The long term sustainability and longevity of the business life cycle depends on correct understanding of the contextual constituents, triggers and dynamics, pre-startup planning, strategy and financial forecasting where business plans are developed to understand and forecast project viability, pitch ideas and secure funding. At the same time there is a lack of one page startup business model. Identifying this need, a ‘startup business strategy frame’ has been developed through learnings and encompassing design thinking, the business canvas and other strategy models and approaches. It provides a logical sequential flow and connects value drivers on environmental, strategic including operational and financial levels of proposed startup in a sequential format. To arrive at the frame as a logical conclusion a website www.strategizestartup.com is developed to build startup business model and plans in a time and cost effective process. The website assists the startups at each stage through a work and guidebook. The website takes the user from creative ideation by charting the context to build the strategy and measuring the financial impact. This startup strategy model will alleviate the problems for the first and second stage entrepreneurs to ideate creatively and strategizing. It also eradicates the issues related to reading and understanding long plans which could be an exhaustive and cumbersome process.

Several design methods and principles have been presented so far, in order to guide the design of autonomous operations. Putting the required efforts into learning and using the methods for designing autonomous operations is a daunting task. Experiences so far have shown that the use of methods meant to the help the design process are often ignored. One reason could be that the design guidelines are too complex and contain much information often not relevant for the project at hand, and therefore there is no easy way to distinguish what is important from what is not. This is an issue that needs to be solved with our approach. In this article, the Autonomous Job Analysis (AJA) method is presented. The proposed methodology is created in order to guide the design of autonomous operations in maritime systems by breaking them down in to sub-operations in order to reveal challenges, needs and limitations regarding autonomous behavior. The canvas contains the categories of the AJA method on a single page format -the canvas- and each category is supported with questions to be asked during the design procedure, as well as example answers. We will describe the AJA method and the AJA canvas in detail, and present a use case scenario of an autonomous operation in order to show how they can be applied. The particular use-case is the design of an autonomous operation for the detection, inspection and tracking of a waste water plume.

The concept of biorefinery constitutes a significant contributing factor to the emerging transition towards a sustainable bioeconomy. In such a context, replacing oil and petrochemicals by biomass may involve several feedstocks, platforms, processes, technologies, as well as final products. This paper concentrates in the complex process of transferring the concept of biorefinery from laboratory to industry, and sheds light on the techno-economic, and complexity management dimensions involved in this endeavor. Towards this end, adopting a systems perspective, the paper presents a structured and comprehensive framework, comprising of the definition of the transformation process, business model development, technoeconomic assessment and strategic positioning and viability assessment, which may be employed to facilitate the engineering at-large and launch of a biorefining venture in a circular bioeconomy context. The framework is applied in the context of a biorefinery plant in a specific region in Southern Greece, which is based on the valorisation of olive mill wastewater (a ‘strong’ and quite common industrial waste in Mediterranean Basin), and produces biopolymers (PHAs) and bioenergy (H2).

Cerebellar ataxia, neuronopathy, and vestibular areflexia syndrome (CANVAS) is caused by biallelic pathogenic expansions, or compound heterozygosity with other pathogenic variants in the RFC1 gene. CANVAS is estimated to be underdiagnosed, both because of the lack of formal diagnostic criteria and molecular challenges that translate to lesser access and high cost of routine testing. Our aim was to address the need for making CANVAS genetic testing routine, by designing a streamlined two-step PCR consisting of a short-allele screening PCR and a confirmatory PCR with fragment capillary electrophoresis detection. Exome sequencing of RFC1 was additionally foreseen to resolve potential compound heterozygosity cases. Specificity of our approach was evaluated using ataxia patients with known non-CANVAS diagnoses, and optimized using Southern blot confirmed CANVAS patients. We evaluated our approach by testing patients consecutively referred for clinically suspected CANVAS using first the two-step PCR, followed by exome sequencing. Our approach was able to accurately identify negative and confirm positive cases in prospectively collected suspected CANVAS patients presenting with at least three typical clinical signs. The proposed testing approach provides an alternative method able to clearly distinguish between CANVAS negative and positive cases and can be easily incorporated into the genetic diagnostic laboratory workflow.
