Hepatocellular Carcinoma is one of the most predominant malignancies with high fatality rate and is rising at an alarming rate because it is quite resistant to radioand chemotherapy. The proliferation, migration and activation of endothelial cells are involved in tumor occurrence and development. Indirubin is the major active anti-tumor ingredient of a traditional Chinese herbal medicine. In this research, Td-ECs were derived from human umbilical vein endothelial cells (HUVECs) by treating HUVECs with the conditioned medium of human liver cancer cell line HepG2. The effects of indirubin on cell proliferation, migration, invasion and angiogenesis of Td-ECs were assessed. Indirubin significantly inhibited Td-EC cell proliferation in a dose- and time-dependent manner. Indirubin also inhibited Td-EC migration and angiogenesis. However, indirubin’s effects on HUVECs were weaker than on Td-ECs.

Endometrial cancer (EC) is the most frequent gynaecological malignancy. The ESGO/ESTRO/ESP 2020 guidelines identify prognostic groups based on morpho-molecular characteristics. This study aims to evaluate the clinical applicability of NGS analysis to define an appropriate risk class and application for a better diagnostic and prognostic stratification of ECs. Cases of serous carcinoma (OHEC), high (HGEC) and low (LGEC) grade endometrioid carcinoma diagnosed with the morphological and immunohistochemical (IHC) protocols were considered. After a standardized pre-analytical phase, the tumor DNA was semi-automatically extracted and analyzed by NGS with a panel of 14 genes. A total of 63 cases were considered. NGS analysis was successful in 60 cases; all of these were classified according to the new diagnostic algorithm. The molecular risk classification showed a good correlation with the morphological (k=0.8). The study showed that the protocols of the pre-analytical and analytical phases used are robust and can lead to molecular results that fall within the standards required for use in clinical practice for a more precise diagnostic-therapeutic management of patients. The implementation of the classification is particularly relevant for better prognostic stratification of HGECs. In addition, the identification of a suspicious VUS in POLE questions the classification of truncating variants.

Soil salinity is a severe soil degradation problem mainly faced in arid and semi-arid regions. About 11 million ha of land in the arid, semi-arid, and desert parts of Ethiopia is salt-affected, especially in the Awash River basin, including Afambo irrigated area. Remote sensing approaches are significant tools for accurately predicting and modeling accurately predicting and modeling soil salinity in various world regions. This study aims to analyze and model soil salinity status in the case of Afambo irrigated areas using Landsat-8 and sentinel-2A, Afar region, Ethiopia, by applying remote sensing with field measurements. Thirty-two soil samples were collected from the topsoil (0-30 cm); out of these, 25 soil samples with various EC ranges were selected for modeling, and the remaining 7 samples were utilized to validate the model. Landsat-8 and Sentinel-2A images acquired in the same month were used to extract soil salinity indices. Linear regression analyses correlated the EC data with corresponding soil salinity spectral index values derived from satellite images. The best-performing model was selected for salinity mapping. The soil salinity indices extracted from both Landsat-8 and Sentinel-2A bands estimated soil salinity with high acceptable accuracy of R² values of SI, 0.78 and 0.81, respectively. The model results in three salinity classes with varying degree of salinity, namely, highly saline, moderately saline, and slightly saline, which covers 15.1%, 39.8% and 45.1% of the total area for Landsat-8, respectively and 26.1%, 32%, and 41.9% for sentinel 2A, respectively. Generally, the results revealed that the expansion rate of salt-affected soils has been increasing. From this study, it is possible to infer that if the present irrigation practice continues, it is expected that total the cultivated lands will become sterile within a short period. Thus, it needs to be monitored regularly to secure up-to-date knowledge of their extent to improve management practices and take appropriate actions.

Due to the high effectiveness of cancer screening and therapies, the diagnosis of second primary cancers (SPCs) has increased in women with endometrial cancer (EC). However, there’s no previous literature mentioned about adequate evidence to support screening for SPCs in endometrial cancer. This study was aimed to develop effective risk prediction models of second primary endometrial cancer in women with obesity (Body-mass index; BMI > 25) and this study includes datasets of the incidence of SPCs and the other risks of SPCs in 4480 primary cancer survivors by a hospital-based cancer registry database. In our study, we found the obesity played a key role in SPCs. There’re 10 independent variables used as predicting variables, which corelated to obesity should be monitored for the early detection of SPCs in endometrial cancer. In conclusion, it is a promising SPCs prediction. The proposed scheme can support the important influence of obesity and clinical data representations in all cases after primary treatments. Our results suggested that obesity is still a crucial risk factor to SPCs in endometrial cancer.

In order to investigate the size distributions and seasonal variations of carbonaceous aerosols (OC and EC), the carbonaceous species were collected and then analyzed by using a 9-stage Anderson-type aerosol sampler and DRI Model 2001A Thermal/Optical Carbon Analyzer on the typical industrial city Nanjing in Yangtze River Delta, China in the summer, autumn and winter of 2013 and spring of 2014. OC, EC, SOC and POC exhibited obvious seasonal variations, with the highest level in winter (39.1±14.0, 5.7±2.1, 23.6±11.7 and 14.1±5.7 μg•m^-3) and the lowest level in summer (20.6±6.7, 3.3±2.0, 12.2±3.8 and 8.4±4.1 μg•m^-3), and were mainly centralized in PM_1.1 in four seasons. The concentrations of OC in PM_1.1 varied in the order of winter > autumn > spring > summer, while EC ranked in the order of autumn > winter > summer > spring. In the PM_1.1-2.1 and PM_2.1-10, the concentrations of OC and EC decreased in the sequence of winter > spring > autumn > summer. The size spectra of OC, EC and SOC had bimodal distributions in four seasons, except for EC with four peaks in summer. The size spectra of POC varied greatly with seasons, exhibiting bimodal distribution in winter, trimodal distribution in spring and summer, and four peaks in autumn. The OC/EC ratios were 7.0, 6.3, 7.6 and 6.9 in spring, summer, autumn and winter, respectively, which demonstrated the abundance of secondary organic aerosols in Nanjing. The sources of carbonaceous aerosol varied significantly with seasons, and were dominated by vehicle exhaust, coal and biomass burning in PM_2.1, and dominant by dust, coal and biomass burning in PM_2.1-10.

The occurrence of ovarian dysfunction is often due to the imbalance between the formation of reactive oxygen species (ROS) and the (in)effectiveness of the antioxidative defense mechanisms. Primary sources of ROS are the respiratory electron transfer and the activity of NADPH oxidases (NOX), while Superoxide dismutases (SOD) are the main key reg-ulators that control the levels of ROS and reactive nitrogen species intra- and extracel-lularly. Because of their central role, SODs are the subject of research on human ovarian dysfunction, but the sample acquisition is low. The high degree of cellular and molecular similarity between Drosophila melanogaster ovaries and human ovaries provides this model organism with the best conditions for analyzing the role of ROS during ovarian function. In this study, we clarify the localization of the ROS producing enzyme dNox within ovaries of Drosophila melanogaster and by a tissue specific knockdown, we show that dNox derived ROS are involved in the chorion hardening process. Furthermore, we analyze the dSod3 localization and show that reduced activity of dSod3 impacts egg laying behavior, but not the chorion hardening process.

The broad and rapid application of new genomic techniques (NGTs) such as CRISPR-Cas has led to a fast increase in the number of genetically modified crops. With the recently published EU Commission’s (EC) regulatory proposal on plants generated with NGTs (July 5, 2023) the process of admission of such genetically modified plants is aimed to get accelerated and simplified. NGT plant applications shall be categorized based on their molecular characteristics into category 1 NGT (NGT1) and category 2 NGT (NGT2), which is in divergence from the current legislation centered around Directive 2001/18/EC. In the regulatory proposal NGT2 plants are envisioned to undergo a reduced risk assessment while for NGT1 plants no risk assessment is foreseen at all. To demonstrate were the path of the proposal leads to in practice, we applied the proposed criteria for categorization to a list of NGT plant applications that are currently in the commercialization pipeline. A key finding is that out of those plant applications affected by the EC proposal, 94% would be classified as NGT1 and thus would receive a market approval without risk assessment, monitoring and sufficient labeling provisions. The remaining 6% of applications would be classified as NGT2 plants for which, in deviation from the current regulation, an adapted risk assessment is proposed. A screening of intended traits in the pipeline highlights that certain NGT1 plants can pose similar environmental risks (e.g. invasiveness) compared to other genetically modified organisms (GMOs), as defined in Directive 2001/18/EC. For example, NGT1 applications based on RNAi technology can exhibit insecticidal effects with potential side effects on non-target organisms (i.e. other insects). Our quantitative and case-specific elaboration on how the current EC regulatory proposal would affect the environment, health or consumer protection will be informative for decision makers and politicians.

Oxidative stress (OS) is reported in several chronic diseases. Extra-cellular superoxide dismutase (ec-SOD) catalyzes the dismutation of superoxide anion with a protective role on endothelial cells. In chronic kidney diseases (CKD), OS and thyroid dysfunction (low T3 syndrome) are fre-quently present, but their relationship has not been investigated. This cohort study evaluated ec-SOD activity in CKD patients during haemodialysis, divided into: “acute hemodialytic pa-tients” (AH, 1-3 months of treatment) and “chronic hemodialytic patients” (CH, treated for a longer period). We also evaluated plasmatic total antioxidant capacity (TAC) and the relation-ships with thyroid hormones. Two basal samples (“basal 1”, 3 days after the last dialysis and “basal 2”, 2 days after the last dialysis) were collected. On the same day of basal 2, a sample was collected 5 and 10 min after the standard heparin dose and at the end of the procedure. The ec-SOD values were significantly higher in CH vs AH in all determinations. Moreover, they pre-sented lower TAC values. Dividing CH patients into two subgroups according to fT3 levels (nor-mal or low) we found significantly lower ec-SOD values in the group with low fT3 at basal, 5 and 10 min samples. Finally, a significant correlation was observed between fT3 and ec-SOD in the basal 1 samples. These data, confirming OS and low T3 syndrome in CKD, suggest that low fT3 concentrations can influence ec-SOD activity and therefore contribute to possible endothelial oxi-dative damage in such a situation.

Controlled drainage (CD) is an important agricultural measure for maintaining soil moisture and nutrients, controlling groundwater level, and increasing crop yield. In arid regions, CD can be used to improve the water supply in agriculture and reduce environmental pollution. In this study, we investigated the effect of CD, including a drainage depth of 40 cm (CWT1) and 70 cm (CWT2) during the plant growth period, free drainage (FD), and open ditch drainage (OD) on the migration of water, nutrients, and salts in the soil; the dynamics of groundwater level; the loss of soil nitrogen; and the growth of oilseed sunflower plants. Compared with FD, CD increased the water and nutrient content in the soil, reduced nitrogen loss, and enhanced the ability of the soil to continuously supply nitrogen to the oilseed sunflower plants, which benefited plant growth at later growth stages and reduced environmental pollution. During the period between irrigation at the budding stage and harvest stage, the average soil water content in the 0–20 cm soil layer in CWT1 increased by 3.67%, 4.78%, and 0.55%, respectively, compared with that in CWT2, FD, and OD. The soil mineral content in CWT1 was 25.17%, 35.05%, and 17.78% higher than that in CWT2, FD, and OD, respectively, indicating that higher soil salinity occurred at the later stage of plant growth in CWT1, which actually had little effect on the plants due to their enhanced salt tolerance and increased need for water and nutrients at that stage. In addition, CD delayed the decline in groundwater level, which allowed the plants to use groundwater at later growth stages, and as a result the yield and water use efficiency were improved. CWT1 significantly increased oilseed sunflower yield by 4.52–11.14% and increased water use efficiency by 1.16–10.8%. Moreover, CWT1 also increased the survival rate of the oilseed sunflower plants by 2.62–2.92%, and the plants demonstrated good growth. Therefore, under CD conditions, plants used soil water and nitrogen more efficiently and, as a result, their productivity was increased, and the water quality was improved.

The sea level rise, a consequence of climate change, is one of the biggest challenges that countries and regions with coastal lowland areas will face in the medium term. This study proposes a methodology for assessing the vulnerability to sea level rise (SLR) on the Atlantic coast of Portugal mainland. Some scenarios of extreme sea level for different return periods and extreme flooding events were estimated for 2050 and 2100, as proposed by the European Union Directive 2007/60/EC. A set of physical parameters are considered for the multi-attribute analysis technique implemented by the Analytic Hierarchy Process, in order to define a Physical Vulnerability Index fundamental to assess coastal vulnerability. For each SLR scenario, coastal vulnerability maps, with spatial resolution of 20 m, are produced at national scale to identify areas most at risk of SLR, constituting key documents for triggering adaptation plans for such vulnerable regions. For 2050 and 2100, it is estimated 903 km² and 1146 km² of vulnerable area, respectively, being the district of Lisbon the most vulnerable district in both scenarios. Results are available through a Web Map Service, for Portuguese public entities, and through a web map viewer for public and communities in general.

This review article is aimed at exploring the possibility of developing an integrated approach in the management of the different needs of endometrial cancer (EC) patients seeking to become pregnant. Life preservation of the woman, health preservation of the baby, a precocious and – as much as possible – minimally invasive characterization of the health and fertility parameters of the patient, together with the concerns regarding the obstetric, neonatal, and adult health risks of the children conceived via assisted reproduction techniques (ART) are all essential aspects of the problem to be taken into consideration, yet the possibility to harmonize such needs through a concerted integrated approach is still highly challenging. This article aims to illustrate the main features of EC and how it affects the normal physiology of pre-menopausal women. Then, we focus on the prospect for a miR-based, molecular evaluation of patient health status, including both EC early diagnosis and staging and, similarly, the receptivity of the woman, discussing the possible evaluation of both aspects using a single specific panel of circulating miR in the patient, thus allowing a relatively fast, non-invasive way to perform such testing with a significantly reduced margin of error. Finally, the ethical and legal/regulatory complexities arising from such innovative techniques require not only a risk-benefit analysis; respect for patient autonomy and equitable health care access allocation are fundamental issues as well.

We report the characterization of phenotype, genotype, and functional parameters such as CD4+ T cell, viral load, HIV-1 genotype, class I HLA genotype, CCR5 polymorphism, and IFN-γ producing cells from a cohort of 37 people living with HIV-1 (PLWH) (viremic controllers, were compared those on antiretroviral therapy (ART) from two sites in São Paulo, Brazil. Half the PLWH (51.4%) were classified as viremic controllers (Controller group - CG), compared to 48.6% were on ART. As expected, controllers showed baseline higher levels of CD4+ T cells and nadir compared to those under ART, confirming the ability of HIV-1 controllers to preserve these cells even in the absence of ART. IFN-γ-producing cells showed greater magnitude to the HIV peptides Gag in controllers (p<0.03). Our results demonstrated that HLA protective alleles are associated with strong production of IFN-γ to Gag, Nef and RT epitopes in people living with HIV controllers.

The spin magnetic moments and spin g factors (g_s = -2) of electron, muon and tau are explained based on the electric charges (EC) and lepton charges (LC) in terms of the three-dimensional quantized space model. The spin g factors of electron, muon and tau are g_s = -2 which is the sum of the EC g factor (g_EC = -1) and the LC g factor (g_LC = -1). The spin g factor (g_s = -2) of the electron is predicted by the Dirac’s equation. The orbit g factors of electron, muon and tau are g_L = g_EC = -1 from the EC g factor (g_EC = -1) without the contribution of the LC g factor (g_LC = -1). The spin g factors of the elementary fermions are calculated from the equation of g_s = g_EC + g_LC + g_CC where g_EC = EC/|EC|, g_LC = LC/|LC| and g_CC = CC/|CC|. For example, the spin g factors of the neutrinos and dark matters are g_s = -1. The spin g factors of the u and d quarks are g_s = 0 and g_s = -2, respectively. The g factor problem of neutrinos with the non-zero LC charges are solved by the LC Coulomb force of F_c(LC) ≈0. It is, for the first time, proposed that the binary motion (fluctuations) of the m_EC and m_LC masses for the electron, muon and tau leptons make the anomalous g factor. This binary motion could be originated from the virtual particle processes including the photons. Also, the weak force (beta) decay is closely related to the binary motion of the m_EC and m_LC for the electron, muon and tau leptons.

Advanced 1H Nuclear Magnetic Resonance (NMR) relaxometry and diffusometry methods and VIS-nearIR spectroscopy combined with pH, electric conductivity (EC) and totally dissolved solids (TDS) measurements were used to assess the properties of wastewater collected from a chicken slaughterhouse in each step of the treatment process (wastewater before treatment, biologically treated wastewater, chemically treated wastewater and discharged wastewater) and from sludge. The 1H NMR Carr-Purcell-Meiboom-Gill (CPMG) and Pulsed-Gradient-Stimulated-Echo (PGSE) decay curves recorded for all samples of wastewater were analyzed by inverse Laplace transform (ILT) to obtain the distributions of transverse relaxation times T2 and diffusion coefficient D. The VIS-nearIR total absorbance, T2-values, D-values, pH, EC and TDS parameters were used for statistical analysis in principal component analysis (PCA). The 1H T2-distributions measured for the slaughterhouse wastewater lie in two main regions reflecting the amount of dissolved solids or the distribution of undissolved solids mobility. The PCA analysis successfully differentiates between polluted and less polluted types of wastewater and sludge. The wastewater treatment applied by the slaughterhouse is efficient. The recommended methods for wastewater monitoring are the NMR T2- and D-distributions and EC, TDS and NMR-D diffusion coefficient. Finally, Machine Learning AI-algorithms are used to provide prediction maps of wastewater treatment stage.
