COS production is carried out enzymatically using chitosanase, but this process is expensive. Production costs can be reduced by using crude enzyme chitosanase, and the resulting COS has depolymerization < 20 and molecular weight < 3900 Da. Sources of crude enzyme chitosanase come from the soil, sediment, and fishery waste. Chitosanase is produced from bacteria and molds isolated from the source or direct isolation process for extracellular enzymes, especially from fishery waste. Microorganisms in the soil produce chitosanase as a biocontrol and self-defense agent. Bacteria and molds have been isolated, including Bacillus cereus, Bacillus mycoides, Paenibacillus sp., and Penicillium janthinellum. Fishery waste, especially parts of the digestive tract, is a source of chitosanase because of its relation to its nature and food, such as small fish, frogs, and several types of crustaceans. In addition, fishery waste can be used as a carbon and nitrogen source (C/N) in microorganism culture media. The chitin content becomes a stimulant for the synthesis of chitosanase by microorganisms. Crude enzyme chitosanase has an active site that hydrolyzes chitosan by breaking glycosidic bonds and producing glucosamine units (GlcN) and N-acetyl glucosamine (GlcNAc). This mechanism can take place through two events, namely inverting and retaining. Crude enzyme chitosanase can break glycosidic bonds, especially with the same groups, for example, bonds between glucosamine or N-acetyl glucosamine. The COS produced is of the partially acetylated COS (paCOS) type, which still contains an acetyl group. The degree of deacetylation is a significant factor in the resulting COS product, COS with the fully deacetylated COS (fdCOS) type of chitosan with a high degree of deacetylation. Other factors to consider are the raw material of chitosan, the incubation time, the screening process, and the production of microorganism cultures.

Life sciences are increasingly benefiting from multidisciplinary research, combining biology, chemistry and physics. At their intersection, biothermodynamics applies the quantitative framework of thermodynamics to life processes and is a potentially interesting subject for life science students. Here a method is proposed to teach undergraduate life science students the basics of biothermodynamics, building and expanding on the concept of Gibbs energy learned as a part of the physical or general chemistry course. The discussion begins with the role of Gibbs energy as the driving force of metabolism and growth. Moreover, a biological example is proposed of the enthalpy-entropy combinations resulting in negative Gibbs energy, which should be interesting to life science students. Gibbs energy is then used to explain microorganism growth rates, using a simple thermodynamic growth model. Finally, multiplication of viruses is considered, including SARS-CoV-2, using Gibbs energy to explain the hijacking of host cell metabolism. Implementation of the proposed biothermodynamics material in classroom is discussed. The paper should be useful as material for making lectures for undergraduate students and as a starting point for anyone beginning to do research in biothermodynamics.

Black variety of tiger nut tubers (Cyperus esculentus) were differently processed in ascorbic acid and alkaline (Ca(OH)2 and Kanwa) solutions at different concentrations and temperatures. The effect of these treatments on the water content (swelling), soluble loss, dehulling efficiency and microbial load was investigated. Water content or swelling of the tiger nut soaked in acid and alkaline solutions can be predicted by Peleg model like with brown variety. This swelling lead to enhance dehulling efficiency of tuber but caused soluble solute loss overall at the highest temperature of soaking in vitamin C solution. Like soaking in water, the constant of Peleg (k1) decrease with the increase of the temperature of soaking, whereas the constant of capacity of Peleg (k2) is approximately 0.018% for all the conditions. Dehulling efficiency also increased with increasing temperatures (65% at 20°C to 70% at 60°C) for tiger nut tubers soaked in water (0 g/L), dehulling efficiency appeared most strongly affected by concentrations and temperatures of soaking solutions. Like dehulling efficiency, ascorbic solutions has given the best results in term of reduction of microbial load compared to the other soaking solutions. This reduction is considerable at the higher temperatures, reaching a maximum of 4.8 log reduction. Soaking and dehulling treatments gave to tiger nut tuber effective decontamination in term of microbial load and obtained good microbiological quality for final process.

Context: The advent of new strains of resistant microbes and the concomitant growth in multidrug resistance have made antimicrobial resistance an urgent public health concern. New antimicrobials are desperately needed to boost the success rates of treating infectious diseases and save lives. There are many intriguing biomolecules with antibacterial action, which are mostly unexplored in microorganisms. Objectives: The objective of the present review article is to describe the importance of natural compounds against microorganisms using advanced techniques to protect people from diseases. Method: We have searched and incorporated many online literature to gather information from SCOPUS, SCI, PUBMED, ScienceDirect and Medline. We have discussed several microorganism sources for antimicrobials and antifungal drugs. In addition, describes microculturing techniques and applications of microbial-based microsystems. Results and Conclusion: Every kind of higher trophic life depends on microorganisms for sustenance. The unseen majority is essential to understanding how humans and other living forms can survive anthropogenic climate change. Antimicrobial substances and the latest methods, techniques, and strategies for developing effective treatments have been discussed in this article. Novel model systems and cutting-edge biomolecular and computational methodologies could help researchers enhance antimicrobial resistance by completely capitalizing on lead antimicrobials.

The spontaneous combustion of coal in goaf is the main cause of mine fire at present. Lignite has become a major hidden danger in mine production safety because of its easy oxidation. In this paper, an innovative strategy was proposed to inhibit the spontaneous combustion of lignite by using the principle of microbial-induced calcium deposition. Based on the optimized culture method, a novel composite inhibitor of Bacillus pasteurelli was prepared. SEM, pore size analysis and FT-IR experiments were carried out simultaneously, and the oxidation properties of lignite before and after inhibition were quantitatively characterized from the perspective of microstructure, and the flame retardant properties of microbial and chemical retardants on coal samples were compared. The results show that a large number of deposited calcium carbonate particles are obviously attached to the surface of lignite after microbial inhibition treatment, which plays a physical oxygen insulation role. At the same time, the total pore volume and specific surface area of the coal sample decreased by 68.49% and 74.01%, respectively, indicating that microbial inhibitors can effectively plug the primary pores of lignite. Based on the peak measurement of 400-4000cm-1 infrared spectrum of coal samples, it is found that the contents of active groups including hydroxyl, carboxyl and methyl/methylene in lignite molecules after microbial inhibition are lower than those in raw coal, especially the methyl/methylene content involved in the initial oxidation reaction decreased by 96.5% compared with the baseline content in raw coal. The results show that the oxidation and self-heating capacity of lignite after microbial inhibition is effectively restrained in the initial spontaneous combustion. The research results of this paper can provide effective solutions for the prevention and control of coal spontaneous combustion risk in mined-out area.

This study was focused on the development of oil-producing microorganisms to generate lipids. The yeast Rhodotorula mucilaginosa (R. mucilaginosa) was selected for liquid-state cultivation, and the conditions for growth of the yeast cells were assessed. Additionally, the relationships between different nutrient elements and the growth of R. mucilaginosa were explored. The lipid accumulation of R. mucilaginosa is increased under nitrogen-restricted conditions. As the concentration of the carbon source increases, the accumulation of lipids is increased. However, if the carbon source concentration is further increased, the growth of yeast is inhibited. From a large-scale liquid fermentation culture with a fixed inoculation amount of 5%, and from a batch of culture experiments, it was determined that a suitable oil-producing culture was obtained on the 6th day, and the optimum conditions involved a carbon source concentration of 60 g/L, a nitrogen source concentration of 0.5 g/L, and a KH2PO4 concentration of 7.0 g/L. After utilizing different carbon sources in this study, it was found that glucose was the carbon source most conducive to the accumulation of R. mucilaginosa lipids. In addition, the extraction method and solvent for the extraction of R. mucilaginosa lipids were chosen. The acid-heat method using the green organic solvent ethyl acetate exhibited the best performance for extraction of yeast lipids under environmentally friendly and safe conditions. The analysis of lipids showed that the fatty acids obtained primarily contained C16:0, C18:1 and C18:2, and especially C18:1 (41%) lipids, indicating that R. mucilaginosa lipids are a good bio-oil source for the production of biodiesel.

In the current scenario of changing climatic conditions and rising global population, there is always a need to explore novel, efficient, and economical natural products for the benefit of human kind. Biosurfactant is one of the latest explored microbial biomolecules that have been used in numerous fields, including agriculture, pharmaceuticals, cosmetics, food processing, and environment-cleaning industries as raw material, lubrication, wetting, foaming softening, making emulsions, and stabilizing dispersions. The amphiphilic nature of biosurfactant biomolecules showed great advantage, distributing themselves into two immiscible surfaces by reducing interfacial surface tension and increasing the solubility of hydrophobic compounds. Furthermore, their eco-friendly nature, low or even no toxic nature, durability at higher temperatures, and wide range of pH fluctuations making the microbial surfactants preferable compared to their chemical counterparts. Additionally, the biosurfactants can obviate the oxidation flow by eliciting the antioxidant property, antimicrobial, anticancer activity, and drug delivery system, further broadening their applicability in the food, pharmaceutical, and pharma industries. Nowadays, biosurfactant has been broadly utilized to improve the soil quality by improving the concentration of trace elements and mixed with pesticides or applied singly on the plant surfaces for plant disease management. In the present review, we summarise the latest aspect of microbial synthesized biosurfactant compounds, limiting factors of biosurfactant production, and their application in improving soil quality, plant disease management.

Regulation (EU) 2017/745, addresses medical devices and clarifies the scope regarding products containing viable biological materials or organisms. This regulation excludes products containing viable cells or tissues of human or animal origin, as well as those containing viable biological materials or organisms of other origins intended to achieve or support the intended purpose of the products. The term "inviable" is defined as incapable of metabolism or multiplication. The regulation mandates that products manufactured using non-viable biological substances must ensure the highest safety standards, including validated methods of elimination or inactivation of transmissible agents during manufacturing. Several Directives and Regulations provide context for the term "viable" in medical devices, emphasizing the device's ability to perform effectively and safely. Notified Bodies in the EU have interpreted that thermally inactivated microorganisms in a product do not classify it as a medical device under Regulation 2017/745. To address this, it is crucial to differentiate between the inactivation of microorganisms and the viability of their metabolites. Demonstrating the inviability of microorganisms through rigorous testing and evidence is essential for regulatory compliance and safe use in medical products. Clarifying these points and providing necessary evidence can establish that inactivated microorganisms are inviable and meet the requirements for use in medical products.

Curdlan is a water-insoluble polymer that has structure and gelling properties useful in a wide variety of applications such as in medicine, cosmetics, packaging and the food and building industries. The capacity to produce curdlan has been detected in certain soil-dwelling bacteria of various phyla, although the role of curdlan in their survival remains unclear. One of the major limitations of the extensive use of curdlan in industry is the high cost of production during fermentation, partly because production involves specific nutritional requirements such as nitrogen limitation. Engineering of the industrially relevant curdlan-producing strain Agrobacterium sp. ATTC31749 is a promising approach that could decrease the cost of production. Here, during investigations on curdlan production, I found that curdlan was deposited as a capsule. Curiously, only a part of the bacterial population produced a curdlan capsule. This heterogeneous distribution appeared to be due to the activity of Pcrd, the native promoter responsible for the expression of the crdASC biosynthetic gene cluster. To improve curdlan production, Pcrd was replaced by a promoter (PphaP) from another Alphaproteobacterium, Rhodobacter sphaeroides. Compared to Pcrd, PphaP was stronger and only mildly affected by nitrogen levels. Consequently, PphaP dramatically boosted crdASC gene expression and curdlan production. Importantly, the genetic modification overrode the strict nitrogen depletion regulation that presents a hindrance for maximal curdlan production and from nitrogen rich, complex media, demonstrating excellent commercial potential for achieving high yields using cheap substrates under relaxed fermentation conditions.

Sterepinic acids A–C (1–3), new carboxylic acids with two primary alcohols, have been isolated from a fungal strain of Stereum sp. OUPS-124D-1 attached to the marine alga Undaria pinnatifida. Dihydro-1,5-secovibralactone (4), a new vibralactone derivative, was isolated from the same fungal metabolites together with known vibralactone A (5), and 1,5-secovibralactone (6). The planar structures of these compounds have been elucidated by spectroscopic analyses using IR, HRFABMS, and NMR spectra. To determine the absolute configuration of the compounds, we used the phenylglycine methyl ester (PGME) method. These compounds exhibited less activity in the cytotoxicity assay against cancer cell lines.

The influence of polysialic acid (PSA) and sialic acid (SA) on the gut microbial community composition and metabolites in healthy human was investigated using a bionic gastrointestinal reactor. The results indicated that PSA and SA significantly changed the gut microbiota and metabolites in different degrees. PSA can increase the relative abundances of Faecalibacterium and Allisonella, whereas SA can increase those of Bifidobacterium and Megamonas. Both can significantly increase the content of short-chain fatty acids. The results of metabolome analysis showed that PSA can upregulate ergosterol peroxide and gallic acid and downregulate the harmful metabolite N-acetylputrescine. SA can upregulate 4-pyridoxic acid and lipoic acid. PSA and SA affect gut microbiota and metabolites in different ways and have positive effects on human health. These results will provide reference for the further development of PSA- and SA-related functional foods and health products.

Early weaning of piglets caused excessive concentration of free radicals, which would oxidize and damage the intestinal tract of piglets, resulting in dyspepsia, low growth, diarrhea, microbiota disorders and intestinal infections. This research studied on effect of different levels of taurine in diets on intestinal oxidative damage in early weaned piglets. A total of 48 weaned piglets (6.71 ± 0.73 kg) of 28-day-old crossbred "Duroc × Landrace × Yorkshire" were assigned to 4 groups with 12 replicates per group and 1 pig per replicate: piglets were fed a basal diet with 0.0% (CON), 0.2% taurine (L-Tau), 0.3% taurine (M-Tau), or 0.4% taurine (H-Tau). The animal experiment lasted for 30 days. With the increase of dietary taurine dosage, the activity of antioxidant enzymes including superoxide dismutase（SOD）,glutathione peroxidase（GPX）,catalase（CAT） and peroxidase（POD） were up-regulated (Linear, P < 0.05), the mRNA abundance of antioxidant enzymes including SOD1, SOD2, GPX1 and heme oxygenase-1（HO-1）were up-regulated (P < 0.05), glutathione (GSH) concentrations was up-regulated (Linear, P < 0.05) , nitric oxide (NO), malondialdehyde (MDA) and oxidized glutathione (GSSG) concentrations were down-regulated (Linear, P < 0.05);The villus height (VH) and villus height/ crypt depth (VH/CD) of jejunum and ileum were increased (Linear, P < 0.05); The expression of tight junction proteins including zonula occludens protein 1(ZO-1), occludin (OCLN) and closure protein 1(CLDN-1) in the jejunum mucosa were up-regulated (Linear, P < 0.05), the mRNA abundance of ZO-1, OCLN and CLDN-1 were up-regulated (P < 0.05),and the D-lactose (D-LA) content in blood was down-regulated (Linear, P < 0.05); the colonization abundance of Firmicutes was increased (Linear, P < 0.05), while Proteobacteria and Spirochaetota was decreased (Linear, P < 0.05); The diarrhea index decreased (Linear, P < 0.05), while the feed conversion rate increased (Linear, P < 0.05). Collectively, supplementation of 0.3% and 0.4% taurine in feed can significantly alleviate intestinal oxidative damage by enhancing antioxidant capacity of piglets, and improve feed utilization of piglets after repairing intestinal damage.

VpStyA1 and VpStyA2B of Variovorax paradoxus EPS is annotated and characterized as the first representative of an E2-type styrene monooxygenase of proteobacteria. It comprises a single epoxidase (VpStyA1) and a fusion protein (VpStyA2B) which serves mainly as NADH:FAD-oxidoreductase. VpStyA2B had a Km of 33.6 ± 4.0 µM for FAD and a kcat of 22.3 ± 1.1 s-1. VpStyA2B and VpStyA1 showed monooxygenase activity on styrene of 0.14 U mg-1 and 0.46 U mg-1 as well as on benzyl methyl sulfide of 1.62 U mg-1 and of 3.11 U mg-1. A putative fusion region at position 408 (AREAV) was mutated to provide insights on VpStyA2B-function. The best mutant (408-AAAAA) obtained showed a 6.6-times higher affinity for FAD while keeping the NADH-affinity and -oxidation activity. Corresponding epoxidase activity increased (1.6-times). But, other mutants showed still NADH:FAD-oxidoreductase activity, but lost mostly their epoxidase activity indicating effects on the monooxygenase-part as well. Thus, this monooxygenase system represents an interesting candidate for biocatalyst development.

Knowledge of the agricultural soil microbiota, of the microbial consortia that comprise it, and the promotion of agricultural practices that maintain and encourage them, is a promising way to improve soil quality for sustainable agriculture and to provide food security. Although numerous studies have demonstrated the positive effects of beneficial soil microorganisms on crop yields and quality, the use of microbial consortia in agriculture remains low. Microbial consortia have more properties than an individual microbial inoculum, due to the synergy of the microorganisms that make them up. This review describes the main characteristics, ecosystem functions, crop benefits and biotechnological applications of microbial consortia composed of arbuscular mycorrhizal fungi, plant growth promoting bacteria and actinobacteria, to promote the restoration of agricultural soils and, consequently, the quality and health of agricultural crops. The aim is to provide knowledge that will contribute to the development of sustainable and sufficiently productive agriculture, which will adapt in a good way to the pace of the growing human population and to climate change.

This study presents the numerical investigation of bioconvective nanofluid (NF) flow compris-ing gyrotactic microorganisms through convective boundary conditions (CBC) enclosing heat and mass transmission analysis above an inclined extending cylinder. Via appropriate transfor-mation, the set of Partial Differential Equations is transformed into a scheme of nonlinear ODEs. A new set of variables are presented in the directive to get the first-order convectional equations and, then solved numerically by using bvp4c. The influence of several factors on velocity, ener-gy, concentration, and the density of motile microorganisms (DMM) is investigated and exam-ined. The analysis describes and addresses all physical measures of concentration such as Skin Friction (SF), Sherwood number (SN), DMM, and Nusselt number (NN). The present conse-quences display a validation with published. For confirmation of bvp4c, the ND-solve approach is also applied. Error analysis is also examined for the confirmation of the mathematical model.

The antimicrobial properties of the new strains of microorganisms isolated from natural sources of various ecological niches in the Moscow Region and the Republic of Tatarstan were studied. An antifungal activity of isolates was established in a test culture of toxin-producing microscopic fungi that cause various plant diseases: Fusarium oxysporum, Aspergillus flavus, Penicillium spp. and Candida albicans. Of the 46 studied microorganisms, 4 isolates (Bacillus subtilis, Propionibacterium freudenreichii, Lactobacillus plantarum and Streptomyces spp.) showed an ability to produce biologically active metabolites with a pronounced antimicrobial potential. This is promising in the growth and protection of the Scots pine, pedunculate oak and small-leaved linden forest crops from diseases caused by fungi Lophodermium pinastri Chev. and Microsphaera alphitoides stimulation. Based on selected isolates, a Bacterial product LRV composition has been developed. A Scots pine, pedunculate oak and small-leaved linden seedlings single and double foliar treatment with Bacterial product LRV at a concentration of 10 ml/l led to an increase in the growth of an above-ground part by 31.8, 51.9 and 25.4 %, respectively, and an underground part – by 25.0, 37.2 and 25.7 %, respectively, compared with the control. The weight of seedling at the end of the study exceeded the control variant by an average of 26.0, 44.0 and 78.0 %, respectively. A use of a plant protection Bacterial product LRV did not have a significant effect on a powdery mildew and Schütte disease damage to forest crops. A Biogeosystem Technique methodology has been developed to improve a long-term forest growing.
