Cardiovascular diseases, which are the leading cause of death in Croatia, are linked to the high prevalence of hypertension. Both are associated with high salt intake which was determined almost two decades ago when Croatian Action on Salt and Health (CRASH) was launched. The main objective of the present study was to evaluate salt, potassium, and iodine intake using 24-hour urine samples in a random sample of adult Croatian population, and to analyse trends in salt consumption after the CRASH has intensively started. Methods: In this national population-based EH-UH 2 study performed in 2018-2021 (participation rate 72 %) in which we analysed a single 24-hour urine sample, 1067 adult participants (mean age 57.12 (SD 13.9), men 35%) were included. Results: Mean salt and potassium intakes were 8.6 g/day (IQR 6.2-11.2) and 2.8 g/day (IQR 2.1-3.5), respectively, with sodium-to-potassium ratio of 2.6 (IQR 1.8-3.3). We detected a decrease of 17.6% (2 g/day less) in salt consumption compared to our previous salt-mapping study. However, only 13.7%, and 8.9 % met the WHO salt and potassium recommended target of 5 g/day, and 3.5 g/day, respectively. Salt intake was higher, and potassium ingestion was lower in rural vs. urban regions, and in continental vs. Mediterranean parts of Croatia. Moderate to severe iodine insufficiency was determined in only 3% adult participants. Conclusion: In the last fifteen years, salt consumption has been significantly reduced in Croatian adult population because of intensive and broad CRASH program led by the Croatian Society of Hypertension and the Croatian Hypertension League. However, salt intake is still too high, and potassium ingestion is too low. With expected more active engagement and help of the government, obtained results could be significantly improved. Salt reduction programs are most cost-effective methods of cardiovascular disease prevention and should be considered more seriously by the government and health policy makers.

myfood24 is a comprehensive self-completed online 24-hour dietary recall tool currently used for nutritional assessments in epidemiological research. However, its clinical application has been unexplored. This mixed methods prospective observational study explores the acceptability and usability of myfood24 in a clinical population, women with gestational diabetes (GDM). Women were recruited at their first diabetes antenatal clinic appointment. To assess acceptability and usability, they were asked to complete five 24-hour dietary recalls using myfood24 over two weeks and a user experience questionnaire; with a subset invited to participate in a one-to-one semi-structured interview. Of the 199 participants, mean maternal age was 33 years, mean booking BMI 29.7kg/m2, 36% primiparous, 57% White, 33% Asian. Of these 121 (61%) completed myfood24 at least once and 73 (37%) completed the user questionnaire; 15 were interviewed. Usability of myfood24 was measured using the System Usability Scale (SUS) and found to be good (mean 70.9, 95%CI 67.1, 74.6). Interviews identified several areas for improvement, including optimising its use for mobile devices. myfood24 appears to be acceptable and have potential to support self-management and behaviour change for women with GDM but requires adaptation to record blood glucose results alongside real-time tracking of diet.

Lead, known as a metal with high neurotoxicity to children, cadmium, which is a carcinogenic and bioaccumulative contaminant, and arsenic, a class 1 carcinogenic according to the International Agency for Research on Cancer, are toxic elements (TEs) whose relevant route of exposure may be diet. We determined the bio-accessible fraction of lead, cadmium, and arsenic from the diet of preschool children from two day care centers (DCC). A cross-sectional study was conducted with 64 one–four-year-old children from two DCCs where the 24-h duplicate diet samples were collected. The diet samples were analyzed by ICP-MS for lead, cadmium, and arsenic total concentrations (n = 64) and their bio-accessibility were analyzed for a subsample (n = 10). The dietary intake (DI) mean for lead, cadmium, and arsenic were 0.18 ± 0.11 µg kg⁻¹ bw, 0.08 ± 0.04 µg kg⁻¹ bw, and 0.61 ± 0.41 µg kg⁻¹ bw, respectively. All DI calculated for TEs, considering total intake, were found lower than the tolerable limits (TL) (European Union, or World Health Organization, WHO, when applicable) except for one child’s Pb intake. Bio-accessibilities ranged between 0% to 93%, 0% to 103%, and 0% to 69%, for lead, cadmium, and arsenic, respectively. Although DI for TEs has been found lower than TL, these reference values have been recently decreased or withdrawn since it was for lead and arsenic whose TL were withdrawn by WHO.

According to WHO, reducing salt intake to less than 5 g/day means less death, disability, and suffering from heart disease and stroke, with around 1.89 million lives saved every year. In this scoping review, we have reviewed the salt consumption pattern of South Asia. For this purpose, we searched online databases and websites and consulted key informants to address relevant unpublished reports. The inclusion criteria are English-language publications between January 2011 and October 2021, as well as research that evaluated salt intake in the South Asian region and reported salt/sodium 24-hr urine or spot urine samples involving at least 100 participants. A total of 2043 studies were screened, out of which 12 studies fulfilled all the criteria. Our review revealed that reported salt intake in the South Asian region ranges from 6.7-13.3 g/day, which was higher than the WHO-recommended maximum level of 5 g/day in all countries. Men consumed more salt than women. This review demonstrates the critical importance of implementing a salt reduction program in South Asian countries.

Progressive loss of retinal ganglionic cells (RGC) causes blindness in glaucoma. Elevated intraocular pressure (IOP) is the most important, treatable risk factor. Currently, the management of glaucoma is centred at reducing the IOP, and drugs in the form of topical drops are the first line of management. Drugs reduce IOP either by suppressing aqueous humour secretion or improving the aqueous humour outflow. Newer drugs added during the past three decades to the armamentarium of glaucoma treatment have targeted the aqueous outflow. With an evolving understanding of the pathogenesis of glaucoma, the role of 24-hour IOP control and other IOP-independent risk factors affecting ocular blood flow and RGC toxicity is also recognised. The role of available drugs in controlling IOP over 24-hours is being evaluated. Improvement of ocular blood flow and neuroprotection are seen as potential drug targets in preventing the loss of RGC. In this article, we review the pharmacotherapy of glaucoma based on current therapeutic principles.

This cross-sectional study evaluated the validity of three alternative methods compared to the gold standard 24-h urine collection for estimating dietary sodium intake, a modifiable risk factor for hypertension, among middle-aged and older adults with elevated blood pressure. These included spot urine collection (using Kawasaki, Tanaka, and INTERSALT equations), 24-h dietary recall, and food frequency questionnaire responses, compared against 24-h urine collection in a subset of 65 participants (aged 50–75 years, 58.5 % women, 61.6 % hypertensive) from the DePEC-Nutrition trial. The validity of the methods was assessed using bias, Spearman correlation coefficient (SCC), intraclass correlation coefficient (ICC), and Bland-Altman analysis. Among the alternative methods, spot urine collection using the Kawasaki equation showed the strongest correlation (SCC 0.238; ICC 0.119, 95 % CI -0.079 to 0.323) but it exhibited a significant bias (1414 mg/day, p-value < 0.001) relative to 24-h urine collection. Conversely, dietary surveys had a smaller bias but wider limits of agreement. These findings underscore the complexities of accurately estimating dietary sodium intake using spot urine collection or dietary surveys in this specific population, suggesting that a combination or refinement of existing methodologies might improve accuracy. Further research with larger samples is necessary to develop more reliable methods for assessing sodium intake in this high-risk group.

The upper ocean heat content in the equatorial Pacific usually serves as a primary precursor for an upcoming El Niño, while the strong atmospheric perturbations such as westerly wind burst and easterly wind surge sometimes play a decisive role in determining the final intensity of the event. The tropical Pacific Ocean has just experienced a rare 3-year La Niña, which accumulated a huge amount of warm water in the western basin by the winter of 2022. Using a state-of-the-art climate prediction system, here we show that the restored warm water is sufficient to boost a strong El Niño toward the end of 2023, and that an extreme event could take place if a few sizable westerly wind bursts would occur in spring and early summer. This prediction is not sensitive to initial errors within the tropical Pacific, but may be subject to some uncertainties brought about by influences from elsewhere.

Universitas Gadjah Mada (UGM) is implementing the concept of green campus by various means including through elements in its sustainable campus development plan. With regards to several factors related to Energy and Climate Change indicators set by UI GreenMetric, this research examined the existing condition of the area of UGM which belongs to academic zone in order to construct a proposal based on a building mass reconfiguration. This proposal was aimed to improve the implementation of energy-specific green campus concept within the Forestry cluster area as a sample case study. This selection was based on the average value of the Building Coverage Ratio (BCR) values in all of the clusters; thus, the scenario was relevant for generalization and could be replicated in other clusters in the university. The study was done in three stages of simulation, and was based on a series of digital simulation of sunlight hour and solar radiation run in Grasshopper using Lady Bug environmental analysis plug-in for a period of five summer months during the building office hours. Following the reconfiguration, results had shown a similar downtrend between the amount of sunlight hour and solar radiation in the area (up to 49% and 45% respectively on building envelopes, and up to 44% and 42% respectively on landscaping surfaces). This reduction indicated a potential of energy efficiency by applying selective building mass reconfiguration as a passive design strategy that goes hand in hand with the campus’s development policy to optimize the use of BCR for a greener UGM campus through its many undergoing and upcoming redevelopment projects.

Longitudinal cutting is a most common process in steel structure manufacturing, and the man-hours of the process provide an important basis for enterprises to generate production schedules.However, currently the man-hours in factories are mainly estimated by experts, and the accuracy of this method is relatively low.In this study,we propose a system that predicts man-hours with history data in the manufacturing process and that can be applied in practical structural steel fabrication.The system addresses the data inconsistency problem by one-hot encoding and data normalization techniques,Pearson correlation coefficient for feature selection,and the Random Forest Regression(RFR) for prediction.Compared with the other three Machine Learning(ML) algorithms, the Random Forest algorithm has the best performance.The results demonstrate that the proposed system outperforms the conventional approach and has better forecast accuracy, so that it is suitable for man-hours prediction.

Background. Metronidazole treats obligate anaerobic bacterial and protozoal infections, with an elimination half-life of around eight hours. The long elimination half-life, the favorable ratio of steady-state serum levels to minimum inhibitory concentration, and the presence of active metabolites lead to consideration of metronidazole use at 12-hour dosage intervals. This systematic review aimed to compare the clinical outcomes of twice-daily and thrice-daily metronidazole dosing. Methods. We used the Preferred Reporting Items for Systematic Reviews and Meta-Analyses checklist. PubMed, Scopus, Science Direct, the Cochrane Database of Systematic Reviews, and the Cochrane Central Register of Controlled Trials to systematically identify all relevant studies published up to June 16, 2023. Results. The final analysis included two published retrospective cohort studies; a single site (n = 200) and a multisite study (n = 85) of ‘good’ quality, as measured by the Newcastle-Ottawa scale. The reported baseline characteristics of the 8-hour and 12-hour dosing groups were comparable, and neither study identified significant differences in primary and secondary clinical outcomes. Meta-analysis of the need to escalate antibiotic therapy also showed no statistically significant differences using the Mantel-Haenszel fixed-effect method (95% CI: 47.6% lower to 6.4 times higher risk, p = 0.342) and inverse-variance method (RR: 1.87, 95% CI: 0.52-6.65, p = 0.336). Conclusions. Dosing metronidazole every 12 hours is as effective as every 8-hour dosing for anaerobic infections. Healthcare systems may consider the adoption of every 12-hour metronidazole dosing with continued evaluation of patient outcomes.

Brassinosteroids (BR) are key phytohormones involved in the regulation of major processes of cell metabolism guiding plant growth. In the past decades, new evidence made it clear that BRs also play a key role in the orchestration of plant responses to many abiotic and biotic stresses. In the present work we analyzed the impact of foliar treatment with 24-epicastasterone (ECS) on the endogenous content of major phytohormones (auxins, salicylic acid, jasmonic acid, abscisic acid) and their intermediates in soybean leaves 7 days following the treatment. Changes in the endogenous content of phytohormones have been identified and quantified by LC/MS. Obtained results point to a clear role of ECS in the upregulation of auxin content (indole-3-acetic acid, IAA) and downregulation of salicylic, jasmonic, and abscisic acid levels. These data confirm that under optimal conditions ECS in tested concentrations of 0,25 µM and 1 µM might promote growth in soybean by inducing auxin contents. Benzoic acid (a precursor of salicylic acid (SA)), but not SA itself, has also been highly accumulated under ECS treatment which indicates an activation of the adaption strategies of cell metabolism to possible environmental challenges.

Low-sodium salt substitute (LSSS) has the potential in regulating sodium and potassium intake and has been available in China market for years. However, its real-world use and impact to sodium\potassium intake is unclear. Baseline data of 4000 adult participants who participated in three similar-designed randomized controlled trails were pooled together for this study. Self-reported awareness and use of LSSS were collected using the uniform questionnaire, and the 24-h urinary sodium and potassium excretion was used to estimate their dietary intake. Mixed-effects models were developed to assess the relationship between LSSS and 24-h urinary sodium and potassium excretion. 32.0% of the participants reported awareness of LSSS and 11.7% reported its current use. After adjusting location, sex, age, and education, compared with the group of participants unaware of LSSS, participants who were aware but not using LSSS and those who were using LSSS had a lower 24-h urinary sodium excretion by –356.1(95% CI: –503.9, –205.9) mg/d and –490.6(95% CI: –679.2, –293.7), respectively (p&lt;0.001). No significant difference was found for 24-h urinary potassium excretion and sodium-to-potassium ratio among the three groups (p&gt;0.05). In conclusion, although associated with lower sodium intake, the awareness and real-world use of LSSS are still very low in China.

Abstract Objectives: Aging may challenge life and even affect individuals' well-being and flourishing. This includes challenges of shrinking social relations and being by yourself in old age while using your personal strengths. The current study examines two important personal resources, i.e., the skill of positive solitude and the 24 character-strengths, that may be associated with flourishing in old age. Methods: A convenience sample of 1085 community-dwelling adults (M = 57.20, SD = 6.24, range = 50-87), completed an online self-report survey with demographic measures and scales measuring personal strengths, the skill of positive solitude, and flourishing. Results: As hypothesized, the 24 character-strengths and positive solitude, were each associated with flourishing, and positive solitude moderated the relationships between the 24 character-strengths and flourishing. The 24 character-strengths and flourishing relationship was stronger among participants with lower levels of positive solitude. Conclusion: These findings highlight that despite the decrease in social relations which often characterizes old age, the skill of positive solitude serves as a valuable resource for flourishing in the later stages of life.

In this paper, a systematic literature review is presented, through a survey in the main digital databases, about modelling methods for Short-Term Load Forecasting (STLF) for hourly electricity demand for residential electricity and to realize the performance evolution and impact of the Artificial Intelligent (AI) in STLF. With these specific objectives, it was developed a conceptual framework on the subject, a systematic review of the literature based on scientific publications with high impact and a bibliometric study directed to the scientific production of AI and STLF. The review of research articles over a 10-year period, which takes place between 2012-2022, uses the Preferred Reporting Items for Systematic and Meta-Analyses (PRISMA) method. This research resulted in more than 300 articles, available in 4 (four) databases: Web of Science, IEEE Xplore, Scopus and Science Direct. The research was organized around 3 (three) central themes, which were defined through keywords: STLF, Electricity, Residential and their corresponding synonyms. 334 research articles were analyzed and identified the year of publication, journal, author, geography by continent and country and the area of application. Of the 334 documents found in the initial research and after applying the inclusion/exclusion criteria, which allowed delimiting the subject addressed in the topics of interest for analysis, 37 (thirty-seven) documents are in English (25 are journal articles and 12 are conference papers). The results point to a diversity of modelling techniques and associated algorithms. The corresponding performance is measured with different metrics and, therefore, cannot be compared directly. Hence, it should be desirable to have a unified data set together with a set of benchmarks with well-defined metrics for a clear comparison of all the modelling techniques and the corresponding algorithms.

Background: Breast cancer is a commonly diagnosed cancer worldwide. Human MutT homolog 1 (MTH1) is found to be elevated in breast tumors and cancer cells are addicted to MTH1 for sur-vival. Pharmacological inhibition of MTH1 may be potentially beneficial in the treatment of breast cancer. Methods: MA-24 was screened by malachite green colorimetric assay from MTH1 inhibitors and the kinetic characteristics of MA-24 was assessed. Binding features of MA-24 with MTH1 was as-certained through molecular docking, and cytotoxic activity of MA-24 was validated in vitro and in vivo. Target engagement assays, comet assay and western blot confirmed that the intracellular target and mechanism of MA-24. Results: MA-24 with potent antitumor bioactivity both in vitro and in vivo. MA-24 competitively inhibited the MTH1 and further induce DNA strand breaks, leading to increased apoptosis of cancer cells depending on upregulation of cleaved-caspase 3 – cleaved-PARP axis. Especially, MA-24 exhibited a powerful efficacy and safety in vivo (tumor growth inhibition rate: 61.8%). Conclusions: MA-24 possesses a broad spectrum of breast cancer cytotoxicity and offered valuable insights for overcoming the challenges of chemotherapy-related toxicity, which holds great po-tential for further development MA-24 as an anti-cancer drug.

We investigated the effect and molecular mechanism of 24-MCF-induced PPAR-γ2 on angiogenesis-related genes in MCF7 cells. cDNA microarray, semi-quantitative reverse transcription (RT)-PCR, and western blotting revealed that 24-MCF mediated the expression of genes related to angiogenesis in MCF-7 cells. Luciferase reporter assay demonstrated that promoter activation of the LIF gene, an anti-angiogenesis factor, was increased upon PPAR-γ2 overexpression and 24-MCF treatment, whereas activation of HoxA7 and VEGF promoters, known pro-angiogenesis factors, decreased upon PPAR-γ2 overexpression and 24-MCF treatment. We identified PPAR-response elements (PPRE) located in the VEGF (-913 to +1), HoxA7 (-1107 to +1), and LIF promoter regions (-9032 to -8403). VEGF promoter activity was abolished by mutation of the PPRE motif. Treatment with 24-MCF inhibited expression of VEGF and inhibited the Akt/mTOR pathway. Treatment with 24-MCF also decreased VEGF secretion in MCF7 cells and PMA-stimulated tube formation in HUVECs. Our findings suggest that 24-MCF induces PPAR-γ2-mediated regulation of anti-angiogenesis via PPRE motifs in VEGF, HoxA7, and LIF promoters or upstream regions. Furthermore, 24-MCF treatment inhibits angiogenesis by blocking VEGF secretion.

In this work, we introduce a scalable and efficient GPU-accelerated methodology for volumetric particle advection and finite-time Lyapunov exponent (FTLE) calculation, focusing on the analysis of Lagrangian Coherent Structures (LCS) in large-scale Direct Numerical Simulation (DNS) datasets across incompressible, supersonic, and hypersonic flow regimes. LCS play a significant role in turbulent boundary layer analysis, and our proposed methodology offers valuable insights into their behavior in various flow conditions. Our novel owning-cell locator method enables efficient, constant-time cell search, and the algorithm draws inspiration from classical search algorithms and modern multi-level approaches in numerical linear algebra. The proposed method is implemented for both multi-core CPUs and Nvidia GPUs, demonstrating strong scaling up to 32,768 CPU cores and up to 62 Nvidia V100 GPUs. By decoupling particle advection from other problems, we achieve modularity and extensibility, resulting in consistent parallel efficiency across different architectures. Our methodology was applied to calculate and visualize the FTLE on four turbulent boundary layers at different Reynolds and Mach numbers, revealing that coherent structures grow more isotropic proportional to the Mach number, and their inclination angle varies along the streamwise direction. We also observed increased anisotropy and FTLE organization at lower Reynolds numbers, with structures retaining coherency along both spanwise and streamwise directions. Additionally, we demonstrated the impact of lower temporal frequency sampling by upscaling with an efficient linear upsampler, preserving general trends with only 10% of the required storage. In summary, we present a particle search scheme for particle advection workloads in the context of visualizing LCS via FTLE that exhibits strong scaling performance and efficiency at scale. Our proposed algorithm is applicable across various domains requiring efficient search algorithms in large structured domains. While this manuscript focuses on the methodology and its application to LCS, an in-depth study of the physics and compressibility effects in LCS candidates will be explored in a future publication.

Background: The immunomodulatory role of 1,25-Dihydroxy vitamin D3 (1,25(OH)2D3) is exerted through its interaction with the Vitamin D receptor (VDR) present on pancreatic and immune cells. While a deficiency in Vitamin D has been linked to Type 1 Diabetes Mellitus (T1DM), the exact molecular mechanism driving this down-regulation in T1DM is yet to be fully understood. This study aimed to decipher differences in the expression of genes associated with Vitamin D metabolism in T1DM patients and to ascertain if there is a correlation between serum 1,25(OH)2D3 levels and the expression of these genes. We also sought to understand the influence of specific microRNAs (miRNAs) on the expression of Vitamin D metabolism genes in peripheral blood mononuclear cells (PBMCs) of T1DM patients. Furthermore, the study delved into the potential implications of altered Vitamin D metabolism genes and miRNAs on autoimmune processes. Methods: Utilizing real-time PCR, we assessed the expression profiles of genes encoding for 1-hydroxylases (CYP27B1) and 24-hydroxylases (CYP24A1), as well as related miRNAs, in PBMCs from 30 T1DM patients and 23 healthy controls. ELISA tests facilitated the measurement of 1,25(OH)2D3, GAD65, and IA-2 levels. Results: Our findings showcased down-regulated CYP27B1 mRNA levels, while CYP24A1 expression remained stable compared to healthy subjects (CYP27B1, p = 0.0005; CYP24A1, p = 0.205, respectively). In T1DM patients, the levels of has-miR-216b-5p were found to be increased, while the levels of has-miR-21-5p were decreased in comparison to the control group. Notably, no correlation was identified between the expression of CYP27B1 in T1DM patients and the levels of has-miR-216b-5p, has-miR-21-5p, and 1,25(OH)2D3. A significant negative correlation was identified between CYP27B1 mRNA levels in PBMCs of T1DM and IA2, but not with GAD65. Conclusions: The study highlights there were reduced levels of both CYP27B1 mRNA and has-miR-21-5p, along with elevated levels of has-miR-216b-5p in the PBMCs of T1DM. However, the absence of a correlation between the expression of CYP27B1, levels of has-miR-216b-5p, and the status of 1,25(OH)2D3 suggests the possible existence of other regulatory mechanisms. Additionally, the inverse relationship between IA2 autoantibodies and CYP27B1 expression in T1DM patients indicates a potential connection between this gene and the autoimmune processes inherent in T1DM.

The paper presents data from a monthly campaign for the elemental composition of PM10 in a specific receptor Kotórz Mały (Opole Voivodeship) located in the vicinity of a moderately inhabited rural area, measured in one-hour samples with the use of the PX-375 analyzer by the Horiba company. The hourly variability of SO2, NO, NO2, CO, O3 concentrations as well as the variability of meteorological parameters were also determined. On average, during the entire measurement period, the elements related to PM10 can be arranged in the following order: As<V<Ni<Pb<Cr<Mn<Cu<Ti<Zn<K<Fe<Ca<Al<Si<S. Trace elements, including toxic elements, such as As, V, Ni, Pb, Cr, Mn, were present in low concentrations, not exceeding 10 ng/m3 (average daily value). These elements had fairly even concentrations, daily and hourly. The concentrations of the main elements in the PM10 in the receptor are subject to strong hourly changes related not only to changes in the structure of the sources identified in the statistical analysis but mainly to wind speed and direction changes (soil and sand particles pick-up and inflow of pollutants from coal combustion). It has been shown that the transport emission in the receptor can have an intense effect on PM10 in the afternoon.

The pH-related metabolic paradigm has rapidly grown in cancer research and treatment. In this contribution, this recent oncological perspective has been laterally for the first time in order to integrate neurodegeneration within the energetics of the cancer acid-base conceptual frame. At all levels of study, molecular, biochemical, metabolic and clinical, the intimate nature of both processes appears to be opposite mechanisms occurring at the far ends of a physiopathological intracellular pH/extracellular pH (pHi/pHe) spectrum. This wide-ranged original approach now permits an increase in our understanding of these opposite processes, cancer and neurodegeneration, and, as a consequence, allows to propose new avenues of treatment based upon the intracellular and microenvironmental hydrogen ion dynamics regulating and deregulating the biochemistry and metabolism of both cancer and neural cells. Under the same perspective, the etiopathogenesis and special characteristics of multiple sclerosis (MS) becomes an excellent model for the study of neurodegenerative diseases and, utilizing this pioneering approach, we find that MS appears to be a metabolic disease even before an autoimmune one. Also within this paradigm, several important aspects of MS, from mitochondrial failure to microbiota functional abnormalities, are analyzed in depth.
