A new approach for the preparation of polymer brush was developed via the union of controlled/“living” radical polymerization and click chemistry. The application of the approach realized the more quickly and accurate design and synthesis of the polymer brushes. A novel functional polymer brush of poly(acrylonitrile-g-glycidyl methacrylate) with tremendous application potential in the field of biology has been successfully synthesized via the new approach. The reaction conditions of click chemistry were optimized with the reaction time and the reactant ratio and judged from UV-vis spectra. Under the optimum of click chemistry, the novel functional polymer brushes were prepared. The GPC, FTIR analyses, 1H NMR spectrum and TGA were employed to ensure the successful synthesis of poly(acrylonitrile-g-glycidyl methacrylate) polymer brushes.

We previously showed that, based on the frequency and distribution of specific cell types, rainbow trout (RT) intestinal mucosa is divided in two regions that form a complex nonlinear 3D pattern and have a different renewal rate. This work had two aims. First, to investigate whether the unusual distribution of cell populations reflects a similar distribution of functional activities. To this end, we determined the protein expression pattern of three well defined enterocytes functional markers: Peptide Transporter 1 (PepT1), Sodium-Glucose/Galactose Transporter 1 (SGLT-1) and fatty acid-binding protein 2 (Fabp2). Second, to characterize the structure of RT intestinal stem cells (ISC) niche and to determine whether the different proliferative rates correlate with a different organization and/or extension of the stem cells population. We studied the expression and localization of well-characterized mammal ISC markers: LGR5, HOPX, SOX9, NOTCH1, DLL1 and WNT3A. Our results indicate that morphological similarity is associated with similar function only between the first portion of the mid-intestine and the apical part of the complex folds in the second portion. Mammal ISC markers are all expressed in RT but their localization is completely different suggesting also substantial functional differences. Finally, higher renewal rates are supported by a more abundant ISC population.

The present article addresses the long-standing problem of the polymer brush bilayers under stationary shear flow at non-linear response regime where the system gets a non-Newtonian fluid. The main idea behind this research would be the fact that the immense lubricity of the polymer brush bilayers originates from a global restructuring that takes place at large shear rates. It is shown here that physical quantities like, stress tensor, viscosity tensor, the friction coefficient and the chain extensions could become dependent on the shear rate at non-Newtonian regime. Apparently, the sub-linear scaling of the physical quantities at large shear rates is solely due to the fact that the chains stretch in the shear direction.

Hypercholesterolemia accelerates atherosclerosis, and extensive research has been undertaken to ameliorate this abnormality. Plant sterols have been shown to inhibit cholesterol absorption and lower plasma cholesterol level since the 1950s. This ingredient has recently been reappraised as a food additive that can be taken daily in a preclinical period to prevent hypercholesterolemia, considering that cardiovascular-related diseases are the top cause of death globally even with clinical interventions. Intestinal cholesterol handling is still elusive, making it difficult to clarify the mechanism for plant sterol-mediated inhibition. Notably, although the small intestine absorbs cholesterol, it is also the organ that excretes it abundantly, via trans-intestinal cholesterol efflux (TICE). In this review, we show a model where the brush border membrane (BBM) of intestinal epithelial cells stands as the dividing ridge for cholesterol fluxes, making cholesterol absorption and TICE inversely correlated. With this model, we tried to explain the plant sterol-mediated inhibitory mechanism. As well as cholesterol, plant sterols diffuse into the BBM but are effluxed back to the lumen rapidly. We propose that repeated plant sterol shuttling between the BBM and lumen promotes cholesterol efflux, and plant sterol in the BBM may disturb the trafficking machineries that transport cholesterol to the cell interior.
