Dengue virus infections are a serious public health problem worldwide. Aedes aegypti is the primary vector of dengue in Cuba. Since there is no vaccine or specific treatment, the control efforts are directed to reduce mosquito populations. The indiscriminate use of pesticides can lead to increase insecticide resistance as well as adverse effects on human health. The sterile insect technique is a species-specific and environmental friendly method of insect control based on the release of large numbers of sterile males. The success of this technique in sustainable control of agricultural pests has encouraged its evaluation for mosquito control. Here, we describe an open field trial to evaluate the effect of the release of irradiated males on a wild population of Aedes aegypti. The case-control study was performed in a suburb of Havana, and compared the mosquito population before and after the intervention, in both control and treated areas. The wild population was monitored by an ovitrap network, recording frequency and density of eggs as well as their hatch rate. A significant induced sterility was observed in the field population, compared to the control. The ovitrap index and the mean eggs/ trap declined dramatically after an expected lag period of twelve and five weeks, respectively. For the last three weeks, no egg was collected in the treated area, evidencing a significant suppression of the wild population. We conclude that the sterile males released competed successfully, and induced enough sterility to suppress the local Aedes aegypti population.

Aedes albopictus is considered one of the major invasive species in the world and can transmit viruses such as dengue, Zika, or chikungunya. The Sterile Insect Technique (SIT) can be used to suppress the native populations of Ae. albopictus. To support the development of the release strategy during the SIT application, Mark-Release-Recapture (MRR) studies are crucial. Mean-while, weather conditions can affect the MRR trial’s results and it is critical to understand the influence of climatic factors on the results. In October 2022 in Faro, Southern Portugal, 84,000 ir-radiated sterile males were released for three consecutive weeks and mosquitoes were recapture by Human Landing Collections (HLC) one, two, four and six days after. Generalised linear models with a negative binomial family and log function were used to estimate the factors asso-ciated with the number of recaptured mosquitoes, prevalence ratios, and the 95% confidence in-tervals (CI). A total of 84,000 sterile male mosquitoes were released, with 528 recaptured (0.8%) by HLC. The prevalence of recaptured mosquitoes was 23% lower when the wind intensity was moderate. Sterile-marked males had an average median distance travelled of 88.7 m. Median probability of daily survival and the average life expectancy were 61.6% and 2.1, respectively. Wild male population estimates was 443.33 males/ ha. Despite no statistically significant asso-ciation was found with humidity, temperature and precipitation, it is crucial to consider weather conditions during MRR trials analyses to obtain the best determinants estimation and a more ef-ficient application of SIT in an integrated vector management program.

In this paper we investigate how the Reionization process is affected by early galaxy formation in different cosmological scenarios. We use a semi-analytic model with suppressed initial power spectra to obtain the UV Luminosity Function in thermal Warm Dark Matter and sterile neutrino cosmologies. We retrace the ionization history of intergalactic medium with hot stellar emission only, exploiting fixed and variable photons escape fraction models ( fesc). For each cosmology, we find an upper limit to fixed fesc, which guarantees the completion of the process at z <6.7. The analysis is tested with two limit hypothesis on high-z ionized hydrogen volume fraction, comparing our predictions with observational results.

This paper investigates an alternative use of sterile aggregate materials which may arise from various construction applications in conjunction with other low-cost mineral raw materials to remediate the acid mine drainage phenomenon. This study is based on the combination of unprocessed mineral raw materials as well as on the basic concept of the cyclic economy where the conversion of a waste into a raw material for another application can be achieved. In this way, the value of mineral raw materials can be prolonged for as long as possible, waste generation and exploitation of natural resources are minimized and resources are kept as far as possible within the existing economy. In this study, an electrically continuous flow driven forced device proposed and demonstrated for the remediation of waste water in lab-scale by using certain mixes of mineral raw materials (serpentinite, andesite, magnesite, peat and biochar). Our results focus on the impact of the studied mineral raw materials and especially on their synergy on the water purification potential under continuous water flow operation. Using the proposed 7-day experimental electrically continuous flow driven forced device with the certain mixes of mineral raw materials, the increase of pH values from 3.00 to 6.82 as well as significant removal of Fe, Cu and Zn was achieved.

Perturbation in the microbial population/colony index has harmful consequences on human health. Both biological and social factors influence the composition of the gut microbiota and promote gastric diseases. Changes in the gut microbiota manifest in disease progression owing to epigenetic modification in host which influences differentiation and function of immune cells adversely. Uncontrolled use of antibiotics; chemotherapeutic drugs and change in the diet pattern usually contribute to the changes in the colony index of sensitive strains known to release microbial content in the tissue micromillieu. Ligands released from dying microbes induce TLR mimicry on interaction with TLR abnormally which skew hypoxia and sterile inflammation contributing to severity of disease like IBD autoimmunity and cancer. Various modalities/interventions practiced across the globe and future strategies for microbiota based therapeutic approaches with special emphasis on tumor and inflammatory diseases are reviewed here. Therefore the major aim and scope of this manuscript is to both discuss various modalities/interventions across the globe and to design future microbiota based therapeutic approaches for mitigating the burden with special emphasis on tumor and Inflammatory diseases.

The field of nephrology has recently directed a considerable amount of attention towards the stimulator of interferon genes (STING) molecule since it appears to be a potent driver of chronic kidney disease (CKD). STING and its activator, the cyclic GMP-AMP synthase (cGAS) are among the most relevant inducers that promote the expression of type I interferons (IFN-Is). These cytokines have been long recognized as part of the mechanism used by the innate immune system to battle viral infections; however, their involvement in sterile inflammation remains unclear. Mounting evidence pointing to the involvement of the IFN-I pathway in sterile kidney inflammation provides potential insights into the complex interplay between the innate immune system and damage to the most sensitive segment of the nephron, the glomerulus. The STING pathway is often cited as the cause of renal disease originating from non-infection related caused by the induction of IFN-I expression. However, other receptors have been implicated in this process, mainly by recognizing host-derived nucleic acids. This review explores the main endogenous inducers of IFN-I in glomerular cells, discusses what exposure to the main autocrine and paracrine cytokines has on these cells, and identifies the pathways that are implicated in the development of glomerular damage.

Visceral Leishmaniasis is a neglected tropical disease affecting 12 million people annually. Even in the second decade of the 21st century, it has remained without an effective vaccine for human use. In the current study, we have designed three multiepitope vaccine candidates by the selection of multiple IFN-γ inducing MHC-I and MHC-II binder T-cell specific epitopes from 3 previously identified antigen genes of Leishmania donovani from our lab, by immune-informatic approach using IFNepiotpe, NET-MHC-1 and NET MHC-2 webservers. We have tested the protective potential of these three multiepitope proteins as vaccine in a hamster model of visceral leishmaniasis. The immunization data revealed that the vaccine candidates induced a very high level of Th-1 biased protective immune response in-vivo in a hamster model of experimental visceral Leishmaniasis, with one of the candidates inducing a sterile immunity. The vaccinated animals displayed highly activated monocyte macrophages with the capability of clearing intracellular parasites due to increased respiratory burst. Additionally, these proteins induced activation of polyfunctional T cells secreting INF-γ, TNF-α, and IL-2 in ex-vivo stimulation of human peripheral blood mononuclear cells, further supporting the protective nature of designed candidates.

We use Big Bang Nucleosynthesis (BBN) to probe Beyond Standard Model physics in the neutrino sector. Recently the abundances of primordially produced light elements D and He-4 were determined from observations with better accuracy. The good agreement between the theoretically predicted abundances of primordially produced and derived from observations light elements allows to update the BBN constraints on Beyond Standard Models (BSM) physics. We provide numerical analysis of several BSM models of BBN and obtain precise cosmological constraints and indications for new neutrino physics. Namely, we derive more stringent BBN constraints on electron neutrino-sterile neutrino oscillations corresponding to 1% uncertainty of the observational determination of the primordial He-4. The cosmological constraints are obtained both for the case of zero and non-zero initial population of the sterle neutrino state. Then in a degenerate BBN model with neutrino νe↔νs oscillations we analyze the change of the cosmological constraints in case lepton asymmetry L is big enough to suppress oscillations. We obtain constraints on the lepton asymmetry L . We discuss a possible solution to the dark radiation problem in degenerate BBN models with νe↔νs oscillations in case L is large enough to suppress neutrino oscillations during BBN epoch. Interestingly, the required value of L for solving DR problem is close to L indicated by EMPRESS experiment and close to the value of lepton asymmetry necessary to relax Hubble tension.

Ulcerative colitis is a chronic immune-mediated disease of unclear etiology, affecting people of different ages and significantly reducing the quality of life. Modern methods of therapy are mainly represented by anti-inflammatory drugs and are not aimed at a specific pathogenetic factor. In this study, we investigated the effect of transplantation of sterile stool filtrate from healthy donors on the induction of anti-inflammatory immune mechanisms. It was shown that performing such a procedure in patients with ulcerative colitis caused the appearance of T helper cells in the blood, which reacted to the content of sterile stool filtrates in an antigen-specific manner and produced IL10. At the same time, cells of the same patients before therapy in response to the addition of sterile stool filtrates were less reactive and predominantly produced IL4, indicating its pro-inflammatory skewing. The obtained data demonstrated the effect of an anti-inflammatory shift in the T-helper response after transplantation of sterile stool filtrate, which increased and persisted for at least three months after the procedure.

Sperm marking represents a valuable monitoring tool for genetic pest control strategies such as the Sterile Insect Technique, but also provides a key tool for reproductive biology studies. Sperm-marked lines can be generated by introducing transgenes that mediate the expression of fluorescent proteins during spermatogenesis. Homozygous lines established by transgenesis approaches are going through a genetic bottleneck that can lead to reduced fitness. Transgenic SIT approaches have mostly focused on Dipteran and Lepidopteran pests so far. With this study, we provide sperm-marked lines for the Coleopteran pest model organism, the red flour beetle Tribolium castaneum, based on the β2-tubulin promoter/enhancer driving red (DsRed) or green (EGFP) fluorescence. The obtained lines are reasonably competitive and were thus used for studies on reproductive biology confriming the phenomenon of ‘last male sperm precedence’ and that the spermathecae are deployed for long term sperm storage enabling the use of sperm from first matings even after secondary matings for a long period of time. The homozygosity and competiveness of the lines will enable future studies to analyze the controlled process of sperm movement into the long time storage organ as part of a post-mating cryptic female choice mechanism of this extremely promiscuous species.

Abstract: Background: COVID-19 is associated with various changes in immunity, including signaling through the pattern-recognition receptors (PRR). However, the role of these processes is poorly understood. The present study aimed to evaluate the relationships of the serum levels of the cyclic dinucleotide 2’3’-cyclic GMP-AMP (cGAMP) marker of PRR activation with immunoglobulin G antibodies against severe acute respiratory syndrome-linked coronavirus (IgG-SARS)-positive status and endothelial dysfunction in apparently healthy subjects and in patients with coronary heart disease (CHD). Methods: The present study investigated selected groups from two cohorts, including cohort 1 of 307 healthy volunteers and cohort 2 of 218 CHD patients. COVID-19 infection was confirmed by the detection of IgG-SARS against SARS-CoV-2 S1 protein receptor-binding domain. Cohort 1 (24–69 years of age; 44.8 ± 8.6 years; 80.4% men) was examined for systematic coronary risk evaluation by European Society of Cardiology (SCORE) starting from 2019 before the onset of the COVID-19 pandemic. Cohort 2 (63±10.9 years of age, 54% men) was processed starting from 2017 (three years prior to the COVID-19 pandemic) in a hospital setting to undergo coronary angiography to assess coronary lesions as Gensini score. The levels of cGAMP and endothelial markers (nitrate and nitrite combined as NOx and endothelin-1) were assessed in the serum to evaluate the associations with IgG-SARS status, SCORE, and extent of coronary lesions by correlation and receiver operating characteristic (ROC) analyses. Results: Serum cGAMP did not discriminate between SARS-positive and SARS-negative healthy subject of cohort 1. Moreover, the level of cGAMP was not associated with endothelial biomarkers in healthy subjects. However, serum cGAMP was associated with atherosclerosis in cohort 2 in patients with CHD (area under the curve (AUC) 0.69; 95% CI 0.587-0.806; P=0.001), indicating that serum cGAMP was lower in patients with coronary stenosis compared to that in patients without coronary lesions. Conclusions: Low levels of serum cGAMP were associated with atherosclerosis and markers of endothelial dysfunction in patients with CHD. However, cGAMP was not associated with endothelial biomarkers or IgG-SASR immune status in healthy subjects, suggesting that cGAMP may be a new biomarker of coronary lesions in the context of sterile inflammation.

Abstract Background: Occipital neuralgia is a debilitating condition and traditional treatments often provide limited or temporary relief. Recently, ultrasound-guided hydrodissection of the greater occipital nerve (GON) has emerged as a promising minimally invasive approach. Objectives: To describe two novel ultrasound-guided hydrodissection with 5% dextrose for GON and discuss their advantages, disadvantages, and considerations. Methods: Two case reports are presented. Case 1 describes a lateral decubitus approach for hydrodissecting the GON between the semispinalis capitis (SSC) and obliquus capitis inferior (OCI) muscles. Case 2 details a cranial-to-caudal approach for hydrodissecting the GON within the SSC and upper trapezius (UT) muscles when the GON pass through these two muscles. Results: Both patients experienced significant and sustained pain relief with improvements in function and quality of life. Conclusions: Ultrasound-guided GON hydrodissection using 5% dextrose is a promising treatment for occipital neuralgia. The lateral decubitus and cranial to caudal approaches provide additional options to address patient-specific anatomical considerations and preferences.
