Macroalgae and macroalgae-associated bacteria together constitute the most efficient metabolic cycling system in the ocean. Their interactions, especially the responses of macroalgae-associated bacteria communities to algae in different geographical locations, is mostly unknown. In this study, metagenomics was used to analyze the microbial diversity and associated algal polysaccharide-degrading enzymes on the surface of red algae among three remote regions. There were significant differences in the macroalgae-associated bacteria community composition and diversity among the different regions. At the phylum level, Proteobacteria, Bacteroidetes, and Actinobacteria had a significantly high relative abundance among the regions. From the perspective of species diversity, samples from China had the highest macroalgae-associated bacteria diversity, followed by those from Antarctica and Indonesia. In addition, in the functional prediction of the bacterial community, genes associated with amino acid metabolism, carbohydrate metabolism, energy metabolism, metabolism of cofactors and vitamins, and membrane transport had a high relative abundance. Canonical correspondence analysis and redundancy analysis of environmental factors showed that, without considering algae species and composition, pH and temperature were the main environmental factors affecting bacterial community structure. Furthermore, there were significant differences in algal polysaccharide-degrading enzymes among the regions. Samples from China and Antarctica had high abundances of algal polysaccharide-degrading enzymes, while those from Indonesia had extremely low abundances. The environmental differences between these three regions may impose a strong geographic differentiation regarding the biodiversity of algal microbiomes and their expressed enzyme genes. This work expands our knowledge of algal microbial ecology, and contributes to an in-depth study of their metabolic characteristics, ecological functions, and applications.

Marnaviridae viruses are abundant algal viruses that regulate the dynamics of algal blooms in aquatic environments. They employ a narrow host range because they merely lyse their own algal host species. This host-specific lysis is thought to correspond to the unique transmission mechanism of the Marnaviridae viruses. Here, we present the atomic structures of the full and empty capsids of Chaetoceros socialis forma radians RNA virus 1 built in 3.0 Å and 3.1 Å cryo-electron microscopy maps. The empty capsid structure and the structural variability provide insights on its assembly and uncoating intermediates. In conjunction with the previously reported atomic model of the Chaetoceros tenuissimus RNA virus type II capsid, we have identified the common and diverse structural features of the VP1 surface between the Marnaviridae viruses. We have also tested the potential usage of AlphaFold2 for structural prediction of the VP1s and a subsequent structural phylogeny for classifying Marnaviridae viruses by their hosts. These findings will be crucial for inferring the host-specific transmission mechanism in Marnaviridae viruses, which is decisive for monitoring and possibly regulating the occurrence and disappearance of algae blooms.

To explore the effect of lily polysaccharide (LP) on immune function and intestinal flora in immunosuppressive mice.The immunosuppressive model was established by intraperitoneal injection of cyclophosphamide (CTX) in mice, and the immunological indexes were measured. The cecal contents of mice were taken, and the DNA of intestinal flora was extracted for 16S DNA sequence analysis.Compared with CTX group, the body weight and thymus index of mice in each treatment group increased significantly, the liver index decreased significantly,the hepatocyte morphology, liver plate and hepatic sinus arrangement of mice in high and low concentration LP groups were relieved in varying degrees, and the contents of IgA and IgG in serum of mice in low concentration LP group and positive control group increased significantly; The contents of TNF-α and IL-6 in colon decreased, while the contents of IL-10 increased significantly. The contents of AST, ALT and MDA in liver decreased significantly, while the content of SOD increased significantly. Compared with the blank group, LP can improve the intestinal flora of immunosuppressive mice , mainly by increasing the abundance of beneficial bacteria such as s24-7, Bacteroides and butyrimionas and reducing the abundance of harmful bacteria such as Prevotella, dorea and adlercreutzia.

Inspired by the process of bone formation in living organisms, many studies have been conducted to develop organic-inorganic composite materials by generating calcium phosphate crystals within solutions or dispersions of polymers with appropriate functional groups. Bones are composite materials consisting of organic polymers (mainly Type I collagen), carbonated apatite, and water, with volume fractions 35-45%, 35-45%, 15-25%, respectively. Carbonated apatite in bone contributes to rigidity, while organic polymers and water contribute to toughness. The inorganic crystal, carbonated apatite, is a plate-shaped crystal with dimensions of 50 nm × 25 nm × 1-4 nm, generating a significant organic-inorganic interface due to its nanoscale size. This interface is believed to absorb externally applied forces to dissipate mechanical energy to thermal energy. Creating such nanometer-scale structures using top-down approaches is challenging, making bottom-up methods like co-precipitation of polymer and inorganic crystals more suitable. In this account, efforts to develop eco-friendly mechanical materials using biomass such as cellulose and starch based on the bottom-up approach to bone-like composites are described.

Chronic renal failure (CRF) is a major public health problem worldwide. In this work, we investigated the effects of a purified Laminaria japonica polysaccharide (LJP61A) on the renal function using adenine-induced CRF mice model. Results exhibited that adenine treatment caused serious renal pathological damages and elevation of serum creatinine and blood urea nitrogen of mice. However, these changes could be significantly reversed by the administration of LJP61A in a dose-dependent manner. Additionally, LJP61A could dramatically reduce the weight loss, improve the urine biochemical index, and regulate the electrolyte disturbance of CRF mice. These results suggested that the renal functions of adenine-induced CRF mice could be improved by LJP61A, which might be developed to a potential therapeutic agent for CRF patients.

The complex structure of the plant cell wall makes it difficult to use the biomass produced by biosynthesis. For this reason, the search for new strains of microorganisms capable of efficiently degrading fiber is a topic of interest. For these reasons, the present study aimed to evaluate both the microbiological and enzymatic characteristics of the fungus Curvularia kusanoi L7strain. For this, its growth in different culture media was evaluated. Wheat straw mineralization was evaluated by gas chromatography assisted by infrared spectroscopy. The production of endo- and exoglucanase, laccase and peroxidase enzymes in submerged solid fermentation of wheat and sugarcane bagasse were characterized. The strain efficiently mineralized raw wheat straw, showing a significant decrease in signals associated with cellulose, hemicellulose and lignin in the infrared spectra. High enzyme productions were achieved in submerged solid fermentation of both substrates, highlighting the high production of endoglucanases in sugarcane bagasse (2.87 IU/mL) and laccases in wheat (1.64 IU/mL). It is concluded that C. kusanoi L7 is an ascomycete with a versatile enzyme production that allows it to exhaustively degrade complex fibers such as raw wheat straw and sugar cane bagasse, making it a microorganism with great potential in the bioconversion of plant biomass.

The drastically increasing amount of plastic waste is causing an environmental crisis that requires innovative technologies for recycling post-consumer plastics to achieve waste valorization while meeting environmental quality goals. Biocatalytic depolymerization mediated by enzymes has emerged as an efficient and sustainable alternative for plastic treatment and recycling. A variety of plastic- degrading enzymes have been discovered from microbial sources. Meanwhile, protein engineering has been exploited to modify and optimize plastic-degrading enzymes. This review highlights the recent trends and up-to-date advances in mining novel plastic-degrading enzymes through state-of-the-art omics-based techniques and improving the enzyme catalytic efficiency and stability via various protein engineering strategies. Future research prospects and challenges are also discussed

Intensive crop production and extensive use of harmful synthetic chemical pesticides create numerous socio-economic problems worldwide. Therefore, sustainable solutions are needed for insect pest control, such as biological control agents. The fungal insect pathogen Beauveria bassiana has shown considerable potential as a biological control agent against a broad range of insects. The insights into virulence mechanism of B. bassiana is essential to show the robustness of its use. B. bassiana has several determinants of virulence, including the production of cuticle-degrading enzymes (CDEs), such as proteases, chitinases, and lipases. CDEs are essential in the infection process as they hydrolyze the significant components of the insect's cuticle. Moreover, B. bassiana has evolved effective antioxidant mechanisms that include enzyme families that act as ROS scavengers, e.g., superoxide dismutases, catalases, peroxidases, and thioredoxins. In B. bassiana, the number of CDEs and antioxidant enzymes characterized in recent years. The enzymatic activities are crucial for the biological control potential and significantly advanced our understanding of the infection mechanism of B. bassiana. This review focuses on the progress detailed in the studies of these enzymes and provides an overview of enzymatic activities and their contributions to virulence.

Watermelon fruits of different ploidy exhibit significant variations in texture. In order to investigate the primary factors influencing texture differences, this study was conducted to investigate the differences in fruit texture development using homozygous autogamous diploid (2X)lines and their autotriploid (3X) and autotetraploid (4X) lines 'Yixuan'. The research encompassed fruit development, flesh texture profile analysis (TPA), flesh cell wall polysaccharide content, and flesh cell microstructure analysis, specifically in 2X, 3X, and 4X watermelon fruits.The results showed that as watermelon fruits matured, several characteristics exhibited an ascending trend, including fruit weight, TSS (total soluble solids) content, rind hardness, flesh cell size, and cell wall polysaccharide contents such as crude fiber, cellulose, hemicellulose, lignin, and protopectin. Conversely, the flesh texture parameters and soluble pectin content of the flesh cell wall gradually declined. Notably, significant differences in fruit flesh texture and flesh cell structure were observed among fruits of different ploidy at the 32d. Specifically, 2X fruits displayed considerably lower rind hardness, reduced flesh texture (Hardness, Fracturability, Chewiness, Gumminess), and cell density compared to 3X and 4X fruits. Additionally, 2X fruits exhibited larger cell sizes than 3X and 4X fruits. However, no significant differences were observed in the content of flesh cell wall polysaccharide contents across various ploidy levels.These findings suggested that the variation in texture among watermelon fruits of different ploidy can be attributed to the size and arrangement of flesh cells. Importantly, this research provided a foundation for further exploration of the intrinsic regulatory factors and molecular mechanisms contributing to texture variation in polyploid watermelon fruits.

Oxidized sucrose (OS) reacts with amino group-containing polysaccharides, including chitosan, without catalyst, resulting in hydrogels entirely composed of carbohydrates. The presence of imine bonds with low structural stabilities and unreacted aldehydes in the structures of these hydrogels hinder their application as biomaterials. Therefore, herein, the chitosan hydrogels (CTSGs) obtained after the crosslinking of chitosan with OS were reduced using sodium borohydride to convert imine bonds to secondary amines and aldehydes to alcohols. Structures of CTSGs were comprehensively characterized by Fourier transform infrared and 13C nuclear magnetic resonance spectroscopies, and results implied that the degree of crosslinking (CR) depended on the OS feed amount used during CTSG preparation. Properties of CTSGs were significantly dependent on CR; with an increase in CR, thermal stabilities and dynamic moduli of CTSGs increased, whereas their swelling properties decreased. CTSGs exhibited antimicrobial properties against the gram-negative bacterium Escherichia coli, and their performances were also dependent on CR. Results indicated the potentials of CTSGs completely based on carbohydrates as antimicrobial hydrogels for various medical and pharmaceutical applications. We believe that this study will contribute to the development of hydrogels for application in the food, medical, and pharmaceutical fields.

Amylin, also known as islet amyloid polypeptide (IAPP), is a metabolic homeostasis-related hormone that is produced and released by the β cells of the pancreas, the same cells that produce insulin. Insulin-degrading enzyme (IDE) is a protease enzyme that plays an essential role in the breakdown and degradation of various peptides, including insulin and amylin. Direct binding & interaction between amylin and IDE is inextricably linked to the degradation of amylin, and research and development effort in this area is crucial to understand the potential therapeutic implications of disrupting the IDE-amylin interaction in the context of conditions where metabolic homeostasis needs to be regulated, such as diabetes and obesity. Here, this article incorporates currently available experimental complex structure of amylin and IDE, and delves deep into the interstructural biophysics underlying the binding interface of the two interacting partners. With a set of comprehensive structural biophysical analysis, this article identified an intriguing region of high electrostatic potential indicative of strong binding sites between the first N-terminal lysine (Lys1, K1) residue of amylin and Glu341 (E341) of IDE. This unique electrostatic hotspot presented herein paves the way for the rational design of drug-like small molecules that can selectively disrupt this interaction, offering a targeted therapeutic strategy for improved metabolic homeostasis, particularly for patients with diabetes and obesity.

In light of the hazardous effects of global warming brought on by climate change, bioethanol production is one of the key alternatives in which the world has expressed a great deal of interest. Two factors—indigenous production and price—are crucial for bioethanol to be appealing in Africa and, for that matter, Ghana. The objective of this research is to produce bioethanol from cheap lignocellulosic materials using onsite enzymes produced from cassava peels degrading fungi. Due to Ghana's prominent position as one of the leading cassava producers in the world, the study focused on utilizing cassava peels, which are underutilized in the country. The research was conducted in multiple stages. In the first stage, the peels were prepared by washing, drying, and manually crushing them using mortar and pestle. The second stage involved analyzing the chemical composition of the peels, including starch and insoluble fibre, which were determined as Acid Detergent Fiber (ADF), Neutral Detergent Fiber (NDF), and Acid Detergent Lignin (ADL). The results showed significant levels of cellulose (39.78%) and starch (31.21%), indicating that cassava peels are valuable raw materials for bioethanol production. The content of hemicellulose (21.11%) and lignin (3.84%) were also determined. In the third stage, using a Petri dish, three fungi (ICPF1, ICPF2, and ICPF3) were isolated from two different cassava peels. ICPF1 was only identified from fresh cassava peels (FCP), while all three fungi were identified from decayed cassava peels (DCP). Morphologically, these fungi were identified as Aspergillus niger, Aspergillus flavus, and Rhizopus stolonifer. The fourth stage focused on optimizing the enzyme activity of the three isolates for potential applications, with A. niger demonstrating the highest enzyme activity with a diameter of zone of clearance of 16 mm. Stage five involved optimizing the production of the onsite enzyme in a 50ml flask using A. niger, basal salt medium (BSM), and cassava peels as a substrate. The DNSA method was used to measure the absorbance of maltose and glucose at 540 nm for various substrate concentrations (1%, 3%, 5%, 8%, and 10%) at specific intervals of 2, 4, 6, 8, and 10 days with a spore concentration of 2.1 x 105 cells/ml. The maltose and glucose concentrations were calculated as 7.138mg/ml and 6.398 mg/ml, respectively, and the corresponding enzyme activity was determined as 4.759U/ml and 4.265U/ml. The optimal conditions of Day 4 and a substrate concentration of 10%, along with a fixed temperature of 30°C and a pH of 5.5, were used to prepare the onsite enzymes in a 500 ml flask for the fermentation process. The onsite enzymes were used for saccharification and Saccharomyces cerevisiae for fermentation under simultaneous saccharification and fermentation (SSF) process. A mixture of 20 ml of onsite enzymes and 1.5 g of S. cerevisiae were added to substrate concentrations of 5%, 10%, and 20%, and the ethanol concentration was analyzed daily for 7 days using Gas Chromatography (GC). The highest ethanol concentration (1.316%) was observed on Day 5 with a substrate concentration of 20%, while the lowest concentration (0.123%) was recorded on Day 1 with a substrate concentration of 5%.

Oxalate, a compound produced by many edible plants and as a terminal metabolite in the liver of mammals, is a toxin that has a detrimental role to human health. Humans and other mam-mals do produce the enzymatic machinery to degrade oxalate. However, numerous oxa-late-degrading bacteria reside in the mammalian gut and thus provide an important function for hosts. The current review focuses on the environmental factors that influence the efficacy of pro-biotic oxalate-degrading bacteria, relative to oxalate metabolism. We describe the mechanism of oxalate catabolism and its consumption by obligate and facultative anaerobic oxalate-degrading bacteria, in both in vitro and in vivo environments. We also explore the environmental variables that impact oxalate degradation. Studies on single species degrade oxalate have not shown a strong impact on oxalate metabolism especially in high oxalate conditions such as consumption of foods high in oxalate (such as coffee and chocolate for humans or halogeton in animal feed). Considering effective variables which enhance oxalate degradation could be used in application of effective probiotic as a therapeutic tool in individuals with hyperoxaluria. This study indicates probiotics can be considered a good source of naturally occurring oxalate degrading agent in human colon

Chlorella vulgaris has considerable promise as a raw material for biofuel production since it has a high productivity per unit area, has a minimum influence on the environment, and does not significantly affect food security. Nevertheless, the low fatty content of their biomass poses a considerable economic obstacle for commercialization. Increasing the amount of biomass and lipids produced by these algae is essential for improving the economic feasibility of using them as biofuel sources. This review highlights the significance of identifying appropriate algal strains, namely those found in local environments, and utilising mutagenesis and genetic engineering techniques to create 'platform strains'. Prior endeavours have primarily concentrated on altering environmental and dietary parameters to enhance the production of biomass and lipids. Here, we review a scientific literature that examines biotechnological approaches and develops methodologies designed to increase lipid production, emphasising the importance of aligning engineering efforts with breakthroughs in DNA manipulation tools. In addition, the review research evaluates the present economic and commercial situation of algal biorefineries, including any related disadvantages. In summary, this thorough research highlights the importance of using creative methods to tackle the intricacies of lipogenesis and enhance the economic viability of producing biofuels from algae.

The stability and harmony of ecological niches rely on intricated interactions between their members. During evolution, organisms have developed the ability to thrive in different environments taking advantage of each other’s metabolic symphonies. Among them, microalgae are a highly diverse and widely distributed group of major primary producers whose interactions with other organisms play essential roles in their habitats. Understanding the basis of these interactions is crucial to control and exploit these communities for ecological and biotechnological applications. The green microalga Chlamydomonas reinhardtii, a well-established model, is emerging as a model organism for studying a wide variety of microbial interactions with ecological and economic significance. In this review, we bring together and discuss current knowledge that points to C. reinhardtii as a model organism for studying microbial interactions.

The introduction of organic and inorganic substances to the environment is a result of human activities such as agriculture, domestic and industrial wastewater which leads to pollution. Treatment processes of these wastewaters are being conducted globally to eliminate easily settled materials and recover nutrients in an attempt to release clear and apparently clean effluent into natural waters. Lack of removing inorganic nitrogen and phosphorus nutrients is the greatest cause of eutrophication in water bodies which inhibits the life of other organisms as well as pose a threat to human life and loss of the economy. Different technologies have been applied and are being developed to recover nutrients as well as heavy metals from wastewater to meet the permissible limits before discharging effluents. Wastewater treatment using microalgae offers an opportunity to provide tertiary bio-treatment and production of valuable biomass. Microalgae use the available inorganic nitrogen and phosphorus for their growth which are then harvested for various uses. Additionally, they have the ability to remove heavy metals and some toxic compounds. The main specific microalgae species in this study is the Chlorella sorokiniana with the Aequorea victoria jellyfish This paper reviews some of the wastewater treatment processes and focus on the use of microalgae and some of the shortcomings of the technologies and how they can be improved to achieve maximum nutrient recovery economically with low energy demand.

The sulfated polysaccharide (SP) was extracted from Gracilaria edulis and tested for its ability to protect against carbon tetrachloride (CCl4)-induced liver injury in Wistar rats. The chemical composition of the sulfated polysaccharide, documented 12.90% of yield, 83.07% of carbohydrate, 0% of protein, 15.02% of ash, and 2.09% of moisture, and the carbon, hydrogen, nitrogen, and sulfur content were 25.90%, 4.28%, 2.65%, and 3.61%. SP has a molecular weight of 41kDa. The monosaccharide and structural properties of SP were investigated using GC-MS, FT-IR, and 1H-NMR spectral analysis. In rats, CCl4 produced hepatotoxicity, as evidenced by an increase in serum ALT, AST, and ALP levels and a decrease in ALB levels. The SP treated showed decreases in ALT, AST and ALP levels, as well as an increase in ALB levels. SOD, catalase, GPx, and GSH levels were suggestively lower in CCL4-induced rats compared to normal control rats. SOD, catalase, GPx and GSH levels in the SP-treated rats, on the other hand, improved to normal levels. The rat liver tissue treated with SP recovered well from hepatic injury. SP can be considered an alternative in the treatment of liver injury or as preventive medicine.

Numerous innovative advancements in dressing technology for wound healing have emerged. Among the various types of wound dressings available, hydrogel dressings, structured with a three-dimensional network and composed of predominantly hydrophilic components, are widely used for wound care due to their remarkable capacity to absorb abundant wound exudate, maintain a moisture environment, provide soothing and cooling effects, and mimic the extracellular matrix. Composite hydrogel dressings, one of the evolved dressings, address the limitations of traditional hydrogel dressings by incorporating additional components, including particles, fibers, fabrics, or foams, within the hydrogels, effectively promoting wound treatment and healing. The added elements enhance the features or add specific functionalities of the dressings, such as sensitivity to external factors, adhesiveness, mechanical strength, control over the release of therapeutic agents, antioxidant and antimicrobial properties, and tissue regeneration behavior. They can be categorized as natural or synthetic based on the origin of the main components of the hydrogel network. This review focuses on recent research on developing natural polysaccharide-based composite hydrogel wound dressings. Their preparation and composition, the reinforcement materials integrated into hydrogels, and therapeutic agents are also explored. Furthermore, their features and the specific types of wounds where applied are discussed as well.

Harmful algal blooms are increasing globally and pose serious health concerns releasing cyanotoxins. Microcystin-LR (MC-LR), one of the most frequently produced cyanotoxins, has recently been detected in aerosols generated by the normal motions of affected bodies of water. MC-LR aerosol exposure has been linked to a pro-inflammatory influence on the airways of mice, however little is understood about the underlying mechanism or the potential consequences. This study aimed to investigate the pro-inflammatory effects of aerosolized MC-LR on murine airways. C57BL/6 and BALB/c mice were exposed to MC-LR aerosols, as these strains are predisposed to type 1/type 17 and type 2 immune responses, respectively. Exposure to MC-LR induced granulocytic inflammation in C57BL/6 but not BALB/c mice, as observed by increased expression of cytokines MIP-1α, CXCL1, CCL2, and GM-CSF compared with their respective vehicle controls. Furthermore, the upregulation of interleukins IL-17A and IL-12 is consistent with Th1 and Th17 driven type 1/type 17 inflammation. Histological analysis confirmed inflammation in the C57BL/6 lungs, with elevated neutrophils and macrophages in the bronchoalveolar lavage fluid and increased pro-inflammatory oxidized lipids. In contrast, BALB/c mice showed no significant airway inflammation. These results highlight the ability of aerosolized MC-LR to trigger harmful airway inflammation, requiring further research.

Seafood, especially from the ocean, is now seen as a greener and more sustainable source of protein causing an increase in its demand. This has also led to people making choices towards seafood as a replacement of carbon intensive protein sources. As a result, the demand for seafood is growing, and the Aquaculture industry is required to increase their produce while keeping the produce safe and sustainable. There are many challenges faced by the aquaculture industry in meeting these increased demands. One such challenge is the presence of harmful algal blooms (HAB) in the ocean which can have a major impact on aquatic life. In this paper we look at the impact of this challenge on aquaculture and the mitigating strategies. We will focus on Abagold Limited, a land-based marine aquaculture business that specializes in large scale production of abalone (Haliotis midae) based in Hermanus, South Africa. HABs are considered a threat to commercial scale abalone farming along the South-African coastline and requires continuous monitoring. The most recent HAB was in February-April 2019, the area experienced a severe red-tide event with blooms of predominantly Lingulodinium polyedrum. We present some of the mitigation strategies employing digital technologies for future proofing the industry.

Since 1998, when Jim van Etten’s team initiated its characterization, Paramecium bursaria Chlorella virus 1 (PBCV-1) had been the largest known DNA virus, both in terms particle size and genome complexity. In 2003, The Acanthamoeba-infecting Mimivirus unexpectedly superseded PBCV-1, opening the era of giant viruses, i.e. with virions large enough to be visible by light microscopy and genomes encoding more proteins than many bacteria. During the 15 following years, the isolation of many Mimivirus-relatives, have made the Mimiviridae one of the largest and most diverse family of eukaryotic viruses isolated from aquatic environments. Metagenomic studies keep suggesting that many more remain to be isolated. As Mimiviridae members are found to infect an increasing range of phytoplanckton species, their taxonomic position compared to the traditional Phycodnaviridae (i.e. etymologically “algal viruses”) became a source of confusion in the literature. Following a rapid history of the key discoveries that established the Mimiviridae family, we describe its current taxonomic structure and propose a set of operational criteria to help in the classification of future isolates.

The overarching aim of this paper is to investigate the lake ecosystem response to different drivers over a long term period by a paleolimnological study in Lake Narlay (46°64N, 5°91E) located in the Jura Mountains of France. It is a small, hard-water lake with a maximum water depth of 40 m and extended anoxic condition of the bottom water. Previous results on sediments analysis have documented a differential response of the lake to the environmental changes that occurred in AD 1600 when major shift in the trophic reliance on methane of the benthic food web were observed. From 1920 with intensification of modern agriculture, animal farming and the construction of a cheese making facility, the lake become eutrophic, with Oscillatoria rubescens bloom. However, the lake showed pronounced changes in an older period that remained unanswered. In this paper we aim at reconstructing in more detail the limnological conditions of this Lake over the last 1200 yrs. using combined analyses of specific algal carotenoids and subfossil diatom remains. A comparison with other proxies (chironomid, pollen, and instrumental climatic reconstruction) will be used to better identify, between the complex combination of climate and anthropogenic pressure, the driving factors that determined the ecological trajectory of Lake Narlay.

In winter 2008-2009, Lake Occhito, a strategic multiple-uses reservoir in South Italy, was affected by an extraordinary Planktothrix rubescens bloom. P. rubescens is a filamentous potentially toxic cyanobacterium which has recently colonized many environments in Europe. A number of studies is currently available on the use of remote sensing techniques to monitor different fresh water cyanobacteria species. By contrast no specific applications are available on the remote sensing monitoring of P. rubescens. In this paper we present a specific algorithm, based on Water Leaving Reflectances (WLR) from MERIS data, atmospherically corrected using the Aerosol Optical Thickness (AOT) retrieved by MODIS data, to detect P. rubescens blooms. The high accuracy in AOT data, provided by MOD09 surface reflectance product, at 1km spatial resolution, allowed obtaining a good correlation between the WLR and the P. rubescens chlorophyll-a concentrations measured in the field, through multiple stations fluorometric profiles. A modified Normalized Difference Chlorophyll index (NDCI) algorithm is presented. The performance of the proposed algorithm has been successfully compared with other specific algorithms for turbid productive waters. We demonstrated how important is to verify the spectral behaviour of bio-optical parameters in order to develop an ad hoc algorithm that better performs with respect to standard algorithms.

Polysaccharide from Ficus carica L. (FCP) exert multiple biological activities, as a biological macromolecule, the available knowledge about the specific structures and mechanisms of biological activity of purified 'Brunswick' fig polysaccharide is currently limited. In the present study, by chemical purification and characteristics identified via specific instruments, and then the impact of FCP on the immunomodulation activity in vitro and in vivo was examined. Structural char-acteristics showed that the molecular weight of the FCP was determined to be 127.5 kDa, the primary monosaccharides present in the FCP were galacturonic acid (GalA), arabinose (Ara), galactose (Gal), rhamnose (Rha), glucose (Glc), and xylose (Xyl) at a ratio of 0.321:0.287:0.269:0.091:0.013:0.011. Based on the investigation of in vitro immunomodulatory ac-tivity, FCP was found to stimulate the production of NO, TNF-α, and IL-6 and increased the pi-nocytic activity of macrophages. Further analysis revealed that FCP activated macrophages by interacting with toll-like receptor 4 (TLR4). Moreover, the in vivo test results indicate that FCP showed significant increase serum pro-inflammatory factors in immunosuppressed mice. Overall, this study suggests that FCP has the potential to be utilized as a novel immunomodulator in the pharmaceutical and functional food industries.

The structure characterization of two new polysaccharides from Agrocybe aegerita (AA-P) and Hygrophorus olivaceoalbus (HO-P) by HPGPC, GC-MS, NMR and FT-IR indicated that AA-P was composed of Galactose, Glucose and Arabinose, in the ratio of 3:2:1. Its skeleton structure was consisted of (1→4)-Arap, (1→4,6)-Glup and (1→6)-Galp with one branched chain. The HO-P was consisted of mannose, galactose and glucose in a ratio of 1:1:2. Its skeleton structure was consisted of (1→6)-Galactose residues, (→1)-glucose residues, (1→4)-glucose residues and (1→4,6)-D-mannose residues. There were two branched chains connected to the main chain. AA-P and HO-P had the best stimulation effect on B cells and RAW264.7 cells, respectively, and could both mainly by impacting and reducing G0/G1 phase which lead to a significant proliferation of B cells, T cells and RAW264.7 cells. In addition, AA-P and HO-P could significantly promote the secretion of TNF-α from T cells, the secretion of IgA, IgD, IgE, IgG and IgM from B cells, and the secretion of TNF-α from RAW264.7 cells, but neither of them could impact the secretion of IL-1β from RAW264.7 cells.

A new one-pot technique for preparation of polysaccharide-based Pd- and Pd-Ag nanocatalysts by sequential supporting natural polymer (2-hydroxyethyl cellulose (HEC), chitosan (Chit), pectin (Pec)) and metals on zinc oxide was developed. The nanocatalysts based on polysaccharide were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), Brunauer–Emmett–Teller (BET), infrared spectroscopy (IRS), X-ray powder diffraction (XRD), X-ray photoelectron spectra (XPS) and elemental analysis methods. The catalyst characterization results indicated complete adsorption of polysaccharide and metal ions on zinc oxide, forming polymer-stabilized Pd nanoparticles of ~2 nm in size, evenly distributed on the support surface. The catalysts were studied in the hydrogenation of 2-hexyn-1-ol under mild conditions (0.1 MPa, 40 °C). The catalysts demonstrated nearly the same conversion of 2-hexyn-1-ol. The selectivity to cis-hexen-1-ol of the catalysts decreases in the following order: 0.5%Pd-Ag(3:1)HEC/ZnO>0.5%Pd-Ag(3:1)Pec/ZnO>0.5%Pd-Ag(3:1)Chit/ZnO. The optimum reaction temperature and catalyst loading for the Pd-Ag catalysts modified with HEC and Chit have been determined (40 °C, 0.05 g).

The present study provides new insights into the growth of brown algal cell wall by showing that cell wall polysaccharide plays an important role in the process of growth consider- ing the physicochemical characteristic of young and old Cladosiphon okamuranus. To determine its structural variation in detail, the cell wall was sequentially fractionated into five fractions: hot water (HW), ammonium oxalate, hemicellulose-I (HC–I), HC-II, and cellulose, and analyzed physicochemically. Results showed that almost 80% of the total recovery cell wall from both young and old thalli was HW and HC-I contained mainly fucoidan composed of Fuc, GlcA, and sulfate in molar ratios of 1.0: 0.3: 0.6~0.7 and 1.0: 0.3: 0.2~0.3, respectively. Fucoidan in HW was a highly sulfated matrix polysaccharide abundance in young thalli, while fucoidan in HC-I was rich in old thalli and functions as hemicellulose in land plants, crosslink with cellulose and strengthen the cell wall. We found that HW and HC-I were particularly involved in the growth and strength of old thalli appeared to be due to the deposition of HC-I and reducing of water content during the growth process.

Agaricus bisporus is well known as a source of polysaccharides that could improve human health. The objective of this study was to explore anti-obesity effect of A. bisporus extract (H2Oslim®) abundant in polysaccharides and its underlying mechanism. Pancreatic lipase inhibitory activity in vitro was determined after treatment with H2Oslim® and chitosan. Treatment with H2Oslim® and chitosan significantly decreased pancreatic lipase activity. Five-week-old male SD rats were randomly divided into three groups for acute feeding with vehicle, H2Oslim® at 80 mg/kg body weight (BW)/day, and H2Oslim® at 160 mg/kg BW/day. H2Oslim® does-dependently increased plasma lipid clearance in an oral lipid tolerance test. Five-week-old male C57BL/6N mice were fed a control diet (CD), a high fat diet (HFD), an HFD with H2Oslim® at 80 mg/kg BW/day, H2Oslim® at 160 mg/kg BW/day, or chitosan at 160 mg/kg BW/day for eight weeks. HFD-fed mice showed significant increases of body weight, fat mass, white adipose tissue, average lipid droplet size, and serum levels of glucose, triglyceride, ALT, and AST compared to those in the CD group. However, H2Oslim® or chitosan administration ameliorated these increases. H2Oslim® or chitosan signifi-cantly reduced dietary efficiency and increased fecal excretion levels of lipid, triglyceride, and total cholesterol. These in vitro and in vivo findings suggest that H2Oslim® might act as an anti-obesity agent by inhibiting pancreatic lipase-mediated lipid absorption, at least in part.

Selenium (Se) is an essential trace element for human beings and Se supplementation is of great significance. In this paper, high selenized Gastrodia elata Blume Polysaccharides (SeGEP) were obtained through pre-sulfation before conducting the selenylation modification process, which owned a maximum Se content reach to 2.29%. UV, FT-IR, NMR, SEM, EDX, HPLC, size-exclusion chromatography and periodate oxidation analysis showed that the seleninic group had been successfully introduced into GEP with the substitution position at C-6 of glucose. Immunostimulating assays were conducted in vitro and in vivo on RAW 264.7 cells and cyclophosphamide (CTX)-induced mice model, respectively. The results showed that Se-GEP enhanced RAW264.7 cells proliferation, cell phagocytosis, and secretion of nitric oxide (NO) and interleukin (IL)-1β factors. In addition, Se-GEP enhanced the immunologic activity of CTX-treated mice by increasing the spleen and thymus indices, enhancing the phagocytic function, and activating both Th1 and Th2 lymphocytes. The current outcome also suggested that Se-GEP with covalently bonding Se group possessing increased safety and has a potential dietary selenium nutrient for improving human immunity.

The Vaal Dam catchment, which is the source of potable water for Gauteng province, holds various human activities including mining, urbanization, agricultural and industrial activities coupled with high fluxes of nutrients from different areas within the catchment. This situation led to an enhancement of Harmful Algal Blooms (HABs) in the dam reservoir. Phosphorus and Nitrogen are the limiting nutrients for HABs in many eutrophic aquatic systems when their concentrations are raised in such waters. In this study, multi-time-series plot types and scutter plots were analysed to reveal the relationship between HABs proxy chlorophyll-a (Chl-a) and the potential nutrients. Also (Chl-a) data extracted from landsat-8 satellite images were visualised to show the spatial distribution of HABs in the reservoir. The results showed that the Vaal Dam HABs productivity is a function of the total phosphorus (TP) and the organic nitrogen (KJEL_N) levels, which were positively correlated with chlorophyll-a (Chl-a) concentration. The long-term analysis of Chl-a in-situ data (1986 – 2022) measured at one point within the reservoir showed an average of 11.25μg/l but at some stochastic dates, the Chl-a concentration jumped to very high values reaching the maximum value of 452.8μg/l. Such high values are associated with high records of TP and KJEL_N concentrations at the same dates which clearly reveals their effect on the productivity levels. The decadal time series and its trend analysis showed that the average productivity level of Chl-a increased during the studded period from 4.75μg/l in the first decade (1990 – 2000) to 10.51μg/l in the second decade (2000 – 2010) reaching 16.7 during the last decade (2010 – 2020). The increasing averages of the decadal values are associated with the increasing decadal average of its driving factors, TP from 0.1043 to 0.1096 to 0.1119mg/l for the three decades, respectively and KJEL_N from 0.80mg/l in the first decade to 1.14mg/l in the last decade. The decadal average of temperature showed no significant increase, 17.9 °C in the first decade and 18 °C in the last decade which suggests that temperature is not the only enhancing factor of the HABs productivity in the Vaal Dam. The satellite data analysis during the last decade revealed that the HABs spatial dynamics is a function of the dam geometry and the levels of the discharges from its two feeding rivers. Higher concentrations were observed where the reservoir is meandering and within the areas of restricted water circulation.

Currently, algae arouse a growing interest in the pharmaceutical and cosmetic area due to the fact that they have a great diversity of bioactive compounds with the potential for pharmacological, cosmetic, and nutraceutical applications. Many of these bioactive compounds are secondary metabolites whose amounts in the algae vary with varying environmental conditions. Free radicals and other active oxygen derivatives are recognized as a natural by-product of aerobic metabolism. However, reactive oxygen species directly participate in mechanisms related to various pathological states such as cancer, diabetes, atherosclerosis, Alzheimer's, and Parkinson's, among others. Diabetes mellitus (DM) is a metabolic disease resulting from changes in glucose metabolism and/or deficient production/action of insulin. This review has as its main objective to reveal the potential antioxidant and antidiabetic capacity of algae extracts.

Background: The effect of rice bran arabinoxylan compound (RBAC), a plant-based immunomodulator, on the quality of life (QoL) in cancer patients and underlying physiological pathways remains unclear. Trial design: The RBAC-QoL study is a double-blind, randomised, controlled pilot feasibility study. The aim is to determine RBAC’s effects on QoL and the associated action mechanisms. Primary outcomes are the EORTC QLQ-C30 functional, symptom, and global QoL scores with inflammatory, nutritional, and cytokine parameters as secondary and exploratory outcomes. Methods: Recruitment targets adults diagnosed with solid organ tumours (≥ stage II) undergoing active treatment in several outpatient centres in New South Wales, Australia. Interventions are RBAC or matched placebo at 3g/day for 24 weeks allocated through stratified randomisation with participants, oncologists, and data collectors blinded. Data is collected from five study visits six weeks apart. The trial is ongoing. An interim intention-to-treat analysis was performed using repeated measure ANOVA with pairwise comparisons where statistical significance is observed and adjusted with covariates. Results: Global QoL scores from currently available data (n = 16; RBAC = 7, placebo = 9) were statistically different between groups (F[1,8] = 8.6, p = 0.019, eta2[g] = 0.267). Pairwise comparisons found significant differences at week 6 (p = 0.032, Cohen’s d = 1.454) and marginally at week 12 (p = 0.069, d = 1.427). Age-adjusted analysis showed a continuous upward trend in QoL improvement over time with RBAC, while the placebo group did not deviate from baseline QoL. Significant elevations of serum white blood cell count (week 18) and total protein (weeks 12 and 18) were detected in the RBAC group compared to placebo. The total protein levels correlated highly with white blood cell count (Pearson’s r = 0.539, p < 0.001) and moderately with the global QoL scores (r = 0.338, p = 0.01). No intervention-related adverse events were reported in both groups. Conclusions: RBAC improves QoL beyond placebo during active cancer treatment, possibly through the immuno-nutritional pathway. Clean, fresh, and safe drinking water is essential to human health and well-being. Occasionally, chemical pollutants taint surface water quality used for consumption. Microcystins (MCs) are toxic heptapeptides produced by freshwater cyanobacteria. These secondary metabolites can reach hazardous concentrations, impairing surface drinking water supplies. Inconsistent screening of MCs is not uncommon in Florida waters as no provisional guidance value is established to protect public health. The occurrence of MCs in Lake Manatee and Lake Washington was monitored over the potential peak algae bloom season (June-August). An indirect competitive enzyme-linked immunosorbent assay (icELISA) quantified total MCs in two drinking water systems. Varied concentrations occurred between June and July, whereas concentrations peaked in August. Overall, MC prevalence was higher in Lake Manatee than Lake Washington. Colorimetric assays measured phosphate and nitrite in environmental water samples. Phosphate and nitrite concentrations strongly correlated with total MCs (p < 0.01). The results indicate the intrinsic nature of environmental MCs in surface drinking water supplies and the need to examine hepatotoxin dynamics to preserve drinking water quality in community served areas.

The procedures for the extraction and separation of lipids and nutraceutics from microalgae using classic solvents have been used many times. However, these production methods usually require expensive and toxic solvents. Based on our studies involving the use of eco-sustainable methodologies and alternative solvents, we select ethanol (EtOH) and cyclopentyl methyl ether (CPME) for extracting bio-oil and lipids from algae. Different percentage of EtOH in CPME favors the production of an oil rich of SFA useful to production biofuel or rich of compounds bioactive. The proposed method for obtain a rich extract of saturated or unsaturated fatty acids from dry algal biomass is disclosed is eco-friendly and allows a good extraction yield. The method is compared both in extracted oil percentage yield and in extracted fatty acids selectivity to extraction by supercritical carbon dioxide.

Rapid haemostasis during surgery is essential when one wants to reduce the duration of opera-tions, reduce the need for transfusions, and above all when one wants to achieve better patient management. The use of haemostatic agents, sealants and adhesives improves the haemostatic process by offering several advantages, especially in vascular surgery. These agents vary widely in their mechanism of action, composition, ease of application, adhesion to wet or dry tissue, im-munogenicity and cost. The most used are cyanoacrylate-based glues (Glubran 2) or polysaccharide hydrogel-microsphere powder (AristaTMAH). This work is based on a retrospective study carried out on a sample of patients with different vascular diseases (FAV, pseudoaneurysm and PICC application) in which two different haemostatic sealants were used. The aim was to assess the safety, the advantages and the ability of both sealants to activate the haemostatic process at the affected site, also in relation to their chemical-physical characteristics. The obtained results showed that the application of Glubran 2 and AristaTMAH as surgical wound closure systems is effective and safe as the success achieved was ≥94% on anastomoses of FAV; 100% on stabilization of Picc catheters; ≤95% on pseudoaneurysms.

Cyclodextrin (CD) is made up of 7 glucose molecules linked in a ring, creating a cone shape. Its interior can house hydrophobic molecules while the exterior is hydrophilic enough to interact with water molecules. Until now, this feature has been used for the development of active and intelligent packaging applied to food, interacting with the product or its environment to improve one or more aspects of its quality or safety. As well, it is used to provide information on monitoring the time in which food is optimal for consumption, an aspect considered very important for the consumer and for the merchant. A known problem with some of these types of packaging is that the bioactive compounds that are housed in the CD can interact with food and can modify its taste, smell, color and/or texture. Therefore, this review will focus on discerning which packaging is most appropriate for each situation.

Due to their increased surface area, extent of swelling and active substance loading capacity and flexibility, nanogels made from natural and synthetic polymers have gained significant interest in the scientific and industrial areas. Especially, customized design and implementation of non-toxic, biocompatible, and biodegradable micro/nano carriers makes their usage very feasible for a range of biomedical applications, including drug delivery, tissue engineering, and bioimaging. The design and application methodologies of nanogels have been outlined in this review. Additionally, the most recent advancements in nanogel biomedical applications have been discussed, with a particular emphasis on applications for the delivery of drugs and biomolecules.

AbstractLarge numbers of urban lakes are facing issues related to cyanobacteria, commonly known as blue-green algae, resulting in significant threats to humans and animal health due to their production of microtoxins and Anatoxin-A. These toxins induce acute effects including gastrointestinal distress, and respiratory problems, underscoring the need to protect urban lakes to safeguard community health and ecosystem well-being. A comprehensive monitoring program is a critical step to address cyanobacteria-related issues. Monitoring results of dissolved phosphorous, nitrates(N), Total Kjeldahl Nitrogen (TKN), total phosphorous (TP), TSS, and Escherichia coli (E. coli) concentrations, are used to understand the importance of different pathways events during dry conditions (no prior rainfall) and wet events (rainfall during or proceeding monitoring events) plus a microbial source tracking procedure, to understand impacts on growth characteristics of cyanobacteria and microtoxins. The Mann-Kendall Trend Test is used to characterize trends of various parameters (using transitions of ‘wet’ and ‘dry’ to the Lake and within the Lake), to identify the impact of various management strategies and/or the impact of various sources from overland land uses for a) wet events to the Lake, b) dry events to the Lake, c) wet events in the Lake, and d) dry events within the Lake. Microbial source tracking methods are used along the shoreline of Fairy Lake to interpret elevated levels of fecal pollution and E. coli bacteria in water samples, aiding in the identification of pollution sources. Blue-green algae have been observed at least once each year from 2018 to 2023. Microtoxins and Anatoxin-A were present in nine samples with a maximum of 1.72 mg/L and 0.02 mg/L, respectively. Frequent beach closures have occurred every year, raising public concerns regarding both ecological and recreational aspects for Fairy Lake.

The wide metabolic diversity of microalgae, their fast growth rates, and cost-effective production make these organisms highly promising resources for a variety of biotechnological applications, addressing critical needs in industry, agriculture, and medicine. The utilization of microalgae in consortia with bacteria is proving to be valuable in different biotechnological fields, including treating various types of wastewaters, producing biofertilizers, and extracting various products from their biomass. Monoculture of the microalgae Chlamydomonas has been a prominent research model for many years, extensively utilized in studying photosynthesis, sulfur and phosphorus metabolism, nitrogen metabolism, respiration, and flagella synthesis, among others. Recent re-search has increasingly recognized the potential of Chlamydomonas-bacteria consortia as a bio-technological tool for various applications. Bioremediation of wastewater using Chlamydomonas, and its bacterial consortia presents significant potential for the sustainable reduction of contam-inants, while also facilitating resource recovery and valorization of microalgal biomass. Using Chlamydomonas and its bacterial consortia as biofertilizers can offers several benefits, such as en-hancing crop yield, protecting crops, maintaining soil fertility and stability, aiding in CO2 miti-gation, and contributing to sustainable agriculture practices. Chlamydomonas-bacterial consortia play a significant role in the production of high-value products, particularly in biofuel and en-hancing H2 production. This review aims to achieve a comprehensive understanding of the po-tential of Chlamydomonas monoculture and its bacterial consortia, identifying current applications, and proposing new research and development directions to maximize its potential.

Since 2015, the invasive alga Rugulopteryx okamurae has triggered the most serious marine macrophyte invasion in Europe. The removal of its huge coastal biomass generates health problems, strong odors, impact on tourism and high clean-up costs. As a circular economy strategy we propose composting with native terrestrial isopods that has not been previously tested. Therefore, specimens of Porcellio laevis were captured in urban parks and kept for 5 years in terrariums with adequate humidity and temperature control. A sample of 150 adult specimens was divided into six batches of 25 animals. Three batches were fed with organic household waste (control) and three with a 50% diet of waste and algae (treatment). P. laevis consumed up to 1.5 times their weight per day on the algae diet with little or no weight loss, but there was high mortality in both mancas and adults. However, the results are promising and we propose to encourage research with isopods because of their great voracity, high prolificacy and rusticity for the recycling of invasive algae. It should be explored in further work what percentage of R.okamurae avoids mortality by the diterpenes of the algae.

The environmental disaster in Kamchatka in the autumn of 2020 was caused by an extensive bloom of harmful microalgae of the genus Karenia. A spectral shape algorithm was used to detect algae. The algorithm calibration of in situ species composition data made it possible to identify areas where harmful algae dominated in biomass. Satellite images of chlorophyll-a concentra-tion, turbidity, specific fluorescence, and spectral shape parameter were computed. The images were used to recognize the stages of algal bloom: intensive growth, blooming, and change in the dominant algal species. Cases of an increase in the concentration of harmful substances in the coastal zone due to wind impact were analyzed. The following explanation of events has been offered. After the stage of intensive growth of microalgae, nutrient deficiency stimulated the production of metabolites that have a harmful effect on the environment. The change of the dominant alga species in the second half of September and the past storm contributed to a sharp increase in the concentration of metabolites and dead organic matter in the coastal zone, which caused an ecological disaster. The subsequent mass bloom of alga species of the same genus, and the regular wind impact leading to the concentration of harmful substances in the coastal zone, contributed to the development of this catastrophic phenomena.

Cystoseira is a relevant alga, which forms dense meadows on rocky substrates up to 100 meters deep. They are widely recognized as bioindicators for their restricted habitat (sub-coastal zone) and low tolerance to pollution. We have monitored Cystoseira sensu lato (sl) spp. in abrasion platforms in Cabo de Las Huertas (Alicante, SE Spain), a total of seven species. A cladogram has been constructed using fifteen phenotypic taxonomic relevant characters. We have also optimized Cystoseira DNA extraction and sequenced amplicons from mt23S, tRNA-Lys, and psbA genes of five Cystoseira sl species from Cabo de Las Huertas. Our phylogenetic supertree, built with these sequences, backs three clades in the genus. Rare species Treptacantha sauvageauana and Treptacantha algeriensis, sequenced for the first time, are included with T. elegans and the Atlantic species T. baccata and T. abies-marina in Cystoseira-VI or Cystoseira-II clades. Our data supports a complex colonization of Cystoseira (sl) spp. in the Mediterranean, perhaps involving Atlantic clades.

The presence of harmful algal blooms (HABs) and cyanotoxins in drinking water sources poses a great threat to human health. The current study employed molecular techniques to determine the occurrence of non-toxic and toxic cyanobacteria species in the Limpopo River basin based on the phylogenetic analyses of 16S rRNA gene. The bottom sediments samples were collected from selected rivers: Limpopo, Crocodile, Mokolo, Mogalakwena, Nzhelele, Lephalale, Sand Rivers (South Africa); Notwane (Botswana), Shashe River and Mzingwane River (Zimbabwe). The physical-chemical analysis of the bottom sediments showed the availability of nutrients, nitrates and phosphates, in excess of 0.5 mg/l for most of river sediments, alkaline pH and salinity in excess of 500 mg/l. The FlowCam showed the dominant cyanobacteria species identified from the samples were Microcystis species, followed by Cylindrospermopsis raciborskii, Phormidium and Planktothrix species and this was confirmed by molecular techniques. Nevertheless, two samples showed the amplification of cylindrospermopsin polyketide synthetase gene (S3 and S9) while two samples showed amplification for microcystin/nodularin synthetase gene (S8 and S13). Thus these findings may imply the presence of toxic cyanobacteria species in the river sediments. The presence of cyanobacteria may be hazardous to human because rural communities and farmers who abstract water from Limpopo river catchment for human consumption, livestock and wildlife watering and irrigation.

Isolation of new plant-derived lactic acid bacteria (LAB) is highly prioritized in developing novel starter cultures for plant-based fermentation. This study explores the diversity of LAB in Danish flowers and their potential use for plant-based food fermentation. 46 flower samples under 34 genera were collected for LAB isolation. By introducing an enrichment step, in total of 61 LAB strains were isolated and identified by using MALDI-TOF and 16S rRNA sequencing. These strains represent 24 species across 9 genera, predominantly Leuconostoc mesenteroides, Fructobacillus fructosus, Apilactobacillus ozensis, and Apilactobacillus kunkeei. Phenotypic screening for exo-cellular polysaccharide production revealed that 40 strains exhibited sliminess or ropiness on sucrose containing agar plate. HPLC analysis confirmed that all isolates produced exo-cellular polysaccharides containing glucose, fructose or galactose as sugar monomer. Therefore, the strains were glucan, fructan and galactan producers. The suitability of these strains for plant-based fermentations was characterized by using almond, oat and soy milk. The results showed successful acidification in all three types of plant-based matrices but only observed texture development in soy by Leuconostoc, Weissella, Lactococcus, Apilactobacillus, and Fructobacillus. The findings highlight the potential of flower-derived LAB strains for texture development in soy-based dairy alternatives.

Cryptococcus neoformans is a facultative intracellular fungal pathogen. Ten-generation-old (10GEN) C. neoformans cells are more resistant to phagocytosis and killing by macrophages than younger daughter cells. However, mechanisms that mediate this resistance and intracellular parasitism are poorly understood. Here, we identified factors important for the intracellular survival of 10GEN C. neoformans, such as urease activity, capsule synthesis, and DNA content using flow cytometry and fluorescent microscopy techniques. Real-time visualization of time-lapse imaging was applied to determine the phagosomal acidity, membrane permeability, and vomocytosis (non-lytic exocytosis) rate in J774 macrophages that phagocytosed C. neoformans of different generational ages. Our results showed that old C. neoformans exhibited higher urease activity, and enhanced capsule formation, which was supported by an increased Golgi complex. In addition, old C. neoformans were more likely to be arrested in the G2 phase, resulting in the occasional formation of aberrant trimera-like cells. To finish, the advanced generational age of the yeast cells slightly reduced vomocytosis events within host cells, which might be associated with increased phagolysosomal pH and membrane permeability. Altogether our results contribute to a better understanding of the strategies used by old C. neoformans to resist phagosomal killing and suggest that old generation C. neoformans intracellular behavior may be driving cryptococcosis pathogenesis.

Plant proteins being considered to become the most important protein source of the future, they must be able to replace the animal-derived proteins currently in use as technofunctional food ingredients. This poses challenges because plant proteins are oftentimes storage proteins with a high molecular weight and low water solubility. One promising approach to overcome these limitations is the glycation of plant proteins. The covalent bonding between the proteins and different carbohydrates created via the initial stage of the Maillard reaction can improve the technofunctional characteristics of these proteins without the involvement of potentially toxic chemicals. However, compared to studies with animal-derived proteins, glycation studies on plant proteins are currently still underrepresented in literature. This review provides an overview of the existing studies on the glycation of the major groups of plant proteins with different carbohydrates using different preparation methods. Emphasis is put on the reaction conditions used for glycation as well as the modifications to physicochemical properties and technofunctionality. Different applications of these glycated plant proteins in emulsions, foams, films, and encapsulation systems are introduced. Another focus lies on the reaction chemistry of the Maillard reaction and ways to harness it for controlled glycation and to limit the formation of undesired advanced glycation products. Finally, challenges related to the controlled glycation of plant proteins to improve their properties are discussed.

In cystic fibrosis (CF), pulmonary infection with Pseudomonas aeruginosa is a cause of increased morbidity and mortality, especially in patients for whom infection becomes chronic and there is reliance on long-term suppressive therapies. Current antimicrobials, though varied mechanistically and by mode of delivery, are inadequate not only due to eradication failure in many cases, but also because they do not halt the progression of lung function decline over time. One of the reasons for this failure is thought to be the biofilm mode of growth of P. aeruginosa, wherein self-secreted exopolysaccharides (EPSs) provide physical protection against antibiotics and an array of niches with resulting metabolic and phenotypic heterogeneity. Targeting the three EPSs secreted by P. aeruginosa (alginate, Psl and Pel) is currently under investigation as a way of disrupting the biofilm extracellular matrix to potentiate the action of antibiotics. In this review, we look at each EPS as a potential therapeutic target for combatting pulmonary infection with P. aeruginosa in CF, with a particular focus on the current evidence for these emerging therapies and barriers to bringing these therapies into clinic.

Phytoremediation, a method of phytomanagement using the plant holobiont to clean up polluted soils, is particularly effective for degrading organic pollutants, such as alkanes and polycyclic aromatic hydrocarbons (PAHS). However, the respective contributions of host plants and their associated microbiota within the holobiont to the efficiency of phytoremediation is poorly understood. Identification of plant-associated bacteria capable of efficiently utilizing these compounds as carbon source while stimulating plant-growth, is a keystone for phytomanagement engineering in order to improve the efficiency of pollutant removal. In this study, we sampled the rhizosphere and the surrounding bulk soil of Salix purpurea and Eleocharis obusta from the site of a former petrochemical plant in Varennes, QC, Canada. Our objectives were to: (i) isolate and identify indigenous bacteria inhabiting these biotopes; (ii) assess the ability of isolated bacteria to utilize alkanes (dodecane and hexadecane) and PAHs (naphthalene, phenanthrene and pyrene) as the sole carbon source, and (iii) determine the plant growth-promoting (PGP) potential of the isolates using five key traits. A total of 438 morphologically different bacterial isolates were obtained, purified, preserved and identified through PCR and 16S rRNA gene sequencing. Identified isolates represent 62 genera, including taxa such as Acinetobacter, Arthrobacter, Bacillus, Enterobacter, Nocardia, Pseudomonas, Rhodococcus, Streptomyces and Variovorax. Approximately, 32% of bacterial isolates, including Arthrobacter, Pseudomonas, Streptomyces, Enterobacter, Nocardia, Acinetobacter and Microbacterium, were able to utilize all five different hydrocarbons compounds. Additionally, 5% of tested isolates belonging to genera Pseudomonas, Acinetobacter, Serratia, Klebsiella, Microbacterium, Bacillus and Stenotrophomonas possessed all five of the tested PGP functional traits. This culture collection of diverse, petroleum-hydrocarbon degrading bacteria, with multiple PGP traits, represents a valuable resource for future use in environmental bio- and phyto-technology applications, including phytoremediation of petroleum hydrocarbons contaminated soils and phytomanagement of anthropized areas.

Research in seaweed cultivation technologies aims to increase production and reduce costs, leading to more efficient and sustainable processes. In this study, we analyzed the outdoor production of Ulva compressa cultured in summertime at different stocking densities of 0.6, 0.8 and 1.0 kg Fresh weight (FW) m-2 in a raceway photobioreactor with 30 m2 surface (3000L) and its relation to pho-tosynthetic activity. Under the experimental conditions of high temperature (> 28-30 oC) and pH > 9 in culture water, higher seaweed density resulted in lower specific growth rate. The biomass pro-duction has been related to photosynthetic activity by using in vivo chlorophyll a fluorescence. Dynamic photoinhibition was observed at noon, which was less severe in cultures with higher algal densities. However, photosynthesis recovered in the afternoon. Seaweeds that were acclimatized for a week to the conditions of 1.0 kg FW m-2 stocking density showed an increase in biomass growth and absence of photoinhibition compared to non-acclimatized thalli. In conclusion, by using high densities (1.0 kg FW m-2), U. compressa presented the highest growth rate in a mid-scale raceway photobioreactor under stressed conditions related to the increase of photosynthetic capacity.

In recent decades, as eutrophication in inland and coastal waters (ICWs) has increased due to anthropogenic activities and global warming, so has the need for timely monitoring. Compared with traditional sampling and laboratory analysis methods, satellite remote sensing technology can provide macro-scale, low-cost, and near real-time water quality monitoring services. The Geostationary Ocean Color Imager (GOCI), integrated onboard the Communication Ocean and Meteorological Satellite (COMS) from the Republic of Korea, was the first geostationary ocean color observation satellite and was operational from April 1, 2011 to March 31, 2021. Over ten years, GOCI has observed oceans, coastal waters, and inland waters within its 2,500 km×2,500 km target area centered on the Korean Peninsula. The most attractive feature of GOCI, compared with other commonly used watercolor sensors, was its high temporal resolution (1h, eight times daily from 0 UTC to 7 UTC), providing the opportunity to monitor the quality of ICWs, where optical properties can change rapidly throughout the day. This systematic review aims to comprehensively review GOCI features and applications in ICWs, analyzing progress in atmospheric correction algorithms and water quality monitoring. Analyzing 123 articles from the Web of Science and China National Knowledge Infrastructure (CNKI) through a bibliometric quantitative approach, we examined GOCI’s strength and performance with different processing methods. These articles reveal that GOCI played an essential role in monitoring the ecological health of ICWs in its observation area in East Asia. GOCI has led the way to a new era of geostationary ocean satellites, providing new technical means for monitoring water quality in oceans, coastal zones, and inland lakes. We also discuss the challenges encountered by geostationary satellites in monitoring water quality and provide suggestions for improvements.

Coastal marine ecosystems worldwide, like Florida Bay, are increasingly affected by tide alteration and anthropogenic disturbances which affect water quality imbalance leading to algal blooms. Increased bloom persistence is a serious threat due to the long-lasting impact on processes like carbon cycle and services like species presence. However, exploring eco-environmental feedback patterns of algal blooms remains challenging and poorly investigated, also due to the paucity of data. Florida Bay, taken as an epitome, has long experienced algal blooms in its central and western regions, and in 2006 an unprecedented bloom occurred in the eastern habitats. We analyzed the occurrence of blooms from three perspectives: (1) the spatial spreading networks of chlorophyll-a (CHLa) that pinpoints source and unbalanced habitats; (2) the fluctuations of water quality factors pre- and post-bloom outbreaks to in assess environmental impacts of ecological dysbiosis and target prevention and control of algal of blooms; and (3) the biogeochemical-spreading network topological co-evolution to quantify ecosystemic stability and ecological shift likelihood in long-term. Here, we propose Transfer Entropy (TE) to quantify dynamical interactions between spatial areas and biogeochemical factors (ecosystem connectome) underpinning bloom emergence and spreading, as well as environmental effects; the Pareto principle is defined for identifying the salient eco-environmental interactions of CHLa. We quantified the spatial dynamics of algal blooms, and thus obtained areas in need for ecological monitoring and potential bloom control. Results show that algal blooms are increasingly persistent over space with long-term negative effects on water quality factor. A dichotomy is reported between spatial ecological corridors of spreading and biogeochemical networks as well as divergence from the optimal eco-organization: randomization of the former due to nutrients' overload and temperature increase leads to scale-free CHLa spreading and extreme outbreaks a posteriori. Subsequently this increases bloom persistence, turbidity and salinity with potentially strong ecological effects on highly biodiverse and vulnerable areas such as tidal flats and marshes. Algal blooms are important ecosystem regulators of nutrient cycles; however, beyond limit chlorophyll-a outbreaks cause aquatic species mortality due to their effect on water turbidity, nutrient balance (nitrogen and phosphorus in particular), salinity and temperature. Beyond compromising local environmental quality, socio-ecological services are also compromised at large scales, yet ecological assessment models like the one presented are in need of application in subtropical and tropical bays worldwide.

The pennate diatom Phaeodactylum tricornutum is a model organism able to synthesize industrially-relevant molecules. Commercial-scale cultivation currently requires large monocultures, prone to bio-contamination. However, little is known about the identity of the invading organisms. To reduce the complexity of natural systems, we systematically investigated the microbiome of non-axenic P. tricornutum cultures from a culture collection in reproducible experiments. The results revealed a dynamic bacterial community that developed differently in “complete” and “minimal” media conditions. In complete media, we observed an accelerated “culture crash”, indicating a more stable culture in minimal media. The identification of only four bacterial families as major players within the microbiome suggests specific roles depending on environmental conditions. From our results we propose a network of putative interactions between P. tricornutum and these main bacterial factions. We demonstrate that, even with rather sparse data, a mathematical model can be reconstructed that qualitatively reproduces the observed population dynamics, thus indicating that our hypotheses regarding the molecular interactions are in agreement with experimental data. Whereas the model in its current state is only qualitative, we argue that it serves as a starting point to develop quantitative and predictive mathematical models, which may guide experimental efforts to synthetically construct and monitor stable communities required for robust upscaling strategies.

Soil contamination with petroleum hydrocarbons (PHCs) has become a global concern in the word due to intensification of industrial activities. This creates a serious environmental issue, therefore there is a need to find solutions, including application of efficient remediation technologies, or to improve current techniques. Rhizoremediation is a sub-category of the phytoremediation which refers to Phytomanagement that uses plants and their associated microbiota. These green technologies have received a global attention as a cost-effective and possible efficient remediation technique that can be applied to cleanup PHCs-polluted soils. The mechanism of rhizoremediation process is that plant roots stimulate soil microbes to mineralize organic contaminants to H2O and CO2. However, this multipartite interaction is much complex because many biotic and abiotic factors can influence microbial processes in the soil, making the efficiency of rhizoremediation unpredictable. This review reports the progress made on rhizoremediation approaches that can overcome the limitations and improve the efficiency of PHCs-contaminated soils. The addressed approaches in this review include: 1) selecting plants with desired characteristics suitable for rhizoremediation, 2) the exploitation and manipulation of plant microbiome by using inoculant containing plant growth-promoting rhizobacteria (PGPR) or hydrocarbon-degrading microbes, or a combination of both types of organisms, and 3) enhancement of the understanding of how host-plant assembles a beneficial microbiome, and how it functions, under pollutant stress.

Damage to peripheral arteries by the atherosclerotic process accounts for about 3% of the world's population, which puts this problem in the category of major social problems. Modified lipoproteins and cholesterol crystals are known to accumulate in the arterial intima and cause the formation of proatherogenic macrophages, foam cells, and inflammation. Defective efferocytosis of apoptotic foam cells leads to the formation of a necrotic core, a sign of failure to resolve inflammation. Resolution of inflammation is mediated by specialized pro-resorption lipid mediators, proteins and signaling gases. Improving the balance between pro-inflammatory and anti-inflammatory factors contributes to the reverse development of local atherogenesis. Active direct intervention in the reconstruction of the adventitial layer of the arterial wall using cholesterol-affinity polysaccharide hydrogels containing a cocktail of growth factors creates conditions for the formation of additional extracellular matrix and reversal of cholesterol mass from the subintimal zone. Creating concentration and electrostatic gradients in the adventitia zone using a hydrogel may be one of the effective ways to degrade early soft atherogenic plaques and locally restore the damaged structure of the vessel wall. Growing scientific interest in the previously insufficiently studied adventitia indicates its important role in angionesis and atherogenesis.

Curdlan is a water-insoluble polymer that has structure and gelling properties useful in a wide variety of applications such as in medicine, cosmetics, packaging and the food and building industries. The capacity to produce curdlan has been detected in certain soil-dwelling bacteria of various phyla, although the role of curdlan in their survival remains unclear. One of the major limitations of the extensive use of curdlan in industry is the high cost of production during fermentation, partly because production involves specific nutritional requirements such as nitrogen limitation. Engineering of the industrially relevant curdlan-producing strain Agrobacterium sp. ATTC31749 is a promising approach that could decrease the cost of production. Here, during investigations on curdlan production, I found that curdlan was deposited as a capsule. Curiously, only a part of the bacterial population produced a curdlan capsule. This heterogeneous distribution appeared to be due to the activity of Pcrd, the native promoter responsible for the expression of the crdASC biosynthetic gene cluster. To improve curdlan production, Pcrd was replaced by a promoter (PphaP) from another Alphaproteobacterium, Rhodobacter sphaeroides. Compared to Pcrd, PphaP was stronger and only mildly affected by nitrogen levels. Consequently, PphaP dramatically boosted crdASC gene expression and curdlan production. Importantly, the genetic modification overrode the strict nitrogen depletion regulation that presents a hindrance for maximal curdlan production and from nitrogen rich, complex media, demonstrating excellent commercial potential for achieving high yields using cheap substrates under relaxed fermentation conditions.

Natural products, especially biologically active compounds derived from plants, have been and continue to be invaluable in anticancer research and therapeutic discoveries. Rice bran arabinoxylan compound (RBAC) is a heteropolysaccharide extract of defatted rice bran obtained through enzymatic treatment with Lentinus Edodes mycelium. RBAC exhibited antitumour properties by arresting in vivo tumour growth in several murine models. In vitro experiments showed RBAC to promote apoptosis in cancer cells by increasing the susceptibility of CD95 (Fas/APO-1) ligands and downregulating Bcl-2 proteins to trigger the permeabilisation of the mitochondrial membrane. RBAC may restore immune dysfunction in cancer patients, most notably by upregulating natural killer cell activity and modulating cytokine production to enhance inflammatory and cytotoxic responses. As such, RBAC acts on the immune system to enhance chemopreventative activity and block tumourigenesis. In treatment, RBAC works synergistically with other natural products and chemotherapeutic agents, such as daunorubicin and paclitaxel, by enhancing drug transport to increase DNA damage in tumours and reduce proliferation. In a clinical trial, combining RBAC with chemoembolization in treating liver cancer showed improved response, reduced recurrence, and prolonged survival time. RBAC has been shown to augment the endogenous antioxidant system to prevent oxidative stress and thus protect against the harmful side effects of radiation therapy. RBAC also has chemoprotective effects, with animal and human studies showing reduced toxicity and side effects from chemotherapy, leading to improved health-related quality of life. Meta-analysing the best available evidence indicates that RBAC treatment increases the odds of survival among cancer patients by 4.02 times (95% CI: 1.67, 9.69) in the first year and 2.89 (95% CI: 1.56, 5.35) times in the second year after commencing treatment, compared to those not treated with RBAC. Notwithstanding, additional research is needed to characterise, quantify, and standardise the active ingredients in RBAC responsible for the anticancer effects. More well-designed, large-scale clinical trials are required to further substantiate the treatment efficacies.

Heavy metal contamination of drinking water is a public health concern that requires the development of more efficient bioremediation techniques. Absorption technologies, including biosorption, provide opportunities for improvements to increase the diversity of metal ions removed and overall binding capacity. Microorganisms are a key component in wastewater treatment plants and they naturally bind metal ions through surface macromolecules but with limited capacity. The long-term goal of this work is to engineer capsule polymerases to synthesize molecules with novel functionalities. In previously published work, we showed that the Neisseria meningitidis serogroup W (NmW) galactose-sialic acid (Gal—NeuNAc) heteropolysaccharide binds lead effectively, thereby demonstrating the potential for using this capsular polysaccharide in environmental decontamination applications. In this study, computational analysis of the NmW capsule polymerase galactosyltransferase (GT) domain was used to gain insight into how the enzyme could be modified to enable the synthesis N-acetylgalactosamine-sialic acid (GalNAc—NeuNAc) heteropolysaccharide. Various computational approaches, including molecular modeling with I-TASSER and molecular dynamics simulations (MD) with NAMD, were utilized to identify key amino acid residues in the substrate binding pocket of the GT domain that may be key to conferring UDP-GalNAc specificity. Through these combined strategies and using BshA, a UDP-GlcNAc transferase, as a structural template, several NmW active site residues were identified as mutational targets to accommodate the proposed N-acetyl group in UDP-GalNAc. Thus, a rational approach for potentially conferring new properties to bacterial capsular polysaccharides is demonstrated.

The two-dimensional, laterally-averaged mechanistic eutrophication model CE-QUAL-W2 version 3.72 was used to predict chlorophyll-a concentrations across two different time periods in the Neuse River Estuary, North Carolina. Chlorophyll calibration was performed for two time periods simultaneously by performing by a full-factorial experiment that tested seven algal kinetic growth parameters over three levels for a single algal group. A cluster of up to six computers each running between two and ten instances of the program was used to complete and manage the data for 2187 runs for each time period. A set of six criteria were used to determine which runs performed acceptably, yielding a group of 27 cases that met all of the criteria. Calibration performance of the set of cases outperformed a previously calibrated model using three algal groups that met only four of the six selection criteria. Calibration performed this way allowed for a more rational specification of model calibration performance and provided uncertainty estimates of model predictions, albeit at the cost of a considerable increase in computational requirements that necessitated the use of a computer cluster.

A recent report from the American Heart Association in 2018 shows that over 103 million American adults have hypertension. The angiotensin-converting enzyme (ACE) (EC 3.4.15.1) is a dipeptidyl carboxylase that, when inhibited, can reduce blood pressure through the renin-angiotensin system. ACE inhibitors are used as a first-line medication to be prescribed to treat hypertension, chronic kidney disease, heart failure among others. It has been suggested that ACE inhibitors can reduce the symptoms in mouse models. Despite the benefits of ACE inhibitors, previous studies also have suggested that alterations in the ACE gene are risk factors for Alzheimer’s disease (AD) and other neurological diseases. In mice, overexpression of ACE in the brain reduces symptoms of the AD-model systems. Thus, we find opposing effects of ACE on health. To clarify the effects, we dissect the functions of ACE as follows: (1) angiotensin-converting enzyme that hydrolyzes angiotensin I to make angiotensin II in the renin-angiotensin system; (2) amyloid-degrading enzyme that can hydrolyze beta-amyloid and reduce amyloid toxicity. The efficacy of the ACE inhibitors is well established in humans, while the knowledge specific to AD remains to be open for further research. We provide an overview of ACE and inhibitors that link a wide variety of age-related comorbidities from hypertension to Alzheimer’s disease to aging. ACE also serves as an example of the middle-life crisis theory that assumes deleterious events during the midlife, leading to age-related later events.

Phytoplankton under the ice of seasonally ice-covered lakes is a key indicator species for maintaining the health of lake ecosystems. However, there is a lack of a unified definition of cold lakes internationally. In addition, there are few studies on the distribution, driving mechanism and simulation prediction of phytoplankton under the ice, which have limited the development of winter limnology and the prevention and control of spring algal blooms. The average temperature of the coldest month, the duration of the ice period and the average annual water temperature were the main indicators for defining cold lakes based on the literatures review. The cross-action of subglacial hydrodynamics and biogeochemistry during the ice period and freeze-thaw period is the key to promoting the phytoplankton growth. The migration and transformation of nutrients and the formation of hypoxic zones of subglacial were driven by physical factors such as available light and water temperature. Cyanobacteria got advantages in the ice period and above the water column through their own gas vesicles and strong adaptation to low temperature and low oxygen. Diatoms are able to multiply rapidly with the increasing of available light and the water temperature in spring. However, the migration and transformation of nutrients induced by physical factors during the freezing-freezing-thawing process, the role of microorganisms in biogeochemical reactions, and the extent and direction of the combined effects of multiple environmental factors on phytoplankton functional groups are still unclear. At present, lake algal blooms have gradually expanded to higher latitudes under the climate change. How to accurately predict the development and response mechanism of algal blooms in cold lakes under the climate change has become a key direction in the development of winter limnology. This review explains the unique physical and chemical processes of phytoplankton growth competition in cold lakes from the perspectives of bibliometrics, mechanism analysis, method construction and future prospects. These findings would provide new insights into the field of winter limnology and provide theoretical support for the future management of spring algal blooms in cold lakes.

The current study investigated the use of two-dimensional spatial distribution mapping representing the chlorophyll-a level in a river generated via an unmanned aerial vehicle (UAV) and an unmanned surface vehicle (USV). A domestically developed UAV (Remo-M, Uconsystem Inc., Korea) and a USV developed by our research team were used to collect data from the Nae Seong stream in Korea. An adaptation of the “Data Cleaner” tool was developed and used for USV data processing and analysis. The operation of the autonomous USV was successful. Four previously described indices for quantifying algal blooms in rivers were utilized to create chlorophyll-a images, the normalized difference vegetation index (NDVI), the normalized green red difference index, the green normalized difference vegetation index (GNDVI), and the normalized difference red edge index. The suitability of the linear regression analysis of the correlation between the spectral indices obtained using the UAV and the in situ chlorophyll-a data obtained using the USV was evaluated with the coefficient of determination (R2) at a significance level of p &lt; 0.001. In field application and correlational analysis the NDVI was strongly correlated with chlorophyll-a (R2 = 0.88, p < 0.001), and the GNDVI was moderately correlated with chlorophyll-a (R2 = 0.74, p < 0.001). The map of chlorophyll-a was successfully quantified using the UAV and USV hybrid platforms.

Background: Recently we demonstrated that Astragalus polysaccharide (PG2), the active ingredient in dried roots of astragalus membranaceus, ameliorates cancer symptom clusters and improve quality of life (QoL) in patients with metastatic disease by modulating inflammatory cascade, against the background that inflammatory cells including macrophages, dendritic cells (DCs), and cytotoxic T lymphocytes (CTLs) in tumor initiation, metastasis, and progression. Nevertheless, the role of PG2 in the modulation of anticancer immunogenicity and therapeutic response remains relatively underexplored and unclear. Purpose: The present study investigates how and to what extent PG2 modulates cellular and biochemical components of the inflammatory cascade, enhance anticancer immunity, and the therapeutic implication of same in patients with lung cancer. Methods and Results: Herein, we demonstrated that PG2 significantly increased the M1/M2 macrophage polarization ratio in non-small cell carcinoma (NSCLC) H441 and H1299 cells. This PG2-induced preferential pharmacologic up-regulation of tumoral M1 population in vitro, positively correlated with the downregulation of tumor-promoting IL-6 and IL-10 expression in NSCLC cell-conditioned medium, with concomitant marked inhibition of cell proliferation, clonogenicity and tumorsphere formation. Our ex vivo results, using clinical sample from our NSCLC cohort, demonstrated that PG2 also promoted the functional maturation of DCs with consequent enhancement of T cell-mediated anticancer immune responses. Consistent with the in vitro and ex vivo results, our in vivo studies showed that mice treated with PG2 exhibited significant time-dependent depletion of the tumor-associated M2 population, synergistically enhanced the anti-M2-based anticancer effect of cisplatin, and inhibition of xenograft tumor growth in the NSCLC mice models. Moreover, in the presence of PG2, cisplatin-associated dyscresia and weight-loss was markedly suppressed. Conclusion: These results do indicate a therapeutically-relevant role for PG2 in modulating the M1/M2 macrophage pool, facilitating DC maturation, and synergistically enhancing the anticancer effect of conventional chemotherapeutic agent, cisplatin; thus laying the foundation for further exploration of the curative relevance of PG2 as surrogate immunotherapy and/or clinical feasibility of its use for maintenance therapy in patients with lung cancer.

Fiber-rich green river Aonori alga is mass-cultivated in Japan; it has been reported to reduce inflammation and improve metabolic health in humans. Peroxisome proliferation-activated receptor (PPARs) are associated with metabolic disorders such as obesity and type 2 diabetes (T2DM). We evaluated the effects of supplementation with NBF2, a formula made of algal and junos citrus biomaterials, on metabolic markers in obese and diabetic rats. An experimental study was performed using 18 obese and hyperglycemic Otsuka Long-Evans Tokushima Fatty (OLETF) rats randomly assigned to high dose NBF2 (20mg/kg; n=6), low dose (10mg/kg; n=6) NBF2 drink for 6 weeks, whereas OLETF control rats received similar amount of tap water (vehicle; n=6). LETO rats were used as normal controls (n=8). Biospecimens were taken for ELISA and real-time PCR. Both high dose and low dose NBF2 increased brown adipose tissue (BAT) mass and adiponectin (APN), improved insulin sensitivity, and significantly improved glycemic and lipid (TG, LDL-c) profiles in NBF2-supplemented OLETF rats after a 6-week supplementation with NBF2 (vs. OLETF controls), as well as LDL-c (p<0.05). During the glucose tolerance test (OGTT), both NBF2 groups and LETO rats had normal glycemia by 90-minute time-point (vs. OLETF controls; p<0.01). Furthermore, intake of NBF2 up-regulated PPARa/g-mRNA and Sirt2-mRNA gene expressions in BAT, and improved blood pressure profile in OLETF rats. Findings from this research suggest that intake of NBF2 formula induces dual PPAR-a/g and Sirt2 activation, providing insights on beneficial prophylactic and therapeutic effects of this NB2 formula on dyslipidemia and hyperglycemia in T2DM.

Algal-bacterial granular sludge, a new biological technology, has been widely recognized due to its highly effective pollutant treatment and energy efficiency. This study investigated the effects of environmental concentrations of Cr(VI) (0.5-2.5 mg/L) on the performance of algal-bacterial gran-ular sludge and self-defensive responses after 90 days of cultivation. The results showed that Cr(VI) affected chemical oxygen demand (COD), ammonia-N and phosphate removal with different trends being apparent. A linear decline in COD removal was observed, whereas an initial de-creasing and then increasing ammonia-N and phosphate removal took place. Algal-bacterial granular sludge effectively removed Cr(VI) from wastewater through biological adsorption and reduction, showing the potential to treat Cr(VI)-contaminated wastewater. Cr(VI) affected the community abundance of the algal-bacterial granular sludge, in which Chlorophyceae and cya-nobacteria were vulnerable under Cr(VI)-induced stress. To reduce the toxicity of Cr(VI), over-produced EPS-PN and antioxidant enzymes (MDA, SOD and CAT) acted as self-defensive responses to resist oxidative damage. This study showed that algal-bacterial granular sludge can remove 00.5 mg/L of Cr(VI) without performance loss. It is hoped that this study can provide useful information for improved engineering feasibility of algal-bacterial granular sludge.

The effects of the consumption of an extract of the brown seaweed Ascophyllum nodosum (BSW) on postprandial glucose and insulin response to white bread were investigated in an acute, randomized, double-blind, three-arm, crossover, controlled trial in healthy, normoglyacemic subjects. Sixteen subjects were administrated either control white bread (50 g total digestible carbohydrates), or white bread with either 500 mg or 1000 mg of BSW extract. Biochemical parameters were measured in venous blood over 3h. Significant inter-individual variation in the glycaemic response to white bread was observed. Analysis of the response of all subjects to either 500 mg or 1000 mg of BSW extract versus control revealed no significant effects of treatments. The variation in response to the control was used to classify individuals into glyacemic responders and non-responders. In the sub-cohort of 10 subjects with peak glucose levels after white bread above 1mmol/L we observed a significant decrease in maximum levels of plasma glucose after the intervention meal with 1000 mg of extract, compared with control. No adverse effects were reported. Further work defining all factors that determine “responders” to the effects of brown seaweed extracts and identify the cohort that would benefit the most from their consumption, is warranted.

Over the past two decades, persistent occurrences of harmful algal blooms (HAB; Karenia brevis) have been reported in Charlotte County, southwestern Florida. We developed data-driven models that rely on spatiotemporal remote sensing and field data to identify factors controlling HAB propagation, provide a same-day distribution (nowcasting), and forecast their occurrences up to three days in advance. We constructed multivariate regression models using historical HAB occurrences (213 events reported from January 2010 to October 2017) compiled by the Florida Fish and Wildlife Conservation Commission and validated the models against a subset (20%) of the reported historical events. The models were designed to specifically capture the onset of the HABs instead of those that developed days earlier and continued thereafter. A prototype of an early warning system was developed through a threefold exercise. The first step involved the automatic downloading and processing of daily Moderate Resolution Imaging Spectroradiometer (MODIS) Aqua products using SeaDAS ocean color processing software to extract temporal and spatial variations of remote sensing-based variables over the study area. The second step involved the development of a multivariate regression model for same-day mapping of HABs and similar subsequent models for forecasting HAB occurrences one, two, and three days in advance. Eleven remote sensing variables and two non-remote sensing variables were used as inputs for the generated models. In the third and final step, model outputs (same-day and forecasted distribution of HABs) were posted automatically on a web-based GIS (http://www.esrs.wmich.edu/webmap/bloom/). Our findings include the following: (1) the variables most indicative of the timing of bloom propagation are bathymetry, euphotic depth, wind direction, SST, chlorophyll-a [OC3M] and distance from the river mouth, and (2) the model predictions were 90% successful for same-day mapping and 65%, 72% and 71% for the one-, two- and three-day advance predictions, respectively. The adopted methodologies are reliable, dependent on readily available remote sensing data sets, and cost-effective and thus could potentially be used to map and forecast algal bloom occurrences in data-scarce regions.

Filamentous manganese (Mn) oxide particles, which occur in the suboxic zone of stratified waterbodies, are important drivers of diverse elemental cycles. Although these particles are considered to be bacteriogenic, the environmental factor responsible for their formation has not been identified. Based on this study of laboratory cultures of a model Mn(II)-oxidizing bacterium, the supply of algal extracellular polymeric substances (EPS) was shown to stimulate Mn(II) oxidation and thus the production of filamentous Mn oxide particles. This observation was consistent with results obtained for naturally occurring particles collected from a near-bottom layer (depth of ~90 m) in the northern basin of Lake Biwa, Japan, that is, most Mn particles resembling delta-MnO2 were associated with an EPS-like gelatinous matrix, which contained dead algal cells and was lectin-stainable. In the lake water column, EPS produced by photosynthesis sank to the bottom layer. The analysis of the quality of water samples, which have been collected from the study site for 18 years, reveals that the annual average total phytoplankton biovolume in the surface layer correlates with the density of filamentous Mn particles in the near-bottom layer. Among different phytoplankton species, green algae appeared to be the key species. The results of this study suggest that algal EPS production induces the formation of filamentous Mn oxide particles in the near-bottom layer of the northern basin of Lake Biwa.

In a constantly changing environment brought about by the climate crisis and escalated anthropogenic perturbations driven by the growing population, harmful algal bloom dynamics and their impacts are expected to shift, necessitating adaptive management strategies and comprehensive research efforts. Similar to primary productivity, HABs have been thought to be driven primarily by major nutrients such as N, P, and Si. However, recent investigations on the role and importance of micronutrients as limiting factors in aquatic environments have been highlighted. This paper provides a review of metal and phytoplankton interactions, with a specific emphasis on pertinent information on the influence of trace nutrients on growth, toxin production, and other underlying mechanisms related to the dynamics of HABs. Low to near-depleted levels of essential nutrients including Fe, Cu, Zn, Se, Mn, Co, and Mo, negatively impact cell growth and proliferation of various marine and freshwater HAB species. However, evidence shows that at elevated levels, these trace elements along with other non-essential ones, could still cause toxic effects to certain HAB species manifested by decreased photosynthetic activities, oxidative stress, ultrastructure damage, and cyst formation. Interestingly, while elevated levels of these metals mostly result in increased toxin production, Co (i.e., yessotoxins, gymnodimine, and palytoxins) and Mn (i.e., isodomoic acid, okadaic and diol esters) enrichments revealed otherwise. In addition to toxin production, releasing dissolved organic matter (DOM) including dissolved organic carbon (DOC) and humic substances was observed as an adaptation strategy, since these organic compounds have been proven to chelate metals in the water column, thereby reducing metal-induced toxicity. Whilst current research center on free metal toxicity of specific essential elements such as in Cu and Zn, a comprehensive account of how trace metals contribute to the growth, toxin production, and other metabolic processes under conditions reflective of in-situ scenarios of HAB-prone areas, would yield new perspectives on the roles of trace metals in HABs. With the growing demands of the global population for food security and sustainability, substantial pressure is exerted on the agriculture and aquaculture sector, highlighting the need for effective communication of information regarding the interactions of macro- and micronutrients with HABs to improve existing policies and practices.

Pseudo-nitzschia spp. blooms are a recurrent problem in many coastal areas globally imposing some significant threats to the health of human, ecosystems and economy. Monitoring programmes have been established, where feasible, to mitigate the impacts caused by Pseudo-nitzschia spp. and other harmful algae blooms. The detection of such blooms from satellite data could really provide timely information on emerging risks but the development of taxa-specific algorithms from available multispectral data is still challenged by coupled optical properties with other taxa and water constituents, availability of ground data and generalisation capabilities of algorithms. Here, we developed a new set of algorithms (PNOI) for the detection and monitoring of Pseudo-nitzschia spp. blooms over the Galician coast (NW Iberian Peninsula) from Sentinel-3 OLCI reflectances using support vector machine (SVM). Our algorithm was trained and tested with reflectance data from 260 OLCI images and 4607 Pseudo-nitzschia spp. match up data points, of which 2171 were of high quality. The performance of no bloom/bloom model in the independent test set was robust, showing values of 0.80, 0.72 and 0.79 for area under the curve (AUC), sensitivity and specificity, respectively. Similar results were obtained by our below detection limit/presence model. We also present different model thresholds based on optimisation of true skill statistic (TSS) and F1-score. PNOI outperforms linear models, while its relationship with in-situ chlorophyll-a concentrations is weak demonstrating poor correlation with the phytoplankton abundance. We showcase the importance of PNOI algorithm and OLCI sensor for monitoring the bloom evolution between the weekly ground sampling and during periods of ground data absence, such as due to Covid-19.

White shrimps are susceptible to outbreaks of vibriosis because they do not have any adaptive immune system, they only have a non-specific innate immune system. The administration of EPS from microalgae Porphyridium cruentum (synonym: P. purpureum) on shrimps Litopenaeus vannamei was investigated to determine the effect of this immunostimulant on their non specific immune response and to test if EPS can be used as a protective agent for shrimp related to Vibrio infection. EPS was given to shrimps by immersion method on day 1 and booster on day 8. Shrimp hemocytes were taken on day 1 (EPS administration), day 7 (no treatment), day 8 (EPS booster) and day 9 (Vibrio infection) and tested for their immune response on each treatment. Result shows an increase in values ​​of all immune parameters in line with the increasing EPS concentration, except the Differential Haemocyte Count (DHC). In detail, an increase was noted in total hemocytes (THC) value, Phagocytotic Activity (PA), Respiratory Burst (RB) in line as the EPS concentration increase. Although there is a decrease after the infection, the value obtained is not lower than the control value. These results indicate that EPS from Porphyrydium enhances immune parameters in shrimp rapidly and has the ability as an immunostimulant or an immunomodulator. It is a good modulator for the non specific immune cells of Pacific white shrimps, and it can be used as a preventive agent against Vibrio.

The extracellular matrix of most bacterial biofilms contains polysaccharides, proteins, and nucleic acids. These biopolymers have been shown to mediate fundamental biofilm-related phenotypes including surface attachment, intercellular adhesion, and biocide resistance. Enzymes that degrade polymeric biofilm matrix components, including glycoside hydrolases, proteases, and nucleases, are useful tools for studying the structure and function of biofilm matrix components and are also being investigated as potential antibiofilm agents for clinical use. Dispersin B is a well-studied, broad spectrum antibiofilm glycoside hydrolase produced by Aggregatibacter actinomycetemcomitans. Dispersin B degrades poly-N-acetylglucosamine, a biofilm matrix polysaccharide that mediates biofilm formation, stress tolerance, and biocide resistance in numerous Gram-negative and Gram-positive pathogens. Dispersin B has been shown to inhibit biofilm and pellicle formation; detach pre-formed biofilms; disaggregate bacterial flocs; sensitize pre-formed biofilms to detachment by enzymes, detergents, and metal chelators; and sensitize pre-formed biofilms to killing by antiseptics, antibiotics, bacteriophages, macrophages, and predatory bacteria. This review summarizes the results of nearly 100 in vitro and in vivo studies that have been carried out on dispersin B since it’s discovery 20 years ago. These include investigations into the biological function of the enzyme, it’s structure and mechanism of action, and its in vitro and in vivo antibiofilm activities against numerous bacterial species. Also discussed are potential clinical applications of dispersin B.

Nitrogen is essential for plant growth, yield and crop quality, and its deficiency limits food production worldwide. In addition, excessive fertilization and inefficient use of N can increase production costs and cause environmental problems. A possible solution to this problem is the application of biofertilizers, which improve N assimilation and enhance biomass and yield. Therefore, the objective of this research was to evaluate the impact of the application of the combination of green and red algal (Ulva lactuca, Solieria spp.), Rhizobium sp., Trichoderma asperel-lum and the combination of the above three biofertilizers on N assimilation. A completely ran-domized design was carried out with 10 plants per treatment and five treatments: T1=control; T2=algal extracts; T3=Rhizobium sp.; T4=T. asperellum; T5=T2+T3+T4. Analyses showed that the Rhi-zobium sp. treatment resulted in higher nitrate reductase enzyme activity quantified in maize leaves, which possibly favored more efficient phosphosynthetic activity, reflecting higher bio-mass accumulation and increased yield. The use of Rhizobium sp. showed increases in biomass (13.4%), yield (11.82%), SPAD values (12%), total chlorophyll (18.4%), carotenoids (13.6%), num-ber of leaves (11.4%) and plant height (11.27%) compared to the control. This results in nitrogen assimilation promises to be a key mechanism for sustainable agricultural practices in the future.

Exopolysaccharides or extracellular polysaccharides (EPS, sPS) represent valuable metabolite compound synthesized from red microalgae. It is a non toxic natural agent and can be applied as immunostimulant. Toxicity test of exopolysaccharides from Porphyridium has been done in-vivo using zebrafish (Danio rerio) embryonic model, or the ZET (Zebrafish Embryotoxicity Test). The administration of extracellular polysaccharide or exopolysaccharides (EPS) from microalgae Porphyridium cruentum (synonym: P. purpureum) on shrimps Litopenaeus vannamei was investigated to determine the effect of this immunostimulant on their non specific immune response and to test if this compound can be used as a protective agent for shrimp related to Vibrio infection. For immune response, exopolysaccharides was given to shrimps by immersion method on day 1 and booster on day 8. Shrimp hemocytes were taken on day 1 (EPS administration), day 7 (no treatment), day 8 (EPS booster) and day 9 (Vibrio infection) and tested for their immune response on each treatment. These results and other previous studies indicate that EPS from Porphyridium is safe and it enhances immune parameters in shrimp rapidly and has the ability as an immunostimulant or an immunomodulator. It is a good modulator for the non-specific immune cells of Pacific white shrimps, and it can be used as a preventive agent against vibriosis.

The European Union Green Deal aims at curbing planet-warming greenhouse gas emissions and introducing clean energy production. But to achieve energy efficiency, the opportunity cost of different energies must be assessed. In this article, we analyse two different systems for the treatment of wastewater that, at the same time, produce energy for its own operation. On the one hand, high-rate algal ponds system (HRAP) is considered; on the other hand, we study a conventional activated sludge system which uses photovoltaic power (AS+PV). This paper offers a viability analysis of both systems based on the capacity to satisfy their energetic consumption. In order to assess this viability, the probability of not achieving the energy consumption threshold at least one day is studied. The results point that the AS+PV system self-sufficiency is achieved using much lesser surfaces than those of HRAP system (for the former, less than 6.500 m2, for the latter 40.000 m2). However, the important AS capital cost makes still the HRAP system more economic, although storage provides a great advantage for using the AS+PV in locations where we have a lot of irradiance. This viability analysis, along with the opportunity cost analysis, will be used to assess these two projects devoted to the treatment of wastewater.

Nanocellulose is cellulose in the form of nanostructures, i.e. features not exceeding 100 nm at least in one dimension. These nanostructures include nanofibrils, e.g. in bacterial cellulose; nanofibers, e.g. in electrospun matrices; nanowhiskers and nanocrystals. These structures can be further assembled into bigger 2D and 3D nano-, micro- and macro-structures, such as nanoplatelets, membranes, films, microparticles and porous macroscopic matrices. There are four main sources of nanocellulose: bacteria (Gluonacetobacter), plants (trees, shrubs, herbs), algae (Cladophora) and animals (Tunicata). Nanocellulose has emerged for a wide range of industrial, technology and biomedical applications, e.g. for adsorption, ultrafiltration, packaging, conservation of historical artifacts, thermal insulation and fire retardation, energy extraction and storage, acoustics, sensorics, controlled drug delivery, and particularly for tissue engineering. Nanocellulose is promising for use in scaffolds for engineering of blood vessels, neural tissue, bone, cartilage, liver, adipose tissue, urethra and dura mater, for repairing connective tissue and congenital heart defects, and for constructing contact lenses and protective barriers. This review is focused on applications of nanocellulose in skin tissue engineering and wound healing as a scaffold for cell growth, for delivering cells into wounds, and as a material for advanced wound dressings coupled with drug delivery, transparency and sensorics. Potential cytotoxicity and immunogenicity of nanocellulose are also discussed.

In a saline semi-isolated Lagoon on the Cape Zeleny (White Sea), the annual dynamics of the vertical hydrological structure, and the seasonal dynamics of phytoplankton were traced, including species composition, and vertical distribution, abundance, nutrition type,and biomass.293 species and supraspecific taxa of algae and cyanobacteria were found. Most of the identified species are marine, and 38 species are freshwater. Taxonomic composition changed in the Lagoon during the year. Dinoflagellates dominated in winter and early June; unidentified cocci and flagellates in July; diatoms, dinoflagellates and unidentified cells in August; dinoflagellates in September; and unidentified cocci and flagellates in October–November. The abundance of algae also changed in the Lagoon during the year. The integrated biomass in the water column varied from 0.01 g C/m2 in January to 0.78 g C/m2 in early September. According to the abiotic parameters, the water column of the Lagoon is subdivided into several zones with different conditions and corresponding phytoplankton communities. Similarity between the communities of different horizons was 32-46% in summer and 7% in winter. The chemocline layer was the most populous. Almost the entire year it contained a maximum of phytoplankton biomass, 1-2 orders of magnitude higher than that in the overlying horizons. Comparison phytoplankton in the lagoon and in the nearby sea area was carried out. Despite the connection between the Lagoon and the sea phytoplankton structure in the surface water layer in the Lagoon and in the sea differs significantly in composition, quantitative parameters, and seasonal dynamics. The similarity of the communities in the sea and the Lagoon never exceeded 50%. In terms of biomass, the Lagoon lags behind the sea until mid-summer, but, starting from August, it outnumbers it, and the phytoplankton development in the Lagoon lasts longer, until late autumn. The phytoplankton structure and dynamics in the Lagoon and the sea by 24.5% was related to the daylight, water salinity, oxygen content and pH, but did not depend on water T, underwater illuminance, and depth. Oxygen content and pH are defined by PhP activity. Salinity serves as a vector of the vertical sequence of ecological niches. The daylight length seems to be the crucial factor of the seasonal PhP dynamics in the semi-isolated coastal stratified lakes and lagoons.

O26 enteropathogenic Escherichia coli (EPEC), possesses a unique pseudocapsule composed of polysaccharides identical to the O antigen in their lipopolysaccharide (LPS) membrane. Therefore, the impact of O26 polysaccharides on the immune evasion mechanisms of capsulated O26 EPEC was investigated. Our findings reveal that the concentration of GlcNAc, an immunodominant unit of O26 polysaccharides capable of activating the MBL (Mannose Biding Lecting) pathway of the complement system, was found to be lower in capsulated EPEC O26 compared to naked shiga toxin-producing E. coli (STEC) O26 strain. It was noted that, unlike the naked STEC strain, the capsulated EPEC O26 strain was protected against lysis by the alternative pathway of the com-plement system by inhibiting C3b adhesion to its bacterial surface. Capsulated EPEC O26 strains were also able to avoid phagocytosis by macrophages. However, the immune evasion provided by the capsule was overcome in the presence of anti-O26 polysaccharide antibodies. Additionally, these antibodies were able to recognize O26 EPEC in the presence of biofilm and inhibit their adhesion to human epithelial cells. Overall, the results indicate that the O26 polysaccharides can generate an effective humoral immune response, making them promising antigens for the de-velopment of a vaccine against capsulated O26 E. coli

We wanted to investigate the current knowledge on the impact of diet on anti-TNF response in inflammatory bowel diseases (IBD), to identify dietary factors that warrant further investigations in relation to anti-TNF treatment response, and, finally, to discuss potential strategies for such investigations. PubMed was searched using specified search terms. One small prospective study on diet and anti-TNF treatment in 56 patients with CD found similar remission rates after 56 weeks among 32 patients with good compliance that received concomitant enteral nutrition and 24 with poor compliance that had no dietary restrictions (78% versus 67%, p = 0.51). A meta-analysis of 295 patients found higher odds of achieving clinical remission and remaining in clinical remission among patients on combination therapy with specialised enteral nutrition and Infliximab (IFX) compared with IFX monotherapy (OR 2.73; 95% CI: 1.73–4.31, p < 0.01, OR 2.93; 95% CI: 1.66–5.17, p < 0.01, respectively). In conclusion, evidence-based knowledge on impact of diet on anti-TNF treatment response for clinical use is scarce. Here we propose a mechanism by which Western style diet high in meat and low in fibre may promote colonic inflammation and potentially impact treatment response to anti-TNF drugs. Further studies using hypothesis-driven and data-driven strategies in observational, animal and interventional studies are warranted.
