A nanostructured electrochemical bi-sensor system for analysis of milks has been developed using the Layer by Layer technique. The non-enzymatic sensor [CHI+IL/CuPcS]2, is a layered material containing a negative film of the anionic sulfonated copper phthalocyanine (CuPcS) acting as electrocatalytic material, and a cationic layer containing a mixture of an ionic liquid (IL) (1-butyl-3-methylimidazolium tetrafluoroborate) that enhances the conductivity and chitosan (CHI) that facilitates the enzyme immobilization. The biosensor ([CHI+IL/CuPcS]2-GAO) results from the immobilization of galactose oxidase on the top of the LbL layers. FTIR, UV-vis and AFM have confirmed the proposed structure and cyclic voltammetry has demonstrated the amplification caused by the combination of materials in the film. Sensors have been combined to form an electronic tongue for milk analysis. Principal Component Analysis has revealed the ability of the sensor system to discriminate between milk samples with different lactose content. Using PLS-1 calibration models, correlations have been found between the voltammetric signals and chemical parameters measured by classical methods. PLS-1 models provide excellent correlations with lactose content. Additional information about other components such as fats, proteins and acidity can also be obtained. The method developed is simple and the short response time permits its use in assaying milk samples on-line.

The detection of hormones, pharmaceutical and personal care products (PPCPs), in aqueous complex media as water bodies is an urgent and mandatory matter because of its harmful impact on the environment. This work presents a sensor device which is able to detect trace of the synthetic hormone 17α-ethinylestradiol (EE2), in a range of concentration of 10-15 to 10-9 M in both mineral and tap waters. These types of water were studied due to their different degrees of complexity, which allow to attain a better understanding of EE2’s behavior and response in different media. The sensor body consists of solid supports having interdigitated electrodes without and with a thin film deposited on it, and device response is evaluated through impedance spectroscopy. The thin films were prepared by the layer-by-layer technique from aqueous solutions of polyethylenimine (PEI) and poly(sodium 4-styrenesulfonate) (PSS) polyelectrolytes. Results demonstrated that it is possible to discriminate clearly the different waters used and to detect EE2, obtained from the second principal component (PC2) as a result of the application of principal component analysis method, with a sensitivity of -0.072 ± 0.009 and -0.44 ± 0.03 per decade of concentration, for mineral and tap water, respectively. Detection limits values were found to be of 8.6 fM (2.6 pg/L) and of 7.5 fM (22.2 pg/L) for TW and MW, respectively and are lower than the ones found in literature. Moreover, the achieved constant proportionality between the PC2 values for each EE2 concentration reveals that one can use this methodology to quantify EE2 concentration in unknown aqueous complex matrices.

The design of multifunctional microcarriers has attracted significant attention because they combine various functions within a single system. In this study, we developed a set of multi-layered hydrogel microcarriers, which were first loaded with chemotherapeutic curcumin (CUR), then using the layer-by-layer (LbL) technique, coated through a polyelectrolyte shell consisting of chitosan (CHIT) or poly(allylamine hydrochloride) (PAH). As an outer layer with antimicrobial function, newly synthesised alkylene quaternary ammonium salt functionalised polyelectrolytes (A-QAS-PEs) were applied. For this purpose, poly(acrylic acid) (PAA) was decorated with dif-ferent hydrophobic side chains (n-hexane and n-dodecane side entities) and different degrees of substitution (m) of quaternary ammonium groups (abbreviated as PAA-C(O)O-(CH2)n-N+(CH3)3(m); n=6,12; m=8-14%). The grafting approach of PAA with the al-kylene quaternary ammonium salt moiety was performed under mild reaction conditions using Steglich esterification followed by quaternisation. The structure of antimicrobial decorated PAA was confirmed by 1H NMR and FTIR, and the mean diameter of all multifunctional microparticles was characterised by SEM. The viscoelastic properties of the functional layers were studied using QCM-D. The release of CUR from the microcarriers was described using a hybrid model, i.e., a combination of first-order kinetics and the Korsmeyer-Peppas model. The antimicrobial activity of functionalised PAA and multilayered CUR-loaded hydrogel microcarriers with quaternary am-monium function was assessed against Staphylococcus aureus and Serratia marcescens by the agar diffusion assay method. Only a limited inhibition zone of PAA was observed, but in the case of both antimicrobial decorated PAA and the corresponding multilayered nanocarriers, the inhibi-tory activity increase was achieved against both strains of bacteria.
