Abstract: Because of outstanding heat resistance and processing qualities, benzoxazine resins are widely employed in a variety of applications. However, traditional benzoxazine resins still confront hurdles in today's engineering applications, such as can’t long-term service in high-temperature settings and inadequate toughness. Four alkyne-functionalized benzoxazines with phthalide side group and cyano group of varying molecular weights were produced in this study. Fourier trans-form infrared spectroscopy (FT-IR) and hydrogen nuclear magnetic resonance spectroscopy ( 1H-NMR ) were used to characterize the resin structure, and differential scanning calorimetry (DSC) was used to investigate the thermal curing kinetics at different warming rates, the apparent activation energy was 116.9 kJ/mol. In-situ FTIR was used to investigate the cure mechanism. Dynamic mechanical analysis (DMA) was used to evaluate the gelationation time of BOZ series resins at various temperatures, and the curing process was designed by combined the results of DSC. The Tg of the composites made using BOZ-1N21 as the matrix was 336°C, which was much higher than the Tg of the BP-a resin made with aniline, phenolphthalein, and formaldehyde (Tg=251°C). As a result, the resin system is expected to be employed in applications requiring high-temperature resistance and toughness.

Cattle occasionally develop brassica-associated liver disease (BALD) and photosensitisation when grazing turnip or swede (Brassica spp.) forage crops. The liver toxin in these brassica varieties has yet to be discovered. Progoitrin is the dominant glucosinolate in incriminated crops. Apart from goitrin, progoitrin hydrolysis yields the nitrile, 1-cyano-2-hydroxy-3-butene (CHB), and the epithionitrile, 1-cyano-2-hydroxy-3,4-epithiobutane (CHEB). The two compounds were custom-synthesised. In a small pilot trial, New Zealand White rabbits were given either CHB or CHEB by gavage. Single doses of 0.75 mmol/kg of CHB or 0.25 mmol/kg of CHEB were subtoxic and elicited subclinical effects. Higher doses were severely hepatotoxic causing periportal to massive hepatic necrosis associated with markedly elevated serum liver biomarkers often resulting in severe illness or death within 24 h. The possibility that one or both of these hepatotoxic nitriles causes BALD in cattle requires further investigation.

Dubermatinib (DMB, TP-0903), a benzenesulfonamide, is an inhibitor of the tyrosine kinase AXL, which is a member of the TAM family, and can prevent GAS6-mediated activation of AXL in cancer cells. Patients with previously treated chronic lymphocytic leukemia are being studied in phase I/II clinical trials to determine its antineoplastic potential (CLL). In the current work, the reactive intermediates of DMB were studied using liquid chromatography-mass spectrometry (LC-MS). Human liver microsomes (HLMs) were exposed to dimethylbenzene in a laboratory setting, and the resulting metabolites were collected through protein precipitation. Intense reactivity toward nucleophilic macromolecules was seen in the metabolites of the piperazine and pyrimidine rings in DMB, iminium and 2,5-quinone-imine, respectively. To assess the toxicities of the possibly reactive metabolites, DMB was incubated with HLMs in the presence of 1.0 mM KCN and 1.0 mM glutathione. The DMB metabolites found by LC-MS/MS were seven in vitro phase I metabolites, three cyano adducts, and two GSH conjugates. Phase I in vitro metabolic reactions included N-demethylation, hydroxylation, and dechlorination. The in vitro metabolism of DMB and the metabolites it produces have not been studied before.
