The construction industry is notable contributors to atmospheric dust particulate emissions. The particulate matter (PM) produced in construction processes contains a range of chemically hazardous substances, posing significant health risks (HR) to individuals. The occupational health hazards research in construction has become a focal point of interest internationally. Initiated in the early 2000s, there has been a growing demand within the construction research community for the creation of a unified PM database that encapsulates a wide array of construction activities. Previous studies have endeavored to establish a PM database for various construction contexts, yet they have fallen short in thoroughly addressing the diversity of construction materials and the levels of toxic substances (TS) within the PM. This study validates the proof of value that HR associated with construction processes can be measured using PM and TS datasets generated from various work processes using a variety of construction materials. This was accomplished through a semi-automated Building Information Modeling (BIM)-based plugin, which streamlines the assessment of occupational HR in construction projects alongside ongoing work processes. To validate the practicality of the Health Risk Index (HRI) determination via this BIM plugin, a case study on a real renovation project was executed. This system provides construction supervisors with a tool to visually assess the HR of daily operations, thereby facilitating the adoption of preemptive measures to protect the health of construction workers.

The construction industry, characterized by its intricate network of stakeholders and diverse workforce, grapples with the challenge of managing information effectively. This study delves into this issue, recognizing the universal importance of safeguarding data, particularly amid rising concerns of unauthorized access and breaches. Aiming to harness the potential of blockchain technology to address these challenges, this study used a hypothetical biographical and safety data of construction workers and securely stored on a Hyperledger Fabric blockchain. Developed within the Amazon Web Services (AWS) cloud platform, this blockchain infrastructure emerged as a robust solution for enhancing data security and privacy. Anchored in the core principles of data security, the model emerges as a potent defender against the vulnerabilities of traditional data management systems. Beyond its immediate implications, this study exemplifies the marriage of blockchain technology and the construction sector, potentially reshaping workforce management, especially in high-risk projects, optimizing risk assessment, resource allocation, and safety measures to mitigate work-related injuries. Practical validation through transaction testing using Hyperledger Explorer validates the model's feasibility and operational effectiveness, serving as a blueprint for the industry's data management. Ultimately, this research not only showcases the promise of blockchain technology in addressing construction data security challenges but also underscores its practical applicability through comprehensive testing, heralding a new era of data management that harmonizes security and efficiency for stakeholders' benefit.

In urban areas, deep excavation-induced ground deformation may damage adjacent existing structures and is conventionally evaluated by levelling at installed settlement points. However, a small number of measurements cannot represent the total change in ground deformation adjacent to excavation sites. Furthermore, significant local subsidence may occur in places where settlement points have not been installed and only noticed after an accident. For deep excavation sites located in urban areas, paved pedestrian sidewalks are often located adjacent to sites, and construction activity can cause these paving blocks to displace. This study introduces a method to detect paving block displacement adjacent to deep excavation sites using terrestrial photogrammetry. A digital camera creating point cloud data (PCD) and an acquisition method satisfying the frontal and side overlap requirements were demonstrated. To investigate the displacement detection and measurement capabilities by PCD analysis, an experimental program was conducted, including a PCD comparison containing the uplift, settlement, and horizontal paving block displacement and reference data. The cloud-to-cloud distance computation algorithm was adopted for PCD comparison. Paving block displacement was detected for displacements of 5, 7.5, and 10 mm in the uplift, settlement, and horizontal directions; however, the horizontal displacements were less clear. PCD analysis enabled satisfactory measurements between 0.024 and 0.881 mm for the vertical-displacement cases, but a significant error was observed for the horizontal-displacement cases owing to the cloud-comparison algorithm. The measurement blind spot of limited settlement points was overcome by the proposed method that detected and measured paving block displacement adjacent to excavation sites.

This study presents a method for the rapid and accurate generation of two-dimensional (2D) orthomosaic maps using selected image data collected by drone-captured video. The focus is on developing a real-time method capable of creating maps more quickly than image selection. The scale-invariant feature transform (SIFT) algorithm is applied to drone images to extract features in various scale regions. For feature point matching, a matching method based on the fast library for approximate nearest neighbors (FLANN) was adopted. A comparison of the computational speed of the FLANN with that of the k-nearest neighbors (KNN) and brute force matcher during the matching process revealed FLANN's superior capability for real-time data processing. The random sample consensus (RANSAC) algorithm was employed to enhance the accuracy of the matching by removing outliers, effectively identifying and eliminating mismatches, and reinforcing the reliability of feature point matching. The combination of SIFT, FLANN, and RANSAC algorithms demonstrates the capacity to process drone-captured image data in real time, facilitating the generation of precise 2D orthomosaic maps. The proposed method was assessed and validated using imagery obtained as the drone executed curvilinear and straight flight paths, confirming its accuracy and operational efficiency concurrently with the image capture process.

The conserved MYST proteins form the largest family of histone acetyltransferases (HATs) that acetylate lysines within the N-terminal tails of histone, enabling active gene transcription. Here, we have investigated the biological and regulatory functions of the MYST family HAT Sas3 in the opportunistic human pathogenic fungus Aspergillus fumigatus using a series of genetic, biochemical, pathogenic, and transcriptomic analyses. The deletion of sas3 results in drastically reduced colony growth, asexual development, spore germination, response to stresses, and the fungal virulence. Genome-wide expression analyses have revealed that the sas3 mutant showed 2,402 significant differentially expressed genes; 1,147 up-regulated and 1,255 down-regulated. The representative up-regulated gene resulting from sas3 is hacA predicted to encode a bZIP transcription factor, whereas the UV-endonuclease UVE-1 was significantly down-regulated by sas3. Furthermore, our Western blot analyses suggest that Sas3 likely catalyzes acetylation of H3K9, K3K14, and H3K29 in A. fumigatus. In conclusion, Sas3 is associated with diverse biological processes and can be a potential target for controlling pathogenic fungi.

Prenatal diagnosis of clubfoot traditionally relied on two-dimensional ultrasonography. To enhance diagnosis and predict postnatal outcomes, we examined the parameters that differentiate pathological clubfoot using three-dimensional ultrasonography. In our retrospective study, we examined the findings of prenatal ultrasound and the postnatal outcomes of pregnancies with suspected congenital clubfoot between 2018 and 2021. Based on the three-dimensional perspective, we measured the angles of varus, equinus, calcaneopedal block and forefoot adduction and compared the sonographic variables between the postnatal treated and non-treated groups. We evaluated 31 pregnancies (47 feet) with suspected clubfoot using three-dimensional ultrasonography. After delivery, a total of 37 feet (78.7%) underwent treatment involving serial casting only or additional Achilles tenotomy. The treated group showed significantly greater hindfoot varus deviation (60.5° vs. 46.6°, p=0.026) and calcaneopedal block deviation (65.6° vs. 26.6°, p <0.05) compared to the non-treated group. The calcaneopedal block had an area under the curve of 0.98 with a diagnostic threshold of 46.2 degrees (sensitivity of 97%, specificity of 90%, positive predictive value of 97%, and negative predictive value of 90%). During prenatal evaluation of clubfoot using three-dimensional ultrasonography, the calcaneopedal block deviation has the potential to predict postnatal treatment.

Glaucoma is a group of ocular diseases that causes irreversible blindness worldwide and is characterized by a multifactorial degeneration of optic nerve and retinal ganglion cells (RGCs), leading to vision loss. One of the significant risk factors in glaucoma pathogenesis is glia-driven neuroinflammation. Impairment of mitochondrial dynamics and bioenergetics is associated with glaucomatous neurodegeneration and is linked to neuroinflammation. Apolipoprotein A-I binding protein (AIBP) binds to toll-like receptor 4 (TLR4) and facilitates the removal of excess cholesterol from the plasma membrane in activated cells, suggesting the regulation of cholesterol-driven inflammation by inhibiting the TLR4-associated lipid rafts. AIBP is localized in mitochondria and regulates mitophagy by ubiquitination of mitofusin 1 and 2. We observed elevated intraocular pressure-induced AIBP deficiency in glaucomatous neurodegeneration. We also observed that AIBP deficiency activated TLR4 in Müller glia, triggered mitochondrial dysfunction in both RGCs and Müller glia and impaired visual function. In mouse models of glaucoma, AIBP treatment protects RGCs by reducing inflammatory responses. Here, our review focuses on investigating the role of AIBP in the retina and examining the underlying mechanism by which AIBP contributes to the pathogenesis of glaucoma, aiming to provide new insight into the therapeutic potential for the treatment of glaucoma.

Although ionizing radiation (IR) is widely used for therapeutic and research purposes, studies on low-dose ionizing radiation (LDIR) are limited compared with those on other IR approaches, such as high-dose gamma irradiation and ultraviolet irradiation. High-dose IR affects DNA damage response and nucleotide-protein crosslinking, among other processes; however, the molecular consequences of LDIR have been poorly investigated. Here, we developed a method to profile RNA species crosslinked to an RNA-binding protein, human antigen R (HuR), using LDIR and high-throughput RNA sequencing. The RNA fragments isolated via LDIR-crosslinking and immunoprecipitation sequencing were crosslinked to HuR and protected from RNase-mediated digestion. Upon crosslinking HuR to target mRNAs, such as PAX6, ZFP91, NR2F6, and CAND2, the transcripts degraded rapidly in human cell lines. Additionally, PAX6 and NR2F6 downregulation mediated the beneficial effects of LDIR on cell viability. Thus, our approach provides a method for investigating post-transcriptional gene regulation using LDIR.

Thin people who undergo implant insertion into the chest wall face a high risk of implant exposure to the external environment. Five months after an 8-year-old boy underwent implantable cardioverter-defibrillator (ICD) implantation in a subcutaneous pocket in the left anterolateral chest wall to manage long QT syndrome, ICD replacement became necessary owing to exposure risk from distal and lateral thinning of the ICD pocket. Pocket rupture and exposure would increase the risk of infection; therefore, we performed ICD removal and primary pocket closure. Two weeks later, a new suprafascial pocket was created, acellular dermal matrix (ADM) was attached to the inner wall to prevent ICD protrusion, and a new ICD was inserted. One year postoperatively, the ADM was engrafted, and no complications were observed. A thin subcutaneous layer increases the risk of ICD implantation complications. Inner wall strengthening with ADM can help prevent pocket rupture.
