The phytophagous insect pests were vectors and could transmit the majority of the phytoviruses to their host plants. The orders of Hemiptera and Thysanoptera were the most common vectors of phytoviruses. The orders Orthoptera, Dermaptera, Coleoptera, Lepidoptera, Diptera, Thysanoptera, and Hemiptera were also the vectors of phytoviruses. Furthermore, aphids, whiteflies, cicadas, spittlebugs, leafhoppers, planthoppers, assassin bugs, plant bugs, stink bugs, lygaeid bugs, and thrips were among the most phytophagous insects that vector and transmit phytoviruses to healthy plants. The occurrence of a single species of these phytophagous insects resulted in one or more phytoviruses in general, and the Hemipteran order, in particular, vectored a lot of phytoviruses species. This review manuscript is focused on vectors of phytoviruses, techniques for their detection, and future directions. It will play a vital role in exploring scientific information concerning the interactions of phytoviruses and vector insects, the effect of phytoviruses on host behavior, mediators of phytoviruses transmission, persistent phytoviruses, some other insect vectors of the phytopathogen, mechanisms of host plant resistance against phytoviruses, and techniques of phytoviruses detection, as well as some important points to be considered in the future sustainably.

Ecologists use the net biotic interactions among plants to predict fundamental ecosystem features. Following this approach, ecologists have built a giant body of theory founded on observational evidence. However, due to the limitations that a phenomenological approach raises both in empirical and theoretical studies, an increasing number of scientists claim the need for a mechanistic understanding of plant interaction outcomes, and a few studies have taken such a mechanistic approach. In this synthesis, we propose a modeling framework to study the plant interaction mechanistically. We first establish a conceptual ground to frame plant-plant interactions, and then, we propose to formalize this research line theoretically developing a family of individual-based, spatially-explicit models in which biotic interactions are an emergent property mediated by the interaction between plants’ functional traits and the environment. These models allow researchers to evaluate the strength and sign of biotic interactions under different environmental scenarios and thus constitute a powerful tool to investigate the mechanisms underlying facilitation, species coexistence, or the formation of vegetation spatial patterns.

Plant-microbe interactions play a crucial role in shaping plant health and survival. In recent years, the role of extracellular vesicles (EVs) in mediating intercellular communication between plants and microbes has emerged as an intriguing area of research. EVs, serve as important carriers of bioactive molecules and genetic information, facilitating communication between cells and even between different organisms. Pathogenic bacteria leverage extracellular vesicles (EVs) to amplify their virulence, exploiting their cargo rich in toxins and virulence factors. Conversely, beneficial microbes initiate EV secretion to stimulate plant immune responses and nurture symbiotic relationships. The transfer of EV-packed small RNAs (sRNAs) has been demonstrated to facilitate the modulation of immune responses. Furthermore, harnessing the potential of EVs holds promise for the development of innovative diagnostic tools and sustainable crop protection strategies. This review highlights the biogenesis and functions of EVs in bacteria, and their importance in plant defense and paves the way for future research in this exciting field.

Global crop yield and food security are being threatened by phytophagous insects. Innovative methods are required to increase agricultural output while reducing reliance on hazardous synthetic insecticides. It appears to be quite effective at reducing production costs and boosting farm profitability to use the ground-breaking CRISPR-Cas technology to create plants that are insect resistant. In contrast, this new technique can modify an insect's genome to either produce gene drive or get beyond an insect's tolerance to various insecticides. This paper reviews and critically discusses the use of CRISPR-Cas genome editing technology in long-term insect pest management. The emphasis of this review is on the prospective uses of the CRISPR-Cas system for insect stress management in crop production by creating genome-edited crops and insects. The potential and difficulties of using CRISPR-Cas technology to reduce pest stress in crop plants are critically examined and discussed.

The prevention of destruction caused by red flour beetle, Tribolium castaneum (Herbst), using essential oils derived from certain herbal plants presents an interesting alternative to synthetic insecticides, known for their harm to both consumers and the environment. The research aimed to investigate the chemical compositions and fumigating effects of essential oils derived from cardamom (Elettaria cardamomum (L.) Maton) and galangal (Alpinia galanga (L.) Willd) against T. castaneum. Chemical compositions were analyzed using GC-MS, while fumigation bioassay was conducted via a vapor-phase test. Experimental setup followed factorial design in CRD with 5 replications. Factor A encompassed 4 types of essential oils: the manually extracted essential oils from cardamom leaves (MCL) and galangal leaves (MGL), the commercially produced essential oils from cardamom seeds (CCS) and galangal rhizomes (CGR). Factor B consisted of 7 concentrations: 0, 50, 100, 150, 200, 250, and 300 µL/L air. Results indicated that MCL contained eucalyptol, trans-calamenene, and isospathulenol as its main compounds. Similarly, CCS was characterized by camphene and eucalyptol. MGL exhibited caryophyllene, aciphyllene, and α-bisanolene. Lastly, CGR was composed of methyl cis-cinnamate, safrole, and p-vinylphenyl isothiocyanate. Study noted that 250 µL/L air over 168 hours of CGR, demonstrated 94% fumigation efficiency against adult T. castaneum. These results suggest the potential of galangal rhizome derived essential oil as an insecticide for managing T. castaneum populations in agricultural product storage.

Proteases are integral enzymes of the plant immune system. Multiple aspects of defence are regulated by proteases, including the hypersensitive response, pathogen recognition, priming and peptide hormone release. These processes are regulated by unrelated proteases residing at different subcellular locations. In this review we discuss ten prominent plant proteases contributing to the plant immune system, highlighting the diversity of roles they perform in plant defence.

Nutrients are the driving force of plant and herbivore interactions. Different elements and fertilization levels in soil, or artificially fertilized substrates can have a direct impact on plant defense responses. Meanwhile, nutrient levels mediated by root associated microbes can indirectly affect plant-herbivore interactions. Nutrient uptake promoted by beneficial mycorrhizal fungi or rhizobacteria interacting with roots can stimulate plant tolerance to herbivore attack, as evidenced by herbivore performance, plant nutrient status, plant biomass allocation and compensatory growth. In return, herbivory affects nutrient fluxes between plants and their associated beneficial microbes. Furthermore, aboveground and belowground herbivory can affect the nutrient exchange between plants and soil microbial communities, mainly for carbon-and nitrogen-based resources supporting herbivores´ life cycles. These ecological interactions are governed by jasmonic acid (JA) and key enzymes on the molecular level. This review summarizes the current scientific literature on the role of nutrients in plant-herbivore interactions. This will support efforts to optimize plant defense strategy via managing nutrient variance.

The concept of omics-based technological approaches has promoted translational research by integrating knowledge from diverse areas to understand their dynamics by exploring the molecular mechanisms underlying various processes paving the way for further improvements in the crops quality by providing sound knowledge on controlling plant diseases. This area is not profoundly investigated a decade back and hence this knowledge has become exceedingly important in modern times as it has got potential for being utilized in crop improvement programs. In this review, the contributions of different omics technologies including genomics, transcriptomics, proteomics, and metabolomics in understanding and materializing the ways involved in the plant pathogen interaction (PPI) is discussed. Furthermore, opportunities, challenges, and perspectives of omics linked to signaling mechanisms have also been highlighted that are significantly linked to plant-microbe interactions.

Global agriculture has been forced to increase food production to feed the growing human population, while confronting various environmental obstacles such as global warming, resistance to pathogens, and constraints on arable land caused by soil salinity, drought, rising sea level, saltwater intrusion, and urbanization. Regarding abiotic stresses, salinity is a worldwide problem for agricultural production. Many efforts, therefore, have been made to cope with the environmental challenges, however, the progress of salinization, which is mainly caused and accelerated by anthropogenic activities, is likely faster than our progress in finding ways to deal with this problem. In addition, drought represents a global threat to the production of major crops. In addition, pests and pathogens cause significant crop losses and diminish global food security. Among the various strategies that have been investigated and applied in plant science, antimicrobial peptides derived from plants have caught widespread attention from scientists since these peptides exhibit beneficial biological activities. In agricultural science, there have been reports on the roles of antimicrobial peptides with active properties against biotic and abiotic stresses. Non-specific plant lipid transfer proteins, thionins, systemins, defensins, cyclotides, and heveins-like antimicrobial peptides are common antimicrobial peptides that have been found to be involved in the defense system against fungi and insect pests. Based on their potential ability to protect crops from pests, bacteria, and pathogenic fungi, the use of antimicrobial peptide genes in creating transgenic plants has been largely conducted during the last decades, and these studies have obtained positive results against the growth of fungi and bacteria. This review will focus on the latest progress in studies of antimicrobial peptides related to biotic and abiotic stress tolerance in plants. We will also update the current progress in the development of antimicrobial peptide-based transgenic crops.

Plant-microbe interactions are essential for ecosystem health and agricultural productivity. Metabolomics has revolutionized our understanding of these interactions by providing powerful methods to decipher the complex chemical signals involved. This review highlights the various metabolomics techniques used to study plant-microbe interactions, such as mass spectrometry, nuclear magnetic resonance spectroscopy, and advanced imaging technologies. We also discuss the role of contemporary bioinformatics tools, extensive databases, and the increasing influence of artificial intelligence and machine learning in the analysis of metabolomics data. Integrating metabolomics with other omics fields, including transcriptomics, proteomics, and metagenomics, allows for a comprehensive understanding of the molecular mechanisms underlying plant-microbe interactions. The review emphasizes the significance of fluxomics in elucidating the dynamic nature of metabolic networks. While there are challenges in data analysis, integration, and interpretation, metabolomics has profoundly expanded our knowledge of the role metabolites play in shaping plant-microbe interactions. As metabolomics technologies advance and integrate further with other omics approaches, we anticipate groundbreaking discoveries that will provide deeper insights into these complex interactions, ultimately leading to innovative strategies for sustainable agriculture and biotechnology.

Beneficial microbes are crucial for improving crop adaptation and growth under vari-ous stresses. They enhance nutrient uptake, improve plant immune responses, and help plants tolerate stresses like drought, salinity, and heat. The yield potential of any crop is significantly influenced by its associated microbiomes and their potential to im-prove growth under different stressful environments. Therefore, it’s crucial and excit-ing to understand the mechanisms of plant-microbe interactions. Maize (Zea mays L.) is one of the primary staple foods worldwide, in addition to wheat and rice. Maize is also an industrial crop globally, contributing 83% of its production for use in feed, starch, and biofuel industries. Maize requires significant nitrogen fertilization to achieve opti-mal growth and yield. Maize plants are highly susceptible to heat, salinity, and drought stresses and require innovative methods to mitigate the harmful effects of environmental stresses and reduce the use of chemical fertilizers. This review summarizes our current understanding of the beneficial interactions between maize plants and specific microbes. These beneficial microbes improve plant resilience to stress and in-crease productivity. For example, they regulate electron transport, downregulate cata-ase, and upregulate antioxidants. We also review the roles of plant growth promoting rhizobacteria (PGPR) enhancing stress tolerance in maize. Additionally, we explore the application of these microbes in maize production and identify major knowledge gaps that need to be addressed to utilize the potential of beneficial microbes fully.

Major disturbances fundamentally alter ecosystems through the transformation of both biotic and abiotic factors. The eruption of Mount St. Helens in 1980 resulted in a cataclysmic restructuring of its surrounding landscapes. The Pumice Plain is one of these landscapes, where tree species such as Sitka willow (Salix sitchensis) and their dependent communities have established along newly-formed streams. Thus, the study of these dependent communities provides a unique opportunity to investigate factors influencing metacommunity assembly during true primary succession. We analyzed the influence of landscape connectivity on metacommunity assembly through a novel application of circuit theory, alongside the effects of other factors such as stream locations, willow leaf chemistry, and leaf area. We found that landscape connectivity structures community composition on willows across the Pumice Plain, while our other factors had varied effects. Most importantly, multiple levels of spatial habitat structure linked via landscape connectivity can predict the presence of organisms lacking high rates of dispersal, such as the invasive stem-boring poplar weevil (Cryptorhynchus lapathi). This is critical for management as we show that the maintenance of a heterogeneous mixture of landscape connectivities and resource locations can facilitate meta-community dynamics to promote ecosystem function and mitigate the influences of invasive species.

Tomato (Solanum lycopersicum L.) is a valuable horticultural crop grown and consumed worldwide. Optimum production is hindered by several factors of which Verticillium dahliae, the cause of Verticillium wilt, is one of the major biological constraints in temperate production regions. V. dahliae is difficult to manage because it is a vascular pathogen, has a broad host range and worldwide distribution, and can persist in soil for years. Understanding the pathogen virulence and genetic diversity, host resistance, and plant-pathogen interactions can ultimately inform the development of integrated strategies to manage the disease. In recent years, considerable research has focused on providing new insight into these processes as well as the development and integration of environment-friendly management approaches. In this review, we discuss and summarize the recent findings on the race and population structure of V. dahliae; pathogenicity factors; host genes, proteins, and enzymes involved in defense; the emergent management strategies, and recent approaches to managing Verticillium wilt in tomatoes.

Bacillus thuringiensis (Bt) is a spore-forming bacterium that produces insecticidal proteins and other virulence factors and is considered one of the most successful bioinsecticides available to control pests in agriculture. Bt strains have been reported as endophyte or rhizospheric bacteria, but little is known about the implications of this property of Bt in crop protection. Here, we review if Bt can establish as an endophyte/rhizobacterium and evaluate if Bt as an endophyte/rhizobacterium can simultaneously act against different phytopathogens (fungi, bacteria, insects and viruses) plus promote plant growth. The implications of the proposed review will broaden our understanding of Bt as a versatile entomopathogen by exhibiting differential behavior depending on context.

Insect meal as a protein source has been considered a sustainable way to feed animals. H. illucens and T. molitor larvae meal are considered high protein sources for poultry, also presenting considerable amounts of fatty acids, vitamins, and minerals. However, other potential components in insect meal and insect oil have been more studied nowadays. Chitin, lauric acid, and antimicrobial peptides can present antimicrobial and prebiotic functions, indicating that low inclusions of insect meal can beneficially affect broilers` health and immune responses. This systematic review was developed to study the impact of insect products on health parameters of broilers, and a metanalysis was conducted to evaluate the effects on performance. A database was obtained based on a selection of manuscripts from 2016 to January 2023, following the mentioned parameters. There were positive effects of both H. illucens and T. molitor meal or oil products on poultry health status, especially in microbiota population, immune responses, and antimicrobial properties. The average daily gain was greater in broilers fed T. molitor meal compared to H. illucens meal (p = 0.002). The results suggest that low levels of insect meal were suitable for broilers, without result in negative effects on performance, while the insect oil can totally replace soybean oil without negative impacts.

Epichloë species form bioprotective endophytic symbioses with many cool-season grasses, including agriculturally important forage grasses. Despite its importance, relatively little is known about the molecular details of the interaction and the regulatory genes involved. VelA is a key global regulator in fungal secondary metabolism and development. In previous studies, we showed the requirement of velA for E. festucae to form a mutualistic interaction with Lolium perenne. We showed that VelA regulates the expression of genes encoding proteins involved in membrane transport, fungal cell wall biosynthesis, host cell wall degradation and secondary metabolism, along with several small-secreted proteins in Epichloë festucae. Here, by a comparative transcriptomics analysis on perennial ryegrass seedlings and mature plants, which are endophyte free or infected with wild type (mutualistic interaction) or mutant ∆velA E. festucae (antagonistic or incompatible interaction), regulatory effects of the endophytic interaction on perennial ryegrass development was studied. We show that ∆velA mutant associations influence the expression of genes involved in primary metabolism, secondary metabolism and response to biotic and abiotic stresses compared to wild type associations, providing an insight into processes defining mutualistic versus antagonistic interactions.

In recent years, the potential of insects has been explored as a sustainable protein alternative to sustain the growing world population. Differences in the way insects are processed can affect their proximate composition and digestibility. This work studies the effects of the combination of different types of slaughter methods and drying temperatures on the proximate composition, organic matter digestibility (OMd), hydrolysis degree (DH/NH2 and DH/100 g DM), total hydrolysis (TH) and hygienic sanitary study of BSFL (black soldier fly larvae). Four types of slaughter methods were used: frozen (F), blanched + frozen (B), Melacide® + frozen (M) and liquid nitrogen (N), each with three drying temperatures (50, 70 and 90 °C). A negative correlation between acid detergent fibre (ADF) and protein digestibility parameters was obtained. 70 °C was the most suitable drying temperature, as it reported higher values of protein digestibility (DH, TH), resulting in hygienic and sanitary conditions suitable for food use. Slaughtering with liquid nitrogen and blanching were more convenient than traditional freezing or the use of Melacide® to achieve higher protein digestibility results.

The populations of clearwing moth borers in Korea have recently exploited and severe damage on pin oaks (Quercus palustris Munchh.). We conducted the field monitoring and the molecular analyses to identify them in an integrated manner. Morphological examination and molecular analyses on COI-gene based intra-and inter-specific genetic divergences (GD) were revealed that the borers were recognized as two invasive species, Sphecodoptera sheni and Paranthrenella pinoakula sp. nov.. The maximum intraspecific GD was found to be 1.9%, whereas the minimum interspecific GD was confirmed as 8.1%, with a distinct barcoding gap. Both MJ network and NJ tree were also showed that 18 haplotypes (HAPs) were detected from the 52 COI sequences. The borers were revealed total 17 HAPs: i) H1-H7 were detected from all seven regions with S. sheni; ii) Wonju and Goyang populations of S. sheni were revealed as more than three HAPs; iii) H7 was closely connected with H8 of Chinese population of S. sheni; iv) H9-H10 were detected as the other samples from Wonju population with P. pinoakula sp. n., and closely located with congeneric species. Maximum likelihood tree also revealed that P. pinoacula sp. n. nested within the congeneric species by genetically separating from the S. sheni.

This paper described a crystallization procedure of the “1-Octadecanol/n-Nonadecane” binary system, an accurate description of the phase change process and the associated analysis of the cooling and melting of this binary system between 20 and 70 degrees Celsius. The introduction section mainly describes the scientific background and principle of plant cuticle wax layer and the former studies on the crystallization of n-Alkanes and n-Alkanols. The experiment part introduces the experimental chemicals, instruments and specific experimental operating procedures required for the preparation of the “1-Octadecanol/n-Nonadecane” binary system. In the following sections, the thermal properties of this binary system are studied. According to the DSC diagram, phase diagrams and absorption spectrum, the specific phase transition and enthalpy of each phase transition were studied. The cooling and heating phase diagrams were plotted. The influence of the different compositions of these two chemicals was analyzed and discussed. The last part is the conclusion of this thesis, which summarizes the experimental research results of the complete text and points out the needed further research.

Notch signaling is a highly conserved pathway which occurs between adjacent cells to distinguish them. The Notch signaling cascade operates in a simple mechanism, yet Notch plays remarkably diverse roles in development to regulate cell fate determination, organ growth and tissue patterning. Originally isolated and characterized in the model insect Drosophila melanogaster, recent studies in various insect species have revealed that Notch signaling is broadly involved in the construction of insect tissues. This review focuses on providing a molecular mechanistic and developmental picture regarding the roles of Notch pathway in insects. The functions of Notch in the formation and patterning of insect embryo, wing, leg, ovary and several specific structures as well as in stress responses are summarized. These results are discussed in developmental context with an eye toward understanding the functionality of Notch signaling pathway in different insect species.

In Poland, there has recently been a lot of interest in the possible use of insects as a new food source. Since they have many nutritional and environmental advantages, insects are a promising and sustainable source of protein. Although, in many parts of the world insects are a highly valued food, in Poland the development of insect-based food has only recently started, and the availability of products on the market is constantly increasing. This study examines the market availability of food products with insect additives in Poland, considering both online and sta-tionary stores. The assortment of products was analyzed based on the country of origin, insect species, form of insects, and product category. Polish consumers have more access to in-sect-based food products than ever before, with whole insects, protein bars, and powdered ver-sions predominating the market's offers..

RNAi is considered a major antiviral defense mechanism in insects but its relative importance compared to other antiviral pathways has not been evaluated comprehensively. Here, it is attempted to give an overview of the antiviral defense mechanisms in Drosophila that involve both RNAi and non-RNAi to acquire a sense of their relative importance. While RNAi is considered important in most viral infections, many other pathways can exist that confer antiviral resistance. It is noted that very few direct recognition mechanisms of virus infections have been identified in Drosophila and that the activation of immune pathways may be accomplished indirectly through cell damage incurred by viral replication. In several cases, protection against viral infection can be obtained in RNAi mutants by non-RNAi mechanisms, confirming the variability of the RNAi defense mechanism according to the type of infection and the physiological status of the host. This analysis invites to investigate more systematically the relative contribution of RNAi in the antiviral response and more specifically to ask whether RNAi efficiency is affected when other defense mechanisms predominate. While Drosophila can function as a useful model, this issue may be more critical for economically important insects that are either controlled (agricultural pests and vectors of diseases) or protected from parasite infection (beneficial insects as bees) by RNAi products.

Genes that affect adaptive traits have been identified, but our knowledge of the genetic basis of adaptation in a more general sense (across multiple traits) remains limited. We combined population-genomic analyses of evolve and resequence experiments, genome-wide association mapping of performance traits, and analyses of gene expression to fill this knowledge gap, and shed light on the genomics of adaptation to a marginal host (lentil) by the seed beetle Callosobruchus maculatus. Using population-genomic approaches, we detected modest parallelism in allele frequency change across replicate lines during adaptation to lentil. Mapping populations derived from each lentil-adapted line revealed a polygenic basis for two host-specific performance traits (weight and development time), which had low to modest heritabilities. We found less evidence of parallelism in genotype-phenotype associations across these lines than in allele frequency changes during the experiments. Differential gene expression caused by differences in recent evolutionary history exceeded that caused by immediate rearing host. Together, the three genomic data sets suggest that genes affecting traits other than weight and development time are likely to be the main causes of parallel evolution, and that detoxification genes (especially cytochrome P450s and beta-glucosidase) could be especially important for colonization of lentil by C. maculatus.

Immunohistochemistry (IHC) helps identify different types of tissues and provides important information about the pattern, shape, and structure of cells in a tissue sample. It is a very useful tool and widely used to uncover histology and pathology diagnosis uses. The IHC is a very useful tool to conduct research in the developmental biology area. Some insects, like butterflies, bees, beetles, ants, wasps, and moths, undergo a unique life cycle called complete metamorphosis, which has four distinct stages: egg, larva, pupa, and adult. Larvae are often wingless and have different habits and forms than adults, which are better suited for growth and development. During the metamorphosis, how does the insect legs change is a really an interesting issue.To better understand insect development, agriculture pest control, and insect pathogen interaction, here we use an insect Bombyx mori (domestic silk moth) as a model to detail how to conduct the IHC assays, such as HE staining, TUNEL to see the cell apoptosis, pH3 and BrdU staining to check cell division. The reagents can be easily found, and this step-by-step protocol is easily reproduced.

In the course of our ongoing research targeting the identification of potential biocontrol agents from entomopathogenic fungi (EPF), we explored a solid-state rice fungal extract of Blackwellomyces roseostromatus BCC56290 derived from infected lepidopteran larvae. Chemical and biological prospections afforded four unprecedently reported natural products differentiated into one dimeric naphthopyran bioxanthracene ES-242 derivative (1) and three cyclodepsipeptides (2-4) in addition to two known cyclodepsipeptides, cardinalisamides B (5) and C (6). Chemical structures of the isolated compounds were elucidated through comprehensive 1D/2D NMR and HR-ESI-MS data together with comparisons to the reported literature. The absolute configuration of the isolated cyclodepsipeptide was determined using Marfey’s method. All isolated compounds were assessed for their antimicrobial, cytotoxic and nematicidal activities with some compounds revealed significant activities.

Tree members of the genus Acacia have benefits that are obvious for enhancing soil fertility in farming, forestry and agroforestry in regions with nutrient-poor soils, and for restoring degraded ecosystems and lands. Nevertheless, the species of the genus Acacia have got the potential to bring about significant adverse impacts on biodiversity or ecosystem functioning when it gets invasive. The ecology of the species in nearly all areas of its created range remains poorly understood. Here we have compiled the information regarding the importance, cultivation and the production of important species of genus Acacia. We hope this information will be useful to get awareness about the crucial trees in the genus Acacia.

Sperm marking represents a valuable monitoring tool for genetic pest control strategies such as the Sterile Insect Technique, but also provides a key tool for reproductive biology studies. Sperm-marked lines can be generated by introducing transgenes that mediate the expression of fluorescent proteins during spermatogenesis. Homozygous lines established by transgenesis approaches are going through a genetic bottleneck that can lead to reduced fitness. Transgenic SIT approaches have mostly focused on Dipteran and Lepidopteran pests so far. With this study, we provide sperm-marked lines for the Coleopteran pest model organism, the red flour beetle Tribolium castaneum, based on the β2-tubulin promoter/enhancer driving red (DsRed) or green (EGFP) fluorescence. The obtained lines are reasonably competitive and were thus used for studies on reproductive biology confriming the phenomenon of ‘last male sperm precedence’ and that the spermathecae are deployed for long term sperm storage enabling the use of sperm from first matings even after secondary matings for a long period of time. The homozygosity and competiveness of the lines will enable future studies to analyze the controlled process of sperm movement into the long time storage organ as part of a post-mating cryptic female choice mechanism of this extremely promiscuous species.

Reproductively lethal mutations (RLMs) are mutations that, upon expression of the encoded lethal phenotypes, cause individuals carrying them to die or to be sterile. An underappreciated fact is that loss-of-function RLMs in protein-coding genes can be phenotypically shielded by favorable environments rendering these genes conditionally non-essential, or by their sister alleles in diploid organisms. Absent of rigorous mitigation, such phenotype-shielding causes the number of genes incurring RLMs to increase over time, and simultaneously allows each RLM to reach high allele frequencies in a conspecific population. Over-accumulation of RLMs then sets the population up for eventual concurrent expression of large numbers of RLMs, and massive deaths in rapid succession, possibly even population-level extinction. This hypothetical scenario in turn predicts that organismal lineages that evolved means to minimize the allele frequencies of phenotypically shielded RLMs are favored by natural selection. We argue that bottlenecking the genome copies destined for reproduction is a universal strategy adopted by all living beings to compel phenotype-based RLM purging. We further postulate that primitive RNA replicons must first evolve bottlenecked reproduction before evolving the capacity to encode diffusible products. In more complex, multicellular organisms, RLM management through bottlenecked reproduction gains additional reinforcement through sexual reproduction. In short, the evidence chronicled in this essay strongly suggest that the Bottleneck, Isolate, Amplify, Select (BIAS) principle, originally proposed to explain intracellular evolutionary dynamics of viruses, may be universally applicable to all living beings.

The molecular mechanisms underlying insect gall formation remain unclear. A major reason for the inability to identify the responsible genes is that most insect galls are produced on woody plants and therefore cannot be experimentally validated in the laboratory. To overcome these problems, we established a new galling insect model, Smicronyx madaranus. Our manipulation experiments using nail polish sealing and insecticide treatment revealed an age-dependent change in gall formation during S. madaranus; adult females and larvae are responsible for gall induction and enlargement, respectively. Furthermore, it has been suggested that substances released during oviposition and larval feeding are involved in each process. Phylogenetic analysis showed that most gall-forming weevils, including S. madaranus, belong to a single lineage, suggesting that gall-forming traits evolved in a specific lineage within Smicronyx. The efficacy of RNA interference (RNAi) in S. madaranus was confirmed by targeting multicopper oxidase 2 gene. It is expected that the mechanisms of gall formation will be elucidated by a comprehensive functional analysis of candidate genes using RNAi and the S.madaranus galling system in the near future.

Plant conciousness is a scenario where people imbibe the culture of engaging plants as a constant part of their day to day activity. The earth supports both plants and animals, but , plants unlike animals are unable to express themselves by shouting, nagging or crying, e.t.c. They are only able to response saliently thus making them oblivious and constantly neglected. Unfortunately, plants play just as much major roles in sustaining the earth as much as people and animals do. The proper handling of plants is much reliant on the ‘’Plant consciousness or obliviousness’’ of people including both the young and the old. Are people well aware of plants, how important they are to the earth, the environment and it sustenance?, the answer is not enough. There is a need for plant knowledge bridges to create more and adequate awareness about plants, their importance, how they should be handled, how they should be cared for, and an understanding that they are partners for the sustaining our dying world. We seek to change this obvious lack of appreciation and understanding of plants, but how can this be done? We discuss methodologies that have and can been used for bridging this knowledge gap to ensure that our world is safe by protecting and caring for plants.

Poplar (Populus) is a genus of woody plants of value for the pulp industry, green energy, urban landscaping, soil phytoremediation and as an ornamental plant. Due to the increasing economic importance of poplar, there is a need to ensure its stable growth by increasing its resistance to pathogens, as one of the main problems in poplar cultivation is its susceptibility to numerous diseases. Currently, breeding poplars with reduced susceptibility to infections is receiving increasing attention. Genetic engineering can produce organisms with improved traits faster than traditional methods, and with the development of CRISPR/Cas-based genome editing systems, scientists have a new highly efficient tool at their disposal to create valuable genotypes. In this review, we summarize recent research data on poplar diseases, the biology of their pathogens, and how these plants resist pathogens. In the final section, we discuss genetic engineering, genome editing, and microbiome strategies that we believe are suitable for obtaining new improved poplar lines.

Pyrops candelaria is one of the common pests of fruit trees, but the research on the pathogenic microorganisms it may carry is very limited. Therefore, it is essential to reveal the pathogenic microbes it carries and their potential hazards. This study found a new virus from the transcriptome of P. candelaria, which was first reported in P. candelaria and named as PyCaV (Pyrops candelaria associated virus). RACE and bioinformatics assay revealed that the full-length of PyCaV is 10855 bp with the polyA tail, containing a single open reading frame (ORF) encoding a polyprotein consisting of 3171 amino acid (aa). The virus has a typical Iflavirus structure, including two rhv domains, a RNA helicase domain (HEL), a 3C cysteine protease domain (Pro), and a RNA-dependent RNA polymerase domain (RdRp). Further phylogenetic analysis revealed that this virus belongs to family Iflaviridae and sequence alignements analysis suggedted PyCaV is new member in unassigned genus of family Iflaviridae. Further in-depth analysis of virus-infection-part showed that PyCaV is distributing in the whole longan lanternfly, including its head, chest, and abdomen, but more PyCaV was identified in the chest. The distribution of PyCaV in different parts of P. candelaria was further explored, which showed that more PyCaV were detected in its piercing-sucking mouthparts and chest viscera. Statistical analysis showed that the PyCaV infection was affected by time and locations.

This study describes the development of an image-based insect trap diverging from the plug-in camera insect trap paradigm. In short, a) it does not require manual annotation of images to learn how to count targeted pests and, b) it self-disposes the captured insects, and therefore is suitable for long-term deployment. The device consists of an imaging sensor integrated with Raspberry Pi microcontroller units with embedded deep learning algorithms that count agricultural pests inside a pheromone-based funnel trap. The device also receives commands from the server which configures its operation while an embedded servomotor can automatically rotate the detached bottom of the bucket to dispose of hydrated insects as they begin to pile up. Therefore, it completely overcomes a major limitation of camera-based insect traps: the inevitable overlap and occlusion caused by the decay and layering of insects during long-term operation, thus extending the autonomous operational capability. We study cases that are underrepresented in literature such as counting in situations of congestion and significant debris using crowd counting algorithms encountered in human surveillance. Finally, we perform comparative analysis of the results from different deep-learning approaches (YOLO7/8, crowd counting, deep learning regression) and we open-source the code and a large database of Lepidopteran plant pests.

Polyurethane (PU) is a polymer widely used by humans whose recycling is highly complex due his chemical structure, being limited to incineration or accumulation in landfills. Biodegradation by enzymes and microorganisms has been studied for decades as an effective method of biological decomposition. In this study, Tenebrio molitor larvae (T. molitor) were fed with polyurethane foams, which gut enzymes and microorganisms were capable of degrading the polymer by 35% in 17 days of treatment, producing a weight loss of 14% in the mealworm. Changes in T. molitor gut bacterial community and diversity were observed, which may be due to colonization of species associated with PU degradation. Physical and structural biodegradation in PU by T. molitor compared to virgin PU, was demonstrated by Fourier Transform InfraRed spectroscopy (FTIR), Thermal Gravimetric Analysis (TGA) and Scanning Electron Microphotography (SEM).

The coffee leaf miner Leucoptera coffeella (Guérin-Mèneville & Perrottet), is a key pest in coffee producing countries. During their development, the larvae feed on the palisade parenchyma of the leaves forming mines. As a result, the photosynthetic area of the plant decreases, affecting coffee production. Despite the severity of the damage caused by coffee leaf miner (CLM), morphological aspects of the larval development and the adult genitalia remain unknown. It is important to identify more susceptible targets to an efficient and narrow control by natural or synthetic approaches, relies on determining the larval instars. Equally important is the sexing of the adult, in experiments aiming efficient ways to control CLM, as the study of pheromone-based control methods. This work presents the first morphological description of the four larval instars and the adult genitalia of L. coffeella. In each larval instar, we measured the Mean ± SD (mm) of the cephalic capsules (1^st 0,14±0,03; 2^nd 0,25±0,04; 3^rd 0,32±0,03; 4^th 0,42±0,03) and observed the following morphological details: primary setae, prolegs, crochets and ecdysial line of the cephalic capsule. In the adults, we observed the sexual structures present in both genitalia: male - bulbus ejaculatorius, valva, anellus, gnathos and aedeagus and female - ovipositor, sclerite and corpus bursae. The dissection of the adult specimens confirmed that the external morphology corresponds to the correct sex attribution in CLM adults. These results may support innovative and improved control strategies for CLM Integrated Pest Management (IPM).

We introduce a device for automatic detection and reporting of crawling insects in urban environments. It is a monitoring device for urban pests that complies with the context of smart homes, smart cities and is compatible with the emerging discipline of the Internet of Things (IoT). We believe it can find its place to every room of a hotel, hospital, military camp and residence. This box-shaped device attracts targeted insect pests, senses the entering insect and takes automatically a picture of the internal space of the box. The picture is communicated through the Wi-Fi commonly found in such establishments to an authorized person/stakeholder receiving the picture to take proper action. The e-trap includes strong attractants (pheromone and/or food) to increase capture efficiency. The insect is trapped on the sticky floor of the device. The device carries the necessary optoelectronic sensors to guard all entrances of the trap. As the insect enters it interrupts the infrared light source. This triggers a detection event; a picture is taken, and a time-stamp is set before reporting the event through the Wi-Fi. The device can be integrated seamlessly in urban environments and operates unobtrusively to human activities. We report results on various insect pests and depending on the insect species, can reach a detection accuracy ranging from 96-99%.

Adaptive radiations occur mostly in response to environmental variation through the evolution of key eco-morphological innovations that allow emerging species to occupy new ecological niches. However, rapid phenotypic evolution and the evolution of key novelties are likely to also occur when a couple or few species are engaged into narrow ecological interactions. To demonstrate coevolution is a difficult task; only elusive evidences confirm that coevolution is a driver of speciation and diversification. Here we propose that the adaptive radiation of the Mediterranean orchid genus Ophrys, which gave rise to ca. 350 species since the apparition of the genus is due to the particular co-evolutionary dynamics between these plants and their pollinators. We suggest that the pollination by sexual swindle used by Ophrys orchids is the main driver of this coevolution. Flowers of each Ophrys species mimic sexually receptive females of one particular insect species, mainly bees. Male bees are attracted by pseudo-pheromones emitted by Ophrys flowers that are similar to the sexual pheromones of their females. Males lured by the flower shape, color and hairiness attempt to copulate with the flower, which glues pollen on their bodies. Pollen is eventually transferred to the stigma of another flower of the same Ophrys species during similar copulation attempts. Three observations led us to propose the scenario of an asymmetric co-evolutionary relationship between Ophrys and their pollinators. Firstly, there is a strong intra-specific competition among Ophrys individuals for the attraction of their species-specific pollinators, which is due to the high learning and memorization abilities of bees that record the pheromone signatures of kin or of previously courted partner to avoid (further) copulation attempts. Mnemonic pollinators induce thus a strong selective pressure for variation in the pseudo-pheromones emitted by individual flowers, which will potentially generate shifts in pollinator species, and hence Ophrys speciation. These pollinator shifts are adaptive for new Ophrys species because they may benefit from a competitor-free space. Secondly, such shifts in pollinator species are due to the random crossing of peaks in the olfactory landscape of the pollinator guild that is syntopic to each particular Ophrys population. This selective process on individual, random variation in pseudo-pheromone bouquets is followed by directional selection on flower phenotypes that will reinforce the attraction of the new pollinator. Thirdly, pollinators use the pseudo-pheromones emitted by Ophrys to locate suitable habitats from a distance within complex landscapes. Pollinators stay fixed for a while in these habitats by the local diversity of pseudo-pheromones, which increases their probability of encounter with a receptive female and hence the reproduction probability of both sexes. Conversely, pollinators disperse out of small suitable habitats once they have memorized the local diversity of sexual pseudo-pheromone bouquet or if fecundated Ophrys flowers repel pollinators, which decreases the probability of geitonogamy (plant advantage) but limit pollinator mating with locally emergent insect females, thus limiting inbreeding and favoring gene flow (pollinator advantage). Finally, we propose several research avenues that emerged according to this scenario of adaptive radiation by assymetric coevolution between Ophrys species and their pollinators.

Mosquitoes are a family of the order Diptera, which includes more than 3500 species. Two groups able to suck human blood and transmit diseases: the anophelines and the culicines comprise several genera such as Culex, Aedes and Aedes communis (Ac). Previous study has demonstrated a significant relationship between Ac sensitization and either extract or single bee venom components suggesting a “bee-mosquito syndrome” occurrence. It is well known that CCD can interference with the measurement of quantitative IgE results. Hence, it has been hypnotized that the presence of sIgE to CCD could have a role in the Bee-Mosquito syndrome. Method Serum from 21 different Mosquito and Ape melifera allergic individuals were combined with extracts of Mosquito (in house) and Apis melifera. SDS-PAGE and Immunoblot (IB) were carried out without and with with CCD-Inhibitor. Results The data would suggest IgE binding to common bands between Culex male, Culex female and Aedes species mosquitos of approximate molecular weight ~21kDa, ~35kDa, ~40kDa and ~55kDa. Whilst IgE binding has been observed to protein bands from the Apis melifera extract, these bands do not align with any present in the mosquito extracts. The use of a CCD-Inhibitor results in differences in IgE binding. Specifically, bands that were previously observed in the Apis melifera venom sample without use of CCD-Inhibitor at 40kDa and 90kDa are no longer observed when CCD-Inhibitor is used. Two bands of the Apis melifera venom remain present between 15-20kDa. No major differences in banding pattern were observed for the mosquito samples with or without CCD-Inhibitor, although the blot appears cleaner with less background when CCD-Inhibitor is used. Conclusion Further experiments have to be done to determine whether the reactive bands from Apis melifera are unrelated proteins or whether the proteins are related homologues of varying molecular weight.

Toxicological studies have shown that the cockroach Periplaneta americana is a classical model for studying the mode of action of commonly used insecticides. In a previous study, we demonstrated that thiamethoxam and clothianidin decreased locomotor activity and decreased the locomotor activity in an open-field-like apparatus. Here, we tested the effect of the neonicotinoid acetamiprid when applied orally, topically, or injected into the haemolymph. We found that acetamiprid decreased locomotor activity in the open-field-like apparatus. When treated with acetamiprid, a strong alteration in locomotor activity was observed 1 h, 24 h and 48 h after haemolymph and topical applications. Oral application of acetamiprid demonstrated that it impaired locomotor activity at 24 h and 48 h. A comparison of the present data with our previously published results showed that neonicotinoids were more active when injected into the haemolymph compared to oral and topical applications. These findings increased our understanding of the effect of neonicotinoid insecticides on insect locomotor activity, and demonstrated that the cyano-substituted neonicotinoid, acetamiprid, was able to alter cockroach locomotor activity.

Glaphyrophlebia victoriensis sp. nov. (Paoliida: Blattinopsidae) is the third Gzhelian representative of the genus and is described based on a beautiful forewing from the Var department in Southern France. Together with the description of another forewing fragment of a Glaphyrophlebia sp. from the Province of León in NW Spain, they improve our knowledge of fossil insects from French and Spanish late Carboniferous deposits. The specimen of Glaphyrophlebia sp. is the first mention of the family in the Carboniferous of Spain and extends the geographical distribution of the genus. These descriptions suggest that the genus Glaphyrophlebia was speciose during the Upper Pennsylvanian, while otherwise, very diverse in the early and middle Permian strata of the Russian Federation. We proposed the first hypothesis to explain the diversification of family and of its most speciose genera, and argue their diversity dynamics were likely linked with the major environmental changes that followed the collapse of the Carboniferous rainforest notably the extension of arid biomes during the Permian period. The exquisite preservation and the fineness of the sediment from Tante Victoire, in which the new species was found, suggests that the locality is suitable for preserving other fossil insects and will require additional investigations.

This paper introduces both a hardware and a software system designed to allow low-cost electronic monitoring of social insects using RFID tags. Data formats for individual insect identification and their associated experiment are proposed to facilitate data sharing from experiments conducted with this system. The antenna's configuration and their duty cycle ensure a high degree of detection rates. Other advantages and limitations of this system are discussed in detail in the paper.

In this paper, the methodology using Insect Intelligent Building (I^2B) technology for establishing energy consumption monitoring system of public buildings is prevailed. The computing process node and distributed algorithm are utilized to implement the energy consumption collection and data transmission and data pre-processing. Taking a commercial building as a case study, CPNs are applied to set up the building energy consumption monitoring system, with the Spanning Tree Algorithm for generating network topology，and BPNN method for solving abnormal data and recovering missing data. The research results demonstrate the proposed method can effectively improve the performance of plug-and-play and self-identified and self-configuration of energy consumption monitoring system.

Black Knot (BK) is a deadly disease of European (Prunus domestics) and Japanese (Prunus salicina) plums caused by the hemibiotrophic fungus Apiosporina morbosa. After infection, the appearance of warty black knots indicates a phytohormonal imbalance in infected tissues. Based on this hypothesis, we quantified phytohormones such as indole-3-acetic acid, tryptophan, indoleamines (N-acetylserotonin, serotonin, and melatonin), and cytokinins (zeatin, 6-benzyladenine, and 2-isopentenyladenine) in temporally collected tissues of susceptible and resistant genotypes belonging to European and Japanese plums during of BK progression. The results suggested auxin-cytokinins interplay driven by A. morbosa appears to be vital in disease progression by hampering the plant defense system. Taken together, our results indicate the possibility of using the phytohormone profile as a biomarker for BK resistance in plums.

Understanding the contribution of protected areas to pollinating insect conservation is crucial, especially in Haiti because of intense deforestation and environmental degradation. We compared pollinating insect assemblage composition and abundance in two protected areas: the suburban Wynne Farm Ecological Reserve (WFER) and the Urban National Park of Martissant (UNPM). Both sites encompass forest patches, agroforestry, and agricultural crops. Using colored pan-traps, we collected 3722 insects from six Orders, with 68.2% identified to family level due to taxonomic limitations. WFER exhibited significantly higher insect abundance than UNPM, though habitat differences varied between sites. Site, habitat, and their interaction significantly influenced insect composition. Among 118 identified families, only 30.51% were shared between sites. Our data suggest combined protection offers broader and more even insect diversity. We discovered patrimonial lepidopteran and hymenopteran species previously unrecorded in Haiti, emphasizing the importance of protected areas and the need for enhanced entomological expertise.

This study addresses the urgent need for sustainable protein sources in aquaculture due to the depletion of marine resources and rising costs. Animal protein sources, particularly poultry by-product meal (PBM) and insect exuviae meal, are investigated as viable alternatives to fishmeal (FM). Research confirms the successful replacement of FM with a combination of PBM and insect exuviae meal (up to 50%) in the diet of European seabass without compromising growth, feed conversion, gut health and liver fat content. In particular, growth performance was robust with both PBM formulations, with the 25% PBM diet showing better results. Histological examinations showed good gut and liver health, contradicting the concerns of previous studies. The study emphasizes the importance of holistic analyzes that go beyond growth parameters and include histomorphological investigations. The results show that PBM in combination with insect/exuviae meal is well tolerated by seabass, which is consistent with literature reports of mitigating negative effects on gut health. A detailed analysis of the microbiota revealed a decrease in the Firmicutes/Proteobacteria ratio due to an increase in potentially pathogenic bacteria. However, the formulation containing insect exuviae partially counteracted this effect by preserving the beneficial Lactobacillus and promoting the synthesis of short-chain fatty acids (SCFA), particularly butyrate. Chitin-rich components from insect exuviae were associated with improved gut health, which was supported by increased production of SCFA, which are known for their anti-inflammatory properties. The study concludes that a combination of PBM and insect/exuviae meal can replace up to 50% of FM in the diet of seabass, supporting sustainable aquaculture practices. Despite some changes in the microbiota, the negative effects are mitigated by the addition of insect exuviae, highlighting their potential as a prebiotic to increase fish productivity and contribute to a circular economy in aquaculture.

The fall armyworm, Spodoptera frugiperda, arrived during the 2016/2017 cropping season in Limpopo, South Africa and it continues to cause devastating damage on maize in this region. The present study investigated knowledge, perception and management of S. frugiperda by smallholder farmers at Dzindi and Tshiombo irrigation schemes in this region. The study was conducted in 16 focus group discussion interviews, 12 at Dzindi and four at Tshiombo irrigation scheme. Although farmers failed to identify S. frugiperda eggs and pupal stages, they positively identified the pest by its larval stage and refer to it by several local names. All farmers relied extensively on insecticides for S. frugiperda control. With the exception of the Dzindi irrigation scheme farmers, who have adopted Bt maize, very few farmers have adopted alternative methods to chemical control such as mechanical control, physical control (crushing egg and handpicking of larvae), biological control, cultural practices (such as Push-Pull Technology), possible indigenous knowledge, or the use of monitoring tools, for the management of S. frugiperda. The study provided valuable information for designing sustainable management practices to control S. frugiperda on maize in smallholder farming communities.

Gall-inducing insects often contain high concentrations of phytohormones, such as auxin and cytokinin, which are suggested to be involved in gall induction, but no conclusive evidence has been obtained. There are two possible approaches to investigating the importance of phytohormones in gall induction: demonstrating either that high phytohormone productivity can induce gall-inducing ability in non-gall-inducing insects, or that gall-inducing ability is inhibited when phytohormone productivity in galling insects is suppressed. In this study, we show that the overexpression of PonAAS2, which encodes an aromatic aldehyde synthase (AAS) responsible for the rate-limiting step in indoleacetic acid (IAA) biosynthesis in a galling sawfly (Pontania sp.) that contains high levels of endogenous IAA, conferred high IAA productivity on Caenorhabditis elegans, as the model system. This result strongly suggests that PonAAS2 can also confer high IAA productivity on low-IAA-producing insects. We also successfully identified an inhibitor of PonAAS2 in a chemical library. This highly selective inhibitor showed stronger inhibitory activity against AAS than against aromatic amino acid decarboxylase, which belongs to the same superfamily as AAS. We also confirm that this inhibitor clearly inhibited IAA productivity in the high-IAA-producing C. elegans engineered here.

Global crop yields are estimated to be reduced by 30–40% per year on account of plant pests and pathogens. Agricultural insect pests raise concerns about constraining global food security and climate changes contributing to the rise of infestation. The current management relies on plant breeding, associated or not with transgenes and chemical pesticides. Both approaches face serious technology obsolescence on the field due to resistance breakdown or development of insecticide resistance. The need for new Modes of Action (MoA) approaches in managing crop health grows each year, driven by market demands to reduce economic losses and phytosanitary requirements to meet the consumer perception. Disabling pest genes by sequence-specific expression silencing is considered a promising tool in the development of environment and health respectful biopesticides. The specificity conferred by long dsRNA-base solutions give support to minimizing effects on off-targeted genes in the insect pest genome and the target gene in non-target organisms (NTOs). In this review, we summarize the current status of gene silencing by RNA interference (RNAi) for agricultural control. More specifically, we focus on the engineering, development and application of gene silencing to control Lepidoptera by the employment of non-transforming dsRNA technologies. Despite some delivery and stability drawbacks of topical applications, we reviewed works showing convincing proof-of-concept results that point to imminent innovative solutions. Considerations about the regulamentation of the ongoing research on dsRNA-based pesticides to produce commercialized products for exogenous application are discussed. Academic and industry initiatives reveal a worthy effort to accomplish controlling Lepidoptera pests with this new mode of action to provide more sustainable and reliable technologies to field management. New data on genomics of this taxon encourage the increment of a customized target genes portfolio. As a case of study, we illustrate how dsRNA and associated methodologies could be applied to control an important Lepidopteran coffee pest.

Background, Leucoptera coffeella (Guerin-Meneville, 1842) is a moth species (Lyonetiidae, Lepidoptera) pest that causes severe losses to coffee crops. Further information about its genomic data is required to allow molecular strategies for the development of sustainable pesticides and to gain in-depth knowledge on phylogenetics. However, the closest complete genome available is within the superfamily level (Yponomeutoidea). Here we report the generation of the first long-read genome, transcriptome and proteome results of L. coffeella and the in silico analysis performed in these molecular levels to investigate genes involved in the siRNA processing. Results, PACBio and paired-end Illumina combined DNA sequencing from pupae samples resulted in more than 436 Gb subreads and 31Mb reads with N50 read length of 15,512 nt, mean read length 13.8 Kb and max read length 420.7 Kb. Additionally, 20Gb data of short DNA sequencing was combined to produce 1,984 contigs comprising 397 Mb in total. The longest and shortest scaffold sizes are 10,809,567 nt and 15,247 nt, respectively (mean size 200,178 nt). The N50 scaffold was 275,598 nt and the GC content was 36.10%. Predicted coding DNA sequences counted 39.930 gene models. Searching of 5286 BUSCO groups revealed 91.7 percent of completeness (single and duplicated genes combined) compared to lepidoptera genomes (lepidoptera_odb10). Flow cytometry showed the 1C DNA content is approximately 295 Mb. RNA-Seq from seven development stages resulted in 28294 identified transcripts. Additionally, proteomics from immature stages resulted in 2045 proteins matching the gene models. Conclusions, This first nuclear genome of the Lyonetiidae family brings valuable molecular resources to study Lepidoptera genomes. Genome, transcriptome and proteome sequencing to raise genome annotation precision may resolve uncovered taxonomic issues. In addition, these combined approaches provide insights into plant-insect interaction players, as horizontally transferred genes (HGT) and endosymbionts. Put together, the generated data enables the development of molecular tools towards sustainable biotechnology solutions for lepidopteran pest control.

In cotton cultivars, an insect that causes irreversible damage is the Spodoptera frugiperda, known as the fall armyworm. Since the visual detection of plants is a burdensome task for human inspection, the spectral information related to plant damage, registered on a spectral scale, can be useful. These measurements, associated with machine learning techniques, produce useful information for a rapid and non-invasive inspection method development. To contribute to this gap fulfillment, this paper proposes a machine learning framework to model the spectral response of cotton plants under the attack of the fall armyworm. Additionally, a theoretical model is presented, built from the results of the machine learning analysis, to infer this damage with up-to-date orbital sensors. The data was composed of the reflectance measurements collected at a cotton field with control plants and plants submitted to Spodoptera frugiperda damage. Their spectral response was recorded with a hand-held spectroradiometer ranging from 350 to 2,500 nm, for eight consecutive days. Different machine learning models were evaluated and the overall best model was defined by accuracies comparisons on a testing-set. A ranking approach was adopted based on the model accuracy, returning the most contributive wavelengths for the classification. Sequentially, an unsupervised neural network (Self-Organizing Map - SOM) was implemented to reduce data-dimensionality and assist in the definition of important spectral regions. The regions were associated with the spectral bands of the two sensors (OLI and MSI) and a theoretical model using a band simulation process with the overall best machine learning model was proposed. The results indicated that the Random Forest (RF) algorithm is the most suitable to predict cotton-plants damaged by insects and that the last day of analysis (8th day) was better to separate it, with F-measure equals 0.912. The ranking approach combined with the SOM method indicated the spectral regions at the red to near-infrared (650 to 1,350 nm) and shortwave infrared (1,570 to 1,640 nm) as the most important regions to the analysis. The proposed theoretical model simulated with the OLI and MSI sensor-bands returned an F-Measure of 0.865 and 0.886, respectively. In conclusion, this framework can be used to map cotton-plants under insect-attack. The theoretical model presents high accuracy to infer the insect-damaged on cotton plants based on multispectral bands from other sensors, being a useful tool for future research that intends to evaluate it in other areas and at different field scales.

The lipid composition of biomembranes influence the properties of the lipid bilayer as well as that of the proteins. In this study, the lipidome and the lipid/protein ratio of membranes from High FiveTM insect cells overexpressing mouse P-glycoprotein was characterized. This provides a better understanding of the lipid environment in which P-glycoprotein is embedded, and thus of its functional and structural properties. The relative abundance of the distinct phospholipid classes and their acyl chain composition was characterized. A mass ratio of 0.57 +/- 0.11 phospholipids to protein was obtained. Phosphatidylethanolamines are the most abundant phospholipids, followed by phosphatidylcholines. Membranes are also enriched in negatively charged lipids (phosphatidylserines, phosphatidylinositols and phosphatidylglycerols), and contain small amounts of sphingomyelins, ceramides and monoglycosilatedceramides. The most abundant acyl chains are monounsaturated, with significant amounts of saturated chains. The characterization of the phospholipids by HPLC-MS allowed identification of the combination of acyl chains, with palmitoyl-oleoyl being the most representative for all major phospholipid classes except for phosphatidylserines, which are mostly saturated. A mixture of POPE:POPC:POPS in the ratio 45:35:20 is proposed for the preparation of simple representative model membranes. The adequacy of the model membranes was further evaluated by characterizing their surface potential and fluidity.

Targeted enrichment of ultraconserved elements (UCE) has emerged as a promising tool for inferring evolutionary history in many taxa, with utility ranging from phylogenetic and phylogeographic questions at deep time scales to population level studies at shallow time scales. However, the methodology can be daunting for beginners. Our goal is to introduce UCE phylogenomics to a wider audience by summarizing recent advances in arthropod research, and to familiarize readers with background theory and steps involved. We define terminology used in association with the UCE approach, evaluate current laboratory and bioinformatic methods and limitations, and, finally, provide a roadmap of steps in the UCE pipeline to assist phylogeneticists in making informed decisions as they employ this powerful tool. By facilitating increased adoption of UCE in phylogenomics studies that deepen our comprehension of the function of these markers across widely divergent taxa, we aim to ultimately improve understanding of the arthropod tree of life.

Pain is a natural bioassay for detecting and quantifying biological activities of venoms. The painfulness of stings delivered by ants, wasps, and bees can be easily measured in the field or lab using the stinging insect pain scale that rates the pain intensity from 1 to 4, with 1 being minor pain, and 4 being extreme, debilitating, excruciating pain. The painfulness of stings of 96 species of stinging insects and the lethalities of the venoms of 90 species was determined and utilized for pinpointing future promising directions for investigating venoms having pharmaceutically active principles that could benefit humanity. The findings suggest several under- or unexplored insect venoms worthy of future investigations, including: those that have exceedingly painful venoms, yet with extremely low lethality – tarantula hawk wasps (Pepsis) and velvet ants (Mutillidae); those that have extremely lethal venoms, yet induce very little pain – the ants, Daceton and Tetraponera; and those that have venomous stings and are both painful and lethal – the ants Pogonomyrmex, Paraponera, Myrmecia, Neoponera, and the social wasps Synoeca, Agelaia, and Brachygastra. Taken together, and separately, sting pain and venom lethality point to promising directions for mining of pharmaceutically active components derived from insect venoms.

Τhe concept of remote insect surveillance at large spatial scales for a number of serious insect pests of agricultural and medical importance is introduced in a series of our papers. We augment typical, low-cost plastic traps for many insect pests with the necessary optoelectronic sensors to guard the entrance of the trap in order to detect, time-stamp, GPS tag, and –in relevant cases- identify the species of the incoming insect from their wingbeat. For every important crop pest there are monitoring protocols to be followed in order to decide when to initiate a treatment procedure before a serious infestation occurs. Monitoring protocols are mainly based on specifically designed insect traps. Traditional insect monitoring suffers in that the scope of such monitoring: is curtailed by its cost, requires intensive labor, is time consuming, an expert is often needed for sufficient accuracy and can sometimes raise safety issues for humans. These disadvantages reduce the extent to which manual insect monitoring is applied and therefore its accuracy, which finally results in significant crop loss due to damage caused by pests. With the term ‘surveillance’ we intend to push the monitoring idea to unprecedented levels of information extraction regarding the presence, time-stamping detection events, species identification and population density of targeted insect pests. Insect counts as well as environmental parameters that correlate with insect’s population development are wirelessly transmitted to the central monitoring agency in real time, are visualized and streamed to statistical methods to assist enforcement of security control to insect pests. In this work we emphasize on how the traps can be self-organized in networks that collectively report data at local, regional, country, continental, and global scales using the emerging technology of the Internet of Things (IoT). This research is necessarily interdisciplinary and falls at the intersection of entomology, optoelectronic engineering, data-science and crop science and encompasses the design and implementation of low-cost, low-power technology to help reduce the extent of quantitative and qualitative crop losses by many the most significant agricultural pests. We argue that smart traps communicating through IoT to report in real-time the level of the pest population from the field straight to a human controlled agency can, in the very near future, have a profound impact on the decision making process in crop protection and will be disruptive of existing manual practices. In the present study, three cases are investigated : monitoring Rhynchophorus ferrugineus (Olivier) (Coleoptera: Curculionidae) using a) Picusan and b) Lindgren trap, and c) monitoring various stored grain beetle pests using the pitfall trap.

Finding a suitable support is a key process in the life history of climbing plants. Climbers that find a suitable support have greater performance and fitness than those that remain prostrate. Numerous studies on climbing plant behavior have elucidated the mechanistic details of support searching and attachment. Far fewer studies have addressed the ecological significance of support-searching behavior and the factors that affect it. Among these, the diameter of supports influences their suitability for twining plants. When support diameter increases beyond some point climbing plants are unable to maintain tensional forces and therefore lose attachment to the trellis. Here we further investigate this issue by putting pea plants in the situation to choose between supports of different diameters while their movement was recorded by means of a three-dimensional motion analysis system. The results indicate that the way climbing plants move can vary depending on whether they are presented with one or two potential supports. Furthermore, when presented with a choice between a thin and a thick support, the plants showed a distinct preference for the former than the latter. The present findings shed further light on how climbers make decisions as far as support search is concerned, and provide evidence that plants adopt one of several alternative plastic responses in a way that optimally corresponds to environmental scenarios.

AMF (Arbuscular Mychorhizal Fungi) are very well known due to their importance in promoting growth and developments of plants especially vegetables. These fungi can be grown easily, stored and multiplied with simple means, also the application of these fungi is generally on the layer of the soil or near the roots in the inner layers of the soils. The growth of the amf fungi is very easy and they are highly adjustable to any soil and environmental conditions. In this review our main focus is on the use of amf for production of vegetables and also the effect of amf against insects and pests. The amf is known to reduce several symptoms caused by different insect pests and also plant diseases thereby promoting healthy growth of the plants. Also use of this amf will increase the uptake of nutrient from the soils through symbiotic relationships between plants and fungi. The uptake of important minerals which are drawn from deeper layers of soils is observed with the help pf amf. This study reveals the benefits of the use of amf under severe disease and pest incidences thereby known as an alternate for harmful chemical pesticides and fungicides.

Antimicrobial Peptides (AMPs), as immune effectors synthesized by a variety of organisms, not only constitute a robust defense mechanism against a broad spectrum of pathogens in the host, but also show promising applications as effective antimicrobial agents. Notably, insects are significant reservoirs of natural AMPs. However, the complex array of variations in types, quantities, antimicrobial activities, production pathways of AMPs, and evolution of AMPs across insect species presents a significant challenge for immunity system understanding and AMP applications. This review covers insect AMP discoveries, classification, common properties, and mechanisms of action. Additionally, we summarize the types, quantities, and activities of im-mune-related AMPs in each model insect. We also conducted the first comprehensive investigation into the diversity, distribution, and evolution of 20 types of AMPs in model insects, employing phylogenetic analysis to describe their evolutionary relationships and shed light on conserved and distinctive AMP families. Furthermore, we summarize the regulatory pathways of AMP production through classical signaling pathways and additional pathways associated with Nitric Oxide, insulin-like signaling, and hormones. This review advances our understanding of AMP as guardians in insect immunity system and also unlocks a gateway to insect AMP resources, facilitating the use of AMPs to address food safety concerns.

The fungal order Entomophthorales in the Zoopagomycotina includes many fungal pathogens of arthropods. This review explores five genera in the Erynioideae subfamily, namely Erynia, Furia, Pandora, Strongwellsea, and Zoophthora. This is the largest subfamily in the Entomophthorales including 125 described species. The species diversity, global distribution, and host range of this subfamily are summarized, revealing that relatively few taxa are geographically widespread, and few have broad host ranges, contrasting with a large number of species with single reports, from one location and one host species. The insect orders infected by the greatest numbers of species are the Diptera and Hemiptera. Across the subfamily relatively few species have been cultivated in vitro and those that have require more specialized media than any fungi. Given their potential to attack arthropods and in the fungal evolutionary tree, which species might be adopted for biological control purposes or biotechnological innovations are highlighted. Current challenges in the implementation of these species in biotechnology include the limited ability or difficulty in culturing many in vitro, a correlated paucity in genomic resources, and considerations regarding the host ranges of different species.

Mosquitoes carrying endosymbiotic bacteria called Wolbachia are being released in mosquito and arbovirus control programs around the world. Open field releases of Wolbachia-infected male mosquitoes have achieved over 95% population suppression, while the replacement of populations with Wolbachia-infected females is self-sustaining and can greatly reduce local dengue transmission. Despite many successful interventions, significant questions and challenges lie ahead. Wolbachia, viruses and their mosquito hosts can evolve, leading to uncertainty around the long-term effectiveness of a given Wolbachia strain, while few ecological impacts of Wolbachia releases have been explored. Wolbachia strains are diverse and the choice of strain to release should be made carefully, taking environmental conditions and the release objective into account. Mosquito quality control, thoughtful community awareness programs and long-term monitoring of populations are essential for all types of Wolbachia intervention. Releases of Wolbachia-infected mosquitoes show great promise, but existing control measures remain an important way to reduce the burden of mosquito-borne disease.

A facile solid-phase synthetic method for incorporating the imidazoline ring motif, a surrogate for a trans peptide bond, into bioactive peptides is reported. The example described is the synthesis of an imidazoline peptidomimetic analog of an insect pyrokinin neuropeptide via a cyclization reaction of an iminium salt generated from the preceding amino acid and 2,4-diaminopropanoic acid (Dap).

Is the extensive decline of insects partially due to an insect-specific feature of their functional organization that is disadvantageous for living in an industrialized environment? The unique way in which gases are supplied in insect tissues is such a special trait. It exposes cells directly to the gas phase, via tracheae that end in micro-tubes, the tracheoles, which have diameters in the same range as particles of ultra-fine dust transported by air of industrialized countries. Number and volume of these particles – calculated to be inhaled by honeybees, e.g., – are indicative to locally impede O2-uptake and CO2-release and thus to restrain physiological activities.

We presented here the first draft genome sequence of the trypanosomatid Herpetomonas muscarum ingenoplastis. This parasite was isolated repeatedly in the black blowfly, Phormia regina. This is the first draft genome of a flagellate from the phylogenetically distinct clade of Trypanosomatidae.

Abstract: Aedes aegypti were exposed to water treated with mosquitocidal chips containing the insecticide pyriproxyfen in a polymer formulation. Chips were tested under different conditions; different water volumes, in containers made of different material, and in water with different levels of organic matter. Treated chips caused 100% mortality of Ae. aegypti during their pupal stage independent of conditions chips were exposed to in water. When tested for longevity, the chips containing 840 µg of pyriproxyfen killed 100% of Ae. aegypti for 4 sequential months of the chips being reused in water. Chips containing 8.4 µg of pyriproxyfen ceased to work after the first week of treatment. When mosquitocidal chips were used in > 25% of the oviposition containers within their cages, there was a significant control of the mosquito populations. Mosquitocidal chips worked in different environments, lasted for extended periods of time, caused significant mosquito population decreases, and were effective in controlling Ae. Aegypti.

Black Knot (BK) is a deadly disease of European (Prunus domestica) and Japanese (Prunus salicina) plums caused by the hemibiotrophic fungus Apiosporina morbosa. Generally, phytopathogens hamper the balance of primary defense phytohormones such as salicylic acid (SA)-jasmonic acid (JA) balance for disease progression. Thus, we quantified the important phytohormones titers in tissues of susceptible and resistant genotypes belonging to European and Japanese plums at five different time points. The results suggested auxin-cytokinins interplay driven by A. morbosa appeared to be vital in disease progression by hampering the plant defense system via perturbance in SA, JA, and gibberellic acid. The results further point out that SA and JA in plant defense are not necessarily antagonistic always, especially in woody perennials. Results also suggest that the changes in SA and JA content following BK disease infection and progression in plums could be utilized as phytohormonal markers in the identification of BK-resistant cultivars.

Common bean (Phaseolus vulgaris L.) is an important crop for food security and for national economics for several countries worlwide. One of the most important factors of risk in common bean production is the fungal disease anthracnose caused by Colletotrichum lindemuthianum, which in some cases, causes complete yield losses; this kind of plant disease is usually managed through the application of chemical products such as fungicides, that are commonly not accepted by society, this rejection is based on the relationship of pesticides with health damage and environmental contamination. In order to help in solving this drawbacks, the present work propose the use of Electrochemically Activated Salt Solutions(EASS) as a safer pathogen control agent in crops due to it have shown an elicitor and biostimulant effect on plants such as tobacco and apple. With this background, this manuscript presented in vitro results of the evaluation of the inhibitory effect for multiple bean pathogens and in vivo results of EASS in the common bean-Colletotrichum pathosystem by in vitro evaluations of the inhibitory effect for multiple bean pathogens and in vivo evaluation of the infection severity and defense activation such as secondary metabolites production and antioxidant activity. EASS presence in growth media had strong inhibitory effect, at beginning of experiment, for some of the evaluated fungi. EASS showed effect against the development of the disease when applied in specific doses to prevent distress in plants.

This paper explores if plants are capable of responding to human movement by changes in their electrical signals. Towards that goal we conducted a series of experiments, where humans over a period of 6 months were performing different types of eurythmic gestures in proximity of garden plants, namely salad, basil, and tomatoes. To measure plant perception, we used the plant spikerbox, a device that measures changes in voltage differentials of plants between roots and leaves. Using machine learning, we find that the voltage differentials over time of the plant predict if (a) eurythmy has been performed, and (b) which kind of eurythmy gestures has been performed. We also find that the signals are different based on the species of the plant. In other words, the perception of a salad, tomato, or basil might differ just as perception of different species of animals differ. This opens new ways of studying plant ecosystems, while also paving the way to use plants as biosensors for analyzing human movement.

Fire-related cues could alter vegetation community composition by promoting or excluding different types of plants. Smoke-derived compounds have been a hot topic in plant and crop physiology. There are some fire-prone areas in Australia, both Americas, some Mediterranean countries and regions with extensive prescribed or illegal burning like central European, which are subjected to a direct impact of fire (heat, minerals accumulation from the burning matter) and/or the indirect one (smoke) and undergo compositional and structural changes. This review first updates information about the effect of the compounds on plants' kingdoms and focuses on the research advances in the field of smoke compounds and attempts to gather and summarize the recent state of research and opinions on the role of smoke-derived compounds in plants' lives. We finish our review by discussing major research gaps, which include: Why plants respond to smoke chemicals? Is the response of seeds to smoke and smoke compounds an evolutionary-driven trait allowing plants to adapt to local environment?

Regulations and guidelines on water quality are used according to physicochemical but scarcely biological variables to protect ecosystems and control water use. This study innovates using in-sects as bioindicators to establish the health status of an aquatic ecosystem in the Latin American's lentic bodies. Water quality in urban lentic bodies in the Metropolitan Region, Chile, was evaluated from of aquatic insect assemblages and physicochemical variables for conserving aquatic life. Evaluations were carried out in parallel in four sampling stations in three water bodies, two seasons, and 2–3-year series, with three replicates. Aquatic insects were randomly sampled and identified by families; abundance, and richness differences were compared with non-parametric tests. Family Biotic Indices (FBI) were calculated. Physicochemical variables were measured using portable multiparametric and laboratory chemical analysis to determine water quality. A new physicochemical variable (total suspended solids) was established, which had a greater incidence with the results obtained for the FBI. Based on the nitrogen and phosphorus levels in the water bodies, all of them were eutrophics. Given the ecosystem diversity and complexity, studies should delve deeper into wetlands to establish methods that contribute to determining water quality, using insects as bioindicators and physicochemical variables.

The conservation status of monarch butterflies in North America is a topic of intense scrutiny and debate. It is clear that winter colonies in Mexico are declining, yet some recent studies suggest that summer breeding populations are relatively stable and similar to historical abundances. One possible explanation for these discordant patterns is that fall migration success has been recently disrupted. Here, we use a relatively unexplored citizen-scientist dataset on the size of monarch “roosts,” which are resting aggregations on vegetation, to infer changes in monarch abundance along the fall migration route over the last 17 years. We found that the timing of migration remained relatively unchanged while the flyway has generally become warmer and greener. Warmer and greener conditions were associated with larger roosts, yet we found steady, dramatic declines in roost sizes through time that were independent of climate and landscape factors. Roost sizes have declined as much as 80%, with losses increasing from north to south along the migration route. These findings suggest that failure during the fall migration could explain the apparent drop in monarch numbers from summer breeding to overwintering populations. This in turn suggests that conservation efforts that support fall migration success are most needed, such as limiting the planting of non-native milkweeds that enhance parasite loads and perhaps dampen migration success. Overall, it appears the fall migration of monarch butterflies is under imminent threat, even if the species’ overall survival is not.

Acetamiprid is a broad-spectrum neonicotinoid insecticide used in agriculture to control aphids. While recent studies have documented resistance to acetamiprid in several aphid species, the underlying mechanisms are still not fully understood. In this study, we analyzed the transcriptome and metatranscriptome of a laboratory strain of the pea aphid, Acyrthosiphon pisum (Harris, 1776), with reduced susceptibility to acetamiprid after nine generations of exposure to identify candidate genes and the microbiome involved in the adaptation process. Sequencing of the transcriptome of both selected (RS) and non-selected (SS) strains allowed the identification of 14858 genes and 4938 new transcripts. Most of the differentially expressed genes were associated with catalytic activities and metabolic pathways involving carbon and fatty acids. Specifically, the alcohol-forming fatty acyl-CoA reductase (FAR) and acyl-CoA synthetase (ACSF2), both involved in the synthesis of epidermal wax layer components, were significantly upregulated in RS, suggesting that adaptation to acetamiprid involves the synthesis of a thicker protective layer. Metatranscriptomic analysis revealed subtle shifts in the microbiome of RS. These results contribute to a deeper understanding of acetamiprid adaptation by the pea aphid and provide new insights for aphid control strategies.

Abstract: The search for alternative protein sources to soy-bean meal (SBM) in animal feeding is a strategic objective to reduce production costs and contribute to a sustainable animal produc-tion. Spirulina, due to the high protein content, has emerged as a potential cost-effective, sus-tainable, viable and high nutritional value food resource for many animal species. Insect larvae (Tenebrio molitor and Hermetia illucens) are also considered potential alternatives to SBM, given their high edible percentage of almost 100%, as well as a protein value higher than that of vegetable proteins. Rapeseed meal and grain legumes, such as faba beans, peas, lupins and chickpea can also be used as locally producible protein ingredients. This study reviews the nutritional value of these potential alternatives to SBM in pig diets, effects on animal perfor-mance, digestion, immune system, physicochemical and sensorial characteristics of meat, in-cluding processed meat products of pork. Limits on their use in pig feeding are also reviewed to indicate gaps to be filled in future research on the supplementation level of these potential al-ternative protein sources into pig diets.

Insect outbreaks are major drivers of natural disturbances in forest ecosystems. Outbreaks can have both direct and indirect effects on the composition of soil arthropod communities, through canopy opening, nutrient addition and predator-prey interactions. In this study, we aim to understand the effects of forest tent caterpillar (Malacosoma disstria; FTC) outbreaks on ant communities in both temperate and boreal forests in Canada. Using pitfall traps and Berlese funnels, we compared the ant communities as well as the surrounding arthropod communities between control and outbreak sites in boreal andboreal and temperate forests (in Québec, Canada). Using the Sørensen dissimilarity index, we determined the alpha and beta diversity of the ant community. Other arthropods collected in the traps were counted to evaluate the richness and abundance of potential prey for the ants and other potential predators of the FTC. We used an indicator species analysis to examine the species associated with sites defoliated by the outbreak. In the boreal forest, we found that FTC outbreaks caused decreases in species richness and increases in the evenness of ant communities in defoliated sites. In the boreal forest sites, species composition varied significantly between control and outbreak sites. This pattern was driven by the presence of other predators. We also saw no changes in beta diversity in the boreal forest but did see a significant decrease in the temperate forest between the outbreak sites and the control sites. A similar, but weaker pattern was observed in the temperate forest. Ant species in the boreal forest tended to exhibit a more marked preference for either control or previously defoliated sites than species in the temperate forest. Our study showed that disturbances like insect outbreaks can drive changes in the ant community. While we saw small effects of outbreaks, manipulation experiments using resource addition could help us validate the mechanisms behind these relationships.

Dengue virus infections are a serious public health problem worldwide. Aedes aegypti is the primary vector of dengue in Cuba. Since there is no vaccine or specific treatment, the control efforts are directed to reduce mosquito populations. The indiscriminate use of pesticides can lead to increase insecticide resistance as well as adverse effects on human health. The sterile insect technique is a species-specific and environmental friendly method of insect control based on the release of large numbers of sterile males. The success of this technique in sustainable control of agricultural pests has encouraged its evaluation for mosquito control. Here, we describe an open field trial to evaluate the effect of the release of irradiated males on a wild population of Aedes aegypti. The case-control study was performed in a suburb of Havana, and compared the mosquito population before and after the intervention, in both control and treated areas. The wild population was monitored by an ovitrap network, recording frequency and density of eggs as well as their hatch rate. A significant induced sterility was observed in the field population, compared to the control. The ovitrap index and the mean eggs/ trap declined dramatically after an expected lag period of twelve and five weeks, respectively. For the last three weeks, no egg was collected in the treated area, evidencing a significant suppression of the wild population. We conclude that the sterile males released competed successfully, and induced enough sterility to suppress the local Aedes aegypti population.

Xenorhabdus nematophila and Photorhabdus luminescens are entomopathogenic symbionts that produce several toxic proteins that can interfere with the immune system of insects. Here, we showed that outer membrane proteins (OMPs) could be involved as virulence factors during bacterial symbiont pathogenesis. Purified OMPs from bacterial culture were injected fifth instar larvae of Spodoptera exigua Hübner. Larvae were surveyed for fluctuations in total haemocyte counts (THC), granulocyte percentage (cellular defence), protease, phospholipase A2 (PLA₂), and phenoloxidase (PO) activities (humoral defence) at specific time intervals. Changes in the expression of the three antimicrobial peptides (AMPs), cecropin, attacin, and spodoptericin, were also measured. Larvae treated with both types of OMPs had more haemocytes than did the negative controls. OMPs of X. nematophila caused more haemocyte destruction than did the OMPs of P. luminescens. The OMPs of both bacterial species initially activated insect defensive enzymes post-injection, their activating fluctuated in different ways. Attacin, cecropin and spodoptericin were up-regulated by OMP injections more than in normal larvae. The expression of these three AMPs was maximal at four hpi with P. luminescens OMPs treatment. Expression of the three AMPs in X. nematophila treatment was irregular and lower than in the P. luminescens OMPs treatment. These findings provide insights into the role of OMPs of entomopathogenic nematode bacterial symbionts in countering the physiological defenses of insects.

Suppressors of cytokine signaling (SOCS) influence cytokine and growth factor signaling by negatively regulating the JAK-STAT pathway. This maintains homeostasis during host immune response. However, functional characterization of SOCS family members in invertebrates is limited. In this study, we discovered the Type-I subfamily of the SOCS genes in the mealworm beetle, T. molitor. The full-length ORFs of TmSOCS5, TmSOCS6, and TmSOCS7 consisted of 1,389, 897 and 1,458 nucleotides, encoding polypeptides of 462, 297 and 485 amino acids, respectively, The C-terminal region of TmSOCS was highly conserved in the SH2 and SOCS box domains. Phylogenetic analysis revealed that the three SOCS genes clustered within the same sub-family and the highest amino acid identity was with the Tribolium castaneum SOCS genes (TcSOCS). While the expression of TmSOCS5 and TmSOCS6 was low in larval, pupal, and adult stages of the insect, TmSOCS7 showed higher expression. The expression of TmSOCS5 and TmSOCS6 was higher in larval hemocytes and adult ovary. The microbes expressed the three TmSOCS genes to varying degrees. C. albicans elicited the strongest response in the host with highest 15-fold expression in TmSOCS7 3 h post-inoculation. Collectively, these data suggest that the Type I TmSOCS could play a role in eliciting host immunity.

Ash trees have considerable economic, cultural and environmental value on the island of Ireland. However, European ash (Fraxinus excelsior L.) is currently under threat from the invasive ascomycete pathogen Hymenoscyphus fraxineus. This pathogen is the causal agent of ash dieback disease, which was initially reported in Poland in 1992. Hymenoscyphus fraxineus has since spread across Europe and the first recorded case of the disease on the island of Ireland was in 2012 at a forestry plantation in Co. Leitrim. The pathogen is now present in all 26 counties in Ireland and 6 counties in Northern Ireland, and it is considered unfeasible to eradicate. The spread of ash dieback disease is reflected in recent policy changes, which focus on management rather than eradication strategies. Since the first formal description of H. fraxineus in 2006, considerable research efforts have been made by the international scientific community to understand the biology of the pathogen and to develop management strategies against it. This review provides an update of current knowledge of H. fraxineus biology and infection. We then explore examples of mitigation techniques that have been trialled in Europe, in order to identify strategies that are feasible for disease management at a local level on the island of Ireland. Finally, we outline five key avenues of research that have the potential to provide breakthroughs in methods to protect valuable F. excelsior resources.

Understanding how plants change their root foraging strategy in the presence of neighbors is of paramount importance for plant ecology and agriculture. The root tragedy of the common (RToC) is a plant behavior predicted by game theory models in which competing plants forage for soil resources inefficiently. The RToC is generally assumed to be induced by non-self root recognition, and researchers consider root overproliferation and reduced fitness with respect to a plant growing solo as the trace left by plants engaging in an RToC in experiments and model results. Herein, I first challenge both notions, and argue that the RToC is a suboptimal phenotypically plastic response of plants that is based in soil resource information exclusively. Second, I discuss how this new perspective carries important implications for the design of experiments investigating the physiological mechanisms underlying observable plant root responses. Finally, I discuss why placing the RToC theory in the context of more general root research is fundamental: The RToC represents a mechanistic foundation for understanding the belowground behavior of plants interacting with neighbors, and a spatially explicit approach to RToC may produce more comprehensive results.

Melatonin, a naturally occurring chemical, has pleiotropic effects across various species. It plays a vital role in animals by regulating the body clock, while in plants, it is known to resist aging. Melatonin is associated with various plant physiological processes, including stress response, plant growth, and flowering time. It has prospects in enhancing and promoting plant growth and development, as well as in strengthening crop yield and increasing sustainability in agriculture. This review summarized the potential roles of melatonin in several plant species, as well as its biosynthetic modes. In addition, we described the recently uncovered melatonin features in plants, summarized the issues and challenges facing plant melatonin research and some feasible solution ideas are proposed.

A substantial amount of phosphorus (P) in soil is not readily available for plant uptake. Certain species may enhance P availability from poorly soluble P forms. This research aimed to compare the P solubilization capacity of the soil of two Lupine species, L. albus, and L. angustifolius, with a control crop of oats (Avena strigosa L.). Plants were planted in pots in four soils with varying texture, organic matter content, acidity, and P availability. Phosphorus solubilization capacity was assessed on variations of P availability (PBray1) at four points in time, comparing soils with lupine to oat-containing soils and their baseline values. At harvest, measurements of plant shoot biomass included their shoot dry weight and some nutrient concentrations such as P, nitrogen (N), manganese (Mn), and lupine nitrogen fixation. Compared to soils containing oats, lupine soils significantly increased PBray1 concentrations; increments ranged from 0.9 to 5.4 mg kg-1, depending on species, sampling time, and soil type. Lupine-induced soil acidification did not fully explain this P increase., As a proxy measurement of organic acid exudation, Manganese concentration was high in both lupine species' shoot biomass; however, these legumes had divergent Mn accumulation responses to soil acidity and P availability. Nitrogen fixed quantity varied with Lupine species (105-240 mg pot-1) and soil type (60-240 mg pot-1). Lupinus albus had a higher ability to mobilize non-labile P and provide N through biological N fixation (BNF), positioning it as a suitable crop for diversifying Uruguay's agricultural crop rotation systems.

The purpose of this investigation was to study the impact of different rates of bio fertilizer on agronomic characteristics and the relations among herbaceous plant diversity and soil properties of Aloysia citrodora in central Greece. For this aim a two – year Completely Randomized Design (CRD) field experiment was carried out in central Greece. The experiments included four biological fertilizer levels, BF0: 0 kg ha-1, BF50: 50 kg ha-1, BF100: 100 kg ha-1 and BF150: 150 kg ha-1 with 3 replication. Plant height, dry stem, dry leaves, dry total yield and Leaf Area Ιndex (LAI) were measured during the two growing years. The results showed that in the 1st studied the maximum plant height was observed in BF150 treatment while the 2nd year the BF150 level provoked a raise of 16.08% and 1.09% compared to BF50 and BF100, respectively. The 1st year the values of total dry yield were better in BF150 application. The 2nd year the total yield increased by 50.27% in BF100 and 50.09% BF150 treatments. Moreover, LAI was ameliorated by applied the BF100 and BF150 treatments both in two studied years. A key finding in this study is that Aloysia citrodora ecosystem favors herbaceous plant species richness. Also, soil factors (soil organic matter, P and K) promote positive herbaceous plant diversity within the Aloysia citrodora ecosystem. Finally, Aloysia citrodora can be a promising medicinal plant, cultivated under the Mediterranean climatic conditions.

The molecular interaction between the nematode and the host plant cells is complex and sophisticated. Initial contact with the plant parasitic nematodes (PPNs) triggers immune response in the host plant system which includes the release of toxic molecules. To put a bridle on this immune response, PPNs trigger pivotal cytoprotective mechanisms, such as antioxidant and detoxification pathways. Mechanisms of these pathways have been studied in PPNs and the specific genes involved have been targeted for gene silencing research in view of developing novel control measures. However, one of the important group of proteins involved in detoxification pathways known as ABC-transporters are not being studied until recently in PPNs. This opinion article focusses on the current knowledge and future prospects of ABC transporters in PPNs.

A synoptic review of plant disease epidemics and outbreaks was made using two complementary approaches. The first approach involved reviewing scientific literature published in 2021, in which quantitative data related to new plant disease epidemics or outbreaks were obtained via surveys or similar methodologies. The second approach involved retrieving new records added in 2021 to the CABI Distribution Database, which contains over a million global geographic records of organisms from over 50,000 species. The literature review retrieved 186 articles, describing studies in 62 categories (pathogen species/species complexes) across >40 host species on 6 continents. Pathogen species with >5 articles were: Bursaphelenchus xylophilus, Candidatus Liberibacter asiaticus, cassava mosaic viruses, citrus tristeza virus, Erwinia amylovora, Fusarium spp. complexes, Fusarium oxysporum f. sp. cubense, Magnaporthe oryzae, maize lethal necrosis co-infecting viruses, Meloidogyne spp. complexes, Pseudomonas syringae pvs, Puccinia striiformis f. sp. tritici, Xylella fastidiosa, and Zymoseptoria tritici. Automated searches of the CABI Distribution Database identified 617 distribution records new in 2021 of 283 plant pathogens. A further manual review of these records confirmed 15 pathogens reported in new locations: apple hammerhead viroid, apple rubbery wood viruses, Aphelenchoides besseyi, Biscogniauxia mediterranea, Ca. Liberibacter asiaticus, citrus tristeza virus, Colletotrichum siamense, cucurbit chlorotic yellows virus, Erwinia rhapontici, Erysiphe corylacearum, Fusarium oxysporum f. sp. cubense Tropical Race 4, Globodera rostochiensis, Nothophoma quercina, potato spindle tuber viroid, and tomato brown rugose fruit virus. Of these, 4 pathogens had at least 25% of all records reported in 2021. We assessed two of these pathogens – tomato brown rugose fruit virus and cucurbit chlorotic yellows virus – to be actively emerging in/spreading to new locations. Although three important pathogens – Ca. Liberibacter asiaticus, citrus tristeza virus and Fusarium oxysporum f. sp. cubense – were represented in the results of both our literature review and our interrogation of the CABI Distribution Database, in general our dual approaches revealed distinct sets of plant disease outbreaks and new records, with little overlap.

Insect-specific viruses (ISVs) include viruses that are restricted to the infection of mosquitoes and are spread mostly through transovarial transmission. Despite using a distinct mode of transmission, ISVs are often closely related to arthropod-borne viruses (arboviruses) that are responsible for human diseases and able to infect both mosquitoes and vertebrates. ISVs can also induce a phenomenon called “superinfection exclusion”, whereby a primary ISV infection in an insect inhibits subsequent viral infections of the insect. This has sparked interest in the use of ISVs for the control of pathogenic arboviruses transmitted by mosquitoes. In particular, insect-specific flaviviruses (ISF) have been shown to inhibit infection of vertebrate-infecting flaviviruses (VIFs) both in vitro and in vivo. This has shown potential as a new and ecologically friendly biological approach to the control of arboviral disease. For this intervention to have lasting impacts for biological control, it is imperative that ISFs are maintained in mosquito populations with high rates of vertical transmission. Therefore, these strategies will need to optimize vertical transmission of ISFs in order to establish persistently infected mosquito lines for sustainable arbovirus control. This review compares recent observations of vertical transmission of arboviral and insect-specific flaviviruses and potential determinants of transovarial transmission rates to understand how the vertical transmission of ISFs may be optimised for effective arboviral control.

The structure and expression of exogenous genes in transgenic crops are critical for the target traits. R7569 was a mutant event identified in this study with deletion at the 3' end of cry1Ac gene compared to the transgenic insect-resistant cotton MON531 event with commercial application. R7569 has the same exogenous insertion structure as MON531, but with a deletion in the 3' end of the cry1Ac gene and the terminator region. R7569 has a truncated cry1Ac gene with the length of 2,554 bp, encoding 881 amino acids. The transcription termination site was mainly concentrated downstream of the truncated position and extended 160-270 bp from the truncated position using rapid-amplification of cDNA ends (RACE). The transcript levels of cry1Ac genes of R7569 and MON531 decreased gradually at seedling, bud and bell stages, and the transcript levels of cry1Ac genes of R7569 were significantly higher than those of MON531 in seedling and bud stages, but there was no significant difference in the boll stage. The content of Cry1Ac protein in R7569 gradually decreased with the seedling, bud and boll stage, and the content of Cry1Ac protein in all three periods was higher than that of MON531. The insect resistance assay showed that the resistance levels of R7569 and MON531 were both at the high level, and the corrected mortality rate against bollworms was 99.5% and 95.2%, respectively, and there was no significant difference between them. The LC50 value of R7569 was 0.732ng/g dw, with a slope of 1.654 indicating a high level of resistance to bollworm. In this study, for the first time, we found a partial deletion of the target gene in commercially applied transgenic crops, and the partial deletion of the 3' end of the cry1Ac gene retained a better transcription, expression level and insecticidal activity, which can provide a specific case for the safety evaluation of transgenic crops.

The consumption of insects in the Democratic Republic of Congo is an ancient practice. The taxonomic identification and characterization of “entomophagy” seem urgent in Kinshasa following the related anthropogenic pressures (demographic pressure, uncontrolled urbanization) leading to a food crisis. This study aims to assess the diversity of edible insects and their consumption by the population of Kinshasa. A total of 360 people were surveyed to identify the insect species consumed in Kinshasa, the ecosystems that provide them, and the preference for species in relation to ethnolinguistic groups, churches (faith) and household size. Three species of insects most consumed in Kinshasa are: Gonimbrasia jamesonii, Cirina forda and Brachytrupes membranaceus. 41.9% of these insects are consumed by the Kongo ethno-linguistic group, 23.6% by the Luba, 20.6% by the Ngala and 13.9% by the Swahili. Inhabitants of the communes of Kassa Vubu (57%), Maluku (54%), Masina (54%) and Mont-Ngafula (39%) mainly consume C. forda and G. jamesonii. Luba and Kongo ethnolinguistic groups mainly consume C. forda while the Swahili and Ngala consume G. jamesonii. Catholic, Revivalist Church, Kimbagu, Islam, Protestant and Johovah's Witness believers mainly consume C. forda and G. jamesonii. This study made it possible to understand the socio-cultural influence on the degradation (pressure) of biodiversity and; contributes to SDGs 2 and 12 by 2023..

Forest-grass intercropping patterns of protected cultivation has been widely used to increase productivity and sustainability in modern agriculture. However, there have been few studies of wolfberry (L. barbarum) intercropping and it is cultivated mostly by clean tillage. This paper takes 10 kinds of wolfberry-forage intercropping patterns, compares the effects of different planting modes on the growth, yield, photosynthetic characteristics, nutritional quality, and the degree of occurrence of diseases and pests, which aims to make clear the effects of wolfberry-forage intercropping patterns on wolfberry. The results showed that different intercropping combinations had different effects on wolfberry. The overall effect of Wolfberry-forage intercropping patterns on the growth of wolfberry were promote the photosynthetic characteristics. Ryegrass could increase the branch number, Lvyuan 5 and mangel increase branch length; Mangel, clover, sweet sorghum, ryegrass and stipas could reduce the damage of powdery mildew by 69%, and alfalfa, stipa and ryegrass could reduce the occurrence of aphids by 67% (P<0.05). Intercropping could increase carotenoids, flavonoids and ascorbic acid contents to improve wolfberry quality by 21%, 53% and 127%, respectively (P<0.05). Among all the performances, wolfberry-mangel intercropping has absolute advantages in promoting wolfberry yield and quality increase. Therefore the best patterns of wolfberry-mangel, wolfberry-ryegrass, wolfberry-alfalfa and wolfberry-clover were chosen.

The whitefly leads extensive damage to plants through direct feeding, honeydew secretion, plant physiological disorders, and vectoring plant viruses. This study aimed to evaluate the physiological characteristics of susceptible and resistant soybean cultivars to B. tabaci. The experiments were conducted in a greenhouse. Eleven soybean cultivars were selected and infested with 100 adults of B. tabaci at the V3 stage. The evaluation of photosynthetic parameters, such as photosynthetic rate, leaf transpiration, stomatal conductance, and internal CO2 concentration, revealed that B. tabaci infestation influenced gas exchange in soybean plants. The photosynthetic rate was higher in cultivars AS3810 and M8349 during the V6 stage. Infestations led to alterations in photosynthetic parameters, suggesting increased energy demand to maintain photosynthetic activity. However, the response to infestation varied among different cultivars, indicating varying levels of resistance and tolerance to the whitefly's damage. Additionally, the impact of infestation was more significant during the vegetative phenological. In conclusion, B. tabaci infestation affects soybean plants' physiology, leading to changes in gas exchange parameters and water use efficiency. The response to infestation varied among soybean cultivars, suggesting potential differences in resistance to the pest. The study highlights the importance of evaluating the physiological impact of whitefly infestations on soybean.

Genetic engineering technology offers opportunities to improve many important agronomic traits in crops, including insect-resistance. However, genetically modified (GM) exogenous proteins in edible tissues of transgenic crops has become an issue of intense public concern. To advance the application of GM techniques in maize, a Cre/loxP-based strategy was developed for manipulating the transgenes in green tissues while locking it in non-green tissues. In the strategy, the site-specific excision can be used to switch on or off the expression of transgenes at specific tissues. In this work, two basic transgenic maize named KEY carrying the Cre gene and LOCK containing the Vip3A gene with a blocked element, were obtained based on their separate fusion gene cassettes. The expression level and concentration of Vip3A were observed with high specific accumulation in green tissues (leaf and stem), and only a small amount in root and kernel tissues in the KEY × LOCK hybrids. The insect-resistance of transgenic maize against two common lepidopteran pests, Ostrinia furnacalis and Spodoptera frugiperda, was assessed in the laboratory and field. The results indicated that the hybrids possessed high resistance levels against the two pests, with mortality rates above 73.6% and damage scales below 2.4 compared with the control group. Our results suggested that the Cre/loxP-mediated genetic engineering approach has a competitive advantage in GM maize. Overall, findings from this study are significant for providing a feasible strategy for transgenes avoiding expression in edible parts and exploring novel techniques toward the biosafety of GM plants.

Triatoma infestans (Klug) is an insect recognized as not only an important vector of South-American trypanosomiasis (Chagas disease) but also a model of specific cellular morphofunctional organization and epigenetic characteristics. The purpose of the present review is to highlight certain cellular processes that are particularly unveiled in T. infestans, such as 1) somatic polyploidy involving nuclear and cell fusions that generate giant nuclei; 2) diversification of nuclear phenotypes in the Malpighian tubules during insect development; 3) heterochromatin compartmentalization into large bodies with specific spatial distribution and presumed mobility in the cell nuclei; 4) chromatin remodeling and co-occurrence of necrosis and apoptosis in the Malpighian tubules under stress conditions; 5) epigenetic markers; 6) unexplained response of heterochromatin to valproic acid, an epidrug that inhibits histone deacetylases and induces DNA demethylation in other cell systems. These cellular processes and epigenetic characteristics emphasize the role of T. infestans as an attractive model for cellular research.

Spodoptera frugiperda is a new invasive and highly polyphagous pest that attacks corn in Indonesia. The availability of abundant plant species allows pests to switch to other host plants to maintain their population. This research aims to examine the development, reproduction, nutritional indices, and life table of S. frugiperda in several plant species. The plants tested were corn, rice, broccoli, oil palm, and baby corn as controls. Ten individual insects were used and repeated five times for each plant species. The test results show that different types of plant feed affect the development time, imago life span, fecundity, and fertility of S. frugiperda. The types of plant feed, that were baby corn fruit and broccoli had higher net reproduction value (R0), intrinsic growth rate (r), gross reproduction rate (GRR), shorter mean generation period (T), and population doubling time (DT) than in corn and rice leaves. On oil palm leaf feed no population parameters can be determined because no larvae developed into adults and had the lowest nutritional indices parameters, so that could not be exploited as a host plant. Also, the nutritional indices of several feed plant species tested provided information that broccoli could be a suitable host when there was no corn in the field.

Epidemiology of Hymenopteran-related deaths in Europe due to bee, wasp and hornet stings (Cause Code of Death: X23) based on official registers from WHO Mortality Database is described. Over a 23-year period (1994-2016), a total of 1,691 fatalities were officially recorded, mostly occurring in Western (42.8%) and Eastern (31.9%) Europe. The victims tended to concentrate in: Germany (n=327; 1998-2015), France (n=211; 2000-2014) and Romania (n=149; 1999-2016). The majority of deaths occurred in males (78.1%), within the age group of 25-64 years (66.7%), and in an “unspecified place” (44.2%). The X23 gender ratio (X23GR) of mortality varied from a minimum of 1.4 for Norway to a maximum of 20 for Slovenia. The highest X23MR, expressed in terms of annual rates and per million inhabitants, were recorded in countries from Eastern Europe (0.35) followed by Western (0.28), Northern (0.23) and Southern Europe (0.2). The countries with the highest and lowest mean X23MR were Estonia (0.61), Austria (0.6) and Slovenia (0.55); and Ireland (0.05), United Kingdom (0.06) and the Netherlands (0.06), respectively. Country-by-country data show that the incidence of insect-sting mortality is statistically low, but not negligible.

The genus Flavivirus contains pathogenic vertebrate-infecting flaviviruses (VIFs) and in-sect-specific flaviviruses (ISF). ISF transmission to vertebrates is inhibited at multiple stages of the cellular infection cycle, via yet to be elucidated specific antiviral responses. The Zinc-finger an-tiviral protein (ZAP) in vertebrate cells can bind CpG dinucleotides in viral RNA, limiting virus replication. Interestingly, the genomes of ISFs contain more CpG dinucleotides compared to VIFs. In this study, we investigated whether ZAP prevents two recently discovered lineage II ISFs, Binjari (BinJV) and Hidden Valley viruses (HVV) from replicating in vertebrate cells. BinJV protein and dsRNA replication intermediates were readily observed in human ZAP knockout cells when cultured at 34 ˚C. In ZAP expressing cells, inhibition of the interferon response via interferon response factors 3/7 did not improve BinJV protein expression, whereas treatment with kinase inhibitor C16, known to reduce ZAP’s antiviral function, did. Importantly, at 34 ˚C both BinJV and HVV successfully completed the infection cycle in human ZAP knockout cells evident from infectious progeny virus in the cell culture supernatant. Therefore, we identify vertebrate ZAP as an important barrier that protects vertebrate cells from ISF infection This provides new insights into flavivirus evolution and the mechanisms associated with host switching.

Mass insect rearing can have a range of applications, for example in biological control of insects. Since the performance of released biological control agents determines efficacy, the competitive fitness of insects post release is a key variable. Here, we tested whether inoculation with a gut symbiont, Enterobacter cloacae, and gnotobiotic rearing of larvae could improve insect growth and male competitive fitness of a transgenic diamondback moth, which has shown variation in fitness when reared in different insectaries. All larvae were readily infected with the focal symbiont. Under gnotobiotic rearing pupal weights were reduced and there was a marginal reduction in larval survival. However, gnotobiotic rearing substantially improved the fitness of transgenic males. In addition, in gnotobiotic conditions, inoculation with the gut symbiont increased pupal weights and male fitness, increasing the proportion of transgenic progeny from 20 to 30% relative to symbiont-free insects. Gnotobiotic conditions may improve the fitness of transgenic males by excluding microbial contaminants, while symbiont inoculation could further improve fitness by providing additional protection against infections, or by normalizing insect physiology. The simple innovation of incorporating antibiotic into diet, and inoculating insects with symbiotic bacteria that are resistant to that antibiotic, could provide a readily transferable tool for other insect rearing systems.

This article presents the analysis of the allocation of important plant areas (IPA), which urgently identifies the most valuable sites for protecting plant objects. The study is based on a comprehensive inventory of the species composition of plants in the flora of the Western Karatau Ridge and the Mangyshlak Peninsula (arid regions)- key sites Akmysh, Samal, and Kogez, considered standard and the best preserved with natural vegetation cover. The flora of these critical sites was analysed using a combination of field surveys, herbarium studies, and a literature review. The results revealed a rich biodiversity, with 104 species from 82 genera and 33 families. Most species belong to 9 families, making up 60.6% of the total taxa. By the nature of life forms, perennial herbaceous plants dominate in all key sites (30 species, 50.0%). The eco-phylogenetic spectrum showed that a significant share of the flora of the studied IPAs is made up of xerophytes (21 species, 35%) and xeromesophytes (9 species, 15%); desert and mountain-steppe species prevail (53.0%). Rare, endangered and Red Book species account for 10-21.3% of those growing in the region. Regarding economic value, fodder, ornamental, honey, medicinal and food species dominate. The Shannon, Simpson, and Jaccard indices assessed alpha-beta and gamma diversity. The index showed that biodiversity in the Samal and Kogez sites is higher and more evenly distributed compared to the Akmysh site. The Whittaker, Harrison and Cody indices confirm significant differences in the species composition between the sites. In light of these compelling results, a clear and urgent call to action is proposed to allocate these areas to the IPA category, a crucial step towards ensuring the long-term existence of natural biocenoses.

Abstract Plant uptake and assimilation of essential nitrogen compounds from the soil are mainly in the form of nitrate and ammonium ions. Plant nitrate transporters (NRTs) play a crucial role in nitrate uptake from the soil into the roots. NRT structural analysis reveals the mechanisms by which these transporters function. Similarly, plant ammonium transporters (AMTs) mediate the uptake of ammonium ions, and their structural analysis has provided valuable information on their mechanisms. After nitrogen uptake by plant roots, the nitrogen metabolism pathway involves the incorporation of nitrogen into organic compounds. Glutamine synthetase (GS) and glutamate synthase (GoGAT) are the master players in this pathway. They work together to convert ammonium ions into glutamine and glutamate, respectively. Different isoforms of GS and GoGAT exist, enabling plants to fine-tune nitrogen metabolism based on environmental cues. Under severe abiotic stress conditions, nitric oxide (NO) has been found to enhance plant survival under drought. Furthermore, the interaction between salinity stress and nitrogen availability in plants has been studied, with NO identified as a potential mediator of responses to salt stress. On the other hand, excessive use of nitrate fertilizers can lead to health and environmental issues. Therefore, alternative strategies, such as establishing nitrogen fixation in plants through diazotrophic microbiota, have been explored to reduce reliance on synthetic fertilizers. Ultimately, genomics can identify new genes related to nitrogen fixation, which could be harnessed to improve plant productivity. By deciphering the genetic basis of nitrogen-fixing traits, researchers aim to develop crops that efficiently utilize nitrogen from the environment.

Medicinal plants are generally defined as rare herbals with potent medicinal activities that can be used as an alternative treatment for diseases. Recent studies exploring novel medicine developments, originating from folk-medicinal practices challenges this notion and suggests that both the circumference of the term medicinal plant and their potential application covers a substantially extensive verse than previously suggested. While medicinal plants are not limited to the borders of any country, Bangladesh and its south-east Asian neighbors do boast a huge collection of potent medicinal plants with considerable folk-medicine history compared to most other countries of the world. MPDB 2.0 is the continuation of MPDB 1.0, it serves as both a data repertoire for medicinal of Bangladesh and a user-friendly interface for researchers, health practitioners, drug developers, and students who wish to study the various medicinal & nutritive plants scattered around Bangladesh and the underlying phytochemicals contributing to their efficacy in folk medicine. While in developing MPDB 2.0 human diseases have been highly focused upon, the information in this database is not limited in its application for human diseases or diseases only, as many of the plants indexed here can serve in developing biofuel or bioremediation technologies or nutritive diets or cosmetics, etc. MPDB 2.0 comprises a collection of more than five hundred medicinal plants from Bangladesh along with a record of their corresponding scientific, family, and local names together with their utilized parts, information regarding ailments, active compounds, and PubMed ID of related publications.

More than 700 thousand human deaths from mosquito bites are observed annually in the world. It is more than 2 times the number of annual murders in the world. In this regard, the invention of new more effective methods of protection against mosquitoes is necessary. In this article for the first time, comprehensive studies of mosquito neutralization using machine vision and a 1 W power laser are considered. Developed laser installation with Raspberry Pi that changing the direction of the laser with a galvanometer. We developed a program for mosquito tracking in real. The possibility of using deep neural networks, Haar cascades, machine learning for mosquito recognition was considered. We considered in detail the classification problems of mosquitoes in images. A recommendation is given for the implementation of this device based on a microcontroller for subsequent use as part of an unmanned aerial vehicle. Any harmful insects in the fields can be used as objects for control.

Cleomella serrulata is a native flowering plant found in North America. Even though this plant is of ecological and native medicinal importance, very little is known about the genomic makeup of Cleomella and the Cleomaceae family at large. Here we report the complete chloroplast genome of Cleomella serrulata and provide an evolutionary comparison to other chloroplast genomes from Cleomaceae and closely related families. This study confirms the taxonomic placement of Cleomella as a distinct genus, but also provides phylogenetic insights that imply potential adaptive strategies and evolutionary mechanisms driving genomic diversity of the Cleomella genus. Whole genome-based and ANI comparisons indicate that the Cleomella species form a distinct clade that is about equidistant from the other Cleomaceae genera as it is from the genera from the nearby Capparaceae and Brassicaceae. This is the first complete chloroplast-based phylogenetic comparison of Cleomella species to other related genera and helps refine the complex taxonomic distinctions of the Cleomaceae.

Segmentation of a region of interest is an important pre-processing step for many colour image analysis techniques. Similarly segmentation of plant in digital images is an important preprocessing step in phenotying plants by image analysis. In this paper we present an analytical study to statistically determine the suitability of colour space representation of an image to best detect plant pixels and separate them from background pixels. Our hypothesis is that the colour space representation in which the separation of the distributions representing plant pixels and background pixels is maximized would be the best for detection of plant pixels. The two classes of pixels are modelled as a Gaussian mixture model (GMM). In our GM modelling we don't make any prior assumption about the number of Gaussians in the model. Rather a constant bandwidth mean-shift filter is used to cluster the data and the number of clusters and hence the number of Gaussians is automatically determined. Here we have analysed following representative colour spaces like $RGB$, $rgb$, $HSV$, $Ycbcr$ and $CIE-Lab$. This is because these colour spaces represent several other similar colour spaces and also an exhaustive study of all the colour space will be too voluminous. We also analyse the colour space feature from the two-class variance ratio perspective and compare the results of our hypothesis with this metric. The dataset for this empirical study consist of 378 digital images of plants and their manual segmentation. Dataset consist of various species of plants (arabidopsi, tobacco, wheat, rye grass etc.) imaged under different lighting conditions, indoor and outdoor, controlled and uncontrolled background. In results we obtain better segmentation of the plants in $HSV$ colour space, which is supported by its Earth mover distance (EMD) on the GMM distribution of plant and background pixels.

An increase in plant-sourced (PS) nutrient intake is promoted in support of a sustainable diet. PS dietary minerals and proteins have bioactive properties that can affect bone health and the risk of fracture. In a group randomised, cross-over, design, this study evaluated the post-ingestion, temporal pattern of change in arterialised ionised calcium (iCa), parathyroid hormone (PTH), C-terminal crosslinked telopeptide of type I collagen (CTX) and procollagen type 1 amino-terminal propeptide (P1NP) for 4 hours following ingestion of a novel supplement (SUPP) containing a PS marine multi-mineral + PS protein isolate. A diurnally matched intake of mineral water was used as a control (CON). Compared to baseline, the change in iCa concentration was 0.022 (95%CI, 0.006 to 0.038, p = 0.011) mmol/l greater in SUPP than CON, resulting in a -4.214 (95%CI, -8.244 to -.183, p = 0.042) pg/mL mean reduction in PTH, a -0.64 (95%CI, -0.199 to -0.008, p = .029) ng/mL decrease in the biomarker of bone resorption, CTX, and no change in biomarker of bone formation, P1NP. Used as a dietary supplement, or by incorporation into a food matrix, the promotion of PS marine multi-mineral and PS protein isolates may contribute to a more sustainable diet and overall bone health.
