Bread wheat (Triticum aestivum L.) is staple of Pakistani people. However, its yield at farmer field is low as compared with its genetic potential. Integration of various crop and soil management strategies might be an option to enhance wheat productivity at farmer field. This 2-year experiment was conducted to check the influence of combine application of natural plant water extracts and biochar on the productvity of wheat during the winter season of 2015-16. The experiment consisted of seven treatment viz. (1) control (2) application of biochar (0.18 kg pot-1) alone, (3) application of sorghum water extract (SWE) alone, (4) application of moringa water extract (MWE) alone, (5) application of biochar + SWE, (6) application of biochar + MWE, (7) application of biochar + SWE+MWE. The results revealed that application of both crop water extracts in combination with biochar improved the growth and grain yield of wheat. Use of MWE in combination with biochar enhanced the grain weight, grain number and grain yield of wheat by 44, 14, and 24%, respectively than the control treatment. In crux, use of MWE in combination with biochar might be a viable option to improve the productivity of bread wheat.

In this study, rheological properties of the bread wheat flour dough from 6 wheat genotypes were determined. For the pereparation of flour, 3 red bread (Pandas, Sagitorya, Pehlivan) and 3 white bread (Kaşifbey, Göktan, Ceyhan-99) were selected as wheat genotype. To determine the rheological properties of the wheat flour dough, farinograph, extensograph, mixolab and glutograph devices were used. According to the results of the Farinograph analysis, the average development time of wheat White and red genotypes were 1.95 minutes and 8.96 minutes, respectively. According to the extensograph results of the flour samples, the most extended stability value was determined with 7.47 min in Ceyhan-99 cultivar. As a result of the research, it was determined that flour yields of red bread varieties were higher other genotypes, gas retention capacities of white bread flours were showed high result in extensograph application and resistance of dough to elongation was higher. In the Mixolab analysis, it determined that white bread wheat varieties have higher values in terms of kneading properties and gluten properties, and red bread wheat varieties have higher values in values of viscosity, amylase value and starch retrogradation.

Wheat gluten contains epitopes that trigger celiac disease (CD). A life-long strict gluten-free diet is the only treatment accepted for CD. However, very low-gluten wheat may provide an alternative treatment to CD. Conventional plant breeding methods are not sufficient to produce celiac-safe wheat. RNA interference technology, to some extent, succeeded in the development of safer wheat varieties. However, these varieties had multiple challenges in their implementation. Clustered Regularly Interspaced Short Palindromic Repeats-associated nuclease 9 (CRISPR/Cas9) is a versatile gene-editing tool that has the ability to edit the immunogenic gluten genes. So far, only a few studies have applied CRISPR/Cas9 to modify the wheat genome. In this article, we reviewed published literature that applied CRISPR/Cas9 in wheat genome editing to investigate the current status of the CRISPR/Cas9 system to produce a low-immunogenic wheat variety. We found that in recent years, the CRISPR/Cas9 system has been continuously improved to edit the complex hexaploid wheat genome. Although some reduced immunogenic wheat varieties have been reported, CRISPR/Cas9 has still not been fully explored in editing the wheat genome. We conclude that further studies are required to apply the CRISPR/Cas9 gene-editing system efficiently for the development of celiac-safe wheat variety and to establish it as a “tool to celiac safe wheat.”

The effects of climate change on yield and quality for different climate regions had high uncertainty. Risk assessment is an effective measure to assess the seriousness of the projected impacts for decision-makers. The modified quality model was used to simulate integrated impacts of climate change, environment and management on wheat yield and quality. Then, the Canadian Earth System Model (CanESM2) was used to forecast the daily meteorological data, and Statistical Down Scaling Model was used for downscaling. CERES-Wheat was combined with the forecasted meteorological data to simulate the future wheat yield and grain protein concentration (GPC). The risk of wheat yield and quality in three climatic regions of Shaanxi combined with two climate change scenarios of CanESM2 were assessed. Temperature increased 0.22-3.34 °C and precipitation increased 10-60 mm for RCP4.5 and RCP8.5. Elevated temperature and precipitation had positive effects on yield in all regions. The yield risk of most regions with climate change decreased 3.8%-25.1%. The GPC risk of all regions with climate change decreased 7.3%-27.2%. Irrigation decreased yield risk greatly in all regions, while had totally different effects for the three climatic regions. Yield risk with irrigation decreased 37.7%-52.1% in different climate. In contrast to previous studies, GPC risk with irrigation increased greatly 25.8%-28.9% in humid region, 3.9%-8.8% in sub-humid region, and decreased 37.7%-52.1% in semi-arid region. Climate change decreased yield risk and GPC risk together. While irrigation decreased yield risk greatly in all regions, had totally different effects for the three climatic regions.

The hexaploid bread wheat Triticum aestivum L. (2n = 6x = 42) and the tetraploid durum wheat Triticum durum (2n = 4x = 28) are two commercially the most important wheat species worldwide. These two species are components of the same gene pool, which evolved under different conditions, which made it possible to acquire different adaptations to a wider range of environmental conditions. Identification and characterization of introgressed fragments of one species in the genome of another can help in the search for new loci responsible for economically valuable traits. SNP microarrays provide information on the genotypes of thousands of positions throughout the genome. This information can be used to obtain the distribution of alleles of one species in the genome of another and to determine the location of introgressed fragments. In our work, we used genotyping data from the SNP microarray to identify durum wheat chromosome fragments containing bread wheat alleles in the offspring from crossing bread and durum wheat. We have studied the distribution of the genetic material of bread wheat in 15 combinations from crosses of bread and durum wheat. Introgressed fragments of bread wheat into the durum wheat genome were identified on chromosomes 1A, 1B, 4A, 5B, 6B, and 7A. The introgressed fragments showed polymorphism in different breeding lines. A functional enrichment analysis of genes in the introgression fragments was performed, which showed the presence of statistically significant enrichment of genes responsible for various molecular functions, biological processes and cellular components. The obtained data can be used to identify new QTL of economically valuable traits for obtaining durum wheat varieties with improved agronomic traits.

Durum wheat is an important food crop in the world and an endemic species of sub-Saharan Africa (SSA). In the highlands of Ethiopia and the oases of the South Sahara this crop has been cultivated for thousands of years. Today, smallholder farmers still cultivate it on marginal lands to assure production for their self-consumption. However, durum wheat is no longer just a staple crop for food security but it has become a major cash crop. In fact, the pasta and couscous industry currently purchase durum grain at prices 10 to 20% higher than bread wheat. Africa as a whole imports over € 4 billion per year of durum grain to provide the raw material for its food industry. Hence, African farmers could obtain a substantial share of this large market by turning their production to this crop. Here, the achievements of the durum breeding program of Ethiopia are revised to reveal a steep acceleration in variety release and adoption in the last decade. Furthermore, the variety release for Mauritania and Senegal is described to show how modern breeding methods could be used to deliver grain yields above 3 t ha^-1 in seasons of just 92 days of length and daytime temperatures always above 32°C. This review describes the ability of releasing durum wheat varieties adapted to all growing conditions of SSA, from the oases of the Sahara to the highlands of Ethiopia. This potential area of expansion for durum wheat production in SSA is not linked to any breeding technology, but rather it remains dependent on the market ability to purchase these grains at a higher price to stimulate farmer adoption. The critical importance of connecting all actors along the semolina value chain is presented in the example of Oromia, Ethiopia, and that success story is then used to prompt a wider discussion on the potential of durum wheat as a crop for poverty reduction in Africa.

Although wheat is used worldwide as a staple food, it can give rise to adverse reactions, for which the triggering factors have not been identified yet. These reactions can be caused mainly by kernel proteins, both gluten and non-gluten proteins. Among these latter, -amylase/trypsin inhibitors (ATI) are involved in baker’s asthma and realistically in Non Celiac Wheat Sensitivity (NCWS). In this paper, we report characterization of three transgenic lines obtained from the bread wheat cultivar Bobwhite silenced by RNAi in three ATI genes CM3, CM16 and 0.28. We have obtained transgenic lines showing an effective decrease of the activity of target genes that, although showing a higher trypsin inhibition as a pleiotropic effect, generate a lower reaction when tested with sera of patients allergic to wheat, accounting for the important role of the three target proteins in wheat allergies. Finally, these lines show unintended changes differences in high molecular weight glutenin subunits (HMW-GS) accumulation, involved in technological performances, but do not show differences in terms of yield. The development of new genotypes accumulating a lower amount of proteins potentially or effectively involved in such pathologies, not only offers the possibility to use them as a basis for the production of varieties with a lower impact on adverse reaction, but also to test if these proteins are actually implicated in those pathologies for which the triggering factor has not been established yet.

In recent years cultivated soils have been increasingly supplemented with nutrients that at low doses are necessary for proper plant functioning but become toxic at high doses. New methods are needed to prevent these destructive actions, and for this reason we studied the effects of two elements – Mn treated as a stressor and Se treated as a potential defense in two wheat cultivars. The intensity of stress was manifested in tissue browning and weight reduction and was determined by an increase in lipid peroxidation and quantitative analysis of hydrogen peroxide levels. It was found that the excess of Mn in the substrate caused more intense changes in these indicators in the root system than in the leaves, and that Se presence partly eliminated the stress evoked effects. Moreover, Mn-treatment was accompanied by a greater absorption of this element by the roots, and a reduced uptake of other elements (K, Fe, S, P), with the exception of Ca, an increase in which was observed especially in the additional presence of Se. It was suggested that the rise in Ca level can lead to modification of cell differentiations and may be one of the steps in defense mechanisms. The change in the direction of cell differentiation in the apical part of the root was observed microscopically under Mn stress and was accompanied by a quantitative increase in 5-met C. Based on DNA methylation profiles detected by MSAP we concluded that various types of methylation sites may be activated under Mn treatment in roots.

Among the rust diseases, leaf rust of wheat caused by Puccinia triticina, is the most prevalent worldwide and causes significant yield losses. This study aimed to determine the genomic location of loci that control adult plant resistance (APR) to leaf rust in the pre-Green Revolution landrace accession, Aus27506, from the ‘Watkins Collection’. An Aus27506/Aus27229-derived F7 recombinant inbred (RIL) population was screened under field conditions across three cropping seasons and genotyped with the iSelect 90K Infinium SNP bead chip array. One QTL on each of chromosomes 1BL, 2B and 2DL explained most of the leaf rust response variation in the RIL population and were named QLr.sun-1BL, QLr.sun-2B and QLr.sun-2DL, respectively. QLr.sun-1BL and QLr.sun-2DL were contributed by Aus27506. QLr.sun-1BL is likely Lr46, while QLr.sun-2DL appeared to be a new APR locus. The alternate parent, Aus27229, carried the putatively new APR locus QLr.sun-2B. Comparisons of average severities among RILs carrying these QTL in different combinations indicated that QLr.sun-2B does not interact with either of the other two QTL; however, the combination of QLr.sun-1BL and QLr.sun-2DL reduced disease severity significantly. In-planta fungal quantification assays validated these results. The RILs carrying QLr.sun-1BL and QLr.sun-2DL did not differ significantly from parent Aus27506 in resistance. Aus27506 can be used as a source of adult plant leaf rust resistance in breeding programs.

Wheat is the essential constituent of cereal-based diets and one of the most significant sources of calories. However, there is an inherently low bioavailability of proteins, mineral, and vitamins in modern wheat grains. Biofortification has earned recognition as an outstanding approach, at the same time as a cure for world hunger. The developments in the identifications of quantitative trait loci (QTL) analysis and understanding of the physiological and molecular basis of QTLs controlling the biofortification traits in wheat has revealed new horizons for the improvement of modern wheat varieties. Within this review, we have compiled the information from the studies carried out in wheat using QTL mapping methodologies that is among the best methods for biofortification traits. We hope this review will serve as an essential reference for the QTLs identified for the several important biofortification traits in wheat.

Wheat (Triticum aestivum L.) being a staple food crop is an important nutritional source providing protein and minerals. It is important to fortify staple cereals like wheat with essential minerals to overcome the problems associated with malnutrition. The experiment was designed to evaluate the status of 11 micronutrients including grain iron (GFe) and zinc (GZn) in 62 wheat cultivars released between 1911 and 2016 in Pakistan. Field trials were conducted over two years and GFe and GZn were quantified by both inductively coupled plasma optical emission spectroscopy (ICP-OES) and energy dispersive X-ray fluorescence spectrophotometer (EDXRF). The GZn ranged from 18.4 to 40.8 mg/kg by ED-XRF and 23.7 to 38.8 mg/kg by ICP-OES. Similarly, GFe ranged from 24.8 to 44.1 mg/kg by ICP-OES and 26.8 to 36.6 mg/kg by EDEXR. The coefficient of correlation was higher for GZn (r=0.90), compared to GFe (r=0.68). Modern cultivars like Zincol-16 and AAS-2011 showed higher GFe and GZn along with improved yield components. Old wheat cultivars WL-711, C-518 and Pothowar-70 released before 1970 also exhibited higher value of GFe and GZn, however their agronomic performance was poor. Multivariate analysis using ten micronutrients (Al, Ca, Cu, K, Mg, Mn, Na and P) along with agronomic traits, and genome-wide SNP markers identified the potential cultivar with improved yield, biofortification trait and wider genetic diversity. Genetic gain analysis identified significant increase in grain yield (0.4% year-1), while there was negative gain for GFe (-0.11% year-1) and GZn (-0.15% year-1) over the span of 100 years. The Green Revolution Rht-B1 and Rht-D1 genes had strong association with plant height, and grain yield (GY), while semi-dwarfing alleles had negative effect on GFe and GZn contents. This study provided a valuable insight into biofortification status of wheat cultivars deployed historically in Pakistan and is a valuable source to initiate a breeding strategy for simultaneous improvement in wheat phenology and biofortification.

Plant growth promoting rhizobacteria (PGPR) are capable to reduce the use of chemical fertilizers input cost of farmer. Keeping in view the study was designed to investigate and evaluate inoculation effect of indigenous rhizospheric bacteria on growth and yield of wheat (Triticum aestivum L.) under in vitro and in vivo conditions using different treatments. Ten potential strains were selected on the basis of their ACC deaminase activity, siderophore production, P-solubilization and production of indole acetic acid (IAA). Further these strains were tested in three different experiments (growth chamber, pot and field). We found significant increase in crop growth response to the inoculants in comparison with un-inoculated control. In pot and field trial we tested PGPR with recommended dose of inorganic fertilizers. The results of present study revealed that inoculation of bacterial strains with wheat seeds significantly increased plant growth and improved crop yield. Results of present study reveal that these strains could be employed in different combinations and can get higher yield in case of half recommended doses of inorganic fertilizers along with consortium of strains in comparison with sole application of recommended dose of fertilizer and with consortium of strains. These strains were further identified by 16Sr RNA gene sequencing, fatty acid profile and biolog. It can be concluded that inoculated bacteria have more potential and contributes in good crop quality, increased yield when they are applied in combination, thus have potential to minimize use of chemical fertilizers.

The endogenous pool of phytoregulators in plant tissues supplied with microbial secondary metabolites may be crucial for the development of winter wheat seedlings during cool springs. Phytohormones may be synthesized by psychrotrophic microorganisms in lower temperatures occurring in temperate climate. Two fungal isolates from the Spitzbergen soils after the microscopic observations and ITS region molecular characterization were identified as Mortierella antarctica (MA DEM 7) and Mortierella verticillata (MV DEM32). To study the synthesis of indoleacetic acid (IAA) and gibberellic acid (GA) Mortierella strains were grown on media supplemented with precursors of phytohormones (tryptophan or methionine) at 9, 15 and 20 °C for 9 days. The highest amount of IAA synthesis was observed in MV DEM 32 9-day culture at 15 °C with 1.5 mM of tryptophan. At the same temperature the significant promoting effect (about 40% root and shoot fresh weight) of this strain on seedlings was observed. However, only MA DEM 7 had the ACC-deaminase activity with the highest efficiency in 9 °C and at this temperature synthesized IAA without tryptophan also at the same conditions the strain confirmed the strong promoting effect (about 40% root and 24% shoot fresh weight) on seedlings. Both strains synthesized GA in all tested terms and temperatures. Tested Mortierella strains had some important traits to consider them as microbial biofertilizers component improving plant growth in difficult temperate climate.

A field experiment was conducted for determination of crop coefficient (KC) and water stress coefficient (Ks) for wheat crop under different salinity levels, during 2015-16. Complete randomized block design of five treatments were considered, i.e., 0.51 dS/m (fresh water) as a control treatment and other four saline water treatments (4, 6, 8 and 10 dS/m), for S1, S2, S3 and S4 with three replications. The results revealed that the water consumed by plants during the different crop growth stages follows the order of FW>S1>S2>S3>S4 salinity levels. According to the obtained results, the calculated values of crop coefficients significantly differed from those suggested by FAO No.56 for the crops. The Ks values clearly differ from one stage to another because the salt stress causes both osmotic stress, due to a decrease in the soil water potential, and ionic stress which the average values of water stress coefficient (Ks) follows this order; FW(1.0)=S1(1.0)>S2(1.0)>S3(0.93)>S4(0.82). Overall, it was found the differences are attributed primarily to specific cultivar, the changes in local climatic conditions and seasonal differences in crop growth patterns. Thus, further studies are essential to determine the crop coefficient values under different variables, to make the best management practice (BMP) in agriculture.

Silicon is an essential trace nutrient for plant growth and is frequently employed to remediate soils contaminated with heavy metals in agriculture. However, silicon’s role and mechanism in reducing heavy metal toxicity have not been well understood, especially for multi-heavy metals. In this study, the effects of silicon-rich materials (silicate, rice husk biochar (RHB), and bentonite) on growth trait, antioxidant response, and heavy metal accumulation and distribution of wheat grown in two soils polluted by multiple heavy metals (Cd, Zn, Pb, and As) were investigated. The results revealed that the addition of silicon-rich materials enhanced plant growth, improved the photosynthetic attributes in leaf tissues, and decreased the contents of Cd, Zn, Pb, and As in wheat shoots and grains. The examination of the subcellular distribution of heavy metals in plants implied that silicon-rich materials transferred heavy metals as intracellular soluble fractions to the cell walls, indicating the reduction of mobility and toxicity of heavy metals in the plants. In addition, the application of the silicon-rich materials reduced oxidative damage in plants by downregulating plant antioxidant response systems and decreasing the production of malondialdehyde (MDA), ascorbic acid (AsA), and glutathione (GSH). Moreover, fractionation analysis of soil heavy metals showed that silicon-rich amendments could convert bioavailable heavy metals into immobilized forms. The results indicated that silicon-rich materials could remediate multi-heavy metal-polluted soils and promote wheat production.

Cadmium (Cd) is a toxic non–essential element to plants, and its accumulation in crops has significant adverse effects on human health. The cross–talk between copper (Cu) and Cd has been reported, but the molecular mechanisms remain unknown. Here, the function of wheat Cu transporter 3D (TaCOPT3D) in Cd tolerance was investigated. The TaCOPT3D transcripts significantly accumulated in wheat roots under Cd exposure. To test whether TaCOPT3D was involved in response to Cd stress, overexpressing lines with altered expression of TaCOPT3D transporters were compared to wild–type (WT) plants. The results showed that, under 20 μM Cd treatment, TaCOPT3D–overexpression lines exhibited more biomass and lower Cd accumulation in roots, shoots, and grains compared to WT plants. In addition, the TaCOPT3D–overexpression lines demonstrated less reactive oxygen species (ROS) and a greater amount of active antioxidant enzymes under Cd conditions than WT plants. Moreover, TaCOPT3D–overexpression lines highly reduced Cd accumulation under exposure to 20 μM Cu. The regulation pathway of TaCOPT3D in response to Cd stress was investigated, and transcription factor (TF) TaWRKY22, which targeted the TaCOPT3D promoter, was identified. Therefore, TaCOPT3D can serve as a candidate gene for decreasing Cd accumulation in wheat through genetic engineering.

Heat stress during the grain-filling period is the main abiotic stress factor limiting grain yield and quality in wheat (Triticum aestivum L.). In this study, 64 wheat genotypes were exposed to heat stress during reproduction caused by delayed sowing in two growing seasons. Grain yield, 1000 grain weight (GW), grain hardness (GH), and grain-quality related traits were investigated using wholemeal flour. Heat stress caused a significant decrease in GW through reducing starch content (SC) and a non-compensating rise in protein content (PC), and thereby resulted in lower yield. In addition, significant increases in flour water absorption (WA), Zeleny sedimentation volume (ZT), ash content (AC), lipid content (LC), loaf volume (LV), wet gluten content (WG), dry gluten content (DG), gluten index (GI), and amylopectin content (APC) were found following heat stress. In contrast, decreases in grain moisture content (MC) and amylose content (AMC) induced by heat stress were observed. The heat-tolerant genotypes were superior in grain yield, GW, SC, AMC, and MC. While the sensitive genotypes contained higher PC, LV, GI and AMP. A group of wheat genotypes characterized with a higher yield, AMC, GW, and SC as well as lower PC, WA, GH, ZT, and LV; and was found to be the most heat tolerant by principal component analysis. Decreases in the ratio of carbohydrates to proteins induced by heat stress, and lower protein content of normal grown wheat genotypes were observed. Therefore, lighter weight and smaller grains produce a smaller starchy endosperm with lower quality (less amylose) and higher grain protein content in heat stress compared to normal conditions. Heat stress caused by delayed sowing improves some of the baking-quality related traits. Whether this improvement in grain quality attributes will translate into better human health outcomes requires further investigation.

The SnRK gene family is a key regulator playing an important role in plant stress response by phosphorylating the target protein to regulate the signalling pathways. The function of SnRK gene family has been reported in many species but is limited to Triticum asetivum. In this study, SnRK gene family in the wheat genome was identified and its structural characteristics were described. One hundred forty-seven SnRK genes distributed across 21 chromosomes were identified in the Triticum aestivum genome and categorised into three subgroups (SnRK1/2/3) based on phylogenetic analyses and domain types. The gene intron-exon structure and protein-motif composition of SnRKs were similar within each subgroup but different amongst the groups. Gene duplication between the wheat, Arabidopsis, rice and barley genomes was also investigated in order to get insight into the evolutionary aspects of the TaSnRK family genes. SnRK genes showed differential expression patterns in leaves, roots, spike, and grains. Redundant stress-related cis-elements were also found in the promoters of 129 SnRK genes and their expression levels varied widely following drought, ABA and light regulated elements. In particular, TaSnRK2.11 had higher and increased expression under the abiotic stresses and can be a candidate gene for the abiotc stress tolerance. The findings will aid in the functional characterization of TaSnRK genes for further research.

The response of salt tolerant wheat genotype (Kharchia 65), and sensitive cultivars (HD2687, HD2009, WL711) to vegetative stage salinity stress (for 4 weeks) were studied at 1.1 (control), 9.1 (S1) and 14.2 (S2) dSm-1 salinity levels. Based on relative change in Membrane stability, PSII efficiency, retention of chlorophyll and carotenoid contents, Kharchia 65 showed better tolerance to salinity than other genotypes considered. To understand the role of different component mechanisms, expression of genes involved in ion exclusion, antioxidant defence and compatible osmolyte synthesis were analysed. Expression of SOS1 (plasma membrane Na+/H+ antiporter), NHX (vacuolar Na+/H+ antiporter), Ionic (sodium exclusion) and tissue tolerance (Sodium compartmentation, compatible solute accumulation and antioxidant defence) mechanisms were analysed in leaves of the genotypes after 4 weeks of salinity stress. Expression assay and the content of respective constituents indicated that apart from the well-known ion exclusion ability, Kharchia 65 also showed high level of tissue tolerance resulting in high early vigour and maintenance of growth rate afterwards. In Kharchia 65, sensing of salinity stress at plasma membrane activates NADPH Oxidase (RBOH) genes and generate ROS at apoplast. Apoplastic ROS triggers calcium influx and activates calcium signaling genes of SOS pathway (SOS1 and NHX). ROS generated from organelles chloroplast, peroxisome and mitochondria triggers cellular oxidative burst. ROS and calcium activates MAPK genes and downstream transcription factors, NAC and bZIP. MAPK signaling induces cellular antioxidant and compatible osmolyte biosynthesis and imparts tissue tolerance to salinity.

Due to increase demand of food grain in the world, assessment of yield before actual production is important in making policies and decisions in agricultural production system. For a large area, forecast models developed from vegetation indices derived from remote sensing satellite data possesses the potential to give quantitative and timely information on crops over large areas. Different vegetation indices are being made used for this purpose, however, their efficiency in estimating crop yield is needed to be certainly tested. In this study, wheat yield forecast was derived by regressing ground truthing yield data against time series of spatial vegetation indices for the 2013 to 2019 growing seasons. These spatial vegetation indices derived from Landsat 8 image data: Normalized Difference Vegetation Index (NDVI) and Soil Adjusted Vegetation Index (SAVI) were compared to evaluate the most appropriate index that performs better in forecasting wheat production at Karcag, Kunhegyes and Ecsegfalva settlements in Jász-Nagykun-Szolnok county, in the Northern Great Plain region of central Hungary. The best time for making wheat yield prediction with Landsat 8- SAVI and NDVI was found to be the beginning of ripening period (160th day of the year) with higher correlation between the vegetation indices and the wheat yield. The validation results revealed that the model from SAVI provides more consistent and accurate forecasts yield compared to NDVI. The SAVI model forecast yield for the validation years, 2018 and 2019 were within 6.00% and 4.41% of the final reported values while that of NDVI model were within 8.31% and 6.27%. Nash-Sutcliffe efficiency index is positive with E1= 0.99 for the model from SAVI and for NDVI, E1=0.57, which connote that the forecasting method developed and evaluated performs acceptable forecast efficiency.

The Septoria Leaf Blotch Complex (SLBC), caused by the two ascomycetes Zymoseptoria tritici and Parastagonospora nodorum, can reduce global yearly yield of wheat by up to 50%. In the last decade in Italy, SLBC incidence has increased; notably, durum wheat has proven to be more susceptible than common wheat. Field fungicide treatment can efficiently control these pathogens, but it leads to the emergence of resistant strains and adversely affects human and animal health, and the environment. Our previous studies indicated that active compounds produced by Trametes versicolor can restrict the growth of mycotoxigenic fungi and the biosynthesis of their secondary metabolites (e.g. mycotoxins). Specifically, we identified Tramesan: a 23 KDa -heteropolysaccharide secreted by T. versicolor that acts as a pro-antioxidant molecule in animal cells, fungi, and plants. Foliar-spraying of Tramesan (3.3 µM) in SLBC-susceptible varieties of durum significantly diminished symptoms of Stagonospora Nodorum Blotch (SNB) and Septoria Tritici Blotch (STB) by 75% and 65%, respectively. Tests were conducted under controlled conditions as well as in field. We show that Tramesan elicits wheat defence against SNB and STB augmenting the synthesis of defence-related hormones, notably JA and SA, that in turn switch on the expression of markers of defence (PR1, PR4 inter alia). In field experiments, yield of durum wheat plants treated with Tramesan was similar to that of untreated ones. The results suggest the use of Tramesan for protecting durum wheat against SLBC.

High antioxidants level in food is gradually becoming popular because of the enhanced risk of oxidative stress in humans. Bread wheat is rich in vital antioxidants but its major bioactive compounds are not available for the human. This study was conducted with the aim to enhance the phytochemical constituents and antioxidative activity of wheat grains by fermenting it with the use of Bacillus subtilis KCTC 13241. The antioxidative potential was determined by DPPH (2,2-diphenyl-1-picryl- hydrazyl) and ABTS (3-ethyl-benzothiazo- line-6-sulfonic acid) radical scavenging assay as well by the concentration of amino acids, flavonoids, minerals, carbohydrates, and phenolic compounds. Different varieties showed different free radical scavenging potential on fermentation, which was significantly high with respect to their corresponding unfermented wheat varieties. The highest potential was found in a fermented wheat variety named Namhae and this combination can be used in the pharmaceutical and food industries.

Due to the global warming and dynamic changes in pathogenic virulence, leaf rust caused by Puccinia triticina has greatly expanded its epidermic region and become a severe threat to global wheat production. Genetic bases of wheat resistance to leaf rust mainly relies on the leaf rust resistance (Lr) gene or quantitative trait locus (QLr). Although these genetic loci have been insensitively studied during the last two decades, an updated overview of Lr/QLr in a genome-wide level is urgently needed. This review summarized recent progresses in genetic studies of wheat resistance to leaf rust. Wheat germplasms with great potentials in genetic improvement of resistance to leaf rust were highlighted. Key information about the genetic loci carrying Lr/QLr were summarized. A genome-wide chromosome distribution map for all the Lr/QLr was generated based on the released wheat reference genome. In conclusion, this review has provided valuable sources for both wheat breeders and researchers to understand the genetics of resistance to leaf rust in wheat.

Application of chemical pesticides to protect agricultural crops from pests and diseases is discouraged due to their harmful effects on human and environment. Therefore, alternative approaches for crop pro-tection through microbial or microbe originated pesticides have been gaining momentum. Wheat blast is a destructive fungal disease caused by Magnaporthe oryzae Triticum (MoT) pathotype, which poses a seri-ous threat to global food security. Screening of secondary metabolites against MoT revealed that antimy-cin A isolated from a marine Streptomyces sp. had significant inhibitory effect on mycelial growth in vitro. This study aimed to investigate the inhibitory effects of antimycin A on some critical life stages of MoT and evaluate the efficacy of wheat blast disease control by this natural product. Bioassay indicated that antimycin A suppressed mycelial growth, conidiogenesis, germination of conidia and formation of ap-pressoria in germinated conidia of MoT in a dose-dependent manner with minimum inhibitory concen-tration 0.005 μg/disk. If germinated, antimycin A induced abnormal germ tubes (4.8%) and suppressed the formation of appressoria. Interestingly, application of antimycin A significantly suppressed wheat blast disease in both seedling and heading stages of wheat supporting the results from invitro study. This is the first report on inhibition of mycelial growth, conidiogenesis, conidia germination, detrimental morphological alterations in germinated conidia, and suppression of wheat blast disease caused by a Triticum pathotype of M. Oryzae. Further study is required to unravel the precise mode of action of this promising natural compound for considering it as a biopesticide to combat wheat blast.

Fusarium graminearum is a dangerous pathogen of the cereals producing mycotoxins (trichothecene and zearalenone) harmful for human and animal health. There were evaluated sixteen winter wheat varieties for their response in conditions of natural infection with F. graminearum in the epidemic year 2019, being well known that accumulation of mycotoxins (DON, ZON and T-2) is induced by different biotic and abiotic factors. Field plot was organized in Latin rectangle randomized with three replicates. For all evaluated wheat varieties were collected field data (incidence, severity and infection degree of the fungus F. graminearum) and laboratory data (mycotoxins concentration in grains) that have been processed using the software JASP (Version 0.14) for descriptive statistics, and exploratory factor analysis (EFA). Microsoft Excel 2019 was used to calculate Pearson’s correlation coefficients. The results showed negative corelation between plants’ density and F. graminearum attack frequency. Positive correlations were found between DON and T-2 and between DON and fungus attack intensity. This work highlights that during a F. graminearum epidemic year some of the most influential factors in the contamination with harmful mycotoxins (DON, ZON and T-2) are: plants density, frequency of the attack on ear, diseased ears and attack intensity on ears.

Fusarium head blight (FHB) can cause contamination of cereal grain with mycotoxins. Winter wheat is also infected with FHB. It is more resistant than durum wheat to head infection and less than other small grain cereals. The aim of this study was to identify winter wheat lines that combine low head infection and kernel damage with low toxin contamination. Resistance to FHB of 27 winter wheat lines and cultivars was evaluated over a three-year experiment established in two locations. At the anthesis stage, heads were inoculated with Fusarium culmorum isolates. The FHB index was scored and the percentage of Fusarium-damaged kernels (FDKs) assessed. The grain was analysed for type B trichothecenes (deoxynivalenol and derivatives, nivalenol) and zearalenone content. The average FHB index was 12.9%. The proportion of FDK was 6.9% (weight) and 8.5% (number). An average content of deoxynivalenol amounted to 3.543 mg/kg and nivalenol to 2.115 mg/kg. In total, it was 5.804 m/kg of type B trichothecenes. The zearalenone content in the grain was 0.214 mg/kg. Relationships between FHB index, FDK and mycotoxin contents were highly significant for wheat lines; however, they were stronger for FDK versus mycotoxins. Breeding lines combing all types of FHB resistance were found, and five of them had resistance similar to that of wheat lines with the Fhb1 gene.

Reticulate evolution through the interchanging of genetic components across organisms can impact significantly on the fitness and adaptation of species. Bread wheat (Triticum aestivum subsp. aestivum) is one of the most important crops in the world. Allopolyploid speciation, frequent hybridization, extensive introgression, and occasional horizontal gene transfer (HGT) have been shaping a typical paradigm of reticulate evolution in bread wheat and its wild relatives, which is likely to have a substantial influence on phenotypic traits and environmental adaptability of bread wheat. In this review, we outlined the evolutionary history of bread wheat and its wild relatives with a highlight on the interspecific hybridization events, demonstrating the reticulate relationship between species/subspecies in the genera Triticum and Aegilops. Furthermore, we discussed the genetic mechanisms and evolutionary significance underlying the introgression of bread wheat and its wild relatives. An in-depth understanding of the evolutionary process of Triticum species should be beneficial to future genetic study and breeding of bread wheat.

Water and nutrients shortage threatens agricultural sustainability in many arid and semiarid areas of the world. It is unknown whether improved water and nutrient conservation practices can be developed to alleviate this issue while increasing crop productivity. In this study, experimental work included the application of straw, biochar and N fertilizer. The straw and biochar were applied alone or combined with N fertilizer (0 and 100 kg N ha-1). Application of biochar and straw in combination with N fertilizer caused a reduction in mean soil temperature by an average of 20.05% and 18.10% relative to soils without carbon. Biochar and straw– amended soils significantly (P < 0.05) increased soil moisture content by 11.04% and 13.68% compared to no carbon treatments. Statistically comparable temperatures and moisture were recorded for both straw and biochar treated plots. Both biochar and straw treated soils produced the lowest bulk density (0–5 cm) at 1.15 g cm–3, and no carbon soils the highest at 1.20 g cm–3. The improved soil quality translated into higher biomass in the biochar (1906 kg ha–1) and biomass (1643 kg ha–1) and soils without carbon the lowest at 1553 kg ha–1. The improvement of soil moisture and the optimization of soil temperature for the two residue treated soils allow us to conclude that combined application of biochar and straw at the rate used in this study can be used as an effective farming model in alleviating water and nutrient shortage in semiarid environments.

Flowering time is a critical stage for crop development as it regulates the ability of plants to adapt to an environment. To understand the genetic control of flowering time, a genome wide association study (GWAS) was conducted to identify the genomic regions associated with the control of this trait in durum wheat (Triticum durum Desf.). A total of 96 landraces and 288 modern lines were evaluated for days to heading, growing degree days, and accumulated day length at flowering across 13 environments spread across Morocco, Lebanon, Mauritania, and Senegal. These environments were grouped into four pheno-environments based on temperatures, day length and other climatic variables. Genotyping with 35K Axiom array generated 7,652 polymorphic SNPs in addition to 3 KASP markers associated to known flowering genes. In total, 34 significant QTLs were identified in both landraces and modern lines. Some QTLs had strong association with already known regulatory photoperiod genes, Ppd-A and Ppd-B and vernalization genes Vrn-A1, and Vrn3. However, these loci explained only 5 to 20% of variance for days to heading. Seven QTLs overlapped between the two germplasm groups in which Q.ICD.Eps-03 and Q.ICD.Vrn-17 consistently affected flowering time in all the pheno-environments, while Q.ICD.Eps-11 and Q.ICD.Ppd-12 were significant only in two pheno-environments and the combined analysis across all environments. These results help clarify the genetic mechanism controlling flowering time in durum wheat and show some clear distinctions to what is known for common wheat (Triticum aestivum L.)

Cistus incanus L. (CI) has been proposed as an innovative functional supplement of food products, and hence the present study aimed to evaluate the effect of the addition of dried CI on the properties of bread. Bread was prepared from white wheat flour supplemented with the addition of 1%, 2%, 3%, 4%, and 5% of ground CI. After the completion of baking process, various characteristics of the obtained bread product, such as yield, volume, porosity, acidity, color, and texture, were evaluated. In addition, total phenolic content (TPC), ABTS radical scavenging activity, CHEL chelating power, and ability to quench OH∙ radicals were measured. The results showed that the addition of CI to bread caused a reduction in the volume of bread, but texture of the crumbs was acceptable. Acidity and moisture content of bread were found to be increased following CI enrichment. Significant changes in the ash content and the color of bread crumbs were also observed. Bread incorporated with CI was characterized by significantly higher TPC and much higher antioxidant activity, as measured by ABTS, CHEL, and OH∙ radicals, compared to control bread. Supplementation of bread with 3% CI produced a product with desirable characteristics which was also favored by consumers.

The wheat aleurone layer is, according to millers, the main bran fraction. It is a source of nutritionally valuable compounds, such as dietary fibres, proteins, minerals and vitamins, that may exhibit health benefits. Despite these advantages, the aleurone layer is scarce on the market, probably due to issues related to its extraction. Many processes exist with some patents, but a choice must be made between the quality and quantity of the resulting product. Nonetheless, its potential has been studied mainly in bread and pasta. While the nutritional benefits of aleurone-rich flour addition to bread agree, opposite results have been obtained concerning its effects on end-product characteristics (namely loaf volume and sensory characteristics), thus ensuing different acceptability responses from consumers. However, the observed negative effects of aleurone-rich flour on bread dough could be reduced by subjecting it to pre- or post-extracting treatments meant to either reduce the particle size of the aleurone’s fibres or to change the conformation of its components.

Cold stress is a major environmental factor affecting the growth, development, and productivity of various crop species. With the current trajectory of global climate change, low temperatures are becoming more frequent and can significantly decrease crop yield. Wheat (Triticum aestivum L.) is the first crop to be domesticated and is the most popular cereal crop in the world. Due to a lack of systematic research on cold response pathways and gene regulatory networks, the underlying molecular mechanisms of cold signal transduction in wheat are poorly understood. This study reviews recent progress in wheat, including the ICE-CBF-COR signaling pathway in cold stress and the effects of cold stress on hormonal pathways, reactive oxygen species (ROS), and epigenetic regulation. This review also highlights outstanding issues that are crucial for understanding the interactions between wheat and low-temperature conditions.

Plant height is significantly correlated with grain traits, which is a component of wheat yield. The purpose of this study is to investigate the main QTLs that control plant height and grain-related traits in multiple environments. In this study, we constructed a high-density genetic linkage map using the Wheat50K SNP Array to map quantitative trait loci (QTLs) for these traits in 198 recombinant inbred lines (RILs). The two ends of the chromosome were identified as re-combination-rich areas in all chromosomes except chromosome 1B. The middle area of the chro-mosomes was identified as the recombination-barren area. Both the genetic map and the physical map showed a significant correlation when p=0.001, with a correlation coefficient between 0.63 and 0.99. However, there was almost no recombination between 1RS and 1BS. In terms of plant height, 1RS contributed to the reduction of plant height by 3.43cm. In terms of grain length, 1RS contributed to the elongation of grain by 0.11mm. A total of 43 QTLs were identified, including 8 QTLs for Plant height(PH), 11 QTLs thousand grain weight(TGW), 15 QTLs for grain length(GL),and 9 QTLs for grain width(GW), which explained 1.36%–33.08% of the phenotypic variation. Seven were environment-stable QTLs, including two loci Qph.nwafu-4B and Qph.nwafu-4D that determined plant height. The explanation rates of phenotypic variation were 7.39%-12.26% and 20.11%-27.08%, respectively. One QTL, Qtgw.nwafu-4B, which influenced TGW, showed an explanation rate of 3.43%-6.85% for phenotypic variation, two co-segregating KASP markers were developed, the physical locations corresponding to KASP_AX-109316968 and KASP_AX-109519968 were 25.888344 MB and 25.847691 MB. Another QTL, Qgw.nwafu-4D, which determined grain width, had an explanation rate of 3.43%-6.85%. Three loci that affected the grain length were Qgl.nwafu-5A, Qgl.nwafu-5D.2 and Qgl.nwafu-6B, illustrating the explana-tion rates of phenotypic variation as 6.72%-9.59%, 5.62%-7.75%, and 6.68%-10.73%, respectively. Two QTL clusters were identified on chromosomes 4B and 4D.

In order to better understand the source-sink flow and relationships of Zinc (Zn) and other nutrients in wheat (Triticum aestivum L.) plants for biofortification and improving grain nutritional quality, effects of reducing photoassimilate source (through the flag leaf removal and spike shading) or sink (through 50% spikelets removal) in the field on accumulation of Zn and other nutrients in wheat grains of two cultivars (Jimai 22 and Jimai 44) were investigated under two soil Zn application levels. The single panicle weight (SPW), kernel number per spike (KNPS), thousand kernel weight (TKW), total grain weight (TGW), concentrations and yields of various nutrient elements (Zn, Fe, Mn, Cu, N, P, K, Ca and Mg), phytate phosphorus (phytate-P), phytic acid (PA) and phytohormones (ABA: abscisic acid, and the ethylene precursor ACC: 1-aminocylopropane-1-carboxylic acid), and C/N ratios were determined. Soil Zn application significantly increased concentrations of grain Zn, N and K. Cultivars showing higher grain yields had lower grain protein and micronutrient nutritional quality. SPW, KNPS, TKW (with an exception of TKW in half spikelets removal), TGW, and nutrient yields in wheat grains were most severely reduced by half spiklets removal, secondly by spike shading, and slightly by flag leaf removal. Grain concentrations of Zn, N and Mg consistently showed negative correlations with SPW, KNPS and TGW, but positively with TKW. There were general positive correlations among grain concentrations of Zn, Fe, Mn, Cu, N and Mg, and bioavailability of Zn and Fe (estimated by molar ratios of PA/Zn, PA × Ca/Zn, PA/Fe, or PA × Ca/Fe). Although concentrations of Zn and Fe were increased and Ca was decreased in treatments of half spikelets removal and spike shading, the simultaneously increased PA limited the increase in bioavailability of Zn and Fe. In general, different nutrient elements interact with each other and are affected to different degrees by source-sink manipulations. Elevated endogenous ABA levels and ABA/ACC ratios were associated with increased TKW and grain-filling of Zn, Mn, Ca and Mg, and inhibited K in wheat grains. However, effects of ACC were diametrically opposite. These results provide basis for wheat grain biofortification to alleviate human malnutrition.

Bread wheat (Triticum aestivum L.) is one of the most important cereal crops for food security. Of all the stresses that curtail wheat productivity, drought has the most detrimental effects. Especially terminal drought stress i.e. at the time of flowering imposes a big challenge to sustain grain production. In the current study, 339 pre-breeding lines derived from three-way crosses of exotics x elite lines were evaluated in the irrigated and drought stress environments at Obregon, Mexico for the year 2016 and 2018. Drought significantly reduced yield (Y), spike length (SL), number of grains per spikes (NGS) and thousand kernel weight (TKW) by 46.4, 19.2, 23.5 and 25.9%, respectively in comparison to irrigated conditions. Kernel abortion (KA), highly correlated with Y, increased significantly (11.6%) under drought stress environment. Population structure analysis in this panel revealed three sub-populations and a genome wide linkage disequilibrium (LD) decay was at 2.5 cM. Single marker and haplotypes-based genome wide association study (GWAS) revealed significant associations on three chromosomes; 4A (HB10.7), 2D (HB6.10) and 3B (HB8.12) with Y, SL and TKW, respectively. Likewise, associations on chromosomes 6B (HB17.1) and 3A (HB7.11) were identified for NGS and on 3A (HB7.12) for KA. Five traits i.e. normalized difference vegetation index (NDVI), canopy temperature depression (CTD) days to heading (DTH), NGS, KA were associated at chromosome 3A both under irrigated and drought conditions however, different haplotypes were estimated. Twenty-six SNPs were part of 10 haplotype blocks associated with Y, SL, TKW, NGS and KA. In silico analysis of the associated SNPs/haplotypes showed hits with candidate genes known to confer abiotic stress resistance in model species and crops. Potential candidate genes include those coding for sulfite exporter TauE/SafE family in Arabidopsis thaliana, TBC domain containing protein in Oryza sativa subsp. Japonica and heat shock proteins in Aegilops tauschii subsp. tauschii were revealed. The SNPs linked to the promising genes identified in the study can be used for marker-assisted selection.

Background: Wheat root rot disease due to soil-borne fungal pathogens leads to tremendous yield losses worth billions of dollars worldwide every year. It is very important to study the relationship between rhizosphere fungal diversity and wheat roots to understand the occurrence and development of wheat root rot disease. Results: A significant difference in fungal diversity was observed between the diseased and healthy groups in the heading stage, but the trend was the opposite in the filling stage. The abundance of most genera with high richness decreased significantly from the heading to the filling stage in the diseased groups; the richness of approximately one-third of all genera remained unchanged, and only a few low-richness genera, such as Fusarium and Ceratobasidium, had a very significant increase from the heading to the filling stage. In the healthy groups, the abundance of most genera increased significantly from the heading to the filling stage; the abundance of some genera did not change markedly, or the abundance of very few genera increased significantly. Physical and chemical soil indicators showed that low soil pH and density, increases in ammonium nitrogen, nitrate nitrogen and total nitrogen contributed to the occurrence of wheat root rot disease. Conclusions: Our results revealed that in the early stages of disease, highly diverse rhizosphere fungi and a complex community structure can easily cause wheat root rot disease. The existence of pathogenic fungi is a necessary condition for wheat root rot disease, but the richness of pathogenic fungi is not necessarily important. The increases in ammonium nitrogen, nitrate nitrogen and total nitrogen contributed to the occurrence of wheat root rot disease. Low soil pH and soil density are beneficial to the occurrence of wheat root rot disease.

In order to realize rapid and nondestructive monitoring of wheat biomass in field, field experiments based on different densities, nitrogen fertilizer and variety treatments were studied. RGB images of wheat in the main growth stage were obtained by UAV, and wheat color and texture feature indices were obtained by image processing, and wheat biomass was obtained by field sampling in the same period. Then the relationship between different color and texture feature indices and wheat biomass was analyzed to select the color and texture feature index suitable for wheat biomass estimation. The results showed that there was a high correlation between image color index and wheat biomass in different stages, and most of them reached a very significant correlation level. However, the correlation between image texture feature index and wheat biomass was poor, only a few indexes reached significant or extremely significant correlation level. Based on the above results, the color indices with the highest correlation to wheat biomass or the combining indices of color and texture feature in different growth stage were used to construct estimation model of wheat biomass. The models were validated using independently measured biomass data, and the correlation between simulated and measured values reached the significant level, RMSE were smaller. This indicated that the estimated results by the models were reliable and accurate. It also showed that the estimation models of wheat biomass combined with color and texture feature indices of UAV image were better than the single color index models. The results would provide a new method for real-time monitoring of wheat field growth and biomass estimation.

Powdery mildew caused by Blumeria graminis f. sp. tritici (Bgt) is one of many severe diseases that threaten bread wheat (Triticum aestivum L.) yield and quality worldwide. The discovery and deployment of powdery mildew resistance genes (Pm) can prevent this disease epidemic in wheat. In a previous study, we transferred the powdery mildew resistance gene Pm57 from Aegilops searsii into common wheat and cytogenetically mapped the gene in a chromosome region with the fraction length (FL) 0.75-0.87, which represents 12% of 2Ss#1 segment on the long arm of chromosome 2Ss#1. In this study, we performed RNA-Seq on three infected and mock-infected wheat-Ae. searsii 2Ss#1 introgression lines with Bgt-isolates inoculation at 0, 12, 24, and 48 hours after inoculation. Then we designed 79 molecular markers based on transcriptome sequences and physically mapped them to Ae. searsii chromosome 2Ss#1- in seven intervals. We used these markers to identify 46 wheat-Ae. searsii 2Ss#1 recombinants induced by ph1b, a deletion mutant of pairing homoelogous (Ph) genes. Analysis of the 46 ph1b-induced 2Ss#1L recombinants with different Bgt-responses using 28 2Ss#1L-specific molecular markers in the interval FL0.72-0.87 where Pm57 is located, and the flanking intervals, we physically mapped Pm57 gene on the long arm of 2Ss#1 in a 5.13 Mb genomic region, which was flanked by markers X67593 (773.72 Mb) and X62492 (778.85 Mb). By comparative synteny analysis of the corresponding region on chromosome 2B in Chinese spring (T. aestivum L.) with other model species we identified ten genes that are putative plant defense-related (R) genes which includes six coiled-coil nucleotide-binding site-leucine-rich repeat (CNL), three nucleotide-binding site-leucine-rich repeat (NL) and a leucine-rich receptor-like repeat (RLP) encoding proteins. This study will lay a foundation for further cloning of Pm57, and benefit the understanding of interactions between resistance genes of wheat and powdery mildew pathogens.

Development and deployment of wheat varieties with high yields, wide adaptability, good quality, multiple-resistance to abiotic and biotic stresses, and efficient response to fertilizers have greatly contributed to global wheat sustainable production. The genomic composition of key commercial wheat variety can help understand the genetic basis underlying the development of new variety and permit increased breeding efficiency. In this study, we report the chromosomal and genomic compositions of BN207, presently the leading wheat variety in the southern region of Huang-Huai River Valley, the most important wheat producing area in China through an integrated analysis using fluorescent in situ hybridization (FISH) and wheat 15 K SNP array. Our results showed that BN207 inherited 55.3% and 40.7% of its genome from its male parent BN64 and female parent ZM16, respectively, and generating 64 novel or recombined loci. Besides, we detected nine chromosomal variations in Bn207 and its parents and ten sister lines, and physically mapped two variations, the pericentric inversion of chromosome 6B, and large tandem repeat sequence block at the long arm of 5A, both had positive effects on agronomic traits, by integration of FISH and SNP loci recombination analyses. These results will provide a reference for breeding of high yield wheat varieties as BN207, and the application of founder parents BN64 and ZM16, which are being utilized frequently in wheat breeding programs in Henan Province and surrounding areas.

Macro and micronutrients, essential for the maintenance of human metabolism, are daily assimilated through the diet. Wheat and other major cereals are a good source of nutrients, such as carbohydrates and proteins, but cannot supply enough amounts of essential micronutrients which includes provitamin A. As vitamin A deficiency (VAD) lead to several serious diseases spread worldwide, the biofortification of a major staple crop, such as wheat, represents an effective way to preserve human health in developing countries. In the present work, a key enzyme involved in the branch of carotenoids pathway producing β-carotene, lycopene epsilon cyclase, has been targeted by a TILLING approach in a “Block strategy” perspective. The null mutant genotype showed a strong reduction in the expression of lcyE gene and also interesting pleiotropic effects on an enzyme (β-ring hydroxylase) acting downstream in the pathway. Biochemical profiling of carotenoids in the wheat mutant lines showed an increase of roughly 75% in β-carotene in the grains of the complete mutant line vs. the control. In conclusions, here we describe the production and the characterization of a new wheat line biofortified in provitamin A obtained through a non-transgenic approach also shading new light on the molecular mechanism governing carotenoids biosynthesis in durum wheat.

Spike shape and morphometric characteristics are among the key characteristics of cultivated cereals associated with their productivity. Identification of the genes controlling these traits requires morphometric data harvesting and analysis for numerous plants, which is automatable using technologies of digital image analysis. A method for wheat spike morphometry utilizing 2D image analysis is proposed. Digital images are acquired in two variants: a spike on a table (one projection) or fixed with a clip (four projections). The method identifies spike and awns in the image and estimates their quantitative characteristics (area in image, length, width, circularity, etc.). Models of sections, quadrilaterals, and radial model are proposed for describing spike shape. Parameters of these models are used to predict spike shape type (spelt, normal, or compact) by machine learning. The mean error in spike density prediction for the images in one projection is 4.61 (~18%) versus 3.33 (~13%) for the parameters obtained using four projections. F1 measure in automated spike classification into three types is 0.78 using logistic regression (one projection) and 0.85 using random forest method (four projections). The early-season area estimation of the winter wheat crop as a strategic product is important for decision makers. Classification of multi-temporal images is an approach which is affected by many factors like appropriate training sample size, proper frequency and acquisition times, vegetation indices (VIs) type, temporal gradient of spectral bands and VIs, appropriate classifier and missed values because of cloudy conditions. This paper addresses the impact of appropriate frequency and acquisition times and VIs type along with the spectral and VI gradient on random forest (RF) classifier when missed values exist in multi-temporal images. To investigate the appropriate temporal resolution for image acquisition, the study area was selected on an overlapping area between two LDCM paths. In our developed method, the miss values of cloudy bands for each pixel are retrieved by the mean of k-nearest ordinary pixels. Then the multi-temporal image analysis is performed by considering different scenarios provided by decision makers in terms of desired crop types that should be extracted at early-season in the study areas. The classification results obtained by the RF decrease by 1.6% when temporally missed values retrieved by the proposed method, which is an acceptable result. Moreover, the experimental results demonstrated that if temporal resolution of Landsat 8 increased to one week the classification task can be conducted earlier with almost better results in terms of OA and kappa. The impact of incorporating VIs along with the temporal gradients of spectral bands and VIs as new features in RF demonstrated that the OA and Kappa are improved 3.1% and 6.6%, respectively. Furthermore, the obtained result showed that if only one image from seasonal changes of crops is available, the temporal gradient of VIs and spectral bands play the main role to discriminate remarkably wheat from barley. The experiments also demonstrated that if both wheat and barley merge to a single class the crop area can be estimated two months earlier with 97.1 and 93.5 in terms of OA and kappa, respectively.

IgE-mediated wheat allergy is a gluten-related disorder. Wheat is one of the five most common food allergens in children. However, the natural history of IgE-mediated wheat allergy has seldom been described in the research literature. This study presents the current state of knowledge about the IgE-mediated wheat allergy in children.

Common root rot caused by Bipolaris sorokiniana infestation of wheat is one of the main reasons of yield reduction in wheat crops worldwide. In current study, strain JK-25 was isolated from soil of wheat rhizosphere and identified as Bacillus halotolerans based on morphological, physiological, biochemical characteristics and molecular identification. The strain showed significant antagonism to B.sorokiniana and broad-spectrum resistance to Fusarium oxysporum, Fusarium graminearum and Rhizoctonia zeae. Inhibition of Bipolaris sorokiniana mycelial dry weight and spore germination rate by JK-25 fermentation supernatant reached 60% and 88% respectively. The crude extract of JK-25 was found by MALDI-TOF-MS to contain the surfactin that exerted an inhibitory effect on B.sorokiniana. The disruption of mycelial cell membranes was observed under microscopy (LSCM) after treatment of B.sorokiniana mycelium with the crude extract. The antioxidant enzyme activity of B.sorokiniana was significantly reduced and the oxidation product MDA content increased after treatment with the crude extract. The incidence of root rot was significantly reduced in pot experiments with the addition of JK-25 culture ferment, which had a significant biological control effect of 72.06%. Its ability to produce siderophores may help to promote wheat growth, and the production of proteases and pectinases may also be part of the strain's role in suppressing pathogens. These results demonstrate the excellent antagonistic effect of JK-25 against B.sorokiniana and suggest that this strain has great potential as a resource for biological control of wheat root rot strains.

During the 1970s, Tunisian durum wheat landraces were replaced progressively by modern cultivars. These landraces are nowadays maintained by smallholder farmers in some ecological niches and are threatened gradually by extinction resulting on the narrowing of the genetic diversity. This study aims to investigate patterns of phenotypic variability using twelve quantitative traits in a panel of 189 durum wheat landraces and seven checks, based on farmer’s population name attribution and genetic structure. Our results showed high phenotypic variability among and within landraces and checks for the majority of the studied traits. The principal components analysis showed similar grouping using farmers name attribution and genetic structure using K = 6, which confirmed the identification of a new gene pool in the oases of Tunisia, represented by the sub-population Jenah Zarzoura and the robustness and high relationships between phenotypic and genome-wide genetic structure using DArTseq method. These findings will enhance the conservation efforts of these landraces and their use in breeding efforts at national and international levels to adapt to dry conditions.

Rice wheat cropping system prevailed in more than fifty percent areas of rice belt of central and upper Punjab. In this area yield of wheat is curtailed every year due to late harvesting of rice crop. In addition to this, lack of machinery for direct sowing of wheat into combine harvested rice is a major limitation for timely wheat sowing due to presence of heavy straw load dropped by the combine harvester. A field experiment was planned and executed during two consecutive years 2017-18 and 2018-19 at Agronomic Research Station Farooqabad to evaluate the various stubble management techniques for the sowing of wheat in a rice-wheat cropping system. The Experiment consist of four treatments Happy seeder (Zero tillage), conventional sowing without burning of rice stubble, Rotavator two times, and Conventional sowing with the burning of rice stubble. The trail was arranged as a Randomized Complete Block Design with four replications having a net plot size of 8 m x 20 m. The Treatments have significant effects on Plant height, Number of productive tillers, 1000- grain weight, and grain yield. Maximum plant height (94.74 and 94.25 cm), numbers of productive tillers/m2 (324.00 and 322.25), 1000-grain weight (36.35 and 36.20 g), grain yield (3780 and 3613 kg/ha), and benefit-cost ratio (2.54 and 2.43) were recorded from the treatment happy seeder (Zero tillage) during both years respectively. It is concluded that the sowing of wheat with a happy seeder (Zero tillage) is an appropriate technique in the rice-wheat cropping system by protecting the environment and soil health from the burning of rice stubbles.

Heat stress occurring during the reproductive stage of wheat has a detrimental effect on productivity. A durum wheat core set was exposed to simulated terminal heat stress by applying plastic tunnels at the time of flowering over two seasons. Mean grain yield was reduced by 54% compared to control conditions, and grain number was the most critical trait for tolerance to this stress. The combined use of tolerance indices and grain yield identified five elites: Kunmiki, Berghouata1, Margherita2, IDON37-141, and Ourgh. The core set was also subjected to genome wide association study using 7,652 polymorphic SNPs markers. The most critical genomic regions were identified in association with spike fertility and tolerance indices on chromosome 1A, 5B and 6B. Haplotype analysis on a set of 208 elites confirmed that lines that carried the positive allele at all three QTLs resulted in a yield advantage of 8% when field tested under daily temperatures above 31° C. Two of the QTLs were successfully validated into KASP markers and explained >10% of the phenotypic variation for an independent elite germplasm set. These genomic regions can now be readily deployed via breeding to improve resilience to climate change and increase productivity in heat-stressed areas.

Dryland winter wheat in Loess Plateau is facing yield reduction due to shortage of soil moisture and delayed sowing time. Field experiment was conducted at Loess Plateau in Shanxi Province, China from 2012 to 2014, to study the effect of subsoiling and conventional tillage and different sowing dates on the soil water storage and contribution of N accumulation and remobilization to yield of winter wheat. The results showed that subsoiling significantly improved the soil water storage at 0-300 cm depth, improved the number of tillers and pre-anthesis N translocation in various organs of wheat and post-anthesis N accumulation, eventually increased the yield up to 17-36%. Delaying sowing time had reduced the soil water storage at sowing and winter accumulated temperature by about 180˚C. The contribution of N translocation to grain yield was maximum in glume+spike followed by in leaves and minimum by stem+sheath. In addition a close relationship was found between the N accumulation and translocation and the soil moisture in the 20-300 cm. Subsoiling during the fallow period and the medium sowing date was beneficial for improving the soil water storage and increased the N translocation to grain, thereby increasing the yield of wheat, especially in dry year.

Although wheat is a staple food for most of human population, some of its components trigger adverse reactions. Among wheat components, the alpha-amylase/trypsin inhibitors (ATI) are important triggers of several allergies and activators of innate immunity. ATI are a group of exogenous protease inhibitors and include several polypeptides. The three ATI polypeptides named CM3, CM16 and 0.28 are considered major allergens, and might also play a role in other common wheat-related pathologies, such as Non Celiac Wheat Sensitivity and even Celiac Disease. On this basis, we pointed to obtain high amounts of them in purity and evaluating their allergenicity potential. We thus isolated the mRNA corresponding to the three ATI genes CM3, CM16 and 0.28 from 28 days post-anthesis wheat kernels and the corresponding cDNAs were used for heterologous expression in Pichia pastoris. The three purified proteins were tested in degranulation assay against human sera of patients with food allergy to wheat. A large range of degranulation values was observed for each protein according to the sera tested. All the three purified proteins CM3, CM16 and 0.28 were active as allergens because able to induce basophils degranulation on wheat allergic patients’ sera, but the highest values of β-hexosaminidase release were observed for CM3 protein.

A remote sensing-based evapotranspiration (ET) study was conducted over the Central Arizona Irrigation and Drainage District (CAIDD), an Arizona agricultural region. ET was assessed means for 137 wheat plots, 183 cotton plots, and 225 alfalfa plots. The remote sensing ET models were the Satellite-Based Energy Balance for Mapping Evapotranspiration with Internalized Calibration (METRIC), the Two Source Energy Balance (TSEB), and Vegetation Index ET for the US Southwest (VISW). Remote sensing data were principally Landsat 5, supplemented by Landsat 7, MODIS Terra, MODIS Aqua, and ASTER. The models produced similar daily ET for wheat, with 6–8 mm/d mid-season. For cotton and alfalfa daily ET showed greater differences, where TSEB produced largest daily ET, METRIC the least, and VISW in the midrange. Modeled cotton ET at mid-season ranged from 9.5 mm/d (TSEB), to 8 mm/d (VISW), and 6 mm/d (METRIC). For alfalfa ET, values at peak cover ranged from 8 mm/d (TSEB), 6 mm/d (VISW), and 5 mm/d (METRIC). Model bias ranged −10% to +18%. Relative to potential ET, FAO-56 ET, and USDA-SW gravimetric-ET, model variability ranged from negligible to 35% of annual crop water use. Model averaging was found a useful way to consider and reconcile all ET estimates.

Iron (Fe) and zinc (Zn) deficiency in cereal grains has deleterious effects on the health of millions of people, especially in developing countries. As wheat, as a staple crop, is consumed in large quantities, its micronutrient content is important. Crops in Africa are often grown under low nitrogen (N) and low phosphorous (P) conditions. The aim of this study was to determine the effect of low N and low P stress on Fe and Zn and phytic acid concentration, in two commercial spring wheat cultivars with excellent baking quality. The two cultivars did not differ significantly for the measured characteristics. Across all treatments the average values for Fe varied between 19.60-28.61 mg kg^-1, Zn between 17.68-33.79 mg kg^-1 and phytic acid between 5.03-6.92 mg g^-1. Low P stress lead to the highest values of Fe and Zn, and the lowest value for phytic acid. Phytic acid:Fe and phytic acid:Zn ratios were also highly significantly reduced under low P stress conditions. Low N conditions caused significantly increased Zn levels. Despite this, the phytic acid:Fe and phytic acid:Zn ratios were relatively high under all conditions, indicating a low bioavailability of both Fe and Zn in these wheat cultivars.

Conservation agriculture (CA) could be very effective for agricultural sustainability under Mediterranean environments, where farmers rely on short rotation based on durum wheat. In this work we investigated the effect of different combination of tillage treatments and crop sequence (conventional tillage and wheat monocropping, CT-WW; conventional tillage, and wheat following faba-bean, CT-WF; zero tillage and wheat monocropping, ZT-WW; zero tillage and wheat following faba-bean, ZT-WF) on yield, grain quality traits as well as on diseases incidence and severity in durum wheat (var. Saragolla). The results of a two-years of data of a long-term experiment (7-year experiment; split-plot design) are discussed. The CA approach (ZT+WF), which induced always the highest grain yields (6.1 t ha-1 and 3.3 t ha-1 in 2016 and 2017) thanks to an increased number of spikes m-2 (296 vs 269 and 303 vs 287 spikes m-2 in 2016 and 2017, respectively) as well as to a more pronounced ear length, demonstrated significant positive influences in terms of grain quality. It promoted grain protein accumulation (12.1% for ZT+WF versus 11.4% for ZT+WW and 12.4% for ZT+WF versus 10.6% for ZT+WW in 2016 and 2017), improved gluten quality (in terms of SDS sedimentation test) and colour of the grain. The abundance of crop residues determined a higher incidence and severity of Zymoseptoria tritici leaf symptoms under CA system; nevertheless the late appearance of infection was the main reason of not affecting yield and quality traits. The presence of faba-bean (WF) in the rotation significantly reduced leaf symptoms of Z. tritici.

Machine learning (ML) is widely used in the field of crop-growing information identification based on high-resolution remote sensing images. With Baoying County in Jiangsu Province, China, as the study area, this paper used Sentinel-2 images during the winter wheat growth period to construct its spectral, textural, and topographic features during its growth period and proposes a winter wheat-growing area extraction method based on the extreme gradient boosting (XGBoost) algorithm, which was compared with traditional ML algorithms such as the support vector machine (SVM), classification and regression tree (CART), and random forest (RF) algorithms. The results indicated that (1) a winter wheat-growing area identification model based on the XGBoost algorithm was successfully constructed based on Sentinel-2 images, considering 27 spectral, textural, and topographic features; (2) the constructed model could effectively extract winter wheat in the study area with an overall accuracy of 93.43% and only a small error compared with the actual winter wheat-growing area in Baoying County, meeting the accuracy requirement for crop identification in the study area; and (3) the deep learning algorithm XGBoost outperformed the three traditional ML algorithms, among which the RF algorithm was better than the SVM and CART algorithms, both of which had poor identification performance and a large error compared with the actual growing area. This paper provides a scientific basis for the accurate extraction of winter wheat-growing areas and further research on winter wheat growth monitoring and yield estimation.

During the response of plants to adverse stresses, aquaporin (AQP) plays a prominent role in membrane water transport based on received upstream signals. In addition, they have various physical parts for dealing with environmental stresses. Due to the importance of the AQP gene family, studies have been conducted investigating the function and regulatory system of these genes. However, many of their molecular aspects are still unknown. This study aims to carry out a genomic-wide investigation of the AQP gene family in durum wheat using bioinformatics tools and to investigate the expression patterns of some members in response to salt stress. Our results showed that there are 80 TtAQP genes in durum wheat, which are classified into four main groups based on phylogenetic analysis. Many duplications were observed between the members of the TtAQP gene family, and high diversity in response to post-translational modifications was observed between TtAQP family members. The expression pattern of TtAQP genes disclosed that these genes are primarily upregulated in response to salt stress. Besides, qPCR data revealed that TtAQPs are more induced in delayed responses to salinity stress. Overall, our findings illustrate that TtAQP members are diverse in terms of their structure, regulatory systems and expression levels.

: Using line × tester analysis, the current research analyses parental genotypes and their combinations in normal conditions and identifies the genes influencing yield characteristics. In the present study, 15 diverse genotypes, including 10 lines, 5 testers, and 50 F1s hybrids, were evaluated for 13 morphological and 2 biochemical traits. A suitable location was taken to study the effect of 15 characters. The results exposed that ability mean squares were significant for all studied additive and non-additive components. In this direction, the general combining ability of PBW-343, DBW-39, K-402, K-1317, KRL-210, and K-68 were higher than the remaining parents. For morphological traits like yield, the top five crosses were described based on SCA effects, namely, HD-3086 × HD-3171, K-402 × K-9107, K-1317 × K-9107, HD-2967 × K-0307 and K-402 × K-68 in F1 generation. In addition, the high value of heritability was estimated for plant height (77.32%), spike length (32.26%), biological yield/plant (59.52%), and grain yield/plant (68.76%). However, the moderate values of heritability were estimated for days to maturity (22.78%) and phenol color reaction (18.00%). The higher genetic advance was not found for recorded characters; however, a moderate genetic advance was recorded for grain yield per plant (13.15%) and harvest index (11.72%). High heritability coupled with moderate genetic advance was recorded for two characters grain yield per plant and harvest index in F1 and F2 generations.

The current and projected climate change that is represented by increasing temperatures, humidity levels and irregular rainfall patterns, promotes the occurrence of preharvest sprouting (PHS) in wheat. PHS results in significant economic losses, globally, which necessitates the need for high-yielding cultivars with increased PHS tolerance, hence this study was conducted. The current study evaluated two doubled-haploid (DH) wheat populations of Tugela-Dn × Elands and Elands × Flamink across six environments in the Free State Province of South Africa to select genotypes with increased PHS tolerance and further map the underlying loci. Significant effects of DH lines (194) and environments (6) were observed for PHS tolerance. The results of this study validate previous findings that PHS is only expressed when environmental conditions are conducive. Quantitative trait loci (QTL) mapping using single nucleotide polymorphism (SNP) and silicoDArT markers revealed three additive QTL with major effects on chromosomes 5B and 7B, and these QTL were detected more than once, when conditions were favourable. These QTL explained a phenotypic variation (PVE) varying between 10.08% and 20.30% (LOD = 2.73 – 3.11). About 16.50% of DH lines performed to the level of Elands (the PHS-tolerant parent) and are recommended for further selection in a pre-breeding or breeding programme. The findings of the study are expected to facilitate the on-going breeding efforts for PHS tolerance in winter wheat.

Main purposes and research question: Wheat is the second largest grain crop by tonnage in the world and the largest in Denmark. Given the observed, adverse impacts on wheat yields of climate change and the importance of wheat in the human diet, the purpose of this study was to use life cycle assessment to compare conventional wheat farming with indoor vertical farming using hydroponics. Methods: Life Cycle Assessment was used to assess the base case systems up to the “farm gate” for 1 tonne of wheat grain. The processes contributing most of the impacts were identified, and scenarios were assessed to determine how much the impacts could be reduced. Results: The conventional system outperformed the base case vertical system in every impact category, due to the electricity consumption in the lighting system. The scenarios included increasing the efficiency of the LED lighting and using 100% wind energy, but the conventional system still outperformed the vertical system by significant margins in all impact categories. This was due to the low photosynthetic conversion efficiency and the high energy density of wheat. Conclusions: Until significant improvements are made to lighting efficiency, the photosynthesis conversion efficiency of wheat, new wheat variants designed for vertical gardens and the sustainability of electricity supply, conventional wheat production will be environmentally preferable and vertical gardens would be advised to focus on food products with low energy densities.

This research was specifically aimed at assessing the influence of sulfur in triple-super phosphate (TSP) on wheat yield. From the results, wheat showed response to sulfur (S) from gypsum (in 67%); and nitrogen (N) from urea in about 100% (of 24 sites). Based on this N was found to be the most limiting element to wheat production followed by sulfur, and then by phosphorus. TSP is tested to contain agronomically up to 2-6% by weight of S. However, wheat didn’t show response to S impurity supplied in the form of TSP. Though, not statistically significant, it is observed that there have always been yield increments by certain percent due to S from TSP in 8 out of 10 target sites, which is depicted in the increasing trends of yield response curves. From this it is learnt that, the benefits of the accidental/incidental application of such high analysis fertilizers can be many-folds in the quality attributes of wheat, if the soils of such investigation at the same time would contain significant amount of organic matter (OM). Indeed, such analysis would be vital in varietal specific nutrient requirement studies in precision-farming and/or in categorizing soils into fertility gradients and fertilizer recommendation domains.

Wheat is one of the global strategic crops and ranks third in terms of cereals production. Wheat crop is exposed to many fungal infections during the cultivation stages, some of which have the ability to secrete numbers of toxic secondary metabolites that threaten the quality of grains, consumer health, producer economics and global trade exchange. Fifty-four random samples were collected from wheat which originated from different countries. The samples included 14 of soft wheat to study the extent of their contamination with Deoxynivalenol (DON) and T-2 toxin by auto-ELISA technology and r-biopharm Microtiter plate. All samples were contaminated with DON toxin except one sample, and the contamination average ranged between 40.7 and 1018.8 µg kg-1. The highest contamination rates were in Lithuanian and the lowest in Indian wheat. Meanwhile the highest average level of T-2 toxin contamination was in Lithuanian wheat grains with a rate of 377.4µg kg-1 and the lowest in Polish wheat with an average of 115.3µg kg-1. GC-MS/MS and multiple reaction monitoring mode (MRM), was used to detect 15 triazole derivatives in collected samples, which may be used to combat fungal diseases on wheat during the growing season. Only 9 derivatives were found; Simeconazole, Penconazole, Hexaconazole, Cyproconazole, Diniconazole, Tebuconazole, Metconazole, Fenbuconazole and Difenoconazole. These derivatives varied according to the origin of wheat samples as well as their its concentration whereas, another 6 derivatives were not detected in all samples. A direct inverse relationship was found between the DON concentration in the samples and the residues of Simeconazole, Penconazole, Diniconazole, Tebuconazole, Metconazole, Fenbuconazole and Difenoconazole and the T-2 toxin showed the same relationship except for Tebuconazole. The safe and rational use of some triazole derivatives may be a new approach and a promising strategy not only to reduce plant diseases and their problems, but also to get rid of some mycotoxins as contaminates.

Stable isotope-assisted approaches can improve untargeted LC-HRMS metabolomics studies. Here, we demonstrate at the example of chemically stressed wheat that metabolome-wide internal standardization by globally 13C-labeled metabolite extract (GLMe-IS) of experimental-condition-matched biological samples can help to improve coverage of treatment-relevant metabolites and aid in the post-acquisition assessment of putative matrix effects in samples obtained upon different treatments. For this, native extracts of toxin- and mock-treated (control) wheat ears were standardized by the addition of uniformly-13C-labeled wheat ear extracts that were cultivated under similar experimental conditions (toxin-treatment and control) and measured with LC-HRMS. The results show that 996 wheat-derived metabolites were detected with the non-condition-matched 13C-labeled metabolite extract, while another 68 were only covered by the experimental-condition-matched GLMe-IS. Additional testing is performed with the assumption that GLMe-IS enables compensation for matrix effects. Although on average no severe matrix differences between both experimental conditions were found, individual metabolites may be affected as is demonstrated by wrong decisions with respect to the classification of significantly altered metabolites. When GLMe-IS was applied to compensate for matrix effects, 272 metabolites showed significantly altered levels between treated and control samples, 42 of which would not have been classified as such without GLMe-IS.

Flour yield determined the profitability of flour mill, but the intrinsic variability of the grain makes it very complex to analysis and estimate wheat grain flour yield. Simulation for flour yield attributes offer considerable advantages in flour mill, if reliable predictions of flour yield can be provided, because the wheat grain characteristics can be tested before milling. If this is possible, the characteristics thus observed could be quantified more reliably and objectively by Structural Equation Modelling (SEM). SEM was used to look for the most important wheat grain characteristics on flour yield, and then these wheat grain characteristics were used to simulate flour yield. Furthermore, the regressive equation was verified by the field experiment. The coefficient of variation of grain characteristics was low and distributed rather closely. The results of SEM showed that test weight had the most significantly effects on flour yield, followed by the hardness index. Test weight and hardness index could excellently estimate flour yield by multiplicative effect of test weight and hardness index, and which could determine 68% of the variation in flour yield. The simulation result can not only predict flour yield, but also look for the important grain characteristics for the flour yield.

Root research on field grown crops is hindered by the difficulty of estimating root biomass in soil. Root washing, the current standard method is laborious and expensive. Biochemical methods to quantify root biomass in soil, targeting species-specific DNA, have potential as a more efficient assay. We combined an efficient DNA extraction method, designed specifically to extract DNA from soil, with well-established quantitative PCR methods to estimate the root biomass of twenty-two wheat varieties grown in field trials over two seasons. We also developed an assay for estimating root biomass for black-grass, a common weed of wheat cultivation. Two robust qPCR assays were developed to estimate the quantity of plant root DNA in soil samples, one specific to wheat and barley, and a second specific to black-grass. The DNA qPCR method was comparable, with high correlations, with the results of root washing from soil cores taken from winter wheat field trials. The DNA qPCR assay showed both variety and depth as significant factors in the distribution of root biomass in replicated field trials. The results suggest that these DNA qPCR assays are a useful, high throughput tool for investigating the genetic basis of wheat root biomass distribution in field grown crops, and the impact of black-grass root systems on crop production.

The domestication and subsequent genetic improvement of wheat led to the development of large-seeded cultivated wheat species relative to their smaller-seeded wild progenitors. While increased grain weight (GW) continues to be an important goal of many wheat breeding programs, few genes underlying this trait have been identified despite an abundance of studies reporting quantitative trait loci (QTLs) for GW. Here we perform a QTL analysis for GW using a population of recombinant inbred lines (RILs) derived from the cross between wild emmer wheat accession ‘Zavitan’ and durum wheat variety ‘Svevo’. Identified QTLs in this population were anchored to the recent Zavitan reference genome, along with previously published QTLs for GW in tetraploid wheat. This genome-based, meta-QTL analysis enabled the identification of a locus on chromosome 6A whose introgression from wild wheat positively affects GW. The locus was validated using an introgression line carrying the 6A GW QTL region from Zavitan in a Svevo background, resulting in >8% increase in GW compared to Svevo. Using the reference sequence for the 6A QTL region, we identified a wheat ortholog to OsGRF4, a rice gene previously associated with GW. The coding sequence of this gene (TtGRF4-A) contains four SNPs between Zavitan and Svevo, one of which reveals the Zavitan allele to be rare in a core collection of wild emmer and completely absent from the domesticated emmer genepool. Similarly, another wild emmer accession (G18-16) was found to carry a rare allele of TtGRF4-A that also positively affects GW and is characterized by a unique SNP absent from the entire core collection. These results exemplify the rich genetic diversity of wild wheat, posit TtGRF4-A as a candidate gene underlying the 6A GW QTL, and suggest that the natural Zavitan and G18-16 alleles of TtGRF4-A have potential to increase wheat yields in breeding programs.

Meta-analysis of plant sterol supplement studies suggests an 8% lowering of LDL cholesterol for 2 to 2.5g/day of plant sterols. Cereal foods have been rarely tested and one study showed a lower LDL lowering of 5.4% with 1.6g of plant sterol in breakfast cereal. We aimed to test a breakfast wheat biscuit with 2g of plant sterols in a single serve of two wholegrain wheat breakfast cereal biscuits. Fifty volunteers with a total cholesterol of >5.5mmol/L were recruited for a randomised crossover study with two 4-week periods with no washout of which 45 successfully completed the study. After exclusion of four outliers the difference in LDL cholesterol between standard wholegrain wheat breakfast cereal biscuit and plant sterol-enriched wholegrain wheat breakfast cereal biscuit was 0.23 mmol/L or 5.6% (P=0.001) with a 95% confidence interval of 2.4-8.9%. Men and daily cereal consumers had greater responses 9.8% vs 3.6% and 7.2% vs 3.8% respectively (P<0.05). The LDL lowering effect of 2g of plant sterol enriched from one serve of wholegrain wheat breakfast cereal biscuit was not significantly different from other food products delivering 2-2.5g of plant sterols daily. Regular cereal consumers have a better response.

Crop yield prediction is relevant subject of current agricultural science. There are various mathematical approaches to crop yield prediction, and regression analysis, notwithstanding the fact that it is somewhat outdated, is still one of the most used ones in this purpose. The quality of predictive model is of great importance, and it is strongly dependent on the rational choice of the target function. The goal of this study is to find out the best regression model for winter wheat, soybeans, and grain corn yield prediction depending on the crops’ water use. The data on true crops’ yields and water use were collected within 1970-2020 at the experimental fields of the Institute of Climate-Smart Agriculture, Kherson region, Ukraine. In total, 145 data pairs were processed by the best subsets regression to find out the best model in terms of fitting quality (assessed by the Pearson’s coefficient of correlation), and prediction accuracy (assessed by the values of the minimum and maximum absolute errors and mean average percentage error). As a result, it was established that the best fitting quality for all the studied crops is attributed to cubic function, while the best accuracy is recorded for hyperbolic (reverse) function in soybeans (mean absolute percentage error is 12.27%), quadratic and hyperbolic functions in winter wheat (mean absolute percentage error is 20.54%), and cubic function in grain corn (mean absolute percentage error is 14.92%). To sum up the results of the study, it is recommended to apply cubic regression function for modeling crops’ yields in agricultural studies.

Fusarium head blight (FHB) is a wheat disease caused by fungi of the genus Fusarium. The aim of the study was to find relationships between the weather conditions in the experimental years and the locations and the amount of F. culmorum DNA and trichothecene genotypes, as well as the proportions between them. A three-year field experiment (2017, 2018 and 2019) was established at two locations (Poznań, Radzików). F. culmorum DNA was detected in all grain samples in an average amount of 20124 pg per 1 g of wheat DNA. The average amount of DNA from the 3ADON genotype was 4879 pg/μg and the amount of DNA from the NIV genotype was 3330 pg/μg. In the three experimental years, a large variability was observed in the coefficients of correlation between DNA concentrations and the FHB index, FDK, ergosterol, and the corresponding toxins. There were significant correlations between disease incidence, fungal biomass (quantified as the total amount of fungal DNA or DNA trichothecene genotypes) and toxins (DON, 3AcDON and NIV) concentrations. The 3ADON trichothecene genotype dominates over the NIV genotype (ratio 1.5); however, this varied greatly depending on environmental conditions.

To evaluate genetic variability and seedling plant response to a dominated Warrior (-) race of yellow rust in Northern and Central European germplasm, we used a population of 229 winter wheat cultivars and breeding lines for genome-wide association study (GWAS). A wide variation in yellow rust disease severity (based on infection types 1-9) was observed in this panel. Four breeding lines TS049 (from Austria), TS111, TS185, and TS229 (from Germany) and one cultivar TS158 (KWS Talent) from Germany were found to be resistant to Warrior (-) FS 53/20 and Warrior (-) G 23/19. GWAS identified five significant SNPs associated with yellow rust on chromosomes 1B, 2A, 5B, and 7A for Warrior (-) FS 53/20, while one SNP on chromosome 5B was associated with disease for Warrior (-) G 23/19. For Warrior (-) FS 53/20, we discovered a new QTL for yellow rust resistance associated with the marker Kukri_c5357_323 on chromosome 1B. The resistant allele G at the marker locus Kukri_c5357_323 on chromosome 1B and the susceptible allele T at the marker locus Excalibur_c17489_804 on chromosome 5B showed the largest effects (1.21 and 0.81, respectively) on the severity of yellow rust detected in Warrior (-) FS 53/20 and Warrior (-) G 23/19. Among 144 putative genes within the flanking sequence of the significant SNPs detected by GWAS, the function of the best candidate genes was determined as protein kinase activity and oxidoreductase activity. Our results provide the basis for knowledge-based resistance breeding in the face of the enormous impact of the Warrior (-) race on wheat production in Europe.

Background: Fusarium head blight (FHB) is a serious fungal disease of crop plants due to substantial yield reduction and production of mycotoxins in the infected grains. The breeding progress in increasing resistance with maintaining a high yield is not possible without a thorough examination of the molecular basis of plant immunity responses; Methods: LC-MS based metabolomics approaches powered by three-way ANOVA and differentially accumulated metabolites (DAMs) selection, correlation network and functional enrichment were conducted on grains of resistant and susceptible to FHB genotypes of barley and wheat as well as model grass Brachypodium distachyon (Bd) still poorly known at metabolomic level; Results: We selected common and genotype-specific DAMs in response to F. culmorum inoculation. Immunological reaction at metabolomic level was strongly diversified between resistant and susceptible genotypes. DAMs common for all tested species from porphyrins, flavonoids and phenylpropanoids metabolic pathways were highly correlated and reflects conservativeness in FHB response in Poaceae family. Resistant related DAMs belonged to different structural classes including tryptophan derived metabolites, pirimidines, amino acids proline and serine as well as phenylpropanoids and flavonoids. Physiological response to F. culmorum of Bd was close to barley and wheat genotypes however, metabolomic changes were strongly diversified. Conclusions: Combined targeted and untargeted metabolomics provides comprehensive knowledge about significant elements of plant immunity with potential of being molecular biomarkers of enhance resistance to FHB in grass family. Thorough examination of Bd21 metabolome in juxtaposition with barley and wheat diversified genotypes facilitate their setting as model grass for plant-microbe interaction.

Biomass is an attractive energy source that can be used for production of heat, power, and transport fuels, and when produced and used on a sustainable basis, can make a large contribution to reducing greenhouse gas (GHG) emissions. Anaerobic digestion (AD) is a suitable technology for reducing organic matter and generating bioenergy in the form of biogas. This study investigates the factors allowing the optimization of the process of biogas production from the co-digestion of wheat straw (WS) and bovine manure. The statistical analysis of the experiments carried out show that ultrasonic processing plays a fundamental role by sonication density and solids concentration leading to improved characteristics of WS by reducing particle size and increasing concentration of soluble chemical oxygen demand. The higher the sonicating power used, the more the waste particles are disrupted. The optimality obtained under mesophilic conditions for WS pretreated with 4% w/w (weight by weight) H2O2 at temperature 36 °C under 10 minutes of ultrasonication at 25 kHz improves the methane yield by 64%.

Previous studies investigated the effects of seed rates and seed recycling practices on the yield and yield related variables. However, higher yield does not always guarantee cost-efficiency. This study aimed at investigating the yield effects of plot-level seed rate along with the cost-benefit analysis of seed recycling practices. This study has introduced the dose-response model to the existing analytical methods used in analyzing the effect of different agrochemicals on crop yield. A multi-stage stratified sampling technique was used to select a total of 450 sample respondents. Data was gathered using a mix of data collection tools. Descriptive statistics along with the dose-response model have been applied for data analysis. Farmers of the study were found to be dissimilar in terms of their seed rate application. The dose-response analysis indicated that the highest average wheat yield has been associated with a seed rate of 50 kg ha-1 above what is recommended. The yield effect of seed recycling has also been assessed and a one-time seed recycling has caused a yield decline of 665 kg ha-1 as compared to the non-recycled seeds. The cost reduced by using recycled seed is by far lower than the economic gains associated with using unrecycled and fresh seeds. The cost-benefit analysis made clear that farmers can reduce their seed costs through seed recycling but their yields and net income can be best improved by using unrecycled CBWS. Thus, farmers must be encouraged to make use of unrecycled seed by establishing agricultural credit schemes geared towards seed procurement and seed price subsidy as key strategies to reduce economically wasteful seed recycling practices.

Metal, primarily Fe and Zn, deficiencies affect over half of the world's population. Human diets with prevalent cereal products cause micronutrient malnutrition. Biofortification is one of the most effective approaches to alleviate malnutrition. Spring wheat genetically stable (M7) mutant lines developed with 100 and 200 Gy gamma treatments to broaden genetic variation and search for new resources were analyzed for nutritionally important minerals (Ca, Mg, K, Fe, and Zn), their bioavailability, and grain protein content (GPC). The variation was 172.3–883.0 mg/kg for Ca, 472.9–1088 mg/kg for Mg, 3128.6–5487.5 mg/kg for K, 40.9–89.0 mg/kg for Fe, and 22.2–89.6 mg/kg for Zn. In mutant lines, among the investigated minerals, the highest increases in concentrations were observed in Fe, Zn, and Ca when compared to the parent. Some mutant lines, mostly in the 100 Gy-derived germplasm, had two to three times higher Fe, Zn, and Ca concentrations, lower phytic acid concentration (1.4–2.1 times), and 6.5–7% higher GPC compare to the parent. Variation was detected for the Ca:Phy, Mg :Phy, Phy:K, Phy:Fe, and Phy:Zn molar ratios, (1.27–10.41, 5.05–18.68, 1.66–4.87, 1.40–5.32 and 1.78–11.78, respectively). The results showed how the genetic variation could be generated through radiation and be useful to develop biofortification by micronutrient varieties with their appropriate bioavailability to overcome malnutrition.

(+)-epi-Epoformin (1), is a fungal cyclohexene epoxide isolated together with diplopimarane and sphaeropsidins A and C, a nor-ent-pimarane and two pimaranes, from the culture filtrates of Diplodia quercivora, a fungal pathogen for cork oak in Sardinia, Italy. Compound 1 possesses a plethora of biological activities including: antifungal, zootoxic and phytotoxic activity. The last activity and the peculiar structural feature of 1 suggested to carry out a structure activity relationship study, preparing eight key hemisynthetic derivatives and their phytotoxicity was assayed. The complete spectroscopic characterization and the activity in the etiolated wheat coleoptile bioassay of all the compounds is reported. Most of the compounds inhibited growth and some of them had comparable or higher activity than the natural product and the reference herbicide Logran. As regards the structure-activity relationship, the carbonyl proved to be essential for their activity of 1, as well as the conjugated double bond, while the epoxide could be altered with no significant loss.

Wheat and barley are among the primary food resources of the world population; therefore, their growth and observation are essential in farms to enhance food security worldwide. On top of that, careful observation of the product is essential to find solutions for the issues faced during their production and to reduce the impacts of weather changes. With the advancement of Remote Sensing technology, the observation and estimation process has increased. In this study, numbers of spectral vegetation indices was used along with canopy biophysical properties ( LAI ) and biochemical properties (chlorophyll), there calculated from (Landsat 8 and Sentinel-2) satellite data. The wheat and barley samples were collected before were be ready for harvest, and a relation with the vegetarian indices was established using the Multi-Linear Regression module, in which the equations used in predicting the harvest were developed and used to create a graph for expected harvest. The result indicated that there is a strong relationship between the vegetation indices of Sentinel-2 and Landsat images and the actual grain yield with R² of 0.77 and 0.71, respectively. The results show that the strongest correlation is observed between the LAI data obtained from Sentinel data and cereal yield data, with an R² 0.68, and the highest correlation for the indices of Landsat images is observed in the NDWI with R² 0.59 and the lowest degree of error was in the root mean square error (RMSE) for the Sentinel-2 and Landsat 8 with 0.57 and 1.54. In addition, this study also showed that the least relationship for grain yield prediction was observed between the NDRI for Sentinel-2 (R² 0.1) and SAVI for Landsat image (R² 0.47).

Soil quality assessment based on crop yields and identification of key indicators of it can be used for better management of agricultural production. In the current research, the weighted additive soil quality index (SQIw), factor analysis (FA) and multiple linear regression (MLR) method are used to assess the soil quality of rainfed winter wheat fields with two soil orders on 53.20 km2 of agricultural land in western Iran. A total of 18 soil quality indicators were determined for 100 soil samples (0-20 cm depth) from two soil orders (Inceptisols and Entisols). The soil properties measured were: pH, soil texture, organic carbon (OC), cation exchange capacity (CEC), electrical conductivity (EC), soil microbial respiration (SMR), carbonate calcium equivalent (CCE), soil porosity (SP), bulk density (BD), exchangeable sodium percentage (ESP), mean weight diameter (MWD), available potassium (AK), total nitrogen (TN), available phosphorus (AP), available Fe (AFe), available Zn (AZn), available Mn (AMn), and available Cu (ACu). Mean wheat grain yield for the two years for all of the 100 sampling sites was also gathered. The SQIw was calculated using two weighting methods (FA and MLR) and maps were created using a digital soil mapping framework. The soil indicators taken in the minimum data set (MDS) were AK, clay, CEC, AP, SMR, and sand. The correlation between the MLR weighting technique (SQI-M) and the rainfed wheat yield (r=0.62) was slightly larger than that the correlation of yield with the FA weighted technique (SQI-F) (r=0.58). Results showed that the means of both SQI-M and SQI-F and rainfed wheat yield for Inceptisols were higher than for Entisols although these differences were not statistically significant. Both SQI-M and SQI-F showed that areas with Entisols had lower proportions of good soil quality grades (Grade I and II), and higher proportions of poor soil quality grades (Grade IV and V) compared to Inceptisols. Based on these results, soil type must be considered for soil quality assessment in future studies to maintain and enhance soil quality and sustainable production. The overall soil quality of the study region was of poor and moderate grades. To improve soil quality, it is therefore recommended that effective practices such as the implementation of scientific integrated nutrient management involving the combined use of organic and inorganic fertilizers in rainfed wheat fields be promoted.

Optimizing the nitrogen (N) timings and rate can improve nutrient uptake, and nutrient-efficiencies, especially of N in wheat under the changing climate scenario. Climatic stress in the form of high temperature and drought resulted in the decreased crop physiology and, ultimately, grain yield. Taking the example of rainfed wheat, we quantified the impact of N application rates as full and split-dose at three variable sites of rainfed Pothwar, Pakistan by conducting field experiments for two years (2013-14 and 201-15). Treatments include, T1 = Control (No fertilizer applied), full dose of N applied at the time of crop sowing, i.e. T2 = 50 kg N ha^-1, T3 = 100 kg N ha^-1 and T4 = 150 kg N ha^-1 and split application of N at different timings during different stages of the crop called as split application of N, i.e. T5: Application of 50 kg N ha^-1 (15 kg N ha^-1 (Sowing) : 20 kg N ha^-1 (Tillering) :15 kg N ha^-1 (Anthesis), T6: Application of 100 kg N ha^-1 (30 kg N ha^-1 (Sowing): 40 kg N ha^-1 (Tillering) : 30 kg N ha^-1 (Anthesis) and T7: Application of 150 kg N ha^-1(45 kg N ha^-1 (Sowing) : 60 kg N ha^-1 (Tillering) : 45 kg N ha^-1 (Anthesis). Three study sites include viz. Islamabad (High rainfall with optimum temperature), University Research Farm (URF)-Koont (Medium rainfall with moderate temperature), and Talagang (low rainfall with high temperature). Results showed that the highest stomatal conductance (0.80 mole m^-2 sec^-1), net photosynthetic rate (20.07 μmole m^-2s^-1), transpiration rate (9.58 mmole m^-2s^-1), intercellular CO2 concentration (329.25 μmole CO₂ mol^-1 air), SPAD values (58.86 %) and proline contents (35.42 μg g^-1) were obtained for split application of N (T6 = Split N100) compared to control and full dose of N treatments. Among sites, these physiological traits remained highest at Islamabad and lowest at Talagang, while among years, maximum values of the measured parameters were obtained in 2013-14. A similar trend was observed for crop total N, N efficiencies, and agronomic traits of the crop. Our results suggest that optimum N application rate and its suitable timings can help to harvest real benefits of N as in our findings, split dose resulted in the maximum performance of the crop from physiological parameters to the agronomic traits of the rainfed wheat crop.

Growing acreage and changing consumer preferences cause increasing interest in the cereal products originating from organic farming. Lack of results of objective test, however, does not allow drawing conclusions about the effects of cultivation in the organic system and comparison to currently preferred conventional system. Field experiment was conducted in organic and conventional fields. Thirty modern cultivars of winter wheat were sown. They were characterized for disease infection including Fusarium head blight, seed sowing value, the amount of DNA of the six species of Fusarium fungi as well as concentration of ergosterol and trichothecenes in grain. The intensity Fusarium head blight was at a similar level in both systems. However, Fusarium colonization of kernels expressed as ergosterol level or DNA concentration was higher for the organic system. It did not reflect in an increased accumulation of trichothecenes in grain, which was similar in both systems, but sowing value of organically produced seeds was lower. Significant differences between analyzed cropping systems and experimental variants were found. The selection of the individual cultivars for organic growing in terms of resistance to diseases and contamination of grain with Fusarium toxins was possible. Effects of organic growing differ significantly from the conventional and grain obtained such way can be recommended to consumers. There are indications for use of particular cultivars bred for conventional agriculture in the case of organic farming, and the growing organic decreases plant stress resulting from intense fertilization and chemical plant protection.

In a changing climate, extreme weather events such as heat waves will be more frequent and could affect grain weight and the quality of crops such as wheat, one of the most significant crops in terms of global food security. In this work, we characterized the response of Triticum turgidum spp. durum wheat to a short-term heat-stress (HS) treatment at transcriptomic and physiological levels during early grain filling in glasshouse experiments. We found a significant reduction in grain weight and size from HS treatment. Grain quality was also affected, showing a decrease in starch content in addition to increments in grain protein levels. Moreover, an RNA-seq analysis of durum wheat grains allowed us to identify 1590 differentially expressed genes related to photosynthesis, response to heat, and carbohydrate metabolic process. A gene regulatory network analysis of HS-responsive genes uncovered novel transcription factors (TFs) controlling the expression of genes involved in abiotic stress response and grain quality, such as a member of the DOF family predicted to regulate glycogen and starch biosynthetic processes in response to HS in grains. In summary, our results provide new insights into the extensive transcriptome reprogramming that occurs during short-term HS in durum wheat grains.

Automated machine learning (AutoML) has been heralded as the next wave in artificial intelligence with its promise to deliver high performance end-to-end machine learning pipelines with minimal effort from the user. However, despite AutoML showing great promise for computer vision tasks, to the best of our knowledge, no study has used AutoML for image-based plant phenotyping. To address this gap in knowledge, we examined the application of AutoML for image-based plant phenotyping using wheat lodging assessment with UAV imagery as an example. We compared the performance of an open-source AutoML framework, AutoKeras in image classification and regression tasks to transfer learning using modern convolutional neural network (CNN) architectures. For image classification which classified plot images as lodged or non-lodged, transfer learning with Xception and DenseNet-201 achieved best classification accuracy of 93.2%, whereas Autokeras had 92.4% accuracy. For image regression which predicted lodging scores from plot images, transfer learning with DenseNet-201 had the best performance (R2=0.8303, RMSE=9.55, MAE=7.03, MAPE=12.54%), followed closely by AutoKeras (R2=0.8273, RMSE=10.65, MAE=8.24, MAPE=13.87%). Interestingly, in both tasks, AutoKeras models had up to 40-fold faster inference times compared to the pretrained CNNs. The merits and drawbacks of AutoML compared to transfer learning for image-based plant phenotyping are discussed.

Uncertain price movement in staple food commodities puts agrarian economies at risk if not monitored and managed consistently. Hence, an attempt has been made to analyze the price behavior and integration across major wholesale and retail markets for rice and wheat in India. Monthly data (July 2000 to June 2022) on prices viz., wholesale and retail were sourced from the Food and Agriculture Organization and analyzed using growth rate, instability index, seasonal price index, Johansen’s test on cointegration, Granger causality test, and impulse response function. Findings indicated strong evidence of price dynamics in the selected markets in terms of spatial and temporal variation, clear-cut seasonality linking to production, and price divergence between wholesale and retail markets. Johansen’s test indicated a strong integration between wholesale and retail markets exhibiting unidirectional-, bidirectional- and no-causality. Impulse response analysis revealed that the selected wheat and rice markets are efficient in terms of ‘price discovery’ which takes place initially in the wholesale market, and then transmitted to the retail market. The study advocates decision-making information to the producers, traders, and consumers who have a potential interest in getting advantage of the price movement. It is concluded that strengthening market intelligence and reducing the distortion in markets will improve the existing overall performance.

In Nitrate Vulnerable Zones (NVZ) site-specific techniques are needed to match N availability with durum wheat (Triticum turgidum subsp. durum Desf.) requirements. Enhanced-efficiency fertilizers (EEF) can improve efficient N supply and reduce leaching, thus contributing to sustainable agriculture. To study the effects of rates, sources and timings of nitrogen application, two-year field experiments were carried out at two Mediterranean NVZs of Central Italy (Pisa and Arezzo). The trial compared: i) two N rates: one based on the crop N requirements (NO), the other on the Action Programmes’ prescriptions of the two NVZ (NAP); ii) three N sources (urea, methylene urea (MU), and nitrification inhibitor (NI) 3,4-Dimethylpyrazole phosphate (DMPP); and two top-dressing timings (1st tiller visible and 1st node detectable). Grain yield and yield components were determined, together with N uptake. Results showed that: i) grain and biomass production were reduced with NAP at both locations; ii) urea performed better than slow-release fertilizers; iii) the best application time varied depending on N source and location: at Pisa enhanced-efficiency fertilizers achieved higher yields when applied earliest, while for urea the contrary was true; at Arezzo different N fertilizers showed similar performances between the two application timings. Different behaviors of top-dressing fertilizers at the two localities could be related to the diverse patterns of temperatures and rainfall. Therefore, optimal fertilization strategies vary according to environmental conditions.

Barley mlo mutants are well known for their profound resistance against powdery mildew disease. Recently, mlo mutant plants were generated in hexaploid bread wheat (Triticum aestivum) with the help of transgenic (transcription-activator-like nuclease, TALEN) and non-transgenic (targeted induced local lesions in genomes, TILLING) biotechnological approaches. While full gene knockouts in the three wheat Mlo (TaMlo) homoeologs, created via TALEN, confer full resistance to the wheat powdery mildew pathogen (Blumeria graminis f.sp. tritici), the currently available TILLING-derived Tamlo missense mutants provide only partial protection against powdery mildew attack. Here we studied the infection phenotypes of TALEN- and TILLING-derived Tamlo plants to the two hemibiotrophic pathogens Zymoseptoria tritici, causing Septoria leaf blotch in wheat, and Magnaporthe oryzae pv. Triticum (MoT), the causal agent of wheat blast disease. While Tamlo plants showed unaltered outcomes upon challenge with Z. tritici, we found allele-specific levels of enhanced susceptibility to MoT, with stronger powdery mildew resistance correlated with more invasive growth by the blast pathogen. Surprisingly, unlike barley mlo mutants, young wheat mlo mutant plants do not show undesired pleiotropic phenotypes such as spontaneous callose deposits in leaf mesophyll cells or signs of early leaf senescence. In conclusion, our study provides evidence for allele-specific levels of enhanced susceptibility of Tamlo plants to the hemibiotrophic wheat pathogen MoT.

If genetic gains in wheat yield are to be achieved in today’s breeding, increasing genetic variability of cultivated genotypes is an essential requisite to meet. To this aim, alien gene transfer through chromosome engineering (CE) is a validated and sound strategy. Attempts to incorporate more than one alien segment into cultivated wheat have been rare, particularly for tetraploid durum wheat. Here we present the agronomic and quality performance of the first successful CE-mediated multiple introgression into the latter species. By assembling into 7AL, 3BS and 1AS arms of a single genotype homoeologous segments of Thinopyrum ponticum 7el1L, Aegilops longissima 3SlS, and Triticum aestivum 1DS arms, respectively, we have stacked several valuable alien genes, comprising Lr19+Sr25+Yp (leaf and stem rust resistance and a gene increasing semolina yellowness), Pm13 (powdery mildew resistance) and Gli-D1/Glu-D3 (genes affecting gluten properties), respectively. Advanced progenies of single, double and triple recombinants were field-tested across three years in a typical durum wheat growing area of Central Italy. The results showed that not only all recombinants had normal phenotype and fertility, but also that one of the triple recombinants had the highest yield through all seasons compared with all other recombinants and control cultivars. Moreover, the multiple introgressions enhanced quality traits, including gluten characteristics and semolina yellow index. Presence of effective disease resistance genes confers additional breeding value to the novel and functional CE products, which can greatly contribute to crop security and safety.

Diseases of cereals caused by pathogenic fungi can significantly reduce crop yields. Many cultures are exposed to them. The disease is difficult to control on a large scale, thus one of the relevant approaches is the crop field monitoring, which helps to identify the disease at an early stage and take measures to prevent its spread. One of the effective control methods is disease identification based on the analysis of digital images with the possibility of obtaining them in field conditions using mobile devices. In this work, we propose a method for the recognition of five fungal diseases of wheat shoots (leaf rust, stem rust, yellow rust, powdery mildew, and septoria), both separately and in combination, with the possibility of identifying the stage of plant development. A set of 2414 images of wheat fungi diseases (WFD2020) was generated, for which expert labeling was performed by the type of disease. WFD2020 data are available freely at http://wfd.sysbio.ru/. In the process of creating this set, a method was applied to reduce the degeneracy of the training data based on the image hashing algorithm. The disease recognition algorithm is based on the convolutional neural network with the EfficientNet architecture. The best accuracy (0.942) was shown by a network with a training strategy based on augmentation and transfer of image styles. The recognition method was implemented by the authors as a bot on the Telegram platform, which allows assessing plants by lesions in the field conditions.
