A Micro holes in a diamond are presented by using a homemade femtosecond (fs) Yb:KGW laser. An fs laser source was used emitting pulse duration of 230 fs at 1030 nm wavelength, whereas the spot size amounted to 8.9 μm. Parameters like pulse energy, and pulse number were varied over a wide range in order to evaluate their influence both on the micro hole geometry like hole diameter, circularity, taper angle, and on the drilling quality. Hourglass-shaped micro holes whose diameters decrease and increase again after a certain depth have important applications. The results demonstrate the feasibility of extending the drilling of an hourglass-shaped hole in a diamond sample, which has similar diameters at the hole entrance (92 μm) and exit (95 μm), but a much smaller diameter (28 μm) at a certain waist section inside the hole.

Birefringence of 3 × 10^-3 is demonstrated inside cross-sectional regions of 100 µm, inscribed by axially stretched Bessel-beam-like fs-laser pulses along the c-axis inside sapphire. A high birefringence and retardance of λ/4 at mid-visible spectral range (green) can be achieved utilizing stretched beams with an axial extension of 30-40 µm. Conditions of laser writing chosen ensure that there are no formations of self-organised nano-gratings. This method can be adopted for the creation of polarisation optical elements and fabrication of spatially varying birefringent patterns for optical vortex generation.

Laser patterning of implant materials for bone tissue engineering purposes has shown to be a promising technique to control cell properties such as adhesion or differentiation, resulting in an enhanced osteointegration. However, the perspective of patterning the bone tissue side interface to generate microstructure effects has never been investigated. In the present study, three different laser-generated patterns were machined on the bone surface with the aim to identify the best surface morphology compatible with osteogenic-related cells recolonization. The laser patterned bone tissue was characterized by electron scanning microscopy and confocal microscopy in order to obtain a comprehensive picture of the bone surface morphology. Cortical bone patterning impact upon cell compatibility and cytoskeleton rearrangement to the patterned surfaces was performed with Stromal Cells from Apical Papilla (SCAPs). Results indicated that laser machining had no detrimental effect upon consecutively seeded cells metabolism. Orientation assays revealed that surface patterning characterized by larger hatch distances was correlated with a higher cell cytoskeletal conformation to the laser-machined patterns. For the first time, to our knowledge, bone is considered and assessed here as a potentially engineered-improvable biological interface. Further studies shall focus on in vivo implications of this direct patterning.

We experimentally demonstrate enhanced spectral broadening of femtosecond optical pulses after propagation through silicon-on-insulator (SOI) nanowire waveguides integrated with two-dimensional (2D) graphene oxide (GO) films. Owing to the strong mode overlap between the SOI nanowires and the GO films with a high Kerr nonlinearity, the self-phase modulation (SPM) process in the hybrid waveguides is significantly enhanced, resulting in greatly improved spectral broadening of the femtosecond optical pulses. A solution-based, transfer-free coating method is used to integrate GO films onto the SOI nanowires with precise control of the film thickness. Detailed SPM measurements using femtosecond optical pulses are carried out, achieving a broadening factor of up to ~4.3 for a device with 0.4-mm-long, 2 layers of GO. By fitting the experimental results with theory, we obtain an improvement in the waveguide nonlinear parameter by a factor of ~3.5 and the effective nonlinear figure of merit (FOM) by a factor of ~3.8, relative to the uncoated waveguide. Finally, we discuss the influence of GO film length on the spectral broadening and compare the nonlinear optical performance of different integrated waveguides coated with GO films. These results confirm the improved nonlinear optical performance for silicon devices integrated with 2D GO films.

We report on the study of ultrafast laser-induced plasma expansion dynamics in a gas microjet. To this purpose, we focused femtosecond laser pulses on a nitrogen jet produced through a homemade De Laval micronozzle. The laser excitation leads to plasma excitation with a characteristic spectral line emission at 391 nm. By following the emitted signal with a detection system based on an Intensified Charge-Coupled Device (ICCD) we captured the two-dimensional spatial evolution of the photo-excited nitrogen ions with a temporal resolution on the nanosecond time scale. We fabricated the micronozzle on fused silica substrate by femtosecond laser micromachining. This technique enables high accuracy and three-dimensional capabilities, thus providing an ideal platform for developing glass-based microfluidic structures for application to plasma physics and ultrafast spectroscopy.

The development of printing technologies has enabled the realization of electric circuit fabrication on flexible substrate. However, the current technique remains restricted to single-layer patterning. In this paper, we demonstrate a fully solution-processable patterning approach for multi-layer circuits using a combined method of laser sintering and ablation. Selective laser sintering of silver (Ag) nanoparticle-based ink is applied to make conductive patterns on a heat-sensitive substrate and insulating layer. The laser beam path and irradiation fluence are controlled to create circuit patterns for flexible electronics. Microvia drilling using femtosecond laser through the polyvinylphenol-film insulating layer by laser ablation, as well as sequential coating of Ag ink and laser sintering, achieves an interlayer interconnection between multi-layer circuits. The dimension of microvia is determined by a sophisticated adjustment of laser focal position and intensity. Based on these methods, the flexible electronic circuit with chip-size-package light-emitting diodes was successfully fabricated and demonstrated with functional operations.

We propose a new approach of extending the laser filament plasma channel. By adding a circular aperture quartz plate before the focusing lens, the extension of the plasma channel is doubled. The effects of different diameters, thicknesses of the circular aperture quartz plate and different pulse energies on the length of the plasma channel were investigated. The experimental results show that the thickness of the quartz plate and the depth of the hole have little effects on the plasma channel of the filament, and the diameter of the hole in the center of the quartz plate has a significant effect on the length of the optical filament. The moving-focus model is used to explain the extension of the optical filament.

We demonstrate a multi-purpose plasmonic sensor based on nanovoid array fabricated via inexpensive and highly reproducible direct femtosecond laser patterning of thin glass-supported Au films. The proposed nanovoid array exhibits near-IR surface plasmon (SP) resonances, which can be excited under normal incidence and optimised for specific application by tailoring array periodicity as well as nanovoid geometric shape. Fabricated SP sensor offers competitive sensitivity of about 1600 nm/RIU at a figure of merit of 12 in bulk refractive index tests, as well as allows for identification of gases and ultra-thin analyte layers making the sensor particularly useful for common bioassay experiments. Moreover, isolated nanovoids support strong electromagnetic field enhancement at lattice SP resonance wavelength, allowing for label-free molecular identification via surface-enhanced vibration spectroscopy.

Transparent and high-hardness materials have become the object of wide interest. Most notably, it concerns technical glasses and crystals. A notable example is a sapphire – one of the most rigid materials having impressive mechanical stability and good optical properties. Nonetheless, using this material for 3D micro-fabrication is not straightforward due to its brittle nature. On the microscale, selective laser etching (SLE) technology is an appropriate approach for such media. Therefore, we present our research on c-cut crystalline sapphire microprocessing by using femtosecond radiation-induced SLE. Here we demonstrate a comparison between different wavelength radiation (1030 nm, 515 nm, 343 nm) usage for modification inscription and various etchants (Hydrofloridic acid, Sodium Hydroxide, Potassium Hydroxide and Sulphuric and Phosphoric acid mixture) comparison. We show that regular SLE etchants such as Hydrofluoric acid or Potassium Hydroxide are unsuitable materials for selective sapphire laser etching. Meanwhile, a 78% sulphuric and 22% phosphoric acid mixture at 270°C temperature is a good alternative for this process. We present the changes in the material after the separate processing steps. Finally, a protocol for advanced sapphire structure formation and a few exemplary structures are presented.

Optical biopsy describes a range of medical procedures in which light is used to investigate disease in the body, often in hard-to-reach regions via optical fibres. Optical biopsies can reveal a multitude of diagnostic information to aid therapeutic diagnosis and treatment with higher specificity and shorter delay than traditional surgical techniques. One specific type of optical biopsy relies on Raman spectroscopy to differentiate tissue types at the molecular level and has been used successfully to stage cancer. However, complex micro-optical systems are usually needed at the distal-end to optimise the signal-to-noise properties of the Raman signal collected. Manufacturing these devices remains a critical challenge, particularly in a way suitable for large scale adoption. In this paper, we describe a novel fibre-fed micro-optic system designed for efficient signal delivery and collection during a Raman spectroscopy based optical biopsy. Crucially, we fabricate the device using a direct-laser-writing technique known as ultrafast laser assisted etching which is scalable and allows components to be aligned passively. The Raman probe has a sub-millimetre diameter and offers confocal signal collection with 71.3 ± 1.5% collection efficiency over a 0.8 numerical aperture. Proof of concept spectral measurements were performed on mouse intestinal tissue and compared with results obtained using a commercial Raman microscope.

An approach employing ultrafast laser hybrid subtractive-additive microfabrication combining ablation, 3D nanolithography and welding is proposed for the realization of Lab-On-Chip (LOC) device. Single amplified Yb:KGW fs-pulsed laser source is shown to be suitable for fabricating microgrooves in glass slabs, polymerization of fine-meshes filter out of hybrid organic-inorganic photopolymer SZ2080 inside them, and, lastly, sealing the whole chip with cover glass into a single monolithic piece. The created microfluidic device proved its particle sorting function by separating 1 μm and 10 μm polystyrene spheres in a mixture. All together, this shows that fs-laser microfabrication technology is a flexible and versatile tool for the manufacturing of mesoscale multi-material LOC devices.

Amorphous silicon (α-Si) film present an inexpensive and promising material for optoelectronic and nanophotonic applications. Its basic optical and optoelectronic properties are known to be improved via phase transition from amorphous to polycrystalline phase. Infrared femtosecond laser radiation can be considered as a promising nondestructive and facile way to drive uniform in-depth and lateral crystallization of α-Si films that are typically opaque in UV-visible spectral range. However, so far only a few studies reported on utilization of near-IR radiation for laser-induced crystallization of α-Si providing no information regarding optical properties of the resultant polycrystalline Si films. The present work demonstrates efficient and gentle single-pass crystallization of α-Si films induced by their direct irradiation with near-IR femtosecond laser pulses coming at sub-MHz repetition rate. Comprehensive analysis of morphology and composition of laser-annealed films by atomic-force microscopy, optical, micro-Raman and energy-dispersive X-ray spectroscopy, as well as numerical modeling of optical spectra, confirmed efficient crystallization of α-Si and high-quality of the obtained films. Moreover, we highlight localized laser-driven crystallization of α-Si as a promising way for optical information encryption, anti-counterfeiting and fabrication of micro-optical elements.

We report on the application of femtosecond laser micromachining to the fabrication of complex glass microdevices, for high-order harmonic generation in gas. The three-dimensional capabilities and extreme flexibility of femtosecond laser micromachining allow us to achieve accurate control of gas density inside the micrometer interaction channel. This device gives a considerable increase in harmonics generation efficiency if compared with traditional harmonic generation in gas jets. We propose different chip geometries that allow to control the gas density and driving field intensity inside the interaction channel to achieve quasi-phase matching conditions in the harmonic generation process. We believe that these glass micro-devices will pave the way to future downscaling of High-order Harmonic Generation beamlines.

We report an easy-to-implement device for SERS-based detection of various analytes dissolved in water droplets at trace concentrations. The device combines an analyte-enrichment system and SERS-active sensor site, both produced via inexpensive and high-performance direct fs-laser printing. Fabricated on a surface of water-repellent polytetrafluoroethylene substrate as an arrangement of micropillars, the analyte-enrichment system supports evaporating water droplet in the Cassie-Baxter superhydrophobic state, thus ensuring delivery of the dissolved analyte molecules towards the hydrophilic SERS-active site. The efficient pre-concentration of the analyte onto the sensor site based on densely-arranged spiky plasmonic nanotextures results in its subsequent label-free identification by means of SERS spectroscopy. Using the proposed device, we demonstrate reliable SERS-based fingerprinting of various analytes, including common organic dyes and medical drugs at ppb concentrations. The proposed device is believed to find applications in various areas, including label-free environmental monitoring, medical diagnostics, and forensics.

We report on space-selective crystallization of congruent and polar Sr2TiSi2O8 crystals in a stoichiometric SrO-TiO2-SiO2 glass induced by (1030 nm, 300 fs) femtosecond laser irradiation. This allows us to compare with non-congruent laser induced crystallization of polar LiNbO3, in non stoichiometric Li2O-Nb2O5-SiO2 glass and gain information on the mechanism of nanocrystals orientation with the laser polarization that we pointed out previously. Using scanning electron microscopy (SEM), Second-harmonic generation (SHG) and electron backscattered diffraction (EBSD), we studied the laser induced crystallization according to the laser processing parameters (pulse energy, pulse repetition rate, scanning speed). We found 1) a domain where the laser track is filled with crystals not perfectly textured (low energy), 2) a domain where an amorphous volume remain surrounded by a crystallized shell (high energy). This arises from Sr out-diffusion and may gives rise to crystallization of both SrTiO3 and Sr2TiSi2O8 phases at low speed. In the one-phase domain (at higher speed), it is found the possibility to elaborate a tube with a perfect fresnoite texture. Significant difference in size and morphology whereas crystallization threshold remains similar are discussed based on glases thermal propeties. Contrarily to Li2O-Nb2O5-SiO2 (LNS) glass, no domain of oriented nanocrystallization controlled by the laser polarization has been found in SrO-TiO2-SiO2 (STS) glass, which is attributed to the larger crystallization speed in STS glass. No nanogratings have also been found that is likely due to the congruency of the glass.

Laser ablation of silicon under an external applied magnetic field with different orientations was investigated in respect to the scanning direction and polarisation of the laser beam, by observation of the ablation patterns and debris deposition. Ultra-short ∼230 fs laser pulses of 1030 nm wavelengths were used in the single and multi-pulse irradiation modes. Ablation with an externally applied magnetic B-field (B ≈ 0.1 T) is shown to strongly affect debris formation. The mechanism of surface plasmon polariton (SPP) wave can explain the ablated periodic patterns observed with alignment along the magnetic field lines.

We show broadband sum-frequency generation (SFG) in the Green-Yellow-Orange-Red spectral range using bismuth triborate, BiB3O6 crystal (BIBO) as nonlinear material. We perform a noncollinear phase-matching configuration within BIBO crystal using the remaining light behind the second harmonic generation stage and the infrared idler of an optical parametric amplifier (OPA). The obtained mixing radiation of ultrafast light sources to generate femtosecond pulses across 520.5 to 742.5 nm region is observed. SFG spectrum from single-pass cross-correlation intensity over such visible range is showed. The SFG wavelengths as a function of tunable wavelength idler OPA agrees with the expectations of parametric conversion condition and open the door to practical multi-beam or multi-color sum-frequency generators.

Micro Injection molding combined with the use of removable inserts is one of the most promising manufacturing process for microfluidic devices, such as Lab-on-a-chip, that have the potential to revolutionize the healthcare and diagnosis system. In this work we have designed, fabricated and tested a compact and disposable plastic optical stretcher. To produce the mould inserts, two micro manufacturing technologies have been used. Micro Electro Discharge machining was used to reproduce the inverse of the capillary tube connection characterized by high aspect ratio. Thanks to the high accuracy of femtosecond laser machining, instead, we manufactured insert with perfectly aligned microfluidic channels and fiber slots, facilitating the final composition of the optical manipulation device. The optical stretcher operation is tested using microbeads and red blood cells solutions. The prototype presented in this work demonstrates the feasibility of this approach that should guarantee a real mass production of ready-to-use- Lab-on-a-chip.

The Human immunodeficiency virus-1 (HIV-1) matrix (MA) domain is involved in the highly regulated assembly process of the virus particles that occur at the host cell’s plasma membrane. High-resolution structures of the MA domain determined using cryo X-ray crystallography have provided initial insights into the possible steps in the viral assembly process. However, these structural studies have relied on large and frozen crystals in order to reduce radiation damage caused by the intense X-rays. Here, we report the first XFEL study of the HIV-1 MA domain’s interaction with inositol hexaphosphate (IP6), a phospholipid headgroup mimic. We also describe the purification, characterization and microcrystallization of two MA crystal forms obtained in the presence of IP6. In addition, we describe the capabilities of serial femtosecond X-ray crystallography (SFX) using X-ray free-electron laser (XFEL) to elucidate the diffraction data of MA-IP6 complex microcrystals in liquid suspension at ambient temperature. Two different microcrystal forms of MA-IP6 complex both diffracted to beyond 3.5 Å resolution, demonstrating the feasibility of using SFX to study the complexes of MA domain of HIV-1 Gag polyprotein with IP6 at near-physiological temperatures. Further optimization of the experimental and data analysis procedures will lead to better understanding of the MA domain of HIV-1 Gag and IP6 interaction at high resolution and provide basis for optimization of the lead compounds for efficient inhibition of the Gag protein recruitment to the plasma membrane prior to virion formation.

Understanding cell migration is a key step to unravel many physiological phenomena and predict several pathologies, like cancer metastasis. In particular, mechanical confinement has been proved to be a key factor in the cellular migration strategy choice. As our insight in the field improves, new tools are needed in order to empower biologists’ analysis capabilities. In this framework, microfluidic devices have been used to engineer the mechanical stimuli and to investigate cellular migration response in a more controlled way. In this work, we will review the existing technologies employed in the realization of microfluidic cellular migration assays, namely soft lithography of PDMS and hydrogels and femtosecond laser micromachining. We will give an overview of the state of the art of these devices, focusing on the different geometrical configurations that have been exploited to study specific aspects of cellular migration. Our scope is to highlight the advantages and possibilities given by each approach and to envisage the future developments in in-vitro migration studies under mechanical confinement in microfluidic devices.

To harness light-matter interaction at nano-/micro-scale better control tools have to be developed. Here, it is shown that by applying external electric and/or magnetic field, ablation of Si and glass under ultra-short (sub-1~ps) laser pulse irradiation can be controlled via the Lorentz force $\mathbf{F} = e\mathbf{E} + e[\mathbf{v}\times\mathbf{B}]$, where $\mathbf{v}$ is velocity of charge $e$, $\mathbf{E}$ is the applied electrical bias and $\mathbf{B}$ is the magnetic flux density. The external electric E-field was applied during laser ablation using suspended micro-electrodes above glass substrate with an air gap for the incident laser beam. The counter-facing Al-electrodes on Si surface were used to study debris formation patterns on Si. Debris was deposited preferentially towards the negative electrode in the case of glass and Si ablation. Also, an external magnetic field was applied during laser ablation of Si in different geometries and is shown to affect ripple formation. The vectorial nature of the Lorentz force widens application potential of surface modifications and debris formation in external E-/B-fields with potential applications in mass and charge spectroscopes.

We report on the application of time-resolved inline digital holography to study the nonlinear optical properties of quantum dots deposited onto the sample glass. Fresnel diffraction patterns of the probe pulse due to noncollinear degenerate phase modulation induced by the femtosecond pump pulse were extracted from the set of inline digital holograms and analyzed. Absolute values of the nonlinear refractive index of both sample glass substrate and the deposited layer of quantum dots were evaluated using the proposed technique. To characterize the inhomogeneous distribution of samples’ nonlinear optical properties we proposed to plot an optical nonlinearity map calculated as a local standard deviation of diffraction patterns intensity induced by noncollinear degenerate phase modulation.
