Essential oils (EOs) have been gaining popularity in the past decades among researchers due to their potential to replace conventional chemicals used in the fight against pests, pathogenic and spoilage microbes and oxidation processes. EOs are complex mixtures with many chemical components the content of which depends on many factors – not just the plant genus, species or subspecies, but also chemotype, locality and climatic conditions, phase of vegetation, method of extraction and others. Due to this fact, there’s still much to study, with antimicrobial effect being one of the key properties of EOs. There are many methods that have been frequently engaged by researchers for in vitro evaluation; however, although the research has been going on for decades, an internationally accepted standard is still missing. Most of methods are based on time-proven standards used for testing of antibiotics. Due to the specific properties of EOs and their components, such as volatility and hydrophobicity, many modifications of these standard procedures have been adopted. The aim of this review is to describe the most common methods and their modifications for testing of antimicrobial properties of EOs and to point out the most controversial variables which can potentially affect results of the assays.

In the post antimicrobial era, increasing attention is paid towards using bacteriophage (phage in short) therapy to control antibiotic-resistant bacteria. The first step in phage therapy applications is isolating highly efficient lytic phages or phage cocktails from various sources. When a double-layer- agar with around 0.7% agar in top agar is employed, it results in a low number of phage isolation with a poor resolution, and in many cases, you miss the phage. To address this problem, a low concentration of agar in top agar is examined for better phage isolation. Here, our results proved the efficiency of isolating phage upon formulating a double-layer agar with 0.3% agar in top agar. A sewage sample was collected then phages were isolated, purified, and spotted on a top layer agar with 0.3% agar. The results showed the possibility of isolating a higher number of phages on 0.3% top agar than 0.7%. The finding advocates using 0.3% top agar for the double-layer agar, as it will provide fast, better, and easy phage screening and isolation.

In this study, a natural-based gelling agent comprised of blended flax seed gum (FSG), konjac glucomannan (KG), and agar gel (AG) was developed for application to control the textural properties of foods. The compound gels, including FSG, KG, and AG, were investigated to determine their physicochemical properties, including minimum gelling concentration, water binding capacity, water soluble index, and swelling power. In addition, we analyzed the rheological properties of the compound gel through texture analysis, frequency sweep, and creep and recovery. The microstructure of the compound gel was identified and compared with the viscoelastic properties of the gel. Overall, these results showed that the F4K6 (4:6:2 of FSG:KG:AG) could serve as an excellent gelling agent, which endowed food gel with the promoted elastic properties, water capacity, and rigid surface morphology. This work suggests that novel gelling agents, including FSG, KGM, and AG, successfully prepared food gels with improved physicochemical properties.

Antibiotic-sparing treatments are required to prevent the further emergence of anti-microbial resistance in Neisseria gonorrhoeae. Commensal Neisseria species have previously been found to inhibit the growth of pathogenic Neisseria species. For example, a previous study found that 3 out of 5 historical isolates of Neisseria mucosa could inhibit the growth of N. gonorrhoeae. In this study, we used agar overlay assays to assess if 24 circulating and historical isolates of Neisseria mucosa could inhibit the growth of 28 circulating and historical isolates of N. gonorrhoeae. Although pitting around each colony of N. mucosa created an optical illusion of decreased growth of N. gonorrhoeae, we found no evidence of inhibition (n=24). In contrast, positive controls of Streptococcus pneumoniae and Escherichia coli demonstrated a strong inhibitory effect against the growth of N. gonorrhoeae.

Background : Probiotics are composed of microorganisms that have positive effects on health when consumed in prescribed quantities. Lately, studies have attempted to establish this role of probiotics in enhancing health. Aim : The aim of this study was to compare the antifungal activity of 3 different probiotics against Candida albicans.Methodology :This was an in-vitro study. 3 types of commercially available probiotics - Bifilac, Pre pro HS and VSL-3 were cultured separately with Candida albicans and were placed under incubation. The maximum zone of inhibition was recorded for each of the probiotic solutions.Results : In this study, it was found that among the three probiotic products, Bifilac exhibited a maximum zone of inhibition of 13mm at concentration of 100µL against C. albicans, while Pre pro HS and VSL-3 showed zone of inhibition of 11mm at concentrations of 50µL and 100µL, respectively.Conclusion : Within the limits of the present study, This study could prove that probiotics can be considered as an alternative and safe treatment option for treatment of Oral candidiasis infections.

Despite advances in conservation-restoration treatments, most surface cleaning tests are subjectively evaluated. Scores according to qualitative criteria are employed to assess results, but these can vary by user and context. This paper presents a range of cleaning efficacy and homogeneity evaluation metrics for appraising cleaning trials, which minimise user bias by measuring quantifiable changes in the appearance and characteristic spectral properties of surfaces. The metrics are based on various imaging techniques (optical imaging by photography using visible light (VIS); spectral imaging in the visible-to-near-infrared (VNIR) and shortwave infrared (SWIR) ranges; chemical imaging by Fourier transform infrared (FTIR) spectral mapping in the mid-infrared (MIR) range; and scanning electron microscopy coupled with energy dispersive X-ray spectroscopy (SEM-EDX) element mapping). They are complemented by appearance measurements (glossimetry and colorimetry). As a case study showcasing the low-cost to high-end metrics, agar gel spray cleaning tests on exposed ground and unvarnished oil paint mock-ups are reported. The evaluation metrics indicated that spraying agar (prepared with citric acid in ammonium hydroxide) at a surface-tailored pH was a safe candidate for efficacious and homogenous soiling removal on water-sensitive oil paint and protein-bound ground. Further research is required to identify a gel-based cleaning system for oil-bound grounds.

Minimum Inhibitory Concentration (MIC) measures the lowest concentration of antibiotic that inhibits in-vitro growth of microorganisms. Canine pyometra is a frequently occurring reproductive disorder where reliable antibiotics along with surgical excision of uterus predicts better prognosis. Scarce literature is available to facilitate proper antibiotic selection to avoid septicemia in clinical cases. The present study was conducted to determine sensitivity and potency of selected antibacterials based on MIC values against bacterial isolates obtained from Canine Pyometra in Kathmandu district, Nepal by Agar Dilution method. Isolates obtained from 63 intra-uterine samples after ovariohysterectomy were Enterococcus spp (n=41), Klebsiella spp. (n=7), Staphylococcus spp. (n=11) and Streptococcus spp. (n=4). Enrofloxacin and Amikacin were found highly sensitive (0% resistance) with a null hypothesis of resistance to all bacterial isolates and were significantly more potent than other antibacterials tested (p<0.05; Kruskal-Wallis test) based on lower MIC ranks. Similarly, ceftriaxone was found to be least potent and was highly resistant to Enterococcus spp. (75.61%) and completely resistant to other bacterial species tested (100%). Comparative results on sensitivity and potency in the study revealed Enrofloxacin and Amikacin to be better antibacterial of choice in pyometra innervation in in-vitro setting. Further clinical trials considering other equally important variables along with MIC should be conducted to predict actual clinical efficacy of antibacterials during therapeutic management of canine pyometra in Nepal.
