Considering the large number of candidates in vaccine testing studies against different pathogens and the amount of time spent in the pre- and clinical trials, there is a pressing need to develop an improved in vivo system to quickly screen vaccine candidates. The model of a polyester-polyurethane sponge implant provides a rapid analysis of the specific stimulus-response, allowing the study of a compartmentalized microenvironment. The sponge implant’s defined measurements were standardized as a compartment to assess the immune response triggered by vaccinal antigen. The LBSap vaccine (composed by Leishmania braziliensis antigens associated with saponin adjuvant) was used in the sponge model to assess the antigen-specific immunological biomarker, including memory generation after initial contact with the antigen. Mice strains (Swiss, BALB/c, and C57BL/6) were previously immunized using LBSap vaccine followed by an antigenic booster performed inside the sponge implant. The sponge implants were assessed after 72 hours, and the immune response pattern was analyzed according to leukocyte immunophenotyping and cytokine production. After LBSap vaccination, the innate immune response of the antigenic booster in the sponge implants demonstrated higher levels in the Ly+ neutrophils and CD11c+ dendritic cells with reduced numbers of F4/80+ macrophages. Moreover, the adaptive immune response in Swiss mice demonstrated a high CD3+CD4+ T-cells frequency consisting of an effector memory component in addition to a cytoxicity response (CD3+CD8+ T-cells), displaying the central memory biomarker. The major cell surface biomarker in the BALB/c mice strain was related to CD3+CD4+ effector memory while the increased CD3+CD8+ effector memory was highlighted in C57/BL6. The cytokine profile was more inflammatory in Swiss mice, with the highest levels of IL-6, TNF, IFN-g, and IL-17, while the same cytokine was observed in in C57BL/6 yet modulated by enhanced IL-10 levels. Similar to Swiss mice, BALB/c mice triggered an inflammatory environment after the antigenic booster in the sponge implant with the increased levels in the ILL-6, TNF, and IFN-g. The results highlighted how genetic background can influence populations involved in the immune response and indicated that it is possible to use this model to optimize and monitor the innate and adaptive immune responses of vaccine candidates. In this sense, these results could guide the choice of the most appropriate experimental model in biomolecule tests, considering that the particularities of each mouse strain influenced the dynamics of the immune response.

Abstract: Flexible pressure sensors are electronic devices that can be attached to various irregular surfaces to measure the pressure. It is widely used in wearable devices and for health detection. In this study, a general carbon nanotubes/polydimethylsiloxane (CNTs/PDMS) sponge electrode was fabricated as the elementary component of the compressible system. The sensitivity of the flexible pressure sensor is improved by optimizing the preparation process of the conductive sponge and surface modification of the CNTs, and the highest sensitivity reached 71.831 kPa-1(0~2 kPa). The flexible pressure sensor can be used to detect subtle pressure quickly and accurately so that it can be used as wearable sensor to measure the radial artery wave of a human body, which has potential applications in health care and other fields.

A hemostatic sponge prototype was successfully synthesized from fish gelatin as an alternative to mammalian gelatin, which was mixed with alginate in certain combinations and double cross-linking of calcium ions and gamma irradiation at a dose of 20 kGy to improve the characteristics and effectiveness of its function as a local hemostatic agent. There was an improvement in the physicochemical and mechanical properties, porosity index, absorption capacity, biodegradation properties, biocompatibility, and hemocompatibility of the FGAS prototypes compared with the pure fish gelatin sponge. Hemostatic activity tests showed that the means of clotting time, prothrombin time, and activated partial thromboplastin time were shorter in the FGAS prototype than in the negative control, and there was no significant difference compared with the commercial gelatin sponge. The hemostatic mechanism of the FGAS prototype combined a passive mechanism as a concentrator factor and an active mechanism through the release of calcium ions as a coagulation factor in the coagulation cascade process.

Gastric perforations and leaks are significant complications that can arise from various gastrointestinal disorders and surgical interventions. Over the past decade, endoscopic techniques have emerged as effective method for managing these conditions. Furthermore, as endoscopic resection techniques have grown in popularity, the risk of acute and delayed perforations has also grown. This review aims to provide an in-depth analysis of the endoscopic management strategies employed in the management of gastric perforations and leaks. We will discuss the etiology, diagnostic modalities, and various endoscopic techniques utilized, and emerging trends in endoscopic management. Our attempt in writing this review paper is to educate and guide clinicians in making informed decisions when faced with gastric perforations and leaks.

Every petroleum processing industry produces sewage sludge containing several types of poly-cyclic aromatic hydrocarbon (PAHs) components. The degradation of PAH components by physical, biological and chemical methods is not efficient. The use of marine sponge symbiont bacteria is considered an alternative method in the degradation and reduction of PAHs com-pared to the previous method. This study aims to explore the potential and performance of a consortium of sponge symbiont bacteria in degrading anthracene and pyrene. There are three types of bacteria (Bacillus pumilus strain GLB197, Pseudomonas stutzeri strain SLG510A3-8, Acineto-bacter calcoaceticus strain SLCDA 976) were mixed to form a consortium. The interaction between the bacterial consortium suspension and PAH components was measured at 5-day intervals for 25 days. The biodegradation performance of bacteria on PAHs samples was determined based on five biodegradation parameters. The analysis results showed a decrease in the concentration of anthracene (21.89%) and pyrene (7.71%), equivalent to a ratio of 3: 1. The data was followed by a decrease in the abundance of anthracene (60.30%) and pyrene (27.52%), an equivalent ratio of 2: 1. The level of degradation of the pyrene component is lower than that of the anthracene compo-nent, presumably due to the higher toxicity of pyrene and the more stable molecular structure, making it difficult for bacterial cells to destroy it. The biodegradation products are organic compounds of alcohol, aldehyde, carboxylic acids and a small proportion of aromatic hydrocarbon components. Three-dimensional graphene (3DG) sponge has attracted increasing attention because it combines the unique properties of cellular materials and the excellent performance of graphene. The preparation of 3DG sponge depends mainly on the self-assembly of graphene oxide sheets. Here, we demonstrate facile fabrication of 3DG sponge with a large-scale and ordered porous structure, exploiting the liquid crystals of large graphene oxide (LGO) and ultralarge graphene oxide (ULGO) sheets. The resulting materials exhibit a low density, high porosity and elasticity. Our work explores a new strategy for organizing the ordered alignment of controlled large GO sheets and exploring the relationship between the microstructures and mechanical properties of 3DG sponge.

The practice of stormwater management has evolved from a singular focus on drainage to a multifaceted approach to support integrated urban development. By contributing to healthy, livable, ecological, and water sensitive cities, it is a key tool to promote Sustainable Development Goal 11 from neighborhood to metropolitan scales. A review of the knowledge base for stormwater management shows several attributes that favor an integrative approach to achieve co-benefits across several sectors. Functional areas of its contributions include drainage, flood control, flood plain management, water quality control, urban ecology, recreation, and city beautification. Legacy path dependance affects the potential to reform land use practices, while stormwater management practice is being affected by climate change, sea level rise, urbanization, inequality, and poor governance. Technical methods for stormwater management are well advanced, but integrative frameworks to address social, ecological and infrastructure needs are more challenging. The sensitivity of ecological issues is most evident in cities in coastal zones. Organizational initiatives are needed to counter neglect of essential maintenance and sustain flood risk reduction in cities. Stormwater management is related to other integrative tools, including IWRM, One Water, One Health, and integrated flood management, as well as the broader concept of urban planning. Stormwater capture and rainfall harvesting offer major opportunities to augment scarce water supplies. Nature-based solutions like low-impact development and the Sponge City concept show promise to transform cities. Major cities face challenges to sustain conveyance corridors for major flows and to store and treat combined sewer runoff. The neighborhood focus of stormwater management elevates the importance of participation and inclusion to advance environmental justice and strengthen social capital. Integrating organizational initiatives from local to city scale and funding improvements to stormwater systems are major challenges that require leadership from higher governance levels, although governments face resistance to change toward integration, especially in countries with poor land use and public works management systems. Finding solutions to neighborhood issues and the connectivity of water systems at larger scales requires complex approaches to urban planning and represent an important agenda for urban and water governance going forward.

The cavitating wake behind a wedge has been simulated employing both incompressible and compressible solvers with the purpose of comparing their performance. To develop the compressible solver, the ZGB cavitation model for incompressible flows has been modified to consider the compressibility effects. First, the equations of state for the vapor and liquid phases have been implemented. Then, a pressure limit and absorbing boundary conditions have been set to prevent a non-physical pressure field. To test the developed compressible solver, a series of verification tests have been conducted comprising a 1D sod cavitating tube and the cavitating vortex shedding behind a circular body at laminar flow conditions. After that, the incompressible and the validated compressible solvers have been applied to simulate the cavitating wake behind a wedge. The obtained results show similar predictions with the two solvers in terms of pressure, vortex shedding frequency and instantaneous and average vapor volume fraction profiles. Moreover, it is interesting to note that the components of the fluid force fluctuations on the body at the lowest frequencies, as predicted by both solvers, are indistinguishable. However, the higher frequency fluctuations of the forces tend to be better resolved and amplified when the compressibility is considered. Overall, both flow solvers have provided comparable cavitation phenomena, yielding results well aligned with experimental observations.

Hydroxytyrosol (HT) is a nutraceutic compound, mainly found in the fruit, leaves and waste from the olive oil industry, that shows one of the highest antioxidant activities among molecules of natural origin. To harness this bioactivity in cosmetics, pharmaceuticals, and the food industry, it is essential to modify the hydrophilicity of HT to enhance its compatibility with lipid-based mixtures. This chemical modification must be done with high selectivity to avoid compromising its radical scavenging activity. This work presents a highly efficient and selective approach to perform the biocatalytic esterification of free fatty acids (FFAs) of different alkyl chain length with HT in a reaction medium based on the SLIL [C12mim][NTf2]. By using a 1:2 (mol/mol) HT:FFA mixture of substrates, the HT monoester derivative was obtained up to 77% yield after 2 h at 80 oC. The adjusted molar ratio of substrates, combined with the ability to recover the SLIL for further reuse, significantly reduces waste accumulation compared to other reported strategies and results in a more sustainable approach as demonstrated by different green metrics. Furthermore, it was demonstrated by a DPPH test how the HT monoester products retain the same antioxidant activity as free HT, being superior to vitamin C.

Cyanobacteria are promising organisms for the sustainable production of various biotechnologi-cal interesting products. Due to their energy production via photosynthesis, the cultivation of cyanobacteria expands the CO2 cycle. Most cyanobacteria form biofilms on surfaces in their natu-ral environment by surrounding the cells with a self-produced matrix of extracellular polymeric substances (EPS) that hold the cells together. These special growth properties need special reac-tors for cultivation. By immobilizing cyanobacteria on carriers, systems currently established in industry could also be used for biofilm formers. Various artificial carriers for immobilized growth of cyanobacteria and microalgae have already been described in the literature. However, the use of waste materials or natural biodegradable carriers would be more sustainable and is, therefore, the focus of this study. Dried Luffa cylindrica, zeolite, and corn stalks were investigated for their use as carriers for cyanobacteria. L. cylindrica was shown to be an excellent natural carrier for (i) Anabaena cylindrica (ii) Nostoc muscorum 1453-12a, and (III) Nostoc muscorum 1453-12b. Higher or at least similar growth rates were achieved when cyanobacteria were cultivated with L. cylindrica compared to submerged cultivation. Additionally, the production of EPS and C-phycocyanin was increased at least 1.4-fold in all strains by culturing on L. cylindrica. The improved growth could be explained on the one hand by the high surface area of L. cylindrica and its properties, and on the other hand by the release of growth-promoting nutrients from L. cylindrica to the medium.

The most widely accepted scientific theory for the origin of life on Earth is that prokaryotic microbes evolved from simple organic compounds in seawater under anoxic conditions. For about 1 billion years thereafter, these microbes consumed the same dissolved organic matter (DOM) from which they had evolved before scarcity of DOM forced the evolution of cyanobacterial photosynthesis followed by eukaryosis. Could the more efficient consumption of DOM have also stimulated the subsequent origin of multicellular animal life? In this report, we synthesize past and recent evidence to propose the “DOM uptake hypothesis” for the origin of metazoans. A choanoflagellate-like protozoan was the likely ancestor of the first sponge-like metazoan to evolve on Earth. Choanoflagellates have outwardly facing flagellae that are subject to viscous water movement, while sponges have choanocytes in chambers with flagellae directed to pump water with greater fluidity across an aquiferous system with a huge cellular surface area. While generally considered particle feeders, both choanoflagellates and sponges absorb DOM, with some sponges relying on DOM for as much as 90% of their diet. We propose that the earliest metazoans may have evolved to survive the dire nutritional conditions of the Cryogenian “snow-ball Earth” period (~700 million years ago) by developing a body plan with the enhanced ability to absorb low concentrations of DOM in seawater from sources such as the viral lysis of microbes, exudates of benthic stromatolites, or refractory DOM compounds. Additionally, species of extant sponges that have a high abundance of microbes living in their bodies consume the greatest amounts of DOM, suggesting that the DOM uptake hypothesis may also be dependent on microbial symbiosis.

Fabrication of durable materials with excellent oil adsorption capacity and separation performance for the treatment of oily wastewater is meaningful based on smart responsiveness. Herein, a solvent-responsive melamine sponge (MS) was developed via silanization followed by the in-situ growth of zeolitic imidazolate framework-8 (ZIF-8). Detailed characterization of the resultant composite MS was conducted using scanning electron microscopy (SEM), energy dispersive spectrometer (EDS), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR). The multiscale hierarchical MS substrate exhibited highly hydrophobic property in the pH range of 1-11, along with a satisfactory adsorption capacity in the range of 65.4-134.2 g/g for different oils. The modified surface transformed from superhydrophobicity/superlipophilicity to superhydrophilicity/underwater superoleophobicity upon ethanol wetting, reverting to its original superhydrophobic state upon drying. The separation flux of the MS substrate was above 1.5×104L/m2h for both oil and water removal, along with the separation efficiency more than 98.7%. No obvious changes of the separation performance after 50 successive immiscible oil/water separation indicated the excellent durability and robustness of the anchored ZIF-8 nanoparticles on the surface of modified MS substrate. More importantly, oil-in-water emulsion separation was successfully carried out via the ZIF-8 MS composite with a high separation efficiency over 99.1%. The developed smart sponge with high oil absorption capacity, excellent chemical stability and fire resistance exhibited a wide range of potential practical applications in the convenient treatment of oily wastewater.

Abstract: Periodontal disease is the main reason for tooth loss in adults. Tissue engineering and regenerative medicine are the advanced technologies used to manage soft and hard tissue defects caused by periodontal disease. We developed a transforming growth factor-β3 chitosan sponge (TGF-β3/CS) to repair periodontal soft and hard tissue defects. We investigated the proliferation and osteogenic differentiation behaviors of primary human periodontal ligament stem cells (hPDLSCs) to discuss the bioactivity and application of TGF-β3 in periodontal disease. We separately used Calcein-AM/PI double-labeling or CM-Dil-labeling coupled with fluorescence microscopy to trace the survival and function of the cells after implantation in vitro or in vivo. The mineralization of osteogenic differentiated hPDLSCs was confirmed by measuring ALP activity and calcium content. The levels of COL I, ALPL, TGF-βRI, TGF-βRII, and Pp38/t-p38 were tested using Western blot to explore the mechanism of bone repair prompted by TGF-β3. When hPDLSCs were inoculated with different concentrations of TGF-β3/CS (62.5–500 ng/mL), ALP activity was the highest in TGF-β3 (250 ng/mL) group after seven days (P < 0.05 vs. control); the calcium content in each group increased significantly after 21 and 28 days (P < 0.001 vs. control). The best result was achieved in the TGF-β3 (500 ng/mL) group. All results showed that TGF-β3/CS can promote osteogenic differentiation of hPDLSC and may be involved in the p38 MAPK signaling pathway. TGF-β3/CS has the potential for application in the repair of incomplete alveolar bone defects.

The advancement in science and technology has led to luffa sponge (LS) being widely used as a natural material in industrial application as its polyporous structure and light texture. In order to enhance the utility of LS fibers as the reinforcement of lightweight composite materials, this study investigate its water absorption, mechanical properties, anatomical characteristic and thermal performance. Hence, moisture regain, tensile properties of LS fiber bundles were measured in accordance with standards and the structural characteristics were investigated via microscopic observation. Scanning electron microscopy (SEM) was used to observe the surface morphology and fracture surface of fiber bundles. Test results shows that the special structure where the phloem tissues degenerate to cavities had a significant influence on the mechanical properties of LS fiber bundles. Additionally, the transverse sectional area occupied by fibers in a fiber bundle (SF), wall thickness and ratio of wall to lumen of fiber cell, and crystallinity of cellulose had an impact on the mechanical properties of LS fiber bundles. Furthermore, the fiber bundles density of LS varies range of 385.46-468.70 kg/m³, much less than that of jute (1360.40 kg/m³) and Arenga engleri (950.20 kg/m³) while LS fiber bundles has superior specific modulus.

Wastewater treatment from oils and oil products, organic mixtures are a very relevant topic that can be successfully used to solve problems of serious environmental pollution, such as oil spills, industrial oily wastewater discharges and water treatment in the water treatment process. In this work, we have developed new superhydrophobic magnetic polyurethane (PU) sponges functionalized with reduced graphene oxide (RGO), MgFe2O4 nanoparticles, and silicone oil (SO) as a selective and reusable sorbent for purification and separation of wastewater from oils and organic solvents. The surface morphology and wettability of the sponge surface were characterized by scanning electron microscopy (SEM) and a contact angle analysis system, respectively. The results showed that the obtained PU sponge PU/RGO/MgFe2O4/SO had excellent mechanical and water-repellent properties, reusable properties (more than 20 cycles), as well as fast (immersion time 20 sec) and excellent adsorption capacity (16.61- 44.86 g / g), and good magnetic properties made it easy to separate the sponge from the water with a magnet. And the presence of RGO in the composition of the nanomaterial improves the separating and cleaning properties of materials, and also leads to an increase in the absorption capacity of oil and various organic solvents. The synthesized PU sponge has great potential for practical applications due to its facile fabrication and excellent oil-water separation property.

This study examines the recovery efforts at Gran Tarajal Harbor following a significant oil spill, employing innovative technologies. Initial response efforts were hampered by inadequate contingency plans and bureaucratic delays, underscoring the need for improved preparedness. Cleanup operations utilized advanced methods such as enhanced absorbent sponges, absorbent granulates, bioremediation with allochthonous species, and a combination of barriers and sponges. These technologies proved effective in minimizing environmental impact, reducing hydrocarbon concentrations in sediments from nearly 60,000 mg/kg to less than 1,600 mg/kg in seven months. Heavy metal analysis revealed lead and copper concentrations associated as well as with routine port activities, while mercury levels linked to the spill decreased over time. Tissue analysis indicated minimal contamination in local organisms, suggesting limited impact from the spill, and evaluation of the biological communities in sediments suggest an ecological recovery. Study underscores the importance of rapid response, advanced technologies, and detailed contingency plans tailored to each port. Despite the successful cleanup and ecosystem recovery, political and bureaucratic challenges remain significant obstacles to implementing these improved methods. The findings advocate for a re-evaluation of current oil spill response protocols to incorporate more effective, sustainable, and cost-efficient technologies, aligning with the principles of a circular economy.

The implementation of grey and green infrastructure is an effective mean to address urban flooding and nonpoint source pollution, but due to the complexity of the process and the diver-sity of benefits, there is a lack of measurement for their comprehensive benefits. Adopting a typ-ical university in Beijing as an example, this paper simulated the multidimensional benefits of the water quantity, water quality, and ecology of grey and green facility renovation by coupling the storm water management model (SWMM) and InfoWorks Integrated Catchment Manage-ment (ICM). Monetization methods and economical means were employed to characterize the comprehensive benefits. The results showed that grey and green infrastructure retrofitting re-duces the number of severe overflow nodes in the study area by 54.35%, the total overflow vol-ume by 22.17%, and the nonpoint source pollution level by approximately 80% under the heavy rain scenario and 60% under the rainstorm scenario. The annual benefits of grey and green infra-structure renovation reached ¥765,600/year, of this amount, ¥83,100/year is from hydrological regulation, ¥454,100/year is from nonpoint source pollution reduction, and ¥228,300/year is from ecological improvement. The benefits of green facilities were higher than those of grey facilities, and the combined benefits were negatively correlated with the rainfall level, with a total bene-fit–cost ratio of 1.19. The results provide methodological and data support for grey and green in-frastructure retrofitting within the context of sponge cities.

Stormwater management is a key urban issue in the world, in line with the global issues of urban sprawl and climate change. It is urgent to investigate the effectiveness in managing stormwater with different strategies for maintain urban resilience. A method based on a storm water management model (SWMM) was developed for assessing the control of stormwater runoff volume and the percentage removal of suspended solids by implementing a Sponge City strategy. An interdisciplinary approach was adopted incorporating Low Impact Development (LID) with urban Green Infrastructure and Gray Infrastructure paradigms in a typical old residential community in Suzhou, China. Four types of sponge facilities for reducing stormwater runoff were bio-retention cells, permeable pavements, grassed pitches, and stormwater gardens. The simulation results indicate that the stormwater pipe system can meet the management standard for storms with a five-year recurrence interval. The volume capture ratio of annual runoff is 91% and the reduction rate of suspended solids is 56%. This study demonstrates that Sponge City strategy is an effective approach for managing stormwater, particularly in old and densely populated urban areas. Implementing spongy facilities with a LID strategy for stormwater management can significantly enhance urban water resilience and increase ecosystem services.

Abstract: Palynomorph assemblages and mineral microfossils from five southern Red Sea coastal environments were studied to demonstrate their distribution pattern and suggest their source. The environments studied were 1. mangrove swamp 2. middle Holocene paleochannel 3. algal mat, 4. Sabia Island coral reef, and intertidal zone. The samples studied were from the top 3 cm semi-consolidated sediments from various environments. Standard palynological maceration technique was used to isolate palynomorphs. These sediment samples are mainly clay and fine sand and yielded low numbers but high diversity of palynomorph assemblages. They are grouped into pollen and spores, dinoflagellate cysts, and algal remains, fungal spores, hyphae and fruit bodies, protists, and invertebrates. A few Ascidian and sponge spicules, and phytoliths (mineral microfossils) occur in the Sabia Island coral reef environment. The protists and invertebrate remain include microforaminifera, thecamoebians, tintinnomorphs, crustacean and annelid palynomorphs. The assemblage also includes six form taxa belonging to the form genera Katora and Mangrovia. These palynomorphs belong to both marine and terrestrial environments and are of autochthonous and allochthonous origins. The source and genetic relationship of each palynomorphs is discussed.

Ultra-low radioactive titanium alloy is a promising material for the manufacture of low-background detectors, which are being developed for experiments in astroparticle physics and neutrino astrophysics. Structural titanium is manufactured on an industrial scale from titanium-sponge. The ultra-low-background titanium-sponge can be produced on an industrial scale with a contamination level of less than 1 mBq/kg of uranium and thorium isotopes. The pathways of contaminants during the industrial production of structural titanium have been analyzed. The measurements have been carried out using two methods: inductively coupled plasma spectroscopy (ICP-MS) and gamma-spectroscopy using high-purity germanium detectors (HPGe). It has been shown that the level of contamination with radioactive impurities does not increase during remelting of titanium-sponge and mechanical processing. We examined titanium-alloys samples obtained at different stages of titanium production, namely, an electrode compaction, a vacuum arc remelting with a consumable electrode (VAR), and a cold rolling of titanium-sheets. It has been found out that all studied samples will be a source of uranium and thorium contamination of the final titanium-alloys. It has been established that the only product allowed obtaining ultra-low-background titanium was the VT1-00 which is manufactured without master alloys addition..

Biological nanoparticles (NPs), such as extracellular vesicles (EVs), exosome-mimetic nanovesicles (EMNVs) and nanoghosts (NGs), are perspective non-viral delivery vehicles for all types of therapeutic cargo. Biological NPs are renowned for their exceptional biocompatibility and safety, alongside their ease of functionalization, but a significant challenge arises when attempting to load therapeutic payloads, such as nucleic acids (NAs). One effective strategy involves fusing biological NPs with liposomes loaded with NAs, resulting in hybrid carriers that offer the benefits of both biological NPs and the capacity for high cargo loads. Despite their unique parameters, one of the major issues of virtually any nanoformulation is the ability to escape degradation in the compartment of endosomes and lysosomes which determines the overall efficiency of nanotherapeutics. In this study, we fabricated all major types of biological and hybrid NPs and studied their response to acidic environment observed in endolysosomal compartment. In this study, we show that EMNVs display increased protonation and swelling relative to EVs and NGs in an acidic environment. Furthermore, the hybrid NPs exhibit an even greater response compared to EMNVs. Short-term incubation of EMNVs in acidic pH corresponding to late endosomes and lysosomes again induces protonation and swelling, whereas hybrid NPs are ruptured, resulting in the decline of their quantities. Our findings demonstrate that in an acidic environment, there is enhanced rupture and release of vesicular cargo observed in hybrid EMNVs that are fused with liposomes compared to EMNVs alone. This was confirmed through PAGE electrophoresis analysis of mCherry protein loaded into nanoparticles. In vitro analysis of NPs colocalization with lysosomes in HepG2 cells demonstrated that EMNVs mostly avoid endolysosomal compartment whereas hybrid NPs escape it over time. To conclude, (1) hybrid biological NPs fused with liposomes appear more efficient in the endolysosomal escape via the mechanism of proton sponge associated with scavenging of protons by NPs, influx of counterions and water, and rupture of endo/lysosomes, but (2) EMNVs are much more efficient than hybrid NPs in actually avoiding endolysosomal compartment in human cells. These results reveal biochemical differences across four major types of biological and hybrid NPs, and indicate that EMNVs are more efficient in escaping or avoiding endolysosomal compartment.

Marine sponges, a well documented prolific source of natural products, harbors numerous microbial communities believed to possess N-acyl homoserine lactones (AHLs) mediated Quorum sensing (QS) as one of the mechanisms of interaction. Bacteria and eukaryotic organisms are known to produce molecules that can interfere with QS signaling, thus affecting microbial genetic regulation and function. In the present study, we established the potential for production of both QS signal molecules as well as QS interfering molecules (QSI) in the same sponge species Sarcotragus spinosulus. A total of eighteen saturated acyl chain AHLs were identified along with six putative unsaturated acyl chain AHLs. Bioassay guided purification led to the isolation of two brominated metabolites with QS-interfering activity. The structures of these compounds were elucidated by comparative spectral analysis of 1HNMR and HR-MS data and was identified as 3-Br-N-methyltyramine (1) and 5,6-dibromo-N,N-dimethyltryptamine (2). The QSI activity of compounds 1 and 2 were evaluated using reporter gene assays for long- and short-chain signals (E. coli pSB1075 and E. coli pSB401) and was confirmed by measuring dose dependent inhibition of proteolytic activity and pyocyanin production in P. aeruginosa PAO1. The obtained results showed the co-existence of QS and QSI in S. spinosulus, a complex network which may mediate the orchestrated function of the microbiome within the sponge holobiont.

As the increase threat of flood risk and environmental safety due to the urbanization, Sponge city research has been attracting extensive attention both in practical and theoretical research field. To date, there are only scattered studies about Sponge city. Moreover, vary names of Sponge city prevalent in different countries, which leads to disconnection of literature in the same field of Sponge city. In this paper, a thorough systematic literature mining of Sponge city is presented. A literature analysis system is created, which includes literature export from Web of Sciences and systematic analysis via NoteExpress and CiteSpace. Some literature statistical results are derived. Challenges and opportunities for future research are anticipated. Our goals are to promote this promising thought, summarize past research, and identify issues for future research to create impacts on the practice of Sponge city.
