Cartilage microfracturation is a surgical technique specifically designed to address chondral defects, which are injuries to the cartilage that covers the ends of bones in joints. These defects can result from traumatic injuries, degenerative conditions such as osteoarthritis, or congenital abnormalities. The primary objective of microfracture surgery is to promote the regeneration of functional cartilage tissue, thereby restoring joint function, alleviating pain, and enhancing mobility. The procedure involves creating small, controlled perforations, or microfractures, in the subchondral bone plate beneath the damaged cartilage. This process, performed with precision to minimize damage to surrounding healthy tissue, penetrates the subchondral bone to reach the bone marrow, which is rich in mesenchymal stem cells (MSCs). MSCs are multipotent cells capable of differentiating into various cell types, including chondrocytes, which are essential for cartilage production. The microfractures provide an access pathway for MSCs, allowing them to migrate from the bone marrow to the defect site. Once at the site, these stem cells differentiate into chondrocytes and initiate the formation of new cartilage tissue. This newly formed tissue helps fill the defect, aiming to restore the smooth surface of the cartilage and improve the joint's structural integrity. Understanding the molecular processes involved in cartilage repair is crucial for optimizing rehabilitation strategies and improving clinical outcomes. These processes include the regulation of various signaling pathways that control stem cell migration, differentiation, and extracellular matrix (ECM) synthesis. Growth factors such as transforming growth factor-beta (TGF-β), bone morphogenetic proteins (BMPs), and fibroblast growth factors (FGFs) play pivotal roles in these pathways, guiding MSCs in their transformation into functional cartilage cells. Additionally, the role of the ECM, composed primarily of collagen type II and proteoglycans like aggrecan, is paramount in providing structural support and biochemical signals. The ECM supports the newly differentiated chondrocytes and helps maintain the cartilage's mechanical properties. Matrix metalloproteinases (MMPs) and their inhibitors, tissue inhibitors of metalloproteinases (TIMPs), are crucial in ECM remodeling, ensuring the balance between synthesis and degradation of the matrix to support tissue repair and integrity. Angiogenesis and vascularization are also critical aspects of the molecular repair process. The initial phase of cartilage repair often includes transient vascularization facilitated by vascular endothelial growth factor (VEGF), ensuring that the newly formed tissue receives adequate nutrients and oxygen. However, as the repair tissue matures, it transitions back to an avascular state characteristic of healthy articular cartilage, which is essential for maintaining its mechanical properties and longevity. Rehabilitation strategies following microfracture surgery are meticulously crafted to facilitate recovery and functional restoration. These strategies include early mobilization techniques, controlled mechanical loading, biological augmentation, nutritional support, and advanced therapeutic modalities. Early mobilization with gentle passive range-of-motion exercises promotes synovial fluid circulation within the joint, preventing adhesions and joint stiffness, and delivering essential nutrients and growth factors. Controlled mechanical loading through gradual weight-bearing activities helps condition the repaired tissue, stimulating chondrocyte proliferation and ECM production while ensuring proper integration and function. Biological augmentation, including platelet-rich plasma (PRP) and hyaluronic acid (HA), supports cartilage repair by enhancing cellular activities and ECM formation. PRP, rich in growth factors, promotes cell proliferation and matrix synthesis, while HA injections improve joint lubrication, reduce pain, and provide a scaffold for new tissue growth. Nutritional support, including amino acids, vitamins, minerals, glucosamine, chondroitin sulfate, and antioxidants, plays a vital role in collagen synthesis and overall cartilage maintenance, protecting chondrocytes from oxidative stress and modulating inflammation. Advanced therapeutic modalities such as low-intensity pulsed ultrasound (LIPUS), pulsed electromagnetic fields (PEMF), photobiomodulation therapy, and cryotherapy offer additional benefits by enhancing chondrocyte activity, reducing inflammation, and promoting ECM production. These modalities work through various molecular mechanisms, such as activating signaling pathways and modulating gene expression, to improve the quality and durability of the repaired cartilage. By comprehensively understanding the molecular and practical aspects of cartilage repair, clinicians can develop more effective treatment plans that optimize cartilage healing and improve patient outcomes. The integration of molecular insights with advanced rehabilitation techniques holds the promise of revolutionizing cartilage repair, offering hope for those suffering from debilitating joint conditions. This review aims to bridge the gap between molecular biology and clinical practice, providing a roadmap for optimizing cartilage repair strategies and ensuring long-term success for patients.

The aim of this study was to analyze the chemical composition of nymphal envelopes (foam) collected from hemipterans from spittlebug belonging to the species Aphrophora alni (Hemiptera family Aphrophoridae). Foam was collected in Poland in the Mazowieckie Voivodeship in 2020- 2022. Foam was collected from nymphs, feeding on young leaves of black alder trees (Alnus glutinosa), from 12 different areas. For the first time, the chemical composition of the foam produced by the spittlebug was quantitatively analyzed, identifying from 73.76 to 93.21 percent. The analyzed samples consisted mainly of a carbohydrate polymer similar in sugar unit composition to fucoidate isolated from brown seaweed. The most important sugar units identified were fucose (17.0 % - 35.6 % of the dry weight of the secretion), glucose (6.1 % - 16.2 %), mannose (1.5 % - 9.9 %), rhamnose (0.4 % - 1.3 %) and xylose (0.06 % - 0.1 %). In addition, various fatty acids, myo-inositol, pinitol, aliphatic hydrocarbons, waxes and aliphatic derivatives of 2(3H)-furanone were found. The collected foam contained from 0.05 % to 0.1 % of dry matter. Significant changes in the chemical composition of nymphal envelopes were found depending on the year of collection, which is probably strongly related to climate change.

Rice (Oryza sativa L.) is a primary energy food for Asian population. One of the most constraints in rice production is soil salinity because of the rice is very susceptible to salt. Meanwhile, many agricultural land in Asia are in the saline area. It is important to identify and develop salt-tolerant rice varieties that can adapt in Asian climate. By combining morphological, physiological, and biochemical assessments for screening the salt tolerant of 116 Asian rice cultivars was able to classify into tolerant, moderate, and sensitive rice cultivars under salinity stress condition, and also for understanding the salt tolerance mechanisms. The rice cultivars which belong to salt-tolerant including Pokkali from India, TCCP 266 from Philippines, IR 45427 also from Philippines, and Namyang 7 from Korea. Whereas, salt-sensitive rice varieties like IR29 from Philippines, IR58 also from Philippines, Daegudo from Korea, and Guweoldo also from Korea. The salt-tolerant varieties showed signs of tolerance, including lower percent reduction of percentage germination, root length, root fresh weight, shoot length, plant biomass, and chlorophyll content. In order to maintain the cellular osmotic balance under saline condition, the salt-tolerant ones exhibited less membrane damage, lower Na/K ratio, high proline and sugar accumulation, lower malondialdehyde (MDA) and hydrogen peroxide (H2O2). Pokkali from India, TCCP 266 from Philippines, IR 45427 also from Philippines, and Namyang 7 from Korea are recommended as valuable germplasm resource for Asian rice breeding program in saline agricultural area.

Biofilms are congregations of bacteria on a surface, and they grow into obstacles for the functionalities of any device or machinery involves anything biological. Biofilms are developed through a biochemical system known as ‘Quorum Sensing’ that accounts for the chemical signaling that direct either biofilm formation or inhibition. Computational models that relate chemical and structural features of compounds to their performance properties have been used to aide in the discovery of active small molecules for many decades. These quantitative structure-activity relationship (QSAR) models are also important for predicting the activity of molecules that can have a range of effectiveness in biological systems. This study uses QSAR methodologies combined with and different machine learning algorithms to predict and assess the performance of several different compounds acting in Quorum Sensing. Through computational probing of the quorum sensing molecular interaction, new design rules can be elucidated for countering biofilms.

Dermal wound healing describes the progressive repair and recalcitrant mechanism of damaged skin and eventually reformatting and reshaping the skin. Many probiotics, nutraceuticals, metal nanoparticles have been associated with improved healing process of intra and inter tissue wounds. Despite the vast nature on material based wound healing mediators, the exact mechanism on material-cellular interaction is still point of repent issue particularly in diabetics and pathological condition. The use of bioengineered alternative agents will likely only continue to dominate the outpatient and perioperative management of chronic, recalcitrant wounds as new additional products continue to cut costs and improve wound healing process. This review article provides an update of the various remedies with confirmed wound healing activities by a diverse group of agents from previous experiments conducted by various researchers

The influence of replacing corn silage with sorghum silage in the diet of dairy buffalo cows on metabolic status and on milk yield, chemical characteristics and fatty acid profile was studied. Forty dairy buffalo cows were included in the trial and divided into two homogeneous groups (SS, sorghum and CC, corn). Blood was collected at the end of the trial (120 days), individual milk yield was registered daily. Samples of milk were monthly collected and analyzed for fat, protein and lactose. Moreover, fatty acid profiles of silages and milk were determined. Buffalo cows fed sorghum silage showed an average milk yield higher than group CS (kg/d 10.120 vs 9.270; P<0.05), probably due to the lower lignin content of sorghum silage (31 vs 47 g/kg dry matter, respectively for SS and CS diets) and by consequence to its energy value, higher than expected. The percentage of linoleic acid was significantly higher in milk of group CS (C18:2: 1.27% vs 2.05%; P<0.01) due to the higher content of these acids in corn than in sorghum silage. The omega 6/omega 3 ratio was significantly lower in milk from buffalo cows fed sorghum than corn silage (7.8 vs 12.9; P<0.01). Serum biochemistry showed no negative effects of the corn replacing with sorghum.

Lipids have not traditionally been considered likely candidates for catalyzing reactions in biological systems. However, there is significant evidence that aggregates of amphiphilic compounds are capable of catalyzing reactions in synthetic organic chemistry. Here we demonstrate the potential for the hydrophobic region of a lipid bilayer to provide an environment suitable for catalysis by means of a lipozyme, or lipid aggregate capable of speeding up a chemical reaction. By bringing organic molecules into the non-polar or hydrophobic region of a lipid bilayer, reactions can be catalyzed by collections of small, nonpolar or amphiphilic molecules. We demonstrate this concept by the ester hydrolysis of calcein-AM to produce a fluorescent product, which is a widely used assay for esterase activity in cells. The reaction was first carried out in a two phase octanol-water system with the organic phase containing the cationic amphiphiles cetyltrimethylammonium bromide (CTAB) or octadecylamine. The octanol was then replaced with DOPC added to the water in the form of vesicles, where the reaction was also found to be carried out. The reaction was monitored using quantitative fluorescence which revealed catalytic turnover numbers on a scale of 10−4s−1 for each system, which is comparable to some slow enzymes. The reaction product was characterized by 1H-NMR measurements which were consistent with ester hydrolysis. The implications of thinking about lipid aggregates as catalytic entities are discussed in the context of biochemistry, pharmacology and synthetic biology.

Opossums, native to the Americas, are unique marsupials with over 100 species. The Common Opossum and Virginia Opossum are notable species. They are nocturnal, omnivorous, and adapt to various habitats. Their reproduction involves a short gestation and marsupial pouch development. Despite their ecological benefits like seed dispersal and insect control, their proximity to human habitats exposes them to pathogens, posing public health risks. These include protozoa, helminths, and arthropods that can spread zoonotic parasites and vector-borne diseases. The marsupial immune system, including opossums, is complex, rivaling eutherian mammals. Key immune genes were identified in the grey short-tailed opossum genome. CD1 protein and VpreB surrogate light chain were also found. Marsupials are proposed as ideal models for studying developmental immunology. A West Indian opossum (Didelphis marsupialis) was found and examined. Blood tests revealed abnormalities, including a low white blood cell count and signs of anemia. The results contribute to understanding the species’ health and adaptability in the wild.

The Kounis syndrome (KS) is an acute coronary syndrome triggered by allergic or hypersensitiv-ity reactions. Incidence rates vary, with studies reporting 19.4 per 100.000 among all admissions and 3.4% among allergy patients. This review explores the expanding understanding of KS, en-compassing various manifestations, and focusing on both clinical data and forensic findings useful in performing the diagnosis. The pathophysiology of this syndrome involves a complex in-terplay between allergic reactions and the cardiovascular system. Mast cell activation, histamine release, leukotrienes, cytokines, and platelet activation can contribute to the coronary events. A classification system with three types (Allergic Angina, Allergic Myocardial Infarction, Allergic Stent Thrombosis) aids in categorizing presentations. The diagnosis of KS relies on clinical presentation, laboratory findings and imaging. Post mortem assessment of KS is based on the in-tegration of circumstantial data, autopsy and histological findings. Biochemical and immuno-histochemical analyses also contribute to post-mortem diagnosis. In conclusion, a combined, multidisciplinary approach, should be gained to ease diagnostic pro-cess which is crucial for forensic practitioners in confirming KS occurrence.

(1) Background: The elderly who have chewing difficulty (CD) are prone to malnutrition, frail-ty, and sarcopenia, and those with insufficient physical activity (IPA) are also prone to frailty, overweight, and high blood cholesterol. There is an urgent need to prepare functional foods and to promote healthy physical activity for the elderly over 70 years of age to prevent poor nutri-tion and frailty. (2) Methods: This study prepared functional foods (FF) with softened chicken moose and milkfish without fin for the elderly and compared the different effects of FF from those of music aerobic exercise (MAE) on the complete blood counts, blood biochemistry, in-cluding nutrition index of albumin, and cholesterol levels, and frailty scores before and after in-stitution of FF and MAE among residents of long-term care institutes. (3) Results: The FF signif-icantly increased the high-density lipoprotein cholesterol and albumin levels. In contrast, MAE significantly increased red blood cells (RBC), and decreased the rates of frailty items on fatigue feeling and low physical activity. In comparison between FF and MAE groups, we found that the FF for 5 weeks significantly improved better on HDL-C and MCV while the MAE for 10 weeks improved significantly better on hematopoiesis as increases in red blood cells and platelets. (4) Conclusions: Results from this study suggest that FF and MAE for the elderly have different ef-fects on nutrition status, blood biochemistry and frailty. A combination of both FF and MAE might provide better benefits of nutrition status, physical function, and frailty for the elderly.

Aim: Abnormal laboratory findings have been shown to be associated with severe COVID-19. However, all aspects of this association have not been reviewed systematically. Therefore, the aim of this meta-analysis was to explore crucial laboratory parameters in severe COVID-19 infection. Methods: We performed the literature review of scientific articles indexed in electronic databases. Scientific search engines were used to perform the electronic literature search. After the removal of duplicates and selection of articles of interest, 30 studies were eligible to include. If heterogeneity was high (I²>50%), a random-effects model was applied to combine the data. Otherwise, a fixed-effects model was used.Results: A total of 5586 individuals were assessed (1555 patients with severe COVID-19 infection and 3452 with non-severe infection). Platelets, lymphocytes and serum albumin were significantly lower in severe patients while other biochemical and immunological parameters including prothrombin time, ALT, AST, total bilirubin, LDH, procalcitonin, CRP, IL-6, and IgA were significantly higher in patients with severe infection. Neutrophil and monocyte counts as well as hemoglobin level, D-dimer, hypersensitive troponin I, IL-2R, IgG and IgM levels were different between two groups; however, the difference was not statistically significant (All P-values >0.05). Conclusions: Lymphopenia, elevated liver enzymes, and high levels of inflammatory biomarkers are associated with severe COVID-19 infection.

Chemistry has been viewed as one of the most fruitful near-term applications to science of quantum computing. Recent work in transitioning classical algorithms to a quantum computer has led to great strides in improving quantum algorithms and illustrating their quantum advantage. Much less effort has been placed on how one finishes these calculations by using the results from the quantum computer (on the active region of the molecule) and embeds them back into the remainder of the molecule in order to determine the properties of the entire molecule. Such strategies are critical if one wants to expand the focus to biochemical molecules that contain active regions that cannot be properly explained with classical algorithms on classical computers. While we do not solve this problem here, we provide an overview of where the field is going to enable such problems to be tackled in the future.

Globally, over the past several months millions of persons contracted the coronavirus disease 2019 (COVID-19) resulting in significant mortality. Health care systems are negatively impacted including the care of individuals with cancers and other chronic diseases such as chronic active hepatitis, cirrhosis and hepatocellular carcinoma. There are various probable pathogenic mechanisms that have been presented to account for liver injury in COVID-19 patients such as hepatotoxicity cause by therapeutic drugs, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection of the bile duct cells and hepatocytes, hypoxia and systemic inflammatory response. Liver biochemistry tests such as AST, ALT, GGT and ALP are deranged in COVID-19 patients with liver injury. Hepatocellular damage results in the elevation of serum AST and ALT levels in early onset disease while a cholestatic pattern that develops as the disease progress causes higher levels of ALP, GGT, direct and total bilirubin. These liver biochemistry tests are prognostic markers of disease severity and should be carefully monitored in COVID-19 patients. We conducted a systematic review of abnormal liver biochemistry tests in COVID-19 and the possible pathogenesis involved. Significant findings regarding the severity, hepatocellular pattern, incidence and related clinical outcomes in COVID-19 patients are highlighted.

The storage of biochemical information, which is a prerequisite for the development of the first cell, is an unsolved problem affecting all concepts of the origin of life. However, if the protected environment in the continental crust is taken into account, completely new possibilities emerge for identifying processes that may have been crucial for the formation of the first cell. Under this background, we can hypothesize that cellular life began, with a self-sustaining cycle of molecular reaction steps and information storage of peptide sequences in RNA in a crustal depth of approximately 1000 m. This cycle was made possible in an open system bound to gas-permeable tectonic fracture zones with a high proportion of CO₂ and/or N₂. It can be assumed that a large number of RNA-like molecular strands formed in the crustal environment, from which a special RNA was selected by flotation processes, which formed the basis of a proto-tRNA. The formation of peptides and vesicles in supercritical CO₂ and the chemical evolution of peptides have already been proven at conditions of the upper continental crust. The behavior of individual amino acids in connection with vesicle formation deserves special interest. Hydrophobic amino acids accumulate in the vesicle membrane, with their position in the membrane depending on the degree of hydrophobicity. Selection of a proto-tRNA with an acceptor arm ending with the bases CCA resulted in a distinctive property. The position of adenine at the tip of the acceptor arm permitted it to penetrate the membrane, allowing it to be linked to an amino acid at the 3'-OH position of the terminal ribose. The penetration depth was controlled by the hydrophobicity of the opposite anti-codon, with adenine always occupying the middle position for the hydrothermally formed hydrophobic amino acids. When the vesicle membrane was closely occupied by proto-tRNAs, the anti-codons acted as templates and stored the sequence of the peptides that form. In the cases of hydrophilic anti-codons, the acceptor arm does not reach far enough into the membrane. This allowed hydrophilic amino acids to be linked to 2´-OH.

Base excision repair (BER) is a cellular process that removes damaged bases arising from exogenous and endogenous sources including reactive oxygen species, alkylation agents, and ionizing radiation. BER is mediated by the actions of multiple proteins that work in a highly concerted manner to resolve DNA damage efficiently to prevent toxic repair intermediates. During the initiation of BER, the damaged base is removed by one of 11 mammalian DNA glycosylases resulting in abasic sites. Many DNA glycosylases are product inhibited by binding to the abasic site more avidly than the damaged base. Traditionally apurinic/apyrimidinic endonuclease, APE1, was believed to help turnover the glycosylases to undergo multiple rounds of damaged base removal. However, in a series of papers from our laboratory we have demonstrated that UV-damaged DNA binding protein (UV-DDB) stimulates the glycosylase activities of human 8-oxoguanine glycosylase (OGG1), MUTY DNA glycosylase (MUTYH), alkyladenine glycosylase/N-methylpurine DNA glycosylase (AAG/MPG), and single-strand selective monofunctional glycosylase (SMUG1), between three-to-five-fold. Moreover, we have shown UV-DDB can assist chromatin decompaction facilitating access of OGG1 to 8-oxoguanine damage in telomeres. This review summarizes the biochemistry, single-molecule, and cell biology approaches that our group used to directly demonstrate the essential role of UV-DDB in BER.

The liver health of Gilthead sea bream and European sea bass fed fishmeal-free diets including various proportions of plant proteins, insect and poultry by-product meals, was investigated through biochemical and histological analyses using a new Liver Index (LI) formula. Four isoproteic (45% DM) and isolipidic (20% DM) diets were compared: a control, plant-based diet (CV) and other three test ones where 40% of a plant protein-rich ingredient mixture was replaced by Hermetia illucens (H40) or poultry by-product (P40) meals alone or in combination (H10P30). The trials lasted 12 and 18 weeks for sea bream and sea bass, respectively. The results so far ob-tained highlighted species-specific differences in the physiological response to dietary changes. In sea bream, the biochemical and histological responses suggest favorable physiological status and liver health, with higher serum cholesterol (CHO) and triglyceride (TAG) levels and moderate hepatocyte lipid accumulation with the H10P30 diet compared to CV (p<0.05). In sea bass, all diets resulted in elevated serum TAG levels and liver lipid accumulation particularly in fish fed the P40 one (p<0.05) which resulted in the highest LI, coupled with higher frequency of severe lipid accumulation, hypertrophy, cord loss, peripheral nuclei displacement and pyknosis. In conclusion sea bream well adapted to the test diets, while sea bass exhibited altered hepatic lipid metabolism leading to incipient liver steatosis, likely due to the high lipid content of the diets including the insect and poultry meals. The LI developed in this study proved to be a reliable tool for assessing the effects of dietary changes on liver health of sea bream and sea bass, consistent with biochemical and histological findings.

Inherited mutations in BRCA1 and BRCA2 genes increase risks for breast, ovarian, and other cancers. Both genes encode proteins for accurately repairing chromosome breaks. If mutations inactivate this function, chromosome fragments may not be restored correctly. Resulting chromosome rearrangements can become critical breast cancer drivers. Because I had data from thousands of cancer structural alterations that matched viral infections, I wondered whether infections contribute to chromosome breaks and rearrangements in hereditary breast cancers. There are currently no interventions to prevent chromosome breaks because they are thought to be unavoidable. However, if chromosome breaks come from infections, they can be treated or prevented. I used bioinformatic analyses to test publicly available breast cancer sequence data around chromosome breaks for DNA similarity to all known viruses. Human DNA flanking breakpoints usually had the strongest matches to Epstein-Barr virus (EBV) tumor variants HKHD40 and HKNPC60. Many breakpoints were near sites that anchor EBV genomes, human EBV tumor-like sequences, EBV-associated epigenetic marks, and fragile sites. On chromosome 2, sequences near EBV genome anchor sites accounted for 90% of breakpoints (p<0.0001). On chromosome 4, 51/52 inter-chromosomal breakpoints were close to EBV-like sequences. Five EBV genome anchor sites were near breast cancer breakpoints at precisely defined, disparate gene or LINE locations. Breakpoint flanking regions resembled known EBV-cancers. Twenty-five breakpoints in breast cancers were within 1.25% of EBV cancer breakpoints. In addition to BRCA1 or BRCA2 mutations, all the breast cancers had mutated genes essential for immune responses. Because of this immune compromise, herpes viruses can activate and produce nucleases that break chromosomes. Alternatively, anchored viral episomes can obstruct break repairs, whatever the cause. The results, therefore, imply proactive treatment and prevention of herpes viral infections may prevent some chromosome breaks and benefit BRCA mutation carriers.
