A rapid and simple high-performance liquid chromatography (HPLC) method using extraction with zinc sulfate has been developed for the determination of Casiopeina III-ia (Cas III-ia) and validated over the linear range 10-120 g/mL in 200 L of rabbit blood. Previously we have reported a HPLC method to quantify CasIII-ia in plasma, but in distribution studies in total blood [20], the Cas III-ia concentrations were higher in total blood that in plasma [19]. The analysis was performed on a Symmetry C (18) (5 microm) column. The mobile phase was Methanol-sodium phosphate buffer (pH 6.5; 0.01 M) (40:60 v/v) was kept at a flow-rate of 0.8 ml/min. The analyses were performed at room temperature. The column effluent was monitored at 262 nm. Acetaminophen was used as internal standard. The results showed that the assay is sensitive at 10 g/mL. Maximum intra-day coefficient of variation was 5.10%. The average recovery obtained in blood was 94.51%. The applicability of this method for distribution in vitro and pharmacokinetic studies in rabbits was demonstrated. Conclusions: the present assay is rapid, simple, precise, and accurate. The pharmacokinetic study was carried out in rabbits and the following pharmacokinetic parameters were obtained: (kel) = 0.0150 min-1, half-life time (T1/2) = 53.92 min, apparent volume of distribution (Vd) = 202.81 mL, clearance (Cl) = 2.08 mL /mi and area under the curve (AUC)=23163.85 µg/mL.min. Contributing to the preclinical characterization of the Cas III-ia.

The aim of this study, for determination of verteporfin in real samples (simulated body fluid, and simulated tears, 0.9% isotonic sodium chloride solution, Lactated Ringer IV solution for infusion, 5% dextrose IV solution for infusion, lemon juice and drinking water) was the method validation and to examined by HPLC-DAD-UV. Metod validation parameters such as specificity, linearity, precision, accuracy, robustness, limit of detection (LOD) and limit of quantitation (LOQ) for verteporfin were validated and developed according to the International Conference on Harmonization (ICH) Q2 R1 guidelines. The LOD and LOQ for verteporfin were found 0.06 µg/L and 0.2 µg/L, respectively. The recovery values of the optimization and validation for verteporfin were found in the range of 97.5-100.7%. The relative standard deviations (RSD) for vertepofin were <1%. The developed method was successfully applied to real samples with high accuracy and the recoveries (%) from real samples were 99.9, 100, 98.2, 99.2, 99.4, 98.8 and 99.4, respectively.

An HPLC method for indicating stability was developed, created, and confirmed for the quantification measurement of recombinant human erythropoietin in large quantities as well as dosage forms. An isocratic separation was accomplished using a Thermo Biobasic C18 (250 mm 4.6 mm i.d., 300Ao, 5 m) column with a flow rate of 1.0 mL min-1 as well as a UV detector to monitor the eluate at 278 nm. Acetonitrile, 0.05 mM potassium dihydrogen phosphate (80:20 v/v), and orthophosphoric acid adjusted to 4.0 made up the mobile phase. The drug was exposed to oxidative stress, hydrolysis, and photodegradation to simulate stress conditionsRecombinant Human Erythropoietin, the parent compound, was eluted at roughly 6.675 minutes, as well as all byproducts have been entirely disconnected inside an overall analytical run time of about 15 minutes. To identify quantification limits of 0.05 and 0.2 g mL-1, respectively, the method was linear over a concentration range of 1-6 g mL-1 (r = 0.9989). Recombinant Human Erythropoietin can be measured accurately, selectively, sensitively, and precisely using this method both in dosage form and in bulk. Stress-induced degradation products did not interact with the identification of Recombinant Human Erythropoietin, indicating that the assay is stable.

Cyanobacterial mass developments in eutrophic ponds and lakes are a major concern for lake management, as many cyanobacteria produce a huge variety of toxic secondary metabolites, e.g. microcystins. The aim of this research was to culture a strain of the cyanobacterium Microcystis sp strain BM25, to observe its biomass production and to isolate and purify protease inhibitors from this cyanobacterial biomass. Different secondary metabolites were isolated following a standard bioassay-guideline. Isolation was performed, with an enzymatic protease assay as bioassay. High performance liquid chromatography was used to identify different fractions of secondary metabolite from the strain BM25. Moreover, protease homogenates were isolated from Daphnia magna in order to test the inhibitors against naturally occurring major digestive proteases trypsin and chymotrypsin. It was measured that 60% MeOH and the 80% MeOH C18-SPE fraction inhibits chymotrypsin activity 98% (6 nmol pNA min^-1 mg^-1) and 99 % (4 nmol pNA min^-1 mg^-1), respectively. In contrast, trypsin activity was not inhibited by methanolic extracts of this cyanobacterium strain.

The aim of this study was characterization of some dairy drinks based on Milk Serum regarding major whey proteins (WP) and free amino acids (FAAs) using reversed phase high performance liquid chromatographic (RP-HPLC) methods. The studied WP, -lactalbumin (-La), bovine serum albumin (BSA), -lactoglobulin A (-Lg A) and -lactoglobulin B (-Lg B) were separated on Aeris XB-C18 column at 214 nm detection. The RP-HPLC method was validated by selectivity, linearity (R2 ≥0.99), sensitivity (LOQ, 1.35–10.08 µg mL−1), accuracy (recovery 96.79-103.07%) and precision (% RSD ≤ 4.13%). The total studied WP in studied dairy drinks varied between 1.42 and 3.047 g·L-1. The chromatographic profile of FAAs (aspartic acid, glutamic acid, serine, histidine, arginine, glycine, threonine, alanine, tyrosine, cysteine, tryptophan, methionine, valine, phenylalanine, isoleucine, leucine and lysine) was determined in lyophilized concentrate of Milk Serum by RP-HPLC using pre-column derivatization reaction with orthophthalaldehyde (OPA). The total studied FAAs in studied samples varied between 1.103 and 1.119 mg·g-1. Moreover, the Milk Serum showed bacteriostatic activity against two bacterial strains Escherichia coli and Staphylococcus aureus. The obtained results confirm that dairy drinks based on the Milk Serum constitutes a valuable sources of bioactive components with benefits for human healthy nutrition.

The leaves of Ligustrum robustum have been applied as Ku-Ding-Cha, a functional tea to clear heat and remove toxins; and served as a folk medicine to deal with diabetes, obesity, and hypertension. Trans-p-hydroxycinnamic acid and its esters, which were isolated from the leaves of L. robustum, might be a new resource to prevent diabetes and its complications because of their strong -glucosidase and fatty acid synthase (FAS) inhibitory activities and antioxidant effect. However, the quantitative analytical method for trans-p-hydroxycinnamic acid and its esters in the leaves of L. robustum hasn’t been reported so far. In addition, it was difficult and troublesome to analyze no less than 34 trans-p-hydroxycinnamic acid esters by usual HPLC. Therefore, a new HPLC-UV method using hydrolyzation with sodium hydroxide for quantitation of trans-p-hydroxycinnamic acid and its esters in the leaves of L. robustum was developed in the present study. The optimal hydrolyzation conditions were as follows: 1 mL sodium hydroxide aqueous solution (1 M) was added into 1 mL the extract of L. robustum and incubated at 80 °C for 2 h , and then 1 mL hydrochloride (1 M) solution was added. HPLC analysis was performed in reverse phase mode using a C-18 column, eluting with methanol-0.1% acetic acid aqueous solution (40:60, v/v) in isocratic mode at a flow rate of 1.0 mL/min, and detecting at 310 nm. The analytical method was simple and rapid, which simplified greatly the analytical process. The method was validated in terms of linearity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy. High linearity was demonstrated with correlation coefficient of 1.000 for trans-p-hydroxycinnamic acid. The LOD and LOQ values were 1.55 and 4.69 μg/mL, respectively. The relative standard deviations (RSD) of intra-day and inter-day variabilities for trans-p-hydroxycinnamic acid were less than 2%. The percentage recovery of trans-p-hydroxycinnamic acid was 101.2% ± 0.8%. Finally, the proposed method was applied successfully to the determination and comparison of the contents of trans-p-hydroxycinnamic acid and its esters in various extracts of the leaves of L. robustum. The 60%-70% ethanol extracts of L. robustum showed the highest contents of free trans-p-hydroxycinnamic acid (3.96-3.99 mg/g), and the 50%-80% ethanol extracts of L. robustum displayed the highest contents of trans-p-hydroxycinnamic acid esters (202.6-210.6 mg/g). The method can be applied also to the quality control of the products of L. robustum.

The surge in popularity of Delta-8 Tetrahydrocannabinol (D8-THC) products, coupled with a lack of stringent regulation and oversight, has given rise to concerns regarding consumer safety and compliance with existing laws. This article delves into the challenges posed by the unregulated sale of D8-THC products, emphasizing the potential for contamination with prohibited constituents, including Delta-9 Tetrahydrocannabinol (D9-THC). The 2018 Farm Bill dictates that D9-THC concentrations must not exceed 0.3%, necessitating precise testing methods for accurate validation. Existing testing procedures, particularly involving High-Performance Liquid Chromatography (HPLC), face difficulties in distinguishing between D8 and D9 peaks, necessitating adjustments in parameters to enhance accuracy. The development of more refined testing methodologies is crucial for companies to ensure compliance, prevent adverse health effects, and provide consumers with accurately characterized cannabinoid profiles in the products they purchase.

The aim of this study was to investigate the parameters affecting the extraction of positive molecules such as chlorogenic acids and antioxidants, as well as potentially carcinogenic substances such as acrylamide, in different coffee brewing methods. Three coffee varieties, each assigned to different roasting degree, were used to prepare coffee brews following eight diverse preparation methods. Acrylamide was quantified by HPLC-MS/MS instrument while chlorogenic acids and caffeine by HPLC-DAD system. Three spectrophotometric analyses were also performed (DPPH, TFC and TPC) to evaluate antioxidant activity as well. The results showed that the main brewing parameters, that have the greatest influence on the final content of these compounds were the volume of water used, more specifically the brewing ratio (coffee to water ratio), the extraction time and the particle size of the coffee powder.

Abstract: Background: The analysis of vitamins in baby food is a challenging task given the com-plexity of the food matrix, vitamin stability, and strict regulations of the European Union regarding permissible deviations from declared values. Vitamins in food exist in different concentrations and forms and have different stabilities, and it is not straightforward to prepare samples for reliable analysis using the same procedure. Therefore, significant attention has been devoted to optimizing sample preparation in the analysis of vitamins. Methods: This study aims to determine which of the seven applied extraction methods is the most efficient for the simultaneous extraction and deter-mination of A, D, E, and K vitamins in milk and baby food using high-performance liquid chro-matography (HPLC). Different samples of baby food were prepared in seven different ways based on four methods (saponification, enzymatic hydrolysis, solvent extraction, and solid-phase extrac-tion). Results and Conclusion: The best preparation method proved to be solid-phase extraction with a C18 stationary phase, and HPLC with a UV-VIS detector was identified as a sufficiently sensitive technique for the identification and quantification of fat-soluble vitamins in milk and baby food.

Under the Japanese health insurance system, medicines undergoing therapeutic drug monitoring (TDM) can be billed for medical fees if they meet the specified requirements. In Japan, TDM of vancomycin, teicoplanin, aminoglycosides, and voriconazole, which are used for the treatment of infectious diseases, is common practice. This means the levels of antimicrobial agents are measured in-house using chromatography. This review describes personalized medicine based on the use of chromatography as a result of the current situation in Japan.

We cultured Parmelina carporrhizans and P. quercina in Corn Meal Agar and 0.2% glucose Malt Yeast Agar for 160 days. Chemosyndrome of natural thalli and mycobiont cultures were analyzed by HPLC. Lecanoric acid, atranorin, chloratranorin and ergosterol were detected in P. carporrhizans thalli, while lecanoric acid, chloratranorin and aliphates were found in P. quercina thalli. The secondary methabolites pattern between thalli and mycobiont culture was completely different in both species. Both species secreted the phenalenone myeloconone C in culture media and was also detected in P. quercina mycobiont aggregates. Interestingly, the phenolic compounds produced by the mycobiont culture of P. carporrhizans are related to those produced by natural thallus by the same biosynthetic pathway, while the chemosyndrome of P. quercina mycobiont implies switch of biosynthetic pathway from acetate-polymalonate pathway to shikimic acid pathway, with pulvinic acid as major compound of mycobiont culture. The role of Myelochonone C, confluentic acid and pulvinic acid produced by mycobiont culture is discussed as possible adaptive vantage in field as photoprotective agent or as byproduct result of stressing artificial culture conditions.

The degradation of the persistent compound perfluorooctanoic acid (PFOA) in water by ultraviolet light in the presence and absence of hydrogen peroxide was examined. A laboratory experiment was designed to answer the question: is UV spectrophotometry feasible for PFOA research? Spectrophotometry was able to measure the degradation of PFOA, but a very high concentration was required. The 1 mg/L used is very concentrated when compared to the 70 ng/L suggested level in drinking water. A UV/Visible diode array scanning spectrophotometer was used to measure changes in PFOA concentrations after 15-minute exposures to ultraviolet light and ultraviolet light with hydrogen peroxide. A noticeable drop in concentration was measured for a 1 mg/L sample. The reaction kinetics were calculated and are likely skewed due to the greater concentration of experimental PFOA. These preliminary results will allow for the next phases of PFOA research to commence, including confirmation through liquid chromatography/mass spectrometry of low concentrations and developing bioremediation techniques.

Cannabis (Cannabis Sativa L.) is now legally produced in many regions worldwide. Cannabis flourishes under high light intensities (LI); making it an expensive commodity to grow in controlled environments, despite its exceptionally high market value. It is commonly believed that cannabis secondary metabolite levels may be enhanced both by increasing LI and by exposing crops to ultraviolet radiation (UV). However, there is sparse scientific evidence to guide cultivators. Therefore, the impact of LI and UV on yield and quality must be elucidated to enable cultivators to optimize their lighting protocols. We explored the effects of LI, ranging from 350 to 1400 μmol m^-2 s^-1 and supplemental UV spectra on cannabis yield and potency. There were no spectrum effects on inflorescence yield, but harvest index under UVA+UVB was reduced slightly (1.6%) vs. the control. Inflorescence yield increased linearly from 19.4 to 57.4 g/plant and harvest index increased from 0.565 to 0.627, as LI increased from 350 to 1400 μmol m^-2 s^-1. Although there were no UV spectrum effects on total equivalent Δ⁹-tetrahydrocannabinol (T-THC) content in leaves, the neutral form, THC, was 30% higher in UVA+UVB vs. control. While there were no LI effects on inflorescence T-THC content, the content of the acid form (THCA) increased by 20% and total terpenes content decreased by 20% as LI increased from 350 to 1400 μmol m^-2 s^-1. High LI can substantially increase cannabis yield and quality, but we found no commercially-relevant benefits of adding supplemental UV radiation to indoor cannabis production.

Review of the existing bibliography shows that the direction and magnitude of the long-term trends of UV irradiance, and their main drivers, vary significantly throughout Europe. Analysis of total ozone and spectral UV data recorded at four European stations during 1996 – 2017 reveals that long-term changes in UV are mainly driven by changes in aerosols, cloudiness, and surface albedo, while changes in total ozone play a less significant role. The variability of UV irradiance is large throughout Italy due to the complex topography and large latitudinal extension of the country. Analysis of the spectral UV records of the urban site of Rome, and the alpine site of Aosta reveals that differences between the two sites follow the annual cycle of the differences in cloudiness and surface albedo. Comparisons between the noon UV index measured at the ground at the same stations and the corresponding estimates from the DWD forecast model and OMI/Aura observations reveal differences of up to 6 units between individual measurements, which are likely due to the different spatial resolution of the different datasets, and average differences of 0.5 – 1 unit, possibly related to the use of climatological surface albedo and aerosol optical properties in the retrieval algorithms.

The analysis of drugs is a priority in public health, particularly antibiotics which develop resistance due, among other things, to bad dosages or poor quality of the active substance. In this study, we evaluated eight antibiotics available in health centers in Djibouti through qualitative and quantitative analysis. The tests were based on high-purity reference products. In qualitative analysis by HPLC, only amoxicillin presented two impurity peaks. This observation is found in the dosage of this active ingredient with a value of 83 ±0.6 mg instead of the 500 mg indicated on the label. It will be necessary to diversify the sample’s provenance to seek the origin of this degradation.

(1) Background: ganoderic acids (GAs) are specific triterpenes of Ganoderma lucidum. The HPLC fingerprint profile of GAs of the fruiting body has been well-known, but the mycelial fingerprinting remains unclear. (2) Methods: the ethanol extract of the mycelium of G. lucidum (YK-01) was further purified by preparative HPLC. The HPLC spectra for four strains of G. lucidum and one strain of G. formosanum (purple lingzhi) were investigated. (3) Results: nineteen lanostane triterpenes, including five new triterpenes, GA-TP (1), ganodermic acid Jc (GmA-Jc) (2), GmA-Jd (3), GA-TQ1 (4), and ganoderal B1 (5), and fourteen known triterpenes 6-19 were isolated from the ethanol extract. A green chemical HPLC analytical method was developed by using ethanol and acetic acid as mobile phase. These triterpenes comprise of a unique HPLC triterpenoid spectral pattern of G. lucidum mycelium. All four G. lucidum strains showed the same HPLC spectral pattern, whereas G. formosanum displayed a different pattern. Quantitation methods for ganoderic acid T (10) and S (12) were also validated. (4) Conclusions: the triterpenoid HPLC analytical method can be used to identify G. lucidum species, and to determine the contents of GA-T and GA-S.

The extracts of Garcinia mangostana L. (mangosteen) are traditionally used for medicinal purposes in Asia. Phenols and xanthones are the two main phenolic compounds with anti-inflammatory activities in the pericarps of mangosteen. A simple and economic reverse-phase high-performance liquid chromatography (RP-HPLC) method has been developed for the simultaneous quantification of phenols (catechin, epicatechin, and procyanidin B2) in mangosteen. The mobile phase was acetonitrile: water (12:88) at 30°C, the flow rate was 1 mL/min, and the detection wavelength was 230 nm. The results showed good linear relationships for catechin from 50-250 ng and for epicatechin and procyanidin B2 from 250-1250 ng. The RP-HPLC fingerprint determination method of total xanthones in mangosteen was also established to provide a new method for quality control of mangosteen extract. The mobile phase was a methanol-water gradient elution at 30°C, the flow rate was 1 mL/min, and the detection wavelength was 320 nm. The similarity of fingerprints of 10 batches of total xanthones was more than 0.90. These methods had good precision, reproducibility, and stability and can be applied in the quality control of mangosteen extracts for medicinal purposes.

A simple, isocratic and robust RP-HPLC method for the analysis of azithromycin was developed, validated and applied for the analysis of bulk samples, tablets and suspensions. The optimum chromatographic conditions for separation were established as mobile phase comprising of acetonitrile-0.1M KH₂PO₄ pH 6.5-0.1M tetrabutyl ammonium hydroxide pH 6.5-water (25:15:1:59% v/v/v/v) delivered at a flow rate of 1.0 ml/min. The stationary phase consisted of reverse-phase XTerra^® (250 mm× 4.6 mm i.d., 5 µm particle size) maintained at a temperature of 43 °C with a UV detection at 215 nm. The method was found to be linear in the range 50-150% (r²=0.997). The limits of detection and quantification were found to be 0.02% (20 µg) and 0.078% (78 µg) respectively with a 100.7% recovery of azithromycin. Degradation products of azithromycin in acidic and oxidative environments at 37 °C were resolved from the active pharmaceutical ingredient and thus the method is fit for the purpose of drug stability confirmation.

Eyes are quintessential complex traits and our understanding of their evolution guides models of trait evolution in general. A long-standing account of eye evolution argues natural selection favors morphological variations that allow increased functionality for sensing light (Darwin 1859; v. Salvini-Plawen and Mayr 1977; Nilsson and Pelger 1994; Nilsson 2013). While certainly true in part, this focus on visual performance does not entirely explain why diffuse photosensitivity persists even after eyes evolve, or why eyes evolved many times, each time using similar building blocks. Here we briefly review a vast literature indicating most genetic components of eyes historically responded to stress caused directly by light, including UV damage of DNA, oxidative stress, and production of aldehydes. We propose light-induced stress had a direct and prominent role in the evolution of eyes by bringing together genes to repair and prevent damage from light-stress, both before and during the evolution of eyes themselves. Stress-repair and stress-prevention genes were perhaps originally deployed as plastic responses to light and/or as beneficial mutations genetically driving expression where light was prominent. These stress-response genes sense, shield, and refract light but only as reactions to ongoing light stress. Once under regulatory-genetic control, they could be expressed before light stress appeared, evolve as a module, and be influenced by natural selection to increase functionality for sensing light, ultimately leading to complex eyes and behaviors. Recognizing the potentially prominent role of stress in eye evolution invites discussions of plasticity and assimilation and provides a hypothesis for why similar genes are repeatedly used in convergent eyes. Broadening the drivers of eye evolution encourages consideration of multi-faceted mechanisms of plasticity/assimilation and mutation/selection for complex novelties and innovations in general.

With the increasing pace of global warming, it is important to understand the role of meteorological factors in influenza virus (IV) epidemics. In this study, we investigated the impact of temperature, UV index, humidity, wind speed, atmospheric pressure, and precipitation on IV activity in Norway, Sweden, Finland, Estonia, Latvia and Lithuania during 2010-2018. Both correlation and machine learning analyses revealed that low temperature and low UV indexes were the most predictive meteorological factors for IV epidemics in the Northern European countries. Our in vitro experiments confirmed that low temperature and UV radiation preserved IV infectivity. Associations between these meteorological factors and IV activity could improve surveillance and promote development of accurate predictive models for future influenza outbreaks in Northern Europe.

Furthermore, catechins have diverse applications across various industries, including food and beverage, cosmetics, agriculture, and materials science. This abstract emphasizes the importance of continued research and innovation in catechin extraction and utilization, which hold promise for advancing human health and technological progress across multiple domains.

Type 2 Diabetes Mellitus (T2DM) is one of the most common metabolic disorders, with a major involvement of oxidative stress in its onset and progression. Pioglitazone (Pio) is an antidiabetic drug which primarily works by reducing insulin resistance while Curcumin (Cur) is powerful antioxidant with o important hypoglycemic effect, also. The both drugs are associated with several drawbacks, such as reduced bioavailability, a short half-life time (Pio) as well as instability and poor water solubility (Cur), which limit their therapeutic use. In order to overcome these disadvantages, a new co-delivery (Pio, Cur) nanosystem based chitosan (CS) (CS-Pio-Cur NPs), was developed, and characterized in reference with simple nanosystems (CS-Pio NPs, CS-Cur NPs). The developed CS-based NPs (CS-Pio-Cur NPs, CS-Pio NPs, CS-Cur NPs) were physico-chemical characterized in terms of particle size (PS), entrapment efficiency (EF%) and loading capacity (LC%). The CS-Pio-Cur NPs proved increased or similar values of EE (88.95% ± 7.79 for Cur; 94.83% ± 9.89 for Pio) and LC% (24.25 % ± 1.62 for Cur; 3.5% ± 0.98 for Pio), in reference with simple nanosystems, CS-Cur NPs (EE = 82.46% ± 1.74; LC = 20.34% ± 0.94) and CS-Pio NPs) (EE = 96.31% ± 0.68; LC = 4.11% ± 0.47), respectively. The identification of APIs (Cur, Pio) loaded into CS matrix was performed using FR-IR spectroscopy and for simultaneously quantification of APIs (Cur, Pio) released from CS-Pio-Cur NPs, a HLPC method was developed and validated. Based on release study performed in different simulated fluids (SGF, SIF, SCF) it can be appreciated that developed CS-APIs NPs have favorable pharmacokinetic profile with good release in gastrointestinal tract.

Sphingolipids are ubiquitous in eukaryotes and certain prokaryotes, where they serve as vital components of biological membranes and as bioactive molecules. Chloroplasts have complex membrane structures that play crucial roles in photosynthesis, but their specific sphingolipidome remains unreported. In this study, we used high-performance liquid chromatography-mass spectrometry (HPLC-MS/MS) to analyze the sphingolipidome of purified Arabidopsis thaliana chloroplasts. We detected 92 chloroplast sphingolipids. The chloroplast sphingolipidome differed from total leaf (TL) samples, having a higher content of free long-chain bases and hydroxyceramides, and a greater proportion of complex sphingolipids with 16C fatty acid (FA) forms. Notably, chloroplast glucosylceramides were predominantly the d18:1 h16:0 and t18:1 h16:0 forms, rather than the 24C FA form found in TL and other cellular structures. Comparing the sphingolipidomes of different cellular structures underscores the inhomogeneity of intracellular distribution of sphingolipids. This provides a robust reference for further elucidating the function of sphingolipids in plant cells.

In searching for compounds with antioxidant and antifungal activity, our study focused on the subshrub species Empetrum rubrum Vahl ex Willd. (Ericaceae), commonly known as Brecillo or Murtilla de Magallanes. We measured the antioxidant activity of its methanolic extract (MEE) obtained from the aerial parts (leaves and stems). The antioxidant activity of MEE was evaluated in vitro by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) free radical and 2,2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) cationic radical. Results were expressed in gallic acid and Trolox equivalents for DPPH and ABTS assays, respectively. The antioxidant activities, for the DPPH and ABTS assays, were also calculated as an inhibitory concentration of extract needed to inhibit 50% of the absorbance (IC50 value). The results of IC50 values measured by the DPPH and ABTS methods were 0.4145 mg mL-1 and 0.1081 mg mL-1, respectively. In relation to the antioxidant activity, the total phenolic content (TPC) in MEE was determined by the Folin-Ciocalteu method, and the presence of secondary metabolites groups, including phenolic compounds, was determined by using different standardized test procedures. Phenolic compounds contained in leaves, stems, and fruits of E. rubrum were determined qualitatively by means of high-performance liquid chromatography coupled with mass spectrometry (HPLC-MS) analysis. The antifungal activity of MEE obtained from aerial parts of E. rubrum was tested against Rhizoctonia solani. In vitro, mycelial growth of this fungus was reduced from 90% to 100% in the presence of MEE.

Leucobryum is a moss genus that exhibits various bioactivities. However, the identification of Leucobryum species with morphology alone remains difficult. Nowadays, chemical profile analysis provides additional tools for plant classification, especially in medicinal plants. This method uses chemical similarities to identify the differences among some plants, especially for the different varieties of the same species. The objective of this study was to obtain the chemical profiles and use chemometrics to identify selected Leucobryum species found in Thailand. Lipophilic extracts from 18 samples of five taxa were analyzed with TLC and HPLC. Cluster analysis and PCA were used to investigate the similarities of the chemical profiles. PERMANOVA was performed to determine the difference among taxa. The results showed that the TLC profile could separate only the genus of the sample, while HPLC chromatograms showed separation among the Leucobryum taxa and consistent patterns within the same species. In addition, the chemical profiles of Leucobryum species were separated at the species level in PCA. The PERMANOVA showed significantly different profiles among the species (P = 0.002). Therefore, this work illustrated the potential of chemometrics as supporting evidence for species delimitation in the moss genus Leucobryum.

An especially important species in southern Brazil, yerba mate (Ilex paraguariensis) is traditionally consumed as tea, chimarrão and tererê. Yerba mate consumption has been stimulated by scientific discoveries that have identified high concentrations of bioactive compounds and their health benefits. There are no studies or scientific evidence to certify or guarantee stability of these compounds’ concentrations during different years in the same plants. We were interested in quantifying caffeine, theobromine, total phenolic compounds and proteins concentrations in leaves of yerba mate genotypes and their stability in the same plants over four consecutive years. Mature leaves from yerba mate genotypes selected on a provenance and progenies trial were collected in August of 2015, 2016, 2017 and 2018. Methylxanthines (caffeine and theobromine), total phenolic compounds and total protein contents were quantified. Our results indicate large variations between genotypes regarding caffeine (0.035 to 2.385 g 100 g^-1), theobromine (0.0004 to 1.772 g 100 g^-1), total phenolic compounds (7.028 to 9.424 g 100 g^-1) and proteins (10.39 to 16.58 g 100 g^-1) contents, and also high stability of those compounds over the four evaluated years. A great variation of the studied bioactive compounds in different Ilex paraguariensis genotypes was found and its stability over four consecutive years was showed.

Plaque psoriasis is a common, long-lasting illness that affects the immune system and causes significant negative impacts on a patient's physical health, well-being, and ability to work effectively. Deucravacitinib (DEU) is the first oral medication used in the treatment of plaque psoriasis, a chronic skin condition that causes red, scaly patches on the skin. DEU is a type of medication called an oral Janus kinase (JAK) inhibitor, which works by blocking specific enzymes that play a role in the inflammation and immune response associated with psoriasis. Therefore, a quick, easy, novel, reliable, sensitive, and straightforward Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS) approach was used to analyse DEU in plasma samples. The LC-MS/MS method for the determination of DEU in human plasma was based on using trimethoprim as an internal standard (IS). The separation of DEU and IS was carried out via liquid-liquid extraction (LLE). The isolated substances were then subjected to the chromatographic system using the ACE-C18 column (4.6x100 mm, 5 µm). The mobile phase employed consisted of methanol and a solution of 2 mM ammonium formate (80:20 v/v, respectively). The flow rate used was set at 0.9 mL min-1. The creative strategy was performed by running an ABSCIEX API 4000 mass spectrometer with an electron spray ionization source in Multiple Reaction Monitoring (MRM) modes. The ion transitions m/z 426.3  358.2 was used for DEU quantitation, while the ion transitions m/z 291.1  261.1 was used for trimethoprim quantitation. The accuracy, precision, linearity, recovery, and selectivity of DEU were deemed acceptable when validated for a concentration range between 0.500 to 601.050 ng/mL, utilizing a weighting factor of 1/x2.

Artic root is a well-known plant adaptogen with multipotential pharmacological properties. Thin-layer chromatography (TLC) – screening followed by diode-array high-performance liquid chromatography and nuclear magnetic resonance spectroscopy proved to be a reliable and convenient method for simultaneous determination of quality of various herbal raw materials and supplements. This combination allowed for comparing and differentiating arctic root samples as well as defining their authenticity. The study provided information on the chemical and biological properties of the seven chosen samples as well as qualitative and quantitative evaluation of the quality markers: rosavin, salidroside, and p-tyrosol. The absence of rosavin, salidroside, and p-tyrosol in three samples was detected using TLC-screening and confirmed by HPLC-DAD and NMR. The paper highlighted the importance of quality control and strict regulation for herbal medicine supplements and preparations.

Nonsteroidal anti-inflammatory drugs (NSAIDs), which block the activity of cyclooxygenase (COX) isoenzymes and inhibit the synthesis of prostaglandin, have been used for pain relief. We have developed a method to separate a mixture of three NSAIDs, such as aspirin, paracetamol, and naproxen, using reverse-phase high-performance liquid chromatography (RP-HPLC). An isocratic mobile phase consisting of acidic water and acetonitrile was selected to run at a low flow rate, such as 0.8 mL/min. The mixture of three NSAIDs was injected at a low volume into a C18 column that was 150 mm in length and characterized using a UV detector at 230 nm. We identified three peaks in the chromatogram indicating the three compounds. The elution time of the peaks was less than 10 minutes. To identify multiple peaks on the isocratic flow using a short column, further studies are required regarding the proposed method to generate microfluidic devices for nanoLC.

Biphasic oily/water nanoemulsions have been proposed as delivery systems for the intranasal administration of curcumin (CUR) and quercetin (QU), due to their high drug entrapment efficiency, the possibility of simultaneous drug administration and protection of the encapsulated compounds from the degradation. To better understand the physicochemical and biological performance of the selected formulation simultaneously co-encapsulating CUR and QU, a stability test of the compounds mixture was firstly carried out using X-ray powder diffraction and thermal analyses, such as differential scanning calorimetry (DSC) and thermogravimetric analyses (TGA). The determination and quantification of the encapsulated active compounds was then required being an essential tool for the development of innovative nanomedicines. Thus, a new HPLC–UV/Vis method for the simultaneous determination of CUR and QU in the nanoemulsions and their evaluation in stability studies in simulated biological fluids was developed and validated. The X-ray diffraction analyses demonstrated that no interaction between the mixture of active ingredients, if any, is strong enough to take place in the solid state. Moreover, the thermal analysis demonstrated that the CUR and QU are stable in the nanoemulsion production temperature range. The proposed analytical method for the simultaneous quantification of the two actives was selective and linear for both compounds in the range of 0.5 – 12.5 µg/mL (R2 &gt; 0.9997), precise (RSD below 3%), robust and accurate (recovery 100 ± 5 %). The method was validated in accordance with ICH Q2 R1 “Validation of Analytical Procedures” and CDER-FDA 2validation of chromatographic methods” guideline. Furthermore, the low detection (LOD &lt; 0.005 µg/mL for CUR and &lt;0.14 µg/mL for QU) and quantification limits (LOQ &lt; 0.017 µg/mL for CUR and &lt; 0.48 µg/mL for QU) of the method were suitable for the application to drug release and permeation studies planned for the development of the nanoemulsions. The method was then applied for the determination of nanoemulsions CUR and QU encapsulation efficiencies (&gt; 99%), as well as for the stability studies of the two compounds in simulated biological fluids over time. The proposed method represents, to our knowledge, the only method for the simultaneous quantification of CUR, and QU in nanoemulsions.

As the global semiconductor market is growing steadily, the need for optimization of the man-ufacturing process is paramount. For example, in the process of transferring the semiconductor chips to the substrate, each chip needs to be individually separated from the dicing tape via a plunger. In this process, however, the fixed chip is not easily removed from the tape due to strong adhesion, and the chip can become damaged if it is forcibly pulled out. Therefore, the present paper reports the development of functional adhesive tapes that initially adhere strongly to the semi-conductor chip on the dicing tape but can be easily removed after transfer. The fabricated functional tape has an initial adhesion of 350 gf, which enables firm fixing of the semiconductor chip to be transferred, and a final adhesion of 0.8 gf after UV curing, thereby facilitating removal of the chip without damage after transfer. Therefore, the fabricated functional tape is expected to contribute to the optimization of the semiconductor manufacturing process and is expected to be useful in the semiconductor package manufacturing process.

In this study we focus on measurements and modeled UV index at the region of Athens, Greece, during a period of a low ozone event. During the period of 12 to 19 of May 2020, total column ozone (TOC) showed extremely low values 35-55 DU (up to 15%) decrease from the climatic mean (being lower than the -2σ). This condition favors the increase of UV erythemal irradiance, since stratospheric ozone is the most important attenuator at the UVB spectral region. A parallel intrusion of Saharan Dust aerosols in the region has masked a large part of the low ozone effect on UV irradiance. In order to investigate the event, we have used spectral solar irradiance measurements from the Precision Solar Radiometer (PSR), total column ozone from the BREWER spectrophotometer and Radiative Transfer Model (RTM) calculations. Model calculations of UV Index (UVI) showed an increase of ~30% compared to the long term normal UVI due to the low TOC while at the same time and for particular days, aerosols masked this effect by ~20%. The study points out the importance of accurate measurements or forecasts of both ozone and aerosols when deriving UVI under unusual low ozone-high aerosol conditions.

The sunscreen industry is achieving remarkable worldwide prominence by responding to the growing need for skin protection with fast-paced innovation. Increased consumer awareness of the harmful effects of sunlight has fueled the demand for improved photo protection. The need for broad-spectrum protection from both UVA and UVB rays has inspired scientists worldwide to research new cosmetic formulations and delivery systems. More effective sunscreen actives, emollients and novel cosmetic and functional ingredients have been regularly added to the formulator’s repertoire. Creativity in innovation has been hindered only by regulatory agencies and patent restrictions worldwide. Familiarity with the current restrictive regulations and patent law infringements has become integral to any research effort attempting to provide improved protection to individuals affected by the sun’s damaging effects. The increasing incidence of skin cancers and photo damaging effects caused by ultraviolet radiation has increased the use of sun screening agents, which have shown beneficial effects in reducing the symptoms and reoccurrence of these problems. Unlike the situation in Europe where sunscreen ingredients are considered under cosmetics guidelines, the FDA is required to define sunscreens as drugs since they are advertised to prevent sunburn and, more recently, the risk of skin cancer. In the USA, the FDA has been regulating this industry since August 25, 1978, with the publication of the Advance Notice of Proposed Rulemaking. Sunscreens are considered drugs and cosmetics and therefore must be governed by the FDA-OTC monograph. With the variety of sunscreen agents used in cosmetic and UV protection products, Australia, Canada, and the European Union (EU) have also developed regulatory protocols on safe sunscreen product use. Unlike the USA though, Australia has approved 34 active sunscreen ingredients and the EU has approved 28 of these ingredients. Current FDA regulations allow labeling of sunscreen products to a maximum of 30þ, despite the many products currently available with numbers as high as 100. From a cosmetic formulation point of view, increasing the SPF number in a product is governed by simple chemical principles.

Controlling the concentration of free radicals in polymerization systems is advantageous for preparing cationic polyacrylamide (CPAM) with high molecular weight and acceptable dissolvability. In this study, a novel ultraviolet (UV)-initiated system characterized with adjustable light intensity and redox-azo complex initiator was used to synthesize a CPAM flocculant named NP. Comparatively, another CPAM flocculant named SP with stable UV light intensity and single initiator was prepared. The chemical structure, morphology, and thermal stability were analyzed through instrumental analysis. Proton nuclear magnetic resonance indicated that NP was successfully prepared, and a small fraction of cationic homopolymer was mixed in the product. Polymerization conditions were optimized, and polymerization mechanism was determined by investigating the effects of various parameters on intrinsic viscosity, conversion, and dissolvability. Results showed that the best performance was obtained at indexes of 0.45 wt‰ redox initiator concentration, 0.2 wt‰ azo initiator concentration, 40.0 wt% of cationic monomer, first- and second-stage light intensities of 8.5 and 13 mW/cm², respectively, and 3 wt% urea. Sludge conditioning performances of NP and SP were comparatively evaluated, and the mechanism was determined by investigating the sedimentation behavior and floc size distribution. High intrinsic viscosity, porous morphology structure, and the mixed cationic homopolymer of NP resulted in better sludge conditioning performance.

The analysis of chemical warfare agents and their degradation products has become increasingly important for public and military safety in recent years. In the article, we drew attention to the possibilities of the HPLC-ICP-MS analytical technique, which has been relatively rarely used for identifying and quantitatively analyzing highly toxic and hazardous substances in environmental samples. This technique enables the identification and determination of minimal amounts (ppt range) of chemicals in various matrices. This fact is important for the analysis of chemical warfare agents and other highly harmful compounds, even small amounts of which can have serious consequences for living organisms. The article critically analyzes the results of scientific research on the identification and quantitative analysis of extremely toxic organophosphorus, organosulfur and organoarsenic substances, their derivatives and degradation products using high-performance liquid chromatography (HPLC) coupled with inductively coupled plasma mass spectrometry (ICP-MS).

Solifenacin (SFC) is a potent muscarinic antagonist that effectively reduces bladder muscle contraction, thereby alleviating symptoms such as frequency of micturition and urgency. Oxidation of SFC leads to the formation of impurities like Impurity K. Effective analysis and control of this impurity is crucial for ensuring compliance with regulatory standards and safeguarding patient health. To address these challenges, we propose a novel one-step synthesis of Impurity K from SFC. Impurity K was synthesized using cerium ammonium (IV) nitrate (CAN) in water/acetonitrile as the solvent. Additionally, we describe a new HPLC-MS method for the detection of Impurity K in solifenacin succinate tablets.

: Nonylphenol is an endocrine disruptor who belongs to alkylphenol group of chemicals and its high occurrence in the environment can caused an adverse effect on human health. The breast milk can serve as a marker to take measure of human exposure to these chemicals through different routes of exposure. In this study, the influence of different factors (kind of wastes mothers drink, consummation of fish, pork and beef; using gloves, mail polish, using vitamins and medication) on the concentration on nonylphenol in breast milk were studied. Concentrations of nonylphenol in breast milk were determined by high-performance liquid chromatography (HPLC) with fluorescence detection. The lowest and highest concentration of nonylphenol in breast milk was 0.97 ng/mL and 4.37 ng/mL, respectively. The selected factors such as pork, beef, fish consumption, vitamin supplementation, medication, using of gloves, nail polish and gel nails were observed in relation to 4-nonylphenol concentration.

The length of accommodation has a statistically significant effect on nonylphenol, concentration in breast milk (p value was 0.02). Other mentioned factors had not a significant influence on nonylphenol values.

The aim of this study, therefore, provides information about Aflatoxin levels in raw cow’s milk in Injibara Town of Awi Administrative zone by using HPLC-FLD. A good linearity of standard calibration was found for AFM1 at a range of 0.5–7 µg/L. Regression coefficient (R2) values were 0.9999, whereas slope and intercept were 2.5278 and 0.1012, respectively. The average recoveries for the spiked samples were range from76.62– 90.98 % and the RSD values ranged between 2.55–7.36 %. The results showed that 15 % of samples (3 out of 20) were contaminated with AFM1 in the range of 0.046–0.22 µg/L. The average contamination level was 0.121 µg/L. The determined mean values of AFM1 in the collected milk samples were above the standard limit of the European Commission and lower than the level established by United States regulations. Further monitoring of Aflatoxin in milk samples from different regions of the country is justified to conclusively determine the actual safe/risks and possibly low Aflatoxins-risk milk production areas.

The pharmacokinetics of salicylic acid (SA) in sheep was evaluated following intravenous (IV) and oral administration of sodium salicylate (sodium salt of salicylic acid) at different doses. Six healthy sheep were administered sodium salicylate (SS) IV at doses of 10, 50, 100 and 200 mg/kg body weight and another six sheep were drenched with 100 and 200 mg/kg of SS orally. Both studies were randomised crossover trials. A one-week washout period between each treatment was allowed in both studies. Blood samples were collected at 0, 15, 30 minutes and 1, 2, 4 and 6 hours after IV and oral SS administrations. Plasma SA concentrations were determined using high performance liquid chromatography with diode array detection method. Pharmacokinetic variables were calculated in a non-compartmental model. The elimination half-life (T_{1/2 el}) of SA after IV administration of 200 mg/kg SS was 1.16 ± 0.32 hours. Mean bioavailability of SA was 64%, and mean T_{1/2 el} was 1.90 ± 0.35 hours, after 200 mg/kg of oral SS. The minimum plasma SA concentration (16.8 µg/mL) required to produce analgesia in humans was achieved after IV administration of 100 and 200 mg/kg SS in sheep for about 0.17 hour in this study. Experiments on pharmacokinetic-pharmacodynamics modelling are required to determine the actual effective plasma concentration range of SA in sheep.

Guayule (Parthenium argentatum Gray) is a shrub native of the arid regions of Mexico. In the last decades, significant attention for its cultivation has risen because it is the raw material for the production of hypoallergenic natural rubber. Guayule biomass contains also high amounts of resin, which is not normally exploited in any way. Among other sesquiterpenic esters, guayulins (i.e. the parteniol esters of cinnamic acid, guayulin A, or of anisic acid, guayulin B) are contained in resin. In addition, minor amounts of guayulin C and guayulin D are formed by degradation/oxidation of guayulins A and B, respectively. Guayulins likely act as cinnamate and p-anisate reservoirs for Guayule shrub, in addition, it has been postulated that they might have a key role in the chemical defense system of Guayule. Furthermore, it seems reasonable that guayulins may possess significant biological properties (e.g. antibacterial and anticancer activities), in close analogy with those shown by sesquiterpene lactones contained in many other species of Parthenum genus. As a matter of fact, guayulins A and B play an important role in the synthesis of antineoplastics used in breast cancer treatment. In this contribution we propose an original and validated RP-HPLC approach to the simultaneous quantification of guayulins A, B, C and D. The procedure of resin extraction from Guayule biomass has been optimized in terms of both extraction method and solvent. RP-HPLC separation has been accomplished by an Ascentis^® C18 column under isocratic elution with a 80:20 (v:v) acetonitrile:water mixture. Validation was carried out in terms of limits of detection and quantification, linearity, precision, and trueness. Finally, the method was tested with a number of fresh and seasoned samples of spontaneous Guayule shrub from Mexico.

Glutamine is the main amino acid which is substrate for gluconeogenesis in postoperative period. It is suggested that this leads to a substantial and a long-term decline in glutamine concentration. Glutamine is a source of energy for the synthesis of nucleotides, lymphocytes and cells of gastrointestinal tract. In this study, 79 patients were qualified to a coronary artery bypass surgery (Group I) or a surgery in the large intestine area (Group II). The objectives of this study were: evaluation of the impact of surgical procedures on the serum concentration of glutamine of the operated patients, assessment of gender, weight and BMI impact on glutamine concentration and analysis of the correlation between glutaminę serum concentration and laboratory parameters. The mean concentration of glutamine before surgery, the 3rd and 5th day after surgery was higher in Group I. CRP level in the 3rd and 5th postoperative day was higher in Group II. There were no significant differences betweengroups when it comes to BMI and the concentration of CRP (p <0.05). Glutamine concentration depends on the severity of inflammation, the operated body cavity and the intensified catabolism which results from different pathophysiology of digestive system diseases and coronary arterial disease.

The antioxidant properties and polyphenol content of some selected aromatic plants grown in Greece were studied. Plants were refluxed with 60% methanol after acid hydrolysis. The phenolic substances were quantified by HPLC-DAD. The antioxidant capacity of the extracts was determined with the Rancimat test using sunflower oil as substrate. Free radical scavenging activity was measured using the stable free radical 1, 1-diphenyl-2-picrylhydrazyl (DPPH). Results were compared with standard BHT and ascorbic acid. Total phenol concentration of the extracts was estimated with Folin-Ciocalteu reagent using gallic acid as standard.

A specific HPLC method has been developed and validated for the determination of vanillyl butyl ether in cosmetic products. The extraction procedure with an isopropanol water 1:1 mixture is described. The method uses a RP-C-18 column with isocratic elution and a UV detector. The mobile phase consists of a mixture of acetonitrile and buffer (Na2HPO4 20mM in water) (30:70 v/v) with a variable flow rate. The method was validated with respect to accuracy, precision (repeatability and reproducibility), specificity and linearity. The procedure described here is simple, selective and reliable for routine quality control analysis and stability tests of commercially available cosmetic products.

The main purpose of this study was to create a prototype of an unmanned aerial system equipped with intelligent hardware and software technologies necessary for monitoring the health and growth of crops in orchards. Another important objective was to use low-cost sensors that accurately measure ultraviolet solar radiation. The device, which needs to be attached to the commercial DJI Mini 4 Pro drone, should be small in size, portable, and have very low energy consumption. For this purpose, the widely used Vishay VEML6075 digital optical sensor was selected and implemented in a prototype, alongside a Raspberry Pi Zero 2W minicomputer. To collect data from these sensors, a program written in Python was used, containing specific blocks for data acquisition from each sensor, to facilitate the monitoring of ultraviolet (UV) radiation, or battery current. By analyzing the data obtained from the sensors, several important conclusions are drawn that may provide valuable pathways for the further development of mobile or modular equipment. Furthermore, the results of the plant condition analysis with proposed models in the Geographic Information System (GIS) environment were also presented. The visualization of maps indicating variations in vegetation condition led to the identification of problem areas like hydric stress.

The nature of the blue color in the iodine-starch reaction (or, in most cases, iodine-iodide-starch reaction, i.e. as I2 as well as I- are typically present) has for decades elicited debate. The intensity of the color suggests a clear charge-transfer nature of the band at ~600 nm, and there is consensus regarding the fact that the hydrophobic interior of the amylose helix is the location where iodine binds. Three types of possible sources of charge transfer have been proposed: (1) chains of neutral I2 molecules, (2) chains of poly-iodine anions (complicated by the complex speciation of the I2 - I- mixture), or (3) mixtures of I2 molecules and iodide or polyiodide anions. An extended literature review of the topic is provided here. According to the most recent data, the best candidate for the “blue complex” is a I2-I5-I2 unit – expected to occur in repetitive manner inside the amylose helix.

Skin aging is one of the most common problems facing humanity. It occurs because of altering the balance between free radicals and antioxidants and increasing the amount of the reactive oxygen species (ROS) in skin cells, which leads to oxidative stress especially when exposed to UV radiation. Antioxidants can neutralize the harmful effects of ROS, and secondary plant metabolites can help protect against UV radiation. In this study, punicalagin was extracted from pomegranate and concentrations of total polyphenolics and flavonoids were determined and antioxidant activities measured. Punicalagin was loaded onto niosomes and its morphology and release were studied. An in vitro study was performed on human fibroblast cell line HFB4 cells with aging induced by H2O2 and UV radiation. Cell cycle arrest was studied and different genes (MMP3, Col1A1, Timp3, and TERT) involved in the skin aging process were selected to measure punicalagin's effect. Punicalagin succeeded in reducing the growth arrest of HFB4 cells, activated production of the Col1A1 and Timp3 genes, maintained collagen level, and lowered of MMP3. Punicalagin increased human TERT concentration in skin cells. Punicalagin is promising as a natural antioxidant to protect human skin from aging.

ATLID (ATmospheric LIDar) is the atmospheric backscatter LIDAR (Light Detection and Ranging) on board of the EarthCARE (Earth Cloud, Aerosol and Radiation Explorer) mission, the sixth Earth Explorer Mission of the ESA (European Space Agency) Living Planet Programme [1-5]. ATLID’s purpose is to provide vertical profiles of optically thin cloud and aerosol layers, as well as the altitude of cloud boundaries [6-10]. In order to achieve this objective ATLID emits short duration laser pulses in the UV, at a repetition rate of 51 Hz, while pointing in a near nadir direction along track of the satellite trajectory. The atmospheric backscatter signal is then collected by its 620 mm aperture telescope, filtered through the optics of the instrument focal plane assembly, in order to separate and measure the atmospheric Mie and Rayleigh scattering signals. With the completion of the full instrument assembly in 2019, ATLID has been subjected to an ambient performance test campaign, followed by a successful environmental qualification test campaign, including performance calibration and characterization in thermal vacuum conditions. In this paper the design and operational principle of ATLID is recalled and the major performance test results are presented, addressing the main key receiver and emitter characteristics. Finally, the estimated instrument, in-orbit, flight predictions are presented; these indicate compliance of the ALTID instrument performance against its specification and that it will meet its mission science objectives for the EarthCARE mission, to be launched in 2023.

Adenoviruses (Ads) have been extensively manipulated for the development of cancer selective replication, leading to cancer cell death or oncolysis. Clinical studies using E1-modified oncolytic Ads have shown that this therapeutic platform was safe, but with limited efficacy, indicating the necessity of targeting other viral genes for manipulation. To improve the therapeutic efficacy of oncolytic Ads, we treated the entire Ad genome repeatedly with UV-light and have isolated AdUV which efficiently lyses cancer cells as reported previously [1]. In this report, we show that no mutations were observed in the early genes (E1 or E4) of AdUV while several mutations were observed within the Ad late genes which have structural or viral DNA packaging functions. This study also reported the increased release of AdUV from cancer cells. In this study, we found that AdUV inhibits tumor growth following intratumoral injection. These results indicate the potentially significant role of the viral late genes, in particular the DNA packaging genes, to enhance Ad oncolysis.

Parthenium incanum (Mariola) is used in Mexico to treat stomach and liver disorders. Reports have shown that the genus Parthenium contains biologically active metabolites. This work aimed to isolate and identify biologically active compounds with trypanocidal activity. The extracts were obtained through static maceration and extraction with successive reflux using different solvents. After that, they were fractionated with column chromatography, and the obtained fractions were compared by TLC; similar fractions were pooled and concentrated. A bioguided assay was performed with S. aureus ATCC 25923, using agar diffusion techniques and bioautography to determine preliminary biological activity. Active fractions were obtained and purified from a preparative TLC plate. The obtained compounds were identified as parthenin and coronopolin by TLC, FT-IR, and HPLC-MS. Finally, they were evaluated against the Ninoa strain of Trypanosoma cruzi, obtaining an IC50 of up to 45±2.5 ppm. It was concluded that P. incanum contains compounds with trypanocidal activity.

The practice of voluntary depigmentation is still prevalent in Africa, with a wide range of lightening cosmetics used. Our objective was to research and quantify three regulated and/or prohibited depigmenting ingredients present in lightening cosmetics sold in Ouagadougou. Twenty-nine lightening cosmetic samples were collected from vendors and HPLC analysis was subsequently conducted to identify and measure the concentrations of hydroquinone, clobetasol propionate, and kojic acid. The presence of hydroquinone was indicated on the label of 13.79% of the products, while 51.72% contained it after analysis. Furthermore, none of the products mentioned a concentration of hydroquinone exceeding 2.00%, even though 27.58% of them contained high concentrations. For clobetasol propionate, its presence was stated on the labels of 13.79% of the products, while 31.03% contained it. One sample had a clobetasol content exceeding 0.05%, although none mentioned a concentration higher than this value. Finally, while 24.13% of the samples claimed to contain kojic acid, only 17.24% did. We also observed that 41.38% of the samples contained combinations of two depigmenting ingredients investigated, with a predominance of the hydroquinone + clobetasol propionate (27.38%). These results demonstrate that manufacturers' declarations regarding the compositions of active ingredients in lightening cosmetics can sometimes be deceptive.

Rivaroxaban and aspirin are commonly used antithrombotic agents that are used in combination for the prevention of coronary artery disease (CAD) and atherothrombotic events in adult patients after an acute coronary syndrome (ACS) with elevated cardiac biomarkers, or with coronary artery disease (CAD) or symptomatic peripheral artery disease (PAD) in high-risk patients. The recommended dosage is 2.5 mg Rivaroxaban twice daily with 75–100 mg aspirin daily. This study aimed to develop a fixed-dose combination tablet of rivaroxaban (2.5 mg) and aspirin (50 mg) to decrease the patient's pill burden and enhance medication adherence. The product formula was developed based on compatibility studies among the active ingredients and the active ingredients' compatibility studies. The formula and the manufacturing procedure were chosen based on the risk assessment for each active substance, wet granulation with both actives intragranular was found to have faster dissolution than direct mix formulae. Furthermore, a validated reverse-phase HPLC stability indicator method was developed to detect APIs and their possible degradants in the formula.

Abstract: The aim of this study was to identify and quantify polyphenolic compounds in skin extracts from four Bulgarian grape varieties and compare to those of seed extracts. The values of total phenolic contents (TPC), flavonoids, anthocyanins, procyanidins, ascorbic acid in grape skin extracts were determined. The antioxidant capacities of skin extracts were evaluated by DPPH, ABTS, FRAP and CUPRAC methods. The TPC values of skin extracts were 2-3 times lower than those of seed extracts. The significant difference between total parameters values of individual grape varieties were also found. According to the TPC and antioxidant capacity of skin extracts the different grape varieties were in the following order: Marselan ≥ Pinot Noir ˃ Cabernet Sauvignon ˃ Tamyanka. The individual compounds in the grape skin extracts were determined by RP-HPLC and compared with those of seed extracts. The determined composition of skin extracts was significant different from seed extracts composition. Qualitative and quantitative evaluation of the procyanidins and catechins in the skins were carried out. A correlation between phenolic contents, individual composition and antioxidant capacity of different extracts was found. The studied grape extracts have a potential to be applied as natural antioxidants in the pharmaceutical and food industries.

This study investigated the phenolic compounds of 15 Chrysanthemum morifolium Ramat cv. ‘Hangbaiju’, including 6 ‘Duoju’ and 9 ‘Taiju’ using high performance liquid chromatography. The antioxidant activities of these ‘Hangbaiju’ were estimated by DPPH, ABTS and FPAR assays. Results showed that a total of 14 phenolic compounds were detected in these flowers, including 3 mono-caffeoylquinic acids, 3 di-caffeoylquinic acids, 1 phenolic acid and 7 flavonoids. ‘Duoju’ and ‘Taiju’ possessed different concentration of phenolic compounds, and ‘Taiju’ exhibited higher caffeoylquinic acids and stronger antioxidant activities than ‘Duoju’. Caffeoylquinic acids showed a strong correlation with the antioxidant activities of the samples. Principal component analysis revealed an obvious separation between ‘Duoju’ and ‘Taiju’ using phenolic compounds as variables. Apigenin-7-O-glucoside, 3,5-di-O-caffeoylquinic acid, luteolin and acacetin were found to be the key phenolic compounds to differentiate ‘Duoju’ from ‘Taiju’.

This contribution is aimed to measure for the first time the amount of biogenic amines (BAs) in one of the most ancient and traditional sheep cheese produced in Sardinia, Italy: the Protected Designation of Origin (PDO) Fiore Sardo. For doing this, an original RP-HPLC-DAD-UV method has been developed, completely validated in terms of LoD, LoQ, linearity, precision and trueness, and tested on 36 real Fiore Sardo PDO cheese samples produced by four different cheesemakers and marketed by four stores. The average total concentration of the eight BAs (i.e. tyramine, tryptamine, histidine, putrescine, cadaverine, 2-phenylethylamine, spermine and spermidine) measured in Fiore Sardo cheese was 700 mg/kg, with a range between 170 mg/kg and 1100 mg/kg. A great variability in the total amount of BAs has been evidenced among the Fiore Sardo marketed in the four stores as well as for the cheeses purchased in different times in the same store. Tyramine (350 mg/kg), putrescine (150 mg/kg), histamine (80 mg/kg) and cadaverine (30 mg/kg) are the most abundant BAs found in this matrix. Among many factors concurring, the dominant microflora of Fiore Sardo PDO is likely the principal cause of the qualitative and quantitative distribution of BAs in this matrix. Finally, the total amount of BAs found in Fiore Sardo PDO is not able to cause any situation of health alert for consumers.

Black yeasts are a highly specified group of fungi, which are characterized by a high resistance against stress factors. There are several factors enabling the cells to survive harsh environmental conditions. One aspect is the pigmentation, besides the melanin black yeasts often display a highly diverse carotenoid spectrum. Determination and characterization of carotenoids depend on an efficient extraction and separation, therefore especially for black yeast, characterized by thick cell walls specific protocols are needed to ensure analyses regarding stress responses in these fungi. Here we present both, a method to extract and analyze carotenoids and the unusual carotenoid composition of the black yeast Knufia petriola A95. Mechanical treatment combined with an acetonitrile extraction gave us very good extraction rates with a high reproducibility. The presented extraction and elution protocol allows the separation of the main carotenoids (7) in K. petricola A95 and should be suitable for the detection of additional carotenoids in other species. K. petricola A95 displays an unusual carotenoid composition, with mainly didehydrolycopene, torulene and lycopene. The pigment composition varied in dependency to oxidative stress but remained relatively constant if the cells were cultivated under low temperature. Black yeasts are a promising source for carotenoid production and other substances. To unravel the potential of these fungi new methods and studies are needed. The established protocol allows the determination carotenoid composition in black yeasts. Oxidative stress results in an adaptation in pigment composition in K. petricola A95. Future experiments have to be carried out to determine if didehydrolycopene functions as a protective agent itself or if it serves as a precursor for antioxidative pigments like torulene and torularhodin, which could be produced after induction under stress conditions.

Background: The cumulative exposure of UV radiation can result in dangerous consequences such as skin burns, photoaging, and skin cancer, hence the importance of using photoprotective formulations. Currently, the formulations seek to be more efficient, innovative and reach a wider audience. Many sunscreens are vegan, and have antioxidant substances to ensure additional photochemoprotective action. To evaluate biological, functional, and mechanical characteristics of them on the skin, biometric and rheologic methods can be used. Objective: Evaluate biophysical, rheological and sensorial parameters of Face Care Facial Moisturizing Cream® (P1) and a vegan formulation (P2) by in vitro and in vivo tests. Methods: A UV scan experiment was performed covering the range of 250 to 400 nm. Sun Protection Factor (SPF) was evaluated by Mansur method. Rheological profiles were obtained using a MARS II (Haake®) controlled shear stress and gradient rheometer. Biophysical parameters were analyzed: skin sebum content, hydration level, transepidermal water loss, erythema and melanin level, skin color, and skin pH. The acceptance profile of the formulations by the volunteers was determined using a 9-point hedonic scale and a 5-point purchase intention test. Results: The formulations effectively absorbed light covering the UV radiation spectrum from UVA to UVB. The SPF of P1 was 25.21 and of P2 was 12.10 by in vitro tests. They had also pseudoplastic and thixotropic behavior, it can contribute to better spreadability and to form a protective film. Biometric tests showed an increase in hydration and skin sebum, decreased erythema, and maintenance of skin pH after application of both formulations. The products also had similarly good acceptance and purchase intention by the volunteers. Conclusion: The comparation of a commercialized product and a vegan test version showed very similar rheological and great acceptance profiles. Therefore, the vegan formulation is a good alternative to reach a different market.

Rutin, a flavonoid compound found in various plants, has gained attention for its potential applications in the cosmetic industry due to its antioxidant, anti-inflammatory, and potential photoprotective properties. Our review explored the use of rutin (rutoside, 3-O-rutinoside-quercetin) in cosmetics/dermocosmetics, focusing on its physicochemical properties and stability, cutaneous permeability, and efficacy in sunscreen systems, involving in vitro tests and the current state of clinical trials. Rutin's ability to scavenge free radicals, prevent peroxidation, and reduce vascular fragility makes this flavonoid a promising ingredient for photoprotection. Studies have shown that rutin can enhance the sun protection factor (SPF) of sunscreen formulations, especially when combined with organic UV filters. The encapsulation of rutin in nanostructures has also been investigated to improve its applicability. Overall, rutin shows potential as a safe and effective ingredient in cosmetic/dermocosmetic formulations, offering protection against the harmful effects of UV radiation and oxidative stress on the skin.

The photodegradation of azithromycin present was carried out in water using H2O2 under UV irradiation. The reaction variables considered in this study were the amount of H2O2 solution and the initial concentration of azithromycin to evaluate the performance of the photodegradation process. The azithromycin degradation was not observed in the dark during stirring for 20 minutes. The study showed efficient photodegradation of azithromycin using H2O2 as an oxidant in the presence of UV irradiation. The azithromycin degradation was affected significantly by the pH of the irradiated solution. The degradation was low at an acidic pH and showed an increasing trend as the pH changed to basic. The azithromycin degradation was increased with a higher amount (higher concentration) of H2O2. The degradation of azithromycin decreased with a higher concentration of azithromycin in the reacting solution. The highest degradation of AZT was achieved in 1 hour using a 1.0 ppm AZT solution containing 3 ml of H2O2. The experimental data obtained was well-fitted to zero-order reaction kinetics. The results of this study were found to be superior when compared with those reported in the literature, with photocatalysis using nanomaterials and photolysis using irradiation and H2O2. The UV/H2O2 system was found to be quite efficient for the photodegradation of azithromycin, and this system can be applied to degrade other organic pollutants present in industrial wastewater. To this point, such a study has not been reported specifically for AZT photodegradation.

In this study, we present comprehensive climatologies of effective ultraviolet (UV) quantities and photosynthetically active radiation (PAR) over Cyprus for the period 2004 to 2023, leveraging the synergy of earth observation (EO) data and radiative transfer model simulations. The EO dataset, encompassing satellite and re-analysis data for aerosols, total ozone column, and water vapor, alongside cloud modification factors, to capture the nuanced dynamics of Cyprus's atmospheric conditions. With a temporal resolution of 15 minutes and a spatial of 0.05°x0.05°, these climatologies undergo rigorous validation against established satellite datasets and are further evaluated through comparisons with ground-based global horizontal irradiance measurements provided by the Meteorological Office of Cyprus. This dual-method validation approach not only underscores the models' accuracy but also highlights their proficiency in capturing intra-daily cloud coverage variations. Our analysis extends to investigating the long-term trends of these solar radiation quantities, examining their interplay with changes in cloud attenuation, aerosol optical depth (AOD), and total ozone column (TOC). Significant decreasing trends in the noon ultraviolet index (UVI), of -2 to -4% per decade have been found in Autumn, especially marked on the island’s Northeastern part, mainly originating from the (significant) positive trends in TOC. The significant decreasing trends in TOC, of -2 to -3% per decade that were found in Spring do not result in correspondingly significant positive trends in the noon UVI since variations in cloudiness and aerosols also have a strong impact on the UVI in this season. The seasonal trends in the Daylight Integral (DLI) were generally not significant. These insights provide a valuable foundation for further studies aimed at developing public health strategies and enhancing agricultural productivity, highlighting the critical importance of accurate and high-resolution climatological data.

The spectrophotometric methods of determination of the active pharmaceutical ingredients meloxicam and nimesulide were reviewed, and a simple UV-spectrophotometric method for the determination of these active pharmaceutical ingredients in industrial equipment cleaning validation samples were proposed. The methods are based on extraction of the residual quantities of meloxicam and numesulide from the manufacturing equipment surface by the concentrated sodium carbonate solution, and the subsequent UV-spectrophotometric determination of the basic forms of the drugs at the wavelength of 362 nm for meloxicam and at 397 nm for nimesulide. The calibration graphs are linear in the range from 5 to 25 mg/L of both nimesulide and meloxicam, the molar attenuation coefficients are 6100 m2/mol for nimesulide and 9100 m2/mol for meloxicam, the limit of detection is 0.8 mg/L for nimesulide and 1.9 mg/L for meloxicam, the limit of quantification is 2.5 mg/L for nimesulide and 5.8 mg/L for meloxicam, the methods are selective with respect to the common excipients, show a good accuracy (the relative uncertainty does not exceed 4%) and precision (the relative standard deviation does not exceed 5%), do not require lengthy sample preparation and sophisticated laboratory equipment and are suitable for the routine analysis of cleaning validation samples.

Evidence has shown that the state of the patient care environment has a direct impact in heightening the risks of hospital acquired infections among patients admitted in hospitals. And in view of the sub-optimal standard of cleanings by housekeeping staff, the quest for a better approach to reliably disinfect environmental surfaces in healthcare facilities. The ultraviolet light has been known for its antimicrobial property, and have been used in water treatment, food processing and in-duct cleaning of ventilations. A recent introduction of its use for surface decontamination has raised interest among hospitals.However, studies have shown that, in spite of its relative success in other applications, there is doubt in its efficacy in decontaminating shadowed areas of the room, and therefore, would not be seen as justifying its capital intensiveness.

Ultraviolet (UV) light with a wavelength of 254 nm has proven to be effective at inactivating microorganisms, and thus has been increasingly employed as a method of disinfection for indoor environments. Solar UV wavelengths (300 to 400 nm) are known to initiate the formation of secondary organic aerosol (SOA) particles from photo-oxidation of volatile organic compounds in the atmosphere, but germicidal wavelengths have not been extensively studied for indoor environments. In this work, toluene was exposed to 254 nm UV light in a laboratory photoreactor, with varying conditions of the air, the duration of UV exposure, and the duration of post-UV time. The number of particles formed in the fine particulate matter (PM2.5) size range was measured, and significant levels of particle formation were observed for UV exposure periods of as short as 5 minutes. The particle formation ranged from 2.4x10⁶ particles/m³ for 5 minutes of UV exposure, to 1449.8x10⁶ particles/m³ for 15 minutes of UV exposure. Particle formation was found to increase with increasing concentrations of gas phase toluene, and at relative humidity of approximately 20% and higher. Variations in the initial number of particles present did not appear to have a significant effect on the particle formation, suggesting that nucleation was not a controlling factor. However, tests in a commercial environment showed no significant detectable PM2.5 formation, indicating that SOA formation during the intermittent use of germicidal UV may not significantly affect indoor air quality.

Compound-specific isotope analysis (CSIA) can provide unique insights into the cycling of elements including carbon and nitrogen. One approach for CSIA is the use of high-performance liquid chromatography (HPLC) to separate compounds of interest, followed by analysis of these compounds using an elemental analyzer coupled to an isotope ratio mass spectrometer. A key component of this technique is the fraction collector, which automatically collects compounds as they are separated by HPLC. Here we present a fraction collector which is a simple adaptation of a 3D printer, and thus can be easily adopted by any laboratory already equipped for HPLC. In addition to the much lower cost compared to commercial alternatives, this adaptation has the advantage for CSIA that the 3D-printer is able to heat the collected fractions, which is not true for many commercial fraction collectors. Heating allows faster evaporation of the solvent, so that the dried compounds can be measured by EA-IRMS immediately. The procedure can be repeated consecutively so that dilute solutions can have the compounds concentrated for analysis. Any computer-controlled HPLC can be integrated to the fraction collector used here by means of AutoIt.

Turmeric, Curcuma longa L., is a type of medicinal plant characterized by its perennial nature and rhizomatous growth. It is a member of the Zingiberaceae family and is distributed across the world’s tropical and subtropical climates, especially in South Asia. Its rhizomes are highly valued for food supplements, spices, flavoring agents, and yellow dye in South Asia since ancient times. It exhibits a diverse array of therapeutic qualities that encompass its ability to combat diabetes, reduce inflammation, act as an antioxidant, exhibit anticancer properties, and promote anti-aging effects. In this study, organic extracts of C. longa rhizomes were subjected to HPLC separation followed by mass spectrometry analysis. The Global Natural Product Social Molecular Networking (GNPS) approach was utilized for the first time in this ethnobotanically important species to conduct an in-depth analysis of its metabolomes based on their fragments. A total of 30 metabolites including 16 diarylheptanoids, 1 diarylpentanoid, 3 bisabolocurcumin ethers, 4 sesquiterpenoids, 4 cinnamic acid derivatives, and 2 fatty acid derivatives were identified. Among 16 diarylheptanoids identified in this study, five of them are reported for the first time in this species.

Wild Chinese prickly ash with elevated antioxidants is a valuable genetic resource for Zanthoxylum bungeanum Maxim improvement. There are rich wild germplasm resources in the Qinling Mountains. In a study with wild germplasm resources from different altitudes and six cultivated varieties, the phenolic and flavonoid compounds were analyzed by high performance liquid chromatography (HPLC). The chromatograms of them were basically the same, although their chemical composition content was greatly different. The thirty samples were divided into three categories through the hierarchical clustering analysis. And catechin, hyperoside and quercitrin were considered to be key compound for the quality evaluation, by contrast, the wild samples with an altitude of 2300±50 m (Ⅳ group) had the highest content of key compounds, and showed stronger antioxidant activity and antibacterial ability, indicating that these wild samples could be used as an excellent breeding resource. This is the first time to evaluate the quality of wild Chinese prickly ash in different altitude areas of Qinling Mountains. These excellent wild germplasm resources provided substantial potential accessions for use directly in Chinese prickly ash breeding programs.

Guava is a vital fruit worldwide, especially in Pakistan, and due to its nutritional value famous in each age group. Due to a very short shelf life, the marketing and export of this fruit faced severe constraints. Therefore, in the current study, edible coating of chitosan (0, 0.5, 1.0, 1.5, and 2.0%) was evaluated on postharvest shelf life when guava fruits were stored (room temperature and 4 °C temperatures) for 12 days. The chitosan treated coating fruits have shown reduced total sugars and malondialdehyde levels compared to untreated control samples. However, a significant difference (p ≤ 0.05) in total sugar and malondialdehyde levels exists between samples stored in m compared to refrigerated temperature (4 °C). The chitosan-coated samples have shown a greater amount of vitamin C, quercetin, rutin, and total phenolic contents than control samples. However, these nutritional parameters' levels were significantly different (p ≤ 0.05) in samples stored at room than samples stored at refrigerated temperature. However, the levels of crude fiber, potassium, and sodium were found statistically nonsignificant (p ≥0.05) in control versus chitosan treated coating treatments. The findings have documented that the coatings of 1.5 and 2.0% were most effective for extension in shelf life and maintaining the nutritional attributes of guava fruit.

Conventional fluorescent lamps used in tissue culture are costly light sources showing excessive wavelength emission-bandwidth that must be replaced by alternative, less costly and much lower power-consuming energy sources. The use of Light-Emitting Diodes (LEDs) is the best option due to their potential role as elicitors of secondary metabolite production in many plant models. Gynura procumbens(G. procumbens) is widely used for treating various diseases. Here, leaf explants were cultivated in MS medium supplemented with 0.5 mg/L of NAA and 2.0 mg/L of BAP for 30 days under white, blue, and red LEDs. Secondary metabolites were analyzed by HPLC and LC-MS. Blue LEDs elicited the highest antioxidant activity, total flavonoid, and phenolic content. Furthermore, the content of cyanidin-monoglucosides increased significantly increased under blue light.

Gold clusters protected by 3-MBA ligands (MBA = mercaptobenzoic acid, -SPhCO2H) have attracted recent interest for their unusual structures and advantageous ligand-exchange and bioconjugation properties. Azubel et al. first determined the core structure of an Au68-complex, which was estimated to have 32 ligands (3-MBA groups). To explain the exceptional structure-composition and reaction properties of this complex, and its larger homologs, Tero et al. proposed a “dynamic stabilization” via carboxyl O-H--Au interactions. Herein, we report the first results of an integrated LC/MS analysis of unfractionated samples of gold / 3-MBA clusters, spanning the narrow size range 13.4 to 18.1 kDa. Using high-throughput procedures adapted from bio-macromolecule analyses, we show that integrated capillary HPLC-ESI-MS, based upon aqueous-methanol mobile phases and ion-pairing reverse-phase chromatography, can separate several major components from the nanoclusters mixture that may be difficult to resolve by standard native gel electrophoresis due to their similar size and charge. For each component, one obtains a well-resolved mass spectrum, nearly free of adducts or signs of fragmentation. A consistent set of molecular mass determinations is calculated from detected charge-states tunable from 3- (or lower), to 2+ (or higher). One thus arrives at a series of new compositions (n, p) specific to the Au/3-MBA system. The smallest major component is assigned to the previously unknown (48, 26); the largest one is evidently (67, 30), vs. the anticipated (68, 32). Various explanations for this discrepancy are considered. A prospective is given for the several members of this novel series, along with a summary of the advantages and present limitations of the micro-scale integrated LC/MS approach to characterize such metallic-core macro-molecules, and their derivatives.

Tamarix nilotica (Ehrenb.) Bunge (Tamaricaceae), an indigenous plant to the Middle East region, is well-known as a medicinal plant for treating many human ailments. The current study aimed at exploring the polyphenols profile of T.nilotica alcohol soluble fraction of aqueous extract, assessing its in-vivo antifibrotic activity and the possible underlying mechanism, to unravel the impact of quantitative difference of sulphated polyphenols content on the antifibrotic activity of T.nilotca grown in two different habitats. Polyphenols profiling of T. nilotica extracts was performed using HPLC-HRESI-QTOF-MS-MS. The major polyphenol components included sulphated flavonoids, phenolic acids and free aglycones. The antifibrotic activity was evaluated through carbon tetrachloride-induced liver fibrosis in rats. Biochemical evaluations revealed that both fractions ameliorated the increased levels of hepatic aminotransferases, lipid peroxidation, hydroxyproline, α-smooth muscle actin (α-SMA), tumor necrosis factor-α (TNF-α), cyclooxygenase-2 (COX-2) and nuclear factor kappa B (NF-κB). Moreover, both fractions reduced catalase activity (CAT) and enhanced hepatic glutathione (GSH) content. Histopathological imaging undoubtedly confirmed such results. In conclusion, T. nilotica polyphenols rich fraction exhibited potential antifibrotic activity in rats. Significant alterations in GSH levels were recorded based on sulphated polyphenol metabolites content.

Solanum nigrum fruits have been conventionally available as a material of beverage due to its nutritional substances such as minerals, vitamins, amino acids, proteins, sugars, polyphenols and anthocyanins. Here has rarely reported on the characterizaton of the components and the regulatory mechanism of Anthocyanins in S. nigrum. In this study, we determined that the peel and flesh of S. nigrum fruits shared the similar HPLC profiles, but the different contents and total antioxidant activities for Anthocyanins. After an efficient purification method mainly including extraction with pH 1.0 distilled water and then desorption with pH 1.0 95% ethanol after a DM-130 resin adsorption step to obtain more pure anthocyanins extracts, the purity of anthocyanins extract from S. nigrum fruits reached to 56.1%. Moreover, eight anthocyanins from S. nigrum fruit were identified with HPLC-MS/MS for the first time. A typical R2R3-MYB transcription factor gene, SnMYB, was also cloned for the first time by RACE-PCR from S. nigrum. Moreover, the contents of anthocyanins was shown a good correlation (r = 0.93) with the expression levels of SnMYB during the fruits developmental stages. Most significantly, SnMYB successfully produced high anthocyanins contents (1.03 mg/g) when SnMYB was transiently expressed in tobacco leaves. Taken together, S. nigrum fruits are a promising resource for anthocyanins extraction and SnMYB is an activator that positively regulates anthocyanins biosynthesis in S. nigrum.

The efficacy and safety of organic UV-filters could benefit from their incorporation into lipid nanoparticles because of the ability of such carriers to reflect solar radiations, thus acting synergically as physical sunscreens. In this work, bemotrizinol (BMTZ), a broad-spectrum UV-filter, was loaded into nanostructured lipid carriers (NLC) whose lipid matrix contained different oils to assess the effects of the lipid core composition on the properties of the resulting NLC. Subsequently, the effects of incorporating different concentrations of optimized BMTZ-loaded NLC on the technological properties of O/W emulsions (pH, viscosity, spreadability, occlusion factor, in vitro BMTZ release and skin permeation, in vitro sun protection factor) were assessed. Optimized BMTZ-loaded NLC showed mean size=190.6±9.8 nm, PDI=0.153±0.013, ζ-potential=-10.6±1.7 mV, loading capacity=8% w/w. The incorporation of increasing concentrations of optimized BMTZ-loaded into emulsions provided slight increase of spreadability, lower viscosity and no change of pH, occlusion factor and BMTZ release compared to the control. No BMTZ skin permeation was observed from all formulations. About 20% increase of sun protection factor values was obtained for vehicles containing BMTZ-loaded NLC. Therefore, BMTZ-loaded NLC incorporation into emulsions could be a promising strategy to develop safer and more effective sunscreen formulations.

The manuscript presents the optical properties of directly deposited films of gold nanoparticles (AuNPs) prepared by ultrasonic spray pyrolysis technology. Four samples were produced, with an AuNPs deposition time of 15 min, 30 min, 1h and 4h on the glass substrate. The morphological characterization of the deposited films showed that the size of the first deposited AuNPs is between 10 and 20 nm, while with a longer duration of the deposition process, larger clusters of AuNPs grow by coalescence and aggregation. The prepared layers were optically characterised with Ultraviolet–visible spectroscopy (UV-vis) and ellipsometry. The ellipsometric measurements showed an increasingly denser and thicker effective thickness of the AuNPs layers. The extinction spectra display a clear local surface plasmonic resonance (LSPR) signature (peak 520-540 nm), indicating the presence of isolated particles in all samples. For all AuNPs layers, the imaginary part of both ε// and ε⏊ is dominated by a central peak around 2.2 eV corresponding to the LSPR of isolated particles and a high-energy shoulder due to Au interband transitions. It is shown that, as the density of particles increases, the extinction cross section grows over the whole spectral range where measurements are taken. Thus response can be explained with an enhanced electromagnetic response among AuNPs, that can be connected to the increase of particle density but also by the formation of clusters and irregular structures.

Highly transparent, conductive nanosheets are extremely attractive for advanced optoelectronic applications. Previously, we have demonstrated that transparent, conductive Au nanosheets can be prepared by UV irradiation of Au nanoparticle (AuNP) monolayers spread on water, which serves as the subphase. In this study, we show that this UV irradiation technique can easily be extended as a nanosheet modification method by using subphases containing metal ions. That is, aqueous solutions containing Au and Ag ions are effective in modifying Au nanosheets. HAuCl4 solutions increased Au nanosheet thickness, which improved conductivity, but reduced transparency. On the other hand, use of aqueous AgNO3 and CH3COOAg solutions yielded Au-Ag hybrid nanosheets; however, their morphologies depended on the electrolytes used. Homogeneous Au-Ag nanosheets from AgNO3 and AuNPs covered with an Ag nanosheet from CH3COOAg were also produced. Further, these Au-Ag hybrid nanosheets had high conductivity without reduced transparency. Therefore, this UV irradiation method, modified by adding metal ions is quite effective at improving and diversifying properties of Au nanosheets.

Sunscreens are topical preparations containing one or more compounds that protect humans from ultraviolet (UV) light. This review focuses on analytical methods, particularly liquid chromatography, aimed at identifying and determining UV filters (UVFs) in environmental and marine biota matrices. A literature review was done using NIH (PubMed and Medline), FDA and EPA databases, Google Scholar, and Federal Regulations. This retrospective literature review is focused on the last five years. UVFs quantification in environmental and biological matrices showed a wide array of methods where liquid chromatography is predominant. The scientific literature identified a large variety of analytical methodologies that are compared in this review, to evaluate the better results in terms of limits of quantification and the possibility to identify as many analytes as possible simultaneously.

Bixa orellana dye was extracted from the seed by method of Soxhlet extraction using ethanol and a percentage yield of 4.3 was obtained. Recrystallisation of the dye was performed using chloroform and ethanol. TLC was used to separate the dye into Bixin and Norbixin with R_f values of 0.56 and 0.42 respectively. The extracted dye was pH sensitive, insoluble in water but soluble in some organic solvents like ethanol and acetic acid . Characterisation by UV-spectroscopy showed maximum absorption for Bixin and Norbixin at 457nm and 453nm respectively. FTIR spectrum of Bixin and Norbixin showed bands confirming similar functional groups for both Bixin and Norbixin except for –CO- stretching and methylene vibration which was absent in Norbixin.

This review emphasizes the significance of dietary carotenoids in promoting skin health and appearance. The applications of carotenoids in health-promoting and nutrient products and their potential health effects are discussed. Food technology, health, nutrition, dermatology, and dermoaesthetics have recently focused on colorless carotenoids, particularly phytoene, and phytofluene, which have the unique ability to absorb UV radiation. These carotenoids, being significant components of the human diet, are readily bioavailable and have demonstrated various health-promoting actions in re-cent studies. Their distinct structures and properties, such as light absorption, oxidation sensitivity, stiffness, aggregation tendency, and even fluorescence in the case of phytof-luene, contribute to their potential benefits. Furthermore, it is recommended to assess the levels of these carotenoids to evaluate skin carotenoid status accurately.

Lichens are unique extremophilic organisms due to their phenomenal resistance to adverse environmental factors, including ultraviolet (UV) irradiation. Melanization plays a special role in the protection of lichens from UV-B stress. In present study, we analyzed the binding of melanins with the components of cell walls of mycobiont of upper cortex in the melanized lichen thalli Lobaria pulmonaria. Using scanning electron and atomic force microscopy, the morphological and nanomechanical characteristics of melanized layer of mycobiont cells were visualized. Melanization of lichen thalli led to the smoothing the surface relief and thickening mycobiont cell walls and resulted in the increase of deformation values and reduction of adhesion properties of lichen thallus. Treatment of thalli with hydrolytic enzymes, especially chitinase and lichenase, enhanced the yield of melanin from melanized thalli and release of carbohydrates, while treatment with pectinase increased the release of carbohydrates and phenols. Our results suggest that melanin by firmly binding with hyphal cell wall carbohydrates, particularly chitin and 1,4-β-glucans, can form associations, strengthen the melanized upper cortex of lichen thalli, and thereby can contribute to lichen survival under UV stress.

Three octahedral iron(III) nitrate complexes with dimethylated urea ligand isomers, [hexakis(N,N’-dimethylurea-O)iron(III)] nitrate (compound 1), trans-[diaquatetrakis(N,N-dimethylurea-O)iron(III)] nitrate (compound 2) and [hexakis(N,N-dimethylurea-O)iron(III)] nitrate trihydrate (compound 3) were prepared and characterized with single crystal X-ray diffraction, IR, Raman and UV-VIS methods. In compounds 1 and 3, six dimethylurea ligands coordinate to the central FeIII ion via the oxygen in octahedral geometry and the ligands are arranged in a propeller-like manner, dividing the complex cations into two sides. In compound 1, the dimethylurea propellers screw in the opposite direction on the two sides of the complex and in compound 3, they are arranged with the same handedness on the two sides. The complexes have helical chirality. The two sides of the complex cations differ not only in the rotation direction of the ligands but also in the hydrogen bond formation. On one side of the complex cation, the ligands form intermolecular hydrogen bonds only with the crystal waters, meanwhile on the other side of the complex, the ligands form hydrogen bonds only with the nitrate ions. In compound 2, [Fe(N,N-dimethylurea)4(H2O)2]3+ cations form layers that are separated by interconnected NO3- ions forming a hydrogen bonding system and connecting the complex cations A-s and B-s. The three crystallographically different nitrate ions each form four hydrogen bonds in a way that they have one bidentate O atom and two monodentate O atoms, however, the anions differ in their hydrogen bonding. The spectroscopic characteristics of compound 2 were determined by IR measurements on the deuterated compound 2 as well.

To investigate the photosynthetic change characteristics of mango leaves under enhanced UV-B radiation, adult ‘Tainong No. 1’ mango (Mangifera indica) trees were treated (N=nine individuals) with simulated enhanced UV-B radiation [24 and 96 kJ/(m2·d)] in the field, and the photochemical reactions, activities of key enzymes in carbon assimilation, and the expression of genes were observed. The results showed that compared with the control, there was a decrease in tree yield and nutritional flavor quality of the fruits under the 96 kJ/(m2·d) treatment, while no significant changes were observed under 24 kJ/(m2·d). After 20 or 40 days, leaves’ net photosynthetic rate (Pn), stomatal conductance (Sc), transpiration rate (Tr), intercellular CO2 concentration (Ci), and chlorophyll a/b under exposure to 96 kJ/(m2·d) of UV-B were significantly lower than in the control, whereas chlorophyll a, chlorophyll b, carotenoids, Hill reaction activity, photochemical quenching coefficient (qP), and Rubisco activities were significantly higher. By contrast, the Hill activity and Rubisco activity under 24 kJ/(m2·d) were significantly higher than the control, while Pn, Sc, Tr, Ci, and the content of photosynthetic pigments were similar to the control. The expression of gene coding the Rubisco big subunit (rbcL) was inhibited by the 96 kJ/(m2·d) treatment. We conclude that stomatal limitation was directly induced by 96 kJ/(m2·d), resulting in the inhibition of photosynthesis and the reduction in yield and deterioration of the quality of mango.

Drug abuse is a major issue worldwide. Methylphenidate (MPH) and alprazolam (ALZ) are commonly prescribed drugs for the treatment of ADHD and anxiety disorders, respectively. The limited studies suggest that abusers primarily use benzodiazepines to counteract adverse effects associated with methylphenidate usage. The main aim of this study was to investigate the interaction of drugs with DNA using spectroscopic methods. Female Wistar rats were administered with MPH (10, 20, 40 mg/kg) and ALZ (5, 10, 20 mg/kg) alone and in combination for a period of 28 days. The FT-IR and UV results reveal some spectral changes in a dose-dependent manner, which indicates interactions of drugs with DNA. Thus, the changes in spectral peaks provide some insight into the mechanism of the interaction of drugs with DNA.

In recent years, the nanoparticles applications have been well recognized in various fields. It is known that nanoparticles as an active ingredient in sunscreens are widely used. Zinc oxide and titanium oxide nanoparticles are common nanoparticles utilized in sunscreens. In this study, we aimed to suggest new nanoparticles for this purpose. Cerium oxide nanoparticles (CeO2-NPs) were synthesized by using Musa sapientum fruit peel extract. Synthesized nanoparticles were identified through Raman, Powder X-ray Diffraction (PXRD), Fourier Transform Infrared spectroscopy (FT-IR), Field Energy Scanning Electron Microscopy (FESEM) and Energy-Dispersive Spectroscopy (EDX). The results showed that size of synthesized nanoparticles are in range 4-13 nm. The cytotoxic activity of synthesized nanoparticles on lung (A549) cancer cell line was performed through MTT assay. The results showed that synthesized nanoparticles are non-toxicity against A549 cell line to below 500 μg/mL of nanoparticles concentration. The Sun protection factor (SPF) was estimated ~ 40 for synthesized CeO2-NPs. So, synthesized nanoparticles can be a good option for use in the cosmetics industry.

The search for molecular biosignatures at the surface of Mars is complicated by an intense irradiation in the mid- and near- ultraviolet (UV) spectral range for several reasons: (i) many astrobiologically relevant molecules are electronically excited by efficient absorption of UV radiation and rapidly undergo photochemical reactions; (ii) even though the penetration depth of UV radiation is limited, aeolian erosion continually exposes fresh material to radiation; and (iii) UV irradiation generates strong oxidants such as perchlorates that can penetrate deep into soils and cause subsurface oxidative degradation of organics.As a consequence, it is crucial to investigate the effects of UV radiation on organic molecules embedded in mineral matrices mimicking the martian soil, in order to validate hypotheses about the nature of the organic compounds detected so far at the surface of Mars by the Curiosity rover, as well as organics that will be possibly found by the next rover missions Mars 2020 and ExoMars 2020. In addition, studying the alteration of possible molecular biosignatures in the martian environment will help to redefine the molecular targets for life detection missions and devise suitable detection methods.Here we report the results of mid-UV irradiation experiments of Mars soil analog samples obtained adsorbing relevant organic molecules on a clay mineral that is quite common on Mars, i.e. montmorillonite, doped with 1 wt% of magnesium perchlorate. Specifically, we chose to investigate the photostability of a plausible precursor of the chlorohydrocarbons detected on Mars by the Curiosity rover, namely phthalic acid, along with the biomarkers of extant life L-phenylalanine and L-glutamic acid, which are proteomic amino acids, and adenosine 5’-monophosphate, which is a nucleic acid component.We monitored the degradation of these molecules adsorbed on montmorillonite through in situ spectroscopic analysis, investigating the reflectance properties of the samples in the Near InfraRed (NIR) spectral region. Such spectroscopic characterization of molecular alteration products provides support for two upcoming robotic missions to Mars that will employ NIR spectroscopy to look for molecular biosignatures, through the instruments SuperCam on board Mars 2020, ISEM, Ma_Miss and MicrOmega on board ExoMars 2020.

ZnO films with Ti atoms incorporated (TZO) in a wide range (0-18 at. %) have been grown by reactive co-sputtering on silicon and glass substrates. The influence of the titanium incorporation in the ZnO matrix on the structural and optical characteristics of the samples has been determined by Rutherford backscattering spectroscopy (RBS), X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD). The results indicate that the samples with low Ti content (< 4 at. %) exhibit the wurtzite-like structure, with the Ti+4 ions substitutionally incorporated into the ZnO structure, forming Ti-doped ZnO films. In particular, very low concentration of Ti (<0.9 at. %) leads to a significant increase of the crystallinity of the TZO samples. Higher Ti contents give rise to a progressive amorphization of the wurtzite-like structure so samples with high Ti content (≥18at. %), displays an amorphous structure indicating the XPS analysis a predominance of Ti-O-Zn mixed oxides. The energy gap, obtained from absorption spectrophotometry, increases from 3.2 eV for pure ZnO films to 3.6 eV for those with the highest Ti content. Ti incorporation in the ZnO samples below 0.9 at. % rises both, the blue (380 nm) and green (550 nm) bands of the photoluminescence (PL) emission, thereby indicating a significant improvement of PL efficiency of the samples.

The effective treatment for chronic wounds constitute one of the most common worldwide health care problem due to the presence of high levels of proteases, free radicals and exudates in the wound, which constantly activate the inflammatory system avoiding the tissue regeneration. In this study, we describe a multifunctional bioactive and resorbable membrane with in-built antioxidant agent for the continuous quenching of free radicals as well as to control inflammatory response helping to promote the wound healing process. To reach that goal synthesized statistical copolymers of N-vinylcaprolactam (V) and 2-hydroxyethyl methacrylate (H) have been conjugated with catechol bearing hydrocaffeic acid (HCA) molecules. The natural polyphenol (catechol) is the key molecule responsible for the mechanism of adhesion of mussels, and provides the functionalized polymer conjugate a continuous antioxidant response, antiinflammatory effect, UV screen and bioadhesion in the moist environment of the human body, all of them key features in the wound healing process. Therefore, these novel mussel-inspired materials have an enormous potential of application and can act very positively, favoring and promoting the healing effect in chronic wounds.

In this study, a new type of temperature sensor device was developed. The circular electrode of the thermal sensitive sensor was modified with TMT and O₂ plasma to enhance the conductivity by forming a thin SnOxCy layer on the electrode surface. The Nano-Au particles were subjected to O₂ plasma pretreatment to form peroxide groups on the surface. The thermally sensitive sensor was made by mixing the above-treated Nano-Au particles with N-isopropylacrylamide (NIPAAm) to form solution and then UV-induced grafting polymerization of the NIPAAm-containing solution onto the electrode substrate. The composite hydrogels on the electrode introduce thermo-sensitive polymeric surface films for temperature sensing. Using ambient environment resistance test to measure the resistance, the LCST (lower critical solution temperature) of Nano-Au (MUA) mixed with NIPAAm hydrogel was found to be 32 °C. At ambient temperatures higher than LCST, the electrode resistance decreases linearly.

Aflatoxin M1 (AFM1), a carcinogenic toxin, mainly contaminates cow milk and poses significant challenges to the dairy industry and, thereby, public health in most tropical countries, including Ethiopia. This study aimed to investigate the prevalence and level of AFM1 in raw cow milk collected from Specialized Dairy Farms and Milk Vendors in three Urban Centers: Chiro town, Dire Dawa, and Harar cities in Eastern Ethiopia. 180 milk samples were collected from two major Milk Sources: Specialized Dairy Farms (N=90) and Milk Vendors (N=90) using a simple random sampling technique. AFM1 was analyzed using high-performance liquid chromatography. The study revealed that, 63.9% (115/810) prevalence and 0.179±0.48µg/L mean of AFM1 level in milk, which 39.13% and 26.08% exceed the tolerable limits of EU and FDA respectively. A 40.0% prevalence and 0.344±0.72µg/L average of AFM1 in the milk from Dire Dawa city was significantly higher than the samples from other Urban Centers. However, 30.43% and 29.57% prevalence as well as 0.055±±0.13µg/L and 0.140±0.33µg/L average of AFM1 in milk from Chiro town and Harar city, respectively were not significant. Furthermore, milk samples from Dairy Farms revealed a higher prevalence (57.39%) and level (0.252±0.64µg/L) of AFM1 than Milk Vendors, with a prevalence of 42.61% and an average of 0.107±0.21µg/L. Therefore, this study concludes that substantial milk samples were contaminated by AFM1 and urges the need to enhance farmers' awareness on mitigation.

Vitamin D is crucial for athletes' health and performance. Although the exact vitamin D requirements for athletes have not been established, maintaining 25(OH)D levels of at least 40 ng/ml is considered beneficial. This study aimed to determine whether an individual loading dose formula for vitamin D supplementation is suitable to meet a target value of 40 ng/ml and is more effective than standardized supplementation. In a 10-week supplementation study during the winter months in Germany, 90 athletes with insufficient vitamin D levels (25(OH)D<30ng/ml) were randomly assigned to receive either a universal dose of 2000 IU/day of vitamin D or a loading dose of 4000 IU/day, followed by a maintenance dose of 1000 IU/day. Total 25(OH)D concentration was measured from dried blood spots at three time points: at baseline, at the computed date of 40ng/ml, and after the 10-week period. Additionally, a vitamin D-specific questionnaire was issued. On the day when 25(OH)D blood concentrations of 40 ng/ml were calculated to prevail, the individualized group had a significantly higher 25(OH)D level than the standardized group (41.1+-10.9 ng/ml vs. 32.5+-6.4 ng/ml, p<0.001). This study demonstrated that the examined formula is suitable for athletes to achieve a 25(OH)D concentration of 40 ng/ml. This indicates that a personalized approach is more effective than a one-size-fits-all approach in restoring adequate vitamin D levels in athletes.

Andean blueberries are wild berries grown and consumed in Ecuador which contain high values of bioactive compounds, mainly anthocyanins, with powerful antioxidant activity. The aim of this study was to evaluate the profile and contents of (poly)phenols and carotenoids in Andean blueberry by HPLC-DAD-MSn and determine a wide range of its biological activities. The antioxidant capacity of this fruit was evaluated in vitro by three different methods and in vivo using the zebrafish animal model, also the toxicity effect was determined by the zebrafish embryogenesis test. Besides, the antimicrobial activity and the capacity of Andean blueberry to produce hemagglutination in blood cells were evaluated. Finally, the bioaccessibility of (poly)phenols and related antioxidant capacity were determined in the different phases of an in vitro digestion. The global results indicated no toxicity of Andean blueberry, weakly bacteriostatic activity, and high contents of anthocyanins and antioxidant capacity, which were partially bioaccesible in vitro (~ 50 % at the final intestinal step), contributing to the knowledge of its health benefits for consumers and its potential use in the food and pharmaceutical industry as functional ingredient.

Opium poppy (Papaver somniferum L.) is an ancient medicinal plant producing pharmaceutically important benzylisoquinoline alkaloids. In the present work we focused on the study of enzyme lipoxygenase (LOX, EC 1.13.11.12) from opium poppy cultures. LOX is involved in lipid peroxidation and lipoxygenase oxidation products of polyunsaturated fatty acids have a significant role in regulation of growth, development and plant defence responses to biotic or abiotic stress. The purpose of this study was to isolate and characterize LOX enzyme from opium poppy callus cultures. LOX was purified by ammonium sulphate precipitation followed by hydrophobic chromatography using Phenyl-Sepharose CL-4B and hydroxyapatite chromatography using HA Ultrogel sorbent. SDS-PAGE analysis and immunoblotting revealed that LOX from opium poppy cultures was a single monomeric protein showing the relative molecular weight of 83 kDa. To investigate the positional specificity of the LOX reaction, purified LOX was incubated with linoleic acid and the products were analysed by high-performance liquid chromatography. LOX converted linoleic acid primarily to 13-hydroperoxy-(9Z,11E)-octadecadienoic acids (78%) and to a lesser extent to 9-hydroperoxy-(10E,12Z)-octadecadienoic acids (22%). Characterization of LOX from opium poppy cultures provided valuable information in understanding of LOX involvement in regulation of signalling pathways leading to biosynthesis of secondary metabolites with significant biological activity.

This study had the objective to evaluate the effect of Piper betle L. powder (PP) at 5 different doses in substrate incubated by sunflower oil as secondary function of PUFA using in vitro gas production technique. The treatments of this study were run as a 2X5 factorial arrangement in a completely randomised design using the PROC GLM procedure of SAS 9.4: (1) control (S1) without supplementation of PP; (2) 15 mg PP (S2); (3) 30 mg PP (S3); (4) 45 mg PP (S4); and (5) 60 mg PP (S5), while sunflower oil was supplemented in all treatments: low 15 mg/incubation and high 30 mg/incubation. A 500 mg of TMR (hay: concentrate, 50:50) was assigned to basal substrate. The PP containing 1.84 mg/g DM quercetin and 1.00 mg/g DM eugenol altered rumen fermentation without change pH (p < 0.001) and methane production was lesser (p < 0.001) about -30% and -25% for DM and OM measurement, respectively. Gas kinetic, degradability, and ammonia level was significantly affected by supplementing PP (p < 0.01). Overall, this study suggested quercetin and eugenol deriving from PP acted three major accelerations: assembled carbon dioxide, behaved antimicrobial role and performed the balance water molecules in the rumen kinetic. This study suggests that PP promotes changing in vitro rumen fermentation and diminishing methane production within recommended doses, 0.1-15 mg/incubation in DM.

Veratrum californicum is a rich source of steroidal alkaloids such as cyclopamine, a known inhibitor of the Hedgehog (Hh) signaling pathway. Here we provide a detailed analysis of the alkaloid composition of V. californicum by plant part through quantitative analysis of cyclopamine, veratramine, muldamine and isorubijervine in the leaf, stem and root/rhizome of the plant. To determine if additional alkaloids in the extracts contribute to Hh signaling inhibition, we replicated the concentrations of these alkaloids observed in extracts using commercially available standards and compared the inhibitory potential of the extracts to alkaloid standard mixtures using Shh-Light II cells. Alkaloid combinations enhanced Hh signaling pathway antagonism compared to cyclopamine alone, and significant differences were observed in the Hh pathway inhibition between the stem and root/rhizome extracts and their corresponding alkaloid standard mixtures, indicating that additional alkaloids present in these extracts contribute to Hh signaling inhibition.

Prunus dulcis is one of the most widely cultivated species in the world. Its fruit (almond) is rich in various nutritious and bioactive compounds that exert several beneficial effects. The aim of this study was to determine the chemical profile and evaluate the biological potential in vitro of almond shell extracts. The chemical analysis of shell extracts led to the identification of 15 com-pounds by HPLC-DAD of which 11 were first detected in the almond plant. Twenty-six volatile compounds were identified by the GC-MS technique, among them; seven were firstly detected in the studied plant. For the biological activities, the extracts demonstrated moderate inhibition potential against the antioxidant, antidiabetic, and cytotoxic activities. The methanol extract at 50 µg/mL showed the highest antioxidant (45%) and antidiabetic activities (45% against alpha-glucosidase and 31% against alpha-amylase extracts), while the cyclohexane and dichloromethane at 50 µg/mL showed the highest cytotoxic activity towards Hela (32.2% with cyclohexane), and RAW 264-7 (45% with dichloromethane). Overall, these findings demonstrate the potential of almond shell extracts as a source of bioactive compounds that could be applied in the pharmaceutical and medical fields.

Natural carotenoids are secondary metabolites that exhibit antioxidant, anti-inflammatory and anti-cancer properties. These types of compounds are in high demand by pharmaceutical, cosmetic, textile and food industries, leading to the search for new natural sources of carotenoids. In recent years, the production of carotenoids from bacteria has become of great interest for industrial applications. In addition to carotenoids with C40-skeletons, some bacteria have the ability to synthesize characteristic carotenoids with C30-skeletons. In this regard, a great variety of methodologies for the extraction and identification of bacterial carotenoids has been reported and this is the first review that condenses much of this information. To understand the diversity of these carotenoids, we present their biosynthetic origin in order to focus on the methodologies employed in their extraction and characterization. Special emphasis has been made on high-performance liquid chromatography-mass spectrometry (HPLC-MS) for the analysis and identification of bacterial carotenoids. We end up this review showing their potential commercial use of bacterial carotenoids. This review is proposed as a guide for the identification of these metabolites, which are frequently reported in new bacteria strains.

Expanding earlier observations, we show that many melanin materials, in vitro synthesized from a wide range of precursors, can be fractionated into a dark-colored precipitate and a near-colorless, dispersible fraction. The dispersible fractions exhibited absorbance in the UVA and UVB range of the electromagnetic spectrum, but none in the visible range. In addition, fluorescent properties were associated with all dispersible fractions obtained. FT-IR spectroscopic analyses were performed to compare both types of fractions. Overall, it appears that some of the properties associated with melanin (UV absorbance, fluorescence) may not necessarily reside in the dark-colored portion of melanin, but in a colorless fraction of the material. It remains to be seen whether any of these in vitro observations have any relevance in vivo. However, we raise the possibility that the presence of a colorless fraction within melanin materials, and their associated properties, may have received inadequate attention. Given the important association between melanin, UV protection, and skin cancer, it is worthwhile to consider this additional aspect of melanin chemistry.

In this work, zinc oxide (ZnO)/ granular activated carbon (GAC) composites at different ZnO concentration (0.25M-ZnO@GAC, 0.5M-ZnO@GAC, and 0.75M-ZnO@GAC) were prepared by an in-situ hydrothermal method and demonstrated synergistic photocatalytic degradation and ad-sorption of Rhodamine B (RhB). The thermal stability, morphological structure, elemental com-position, crystallographic structure, and textural properties of developed catalysts were charac-terized by thermal gravimetric analysis (TGA/DTG), scanning electron microscopy equipped with energy dispersive-x-ray (SEM-EDS), x-ray diffraction (XRD), and Brunauer–Emmett–Teller (BET) analysis. Successful loading of ZnO onto GAC was confirmed by SEM-EDS and XRD analysis. The BET surface areas of GAC, 0.25M-ZnO@GAC, 0.5M-ZnO@GAC, and 0.75M-ZnO@GAC were 474 m2/g, 450 m2/g, 453 m2/g, and 421 m2/g, respectively. The decrease in GAC could be attributed to the successful loading of ZnO on the GAC surface. Notably, 0.5M-ZnO@GAC exhibited the best photocatalytic degradation efficiency of 82% and 97% under UV-A and UV-C light over 120 min, attributed to improved crystallinity and visible light absorption. The photocatalytic degradation parameters revealed that lowering the RhB concentration, raising the catalyst dosage and pH be-yond the point of zero charge (PZC) would favor the RhB degradation. Photocatalytic reusability was demonstrated over 5 cycles. Scavenger tests revealed that the hydroxyl radicals (•OH), su-peroxide radicals (O2−•), and photoinduced hole (h+) radicals play a major role during the RhB degradation process. Based on the TOC results, the RhB mineralization efficiency of 79.1% was achieved by 0.5M-ZnO@GAC. Additionally, GAC exhibited a strong adsorptive performance towards RhB with adsorption capacity and RhB removal of 487.1 mg/g and 99.5% achieved within 90 min equilibrium time. The adsorption characteristics were best described by pseu-do-second-order kinetics, suggesting chemical adsorption. This research offers a new strategy for the development of effective photocatalyst materials with potential for wider wastewater treat-ment applications.

Seaweeds have been the target of much attention due to the known bioactivities of some of their compounds. Although many studies about the application of seaweeds in various areas are available, these data are mostly about the raw extracts and not about specific compounds, such as seaweeds pigments. Moreover, the fungal infections of the skin, nails and hair caused by dermatophytes, are the most common fungal infections in humans. These pathologies require long periods of topical and/or systemic treatment that are associated with adverse effects and contraindications. This study had two objectives, the first was to evaluate the possibility of isolating and characterizing the pigments of Calliblepharis jubata and Fucus vesiculosus; the second was to assess their antifungal activity. The extraction of pigments was performed using a method of extraction by exhaustion; the purification was carried out by column chromatography. Three techniques were used to characterize the pigments, thin layer chromatography (TLC), UV-visible spectrophotometry and Fourier-transform infrared spectroscopy (FTIR). The antifungal activity was evaluated using the micro-dilution method, following the EUCAST international standards, on three dermatophytes, Trichophyton rubrum, Trichophyton mentagrophytes and Microsporum canis. It was possible to observe that the extract obtained from the seaweed C. jubata and F. vesiculosus, corresponding to the purified pigment phycobiliprotein, and the crude extract, an enriched extract, obtained from the seaweed C. jubata showed antifungal activity against the three fungal agents of human skin infection.

This study aimed to investigate the atherogenicity (quality) of LDL particles in patients with acute and recovered from COVID-19 infection. The participants were adults, aged 18 years or older of both sexes. Those with positive RT-PCR results at baseline were included in the Acute COVID-19 group (n=33), and those with negative RT-PCR six months after acute infection, were included in the Recovered COVID-19 group (n=30). The LDL quality was evaluated using three validated methods: Z-scan, UV-visible spectroscopy, and Lipoprint. The Recovered COVID-19 group showed significantly higher numbers of large LDL particles (less atherogenic) than the Acute COVID-19 group (P&lt;0.05). We also found that COVID-19 infection was associated with the oxidative modification of LDL particles. D-dimer and CRP levels were correlated with Z-scan results and antioxidant-amount estimate. Moreover, we noticed that the infection left a sequel in LDL quality, even after six months of recovery. These findings highlight the importance of monitoring lipids during and after recovery from COVID-19 infection, and their potential deleterious effect on the LDL profile might correlate with the progression of atherosclerosis and poor clinical outcomes.

The emergence of Non-Ionizing Radiation (NIR) has stimulated the growth and advancement of technological civilization. NIR is ubiquitous; it has helped improve all fields and even deepened democratic processes through the transfer of data and electronic transmission of results etc. A breathtaking part of NIR is its recent application in the treatment of adverse health effects through radiofrequency ablation, phototherapy, etc. as summarized in Table 1. The benefits of NIR are enormous, however, technological civilization has not come without a cost; some of the adverse health effects associated with NIR include skin cancers, sleep disorders, photo-aging, etc. as summarized in Table 2. Therefore, with the rapid and sporadic increase in the sources of NIR from Natural and Artificial sources including fossil fuel burning, Starlink technology, mobile phones, mobile base antennas, etc., background radiation is expected to rise beyond the exposure limit leading to health illnesses. Intrinsically, there is a dare need to marry concerns that evolve with this growth. Hence, this review article aims to congregate the health hazards associated with NIR and the state-of-the-art applications of NIR in medicine and the health sector in Nigeria being a developing country. Part of our recommendations is that the government at all levels should set up enforcement agencies and policies to drive strict adherence to NIR as provided by ICNIRP; frequent-periodic assessments of background radiation in public places should be carried out due to the increasing sources. In addition, further research is needed to ascertain the health hazard from emerging sources of NIR like Starlink technology and to substantiate findings of non-thermal effects.

The Ramazzottius varieornatus tardigrade is an extremotolerant terrestrial invertebrate belonging to the phylum of Tardigrada. At a length of 0.1-1.0 mm, tardigrades are small animals with an exceptional tolerance to extreme conditions such as high pressure, chemicals and irradia-tion. These properties have been attributed to the recently-discovered Dsup protein. Dsup is a nucleosome-binding protein that prevents DNA damage against X-ray and oxidative stress without impairing cell life, also in Dsup-transfected animal and plant cells. However, the precise “protective” role of this protein is still under study. We performed experiments on human cells and shows that, as compared to control cells, Dsup+ cells are more resistant to UV-C exposure and H2O2. Real-time PCR identified different expression patterns of endogenous genes involved in apoptosis, cell survival and DNA damage repair in Dsup+ cells in response to H2O2 and UV-C. While H2O2 treatment in Dsup+ cells only marginally involved the activation of pathways responsible for DNA repair reinforcing the idea of a direct protective effect of the protein on DNA, in UV-C exposed cells, Dsup efficiently upregulates DNA damage repair genes. In conclusion, our data may help to delineate the different mechanisms by which the Dsup protein operates in response to different insults.
