjected to one-way ANOVA, and independent t-test for comparison STATA V. 17.0. Of 31 woody species, (16 families in 26 genera) observed, 26 woody species belonging to 13 families in 22 genera from midland while 18 woody species belonging to 12 families in 16 genera from highland parklands were observed. Species richness 6.8, 3.7; Shannon 1.82, 1.68; Simpson 0.85, 0.73; evenness 0.83, 0.7 were observed in midland, and highland parklands, respectively. Poorly regenerated stands were observed along Agroecology. Pruning, pollarding, coppicing, lopping, cutting, fertilizing, watering were common management practices. In conclusion, woody species diversity, and structure were affected by Agroecology, and socioeconomic factors needing intervention to enhancing woody species conservation.

Background: RNA helicase DHX15 plays a significant role in vasculature development and lung metastasis in vertebrates. In addition, several studies have demonstrated the overexpression of DHX15 in the context of hepatocellular carcinoma. Therefore, we hypothesized that this helicase may play a significant role in liver regeneration, physiology and pathology. Methods: Dhx15 gene deficiency was generated by CRISPR/Cas9 in zebrafish and by TALEN-RNA in mice. FANA Antisense-Oligonucleotides were used to silence Dhx15 in wild-type mice. The hepatocellular carcinoma tumor induction model was generated by subcutaneous injection of Hepa 1-6 cells. Results: Homozygous Dhx15 gene deficiency was lethal in zebrafish and mouse embryos. Dhx15 gene deficiency impaired liver organogenesis in zebrafish embryos and liver regeneration after partial hepatectomy in mice. Also, heterozygous mice presented decreased number and size of liver metastasis after Hepa 1-6 cells injection compared to wild-type mice. Dhx15 gene silencing with FANA Antisense-Oligonucleotides in wild-type mice resulted in 80% reduced expression in the liver and a significant reduction in other major organs. In addition, Dhx15 gene silencing significantly hindered primary tumor growth in the hepatocellular carcinoma experimental model. Regarding the potential use of DHX15 as a diagnostic marker for liver disease, patients with hepatocellular carcinoma showed increased levels of DHX15 in blood samples compared with subjects without hepatic affectation. Conclusion: Dhx15 is a key regulator of liver physiology and organogenesis, is increased in the blood of cirrhotic and hepatocellular carcinoma patients and has a key role in controlling hepatocellular carcinoma tumor growth and expansion in experimental models.

The porous structure is an important three-dimensional morphological feature of the peripheral nerve guidance conduit (NGC), which permits infiltration of cells, nutrients, and molecular signals, and the discharge of metabolic waste. Porous structures with precisely customised pore sizes, porosities, and connectivities were used to construct fully permeable, semi-permeable, and asymmetric peripheral NGCs to replace traditional nerve autografts in treatment of long-segment peripheral nerve injury. In this review, the features of porous structures and classification of NGCs based on these characteristics are discussed. Common methods for constructing 3D porous NGCs in current research are described, as well as the pore characteristics and the parameters used to tune the pores. The effects of the porous structure on the physical properties of NGCs including biodegradation, mechanical performance, and permeability were analysed. Pore structure affects the biological behaviour of Schwann cells, macrophages, fibroblasts, and vascular endothelial cells during peripheral nerve regeneration. Construction of ideal porous structures is a significant advancement in the regeneration of peripheral nerve tissue engineering materials. The purpose of this review is to generalise, summarise, and analyse the preparation methods for porous NGCs and their biological functions in promoting peripheral nerve regeneration to guide development of medical nerve repair materials.

Windthrows in the forestry fund, which have become more frequent due to the increase in extreme weather events, have had and continue to have negative economic and ecological effects, making them a pressing issue in forestry research. Their urgency has been amplified in light of the need to develop sustainable forest management systems. The main objectives of this study are to evaluate the effects of windthrows on some microbiological properties of forest soils and to monitor the evolution of the degraded tree regeneration, 4 years after the event, for three tree species: Norway spruce (Picea abies L.), sessile oak (Quercus petraea), and European beech (Fagus sylvatica L.). The experimental plot used is arranged in dispersed blocks and subdivided plots, with three repetitions, bifactorial, factor A representing the tree species, and factor B the windthrows: in two situations: affected (AW) and not affected by felling (WW). Three representative soil profiles were studied for each tree species, including dehydrogenase activity (DA) and the number of fungi (NF) in the organic horizon at the soil surface. Dehydrogenase activity was determined by the extraction of triphenylformazan, and NF was evaluated using the Plate Count Method (Petri plates) and the Sabouraud Agar culture medium. The values of Current Dehydrogenase Activity (CDA) and Potential Dehydrogenase Activity (PDA) did not show significant statistical differences in relation to the windthrow factor, but were distinctly significant (p &lt; 0.01) for the tree species factor. On the other hand, NF showed statistical significance for both tree species and windthrows factors, at a level of p &lt; 0.01. Correlations were highlighted between the differences in AW and WW of CDA, PDA, and NF with the number of naturally regenerated seedlings (NRS), and the type of soil. The regressions established between NRS and the differences in CDA and PDA, the dependence of the differences in PDA on the differences in NF, and the regression between the differences in NF and the type of soil demonstrate the importance of soil characteristics in the natural regeneration process of the main tree species - Norway spruce, sessile oak, and European beech. Stimulating NRS in AW forests and increasing the volume of terrestrial organic carbon (TOC) biomass is directly dependent on soil fertility, primarily determined by soil organic carbon (SOC), which accumulates in the soil as a result of organic matter, deposited on the surface. Sustainable forest management of AW plots should stimulate the accumulation of SOC, including the partial or total preservation of dead trees, provided that the attack of specific diseases and pests is avoided.

Cytokines secreted by individual immune cells regulate tissue regeneration and allow communication between various cell types. Cytokines bind to cognate receptors and trigger the healing process. Determining the orchestration of cytokine interactions with their receptors on their cellular targets is essential to fully understand the process of inflammation and tissue regeneration. To this end, we have investigated the interactions of Interleukin-4 cytokine (IL-4)/Interleukin-4 cytokine receptor (IL-4R) and Interleukin-10 cytokine (IL-10)/Interleukin-10 cytokine receptor (IL-10R) using in situ Proximity Ligation Assays in a regenerative model of skin, muscle and lung tissues in the mini-pig. The pattern of protein-protein interactions was distinct for the two cytokines. IL-4 bound predominantly to receptors on macrophages and endothelial cells around the blood vessels while the target cells of IL-10 were mainly receptors on muscle cells. Our results show that in situ studies of cytokine-receptor interactions can provide unravel the fine details of the mechanism of action of cytokines.

The paper proposes, through an experimental method, a conceptualisation of theoretical-methodological and practical guidelines for the project on the system of collective open spaces, residual areas, abandoned areas that characterise the contemporary city, both in the urban contexts of the historical city, and in the more marginal areas of the modern suburbs. This methodology was experimented on an historical neighbourhood in Rome (San Lorenzo discrict) starting from the results of some researches conducted in collaboration between Sapienza University of Rome and the Escuela de Architettura, Universidad San Jorge and some design experiments conducted by Gràvalosdimonte design studio. This methodology and experimentation combines the theme of the inter-scalarity of the project (territorial, urban and local scale) with the themes of the flexibility and proximity of functions with respect to local instances and contingencies caused by the pandemic situation and by the sustainability perspectives, with particular reference to the need for urban health, the dialectic between public and private space in the perspective of the notion of common good, the bottom-up project for the construction of proximity spaces and plurality in the decision-making process.

In bone tissue engineering, ceramics have been the choice due to their excellent biological properties. But the paradigm changed with the discovery of bioactive glasses (BGs) in 1969 by Larry Hench and co-workers, due to their ability to bond to living tissues through the formation of an interfacial bone-like hydroxyapatite layer when the bioglass was put in contact with biological fluids in vivo. Among a number of tested compositions, the one exhibiting the highest bioactivity index is the well-known trademarked 45S5 Bioglass®. The topic received increasing attention particularly after 1985 when this material entered in the market of biomedical devices, inspiring many other investigations aiming at further exploring the in vitro and in vivo performances of this BG, or developing other related BG compositions. The research efforts gradually revealed a number of shortcomings of 45S5 Bioglass®, mostly derived from its high sodium content, initially intended to decrease the melting temperature and accelerating the degradation of the silicate network over time. But the extensive release of sodium from 45S5 Bioglass® in the biological fluids creates a high pH cytotoxic environment. Other serious drawbacks include a fast degradation rate, and a poor sintering ability, which hinders the reliable fabrication of porous scaffolds. Therefore, sol-gel was regarded as an attractive alternative to prepare alkali-free BG compositions. The process uses inorganic and/or organic precursors, which undergo hydrolysis and condensation at room-temperature, being less costly. When properly conducted, the sol-gel process might result in amorphous structures with all the components intimately mixed at the atomic scale. Moreover, developing new better performing materials for bone tissue engineering is a growing concern, as the ageing of the world’s population leads to lower bone density and osteoporosis. This work describes the sol-gel synthesis of a novel quaternary silicate-based BG with the composition 60 SiO2 – 34 CaO – 4 MgO – 2 P2O5 (mol%) was prepared using acidified distilled water as single solvent. By controlling the kinetics of the hydrolysis and condensation steps, an amorphous glass structure could be obtained. The results of XRD of samples calcined within the temperature range from 600-900 ºC demonstrated that amorphous nature was maintained until 800 ºC, followed by partial crystallization at 900 ºC. The specific surface area, an important factor in osteoconduction, was also evaluated over different temperatures, ranging from 160.6 ± 0.8 m2/g at 600 ºC down to 2.2 ± 0.1 m2/g at 900 ºC, being accompanied consistent changes in average pore size and agreeing pore size distribution. The immersion of the BG particles in simulated body fluid (SBF) led to the formation of an extensive apatite layer on its surface. These overall results indicate the proposed material is very promising for biomedical applications in bone regeneration and tissue engineering.
