Aflatoxin contamination (AC) increases as the severity of drought stress increase in peanut. Identifying drought-tolerant (DT) genotypes with resistance to Aspergillus. flavus in semi-arid tropics may aid in development of peanuts that minimizing aflatoxin contamination. The goal is to identify the DT genotypes and resistant to aflatoxin contamination. The experiments curried out at ICRISAT Sahelian Center on fifty-five genotypes assessed in adjacent intermittent water-stressed (WS) from 60th day after sowing to maturity and well-watered (WW) conditions in a randomized complete block design. The yields and components, incidence of A. flavus colonisation, aflatoxin contamination and the seed coat total pthe olyphenol (SCTPP) were estimated. Water deficit reduced pod yield, seed yield, and haulm yield up to 19.49%, 27.24% and 22.07% respectively and increased the immature pods plant-1, (IMPN) and aflatoxin contamination up to 67.16% and 54.95%. The genotypes ICG 2106, ICG 311, ICG 4684, ICG 4543, and ICG 1415 maintained high yield under WS, less of IMPN and the lowest AC variation between WW and WS. The drought tolerant genotypes showed the relationship with aflatoxin resistance and SCTPP (r2=0.80; r2=0.82) under WW and WS. These findings can be used to select the genotype that combine drought tolerance and minimizing aflatoxin contamination.

Background and Objectives: Peanut allergy is the most common single cause of anaphylaxis in children. The risk factors for anaphylaxis in children with peanut allergy are not well defined. Therefore, we aimed to identify epidemiological, clinical and laboratory characteristics of children with peanut allergy that may predict the severity of allergic reaction and anaphylaxis. Materials and Methods: We conducted a cross-sectional study and included 94 children with peanut allergy. Allergy testing was performed, including skin prick testing and determination of specific IgE levels to peanut and its Ara h2 component. In case of discordance between patient history and allergy testing, an oral food challenge with peanut was performed. Results: Anaphylaxis, moderate and mild reactions to peanuts occurred in 33 (35.1%), 30 (31.9%) and 31 (33.0%) patients, respectively. The severity of the allergic reaction only weakly correlated (p = 0.04) with the amount of peanuts consumed. The median number of allergic reactions to peanut was 2 in children with anaphylaxis compared with 1 in other patients (p = 0.04). The median level of specific IgE to Ara h2 was 5.3 IU/ml in children with anaphylaxis compared to 0.6 IU/ml and 10.3 IU/ml in children with mild and moderate peanut allergy (p = 0.06). The optimal cutoff for distinguishing between anaphylaxis and a less severe allergic reaction to peanut was a specific IgE Ara h2 level of 0.92 IU/ml with 90% sensitivity and 47.5% specificity for predicting anaphylaxis (p = 0.04). Conclusions: Epidemiological and clinical characteristics of the patient cannot predict the severity of allergic reaction to peanut in children. Even standard allergy testing, including component diagnostics, is a relatively poor predictor of the severity of allergic reaction to peanuts. Therefore, more accurate predictive models including new diagnostic tools are needed to reduce the need for oral food challenge in most patients.

: F-box proteins are a large gene family in plants, and play crucial roles in plant growth, development, and stress response. However, a comprehensive investigation of F-box family genes in cultivated peanut (Arachis hypogaea L.), has not been undertaken. In this study, we used a bioinformatics approach to identify and analyze F-box protein family genes in the cultivated peanut genome, and examined their expression profiles. Herein, a total of 95 F-box protein family members on 18 chromosomes were identified in cultivated peanut, which were sequentially designed as AhFBX1-AhFBX95. Phylogenetic analysis classified these AhFBXs into four different groups (Group I - IV), and eight subfamilies based on the structural domains of the C-terminal. Structural analysis revealed the presence of ten unique motifs within these genes. Additionally, the investigation of non-synonymous and synonymous substitution rates (Ka/Ks), and synteny in the F-box proteins, provided valuable insights into the evolutionary and domestication processes of AhFBX genes in peanut. Furthermore, in silico expression profiling of 22 different tissues demonstrated that most genes were predominantly expressed in leaves, pistils, and the aerial gynophore tip, while some members showed tissue-specific expression at the apex of both vegetative and reproductive shoots. Subsequently, we validated the expression of 9 differentially expressed genes (DEGs) using transcriptomeic data of lateral branch development and qRT-PCR analysis. These DEGs may be involved in regulating the development of lateral branches and subsequently affect the architecture of peanut plants.

Aqueous enzymatic method is a green oil extraction technology with limited industrial application owing to the need for demulsification of the oil body intermediate product. Existing demulsification methods have problems including low demulsification rates and high costs, such that new methods are needed. Free fatty acids produced by lipid hydrolysis can affect the stability of peanut oil body (POB) at a certain concentration. After screening even-carbon fatty acids with carbon chain lengths below ten, caproic acid was selected for demulsification of POB using response surface methodology and a Box–Behnken design. Under the optimal conditions (caproic acid concentration, 0.22%; material-to-liquid ratio, 1:4.7 (w/v); time, 61 min; and temperature, 79 C), a demulsification rate of 97.87% was achieved. Caproic acid not only adjusted the reaction system pH to cause aggregation of the POB interfacial proteins, but also decreased the interfacial tension and viscoelasticity of the interfacial film with increasing caproic acid concentration to realize POB demulsification. Compared with pressed oil and soxhlet-extracted oil, the acid value and peroxide value of caproic acid demulsified oil were increased, while the unsaturated fatty acid content and oxidation induction time were decreased. However, the tocopherol and tocotrienol contents were higher than those of the soxhlet-extracted oil. This study provides a new method for the demulsification of POB.

Crop yield data is critical for managing sustainable agriculture and assessing national food security. Current study aims to increase Peanut productivity from current levels by analyzing the yield gap of production potential between theoretical yield and actual farmers’ yields. The spatial yield gap of Peanut for Thiruvannamalai district of Tamil Nadu is examined in this paper by integrating the products of microwave remote sensing (SAR Sentinel-1A) with DSSAT CROPGRO peanut simulation model. CROPGRO Peanut model was calibrated and validated by conducting field experiment at Oilseeds Research Station, Tindivanam during Rabi 2019 for predominant cultivars viz. TMV 7, TMV 13, VRI 2 and G 7. Actual attainable yield was recorded by organizing CCE with help of Department of Agriculture Economics and Statistics in the respective monitoring Villages. Regression analysis between maximum recorded DSSAT Leaf Area Index (LAI) at peak flowering stage of peanut and yield recorded by Crop Cutting Experiment (CCE) for spatial yield estimation of Peanut in Thiruvannamalai district of Tamil Nadu during Rabi 2021 was carried out using ArcGIS 10.6 software. The results showed that the simulated potential yield ranged from 3194 to 4843 kg/ha, whereas actual yield ranged from 1228 to 3106 kg/ha, with a considerable disparity between the actual and potential yield levels (1217 to 2346 kg/ha) of the monitored locations. The minimum, maximum and average yield gaps in Peanut for Thiruvannamalai district was assessed as 1890, 2324 and 2134 kg/ha, respectively. To reduce the production difference (Yield gap) of Peanut cultivation, farmers should focus more on management issues such as time of sowing, irrigation or water management, quantity and sources of nutrients, cultivar selection and availability of quality seeds tailored to each region.

Peanut production contributes to food security in northern Ghana due to its ability to tolerate drought and survive on marginal lands. This notwithstanding, poor handling along the value chain favors aflatoxin contamination, a threat to human and animal life. Farmer-led improved storage practice, a potential solution to aflatoxin contamination, have been promoted in northern Ghana. However, there is limited evidence on the factors influencing the use of the improved storage system that may guide dissemination efforts. Using the Cragg’s two stage model, our results show that the probability and intensity of adopting the improved storage structure is principally and significantly influenced by economic active household members, storing of peanut in other places relative to the field, and location of farmer residence. In disseminating the improved peanut storage structure, the identified factors must be incorporated in the selection criteria to ensure maximum uptake and usage. Farmer training programs that incorporate both good agronomic practices and good storage practices must be intensified to reduce postharvest losses due to aflatoxin contamination. These strategies will help mitigate the harmful effect of aflatoxin, ensure economic sustainability, and enhance food security.

Peanut (Arachis hypogaea) is a crop plant with high economic value, but the epigenetic regulation of its growth and development has only rarely been studied. The peanut histone deacetylase 1 gene (AhHDA1) has been isolated and is known to be ABA- and drought-responsive. In this paper, we investigate the role of AhHDA1 in more detail, focussing on the effect of altered AhHDA1 expression in hairy roots at both the phenotypic and transcriptional levels. Agrobacterium rhizogenes-mediated transformation of A. hypogaea hairy roots was used to analyse how overexpression or RNA interference of AhHDA1 affects this tissue. In both types of transgenic hairy root, RNA sequencing was adopted to identify genes that were differentially expressed, and these genes were assigned to specific metabolic pathways. AhHDA1-overexpressing hairy roots were growth-retarded after 20 d in vitro cultivation, and superoxide anions and hydrogen peroxide accumulated to a greater extent than in control or RNAi groups. Overexpression of AhHDA1 is likely to accelerate flux through various secondary synthetic metabolic pathways in hairy roots, as well as reduce photosynthesis and oxidative phosphorylation. Genes encoding the critical enzymes caffeoyl-CoA O-methyltransferase (Araip.XGB85) and caffeic acid 3-O-methyltransferase (Araip.Z3XZX) in the phenylpropanoid biosynthesis pathway, chalcone synthase (Araip.B8TJ0) and polyketide reductase (Araip.MKZ27) in the flavonoid biosynthesis pathway, and hydroxyisoflavanone synthase (Araip.0P3RJ) and isoflavone 2'-hydroxylase (Araip.S5EJ7) in the isoflavonoid biosynthesis pathway were significantly upregulated by AhHDA1 overexpression, while their expression in AhHDA1-RNAi and control hairy roots remained at a lower level or was unchanged. Our results suggest that alteration of secondary metabolism activities is related to overexpression of AhHDA1, which is mainly reflected in phenylpropanoid, flavonoid and flavonoid biosynthesis. Future studies will focus on the function of AhHDA1 interacting proteins and their action on cell growth and stress responses.

Aflatoxins, which have been classified as a group-1 carcinogen are the well-known mycotoxins produced by Aspergillus flavus. Aflatoxins have been linked to liver diseases, acute hepatic necrosis, resulting in cirrhosis or hepatocellular carcinomas due to which it incurs a loss of value in international trade for peanuts contaminated with it. The four main aflatoxins are B1, B2, G1, and G2 of which B1 is predominant. In plants, the cell wall is the primary barrier against pathogen invasion. Cell wall fortifications such as deposition of callose, cellulose, lignin, phenolic compounds and structural proteins help to prevent the pathogen infection. Further, the host cell’s ability to rapidly repair and reinforce its cell walls will result in a reduction of the penetration efficiency of the pathogen. Peanut seed coat acts as a physical and biochemical cell wall barrier against both pre and post-harvest pathogen infection. The structure of seed coat and the presence of polyphenol compounds have been reported to inhibit the growth of A. flavus, however, not successfully employed to develop A. flavus resistance in peanut. A comprehensive understanding of peanut seed coat development and biochemistry will provide information to design efficient strategies for the seed coat mediated A. flavus resistance and Aflatoxin contamination.

Cholangiocarcinoma is an aggressive cancer with limited treatment options. Current therapies, such as cisplatin and 5-fluorouracil (5-FU), often cause side effects and drug resistance. This study investigated a potential new approach by combining a peanut skin extract (KK4-PSE) with these drugs. The extract effectively inhibited the growth of cholangiocarcinoma cells in laboratory studies and showed even greater selectivity for cancer cells compared to healthy cells. When combined with cisplatin or 5-FU, KK4-PSE enhanced the drugs' effects, leading to cell cycle arrest and increased cell death. These combined treatments also reduced levels of proteins that promote cancer cell survival. In animal models, KK4-PSE combined with 5-FU showed superior tumor suppression compared to the combination with cisplatin. Overall, these findings suggest that KK4-PSE may be a promising synergistic agent to improve the effectiveness of chemotherapy for cholangiocarcinoma, warranting further clinical investigation.

To realize the utilization of peanut shell, this study investigates the combustion behavior, chemical kinetics, and thermodynamic parameters of peanut shell using TGA under atmospheric air. Results indicate that increasing the heating rate leads to higher ignition, burnout, and peak temperatures, as observed in the TG/DTG curves shifting to the right. Analysis of combustion performance parameters suggest that higher heating rates can enhance combustion performances. Kinetic analysis using two model-free methods, KAS and FWO, shows that the activation energy (Eα) ranges from 93.30 to 109.65 kJ/mol for FWO and 89.72 to 103.88 kJ/mol for KAS. The data fit well with coefficient of determination values (R2) close to 1. Pre-exponential factors using FWO range from 2.19×106 to 8.06×107 s–1, and for KAS range from9.72×105 to 2.25×107 s–1. Thermodynamic analysis indicates a low-energy barrier (≤ ±6 kJ/mol) between activation energy and enthalpy changes, suggesting easy reaction initiation. Furthermore, variations in enthalpy (ΔH), Gibbs free energy (ΔG), and entropy (ΔS) upon conversion (α) suggest that peanut shell combustion is endothermic and non-spontaneous, with the generation of more homogeneous or well-ordered products as combustion progresses. These findings offer a theoretical basis and data support for the further utilization of agricultural biomass.

Cultivated Peanut (Arachis hypogaea L.) is an important oil and cash crop in worldwide. Hundred pod and seed weight are important components for peanut yield. In order to dissect the genetic control of hundred pod weight (HPW) and hundred seed weight (HSW), a RIL population containing 188 individuals was developed from a cross between Jihua 5 (JH5, large pod and seed size) and M130 (small pod and seed size). An integrated genetic linkage map was constructed by using SSR, AhTE, SRAP, TRAP and SNP markers. The map contained 3,130 genetic markers which were assigned to 20 chromosomes and covered 1,998.95 cM with an average distance 0.64 cM. Based on the genetic map, a total of 31 QTLs for HPW and HSW were identified on 7 chromosomes, explaining 3.7-10.8% of the phenotypic variation by each QTL. Among these, seven QTLs were detected under more than two environments, two major QTLs were found on B04 and B08, respectively. A QTL hotspot containing seven QTLs was detected on chromosome A08 spanning a 2.74 cM genetic interval with 0.36 Mb physical map which included 18 candidate genes. Among these, Arahy.D52S1Z, Arahy.IBM9RL, Arahy.W18Y25, Arahy.CPLC2W and Arahy.14EF4H may be involved in regulating peanut pod and seed weight. These results may help further analysis of genetic mechanisms for pod and seed size in cultivated peanut.

Despite the allergen labeling of peanuts, reducing peanut allergenicity through material selection and breeding may provide an avenue to reduce the severity of allergy due to consumption of foods containing peanuts. This study’s objective is to select peanut lines (cultivar/variety) with lower potent allergenic peptides. One hundred and twenty-two peanut lines (harvested in 2015, New Mexico State, USA.), including 97 lines from the Core USDA- Griffin collections were used for SDS-PAGE analysis in order to find low allergen peanut lines. Eleven peanut lines (Coded MS-1~MS-11) were identified as having lower allergen (Ara h 1, Ara h 2 and Ara h 3) levels and were re-planted in 2016 for further analysis. One line named MS-7 was selected for lower Ara h 1 (8.5-9.5% of total protein) and Ara h 2 (4.2-6.6% of total protein) content in 2015 and 2016. Two-dimensional (2-D) gel electrophoresis (isoelectric focusing plus SDS-PAGE) was conducted to compare the protein (allergens) distributions of MS-7 and check line (MS-9). The 2-D results supported the SDS-PAGE analysis that MS-7 had lower quantities of Ara h 1 and Ara h 2, the two major peanut allergens. The identified peanut line (MS-7) has potential for processing into lower allergenic peanut food products.

Peanut leaf spot is a worldwide disease whose prevalence poses a major threat to peanut yield and quality, and accurate prediction models are urgently needed for timely disease management. In this study, we proposed a novel peanut leaf spot prediction method based on an improved long short-term memory (LSTM) model and multi-year meteorological data combined with disease survey records. Our method employed a combination of convolutional neural network (CNN) and LSTM to capture spatial-temporal patterns from the data and improve the model ability to recognize dynamic features of the disease. In addition, we introduced an attention mechanism module to enhance model performance by focusing on key features. Through several hyper-parameter optimization adjustments, we identified a peanut leaf spot disease condition index prediction model with a learning rate of 0.001, a number of cycles (Epochs) of 800, and an optimizer of Adma. The results showed that the integrated model demonstrated excellent prediction ability, obtaining an RMSE of 0.063 and an R2 of 0.951, which reduced the RMSE by 0.253 and 0.204, and raised the R2 by 0.155 and 0.122, respectively, compared to the single CNN and LSTM. Predicting the incidence and severity of peanut leaf spot disease based on the meteorological conditions and neural networks is feasible and valuable to help growers made accurate management decisions and reduced disease impacts through optimal fungicide application timing.

Peanut (Arachis hypogaea) contains allergenic proteins, which make it harmful to the sensitised population. The presence of peanut in foods must be indicated on label, to prevent accidental consumption by allergic population. In this work, we use chloroplast markers for specifically detection of peanut by real-time PCR, in order to increase the assay sensitivity. Binary mixtures of raw and processed peanut flour in wheat were performed at concentrations ranging from 100000 to 0.1 mg/kg. DNA isolation from peanut, mixtures and other legumes was carried out following three protocols for obtaining genomic and chloroplast-enrich DNA. Quantity and quality of DNA was evaluated, obtaining better results for protocol 2. Specificity and sensitivity of the method has been assayed with specific primers for three chloroplast markers (mat k, rpl16 and trnH-psbA) and Ara h 6 peanut allergen-coding region was selected as nuclear low-copy target and TaqMan probes. Efficiency and linear correlation of calibration curves were within the adequate ranges. Mat k chloroplast marker yielded the most sensitive and efficient detection for peanut. Moreover, detection of mat K in binary mixtures of processed samples was possible up to 10 mg/kg even after boiling, and autoclave 121°C 15 min, with acceptable efficiency and linear correlation. Applicability of the method has been assayed in several commercial food products.

Tree nuts confer many health benefits because of their high content of vitamins and antioxidants and they are increasingly consumed in the last the years. Food processing is an important industrial tool to modify allergenic properties of foods, in addition to ensure safety and to enhance organoleptic characteristics. The effect of high pressure, without and with heating, on SDS-PAGE and immunodetection profile of potential allergenic proteins (anti-11S, anti-2S and anti-LTP) of pistachio, cashew, peanut, hazelnut, almond and chestnut was investigated. Processing based on heat and /or pressure and Ultra high pressure (HHP, 300-600 MPa) without heating was applied. After treating the six tree nuts with pressure combined with heat a progressive diminution of proteins with potential allergenic properties was observed. Moreover, some tree nuts proteins (pistachio, cashew and peanut) seemed to be more resistant to technological processing than others (hazelnut and chestnut). Differences among tree nut varieties were found regarding to protein content, SDS and immunoblotting profile. High pressure combined with heating processing markedly reduce tree nut allergenic potential as the pressure and treatment time increases. HHP do not alter hazelnut and almond immunoreactivity

Groundnut (Arachis hypogaea L.) is a major food and cash crop in Burkina Faso. Due to growing demand for raw oilseeds, there is an increasing interest in groundnut production from traditional rain-fed areas to irrigated environments. However, despite implementation of many initiatives in the past to increase groundnut productivity and production, the groundnut industry still struggles to prosper, due to several constraints including minimal development research and fluctuating markets. Yield penalty due to drought and biotic stresses continue to be a major drawback for groundnut production. This review traces progress in the groundnut breeding that started in Burkina Faso before the country’s political independence in 1960 through to present times. Up to the 1980s, groundnut improvement was led by international research institutions such as IRHO (Institute of Oils and Oleaginous Research) and ICRISAT (International Crops Research Institute for the Semi-Arid Tropics). However, international breeding initiatives were not sufficient to establish a robust domestic groundnut breeding programme. This review also provides essential information about opportunities and challenges of groundnut research in Burkina Faso, emphasising the need for institutional attention to genetic improvement of the crop.

Abstract: This study aimed at investigating the effects of Valencia KK4-type peanut skin ethanolic extract (KK4-PSE) in combination with cisplatin or 5-fluorouracil (5-FU) against HeLa cells in vitro and in xenograft models. MTT assay showed that KK4-PSE, cisplatin, and 5-FU suppressed proliferation of HeLa cells in a dose and time dependent manner. Drug interactions by Chou-Talalay method demonstrated that KK4-PSE enhanced antiproliferative activity of 5-FU against HeLa cells with combination index (CI) values of 0.49 (48 h) and 0.60 (72 h), indicating a synergistic effect, while KK4-PSE combined with cisplatin exhibited an additive effect (CI = 1.02) (72 h) and antagonistic effect at 24 and 48 h exposures (CI = 1.12 and 1.18, respectively). In nude mouse xenograft models, the combination of 5-FU and KK4-PSE markedly reduced HeLa tumor weights compared with control and single agent treatments. The combination of KK4-PSE and 5-FU achieved greater tumor growth inhibition than that of the KK4-PSE-cisplatin combination. KK4-PSE mitigated hepatotoxicity induced by both cisplatin and 5-FU in nude mice. The spleen hyaloserositis was significantly reduced in the combination treatment of 5-FU and KK4-PSE. These findings suggest a potential of KK4-PSE to inhibit cervical cancer cell growth while alleviating the toxicities of cisplatin and 5-FU.

Intercropping of maize (Zea mays L.) and peanuts (Arachis hypogaea L.) (M||P) significantly enhances crop yield. In a long-term M||P field experiment with two P fertilizer levels, we examined how long-term M||P affects topsoil aggregate fractions and stability, organic carbon (SOC), available phosphorus (AP), and total phosphorus (TP) in each aggregate fraction, along with crop yields. Compared to their respective monocultures, long-term M||P substantially increased the proportion of topsoil mechanical macroaggregates (7.6–16.3%) and water-stable macroaggregates (>1 mm) (13.8–36.1%), while reducing the unstable aggregate index (ELT) and the percentage of aggregation destruction (PAD). M||P significantly boosted the concentration (12.9–39.9%) and contribution rate (4.1–47.9%) of SOC in macroaggregates compared to single crops. Moreover, the concentration of TP in macroaggregates (>1 mm) and AP in each aggregate fraction of M||P exceeded that of the respective single crops (P<0.05). Furthermore, M||P significantly increased the Ca2-P, Ca8-P, Al-P, and Fe-P concentrations of intercropped maize (IM) and the Ca8-P, O-P, and Ca10-P concentrations of intercropped peanuts (IP). The land equivalent ratio (LER) of M||P was higher than one, and M||P stubble improved the yield of subsequent winter wheat (Triticum aestivum L.) compared with sole-crop maize stubble. P application augmented the concentration of SOC, TP, and AP in macroaggregates, resulting in improved crop yields. In conclusion, our findings suggest that long-term M||P combined with P application, sustains farmland productivity in the North China Plain by increasing SOC and macroaggregate fractions, improving aggregate stability, and enhancing soil P availability.

An on-site, ultra-sensitive, and quantitative sensing method was developed based on quantum dot nanobeads (QDNBs) and test strip for the determination of total aflatoxins (AFTs) in rice and peanut. The monoclonal antibody against AFT (mAbAFT) was home-made and labeled with QDNB. After the pre-coating of the AFT antigen on the test line (T line), the competitive immunoreactions were conducted between AFT and AFT antigen on the T line with QDNBs-mAbAFT. Under optimal conditions, this approach allowed a rapid response towards AFT with a considerable sensitivity of 1.4 pg/mL and 2.9 pg/mL in rice and peanut matrices, respectively. The put-in and put-out duration were within 10 min. The recoveries for AFT in rice and peanut samples matrices were recorded from 86.25–118.0% with the relative deviations (RSD) below 12%. The assay was further validated via the comparison between this QDNB strip and the conventional HPLC method using spiked samples. Thus, the design provided a potential alternative for on-site, ultra-sensitive, and quantitative sensing of AFT that could also be expanded to other chemical contaminants for food safety.
