The present context regarding development of the food products trade at European Union level presumes manifestation of some tendencies and challenges meant to claim at the highest level the adaptation capacity of economical agents - both producers and traders. Among these we can take into account their need to comply with organic product labeling standards as an integral part of food products safety policies at Union level. In order to be able to implement a proper label type, not only from the point of view of standards but especially from the perspective given by the consumers perception and motivation toward a healthy sustainable food consumption, producers have to constantly innovate, to find new design solutions for food products labels. This article propose an exploratory research in the form of a field survey that highlights the perceptions of Romanian and Serbian consumers, altogether from the urban environment in relation to organic food labeling and how they influence purchasing decision-making. A random number of 373 Serbian consumers’ questionnaires, respectively 1262 Romanian consumers’ questionnaires have been validated, the results showing interesting results from the point of view of national consumption profile differences due to different cultural, economic and social factors as well as different levels of integration within European economy for the both countries. Main conclusions of the research stress the fact that perceptions regarding organic food products labels are constructed around variables like: type of information to appear within the label, category of information that are missing from labels, degree of producers interest in proper labeling of organic food products, relevance of the information on the organic food labels as regards the decision to purchase the products etc. Conclusions propose some possible developments for innovative future characteristics for organic food products labels.

Convenient labeling of proteins is important for observing its function under physiological conditions. In tissues particularly, heptamethine cyanine dyes (Cy-7) are valuable because they absorb in near infrared (NIR) region (750 – 900 nm) where light penetration is maximal. In this work, we found Cy-7 dyes with a meso-Cl functionality covalently binding to proteins with free Cys residues under physiological conditions (aqueous environments, at near neutral pH, and 37 °C). It transpired that the meso-Cl of the dye was displaced by free thiols in protein, while nucleophilic side-chains from amino acids like Tyr, Lys, and Ser did not react. This finding shows a new possibility for convenient and selective labeling of proteins with near-IR fluorescent probes.

Urbanization influences food culture, particularly in low- and middle-income countries, where there is an increasing consumption of processed and pre-packaged foods. This shift is contributing to a rise in non-communicable diseases. Food labeling standards are crucial for regulating manufacturing practices and aiding consumers in making healthy food choices. We aimed to assess the compliance of local and imported pre-packaged snacks with Tanzanian and international labeling standards in Dar es Salaam, Tanzania. A cross-sectional study was conducted on 180 snack products. A checklist based on Tanzanian and Codex labeling standards was used to evaluate adherence. We also examined factors influencing adherence, such as product origin, price, category, purchase location, and package size. The majority of the snacks demonstrated partial adherence to labeling standards. Imported products showed significantly better full adherence to both Tanzanian and International standards. Notably, over half of the products used English for labeling, and infrequently used the recommended World Health Organization Front-of-Pack nutrition Labeling. Product category, origin, and package size were significantly associated with higher levels of International standard adherence (p<0.05). The inadequate adherence to mandatory labeling standards, scarce use of Swahili and FoPL highlight the need to strengthen labeling practices and potential challenges faced by consumers in understanding nutritional information. Thus, strengthening and emphasizing good labeling practices is of urgency as we seek to address diet-related Non-communicable diseases.

Mitochondria, crucial intracellular organelles, are central to energy metabolism, signal transduc-tion, apoptosis, calcium homeostasis, and a myriad of other biological processes, making them a focal point in diverse research fields. The capacity to fluorescently labeling and visually track mitochondria is crucial for understanding their biological roles. We present Mulberrin-Cy3, a novel small molecule fluorescent probe that selectively labels mitochondria in animal cells, in-cluding cancer cells, with relative ease. This protocol details the synthesis of Mulberrin-Cy3 and its use for visualizing mitochondria in living cells. The synthesis is straightforward and time-efficient, and the labeling method is more accessible than traditional approaches, providing a cost-effective option for mitochondrial visualization at room temperature. The labeling is rapid, with effective labeling achieved within 5 minutes of incubation. The fluorescent signal is stable and brighter, offering a significant advantage over existing methods. Mulberrin-Cy3 represents a promising mitochondrial labeling compound, providing researchers with a novel experimental tool to explore the complex biological functions of mitochondria. This innovation has the potential to significantly advance our comprehension of mitochondrial dynamics and their role in cellular health and disease.

The inherent dependency of deep learning models to labeled data is a well-known problem and one of the barriers that slows down the integration of such methods into different fields of applied sciences and engineering, in which experimental and numerical methods can easily generate a colossal amount of unlabeled data. This paper proposes an unsupervised domain adaptation methodology that mimics the peer review process to label new observations in a different domain from the training set. The approach evaluates the validity of a hypothesis using domain knowledge acquired from the training set through a similarity analysis, exploring the projected feature space to examine the class centroid shifts. The methodology is tested on a binary classification problem, where synthetic images of cubes and cylinders in different orientations are generated. The methodology improves the accuracy of the object classifier from 60% to around 90% in the case of a domain shift in physical feature space without human labeling.

Flaxseed (Linum usitatissinum L.) oil is an important source of α-linolenic (C18:3 ω-3), this polyunsaturated fatty acid is well known for its nutritional role in human and animal diet. Understanding storage lipid biosynthesis in developing flaxseed embryos can lead to an increase in seed yield. While a tremendous amount of work has been done on different plant species to highlight their metabolism during embryos development, flaxseed metabolic flux analysis is still lacking. In this context, we have developed an in vitro cultured developing embryos of flaxseed and determined net fluxes by performing three complementary parallel labeling experiments with 13C-labeled glucose and glutamine. Metabolic fluxes were estimated by computer- aided modeling of the central metabolic network including 11 cofactors of 118 reactions of the central metabolism, 12 pseudo fluxes. A focus on lipid storage biosynthesis and the associated pathways was done in comparison with rapeseed, arabidopsis, maize and sunflower embryos. In our conditions, glucose was the main source of carbone of flaxseed embryos, leading to the conversion of phosphoenolpyruvate to pyruvate. The oxidative pentose phosphate pathway (OPPP) was identified as the producer of NADPH for fatty acid biosynthesis. Overall, the use of 13C-metabolic flux analysis provided new insight into flaxseed embryos metabolic processes involved in storage lipids biosynthesis. The elucidation of the metabolic network of this important crop plant reinforces the relevance of the application of this technique to the analysis of complex plant metabolic systems.

The present study set out to explore the option of developing food portion size for nutritional labelling purposes using two European Union (EU) dietary surveys. The surveys were selected as they differed in (a) methodologies (food diary v food frequency questionnaire), (b) populations (Irish National Adult Nutrition Survey: NANS) versus a seven-country based on the pan EU study Food4Me), (c) food quantification (multiple options v, solely photographic album) and (d) duration (4 consecutive days v recent month). Using data from these studies, portion size was determined for 15 test foods, where portion size was defined as the median intake of a target food when consumed. The median values of the portion sizes derived from both the NANS and Food4Me surveys were correlated (r = 0.823; P<0.00) and the mean of the two survey data sets were compared to US values from the Recognized as Customarily Consumed (RACC) database. There was very strong agreement across all food categories between the averaged EU and the US portion size (r = 0.947; P<0.00). It is concluded that notwithstanding the variety of approaches used for dietary survey data in the EU, the present data supports using a standardized approach to food portion size quantification for food labelling in the EU.

Identifying the total number of fruits on trees has long been of interest in agricultural crop estimation work. Yield prediction of fruits in practical environment is one of the hard and significant tasks to obtain better results in crop management system to achieve more productivity with regard to moderate cost. Utilized color vision in machine vision system to identify citrus fruits, and estimated yield information of the citrus grove in-real time. Fruit recognition algorithms based on color features to estimate the number of fruit. In the current research work, some low complexity and efficient image analysis approach was proposed to count yield fruits image in the natural scene. Semi automatic segmentation and yield calculation of fruit based on shape analysis is presented. Color and shape analysis was utilized to segment the images of different fruits like apple, pomegranate obtained under different lighting conditions. First the input sectional tree image was converted from RGB colour space into the colour space transform (i.e., YUV, YIQ, or YCbCr). The resultant image was then applied to the algorithm for fruit segmentation. After it is applied Morphological Operations which is enhanced image then execute Blob counting method which identify the object and count the number of it. Accuracy of this algorithm used in this thesis is 82.21% for images that have been scanned.

We investigate new graph characteristics namely total (vertex, edge) face irregularity strength of gen- eralized plane grid graphs Gmn under k-labeling Phi of type (Alpha, Beta, Gamma). The minimum integer k for which a vertex-edge labelled graph has distinct face weights is called the total face irregularity strength of the graph and is denoted by tfs(Gmn). In this article, the graphs G = (V;E; F) under consideration are simple, finite, undirected and planar. We will estimate the exact tight lower bounds for the total face irregularity strength of some families of generalized plane grid graphs.

n our previous paper, we showed that the so-called quantum entanglement also exists in classical mechanics. The inability to measure this classical entanglement was rationalized with the definition of a classical observer which collapses all entanglement into distinguishable states. It was shown that evidence for this primary coherence is Newton’s third law. However, in reformulating a "classical entanglement theory" we assumed the existence of Newton’s second law as an operator form where a force operator was introduced through a Hilbert space of force states. In this paper, we derive all related physical quantities and laws from basic quantum principles. We not only define a force operator but also derive the classical mechanic's laws and prove the necessity of entanglement to obtain Newton’s third law.

Significance: Electrophile signaling is coming into focus as a bona fide cell signaling mechanism. The electrophilic regulation occurs typically through a sensing event (i.e., labeling of a protein) and a signaling event (the labeling event having an effect of the proteins activity, association, etc.). Recent advances: Herein, we focus on the first step of this process, electrophile sensing. Electrophile sensing is typically a deceptively simple reaction between the thiol of a protein-cysteine, of which there are around 200,000 in the human proteome, and a Michael acceptor, of which there are numerous flavors, including enals and enones. Recent data overall paint a picture that despite being a simple chemical reaction, electrophile sensing is a discerning process, showing labeling preferences that are often not in line with reactivity of the electrophile. Critical issues: With a view to trying to decide what brings about highly electrophile-reactive protein-cysteines, and how reactive these sensors may be, we discuss aspects of the thermodynamics and kinetics of covalent/non-covalent binding. Data made available by several laboratories indicate that it is likely that specific proteins exhibit highly stereo- and chemoselective electrophile sensing, which we take as good evidence for recognition between the electrophile and the protein prior to forming a covalent bond. Future directions: We propose experiments that could help us gain a better and more quantitative understanding of the mechanisms through which sensing comes about. Low energy consumption is a significant contribution of new and retrofitted buildings that place an emphasis on energy efficiency. Although the mandatory requirements of the U-factor and R-value are described in the Vietnamese National Technical Regulation on the energy efficiency of buildings—QCVN09:2017/BXD, these values are not widely publicized, leading to designers facing difficulties when choosing suitable materials at the design stage. This study focuses on the method used to declare the heat transfer values of building materials based on the energy labeling process in order to create an energy labeling program for building materials. To accomplish this objective, criteria and labeling procedures for windows, doors, walls, and building roofs have been developed. In addition, a case study of insulation product labeling was used to evaluate the applicability of the developed labeling program in the implementation phase.

If one considers chemical-biology toolsets that have had the greatest impact on numerous fields of life sciences over the most recent years, proximity-labeling tools, such as APEX, and Bio-ID arguably lead the way. This article reflects upon the current state-of-the-art and discusses key limitations underlying these emerging approaches, in particular, the limited functional knowledge they provide in understanding local proteomes / interactomes. This limitation is directly linked to the use of non-biologically- or non-pharmaceutically-relevant reactive intermediates in the course of covalently labeling the local proteomes. As such, these methods cannot report on specific functions of localized protein players, nor can they scrutinize whether the specific functions of such proteins/interactomes can be directly manipulated by pharmacologically-relevant small-molecule ligands. The latest data hint that precision localized electrophile delivery concept ushers a means to address this limitation with high spatiotemporal resolution, and ultimately, in relevant live animals.

Novel cultivation technologies demand the adaptation of existing analytical concepts. Metabolic flux analysis (MFA) requires stable-isotope labeling of biomass-bound protein as the primary information source. Obtaining the required protein in cultivation set-ups where biomass is scarce or inaccessible due to low cell densities and cell immobilization is difficult to date. We developed a non-disruptive analytical concept for 13C-based metabolic flux analysis based on secreted protein as an information carrier for isotope mapping in the protein-bound amino acids. This "metabolic flux probe" (MFP) concept was investigated in different cultivation set-ups with a recombinant, protein-secreting yeast strain. The obtained results grant insight into intracellular protein turnover dynamics. Experiments under metabolic but isotopically nonstationary conditions in continuous glucose-limited chemostats at high dilution rates demonstrated faster incorporation of isotope information from labeled glucose into the recombinant reporter protein than in biomass-bound protein. Our results suggest that the reporter protein was polymerized from intracellular amino acid pools with higher turnover rates than biomass-bound protein. The latter aspect might be vital for 13C-flux analyses under isotopically nonstationary conditions for analyzing fast metabolic dynamics.

Parts of shotgun cartridges are a significant source of plastic litter in the marine environment. Empty cartridge shells may not be picked up by the hunter who fired them, and plastic wads that serve to separate the propellant from the shot load, are lost down-range when a shot is fired. Such litter items constitute a cosmetic and aesthetic problem on coastlines and may cause harm to marine animals and in the later stages of decomposition break down into harmful micro plastic particles. There exists no reliable estimate of the global exposure of marine ecosystems to this plastic source. However, in some countries it has been subject to closer examination, including for example, Denmark where the annual contribution of plastic wads into marine systems was estimated to 600,000 pieces (c2 tonnes), and the accumulated density of washed-up items (both wads and shells) was 3.7 items per 100 m coastline. Increasing awareness of this problem has caused scientists, hunters’ communities and governments to suggest altered practice including transition to the use of biodegradable cartridge components, first and foremost wads as this item is invariable lost during hunting. Several manufacturers provide shotgun cartridges containing biodegradable wads based on different types of materials, including fibers and various types of plastics, for example PVAL (poly(vinyl alcohol)) and PHA (polyhydroxyalkanoate). In this paper, we review the most recent literature on the amounts and related environmental hazards of plastic dispersed from hunting ammunition into marine ecosystems. We summarize the market availability of shotgun cartridges with biodegradable wads and discuss chemical, technical, economical and legal aspects of a transition to the use of such products.

IIAEK (Ile-Ile-Ala-Glu-Lys, lactostatin) is a novel pentapeptide from bovine milk β-lactoglobulin which lowers cholesterol levels. However, the molecular mechanisms underlying the suppression of intestinal cholesterol absorption by IIAEK are unknown. Therefore, we evaluated the effects of IIAEK on intestinal cholesterol metabolism in Caco-2 cells in a human intestinal model. We found that IIAEK significantly reduced the expression of intestinal cholesterol metabolism-associated genes, particularly that of the ATP-binding cassette transporter A1 (ABCA1) protein. Subsequently, we chemically synthesized a novel molecular probe, IIXEK, which can visualize a complex of target proteins interacting with photoaffinity-labeled IIAEK by fluorescent substances. Photoaffinity labeling and MS analysis with IIXEK for the rat small intestinal mucosa and intestinal lipid raft fractions of Caco-2 cells, we identified intestinal alkaline phosphatase (IAP) as a specific molecule interacting with IIAEK and discovered IIAEK common binding amino acid sequence, GFYLFVEGGR. Transfection of IAP siRNA counteracted the decrease in ABCA1 mRNA levels in Caco-2 cells. IIAEK significantly increased IAP mRNA and protein levels, and significantly decreased ABCA1 mRNA and protein levels in Caco-2 cells. In conclusion, we found that IIAEK targets IAP to improve cholesterol metabolism via a novel signaling pathway with a specific activation of IAP and down-regulation of intestinal ABCA1.

Depressive disorder is a disease that causes a decrease in daily function, and the prevalence of depressive disorder in Korea is the highest among OECD countries. However, due to lack of manpower and negative social awareness, timely treatment is not being performed properly. As a solution to this, AI counseling chatbots are emerging, and it is essential to use labeled datasets to create chatbots. Currently, many countries around the world, including Korea, use the Diagnostic and Statistical Manual of Mental Disorders Fifth Edition, DSM-5, but there are no nine labeled datasets according to the DSM-5 depression disorder diagnosis criteria. This study collects sympathetic datasets, analyzes morphemes using Kind Korean Morpheme Analyzer and augments and builds a word dictionary using Word2Vec. As a result, we have labeled the dataset with nine criteria for diagnosing DSM-5 depressive disorder, which will enhance the performance of the counseling chatbot.

For highly time-constrained very short-term investors, reading and extracting valuable information from financial news poses significant challenges. The wide range of topics covered in these news articles further compounds the difficulties for investors. The diverse content adds complexity and uncertainty to the text, making it arduous for very short-term investors to swiftly and accurately extract valuable insights. Variations in authors, media sources, and cultural backgrounds also introduce additional complexities. Hence, performing a bull-bear semantic analysis of financial news using text mining technologies can alleviate the volume, time, and energy pressures on very short-term investors while enhancing the efficiency and accuracy of their investment decisions. This study proposes labeling bull-bear words from a futures corpus detection method that extracts valuable information from financial news, allowing investors to understand market trends quickly. Generative AI models are trained to provide real-time bull-bear advice, aiding investors in adapting to market changes and devising effective trading strategies. Experimental results show the effectiveness of various models, with Random Forest and SVMs achieving an impressive 80% accuracy rate. MLP and Deep learning models also perform well. By leveraging these models, the study reduces the time spent reading financial articles, enabling faster decision-making and increasing the likelihood of investment success. Future research can explore the application of this method in other domains and enhance model design for improved predictive capabilities and practicality.

Stable-isotope dimethyl labeling is a highly reactive and cost-effective derivatization procedure that could be utilized in proteomics analysis. In this study, a liquid chromatography-tandem mass spectrometry in multiple reaction monitoring mode (LC-MS-MRM) platform for the quantification of kiwi allergens was first developed using this strategy. Three signature peptides for target allergens Act d 1, Act d 5, and Act d 11 were determined and were derivatized with normal and deuterated formaldehyde as external calibrants and internal standards, respectively. The results showed that sample preparation with the phenol method provided comprehensive protein populations. Recoveries at four different levels ranging from 72.5–109.3% were achieved for the H-labeled signature peptides of Act d 1 (SPA1-H) and Act d 5 (SPA5-H) with precision ranging from 1.86–9.92%. The limit of quantification (LOQ) was set at 8 pg mL–1 for SPA1-H and at 4 ng mL–1 for SPA5-H. The developed procedure was utilized to analyze seven kinds of hand-made kiwi foods containing 0.0175–0.0515 mg g–1 of Act d 1 and 0.0252–0.0556 mg g–1 of Act d 5. This study extended the applicability of stable-isotope dimethyl labeling to the economical and precise determination of food allergens and peptides.

The generalization of Farey graphs and extended Farey graphs are all originated from Farey graph and scale-free and small-world simultaneously. We propose a labeling of the vertices for it that allows determining all the shortest paths routing between any two vertices only based on their labels. The maximum number of shortest paths between any two vertices is huge as the product of two Fibonacci numbers, however, the label-based routing algorithm runs in linear time O(n). The existence of an efficient routing protocol for Farey-type models should help the understanding of several physical dynamic processes on it.

Global warming poses a threat to plant survival, impacting growth and agricultural yield. Protein turnover, a critical regulatory mechanism balancing protein synthesis and degradation, is crucial for cellular response to environmental changes. We investigated the effects of elevated temperature on proteome dynamics in Arabidopsis thaliana seedlings using 15N-stable isotope labeling and ul-tra-performance liquid chromatography-high resolution mass spectrometry, coupled with the ProteinTurnover algorithm. Analyzing different cellular fractions from plants grown under 22°C and 30°C growth conditions, we found significant changes in the turnover rates of 571 proteins, with a median 1.4-fold increase, indicating accelerated protein dynamics under thermal stress. Notably, soluble root fraction proteins exhibited smaller turnover changes, suggesting tissue-specific adap-tations. Significant turnover alterations occurred with redox signaling, stress response, protein folding, secondary metabolism, and photorespiration, indicating complex responses enhancing plant thermal resilience. Conversely, proteins involved in carbohydrate metabolism and mito-chondrial ATP synthesis showed minimal changes, highlighting their stability. This analysis high-lights the intricate balance between proteome stability and adaptability, advancing our under-standing of plant responses to heat stress and supporting the development of improved thermo-tolerant crops.

RNA, as an important substance for regulating biological growth and development, has significant implications for visualization research. However, many spontaneously fluorescent substances in plants greatly interfere with the effectiveness of plant bioimaging. Aggregetion- induced emission luminogens (AIEgens), due to their luminescent properties, tunable molecular size, high fluorescence intensity, good photostability, and low cell toxicity, have been widely applied in the animal and medical fields. We have found that AIEgens have great potential as RNA fluorescent probes for efficient imaging in plants. In this review, we first introduce several common RNA labeling forms and point out their pros and cons. We briefly describe the development of AIEgens and the AIE mechanism, and then present various practical applications of AIEgens, including detailed examples of their use as biological markers. To further promote the application of AIE in the field of RNA, we suggest the use of AIEgens to modify target RNA via techniques such as click chemistry or CRISPR/Cas, to achieve RNA visualization in plants. highly possible to modify target RNA with AIEgens in vivo for RNA visualization.

Beginning with an introduction that underscores the pivotal role of stem cell tracking in regenerative medicine, this review introduces quantum dots as promising tools for this purpose. It proceeds to explore the fundamental properties of quantum dots, including the quantum confinement effect, size-dependent emission, and exceptional brightness, which render them ideal candidates for stem cell tracking. The historical evolution and recent advancements of quantum dot technology in biomedicine are discussed, emphasizing their growing importance in terms of biocompatibility and safety. Furthermore, various labeling strategies for stem cells using quantum dots are detailed, encompassing surface modification and internalization techniques, with a comparison of their respective advantages and drawbacks. A comprehensive exploration of the wide range of imaging modalities that synergize with quantum dots for stem cell tracking is presented, accompanied by an evaluation of their strengths and limitations. Real-world applications of quantum dot-based stem cell tracking in regenerative medicine, spanning tissue regeneration to disease modeling, are showcased, with illuminating case studies. Current challenges and limitations in quantum dot-based stem cell tracking, including long-term stability and regulatory considerations, are addressed, and innovative directions and emerging trends in quantum dot technology are proposed. Safety considerations and strategies to enhance quantum dot biocompatibility within biological systems are discussed, while ethical and regulatory implications related to their use in stem cell tracking are explored, promoting responsible research and development practices. Finally, the review concludes by summarizing key insights and takeaways, reaffirming the transformative potential of quantum dots in advancing stem cell tracking and the field of regenerative medicine.

The Kuwaiti dialect is a particular dialect of Arabic spoken in Kuwait; it differs significantly from standard Arabic and the dialects of neighboring countries in the same region. Few research papers with a focus on the Kuwaiti dialect have been published in the field of NLP. In this study, we created Kuwaiti dialect language resources using Q8VaxStance, a vaccine stance labeling system for a large dataset of tweets. This dataset will fill this gap and provide a valuable resource for researchers studying vaccine hesitancy in Kuwait. Furthermore, it will contribute to the Arabic natural language processing field by providing a dataset for developing and evaluating machine learning models for stance detection in the Kuwaiti dialect. The proposed vaccine stance labeling system combines the benefits of weak supervised learning and zero-shot learning; for this purpose, we implemented 52 experiments on 42815 unlabeled tweets extracted between December 2020 and July 2022. The results of the experiments show that using keyword detection in conjunction with zero-shot model labeling functions is significantly better than using only keyword detection labeling functions or just zero-shot model labeling functions. Furthermore, using the Arabic language in both the labels and prompt or a mix of Arabic labels and an English prompt is statistically significant compared to using English in both the labels and prompt for the total number of generated labels evaluation metric. Finally, the best accuracy for Macro-F1 values were found in the experiments KHZSLF-EE4 and KHZSLF-EA1, with values of 0.83 and 0.83, respectively. And, for the total automatically labeled data evaluation metric, experiment KHZSLF-EE4 labeled 42,270 tweets, while experiment KHZSLF-EA1 was able to generate 42,764 labels.

This study has investigated the effect of nutrition labeling on consumer guilt, in situations where consumers ingest products that have an unhealthy image. The first investigation has examined the effects of types of nutrition labeling (adding healthy ingredients/reducing unhealthy ingredients). The second investigation has examined the influence of two goal-activation types(utilitarian and hedonic) and nutrition labeling on consumer guilt. The findings reveal that the utilitarian-goal-activation group felt less guilty about consuming a product with a reduced unhealthy ingredient than they did about consuming a product with an added healthy ingredient. In addition, there were interactions between the two variables, since consumer goal activation had more effect on consumer guilt about products with reduced unhealthy ingredients than on products with added healthy ingredients. Based on these results, the academic and practical implications of the present study are discussed. and proposes several directions for future research.

Accurate determination of protein localization, levels, or protein-protein interactions is pivotal for the study of their function, and in situ protein labeling via homologous recombination has emerged as a critical tool in many organisms. While this approach has been refined in various model fungi, the study of protein function in most plant pathogens has predominantly relied on ex-situ or overexpression manipulations. To dissect the molecular mechanisms of development and infection for Verticillium dahliae, a formidable plant pathogen responsible for vascular wilt diseases, we have established a robust, homologous recombination-based in situ protein labeling strategy in this organism. Utilizing Agrobacterium tumefaciens-mediated transformation (ATMT), this methodology facilitates the precise tagging of specific proteins at their C-termini with epitopes, such as GFP and Flag, within the native context of V. dahliae. We demonstrate the efficacy of our approach through the in situ labeling of VdCf2 and VdDMM2, followed by subsequent confirmation via subcellular localization and protein-level analyses. Our findings confirm the applicability of homologous recombination for in situ protein labeling in V. dahliae and suggest its potential utility across a broad spectrum of filamentous fungi. This labeling method stands to significantly advance the field of functional genomics in plant pathogenic fungi, offering a versatile and powerful tool for the elucidation of protein function.

In this paper, we present a framework that automatically labels Latent Dirichlet Allocation (LDA) generated topics using sentiment and aspect terms from COVID-19 tweets to help the end-users by minimizing the cognitive overhead of identifying key topics labels. Social media platforms especially Twitter are considered as one of the most influential sources of information for providing public opinion related to a critical situation like the COVID-19 pandemic. LDA is a popular topic modelling algorithm that extracts hidden themes of documents without assigning a specific label. Thus automatic labelling of LDA-generated topics from COVID-19 tweets is a great challenge instead of following the manual labelling approach to get an overview of wider public opinion. To overcome this problem, in this paper, we propose a framework named \texttt{SATLabel} that effectively identifies significant topic labels using \textit{top unigrams features of sentiment terms and aspect terms clusters from LDA generated topics} of COVID-19 related tweets to uncover various issues related to the COVID-19 pandemic. The experimental results show that our methodology is more effective, simpler, and traces better topic labels compare to the manual topic labelling approach.

Tumor-associated macrophages are a key component in the tumor microenvironment, secreting extracellular vesicles (EVs) such as exosomes and other various factors for intercellular communication. However, macrophage-derived EVs heterogeneity and their cytotoxicity to cancer cells has not been well understood. Here, we aimed to separately isolate various types of macro-phage-EVs by size exclusion chromatography (SEC) method and investigate EV transmission and cytotoxicity to oral cancer cells. For fluorescence-labeling of cellular and EV membranes, palmitoylation signal-fused GFP and tdTomato were expressed in THP-1 monocytic cells and HSC-3 oral cancer cells, respectively. We found that fluorescence-labeled EVs secreted by macrophages were highly transmissive to oral cancer cells than those from parental monocytic cells. In a co-culture system and conditioned medium (CM), a macrophage-secreted unidentified factor was cytotoxic to oral cancer cells. We fractionated macrophage-derived EVs by the SEC method and performed western blotting to characterize various EV types. Three fractions were characterized: small exosomes (EXO-S: < 50 nm) fraction containing HSP90α, HSP90β, CD63 (EV marker) and β-actin; large exosomes (EXO-L: 50-200 nm) fraction containing CD9 (EV marker) and HSP90β; large EVs (100-500 nm) fraction. Notably, the macrophage-derived small exosomes fraction was cytotoxic to oral cancer cells, while large exosomes and large EVs were not. There-fore, it was implicated that macrophage-derived small exosomes are cytotoxic with high trans-mission potential to cancer cells.

NASA’s ICESat-2space-borne photon-counting lidar mission is providing global elevation measurements that will provide significant benefits to a variety of bio-geoscience research applications. Given the novelty of elevation and the derived data products from the ICESat-2 mission, the research community needs software tools that can facilitate photon-level analyses to support product validation and development new analysis methods. Here, we describe PhotonLabeler, a free graphic user interface (GUI) for manual labeling and visualization of ICESat-2 Geolocated Photon data (ATL03). Developed in MATLAB, the GUI facilitates the reading and display of ATL03 Hierarchical Data Format (HDF) files, the manual labeling of individual photons into target classes of choice using a number of point selections tools and enables eventual saving of labeled data in ASCII format. Other capabilities include saving and loading of labeling sessions to manage labeling tasks over time. We expect labeled data generated using the application to serve two main purposes. First, serve as ground truth for validating various products from ICESat-2 mission, especially for study sites around the world that do not have existing reference datasets such as airborne lidar. Second, serve as training and validation data in the development of new algorithms for generating various ICESat-2 data products. We demonstrate the first use case through a validation case study for the land and vegetation product (ATL08), which provides canopy and terrain height estimates, over two sites. For the first site, located in northwestern Zambia, we used ICESat-2 ATL03 data acquired at night and for our second site in Texas, US, we used ATL03 data acquired during the day. The PhotonLabeler application is freely available as a compiled MATLAB binary to enable free access and utilization by interested researchers.

Overhead power transmission line icing (PTLI) disasters are one of the most severe dangers to power grid safety. Automatic iced transmission line identification is critical in various fields. However, existing methods primarily focus on the linear characteristics of transmission lines, employing a two-step process involving edge and line detection for PTLI identification. Nonetheless, these traditional methods are often difficult when confronted with challenges such as background noise or variations in illumination, leading to incomplete identification of the target area, missed target regions, or misclassification of background pixels as foreground. We propose a new iced transmission line identification scheme to overcome this limitation. In the initial stage, we integrate the image restoration method with image filter enhancement to restore the image's color information. This combined approach effectively retains valuable information and preserves the original image quality, thereby mitigating noise presented during the image acquisition. Subsequently, in the second stage, we introduce an enhanced multi-threshold algorithm to separate background and target pixels. After image segmentation, we enhance the image and obtain the region of interest (ROI) through connected component labeling modification and mathematical morphology operations, eliminating background regions. Our proposed scheme achieves an accuracy value of 97.72%, a precision value of 96.24%, a recall value of 86.22%, and the specificity value of 99.48% based on the average value of test images. Through object segmentation and location, the proposed method can avoid background interference, effectively solve the problem of transmission line icing identification, and achieve 90% measurement accuracy compared to manual measurement on the collected PTLI dataset.

As matrix representations of magic labelings of related hypergraphs, magic squares and their various variants have been applied to many domains. Among various subclasses, trimagic squares have been investigated for over a hundred years. The existence problem for trimagic squares with singly even orders and orders 16n has been solved completely. However, very little is known for the existence of trimagic squares with other even orders except for the only three examples and three families. We construct normal trimagic squares by using product constructions, row-square magic rectangles, and trimagic pairs of orthogonal diagonal Latin squares. We give a new product construction: For positive integers p, q, and r having the same parity other than 1, 2, 3, 6, if normal p × q and r × q row-square magic rectangles exist, then a normal trimagic square with order pqr exists. As its application, we construct normal trimagic squares of orders 8q^3 and 8pqr for all odd integers q not less than 7 and p,r ∈ {7, 11, 13, 17, 19, 23, 29, 31, 37}. Our construction can easily be extended to construct multimagic squares.

Trans fatty acids (TFA) are a concern for public health due to their negative impact on cardiovascular health. This study aimed to assess the levels of TFA and other fatty acids in popular edible oils and fried foods commonly consumed in Tanzania. A total of 57 samples from 38 brands of edible oils and 20 samples of fried street foods and fast foods were collected in Dar es Salaam, Tanzania. Capillary gas chromatography was used to determine TFA levels, while nutritional labeling and other fatty acid classes were also evaluated. Among the analyzed edible oils, 21% exceeded the recommended TFA levels, primarily found in popular brands of margarine, industrially refined sunflower oils, animal butter, and peanut butter. Only two food items surpassed the 2% TFA limit, but a majority of the foods had high levels of saturated fatty acids and a fatty acid profile similar to palm oil. Additionally, only 22% of the edible oils provided TFA content information. These findings indicate the persistence of TFA levels beyond recommended limits and inadequate TFA nutritional labeling in Tanzanian food products. Urgent interventions are necessary to reduce TFA levels and improve nutritional labeling practices to safeguard public health.

We introduce the Open Research Knowledge Graph Agriculture Named Entity Recognition (the ORKG Agri-NER) corpus and service for contribution-centric scientific entity extraction and classification. The ORKG Agri-NER corpus is a seminal benchmark for the evaluation of contribution-centric scientific entity extraction and classification in the agricultural domain. It comprises titles of scholarly papers that are available as Open Access articles on a major publishing platform. We describe the creation of this corpus and highlight the obtained findings in terms of the following features: 1) a generic conceptual formalism focused on capturing scientific entities in agriculture that reflect the direct contribution of a work; 2) a performance benchmark for named entity recognition of scientific entities in the agricultural domain by empirically evaluating various state-of-the-art sequence labeling neural architectures and transformer models; and 3) a delineated 3-step automatic entity resolution procedure for the resolution of the scientific entities to an authoritative ontology, specifically AGROVOC that is released in the Linked Open Vocabularies cloud. With this work we aim to provide a strong foundation for future work on the automatic discovery of scientific entities in the scholarly literature of the agricultural domain.

ATTO 565, a Rhodamine-type dye, has garnered significant attention due to its remarkable optical properties, such as a high fluorescence quantum yield, it is a relatively stable structure, and has low biotoxicity. ATTO 565 has found extensive applications in combination with microscopy technology. In this review, the chemical and optical properties of ATTO 565 are introduced, along with the principles behind them. Then, the functionality of ATTO 565 in confocal microscopy, stimulated emission depletion (STED) microscopy, single-molecule tracking (SMT) techniques, and fluorescence correlation spectroscopy (FCS) is discussed. These studies demonstrate that ATTO 565 plays a crucial role in areas such as biological imaging and single-molecule localization, thus warranting further in-depth investigation. Finally, we present some prospects and concepts for the future applications of ATTO 565 in the fields of biocompatibility and metal ion detection.

This research aims to understand the Cambodian green economy transition and analyze its strengths, weaknesses, opportunities, and threats (SWOT). The research showed that Cambodia has been significantly transitioning its green economy since 2009 as evidenced by the National Green Growth Roadmap. Cambodia also established the National Council for Green Growth in 2012 and upgraded it to the National Council for Sustainable Development in 2015 in alignment with 2030 Agenda for Sustainable Development of the United Nations. The members of the Council are from all the government’s ministries and relevant inter-ministerial councils/committees, which are significant for policymaking. All governors of the capital and provincial administrations are also its members, which are significant for supporting and implementing the works at the subnational levels. Under the structure of the Council’s Secretariat, Department of Green Economy is playing an important role in promoting green economic development in Cambodia by looking at national policies and strategic plans, undertaking projects, and mobilizing resources. Through coordination between this Department and UNIDO, Cambodia in 2022 became a Member of the United Nations Partnership for Action on Green Economy. The findings showed that Cambodia is transitioning its green economy at the right time, especially the new government’s Pentagonal Strategy strongly supported this green economy transition with a clear and strong political commitment. The national inter-ministerial council for sustainable development is also very significant for cross sectoral policy-making and implementing levels, inclusively for promoting green economic development.

This paper reports aligned activities to promote energy and resource-efficient construction practice in Vietnam. First, the governance framework is introduced, including government decrees and technical standards. Then, a laboratory with building physics measurement technology is designed and partly set up at the local partner Vietnam Institute for Building Materials (VIBM), which can be used to determine the essential characteristic values required for the implementation of energy standards. The basis for the prioritization of the characteristic values are the requirements of the National technical regulation on energy efficient buildings of Vietnam – QCVN09:2017/BXD. Furthermore, basic characteristic values from international standards are described, which can also be used for calculations to optimize the energy consumption of buildings. In order to be able to carry out transient hygrothermal computer simulations, special characteristic values were also included. These are particularly useful for the research and development of new building materials and the evaluation of entire buildings in terms of thermal and moisture protection In order to communicate the material properties to the market and enable informed decision making by the stakeholders, a labeling system has been developed. Thereby, the practical means for implementing governance instruments are provided, and the related technical applications are supported.

Reducing fertilizer-N rate, applying nitrification inhibitor (NI), and incorporating straw are widely recommended to improve N use efficiency of crops and decrease N losses. However, how these practices affect crop N uptake by regulating the fertilizer-N fate during the growing season is not well understood. We conducted a field 15N tracer study to explore the effectiveness of reduced N application, NI, and straw incorporation on fertilizer-N fates for maize cultivation in Northeast China. The following six treatments were used: 1) no N fertilization (control); 2) 200 kg urea-N ha−1 (100%N); 3) 200 kg urea-N ha−1 and 2400 kg dry-straw ha−1 (100%N+S); 4) 160 kg urea-N ha−1 (80%N, i.e., 20% reduction of N fertilization); 5) 160 kg urea-N ha−1 and nitrification inhibitor (Nitrapyrin) (80%N+NI); 6) 160 kg urea-N ha−1, nitrification inhibitor and 2400 kg dry-straw ha−1 (80%N+NI+S). Our results showed that the five N fertilization treatments yielded 16%–25% more grain and 39%–60% more crop N uptake than the control receiving no N fertilizer, but the differences among the five treatments were not statistically significant. In contrast to plant growth, changes in fertilizer-N fates in soil and plant pools were more pronounced. Compared with the 100%N treatment, 20% fertilizer-N reduction (80%N) decreased 15N concentration in topsoil and plant pools but increased the proportion of plant 15N recovery at harvesting (N use efficiency, NUE15N, 60% vs. 50%) (P &lt; 0.05). Nitrapyrin co-application (80%N+NI) delayed soil nitrification and increased soil 15N retention at harvesting (52% vs. 36%), thereby decreasing NUE15N significantly (P &lt; 0.05). Straw incorporation (100%N+S) decreased fertilizer-N retention compared with NI co-application because it promoted NUE15N significantly (P &lt; 0.05). The combined application of NI and straw (80%N+NI+S) was more conducive to fertilizer-N retention in soil. Our findings demonstrate that NI and straw additions are efficient strategies for stabilizing fertilizer-N in soils and potentially minimizing N loss; however, their effects on NUE15N vary and the related mechanism must be further clarified in long-term trials.

The illegal use of explosives by terrorists and other criminals is an increasing issue in public spaces, such as airports, railway stations, highways, sports arenas, theaters, and other large buildings. Security in these environments can be achieved by a set of different means, including the installation of scanners and other analytical devices to detect ultra-small traces of explosives in a very short time-frame to be able to take action as early as possible to prevent the detonation of such devices. Unfortunately, an ideal explosive detection system still does not exist, which means that a compromise is needed in practice. Most detection devices lack the extreme analytical sensitivity, which is nevertheless necessary due to the low vapor pressure of nearly all explosives. In addition, the rate of false positives needs to be virtually zero, which is also very difficult to achieve. Here we present an immunosensor system based on kinetic competition, which is known to be very fast and may even overcome affinity limitation, which impairs the performance of many traditional competitive assays. This immunosensor consists of a monolithic glass column with a vast excess of immobilized hapten, which traps the fluorescently labeled antibody as long as no explosive is present. In the case of TNT occurring, some binding sites of the antibody will be blocked, which leads to an immediate breakthrough of the labeled protein, detectable by highly sensitive laser-induced fluorescence with the help of a Peltier-cooled CMOS camera. Liquid handling is performed with high-precision syringe pumps and chip-based mixing-devices and flow-cells. The system achieved limits of detection of 1 pM (1 ppt) of the fluorescent label and around 100 pM (20 ppt) of the explosive 2,4,6-trinitrotoluene (TNT). The total assay time is less than 8 min. A cross-reactivity test with 5000 pM solutions showed no signal by PETN, RDX, and HMX. This immunosensor belongs to the most sensitive and fastest detectors for TNT with no significant cross-reactivity by non-related compounds.

The trafficking of illegal drugs by criminal networks at borders, harbors, or airports is an increasing issue in public health as these routes ensure the main supply of illegal drugs. The prevention of drug smuggling, including the installation of scanners and other analytical devices to detect ultra-small traces of drugs within a reasonable time frame, remains a challenge. The presented immunosensor is based on a monolithic affinity column with a large excess of immobilized hapten, which traps fluorescently labeled antibodies as long as the analyte cocaine is absent. In the presence of the drug, some binding sites of the antibody will be blocked, which leads to an immediate breakthrough of the labeled protein, detectable by highly sensitive laser-induced fluorescence with the help of a Peltier-cooled complementary metal-oxide-semiconductor (CMOS) camera. Liquid handling is performed with high-precision syringe pumps and microfluidic chip-based mixing devices and flow cells. The biosensor achieved limits of detection of 23 pM (7 ppt) of cocaine with a response time of 90 seconds and a total assay time below 3 minutes. With surface wipe sampling, the biosensor was able to detect 300 pg of cocaine. This immunosensor belongs to the most sensitive and fastest detectors for cocaine and offers near-continuous analyte measurement.
