The farnesoid X receptor (FXR)/βKlotho/fibroblast growth factors (FGFs) pathway is crucial for maintaining the intestinal barrier and preventing colorectal cancer (CRC). We used an FXR ago-nist, GW4064, and FXR knockout (KO) mice to investigate the role of FXR/Klothos/FGFs pathways in lipopolysaccharide (LPS)-induced intestinal barrier dysfunction and colon carcinogenesis. The results showed that upregulation of FXR in enterocytes effectively ameliorated intestinal tight junction markers (claudin1, Zonula occludens-1), inflammation, and bile acid levels, thereby protecting mice from intestinal barrier dysfunction and colon carcinogenesis. GW4064 treatment increased FXR, αKlotho, βKlotho, FGF19, FGF21, and FGF23 in wild-type mice exposed to LPS, while FXR KO mice had decreased levels. FXR KO mice exhibited elevated colon cancer markers (β-catenin, LGR5, CD44, CD34, Cyclin D1) under LPS, underscoring the pivotal role of FXR in inhibiting the development of colon tumorigenesis. The varying gut microbiota responses in FXR KO mice versus wild-type mice post-LPS exposure emphasize the pivotal role of FXR in pre-serving intestinal microbial health, involving Bacteroides thetaiotaomicron, Bacteroides acidifaciens, and Helicobacter hepaticus. Our study validates the effectiveness of GW4064 in alleviating LPS-induced disruptions to the intestinal barrier and colon carcinogenesis, emphasizing the importance of the FXR/αKlotho/βKlotho/FGFs pathway and the interplay between bile acids and gut microbiota.

This paper presents a novel approach for semantic segmentation of building roof in dense urban environment with Deep Convolution Neural Network (DCNN) using imagery acquired by a Chinese Very High Resolution (VHR) satellite mission, i.e. GaoFen-2 (GF-2). To provide an operational end-to-end work flow for accurate build roof mapping with feature extraction as well as image segmentation, a fully convolutional DCNN with both convolutional and deconvolutional layers is designed to perform the VHR image analysis for labeling pixels. Since the diverse urban patterns and building styles in large areas, sample image data sets of building roof and non-building roof are collected over different metropolitan regions in China. We selected typical cities with dense urban environment in each metropolitan region as study areas for collecting training and test samples. High performance cluster with GPU-mounted workstations is employed to perform the model training and optimization. With the building roof samples collected over different cities, the predictive model with multiple NN layers is developed for building roof labeling. The validation of the building roof map shows that the overall accuracy(OA) and the mean Intersection Over Union( mIOU) of DCNN based segmentation are 94.67%, 0.85 respectively, while CRF-refined segmentation achieved OA of 94.69% and mIOU of 0.83. The results suggest that the proposed approach is a promising solution for building roof mapping with VHR images over large areas across different urban and building patterns. With the operational acquisition of GF2 VHR imagery, it is expected to develop an automated pipeline for operational built-up area monitoring and timely update of building roof map over large areas.

Crossostephium chinense is a wild species with strong salt tolerance, which has great potential to improve the salt tolerance of cultivated chrysanthemums. Conversely, the unique salt-tolerant molecular mechanisms of Cr. chinense are still unclear. This study performed a comparative physiological and transcriptome analysis of Cr. chinense, Chrysanthemum lavandulifolium and their hybrids. The physiological results showed that Cr. chinense maintained a higher superoxide dismutase (SOD) activity, to alleviate the oxidative damage to membrane. KEGG enrichment analysis showed that the plant hormone signaling transduction and the MAPK signaling pathway were mostly enriched in Cr. chinense and hybrids under salt stress. Further weighted gene co-expression network analysis (WGCNA) of DEGs suggested that the abscisic acid (ABA) signaling transduction may play a significant role in the salt-tolerant mechanisms of Cr. chinense and hybrids. The tissue-specific expression patterns of the candidate genes related to ABA signaling transduction and MAPK signaling pathway indicated that genes related to ABA signaling transduction performed significant expression levels under salt stress. This study provides a theoretical foundation for future research into the molecular mechanism of salt tolerance in chrysanthemums under salt stress.

Ecological stoichiometry is an important index to reflect ecological interaction and the balance of various elements in ecological processes. At present, the stoichiometric spatial distribution of soil carbon (C), nitrogen (N), phosphorus (P) and potassium (K) in farmland and grassland of Inner Mongolia and its influencing factors are still unclear. In this study, the spatial distribution of soil nutrient content and stoichiometry and the effects of environmental factors on soil C:N:P:K stoichiometry were analyzed under different land use patterns in four agro-pastoral areas. The results showed that there was a geographical gradient in the ecological stoichiometric relationship between farmland and grassland. RCN, RCP, RCK, RNP and RNK showed an "N" distribution from west to east, while RPK showed a "V" distribution. The difference in spatial distribution was more significant than the difference in land use pattern. The ecological stoichiometric relationship of grassland was mainly affected by the content of C and N, and was related to the similarity of soil properties. The field ecological stoichiometry was mainly affected by the content of nitrogen and phosphorus, and had a great relationship with fertilization. The effects of MAT and MAP on soil ecological stoichiometry were related to the inter-regional environmental heterogeneity. The influence of MAP on farmland and MAT on grassland was more significant. It was of great significance for field management according to local conditions, rational utilization of land resources and sustainable development of agriculture to clarify the ecological stoichiometric characteristics of farmland and grassland in agro-pastoral ecotone.
