Survival following Ebola virus (EBOV) infection correlates with the ability to mount an early and robust interferon (IFN) response. The host IFN-induced proteins that contribute to controlling EBOV replication are not fully known. Among the top genes with the strongest early increases in expression after infection in vivo is IFN-induced HERC5. Using a transcription- and replication-competent VLP system, we showed that HERC5 inhibits EBOV virus-like particle (VLP) replication by depleting EBOV mRNAs. The HERC5 RCC1-like domain was necessary and sufficient for this inhibition and did not require zinc finger antiviral protein (ZAP). Moreover, we showed that EBOV (Zaire) glycoprotein (GP) but not Marburg virus GP antagonized HERC5 early during infection. Our data identifies a novel ‘protagonist-antagonistic’ relationship between HERC5 and GP in the early stages of EBOV infection that could be exploited for the development of novel antiviral therapeutics.

Marburg virus disease (MVD) is a hemorrhagic fever and cause death up to 88% of people and is the same as Ebola virus, It is transmitted through skin contact or mucous membrane in the eyes, nose or mouth with blood or body fluids like urine, saliva sweat, feces and by object contaminated with body fluids. Despite these facts, yet there is no approved vaccine have been developed for the eradication of Marburg virus infections. Therefore, this study described a multi epitope-based peptide vaccine against Marburg virus viral protein 35, using several immunoinformatics tools combined with molecular docking studies. Utilizing Vaxijen 2.0 server, the V35 protein revealed to be antigenic with a score of 0.4316. Prediction of the T-cell and B-cell epitopes was then conducted. RTFDAFLGV epitope was found to be the most promising one with binding affinity to the highest numbers of MHC I alleles, a positive score in the Class I immunogenicity study and non-allergen. These results were further confirmed by the good interaction of RTFDAFLGV to the groove of HLA-A*02:01 with binding energy of -8.1 kcal/mole. Finally, the vaccine was cloned in silico to ensure its validity and efficiency of expression. To ensure its safety and immunogenic profile, in-vitro and in-vivo bioassays are recommended to confirm these findings.

Marburg virus (MARV) is a lipid-enveloped negative sense single stranded RNA virus, which can cause a deadly hemorrhagic fever. MARV encodes seven proteins, including VP40 (mVP40), a matrix protein that interacts with the cytoplasmic leaflet of the host cell plasma membrane. VP40 traffics to the plasma membrane inner leaflet, where it assembles to facilitate the budding of viral particles. VP40 is a multifunctional protein that interacts with several host proteins and lipids to complete the viral replication cycle, but many of these host-interactions remain unknown or are poorly characterized. In this study, we investigated the role of a hydrophobic loop region in the carboxy-terminal domain (CTD) of mVP40 that shares sequence similarity with the CTD of Ebola virus VP40 (eVP40). These conserved hydrophobic residues in eVP40 have been previously shown to be critical to plasma membrane localization and membrane insertion. An array of cellular experiments and confirmatory in vitro work strongly suggests proper orientation and hydrophobic residues (Phe281, Leu283, and Phe286) in the mVP40 CTD are critical to plasma membrane localization. In line with the different functions proposed for eVP40 and mVP40 CTD hydrophobic residues, molecular dynamics simulations demonstrate large flexibility of residues in the EBOV CTD whereas conserved mVP40 hydrophobic residues are more restricted in their flexibility. This study sheds further light on important amino acids and structural features in mVP40 required for its plasma membrane localization as well as differences in the functional role of CTD amino acids in eVP40 and mVP40.

During virus outbreaks in the recent past web behavior mining, modeling, and analysis have served as means to examine, explore, interpret, assess, and forecast the worldwide perception, readiness, reactions, and response linked to these virus outbreaks. The recent outbreak of the Marburg Virus disease (MVD), the high fatality rate of MVD, and the conspiracy theory linking the FEMA alert signal in the United States on October 4, 2023, with MVD and a zombie outbreak, resulted in a diverse range of reactions in the general public which has transpired in a surge in web behavior in this context. This resulted in “Marburg Virus” featuring in the list of the top trending topics on Twitter on October 3, 2023, and “Emergency Alert System” and “Zombie” featuring in the list of top trending topics on Twitter on October 4, 2023. No prior work in this field has mined and analyzed the emerging trends in web behavior in this context. The work presented in this paper aims to address this research gap and makes multiple scientific contributions to this field. First, it presents the results of performing time series forecasting of the search interests related to MVD emerging from 216 different regions on a global scale using ARIMA, LSTM, and Autocorrelation. The results of this analysis present the optimal model for forecasting web behavior related to MVD in each of these regions. Second, the correlation between search interests related to MVD and search interests related to zombies (in the context of this conspiracy theory) was investigated. The findings show that there were several regions where there was a statistically significant correlation between MVD-related searches and zombie-related searches (in the context of this conspiracy theory) on Google on October 4, 2023. Finally, the correlation between zombie-related searches (in the context of this conspiracy theory) in the United States and other regions was investigated. This analysis helped to identify those regions where this correlation was statistically significant.
