Abstract

Background: COVID-19, a member of corona virus family is spreading its tentacles across the world due to lack of drugs at present. Associated with its infection are cough, fever and respiratory problems causes more than 15% mortality worldwide. It is caused by a positive, single stranded RNA virus from the enveloped coronaviruse family. However, the main viral proteinase (Mpro/3CLpro) has recently been regarded as a suitable target for drug design against SARS infection due to its vital role in polyproteins processing necessary for coronavirus reproduction.Objectives: The present in silico study was designed to evaluate the effect of Eucalyptol (1,8 cineole), a essential oil component from eucalyptus oil, on Mpro by docking study.Methods: In the present study, molecular docking studies were conducted by using 1-click dock and swiss dock tools. Protein interaction mode was calculated by Protein Interactions Calculator.Results: The calculated parameters such as RMSD, binding energy, and binding site similarity indicated effective binding of eucalyptol to COVID-19 proteinase. Active site prediction further validated the role of active site residues in ligand binding. PIC results indicated that, Mpro/eucalyptol complexes forms hydrophobic interactions, hydrogen bond interactions and strong ionic interactions.Conclusions: Therefore, eucalyptol may represent potential treatment potential to act as COVID-19 Mpro inhibitor. However, further research is necessary to investigate their potential medicinal use.

Comment: This article is interesting, regarding the empirical experience of my uncle who is currently a covid-19 survivor. On June 25, 2020, he was rushed to a special hospital to handle covid-19 at the Hasanuddin University Teaching Hospital with reports of severe shortness of breath and a severe dry cough. According to his confession, when he was taken to the hospital it was the fourth day after the symptoms he felt.
On that day I then responded to the news that my uncle had a Covid-19 attack. I went to his house and met his wife and then I handed him two bottles of pure eucalyptus oil and a mixture of eucalyptus oil and eucalyptus oil with a composition of 60% eucalyptus and 40% eucalyptus oil (greenish color). I give the following instructions:
1. Give pure eucalyptus (clear origin china) droplets on a tissue to be inhaled deeply slowly remembering that the patient feels pain when inhaling.
2. Give a drop and then stick it on the inside of the tongue and then taste it like eating candy at intervals of 2-3 hours at a time.
3. Apply eucalyptus + eucalyptus on the chest, neck, shoulders and back at all times.
The next day (26 June 2020) I tried to communicate with him via Whats Up (social media) to ask how he was. He himself replied to my chat. Information conveyed that;
1. The dry cough has stopped, the mucus starts to feel in the throat from before.
2. You can already feel the smell (pungent smell of eucalyptus globulus).
3. Breathing is better but still panting.
I contacted him again on August 2, 2020 to ask about the news and developments in his health recovery after the Covid-19 attack. He gave a report that his condition was getting better and according to the doctor in charge that his lungs were still recovering by administering medicine (the name of the drug was not disclosed but the doctor said it contained pork extract).
My communication with him on August 17, 2020, was that he was no longer using a breathing apparatus. He continues to inhale eucalyptus essential oil, as I instructed him.
On 23 August 2020, he was declared cured and returned home.

My own experience is every day sipping and tasting a drop and sticking it on my tongue then I have been tasting it since April 2020.
My daily activities are interacting with many people as usual. Although I believe in the properties of pure eucalyptus I still apply the protocol using a mask.
That is a little empirical experience that I convey, hopefully it can contribute.
