Corn starch-based nanocomposite films usually have low moisture barrier properties. The addition of virgin coconut oil (VCO) as a hydrophobic component can improve the nanocomposite film's characteristics, especially the film's permeability and elongation properties. This study aimed to determine the role of VCO with various concentrations (0,3,5 wt%) on the physical, mechanical, and water vapor transmission characteristics of corn starch/NCC-based nanocomposite films. The results showed that the concentration of 3 wt% VCO was most effective in reducing the WVTR value of nanocomposite films with a value of 4.721 g/m2.h. While the value of tensile strength was 4.243 MPa, elongation 68.58%, modulus of elasticity 0.062 MPa, thickness 0.219 mm, lightness 98.77, and water solubility 40.51%. However, the addition of 5 wt% VCO was more effective in increasing the elongation properties of the film. The addition of VCO gave the appearance of a porous film surface and a finer structure was formed. The FTIR test on corn starch nanocomposite films with the addition of VCO did not show any new absorption peaks. The results of this study may provide opportunities for the development of nanocomposite films as biodegradable packaging in the future.

Virgin coconut oil (VCO) is a functional food with important health benefits. Its economic interest encourages fraudsters to deliberately adulterate VCO with cheap and low-quality vegetable oils for financial gain, causing health and safety problems for consumers. In this context, there is an urgent need for rapid, accurate and precise analytical techniques to detect VCO adulteration. In this study, the use of Fourier Transform Infrared (FTIR) spectroscopy combined with Multivariate Curve Resolution - Alternating Least Squares (MCR-ALS) methodology was evaluated to verify purity or adulteration of VCO with reference to several low-cost commercial oils such as sun-flower, maize and peanut oils. Control charts were developed to assess the purity of oil samples using MCR-ALS scores values calculated from a data set of pure and adulterated oils. In addition, quantification models were developed using MCR-ALS with correlation constraint for adulterated coconut oil to assess the blend composition. Different data pre-treatment strategies were tested in order to best extract the information contained in the sample fingerprints, and the calibration models were optimised using a genetic algorithm (GA) to select the most important variables. The models gave satisfactory results in external validation procedure, with absolute errors of less than 4.6 % for samples adulterated with sunflower, maize and peanut oils.

Nowadays, the fingerprinting methodologies of olive oils are dominated. They consider the entire analytical signal, which is acquired and recorded by the analytical instrument, directly from olive oil or isoleted fraction, i,e chromatogram. The shape and intensity of the recorded signal the instrumental fingerprint from the whole olive oil adulteration. Therefore, the methodolygy is based on the chemical composition (Fatty acids and Triglycerides compositions). However, Fatty acids composition as an indicator of purity suggests that linolenic acid content could be used as a parameter for the detection of extra virgin olive oil fraud with 5% of soybean oil. The adulteration could also be detected by the increase of the trans-fatty acid contents with 3% of soybean oil, 2% of corn oil and 4% of sunflower oil. The use of the ∆ECN42 proved to be effective in the Chemlali extra-virgin olive oil adulteration even at low levels: 1% of sunflower oil, 3% of soybean oil and 3% of corn oil. Therefore, compared to classical methods PCA and new approach of using LDA application could represent an alternative and innovative tool for faster and cheaper evaluation of extra-virgin olive oil adulteration.

Extra virgin olive oil, water, and water/oil mixture (W/O) were used to fry, boil and sautée Mediterranean vegetables. We determined the variations between unused and used water and oil for the contents of total (TPC) and individual phenolic compounds measured by HPLC, and the antioxidant capacity (AC) by DPPH, FRAP and ABTS methods. The highest TPC value was found in water used to boil tomato, whereas the lowest in the EVOO from the W/O used for boiling potatoes. The TPC decreased in the processed EVOO, whereas the water was enriched after boiling. After processing, the concentrations of phenols exclusive to EVOO diminished with differentiated behavior, from complete elimination of hydroxytyrosol and tyrosol after sautéeing and W/O boiling, to conservation of pinoresinol. The transfer of phenols such as chlorogenic, gallic, dihydroxybenzoic, hydroxybenzoic and hydroxyphenylacetic acids, as well as luteolin, apigenin and vanillic acid from the vegetable to the oil was frequent when eggplant, tomato and pumpkin were cooked. The cooking water was enriched in most of the phenols analysed, as the case of chlorogenic acid and phenols exclusive to EVOO after W/O boiling. The values of AC in the fresh oil decreased or were maintained after being used to cook the vegetables. With some exceptional increases in the oil used for frying (p<0.05). The cooking techniques were classified in decreasing order according to the AC as follows: raw>deep frying>sautéing>boiling. This pattern was more consistent with DPPH results (from potato, eggplant and pumpkin) than in those of ABTS (pumpkin) and FRAP (eggplant). The water fraction recovered from boiling was enriched in Trolox equivalents being higher when EVOO was added. Phenolic content and AC in the EVOO decreased after cooking Mediterranean Diet vegetables, whereas the water was enriched after the boiling processes, particularly when oil was included.

Epidemiological studies have shown that consuming olive oil rich in phenolic bioactive compounds is associated with a lower risk of neurodegenerative diseases and better cognitive performance in aged populations. Since oxidative stress is a common hallmark of age-related cognitive decline, incorporating exogenous antioxidants could have beneficial effects on brain aging. In this review, we firstly summarize and critically discuss the current preclinical evidence and the potential neuroprotective mechanisms. Existing studies indicates that olive oil phenolic compounds can modulate and counteract oxidative stress and neuroinflammation, two relevant pathways linked to the onset and progression of neurodegenerative processes. Secondly, we summarize the current clinical evidence. In contrast to preclinical studies, there is no direct evidence in humans of the bioactivity of olive oil phenolic compounds. Instead, we have summarized current findings regarding nutritional interventions supplemented with olive oil on cognition. A growing body of research indicates that high consumption of olive oil phenolic compounds is associated with better preservation of cognitive performance, conferring an additional benefit, independent of the dietary pattern. In conclusion, the consumption olive oil rich in phenolic bioactive compounds has potential neuroprotective effects. Further research is needed to understand the underlying mechanisms and potential clinical applications.

Virgin Argan Oil (VAO) is extracted from the fruits of the Moroccan endemic species argan [Argania spinosa (L.) Skeels]. It is known for its richness in polyunsaturated fatty acids and its unique composition in tocopherols conferring a panoply of pharmacological properties, documented in several studies. The aim of this study was to investigate the potential protective effect of VAO against oxidative and nitrosative stress induced in cells of the model species Tetrahymena pyriformis. As a comparison, well-known Virgin Olive Oil (VOO) from Olea europaea L. was used instead. Both oils were subjected to a preliminary analysis of phytochemicals and properties of interest. Oxidative stress in T. pyriformis cells was induced by hydrogen peroxide (H2O2) at 350µM, whereas sodium nitroprusside (SNP) nitrosative stress was induced using a 1mM concentration. Neither of these concentrations caused relevant changes in cell viability. The level of reactive oxygen species (ROS) was evaluated in the cell cultures by using H2-DCFDA dye. The activity and cell localization of the antioxidant enzymes catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPX) was evaluated as well as the levels of glutathione (GSH) and the malondialdehyde (MDA) generated after lipid peroxidation. Finally, the presence and localization of intracellular lipid droplets was assessed by using Nile red. The treatment of cultures with H2O2 produced significant increases in the activity of CAT, SOD and GPX and in the level of GSH, as also did the SNP treatment. MDA level was increased by both the treatments. VAO and VOO treatment were found to protect T. pyriformis from oxidative stress and increased cells defense in nitrosative stress. In another hand VAO and VOO decreased significantly MDA level increased by H2O2 treatment and failed after SNP treatment. The quantification of fluorescence signal obtained from immunolocalization of antioxidant enzymes confirmed the results obtained after the evaluation of their activities. Interestingly the level of ROS and the number of lipid droplets increased by H2O2 treatment was significantly decreased by VAO and VOO co-treatments. VAO and VOO represent strong antioxidants, playing an important role in protecting cells against oxidative stress.

Oleuropein (Ole), a secoiridoid glucoside present in Olea europaea leaves, gained the interest of many scientists thanks to its several biological properties, including the anticancer one. We verified whether Ole might potentiate cytotoxicity of conventional drugs used to treat melanoma, disclosing new potential therapeutic strategy. We tested the cytotoxic action of Ole alone or in combination with chemotherapeutics on A375 human melanoma cells. We found that Ole was able, at a dose of 500 μM, to stimulate apoptosis in melanoma cells, while at a non-toxic dose of 250 μM, it affected cell proliferation and induced the downregulation of pAKT/pS6 pathway. 250 μM Ole did not potentiate the effect of Vemurafenib (PLX4032), but it succeeded in increase the cytotoxic effect of Dacarbazine (DTIC). The mayor effect was found in the association between Ole and Everolimus (RAD001), also on PLX4032-resistant BRAF melanoma cells, possibly cooperating in the inhibition of pAKT/pS6 pathway. Of interest, an olive leaf extract enriched in equimolar Ole was more effective and able to further improve DTIC and, particularly, RAD001 efficacy on BRAF melanoma cells than Ole alone. Therefore, Ole represents a natural product able to potentiate a wide array of chemotherapeutics against BRAF melanoma cells affecting pAKT/pS6 pathway.

The health benefits of extra virgin olive oil (EVOO) are related with its chemical composition and the presence of bioactive compounds with antioxidant properties. The aim of this study was evaluate antioxidant compounds (pigments, CoQ10 and phenolic compounds) and antioxidant properties of EVOO from the same region comparing different cultivars (Hojiblanca and Arbequina), harvest year and crop stage. Antioxidant properties of oils were studied before and after a gastrointestinal digestion process, by in vitro assays (DPPH, ABTS and FRAP) and antioxidant markers in Caco-2 cells (reactive oxygen species production). The content of bioactive compounds measured was significantly affected by cultivar and harvest year (except for carotenoids) and by the crop stage (except for coenzyme Q10). Higher amount of coenzyme Q10 was observed in Hojiblanca than in Arbequina EVOO. Total phenol content and antioxidant properties were also different depending on cultivar and harvest year and the in vitro digestion process strongly improved antioxidant marker values. Antioxidant potential in bioaccessible fractions was mainly related with content of coenzyme Q10 and phenolic compounds in EVOO. Chemometric analysis showed that the oils were clearly classified by cultivars, harvest and crop stage, according with chemical composition and antioxidant activity analyzed in the present study

Background: Extra virgin olive oil is a fundamental component of the Mediterranean diet. It contains several molecules that sustain human well-being by modulating cellular metabolism and exerting antioxidant, anti-inflammatory, anti-ageing effects to protect normal tissues and can exert anti-angiogenic, and pro-apoptotic effects on cancer cells. Metabolites found in different parts of the olive tree, including leaves, also possess properties that might help in cancer prevention and promote wellness in aging. Olive mill wastewater (OMWW), a liquid residue produced during olive oil extraction, represents an environmental issue. However, it is enriched in phytochemicals with potential beneficial properties. Dietary supplements based on OMWW can be produced for nutritional supplementation with advantages to the ecology. Purpose: This work aims to measure hematochemical, anthropometric and metabolomic parameters in volunteers assuming an OMWW dietary supplement, OliphenoliaⓇ (OMWW-OL). Methods: The supplementation of OMWW-OL 25 mL twice daily for 30 days was tested on a pilot cohort of volunteers with characteristics close to metabolic syndrome. Hematochemical, anthropometric, and metabolomic parameters were analyzed before the intervention, at 30 days and 30 days after stopping consumption. Results: 29 volunteers were enrolled and 23 completed the study. The participants’ parameters at baseline were measured, and then twice daily at 30 days of treatment and 30 days after assumption discontinuation. Although treatment was with an olive derivative, weight and body mass index instead of increasing slightly decreased, particularly in women. Also, hydration increased, especially in women, while blood pressure, glycemia and insulin decreased. Cholesterol, low-density and high-density lipoproteins and triglycerides were stable while vitamin D levels, alongside calcium, sensibly increased. Albumin also increased and parameters related to sarcopenia displayed promising, although not statistically significant, trends toward positive effects. All the values were in support of an equilibrium with no damaging effects. By mass spectrometry analysis, we also found favorable changes in the vitamin D/histamine and homocysteine/methionine ratios, an increase in a new metabolite of unknown formula, and vitamin D/unknown metabolite ratio. Conclusions: Supplementation of OMWW-OL has no detrimental effects and, although with limited potency of a pilot study, might imply the beneficial modulation of several biological parameters, being potentially valuable for people at risk of metabolic syndrome. Some of these parameters could also be relevant as anti-ageing and in cancer prevention.

The nuclear magnetic resonance (NMR) metabolomic analysis was applied to investigate the differences within nineteen Sicilian Nocellara del Belice mono varietal Extra Virgin Olive Oils (EVOO), grown in two zones different for altitude and soil composition. Several classes of endogenous olive oil metabolites were quantified through a nuclear magnetic resonance (NMR) three-experiment protocol coupled with a yet developed data-processing called MARA-NMR. This method, taking around one-hour experimental time per sample, faces the possible quantification of different class of compounds at different concentration’s range which would require at least three alternative traditional methods. NMR results were compared with the data of traditional analytical methods to quantify free fatty acidity (FFA), fatty acid methyl esters (FAMEs), and total phenol content. The presented NMR methodology is compared with traditional analytical practices and its consistency is also tested through slightly different data treatment. Despite the rich literature about the NMR of EVOOs, the paper points out that there are still several advances potentially improving this general analysis and overcoming the other cumbersome and multi-devices analytical strategies. Monovarietal EVOO’s composition is mainly affected by pedoclimatic conditions in turn relaying upon the nutritional properties, quality, and authenticity. Data collection, analysis and statistical processing are discussed touching the important issues related to the climate changes in Sicily and to the specific influence of pedoclimatic conditions.

We have studied the very large amount of literary works written by the Italian mystic Maria Valtorta to assess similarities and differences in her writings because she claims that most of them are due to mystical visions. We have used mathematical and statistical tools developed for specifically studying deep linguistic aspects of texts. The general trend indicates that the literary works explicitly attributable to Maria Valtorta differ significantly from her other literary works, that she claims are attributable to the alleged characters Jesus and Mary. Mathematically, they seem to have been written by different authors. The comparison with the Italian literature is very striking. A single author, namely Maria Valtorta, seems to be able to write texts so diverse to cover the entire mathematical range (suitable defined) of the Italian literature of seven centuries.

Lutein, a plant-derived xanthophyl-carotenoid, is an exceptional antioxidant and anti-inflammatory constituent found in food. Elevated concentrations of lutein found in human blood and plasma, due to high dietary intake, are beneficial against eye disease and improve cardiometabolic health. Lutein plays an important protective role against the development of neurological disorders, including Alzheimer’s disease (AD), multiple sclerosis (MS) and Parkinson’s disease (PD). It has also been shown to be beneficial for liver, kidney and respiratory health. Lutein, acting as a very strong antioxidant, can alleviate oxidative stress and downgrade reactive oxygen species (ROS). Oxidative stress is one of the key pathogenic mechanisms in post-COVID and mRNA vaccine injury syndromes. Recent in silico studies suggest that lutein and other naturally derived antioxidants, by docking at the site where the SARS-CoV-2 spike protein (SP) binds to the angiotensin enzyme type 2 (ACE2) receptor, may neutralize the SP-ACE2 interactions. Lutein can be added to a detoxification regimen to aid in clearing Spike protein and relieving symptoms. In agreement with Hippocrates’ dictum to “Let food be thy medicine,” this review establishes dietary lutein as a valuable therapy in the treatment of post-COVID syndrome, mRNA vaccine injury syndromes, and a wide range of other chronic illnesses.
