Nephrotoxicity is a common adverse effect associated with gentamicin administration. The current study investigated the potential protective effects of dates fruit against Gentamicin-induced renal nephrotoxicity in female albino rats. Date fruit antioxidants were measured using a spectropho-tometer, and GC-MS was performed using a Perkin Elmer Clarus 600 GC and Turbomass mass spectrometer. Thirty experimental animals were assigned at random to one of three Prevention and Treatment subgroups. Gentamicin (GM) administration at a dose of 100mg/kg resulted in renal toxicity, as evidenced by alterations in kidney function tests and histological changes in the proximal convoluted tubules. Date fruit improved kidney function (albumin, total protein, uric acid, urea, creatinine) and tubule histology, according to the findings. The GC-MS analysis reveals the presence of 20 chemicals. Date fruit antioxidants include 14mg/gallic acid total phenolic, 32.22 mg/rutin equivalent total flavonoid, and 1000 ug/ml DPPH free radical scavenging. According to the study, date fruit and Gentamicin performed the best. Date fruit had a significant impact on prevention during Gentamicin treatment.

All neurological diseases have their pathological progress influenced by microglial cells, which react to insults with multiple morphofunctional phenotypes. However, the complete morphological spectrum of the reactive microglia, revealed by the three-dimensional microscopic reconstruction, has not been explored in detail in virus limbic encephalitis. Here, we used anatomical series of brain sections from a previous study using Piry arbovirus encephalitis, expanding the original investigation to CA1/CA2. The morphological response of the homeostatic and reactive microglia was investigated 8 days after infection. The hierarchical cluster and linear discriminant function analysis of multimodal morphometric features allowed to distinguish between the microglial morphology of infected individuals and controls. In order to represent more broadly the morphological spectrum of the microglia of each cluster, representative cells of the homeostatic and reactive microglia were chosen, using the sum of the distances of each cell in relation to all the others. Overall, the multivariate statistical analysis showed that the reactive microglia of infected animals have more complex trees and thicker branches, that cover a larger volume of tissue than the microglia of control animals. With this approach a more reliable representation of the microglia dispersion in the Euclidean space, would be closer to the spectrum of morphotypes of the control and infected groups, and help to avoid hiding the morphological kaleidoscope of surveillant and reactive microglia. As form precedes function in nature, our findings are a good starting point for future research with integrative approaches to microglia form and function.

The chloroplast is important organelle responsible for photosynthesis and various bio- synthetic functions essential to green plants. However, the regulatory mechanism of chloroplast development remains elusive. Here, we characterized a mutant with defective chloroplasts in rice (Oryza sativa L.) named it albino and lethal 13(osal13), which exhibited a distinct albino phenotype in leaves, eventually leading to seedling lethality. Molecular cloning revealed that OsAL13 encodes a novel protein in rice for which no homologous gene or known conserved domain was found. Analysis of the biological characteristics showed that the RNAi2 leaf was completely albino phenotype, the root and shoot lengths were significantly inhibited compared with wild type. Transmission electron microscopy indicated that RNAi2 were defective in chloroplast development. Overexpression of OsAL13 resulted the photosynthetic rate increase, So the total grain number per panicle also would be significantly increase. Besides, OsAL13 was located in chloroplast and constitutively expressed in various tissues, including green and non-green tissues. In addition, disruption of OsAL13 resulted in altered expression of plastid-encoded polymerases and nuclear-encoded chloroplast genes, OsAL13 also affected hormone levels in plant tissues. Therefore, our results provide new insights into the role of OsAL13 in regulating chloroplast development in rice.

We studied short- and long-term effects of intravitreal injection of N-methyl-D-aspartate (NMDA) on melanopsin-containing (m⁺) and non-melanopsin-containing (Brn3a⁺) retinal ganglion cells (RGCs). In adult SD-rats, the left eye received a single intravitreal injection of 5µL of 100nM NMDA. At 3 and 15 months, retinal thickness was measured in vivo using SD-OCT. Ex vivo analyses were done at 3, 7, 14 days or 15 months after damage. Whole-mounted retinas were immunolabelled for Brn3a and melanopsin, the total number of Brn3a⁺RGCs and m⁺RGCs were quantified and their topography represented. In control retinas, the mean total numbers of Brn3a⁺RGCs and m⁺RGCs were 78,903±3,572 and 2,358±144 (mean ± SD; n=10), respectively. In the NMDA injected retinas, Brn3a⁺RGCs numbers diminished to 50% and 25%, at 3 and 14 days, respectively, but there was no further loss up to 15 months. The number of immunoidentified m⁺RGCs decreased significantly at 3 days, recovered between 3-7 days and was back to normal thereafter. OCT measurements revealed a significant thinning of the left retinas at 3 and 15 months. Intravitreal injections of NMDA induce a rapid loss of 75% of Brn3a⁺RGCs, a transient downregulation of melanopsin expression but not m⁺RGC death, and a thinning of the inner retinal layers.
