In this work, PMMA(Poly methyl methacrylate) microfluidic system was used as a micro-reactor for urea hydrolysis by urease enzyme with use conductivity principle and utilize sound level meter(SLM) App in smartphone as a novel detector by considering the peak height in the App as an indicate for urea concentration. the advantage of use small volume and how the reaction carried on in the microfluidic system with simple and low cost are discussed, and the results were analyzed and statically determine. The linearity, detection limit (3×noise )and Correlation Coefficient, 62.5-500 ppm, 31.25 ppm and 0.992 respectively also, recovery studied were between(98.5-100.13%).

Immobilization of enzymes is a good field of study to expand the life of enzyme in and lowering the cost of the chemical processes such as separation processes. Urease is an important enzyme with medical and industrial applications. The aim of the present study is to prepare an immobilized urease on a strong cation exchange resin (Amberlite IR120 -Na) and study of its activity and stability. We monitored the liberation of Na ions in the collected fractions and searching for protein in the fractions as an indicators of immobilization by ion exchange phenomenon. Sodium is measured using atomic absorption spectroscopy techniques, while protein tested by Bradford’s method. Immobilized urease activity was evaluated by salicylate-hypochlorite method. The results indicated a complete immobilization of urease enzyme on the resin surface with reserving 92% of the activity of free enzyme. The immobilized urease enzyme on resin showed good stability and it has a 62% of its activity after 154 days of storage at room temperature. It is concluded that a new immobilized urease enzyme system is prepared with good enzyme activity and stability.

Development of carbon nanomaterials for adsorption thus removal of organic pollutants from water is a progressive research subject. In this regard, carbon nanomaterials with bifunctionality towards polar and non-polar or even amphiphilic undesired materials is indeed attractive for further study and implementation. Here, we created carbon buckypaper adsorbents comprising amphiphilic (oxygenated amorphous carbon (a-COx)/graphite (G)) nanofilaments that can dynamically adsorb organic biomolecules (i.e. urease enzyme) and thus purify the wastewaters of relevant industries. Given the dynamic conditions of the test, the adsorbent was highly efficient in adsorption of the enzyme (88%) while permeable to water (2382 L.h-1.m-2), thus holds a great promise for further development and upscaling. A subsequent citric acid functionalization declined selectivity of the membrane to urease, implying the biomolecules adsorb mostly via graphitic domains rather than oxidized, polar amorphous carbon ones. The devised platform i.e. the urease functionalized buckypaper is optimally conductive (13 S.cm-1) and can be further employed as a biosensor. Accordingly, water treatment can be linked to biosensing via a nanostructured membrane.

Cryptococcus neoformans is a facultative intracellular fungal pathogen. Ten-generation-old (10GEN) C. neoformans cells are more resistant to phagocytosis and killing by macrophages than younger daughter cells. However, mechanisms that mediate this resistance and intracellular parasitism are poorly understood. Here, we identified factors important for the intracellular survival of 10GEN C. neoformans, such as urease activity, capsule synthesis, and DNA content using flow cytometry and fluorescent microscopy techniques. Real-time visualization of time-lapse imaging was applied to determine the phagosomal acidity, membrane permeability, and vomocytosis (non-lytic exocytosis) rate in J774 macrophages that phagocytosed C. neoformans of different generational ages. Our results showed that old C. neoformans exhibited higher urease activity, and enhanced capsule formation, which was supported by an increased Golgi complex. In addition, old C. neoformans were more likely to be arrested in the G2 phase, resulting in the occasional formation of aberrant trimera-like cells. To finish, the advanced generational age of the yeast cells slightly reduced vomocytosis events within host cells, which might be associated with increased phagolysosomal pH and membrane permeability. Altogether our results contribute to a better understanding of the strategies used by old C. neoformans to resist phagosomal killing and suggest that old generation C. neoformans intracellular behavior may be driving cryptococcosis pathogenesis.

Our study aimed to assess sprinkler irrigation's impact on the activity of selected soil enzymes concerning nitrogen metabolism and oxidation-reduction processes in the soil with different doses of inorganic nitrogen fertilizers. The soil Alfisol was sampled from the experimental field of spring barley in the University Research Center in the central part of Poland in the moderate transitory climate during the growing seasons 2015–2017. The index resistance (RS) was derived to recognise the resistance enzymes of drought. In the maturity phase, nitrate reductase activity was at an 18% higher level in irrigated soils and the activity of other enzymes was higher in no-irrigated treatments by 25% in dehydrogenase, 22% in peroxydase 33% in catalase and 17% in urease case. The development stage of barley influenced nitrate reductase activity. Enzymatic activity has been changing in the examined years of the research, depending on the weather conditions. During the maturity stage, the soil's lower ammonium nitrogen content resulted from higher spring barley uptake due to drought stress. Irrigation probably contributes to increased leaching of nitrate in soil. The highest index of resilience was presented in the soil catalase activity.

Ureaplasma species (Ureaplasma spp.) are commonly found as commensals in the human urogenital tracts , but their overgrowth can lead to infection in the urogenital tract and at distal sites. Treatment of Ureaplasma spp. infections is difficult. The lack of a cell wall confers resistance to all β-lactam and glycopeptide antibiotics whereas the lack of de novo synthesis of folic acid makes them resistant to sulfonamides and diaminopyrimidines. Thus, there is a need to search for new drugs effective against these bacteria. In the presented studies, the effect of N,N-diethyldithiocarbamate (DDC) at a concentration of 2 µg/ml, 20 µg/ml and 200 µg/ml on the growth and survival of Ureaplasma urealyticum and Ureaplasma parvum was tested. Our results indicated that all DDC concentrations used showed both bacteriostatic and bactericidal activity against both tested strains In brief, for the first time we showed the bacteriostatic and bactericidal activity of DDC against Ureplasma urealyticum and Ureaplasma parvum. Based on literature data, we conclude that this DDC activity is related to the inhibition of urease activity, which is an indispensable enzyme for these bacteria. Further research is needed and the results obtained in the present study should be verified in in vivo models of infection.

Ten new Schiff bases, including thiocarbohydrazone derivatives (1–10), were synthesized using an efficient, uncomplicated, and environmentally friendly approach using the reaction of alkyl-substituted aldehydes. The thiocarbohydrazones that were obtained were derived from o-phthalaldehyde. The chemical structures of the compounds were identified by the utilization of UV-Vis spectroscopy, Fourier transform infrared (FT-IR), ¹H nuclear magnetic resonance (¹H NMR), ¹³C nuclear magnetic resonance (¹³C NMR), and elemental analysis methods. The thermal stability of thiocarbohydrazones was examined using thermogravimetric analysis (TGA-DSC). Investigations were conducted on urease inhibition tests, pharmacokinetic property research, and molecular docking. The collection of samples with the urease enzyme revealed significant molecular docking findings, with MolDock scores ranging from -153 to -189. The compounds have the potential to be effective drug candidates for treating certain diseases, as indicated by the consistent results of molecular docking, urease inhibition tests, and pharmacokinetic characteristics investigations.

A portable urea sensor for use in the fast flow condition was fabricated using porous polytetrafluoroethylene (PTFE) membranes coated with amine-functionalized parylene, parylene-A, by vapor deposition. To generate a specific electrochemical sensor signal from urea, the urea-hydrolyzing enzyme urease was immobilized on the parylene-A-coated PTFE membranes via chemical crosslinking using glutaraldehyde. The urease-immobilized membranes were assembled in a polydimethylsiloxane (PDMS) fluidic chamber, and a screen-printed carbon three-electrode system was used for electrochemical measurements. The success of urease immobilization was confirmed using fluorescence microscopy, scanning electron microscopy, and Fourier-transform infrared spectroscopy. The optimum concentration of urease for immobilization on the parylene-A-coated PTFE membranes was determined to be 48 mg/mL, and the optimum number of membranes in the PDMS chamber was found to be 8. Using these optimized conditions, we fabricated the urea biosensor and monitored urea samples under various flow rates ranging from 0.5 to 10 mL/min in the flow condition using chronoamperometry. To test the applicability of the sensor for physiological samples, we used it for monitoring urea concentration in the waste peritoneal dialysate of a patient with chronic renal failure, at a flow rate of 0.5 mL/min.

The soil microbial community serves as a crucial indicator for assessing soil fertility and health. The composition and activity of soil microorganisms reflect the soil's well-being and its ability to provide proper nutrients for crops. Fertilization plays a prominent role in enhancing soil fertility, and novel types of fertilizers, such as stabilized fertilizers with nitrification/urease inhibitors and coated fertilizers, have gained popularity in rice and maize cultivation. However, the long-term effects of these stabilized and coated urea fertilizers on soil chemical properties, microbial di-versity and community structure, as well as nitrogen cycling functional genes, remain unclear. Therefore, it is essential to investigate the impact of extended use of stabilized and coated urea fertilizers on soil fertility and microbiota in rice paddy fields. This research will provide scientific and theoretical support for the development and promotion of stabilized and coated urea ferti-lizers. To examine the effects of long-term application of these stabilized and coated urea fertilizers on soil chemical properties and microorganisms in rice paddy fields, soil samples were collected from brown soil rice paddy fields treated with various urease and nitrification inhibitors, stable urea, sulfur-coated urea (SCU), and resin-coated urea (PCU). The study revealed that 16 years of long-term use of conventional urea nitrogen fertilizer led to a considerable reduction in soil TP. On the other hand, NBPT and conventional urea fertilizers, when applied for an extended period, significantly increased soil organic matter (SOM). Moreover, except for HQ and NBPT+DMPP, the prolonged application of new urea fertilizers also signifi-cantly enhanced soil total potassium (TK). Notably, among all the treatments, PCU treatment had higher values for various soil chemical properties. In the case of SCU fertilizer used in brown soil rice paddy fields, it resulted in a significant decrease in soil pH over time. However, this change in pH did not affect the population of ammonia-oxidizing bacteria (AOB), as it was primarily in-fluenced by soil available nitrogen. DMPP, HQ+DCD, NBPT+DMPP, SCU, and PCU significantly reduced the copy number of bacterial 16S rRNA genes in the soil, with the coated urea fertilizers (SCU and PCU) having a greater impact. The long-term use of stabilized urea fertilizers containing HQ significantly reduced the bacterial community in rice paddy soil. Conversely, HQ+DCD stable urea fertilizer significantly increased the population structure and abundance of Basidiomycota fungi while decreasing the population structure and abundance of Rozellomycota fungi. DMPP stabilized urea fertilizer notably increased the population structure and abundance of Ascomycota fungi while decreasing the population structure and abundance of Rozellmycota and Chytridiomycota fungi. Further-more, HQ stabilized urea fertilizer significantly reduced the population structure and abundance of Chytridiomycota fungi. It is important to note that SCU fertilizer is not suitable for long-term application in neutral to slightly acidic meadow brown soil with a background pH. On the other hand, stable urea fertilizers and resin-coated urea fertilizers containing HQ, NBPT, DCD, DMPP, and their combinations are suitable for long-term application in neutral to slightly acidic meadow brown soil.

Alzheimer’s disease (AD) causes dementia and memory loss in the elderly. Deposits of beta-amyloid peptide and hyperphosphorylated tau protein are present in AD’s brain. A filtrate of Helicobacter pylori’s culture was previously found to induce hyperphosphorylation of tau in vivo, suggesting that bacterial exotoxins could permeate the blood brain barrier and directly induce tau’s phosphorylation. H. pylori, which infects ~60% of the world population and causes gastritis and gastric cancer, produces a pro-inflammatory urease (HPU). Here the neurotoxic potential of HPU was investigated in cultured cells and in rats. SH-SY5Y neuroblastoma cells exposed HPU (50-300 nM) produced reactive oxygen species (ROS) and had an increased [Ca2+]i. HPU-treated BV-2 microglial cells produced ROS, cytokines IL-1β and TNF-α, expressed Iba1 and showed reduced viability, consistent with a neurotoxic effect of HPU. Rats received daily i.p. HPU (5 µg) for 7 days. Hyperphosphorylation of tau at Thr205, Ser199 and Ser396 sites was seen in hippocampal homogenates of treated rats, with no alterations in total tau or GSK-3b levels. HPU was not detected in the brain homogenates. Behavioral tests were performed to assess cognitive impairments. Our findings support previous data suggesting an association between infection by H. pylori and tauopathies such as AD, possibly mediated by its urease.
